Description
FcγRIIIAは、特にIgG結合ドメインの48位および158位において、いくつかの対立遺伝子多型を示すことが知られている。前記の抗CD16抗体は、異なるFcγRIIIAの多型フォームに異なる親和性で結合することが示され、Koeneらの研究(1997、Blood 90:1109−1114)は、これらの抗体のうちのいくつかの結合の違いの原因は158位での多型であるとしている。FcγRIIIA158FアレルおよびFcγRIIIA158Vアレルの遺伝子頻度は、それぞれ約0.6および0.4であるので、任意のCD16A抗体について、両方のアレル型を認識することは、あらゆる患者において、抗CD16A 抗体がNK細胞集団に結合し活性化することを確実にする。
好ましい実施態様において、FcγRIIIAに対する特異性は、抗体またはその抗原結合断片によって与えられる。したがって、本発明の結合分子は、FcγRIIIAに特異性を有するが、FcγRIIIBに特異性を有さない、少なくとも一つの抗体(またはその抗原結合断片)を含有するか、ある実施態様においては、少なくとも一つの抗体(またはその抗原結合断片)からなる。

本発明の更なる態様は、FcγRIIIA158FアレルおよびFcγRIIIA158Vアレルにほぼ同じ親和性で結合可能な、前記の態様のいずれか1つ記載の結合分子を提供する。すなわち、FcγRIIIAの2つの158アレルに対する前記結合因子の親和性の違いは2倍以下である。本発明の本態様にしたがった好適な具体例において、FcγRIIIAに対する特異性は、本明細書に記載の4−LS21 scFv由来の抗体または抗原結合断片によって与えられる。本発明のこの態様に従った好ましい実施態様では、FcγRIIIAに対する特異性は、本明細書中に記載の、4−LS21 scFvに由来する抗体または抗原結合断片によって与えられる。特に好ましい実施態様においては、本発明の当該態様に従った結合分子は、TandAb型であり、本明細書中に記載の4−LS21 scFvに由来する抗原結合断片およびCD19またはCD30に対する特異性を有する抗原結合断片を含有する。
上記の方法に代えて、または加えて、免疫原性を減らすために用いられうるもう1つの技術が、デイムニゼーション(deimmunisation)の技術である。デイムニゼーション技術は、抗体および他のタンパク質生物学治療剤からの、Tヘルパー(Th)細胞エピトープの同定と分離を伴う。Th細胞エピトープは、MHCクラスII抗原に結合する能力を有する、タンパク質の中の短いペプチド配列を含有する。ペプチド−MHCクラスII複合剤はT細胞によって識別されることができ、そしてTh細胞の賦活化と分化も引き起こすことができ、これは、B細胞との相互作用を介する免疫原性の獲得と維持に必要とされ、投与された生物学的治療剤に特異的に結合する抗体の分泌物をもたらす。抗体デイムニゼーションの目的で、Th−細胞エピトープは、例えばペプチドのヒトMHCクラスII抗原への結合を予測するコンピューターに基づく方法によって抗体配列の中で識別される(Altuviaら,1995,J.Mol.Biol.,249:244−2450,およびSchueler−Furmanら,2000,Protein Sci.,9:1838−1846を参照)。T細胞による認識を避けるために、このようにして同定されたTh細胞エピトープは、アミノ酸置換によってタンパク配列から排除される。これは、例えば、治療用タンパク質におけるTh細胞エピトープをコードする核酸配列を改変する部位特異的突然変異誘発法のような標準的な分子生物学テクニックの使用を通じて達成できる。このようにして、HAMA(ヒトの抗マウス抗原性)および/または抗イディオタイプ応答が抑制または回避されるように、抗体または抗原結合断片を修飾できる(Banderら,2003,Semin.Oncol.,30:667−676;Nanusら,2003,J.Urol.,170:S84−S89)。このように、更なる実施態様では、本発明の結合分子は、修飾されて、それらの配列に存在しているTh細胞エピトープが除去されている。本明細書中、このような結合分子を、デイムナイズされた結合分子と称する。
更なる態様では、本発明の結合分子を、標識分子、あるいは毒素に結合させることができる。標識分子または毒素がタンパク質である場合、結合分子への結合は、ペプチド結合または化学的結合によって生じさせうる。したがって、本発明の当該態様によれば、結合分子は、標識分子または毒素が、ペプチド結合、好ましくはペプチドリンカーによって結合分子に連結された融合タンパク質の形態であるか、または化学的複合体の形態であってもよい。疑義の回避のためであるが、本明細書中、複合体なる用語は、2つのコンポーネントが、ペプチド結合(したがって、複合体は融合タンパク質を包含する)、化学結合、エステル結合、またはジスルフィド架橋によって物理的に連結されていることを意味するために用いられる。
放射能標識または螢光もしくは発光標識(化学発光を含む)のような標識分子への本発明の結合分子の結合は、当該結合分子が免疫学的染色試薬として使われることを可能にする。このような試薬は、例えば、FcγRIIIAを発現している組織潜入NK細胞、マクロファージ、またはマスト細胞の検出に、あるいは当該結合分子が更なる抗原に特異性を示す場合は、NK細胞−結合分子−更なる抗原の複合体の検出に用いられうる。後者の検出は、疾患の診断、または疾患の進行もしくは寛解のモニタリングにおいて、特に有用たりうる。

適当な細菌の分泌シグナルは技術文献に記載されており、そのような一例は、PelBリーダー配列である。他の態様では、例えば、真核生物での発現に関連して、結合分子の発現が小胞体(ER)で保持されることが望まれうる。この場合、シグナル配列は“KDEL”(配列番号54)モチーフ、特に“SEKDEL”(配列番号55)コードする配列をコードするER保留シグナルを含有する。
本発明のポリヌクレオチドはまた、選択された特定の発現宿主に適合するように変更されたコドンバイアスを用いて、当該技術分野で容易に利用可能な方法に従って最適化された、コドンであってもよい。
細菌細胞では、適当な技術としては、塩化カルシウム形質転換、エレクトロポレーション法、またはバクテリオファージを使ったトランスフェクションが挙げられる。植物細胞では、形質転換される特定の植物種に依存して、アグロバクテリア媒介形質転換、エレクトロポレーション法、植物細胞およびプロトプラストの微量注射、微粒子銃、細菌ボンバートメント(特に、「ファイバー」法または「ウィスカー」法)が挙げられる。したがって、更なる態様では、本発明は、本発明のポリヌクレオチドまたはベクターを含有する宿主細胞を提供する。したがって、下記の生物体に由来し、本発明のポリヌクレオチドまたはベクターを含有する宿主細胞は、当該態様の具体例であると考えられる。

分泌・可溶型のCD16A−Fc融合タンパク質を、ヒトIgG1のFc部に融合したヒトCD16AをコードするプラスミドpCDM−CD16−Fc(Mandelboimら,1999、上掲)のトランジェントな形質転換の後、HEK−293細胞中で生産した。
CD16A−Fc融合タンパク質を用いて、大分子ヒトIgM由来scFv ファージディスプレイライブラリーのスクリーニングを行った。これは、前記と同様の方法を用いて生産した(Doersamら,1997 FEBS Letts 414:7−13、Littleら,1999 J.Immunol.Methods 231:3−9、Schwarzら,FASEB J.18:1704−6)。当該ライブラリー中のクローンは、M13ファージのpIII遺伝子とscFvコード配列との間のヌクレオチド配列を含んだ。これは、ヘキサヒスチジンタブ、アンバー終止コドン、およびc−mycエピトープをコードする。
クローン50NI由来のペリプラズマ抽出物を前記のように調製し、CD16A−Fc 融合タンパク質およびヒトIgGへの結合能を試験した。抗−CD16マウスモノクローナル抗体A9(Hombachら,1993 Int.J.Cancer 55:830−836)を対照として用いた。Maxisorb(商品名)ELISAプレートを、ウェルあたり、100mM NaHCO3,pH8.6中、500ngのストレプトアビジンおよび300ngのヒトIgG(Sigma−Aldrich社,Taufkirchen,ドイツ)でプレコートした。次いで、300ngのビオチン化CD16A−Fcを30分間、室温、PBS中で捕捉した。
結果を図1に示した。MAb A9およびscFv 50NIは明らかにビオチン化ストレプトアビジン捕捉CD16A−Fc融合タンパク質を認識した。scFv 50NIは、ヒトIgG上のシグナルもストレプトアビジン単独のシグナルもどちらも示さなかった。2次試薬として用いた抗−ペンタHis HRP−複合体は、抗原への非特異的結合を示さなかった。しかし、MAb A9 検出用の2次試薬として用いたヤギ抗−マウス−HRP複合体は、CD16融合タンパク質のヒトFc部およびネガティブコントロールとして用いたヒトIgGとの交差反応性によるバックグラウンド染色を起こした。これらの結果は、scFv 50NIが当該融合タンパク質のCD16A部に特異性を示し、Fc部に特異性を示さないことを強く示している。
50NI scFvが細胞表面上のネイティブCD16Aタンパク質への結合を示すかどうかを確かめるため、scFvの粗精製ペリプラズム抽出物を、それぞれCD16AまたはCD16Bを発現する2つの異なるセルラインにおけるフローサイトメトリーによって試験した。この目的のため、CD16Aを発現するセルラインBW/CD16A(Mandelboimら,2001,Nature 409:1055−1060)およびセルライン293−CD16(ヒトCD16BアイソフォームのNA−2対立形質をコードするプラスミドで安定に形質転換されたHEK−293細胞)を用いた。50NI scFvがナチュラルキラー(NK)細胞の表面上で発現するCD16Aおよび/または好中球顆粒球の表面上に存在するCD16Bへの結合能を有するかどうかを評価するため(van de WinkelおよびCapel,1993,上掲;van Sprielら,2000,Immunol.Today 21(8):391−397)、新たに単離した10−20%のNK細胞を含む末梢血単核球細胞(PBMC)、および95%以上のCD16Bポジティブの好中球顆粒球を表す多形核細胞(PMN)への結合を、フローサイトメトリーを用いて試験した。マウスMAb A9およびマウスクローンscFv18 A9(MAb A9から得られたscFv)由来のペリプラズム抽出物をコントロールとして用いた。
BW/CD16A細胞を、10%の熱不活性化ウシ胎児血清(FCS)、2mMのL−グルタミン、100U/mLのペニシリンGナトリウム、および100μg/mLの硫酸ストレプトマイシン、1%の非必須アミノ酸、および5mg/mLのG418を添加したDMEM培地中で培養した。293/CD16B細胞を、10%の熱不活性化ウシ胎児血清(FCS)、2mMのL−グルタミン、100U/mLのペニシリンGナトリウム、100μg/mLの硫酸ストレプトマイシン、および5mg/mLのG418を添加したDMEM培地中で培養した。非形質転換の親セルラインBWおよび293を、G418を欠く培地である以外は、安定形質転換体についての前記のように培養した。
粗精製のscFv含有ペリプラズム抽出物を5mLの細菌培養物から単離した。新たに接種した細菌を、50mMのグルコース、100μg/mLのアンピシリンおよび20μg/mLのテトラサイクリンを含有するLB培地中、37度で一晩生育させた。一晩培養した培養物をアンピシリンおよびテトラサイクリンを含有するLB培地でOD600nm=0.1まで希釈し、更に37℃でOD600nm=0.8まで生育させた。この細菌を遠心分離によって回収し、0.1mM イソプロピル−β−D−チオガラクトピラノシド(IPTG)、100μg/mL アンピシリン、および20μg/mL テトラサイクリンを含有するLB培地に再懸濁した。形質導入細菌の21℃での一晩のインキュベーション後、300μLのペリプラズム抽出物をELISA用に前記のように単離した。FACS分析での使用の前に、ペリプラズム抽出物をPBSに対して一晩透析した。
フローサイトメトリー分析用に、1x106細胞のBW、BW/CD16A、HEK−293、293/CD16B、PBMCおよびPMNを、モノクローナル抗体A9、またはscFv18A9および50NIを含有するペリプラズム抽出物で染色した。1x106個の細胞を100μLの透析した抽出物またはMAb A9(10μg/mL)とともに45分間、氷上でインキュベーションした。PBMCおよびPMNとの抗体/scFvインキュベーションに、1mg/mLのポリクローナルのヒトIgGを添加し、PBMCおよびPMNのサブ集団の表面上のFc受容体をブロックした。FACS緩衝液(2%の熱失活FCSおよび0.1%のアジドナトリウムを添加したPBS)での洗浄の後、細胞を氷上で45分間、0.1mLの0.01mg/mL 抗−(His)6マウスMAb 13/45/31−2(Dianova社,Hamburg,ドイツ)を用いて、同じ緩衝液中で、インキュベーションした。第2の洗浄サイクル後、細胞を同じ条件下で、0.1mLの0.015mg/mL FITC−複合ヤギ抗−マウスIgG抗体(Dianova社,Hamburg,ドイツ)を用いてインキュベーションした。次いで、この細胞を再び洗浄し、0.5 mLの2μg/mL ヨウ化プロピジウム(Sigma−Aldrich社)含有FACS緩衝液に懸濁して、死細胞を除去した。1×104個の染色細胞の蛍光をBeckman−Coulter EpicsXLフローサイトメーターを用いて測定した。平均蛍光強度をSystem−IIおよびExpo32ソフトウェア(Beckman−Coulter社,Krefeld,Germany)を用いて計算した。第2試薬のみの細胞のバックグラウンド染色によって生じる蛍光を結果および分析の補助のためにプロットしたヒストグラム(図示せず)から差し引いた。
マウスモノクローナル抗−CD16抗体A9およびポジティブコントロールとして用いたA9誘導抗−CD16 scFv18は、非形質転換親細胞(BW)との比較においてBW/CD16Aへの明らかな結合を示した。scFv 50NIは、A9に匹敵するレベルでBW/CD16A細胞の明確な染色も示した。
実施例3に示すように50NIによって認識されるPBMCサブ集団がナチュラルキラー(NK)細胞を含有することを示すため、NK細胞の高濃度化集団PBMCから単離し、フローサイトメトリーによってscFv 50NIへの結合を分析した。健常ドナーのヘパリン化末梢血から密度匂配遠心によってPBMC類を単離した。血液サンプルをPBSで2回洗浄し、Histopaque−1077(Sigma−Aldrich社)のクッション上に重層し、800gで25分間、遠心分離した。界面中に位置するPMBCを回収し、PBSで3回洗浄した。NK細胞の単離のため、PBMCを2%のFCS/PBS中に再懸濁し、製造者の説明書に従い、EasySEP(商品名)ネガティブNK細胞濃縮キット(CellSystems社)を用いた1回のネガティブ選択に付した。PBMCおよびNK細胞の染色ならびにサイトメトリー分析を本質的に前記実施例3の記載と同様に行った:1×106個の細胞を10μg/mLのMAb A9または抗−CD56モノクローナル抗体MAb B159(BD Biosciences/Pharmingen社)、ついで15μL FITC−複合化ヤギ抗−マウスIgGで染色した。マウスscFv18 A9およびヒトscFv 50NIを50μg/mLの濃度で染色に用い、細胞に結合したscFvを10μg/mL MAb 抗−(His)6、ついで15μL FITC−複合化ヤギ抗−マウスIgGで検出した。全ての染色は、1mg/mLのポリクローナル ヒトIgGの存在下で実施した。死細胞を前記のようにヨウ化プロピジウム染色によって除外した。データ分析は、前記の実施例3に記載のように実施した。
抗−CD56抗体および抗−CD16抗体によるPBMCの染色は、NK細胞フラクションを示す小さなサブ集団を明確に同定した。scFv 50NIは、NK細胞への結合を示す類似した小さな細胞集団もまた認識した。
NK細胞のネガティブな高濃度化の後、全ての4つの抗体/scFvは明確な染色を示した。抗CD56 MAbが完全なNK細胞集団への結合をしめした一方で、細胞の小さなフラクションは、CD16に対する抗体を用いて染色されなかった。これは、CD16−ネガティブの表現型を有するいくらかの細胞の存在を反映している可能性がある。
IgGに対する低親和性受容体であるCD16Aは、FcεRI由来のγ鎖またはTCR/CD3由来のζ鎖を含有する免疫受容体チロシンベース活性化モチーフ(ITAM)に関連して、多数のヒト末梢血NK細胞上で発現する。抗体または免疫複合体へのCD16Aの結合は、NK細胞傷害性およびサイトカイン分泌のきっかけとなる。この活性化は、γ鎖またはζ鎖の各々のリン酸化、およびこれに続く細胞内のシグナル変換(例、Ca2+動員)のきっかけとなる。
新たに単離したNK細胞で得られた蛍光トレーシングは、明確に、MAb A9が、抗−マウスIgG抗体との架橋において細胞内カルシウム濃度の強い増加を誘起することを示した。これは、より高い蛍光シグナルによって反映される。MAb A9と対照的に、A9由来のscFv(scFv18 A9)は、抗−(His)6、抗−c−mycおよび抗−マウスIgG抗体との架橋後も、ほとんどまたは全く細胞内カルシウムの増加を誘導しなかった。しかし、ヒトscFv 50NIは、細胞表面に結合したscFvの、架橋抗体への連結の後に、明確な(しかし若干弱い)蛍光シグナルを誘導した。このことは、ヒトscFv 50NIが、CD16Aを介するNK細胞活性の作動のために適当な結合特性を有することを示す。
親和性が増加している抗−CD16 scFvを得るため、50NI VH 鎖のVL遺伝子レパートリーとの再編成により、クローン50NIに基づくチェーン・シャッフルド・ライブラリー(chain shuffled library)を生産した。VκおよびVλ遺伝子レパートリーを、前記の巨大なヒト・ナイーブscFvライブラリー(Schwarzら,上掲、2004)を由来とするpEXHAM1 DNAから切り出した。このレパートリーは、それぞれ約105の個々のVκおよびVλ鎖を反映する。pEXHAMのMluIおよびNotI部位へのクローニングによる50NI重鎖を用いた再編成と、1μgのE.coli XL1 blueへの形質転換(1.8kV,0.1cmギャップキュベット、25μF、200Ω)により、2.6×106(Vκ)および2.2×106(Vλ)の数のライブラリーを得た。
形質転換後にランダムに選択した32クローンの配列分析は、各クローンが個々の軽鎖を含有することを示し、および24クローンが、抗−His抗体を用いたウェスタン・ブロット(Qiagen社,Hilden,ドイツ)において、検出可能な完全長のscFvの発現を示した。

他の全てのグループは、親クローンに比べて増強したELISAシグナルを示す新たに編成された軽鎖を含有する。グループCおよびDからのクローンは、第3回および第4回の選択後、同程度の数が見出された。
V−ベース・アラインメント・ソフトウエア(http://vbase.mrc−cpe.cam.ac.uk)を用いた配列の分析により、全ての新たなVλ鎖が、親クローンのVL鎖と同様に、VL3ファミリーに属することが明らかになった。
全てのクローンが持つ共通のVH鎖のアミノ酸配列は、本明細書中で、配列番号9として、ならびにクローン50NI,3−LB3,3−LB5,3−LB6,3−LS49,4−LS14および4−LS21の特異的VL鎖は、本明細書中で、それぞれ配列番号10〜16として与えられる。共通のVH鎖をコードする核酸配列は、本明細書中で、配列番号1として、ならびにクローン50NI,3−LB3,3−LB5,3−LB6,3−LS49,4−LS14および4−LS21の個々のVL鎖は、本明細書中で、それぞれ配列番号2〜8として与えられる。個々の重鎖および軽鎖CDRは表1に与えられ、共通の重鎖CDRは、この上の最初の項目として示され、ついでクローン50NI,3−LB3,3−LB5,3−LB6,3−LS49,4−LS14および4−LS21の軽鎖CDRが示されている(それぞれ、項目2〜8として示される)。

サブクローンを含有するベクターを、E.coli RV308に形質転換し、2L振とうフラスコ培養で発現させ、Le Gallら(2004,J.Immunol.Methods 285:111−127)に従い、固定化金属イオン親和性クロマトグラフィーによって精製した。
改良scFvが、真核細胞で発現したCD16Aへの特異的結合能を保持しているかどうか評価するため、ドナー血液から新たに単離したPMNおよび強化NK細胞を前記のようにIMAC−精製scFvを用いて染色した。NK細胞の濃縮は、前記のネガティブ選択によって実施した。フローサイトメトリー分析で得られた平均蛍光強度を図5に示した。
scFvのCD16A特異性に関する更なるサポートデータを得るため、CD16A−Fc融合タンパク質(実施例1)および市販の組換CD16B−Fc(対立遺伝子NA−2;BSAフリー;RD Systems)を用いて、ELISAを行った。gp34−Fc融合タンパク質をネガティブコントロールとして用いた。この組換タンパク質を、pH8.8で、100mM NaHCO3中、80 nMの濃度で一晩、コートした。
更なる性質決定のため、組換CD16A−FcおよびNKp46−Fcにおけるウェスタン・ブロットでの結合について、全ての抗−CD16A scFvを試験した。CD16A−FcおよびネガティブコントロールとしてのNKp46−Fcを、還元条件下の12% SDS−ゲルおよび非還元条件下の8%ゲルによって分離した。これらのタンパク質を、エレクトロブロッティングによって、ニトロセルロース膜に移した。ポンソーS染色後、この膜を5mm幅片に切断した。各ニトロセルロース片をPBS、0.1% tween、2%スキムミルク中のscFv(4μg/ml)とともに個々にインキュベーションした。検出は、抗−ペンタHis HRP−複合体(1μg/mL)を用い、ジアミノベンジジン(DAB)ペルオキシダーゼを基質として用いて行った。コントロール片をA9(1μg/mL)または抗−NKp46 195314、ついでヤギ抗−マウスIgG HRP−複合体(0.2μg/mL,最小交叉反応性)と共にインキュベーションした。更なるポジティブコントロールとして、ヤギ抗−ヒトIgG HRP−複合体(0.16μg/mL)を、融合タンパク質のFc部の検出に用いた。モノクローナル抗体A9および195314は、非還元条件下と同様に還元条件下で、それらの抗原を認識した。
scFv変種のより高い親和性が主に改良された一価結合によるかどうかを決定するため、タンパク質調製物を、そのオリゴマー組成物について、スーパーデックス200ゲルろ過カラムにおいて分析した。LMWゲルろ過キャリブレーションキット(Amersham・Pharmacia Biosciences社、Freiburg、ドイツ)をスタンダードに用いた。表3は、個々のタンパク質調製物に存在する分子形式の概要を示す。
scFvの親和性定数(KD値)を、BIAcore2000バイオセンサーシステム(Amersham−Pharmacia社)を用いたSPRによって測定した。CD16A−Fc融合タンパク質およびネガティブコントロールとして用いたBSAをアミンカップリング法のための慣用のNHS/EDC化学を用いてカルボキシメチルデキストランでコートしたセンサーチップCM5(BIAcore社)上に固定化した。精製scFvをHBS−EP緩衝液(BIAcore社、Uppsala、スウェーデン)で希釈した。固定化抗原に対して、scFv抗体の変動濃度(100nM、200nM、400nM、800nMおよび1600nM)の経過により、結合データを生じさせた。ポジティブコントロールとして抗−CD16 MAb A9を用いた。全てのSPR測定は、20μl/minの一定の流速で、HBS−EP緩衝液中、25℃で行った。各注入試料(100μl)は、抗原に5分間接触した。ついで、解離は30分間であった。各サイクルの後、センサーチップ表面をHBS−EP緩衝液でフラッシングした。BSAでコートされたコントロールチャネルからのシグナルを自動的に差し引いた。速度定数を、1/1 ラングミュア結合モデルにしたがってBIAevaluation バージョン3.0ソフトウェア(BIAcore社)を用い、計算した。計算されたオフーおよびオン−速度定数、および各KD値を表3にまとめた。MAb A9について、コントロールとして用いて、1.75×10−9MのKDを、文献記載の1.55×10−9Mの値(Rennerら,2001 Cancer Immunol.Immunother.50:102−108)と関連付けて計算した。更に、scFv 50NI、4−LS21および3−LB6のSPRデータについての定常状態分析から,平衡解離定数を推論した。

親抗−CD16 scFv 50NIについて示されている親和性成熟scFvが末梢血NK細胞におけるCa2+動員を誘発する能力を保持しているかどうかを評価するために、末梢血からNK細胞を単離および濃縮し、および実施例5に前記記載のフローサイトメトリーによるCa2+流量測定に用いた。
親和性成熟抗−CD16 scFv(3−LS49または4−LS21、参照例として用いられる)が適用された場合、抗−(His)6ついでGAMによる架橋後に誘導されたシグナルの増加は50NIによって引き起こされたシグナルと少なくとも同じ強さであった。したがって、鎖−シャッフリングによってヒトscFv 50NIから導かれた全てのscFvがNK細胞を活性化し、およびCD16Aへの結合による細胞内カルシウムカルシウム動員のきっかけとなる能力を保持することが結論付けられる。
CD16AはNK細胞に媒介される抗体依存細胞傷害性(ADCC)に関与する。インビトロでは、CD16に特異的なIgGアイソタイプのモノクローナル抗体の活性化が、FcR−ポジティブ標的セルラインの殺滅を引き起こせることが示された。NK細胞においてカルシウム動員を誘導する親和性成熟抗−CD16A scFvがNK細胞傷害性も媒介できるかどうかを確認するため、これらのscFvを、マウスFcR−ポジティブP815細胞に対して新たに単離したNK細胞を用いたリダイレクトされた殺細胞アッセイにおいて試験した。scFvは、IgGのFc部を欠くので、抗−(His)6 マウスモノクローナル抗体と一緒に用いた。このリダイレクトされた細胞溶解を、カルセイン−放出アッセイで測定した。
ヒトFcγRIIIは、2つのアイソフォーム、FcγRIIIA(膜貫通タンパク質)およびFcγRIIIB(GPI−アンカー型)、として存在し、これらは、その細胞外の免疫グロブリン結合領域において96%の配列同一性を有する(van de WinkelおよびCapel,1993,Immunol.Today 14(5):215−221)。これらの2つのアイソフォームの異なる対立遺伝子変種は開示されている(Koeneら,1997 Blood 90:1109−1114;Koeneら,1998 Blood 91:673−679)。図6は、AアイソフォームおよびBアイソフォームの対立遺伝子変種のアミノ酸配列アラインメントを示す。
CD16Aアイソフォームに対するscFv 4−LS21の特異性を示すため、異なる対立遺伝子変種のCD16AおよびCD16Bにおいてウエスタンブロット分析を実施した。それぞれ750ngのCD16A−Fc48R/158V、CD16A−Fc48R/158F、CD16B−FcSH、CD16B−FcNA1、CD16B−FcNA2およびNKp46−Fc(ネガティブコントロール)を非還元条件下で8%SDSゲル上にロードし、および膜上へブロッティングした。
更にCD16Aアイソフォームに対するscFv 4−LS21の特異性を示すため、異なる対立遺伝子変種を用いてELISAを実施した。Maxisorp(商品名)プレートのウエルをそれぞれ200ngのCD16A−Fc48R/158V、CD16A−Fc48R/158F、CD16B−FcSH、CD16B−FcNA1、CD16B−FcNA2またはNKp46−Fc(ネガティブコントロール)を含有する100mM NaHCO3でコーティングした。
実施例12に記載のプロトコールに従い、BIAcore2000バイオセンサーシステム(Amersham−Pharmacia社)を用いて、対立遺伝子変種CD16A−Fc48R/158V、CD16A−Fc48R/158F、CD16B−FcSH、CD16B−FcNA1およびCD16B−FcNA2についてのSPRにより、scFv 4−LS21およびMAb A9の親和性定数(KD値)を測定した。計算したオフ−およびオン−速度定数および各KD値を、表5にまとめた。

抗−CD30および抗−CD16A抗体のVHおよびVL可変領域をコードする遺伝子を、ハイブリドーマHRS−3またはヒトscFvライブラリーから,それぞれ、プラスミドpHOG_scFv(G2S)3 αCD30(HRS−3)およびpSKK2_scFv αCD16A(LS21)を用いて取り出した。
抗CD19抗体のVHおよびVL可変領域をコードする遺伝子を、プラスミド pSKK3_scFv(G2S)3α CD19を使ったハイブリドーマ HD37(HD37)から得た。プラスミドpSKK3_scFv(G2S)3α CD19(HD37)を、NcoI/BsmBIによって切断し、挿入断片をアガロースゲルにおいて精製した。ついで、精製した断片を、制限酵素 EcoRVで消化した。EcoRV/BsmBI消化断片を直線化EcoRV/BsmBIプラスミド pHOG CD30xCD16 LS21 TandAb/(G2S)3にクローニングし、プラスミド pHOG CD19xCD16 LS21 TandAb/(G2S)3を得た。プラスミド pHOG CD19xCD16 LS21 TandAb/(G2S)3 をNcoI/XbaIで切断し、NcoI/XbaI 線形化プラスミド pSKK3にライゲーションした。得られたプラスミドは、抗CD19×抗CD16A TandAbをコードするpSKK3 CD19xCD16ALS21 TandAb/(G2S)3である。
発現プラスミド pSKK3 CD30xCD16 LS21 TandAb/(G2S)3およびpSKK3 CD19xCD16 LS21 TandAb/(G2S)3で形質転換したE.coli K12株RV308(Maurerら,1980、J.Mol.Biol.139:147−161)のサンプルを、50μg/mlアンピシリンおよび100mM グルコース(2×YTGA)を含有する2×YT培地中、28℃で、一晩生育させた。2xYTGA中の一晩培養物の希釈物(1:50)を、28℃、200rpm振とうでフラスコ培養として生育させた。培養物が、OD6000.8に達した時点で、細菌を9500g、15分間、20℃の遠心で沈降させ、同容量の新しい、50μg/mlアンピシリンを添加したYTBS培地(1M ソルビトールおよび2.5mM グリシンベタインを含有する2×YT;Blacwell & Horgan、1991、FEBS Letters 295:10−12)に再懸濁した。IPTGを最終濃度0.2mMで添加して、培養を21℃、18−20時間継続した。細胞を、14000g、20分間、4℃の遠心分離で回収した。可溶性周辺質タンパク質を単離するため、沈降した細菌を5%の初期容量の氷冷200mM Tris−HClTris−HCl、20% スクロース、1mM EDTA、pH8.0に再懸濁した。随時攪拌しながら氷上で1時間インキュベーションした後、スフェロプラストを14000g、60分間、4℃で遠心分離し、可溶性ペリプラズム抽出物を上清中に残し、このスフェロプラストにペレット中の不溶性ペリプラズム物質を加えた。ペリプラズム画分を開始緩衝液(50mM Tris−HClTris−HCl、1M NaCl、50mM イミダゾール pH7.0)に対して、4℃で透析した。組換え産物を含有する透析溶液を、14000g、30分間、4℃で遠心分離した。Cu2+で荷電し、50mM Tris−HCl、1M NaCl、pH7.0(開始緩衝液)で平衡化した、Chelating Sepharose Fast Flow(GE Healthcare Biosciences社、Freiburg、ドイツ)の1mlカラムを用いて、固定化した金属親和性クロマトグラフィー(IMAC)を4℃で、実施した。試料を、カラムを通過させることによって、ロードした。ついで、これを20カラム容量の開始緩衝液、ついで溶出液の吸収(280nm)が最小(約30カラム容量)になるまで50mMイミダゾールを含有する開始緩衝液で洗浄した。吸収された物質を、50mM Tris−HCl、1M NaCl、300mM イミダゾール、pH7.0で溶出させた。このTandAb分子を含有する溶出画分を、ペンタHis(商品名)抗体 BSA不含有(Qiagen、Hilden、ドイツ)、ついで西洋ワサビペルオキシダーゼで標識したヤギ抗−マウスIgG(Dianova社、Hamburg、ドイツ)を用いて、ウエスタンブロット分析で同定した。
CD16Aへの結合を測定するため、実施例7に記載したように、HEK−293 細胞をpcDNA3−CD16A−ゼータ プラスミド(Dr.O.Mandelboim、Jerusalem大学、イスラエル)を用いてリン酸カルシウム法でトランジェントに形質転換し、フローサイトメトリーのため形質転換の40時間後に回収した。有毛状細胞性白血病セルラインJOK−1(Schwartz−Albiez R,Dorken B,Monner DA、Moldenhauer G、(1991) Int Immunol 3:623、Dr.G.Moldenhauer、DKFZ、Heidelbergの親切なる贈り物)を用いて、CD19への結合を測定した。CD30での反応性を試験するため、染色をヒト・ホジキン・セルラインL540CY(V.Diehl博士、Cologne大学、ドイツから親切に提供を受けた;Kapp U、Wolf J、von Kalle C、Tawadros S、Rottgen A、Engert A、Fonatsch C、Stein H、Diehl V.(1992) Ann Oncol. Sep;3 Suppl 4:21−3)を用いて行った。細胞の染色および分析を、基本的に上記のように行った。全ての組換え抗体を10μg/mL MAb 抗−ヘキサ His 13/45/31−2 (Dianova、Hamburg、ドイツ)、ついで15μg/mL FITC−複合化ヤギ 抗−マウスIgG(Dianova)で検出した。
フローサイトメトリー分析は、CD30xCD16LS21 TandAbおよびCD19xCD16LS21 TandAb抗体の両方の、対応抗原を発現する細胞への強い結合を示した。TandAb抗体の両方は、CD16A−発現細胞に、ほぼ等しい親和性で結合した。
対立遺伝子変種、48R/158Vおよび48R/158F、におけるscFv 4−LS21のKD値の測定により、表面プラスモン共鳴で測定されたように、同等の約5×10−8MのKD値が示され、両方の対立遺伝子変種は、ほぼ同じ親和性(実施例12を参照)で認識されることを示した。この知見は、TandAb CD30xCD16LS21由来の4−LS21で確認され、これもまたCD16Aの両方の対立遺伝子変種にほとんど同一の親和性での結合を示した。4−LS21 scFvと比較して、結合性の増加による、ファクター10以上の親和性における改良が、TandAb抗体で観察された。
NK細胞を標的細胞に集めること、および受容体の架橋によるNK細胞の活性化によって、CD30xCD16LS21およびCD19xCD16LS21 TandAb抗体が、インビトロの細胞傷害性を活性化するかどうかを確認するため、非−放射活性細胞傷害性アッセイを、基本的にT.Dreierら(2002、Int J Cancer 100:690−697)に従って、実施した。前記のようにエフェクター細胞として用いたNK細胞を、PBMCから濃縮した。それぞれのケースで濃縮したNK細胞の純度および抗原発現をフローサイトメトリーでチェックした(データは示していない)。
この結果は、TandAb−活性化NK細胞による標的細胞の特異的溶解を示す。対照的に、CD19xCD16LS21 TandAbは、CD19−L540CY標的細胞の溶解を誘導せず、逆もまた同様であり、CD30xCD16LS21TandAbは、NK細胞のCD30− JOK−1細胞の殺滅を誘導しなった。NK細胞の不存在下では、CD30xCD16LS21 TandAbもCD19xCD16LS21 TandAbも標的細胞細胞傷害性活性を示さなかった。
Claims (26)
- 少なくとも1つの抗原に対する特異性を有する抗体又は抗原結合断片であって、
当該少なくとも1つの抗原がFcγRIIIAであり、かつ特異的にFcγRIIIBに結合しない抗体又は抗原結合断片であり、
(a)アミノ酸配列重鎖CDR1は配列番号17であり、アミノ酸配列重鎖CDR2は配列番号18であり、アミノ酸配列重鎖CDR3は配列番号19であり、
アミノ酸配列軽鎖CDR1は配列番号20、23、26、29、33、34からなる群から選択され、
アミノ酸配列軽鎖CDR2は配列番号21、24、27、30、31、35からなる群から選択され、
アミノ酸配列軽鎖CDR3は配列番号22、25、28、32からなる群から選択される、6つのCDRを含み、
(b)配列番号9のアミノ酸配列を有するヒト重鎖可変部を含み、
(c)配列番号10〜16からなる群から選択されるアミノ酸配列を有するヒト軽鎖可変部を含む、
抗体又は抗原結合断片。 - FcγRIIIA<158F>対立遺伝子変種及びFcγRIIIA<158V>対立遺伝子変種に結合することができ、FcγRIIIAの2つの158アレルに対する親和性の違いは2倍以下であり、特異的にFcγRIIIBに結合しない、
請求項1記載の抗体又は抗原結合断片。 - 配列番号9のアミノ酸配列可変重鎖及び配列番号16のアミノ酸配列可変軽鎖を含む、請求項2記載の抗体又は抗原結合断片。
- 抗原結合断片が、Fv、Fab、di−Fab、Fab’、F(ab’)2、scFvからなる群から選択される、請求項1又は3のいずれか1項に記載の抗体又は抗原結合断片。
- 抗体または抗原結合断片が完全ヒト型である、請求項1〜4のいずれか1項に記載の抗体又は抗原結合断片。
- 少なくとも1つの更なる抗原に対する特異性を有する、請求項1〜5のいずれか1項に記載の抗体又は抗原結合断片。
- 少なくとも1つの更なる抗原が細胞表面抗原である、請求項6記載の抗体又は抗原結合断片。
- 細胞表面抗原がCD19、CD20、CD30、ラミニン受容体前駆体、EGFR1、EGFR2、EGFR3、Ep−CAM、PLAP、トムセン−フリーデンライヒ抗原、MUC−1、IL4−Rアルファー、IL13−R、IGFR、FcεRIおよびCD5から選択される、請求項7記載の抗体又は抗原結合断片。
- 少なくとも1つの更なる抗原が病原体由来である請求項6または請求項7記載の抗体又は抗原結合断片。
- 病原体がウイルス、細菌、真菌、マイコプラズマ、寄生体またはプリオンである請求項9記載の抗体又は抗原結合断片。
- 抗原結合断片がダイアボディまたはTandAbである請求項6〜10のいずれか1項に記載の抗体又は抗原結合断片。
- 前記抗原結合断片が更に機能ドメインを含有する、請求項1〜11のいずれか1項に記載の抗体又は抗原結合断片。
- 機能ドメインが、抗体Fcドメイン、抗体依存酵素プロドラッグ治療における使用のための酵素、Fc受容体に結合可能なペプチド、前記抗体又は抗原結合断片の血中半減期を増加させるためのタンパク質またはペプチドから選択される、請求項12記載の抗体又は抗原結合断片。
- 抗体又は抗原結合断片が標識分子または毒素に結合している、請求項1〜13のいずれか1項に記載の抗体又は抗原結合断片。
- 請求項1から14のいずれか1項に記載の抗体又は抗原結合断片と、薬学的担体、賦形剤、希釈剤および/または安定剤と、を含有する薬学的組成物。
- 請求項1〜14のいずれか1項に記載の抗体又は抗原結合断片をコードするポリヌクレオチド。
- 配列番号2〜8からなる群から選択される核酸配列によってコードされる第1の軽鎖可変領域、配列番号1の核酸配列によってコードされる第1の重鎖可変領域、第2の軽鎖可変領域、および第2の重鎖可変領域をコードするポリヌクレオチドであって、当該第2の軽鎖および重鎖の可変領域がCD19に対する親和性を有する、請求項16記載のポリヌクレオチド。
- 配列番号2〜8からなる群から選択される核酸配列によってコードされる第1の軽鎖可変領域、配列番号1の核酸配列によってコードされる第1の重鎖可変領域、第2の軽鎖可変領域、および第2の重鎖可変領域をコードするポリヌクレオチドであって、当該第2の軽鎖および重鎖の可変領域がCD30に対する親和性を有する、請求項16記載のポリヌクレオチド。
- 作動可能にプロモーター領域および転写終結領域に連結された請求項16〜18のいずれか1項に記載のポリヌクレオチドを含有するベクター。
- 請求項16〜18のいずれか1項に記載のポリヌクレオチド、または請求項19のベクターを含有する宿主細胞。
- 請求項16〜18のいずれか1項に記載のポリヌクレオチド、または請求項19のベクターを宿主細胞へ導入することと、抗体又は抗原結合断片が発現される条件下で宿主細胞を培養することとを含有する、請求項1〜14のいずれか1項に記載の抗体又は抗原結合断片の製造方法。
- 更に抗体又は抗原結合断片の単離を含有する請求項21記載の方法。
- 自己免疫疾患、炎症疾患、感染症、アレルギーまたはがんの診断または治療における使用のための、請求項1〜14のいずれか1項に記載の抗体又は抗原結合断片、または請求項15記載の組成物。
- がんが、非ホジキンリンパ腫、慢性リンパ性白血病、悪性リンパ肉芽腫症、固形腫瘍、微小残存病変、および転移性腫瘍からなる群から選択される、請求項23記載の抗体又は抗原結合断片、あるいは組成物。
- 生体外での患者細胞プロファイルの分析ための試薬としての請求項14記載の抗体又は抗原結合断片の使用。
- 請求項1〜14のいずれか1項に記載の抗体又は抗原結合断片およびFcγRIIIAへの結合時の抗体又は抗原結合断片を検出する手段を含有するキット。
