Description
(a)前記アナライトを含んでいてもよい試験サンプルと、各アリコートが異なる既知の量の前記アナライトを含む少なくとも2の異なるアナライト含有アリコートを含むキャリブレーションサンプルとを混合する工程であって、前記試験サンプルと前記キャリブレーションサンプルの各アリコートとを質量分析法によって区別できるように、前記サンプル及び各アリコートを、質量分析上種類の異なる質量マーカー基をそれぞれ有する1以上の同重質量標識体を用いて相互に異なるように標識する工程と、
(b)質量分析法によって、前記試験サンプル中の前記アナライトの量及び前記キャリブレーションサンプルの各アリコート中の前記アナライトの量を測定し、前記キャリブレーションサンプルのアリコート中の既知且つ測定されたアナライトの量に対して、前記試験サンプル中のアナライトの量を求める工程とを含むことを特徴とする方法を提供する。
(i)質量分析計において、質量標識体で標識したアナライトに相当する質量電荷比のイオンを選択しそしてフラグメント化し、フラグメントイオンの質量スペクトルを検出及び作製し、及び前記質量標識体の質量マーカー基に相当するフラグメントイオンを同定する工程と、
(ii)質量スペクトルにおけるキャリブレーションサンプルのアリコートの質量マーカー基の量に対する、同一の質量スペクトルにおける各試験サンプル中のアナライトの質量マーカー基の量に基づいて、前記各試験サンプル中のアナライトの量を測定する工程とを含むのが好ましい。
加えて、1以上のタンパク質カチオンを低エネルギー電子で中和し、結合の特異的開裂を引き起こし、例えば衝突活性化解離(CAD;衝突誘起解離(CID)としても知られる)などの他の技術によって形成されるb、y生成物とは対照的な、c、z生成物を形成させることができる。RF 3D四重極型イオントラップ(QIT)機器、四重極型飛行時間機器、及びRFリニア2D四重極型イオントラップ(QLT)機器のいずれかのRF場に導入される熱電子は、その熱エネルギーを1マイクロ秒の何分の1かの間しか保持できず、これらの装置にトラップされないので、依然としてECDは、最も高価な種類のMS機器であるFTICRで排他的に用いられる技術である。
電子移動の結果、ETDでは、ECDと同様なフラグメント化パターン、即ちいわゆるc及びzイオンの形成を生じる。電子移動方法が異なることに基づいて、ETDを、ECDには適切でない四重極型イオントラップ(QIT)機器及びRFリニア2D四重極型イオントラップ(QLT)機器のいずれかなどの様々な「低価格」質量分析計に用いることができる。適切な参考文献としては、John E.P.Syka,Joshua J.Coon,Melanie J.Schroeder,Jeffrey Shabanowitz,及びDonald F.Hunt, PNAS,第101巻,26号,pp.9528−9533を参照のこと。
X−L−M
(式中、Xが質量マーカー部分、Lが開裂可能リンカー、そしてMが質量正規化部分を示す)からなる実施形態が好ましい。Lは、単一結合であっても、Xの一部分であっても、Mの一部分であってもよい。これらの質量標識体は、その任意の部分で、例えばM、L、及びXのいずれかなどを介して、試験サンプル及びキャリブレーションサンプルのいずれかの中のアナライトに結合することができる。しかしこれらは、Mを介して結合させることが好ましく、例えば標識体は以下の構造からなる:

この例において、増感基を用いる目的は、質量分析計において質量マーカー部分の検出感度を高めるためであるので、増感基は、通常、質量マーカー部分に結合させられる。反応性官能基は増感基とは異なる部分に結合し存在するように示されているが、質量標識体はこの配置に限定される必要はなく、幾つかの場合増感基を反応性官能基と同一の部分に結合させてもよい。

7.これらの選択されたイオンの解離を、衝突及び電子移動のいずれかによって誘起する工程と、
8.質量標識体を示す衝突生成物イオンを同定するために前記衝突生成物を検出する工程と、
9.前記質量標識体を示す前記衝突生成物イオンの強度に基づいて、イオンの強度と生体分子の量の関係を示す標準曲線を作製する工程と、
10.前記生体分子及び生体分子の混合物のいずれかの絶対存在量及び相対存在量を計算する工程とを含む。
好ましい実施形態において、本発明は、上述のように定義された質量標識体であって、質量マーカー部分の分子量が300ダルトン以下、好ましくは250ダルトン以下、より好ましくは100ダルトン〜250ダルトン、最も好ましくは100ダルトン〜200ダルトンである質量標識体を使用する。質量マーカー部分の分子量は、125、126、153、及び154ダルトン、及び1以上又は全ての12C原子を13C原子で置換した場合における分子量のいずれかであることが特に好ましい。例えば、分子量が125である非置換質量マーカー部分では、その置換された化合物の質量は、1、2、3、4、5、及び6の13C原子でそれぞれ置換された場合及び1以上又は全ての14N原子を15N原子により置換した場合の少なくともいずれかにおいて、それぞれ126、127、128、129、130、及び131ダルトンとなる。

本発明において使用される質量標識体の開裂可能リンカーは、特に制限されない。開裂可能リンカーが、衝突誘起解離、表面誘起解離、電子捕獲解離(ECD)、電子移動解離(ETD)、及び高速原子衝突のいずれかにより開裂可能であるリンカーであることが好ましい。前記結合がCIDにより開裂可能である実施形態が最も好ましい。前記リンカーは、アミド結合からなってもよい。
質量標識体が所望の総質量を有することを確実にするのに適したものであれば、本発明において使用される質量標識体の質量正規化部分の構造は特に限定されない。しかし、質量正規化部分は、直鎖及び分岐のいずれかのC1−C20置換及び非置換のいずれかの脂肪族基及び1以上の置換及び非置換のいずれかのアミノ酸の少なくともいずれかからなることが好ましい。
質量分析法により生体分子を標識し検出する本発明の反応性質量標識体は、通常、上述のように定義された質量標識体、及び質量標識体の生体分子への結合又は結合を容易にする反応性官能基からなる。本発明の好ましい実施形態において、反応性官能基により、質量標識体が、好ましくはアミノ酸、ペプチド、及びポリペプチドのいずれかのアナライトと共有結合的に反応することが可能になる。反応性官能基はリンカーを介して質量標識体に結合してもよく、リンカーは開裂可能であってもなくてもよい。反応性官能基は、質量標識体の質量マーカー部分に結合しても、質量標識体の質量正規化部分に結合してもよい。
M(A)y−L−X(A)z
(式中、Mは質量正規化部分であり、Xは質量マーカー部分であり、Aは質量調整部分であり、Lは開裂可能リンカーであり、y及びzは0以上の整数であり、y+zは1以上の整数である)を有する質量標識体からなる。Mがフラグメンテーション耐性基であり、Lが他の分子及び原子のいずれかと衝突した際にフラグメンテーション感受性であるリンカーであり、Xが予備イオン化されたフラグメンテーション耐性基であることが好ましい。
(a)質量マーカー部分及び質量正規化部分の少なくともいずれかの内に位置する同位体置換基、並びに

本発明の原理を説明するために、ウシ血清アルブミン(BSA)に対する参照試薬のセットを調製した。1mgのBSAを緩衝液中に溶解し、還元し、アルキル化し、次にトリプシンで消化した。当業者であれば、タンデム型質量分析法による分析に適合性があり、トリプシンペプチドを調製するのに適切な如何なる方法も使用できることを十分に理解するであろう。

各分析のためには、10μLの参照材料を試験サンプルに添加し、これにより参照TMT−標識消化物の量が0.156μg、0.469μg、1.406μg、及び4.219μgとなるようにした。
実施例1において調製したBSA標準を用いた定量の正確度を、既知のBSA濃度を有する1連の溶液にBSA標準溶液を添加して分析することによって判定した。
粒子サイズが3μm、流速が300μL/minでMicromass QTOF IIに連結した、サイズが75μm、150mmのRP−C18カラムを有するWaters Cap−LCからなる本発明者らのパイプラインを介して、MS/MSデータを発生させた。MS/MS実験は、データ依存測定モード(DDA)により実施した。稼動の間、MS/MS実験を、1MSスキャンに対し1.0秒のデータ収集時間、及び続くそれぞれ1.4秒の4種の最も存在量が多かったイオン種の4連続MS/MSスキャンという条件で実施した。図2に、BSAトリプシンペプチド AEFVEVTK のMS/MSプロフィールを示す。上段にフルMS/MSスペクトルを示す。下段に同重質量マーカー部分領域の拡大表示を示すが、ここでは同一ペプチドの試験サンプル(126)中及び参照材料(128、129、130、及び131)中での異なる存在量を反映した異なる強度のピークが示されている。
10個の粗ヒト血漿サンプルを使用した。
本発明を使用して定量すべき対象のアナライトは、タンパク質のクラスタリンとした。以下に示すアミノ酸配列を有する1個のクラスタリンペプチドを、参照物質として使用した。

1.66mgのペプチドから1μg/μLのペプチドストック液を製造した。それぞれ200μLの4ポーションを、それぞれTMT6−128、TMT6−129、TMT6−130、及びTMT6−131で標識した。これらのペプチドをNH2OHで処理し、生じ得るチロシン、セリン、及びトレオニンへの標識化を元に戻した。次に、相互に異なるように標識したペプチドサンプルを、1:2:4:8の比で混合した。図6は、使用した方法を例示する概略図である。LC−MS/MSによる初期の分析から、部分的に生じる過剰標識化状態を完全に元に戻すことはできず、従って第2の処理が必要であることが示された。このキャリブレーションサンプルは、TMT6−128標識血漿サンプルに混合する前に、528倍希釈した。
ELISAによって測定されるクラスタリン含量に基づいて、10個の血漿サンプルをコホートから選択した。1.66μLの各血漿サンプルを198.33μLの緩衝液(100mM TEAB、0.1%SDS、pH 8.5)で希釈し、全タンパク質量を142μg〜200μg(タンパク質濃度は、0.71μg/μL〜1.00μg/μL)とした。次に、各血漿サンプルをTMT6−126で標識し、最適条件に従ってNH2OH処理を実施した。次に、4μLの希釈キャリブレーションサンプルを10%の各サンプルに添加した。次に、このサンプルを逆相クロマトグラフィー並びに強カチオンイオン交換クロマトグラフィーによって精製した。
LC−MS/MSを、Qtof−2質量分析計(Waters,マンチェスター、イギリス)に連結したCapLCを用いて実施した。
1稼動当り5μLの精製サンプル(40nLの粗血漿に相当)を注入した。図7は、クラスタリン由来の標識ペプチドの保持プロフィールの質量スペクトルである。クラスタリン由来TMT−標識ペプチドから標的とするものを選んで行うMS/MSデータ収集を、次に、クラスタリン由来のTMT−標識ペプチドのm/zのリストを用いて実施した。衝突エネルギーパラメーターの最適化を行い、質量マーカー基イオン強度を増加させた。図8は、クラスタリン由来の標識ペプチドのMS/MSスペクトルである。挿入図は、質量マーカーイオンを示すMS/MSスペクトルの領域を示す。
全ての対応するMS/MSスキャンを手作業で蓄積し、1稼動当り1個のMS/MSファイルを作製した。次に、ピーク処理及びIDを、標準的方法を使用して実施した。各MS/MSファイルに対して、1本の検量線を、質量マーカーイオン強度128〜131(直線回帰)に基づいて作製した(図9)。各サンプル中に存在するペプチドの量を、次に、前記検量線を使用して決定した(表3)。最後に、μg/mLで表したサンプル当りのクラスタリン濃度を、クラスタリンα鎖の分子量に基づいて計算した。
多くの状況下、例えば初期バイオマーカーの発見ワークフローにおいては、絶対定量参照標準を準備することは必ずしも必要ではなく、むしろ代表的、均一な標準であって、如何なる所定のアナライトの絶対量も知られておらず、参照サンプルの通常の範囲内にあると考えられる分析する全プロテオームをカバーする標準が準備される。このような全プロテオーム標準の1例が、ヒト血漿である。本発明を使用することにより、全てのタンパク質及びペプチドの少なくともいずれかが同重標識多重定性標準として存在する、一般的で均一な参照標準血漿を調製することが可能である。このような標準を、全てのタンパク質及びペプチドの少なくともいずれかを前記参照標準血漿に用いたものと同一の同重標識体のセットからの異なる標識体で標識した試験サンプルに添加する場合、MSにおいて検出される全ての前駆体イオンについての定量的MS/MS分析を実施することが可能であり、そして試験サンプル中のアナライトの参照標準に対する相対存在量を決定することも可能である。
1)リボヌクレアーゼA、タイプI−AS:ウシ膵臓から(Sigma、R−5503)、分子量13.7kDa;pI 9.6;純度85%。リボヌクレアーゼA、タイプI−ASを1.8mg、170μLの水に溶解し、この溶液の10μLを、高存在量タンパク質の除去前の血漿1mLに添加した。
2)トリプシンインヒビター、タイプI−S:ダイズから(Sigma、T−9003);分子量20.1kDa;pI 4.5;タンパク質含量90%;α鎖分子量20,090Da;β鎖分子量20,036Da;γ鎖分子量20,163Da。トリプシンインヒビター、タイプI−Sを8.8mg、196μLの水に溶解し、この溶液の10μLを、高存在量タンパク質の除去前の血漿1mLに添加した。
タンパク質処理を、以下の標準プロトコールに従い実施した。タンパク質を希釈して、100mMホウ酸塩緩衝剤及び0.1%ドデシル硫酸ナトリウム中に溶解したpH7.5の1g/Lタンパク質溶液とした。システインの1mM TCEPによる還元を、室温で30分間かけて実施した。次に、このシステインを、ヨードアセトアミドを用いて、室温で1時間かけてアルキル化した。最後に440μgのトリプシンを添加し、37℃で18〜24時間、インキュベートした。
トリプシン消化の後、タンパク質除去血漿を4個のアリコートに分け、各アリコートを独立に異なる同重標識体TMT6plex試薬、即ちTMT6−128、TMT6−129、TMT6−130、及びTMT6−131で標識した。標識処理の後、アリコートをそれぞれ1:2:4:8の体積比で混合し、血漿の4点キャリブレーション混合物を作製した(図1)。この参照材料のプロテオームカバー率を測定するために、この参照材料を多次元クロマトグラフィー及びタンデム型質量分光分析法によって分析した。
質量分析法に先立って、SCXカラム(Poly LC、4.6mm(内径)×100mm;ポリスルホエチルA)に装着したBioCAD Vision HPLC(Applied Biosystems)を用いて、第1の分離を実施した。サンプルをWaters Sunfire RP PreColumn(4.0mm(内径)×10mm)に捕獲し、50%アセトニトリルのパルスを与えることでSCXカラムへ溶出した。PreColumnをオフラインに切替え、SCXカラムのための勾配をかけた溶出を開始した。SCX勾配は、溶媒C(水75%、アセトニトリル25%、KH2PO4を5mM、pH3)と30分間で濃度を0%から50%に変化させる溶媒D(水75%、アセトニトリル25%、KH2PO4を5mM、pH3、KClを500mM、pH3)とを用いて形成した。この分離工程で、24のSCX分画を採取した。各分画を、次に逆相分離にかけた。
第2の分離システムは、Waters nanoAQUITY UPLC Systemによる逆相クロマトグラフィーである。MALDIワークフローにおいては、クロマトグラフィーとMS測定が連結されていないので、HPLCの条件を高ピーク容量が得られるように最適化することができた。カラム:75μm(内径)×250mm、1.7μmBEH 130 C18充填剤(Waters,part Nr.:186003545)を充填。カラムオーブン温度60℃。5μLの各SCX分画を、如何なるプレカラムも装着していないUPLCカラムに、直接注入した。
nanoAQUITY UPLC Systemの前記分離カラムを、Dionex Probot spotterの注入口に連結し、MALDI調製物中のペプチドを、4800 Tof/Tof MALDI機器のためのMicrotiter format MALDIサンプル標的上で分画した。MALDI標的上においては、1920の個々の分画を採取することができる。1のMALDI標的に対してそれぞれ480のスポットを有する4のRPクロマトグラムを作製した。スポッティングは、保持時間50分から開始し、保持時間210分で終了し、1のスポットに対して20秒間のスポッティング持続期間で実施された。前記スポッター上で、0.35μL/minの溶離液流に0.6μL/minのMALDIマトリックス溶液流(80%アセトニトリル中に溶解したα−シアン−4−ヒドロキシ桂皮酸の5g/L溶液、19.8%の水、0.2%のトリフルオロ酢酸)を混合した。各MALDI調製物の体積は、約330nLであった。
4800 Tof/Tof機器で、スポットに対しては2のモードの稼動が実施された。第1の稼動においては、リフレクターモードの従来からのMSスペクトルが記録された。個々のMSスペクトルそれぞれに対して、合計1,000のMALDIショットが行われた。スペクトルを、m/zが568.138のマトリックストリマーシグナルを用いて内部で較正した。ペプチドピークの溶出プロフィールの計算を含むLC MALDIストラテジー(分画対分画の質量トレランスが100ppm、200解像度内の前駆体の排除)を用いて、“4000 Series Explorer”機器ソフトウェアの解釈ツールにより、MS/MS実験の前駆体リストが発生させられる。後のMS/MS分析のために選択される前駆体として、1個のスポットに対して5個以下の前駆体が与えられた。MS/MSデータ収集を、最初に最も強い前駆体から行った。1のスペクトルに対して1,000のレーザーショットが必要であった。このMS/MSスペクトルを、TMT6−131フラグメントイオンの理論質量値131.1387を用いて内部で較正した。
15,818クエリーのデータベース検索をMASCOT Version 2.1.04を用いて実施した。ヒトタンパク質をIPIヒトデータベース(IPI_Human_20071024; 68348配列; 28969400残基)を用いた検索において同定した。添加したタンパク質は、両方ともIPIヒトデータベース中には存在しなかった;これらは、Swissprotデータベースにおいて、「他の哺乳類」という分類上のキーを使用して検索した。質量126、127、128、129、130、131のピーク面積を、Sequest ToolboxからのTOFTOF Matcherにより抽出した。
<定量分析>
GPS Oracle データベースからのピーク面積の抽出によって、質量128Da、129Da、130Da、及び131Daに存在するレポーターピークの計量を行った。ピーク面積の計算の後、検量線(1:2:4:8の比)の品質を確認するために、回帰分析を行った。レポーターピークの直線への適合について解析することによって、各ペプチドMS/MS実験の成功度を調べた。全てのMS/MSスペクトルについて、直線回帰が達成された。
Claims (45)
- アナライトを分析する方法であって、
(a)前記アナライトを含んでいてもよい試験サンプルと、各アリコートが既知量の前記アナライトを含む少なくとも2個の異なるアナライト含有アリコートを含むキャリブレーションサンプルとを混合する工程であって、前記試験サンプルと前記キャリブレーションサンプルの各アリコートとを質量分析法によって区別できるように、前記試験サンプル及び各アリコートを、質量分析上種類の異なる質量マーカー基をそれぞれ有する1以上の同重(isobaric)質量標識体を用いて相互に異なるように標識する工程と、
(b)質量分析法によって、前記試験サンプル中の前記アナライトの量及び前記キャリブレーションサンプルの各アリコート中の前記アナライトの量を測定し、前記キャリブレーションサンプルのアリコート中の既知且つ測定されたアナライトの量に対して、前記試験サンプル中のアナライトの量を較正する工程とを含むことを特徴とする方法。 - 試験サンプルが複数の異なるアナライトを含むことができ、各異なるアナライトに対して1個のキャリブレーションサンプルが与えられ、各異なるアナライトに対して工程(b)を繰り返す請求項1に記載の方法。
- 複数のアナライトが、工程(a)より前にタンパク質及びポリペプチドのいずれかの化学的処理及び酵素的処理のいずれかによって生成される前記タンパク質及びポリペプチドのいずれかのペプチド断片である請求項2に記載の方法。
- 各キャリブレーションサンプルの異なるアリコートは、前記各キャリブレーションサンプルのアナライトの量の範囲が他のキャリブレーションサンプルと異なるように選択される請求項2から3のいずれかに記載の方法。
- 複数の試験サンプルにおいて1のアナライトを分析する請求項1から4のいずれかに記載の方法。
- 複数の試験サンプルにおいてそれぞれ分析されるアナライトが同一である請求項5に記載の方法。
- 試験サンプルのそれぞれを、1以上の同重質量標識体で相互に異なるように標識し、工程(a)において単一のキャリブレーションサンプルと混合し、工程(b)において各サンプル中のアナライトの量を同時に測定する請求項6に記載の方法。
- 各試験サンプルを同一の質量標識体で標識し、各異なる試験サンプルに対して工程(a)及び(b)を繰り返す請求項6に記載の方法。
- 分析される各試験サンプルに対して同一のキャリブレーションサンプルを使用する請求項8に記載の方法。
- 工程(a)より前に、1以上の同重質量標識体で、各試験サンプル及びキャリブレーションサンプルの各アリコートのいずれかを相互に異なるように標識する工程を更に含む請求項1から9のいずれかに記載の方法。
- 工程(a)より前に、相互に異なるように標識したアリコートを混合してキャリブレーションサンプルを調製する工程を更に含む請求項10に記載の方法。
- 工程(b)が、
(i)質量分析計において、質量標識体で標識したアナライトに相当する質量電荷比のイオンを選択しそしてフラグメント化し、フラグメントイオンの質量スペクトルを検出及び作製し、及び前記質量標識体の質量マーカー基に相当するフラグメントイオンを同定する工程と、
(ii)質量スペクトルにおけるキャリブレーションサンプルのアリコートの質量マーカー基の量に対する、同一の質量スペクトルにおける各試験サンプル中のアナライトの質量マーカー基の量に基づいて、前記各試験サンプル中のアナライトの量を測定する工程とを含む請求項1から11のいずれかに記載の方法。 - キャリブレーションサンプルの各アリコート中のアナライトの量が既知の絶対量である請求項1から12のいずれかに記載の方法。
- 試験サンプル中のアナライトの絶対量を工程(b)において決定する請求項13に記載の方法。
- キャリブレーションサンプルの各アリコート中のアナライトの量が既知の定性的量(qualitative quantity)である請求項1から12のいずれかに記載の方法。
- 定性的量が、特定の状態を有する被検体中のアナライトに予測される範囲の量である請求項15に記載の方法。
- 較正工程が、キャリブレーションサンプルのアリコート中のアナライトの既知の定性的且つ測定された量に対して試験サンプル中のアナライトの量を較正する工程を含む請求項15から16のいずれかに記載の方法。
- 試験サンプル中のアナライトの量のパーセント変化率を決定する請求項17に記載の方法。
- 各異なるアリコート中のアナライトの量が、試験サンプルにおけるアナライトの量の既知の変動及び推測される変動のいずれかを反映するようにして、選択される請求項1から18のいずれかに記載の方法。
- 健常被検体及び罹患被検体のいずれかの試験サンプル中に見出されるアナライトの既知の量及び推測される量のいずれかの範囲内の上限及び下限、並びに任意に中間点に相当する量のアナライトを含むアリコートが提供される請求項19に記載の方法。
- 異なるアリコート中に存在するアナライトの異なる量が、異なる期間インキュベートした試験サンプル中に存在するアナライトの既知の量及び推測される量のいずれかに相当する請求項1から20のいずれかに記載の方法。
- アリコートが試験サンプルの標準化形態のサンプルから採取される請求項1から21のいずれかに記載の方法。
- 試験サンプル及びキャリブレーションサンプルのアリコートの少なくともいずれかが植物由来及び動物由来のいずれかである請求項1から22のいずれかに記載の方法。
- 動物がヒトである請求項23に記載の方法。
- キャリブレーションサンプルが、治療の有効性及び毒性の少なくともいずれかを示す量のアナライトを含む請求項1から24のいずれかに記載の方法。
- 試験サンプル及びキャリブレーションサンプルの少なくともいずれかが、ヒト及び動物のいずれかの組織、血液、血漿、血清、脳脊髄液、滑液、眼液、尿、涙、涙管滲出物、肺吸引物、母乳、乳頭吸引液、精液、洗浄液、細胞抽出物、組織培養抽出物、植物組織、植物体液、植物細胞培養抽出物、細菌サンプル、ウイルスサンプル、真菌、発酵もろみ液、食料品、及び医薬組成物のいずれかである請求項1から25のいずれかに記載の方法。
- アナライトがタンパク質、ポリペプチド、ペプチド、アミノ酸及び核酸のいずれか、ペプチド核酸、糖、澱粉、複合糖質、脂質、高分子、並びにこれらの断片のいずれかを含む請求項1から26のいずれかに記載の方法。
- 工程(a)の後であるが工程(b)より前に、同重標識したアナライトを電気泳動及びクロマトグラフィーのいずれかによって分離する工程を更に含む請求項1から27のいずれかに記載の方法。
- キャリブレーションサンプルが、MSスキャンの間及びMS/MSスキャンを開始するための非スキャンMS/MSの間のいずれかの間にトリガーの役目を果たす量のアナライトを含む更なるアリコートを、含む請求項1から28のいずれかに記載の方法。
- 更なるアリコート中のアナライトが同重質量標識体で標識されている請求項29に記載の方法。
- 更なるアリコート中のアナライトが、キャリブレーションサンプルのその他のアナライトの同重質量標識体と化学的には同一だが異なる同位体を有し質量が異なる質量標識体で、標識されている請求項29に記載の方法。
- 試験サンプル中のアナライトがタンパク質であり、キャリブレーションサンプル中のアナライトが前記試験サンプル中の前記タンパク質の組み換え型である請求項1から31のいずれかに記載の方法。
