本発明には、表現型的にCD45RA+の血液細胞の集団から調節性T細胞を単離する方法が包含され、ここで、該Treg細胞はT細胞増殖を抑制する。本発明の方法は、ヒト臍帯血サンプルから単核細胞の集団を単離すること;単核細胞−抗体複合体の形成に適する条件下でCD25に特異的に結合する抗体と単核細胞の集団を接触させること;および単核細胞の該集団から単核細胞−抗体複合体を実質的に分離し、それにより表現型的にCD45RA+の血液細胞の集団から調節性T細胞を単離することを含んでなる。本発明の1つの態様において、表現型的にCD45RA+の血液細胞の集団は臍帯血、好ましくはヒト臍帯血サンプルからである。
本発明には、CD45RA+表現型を有するT調節性細胞(Treg)の増大された集団の単離および増殖のための方法およびキットが包含される。「増大された」という用語は、本明細書において用いる場合、増大されない対象より少なくとも20%多い、より好ましくは30%、好ましくは40%、さらにより好ましくは50%、さらにより好ましくは約60%、さらにより好ましくは70%、さらにより好ましくは約80%、さらにより好ましくは90%、そしてさらにより好ましくは100%多いことをさす。例として、調節性細胞の増大された集団は、増大されない集団より少なくとも20%多い調節性細胞を有する。そのような細胞の好ましい供給源は、ヒト臍帯血であることが見出された。成人ヒト血液のような他の供給源から多数の活性のある強力なサプレッサーTreg細胞を製造する方法は、低〜中程度レベルでCD25を発現するメモリーT細胞(CD25dim細胞)、およびTreg細胞精製を妨げる通常の活性化T細胞(CD25+)の存在により複雑にされていた。「Treg」という用語は、CD4(CD4+)およびCD25(CD25+)の両方を発現する調節性T細胞をさすために本明細書において使用する。しかしながら、本明細書に開示されるデータにより示されるように、T細胞のこれらのタイプは両方とも(メモリーおよび活性化)、一般に臍帯血において欠如しているかもしくはより低い数で存在する。これは、本明細書に開示されるように、臍帯血は保護された環境において発生し、そして免疫学的にナイーブであるからである。従って、本発明は、臍帯血におけるCD25+Treg細胞がヒト成人末梢血のような他の供給源由来のものより容易に精製されることを示す。

また、所望のペプチドをコードするDNAをクローン化し、そして大量のペプチドの生成に適当な細胞において適切なプロモーター配列から発現させることができる。ペプチドに対するモノクローナル抗体は、本明細書に参考文献を載せるような標準的な方法を用いてペプチドで免疫感作したマウスから生成される。
分離装置に全サンプルを通すことにより抗原について陽性のTregが選択される。装置からの自由溶液の排出もしくは溶出の際に、磁気的に保持されるマーカーを含有する細胞のみが残る。次に、抗原を含有するTreg細胞を装置から溶出し、Treg細胞の濃縮された、単離されたもしくは精製された集団がもたらされる。本発明の1つの態様において、TregマーカーはCD25である。

培養用のCD25 + およびCD25 − 細胞のMACS精製
健常なボランティアドナー(Memorial Blood Centers,Minneapolis,MN)の全血から得られたバフィコート調製物から末梢血単核細胞(PBMC)を単離した。白血球に富んだバフィコート細胞をフィコール−ハイパック層上で遠心分離してPBMCを集めた。以下の間接抗体に基づくマイクロビーズを用いてCD25+細胞を単離した。PBMCを抗CD25−FITC、クローン2A3(Becton Dickinson Immunocytometry Systems,San Jose,CA)で染色し、洗浄し、そして次に抗FITCマルチソートマイクロビーズ(5マイクロリットル/107細胞、Miltenyi Biotec,Auburn,CA)に二次的に結合させ、そして陽性選択した。CD25+細胞を第二のカラムに再びかけ、洗浄し、そして再溶出した。カラム精製後に、抗FITCマルチソートビーズを脱離させた。CD25+細胞を系統枯渇(lineage depletion)用のmAb被覆マイクロビーズの混合物でCD8、CD14、CD19、CD20およびCD56を発現する細胞についてさらに枯渇させた。次に、これらのCD25+系統枯渇細胞(DC−8x−マイナス)を抗CD45RAマイクロビーズ(20マイクロリットル/107細胞、Miltenyi)での直接陽性選択によりCD45RAについて選択した。ある場合には、抗HLA−DR+細胞のさらなる精製を抗HLA−DRマイクロビーズ(20マイクロリットル/107細胞、Miltenyi)での陽性選択によりCD45RA−細胞から単離した。CD25−細胞を直接抗CD25マイクロビーズ(20マイクロリットル/107細胞、Miltenyi)での2回目の枯渇によりCD25についてさらに枯渇させた。CD25枯渇後にこれらの細胞を次に直接抗CD4マイクロビーズ(20マイクロリットル/107細胞、Miltenyi)でCD4について陽性選択した。
単離されたCD25+細胞、CD25+サブセットもしくはCD4+CD25−コントロール細胞を24ウェルプレートにおいて百万個の総細胞/mlで培養した。等しい数の
放射線照射したCD4+CD25−支持細胞を培養物に加えた。抗CD3/CD28mAb被覆ダイナビーズ(University of Pennsylvania,Philadelphia,PA)を増殖刺激として3:1のビーズ:細胞比で加え、そして中程度の用量のIL−2を新鮮培地(Chiron,Emeryville,CA)において50IU/mlで3日目に加えた(Godfrey,et al.,2004,Blood,104:453−461;Levine,et al.,1998,J.Hematother.,7:437−48)。臍帯血培養物では、培養もしくはサプレッサー機能の維持に支持細胞が必要とされず、従って、使用しなかった。細胞培養物は、急増殖期中に3日ごとに約1/3、必要に応じて分割した。培養培地は、10%のFCS(Invitrogen−Gibco)、ならびにL−グルタミン、ペニシリンおよびストレプトマイシンを補足したRPMI−1640(Invitrogen−Gibco,Carlsbad,CA)であった。
7百万個のCD25+系統陰性細胞、5百万個のCD45RA−マイナス細胞および百万個のCD45RA+細胞が得られた。2人のドナーからの精製を示す(図1)。これらの様々なCD25+集団を抗CD3/28ビーズでの1回の刺激で培養した。3週後に、アロMLRを抑制する能力についてこれらの細胞系を試験した。
CD25+細胞サブセットのさらなる分析により、CD45RA+細胞は古典的に記載されているヒトTreg細胞(Baecher−Allan,et al.,2001,J.Immunol.,167:1245−1253;Godfrey,et al.,2004,Blood 104:453−461;Hoffmann,et al.,2004,Blood 104:895−903)、CD25+brightと異なることが示された。前の報告は、これらの細胞をCD25+hiでありそしてCTLA−4陽性であると記載している(Jonuleit,et al.,2001,J.Exp.Med.,193:1285−1294)。本明細書に開示されるデータは、CD45RA細胞が異なることを示す。2色ドットプロットにおいて、CD45RA細胞は細胞内CTLA−4陰性(図4)、HLADR陰性(図4)であることが示される。HLA−DRに対するCD25の二重染色は、HLA−DR+細胞がCD25+bright細胞であることを示し(図4)、そしてそれらは細胞内CTLA−4について二重染色する(図4)。これは、2つの異なる細胞サブセットを示す。
CD4+CD25−応答T細胞を健常なボランティアドナー(Memorial Blood Centers,Minneapolis,MN)の全血由来のバフィコート調製物から単離した。細胞をフィコール−ハイパック上で遠心分離してPBMCを集めた。CD4+T細胞を抗CD4mAb被覆磁気マイクロビーズ(Miltenyi−Biot
ec)で陽性選択により単離する前に、PBMCを抗CD25mAb被覆マイクロビーズ(Miltenyi−Biotec)でCD25+細胞について最初に枯渇させた。細胞は、FACS分析により通常96〜98%純粋であった。磁気ビーズに基づく精製(Miltenyi−Biotec)により、PBMCから単離された、CD14+単球から未熟樹状細胞(DC)を生成せしめ(Sallusto and Lanzavecchia,1994,J.Exp.Med.,179:1109−1118)、そしてX−vivo−15(BioWhittaker,Walkersville,MD)培地においてGM−CSF(最終50ng/ml)およびIL−4(最終20ng/ml;R&D Systems,Minneapolis,MN)を補足して百万個の細胞/mlで培養した。MLRにおいてスティムレーター(stimulator)として使用する前に細胞を5〜10日間培養した。ある実験では、DCをLPS(Sigma,St.Louis,MO)(100ng/ml)、もしくはTNF−アルファ(最終20ng/ml)およびポリI:C、トル様受容体(TLR)−3アゴニストリガンド(最終20μg/ml)(Sigma)で2日間成熟させた(Godfrey,et al.,2004,Blood 103:1158−1165;Cella,et al.,1999,J.Exp.Med.,189:821−829)。DC刺激細胞に30Gyで放射線照射した。
5x104の応答CD4+CD25−T細胞および5x103のDC刺激抗原提示細胞(APC)を96ウェルU底プレートにおいてウェル当たり培養した。培養したサプレッサーもしくは通常のT細胞系を標準アッセイではウェル当たり2.5x104で、もしくは滴定実験では段階的な数において加えた。抗体ブロッキング実験では、104もしくは5x103のサプレッサー細胞を用いた。培地は、10%のFCSを補足したRPMI−1640であった。ウェルを3、5、6および7日目に培養の最後16時間にわたって3H−チミジンでパルス標識した。各時間点は、6つの反復実験を有した。結果は分当たりのカウントにおいて表した。データを直接ベータカウンター(液体シンチレーションなし)で集め、従って、結果の大きさはより低いが比例的に正確であった。
MLR培養上清を細胞なしに回転させ、そしてアリコートを−80℃で凍結させた。再刺激には、抗CD3/CD28ビーズを1:1のビーズ:細胞比で用いた。上清をラテックスビーズに基づく多検体システム(R&D Systems,Minneapolis,MN)でルミネックスアッセイシステムにより評価した。
抑制効能をさらに評価するために滴定実験を実施した。標準的なDC刺激MLR培養物に加えるサプレッサーの数を下げることにより、臍帯血由来のサプレッサーのほぼ完全な抑制活性は1:16もしくは1:32(50,000の応答細胞に対してわずか1500のサプレッサー)の比率まで(out to)維持されることが示された。これは、選択された最も強力な成人由来の細胞系(CD25+lineage−)よりわずかに強力である(〜2倍)。
MLRアッセイにおける応答T細胞へのサプレッサー細胞系の効果を決定するために、培養上清を複数のサイトカインの存在および量について評価した。MLRアッセイへのサプレッサー細胞添加は、IL−2、IFN−ガンマ、GM−CSF、TNF−αおよびIL−10を包含する、T細胞活性化依存性サイトカインの蓄積を顕著に減少した。活性化依存性サイトカイン蓄積は、抑制されたMLRの間の全ての時間点で最小であった。これらのデータは、T細胞活性化の顕著な機能障害を示す。
通常のT細胞系に対する抑制細胞系の機能的能力を決定するために、再刺激後のサイトカイン生産および細胞表面分子発現のそれらの能力を評価した。細胞系を強力な再活性化のために抗CD3/CD28ビーズで再刺激し、そしてルミネックスビーズに基づくアッセイによるサイトカイン含有量の分析用に特定の時間点で上清を採取した。CD45RA+由来の細胞系はIL−2、IFN−γもしくはIL−10を本質的に生産せず、一方、コントロールのCD25−由来の細胞系は高レベルのこれらのサイトカインを生産した。TNF、GM−CSFおよびIL−5、ならびにケモカインIL−8の蓄積もまた、コントロール細胞系と比較した場合に著しく減少された。
臍帯血免疫学および移植におけるTreg細胞の重要な役割は、一般に理解されていない(Barker,et al.,2003,Crit.Rev.Oncol.Hematol.,48:35−43)。多数の研究者は、免疫学的特性化の目的のために真にナイーブのT細胞の代表的なモデルとして全CD4+選択臍帯血集団を使用する(Kaminski,et al.,2003,Blood 102:4608−4617)。しかしながら、本明細書に開示されるデータに照らしてみると、いくつかの前の研究はCD25枯渇CD4+細胞で再評価される必要があり得る(Jonuleit,et al.,2000,J.Exp.Med.,192:1213−1222)。これらの細胞は、臍帯血移植(CBT)において経験されるGVHDの低い割合への寄与因子であり得る(Wadlow,et al.,2002,Biol.Blood.Marrow.Transplant.,8:637−647)。
CD25+およびCD25−CD4+T細胞を臍帯血(Red Cross,Saint Paul,MN)から単離した。臍帯血単核細胞を製造業者の使用法に従ってフィコール−ハイパック上で遠心分離により調製した。磁気マイクロビーズ(Miltenyi
Biotec,Auburn,CA)でのCD34+枯渇後に、直接結合した抗CD25磁気マイクロビーズ(107の細胞当たり4マイクロリットル;Miltenyi Biotec)での陽性選択によりCD25+細胞を単離した。次に、細胞を第二の磁気カラムにかけ、洗浄し、そして再溶出した。二重カラム方法の後に、細胞はFACS分析により通常>90%純粋であった(CD4/CD25について)。次に、抗CD4mAb被覆マイクロビーズ(Miltenyi Biotec)での陽性選択によるCD4+CD25−細胞の単離の前に、あらゆる残留するCD25+細胞を枯渇させるために非CD25画分を別の磁気カラムにかけた。成人CD25+細胞のストリンジェントな精製は、抗CD25−FITCおよび抗FITCマイクロビーズ(107の細胞当たり2マイクロリットル)、ならびに2サイクルにわたる磁気カラム上の通過および溶出を使用した。この後に磁気ビーズを遊離させ、そして続いてGodfrey,et al.(2004,Blood 104:453−461)に記載のとおり抗CD8、CD14、CD19およびCD56直接結合マイクロビーズで系統枯渇させた(CD4+CD25++lin−)

単離されたCD4+CD25+細胞もしくはコントロールCD4+CD25−細胞を3:1のビーズ:細胞比で抗CD3/CD28mAb被覆ダイナビーズ(University of Pennsylvaniaから入手し、そしてLevine,et al.,1998,Hematother.,7:437−448に記載される)とGodfrey,et al.(2004,Blood 104:453−461)に記載のとおり培養した。細胞は、24ウェルプレートにおいて百万個の総細胞/mlで培養した。IL−2を50IU/ml(Chiron,Emeryville,CA)で3日目に加えた。前の研究とは対照的に、全ての系を支持細胞なしに培養した。臍帯血培養物では、放射線照射した支持細胞(CD4+CD25−細胞)(Godfrey,et al.,2004,Blood 104:453−461)の添加は増殖にもサプレッサー機能の維持にも役立たず、従って、培養プロトコルを簡素化するために支持細胞を省いた。細胞培養物は、急増殖期の間3日ごとに約1:3、必要に応じて分割した。培養培地は、10%のウシ胎仔血清(FCS;Invitrogen−Gibco)、ならびにL−グルタミン、ペニシリンおよびストレプトマイシンを補足したRPMI−1640(Invitrogen−Gibco,Carlsbad,CA)であった。
臍帯血単核細胞(CBMC)は、CD25を共発現するCD4+T細胞を含有することが示されている。従って、本発明の結果は、サプレッサー機能およびサプレッサー細胞系生成のこれらの細胞の能力の評価を開示する。FACS分析を用いてCBMCはCD25+を共発現するCD4+T細胞の顕著な集団を含有すること、およびこれは細胞の別個の集団であることが確かめられた(図6A)。対照的に、成人血液は、非抑制性CD25−dim細胞の大きな集団を包含する、広範囲のレベルのCD25を有するCD4+T細胞を含有する(図6B)。臍帯血CD4+T細胞の約5%は、CD25+を明確に発現する(平均5.2%、範囲2.3〜9.5%、n=20)(成人末梢血CD4+T細胞に存在する(CD4+の平均3.8%)よりわずかに高いパーセンテージ)。精製の直接比較のために、CD25+Treg細胞を同一のMACSに基づくプロトコル(直接抗CD25に基づく選択)を用いて成人および臍帯血の両方から単離した。臍帯血から精製されたCD25+細胞は、CD25+bright細胞(MFI320vs130)のより収束した集団、およびより少ないCD25−dimもしくはCD25−陰性細胞を含有した(図6C)、それぞれ平均9%(範囲5〜21%、n=10)および3%(範囲1〜9%、n=10)。比較して、成人血液由来のCD25+細胞はより多くのCD25−dimおよびCD25−陰性細胞を含有することが見出された(図6D)、それぞれ平均30%(範囲25〜38%、n=10)および10%(範囲2〜24%、n=10)。
臍帯血CD25+細胞を直接選択により精製した。それらをCD25およびCD45RAについて染色した。細胞は主にCD45RA+である(図7)。
IL−2を補足して、抗CD3/CD28被覆ビーズでストリンジェントに精製された成人由来のCD25+Treg細胞を刺激することは、著しい増殖を誘導する。精製された成人もしくは臍帯血CD4+CD25+細胞、ならびに比較のための臍帯血CD4+CD25−細胞から細胞系を生成するためにこの培養戦略を用いた。臍帯血由来のCD4+CD25+細胞は培養において容易に増殖した;単一の初期刺激で3週にわたって約100倍。3〜4週後に細胞系は数を増殖するのを停止し、そしてIL−2において維持され
た。従って、これらの細胞系の増殖曲線は、成人血液由来のCD4+CD25+細胞系のものと同様であった。

細胞系のサプレッサー機能を評価するために、HLA不適合アロ−MLRアッセイを機能読み取りとして用いた。CD4+応答およびDC刺激MLRアッセイは非常に強力であり、そしてドナー間で一貫性があり、従って、抑制の標準的測定として役割を果たした。2〜3週の培養後に全ての細胞系をMLRにおいてサプレッサー活性について最初にスクリーニングし、そしてそれから次の3〜4週にわたってさらに分析した。驚くべきことに、臍帯血CD25+細胞由来の細胞系は、一貫してそして強力に抑制性であった(図8A)。強力な抑制性細胞系は30のドナーのうち29から単離され、ここで、増殖の抑制は典型的に>95%であった(図8B)。成人もしくは臍帯血CD25−細胞由来のコントロール細胞系は、抑制性ではなかった(図8B)。成人CD25+細胞(直接単離された)由来の細胞系は、弱いそして変わりやすい抑制活性を示した(図8Aおよび8B)。成
人ドナーのサブセットにおいて顕著な、強力なサプレッサー細胞系を生成するためにストリンジェントなMACSに基づく選択が必要とされることが以前に報告された。これらの系(CD25++lineage−)により媒介される抑制は図8Aおよび8Bに示され、それは臍帯血由来の細胞系の驚くべき一貫性を示す。
MLRアッセイにおける応答T細胞へのサプレッサー細胞系の効果を決定するために、上清を複数のサイトカインの存在および量について評価した。MLRアッセイへのサプレッサー細胞添加は、IL−2、IFN−ガンマ、GM−CSF、TNF−αおよびIL−10を包含する、T細胞活性化依存性サイトカインの蓄積を顕著に減少した。重要なことには、TGF−ベータ−1蓄積のレベルは改変されないようであった。活性化依存性サイトカイン蓄積は、抑制されたMLRの間の全ての時間点で最小であり、コントロールDC−MLRにおけるピーク検出時での結果を示す(図9A)。これらのデータは、T細胞活性化の顕著な機能障害を示す。選択されたケモカイン発現への抑制の効果を決定するために、MLR上清をIL−8(CXCL8)、MIP−1a(CCL3)およびRANTES(CCL5)、前炎症性ケモカインの発現について評価した。早い時間点で、蓄積への効果は認められなかった(図9B)。しかしながら、MLRにおける遅い時間点で、蓄積は約50〜75%減少された。従って、ケモカイン生産は抑制によりそれほど影響されないようであり、そしてサプレッサー細胞の効果においてある程度の選択性がある。

培養したサプレッサー細胞系は既知の可溶性免疫抑制性もしくは細胞表面の負の調節タンパク質を介して働くかどうかを決定するために、DC−MLRサプレッサーアッセイを中和もしくはブロッキングモノクローナル抗体で処理した。アッセイは、増殖の回復による抑制の取り消しについて評価した。最初に、IL−10、IL10RおよびTGF−ベータ1,2,3に対する抗体を単独でもしくは最小効果を有する様々な組み合わせで試験した。高用量(30μg/ml)で3つの抗体全てを組み合わせることによってのみ、抑制効果のあまり多くないそしておそらく非特異的な取り消しが見られた。3つの抗体全てで処理した処理コントロール培養物はまた、3つの抗体全てで処理したTreg細胞と同様に、増加した増殖を明白に示した。
免疫蛍光染色のために、細胞を4℃で30分間染色した。細胞を洗浄し、そしてFACS Calibur血球計算器(Becton Dickinson)上で実行した。データは、FlowJoソフトウェアバージョン4.5(Treestar,Ashland,OR)により分析した。細胞内染色は2%パラホルムアルデヒド固定細胞を用いて行い、続いて0.1%サポニン含有バッファーにおいて透過化しそして染色した。
全てのエラーバーは、平均より上および下の1つの標準偏差を表す。
細胞系をサプレッサー細胞表現型と関連する抗原についてフローサイトメトリーにより
分析した。通常のT細胞コントロールとして役割を果たすCD4+CD25−由来の細胞系と並行してサプレッサー細胞系を培養した。細胞系は、次の3〜5週にわたって比較的安定な表現型および機能を維持した。CD45RA+由来のCD25+細胞系は、CD25、細胞内CTLA4、CD27およびCD62Lを包含する複数の抗原の著しく均一な発現を維持する。比較して、CD45RA(−)由来の細胞系は、これらの抗原について不均一であった。
TR1−試薬(Molecular Research Center,Cincinnati,OH)もしくはRNAeasy(Qiagen,Valencia,CA)を用いて全RNAを抽出した。Taqmanユニバーサルマスターミックス(Applied Biosystems,Foster City,CA)を用いて1μgの各RNAサンプルからcDNAを合成した。10ngを各qPCR反応において使用した。全てのサンプルを二重反復で行った。FoxP3およびサイクロフィリンAのプライマーおよびプローブは、Applied Biosystemsから購入した。リアルタイムPCRは、ABI Prism 7900(Advanced Biosystems)を用いて行った。FoxP3メッセージレベルは、サイクロフィリンAにデータを正規化した後に決定した。
Active Motif(Carlsbad,CA)を用いて製造業者の使用法に従って核抽出物を調製し、そしてレーン当たり70μgのタンパク質を載せた。サンプルをNuPage 10%Bis−Trisミニゲル(Invitrogen)上で泳動した。タンパク質をPVDF膜に移し、そしてヤギ抗FoxP3抗体(AB2481)(Abeam,Cambridge,MA)、続いてウサギ抗ヤギIgG西洋ワサビペルオキシダーゼとインキュベーションした。ブロットをSuperSignal WestPico化学発光基質(Pierce,Rockford,IL)で発色させた。
MLR培養上清を細胞なしに回転させ、そしてアリコートを−80℃で凍結させた。再刺激のために、抗CD3/CD28ビーズを1:1のビーズ:細胞比で用いるか、もしくは2ng/mlのPMAおよび500ng/mlのイオノマイシンを用いた。細胞を24ウェルプレートにおいて百万個の細胞/ml培地で培養した。上清をラテックスビーズに基づく多検体システム(R&D Systems,Minneapolis,MN)でルミネックスアッセイシステムにより評価した。

CD25−由来の細胞系でも認められた(Godfrey,et al.,2004,Blood 104:453−461)。
通常のT細胞系に対する抑制性細胞系の機能的能力を決定するために、これらの細胞を再刺激後のサイトカイン生産および細胞表面分子発現のそれらの能力について評価した。細胞系を強力な再活性化のために抗CD3/D28ビーズで再刺激し、そしてルミネックスビーズに基づくアッセイによるサイトカイン含有量の分析のために特定の時間点で上清を採取した。CD25+由来の細胞系はIL−2、IFN−γもしくはIL−10を本質的に生産せず(図12A)、一方、コントロールのCD25−由来の細胞系は高レベルのこれらのサイトカインを生産した。TNF、GM−CSF、IL−5およびケモカインIL−8の蓄積もまた、コントロール細胞系と比較した場合に著しく減少された(図12A)。対照的に、TGF−ベータ、ならびにケモカインMIP1aおよびRANTESの蓄積は、異なる細胞系間で著しく異ならなかった。

み、あまり多くないそしておそらく非特異的な効果が見られた(図13A)。処理したコントロール培養物もまた、増加した増殖を明らかに示した。負の共刺激シグナル伝達分子CTLA4およびPD1に対する抗体もまた試験し、そしてそれら自体への効果を本質的に有さないことが見出されたが、この場合も同様に組み合わせにおいてのみ抑制へのわずかな効果が認められた(図13B)。そのシグナル伝達が休止マウスTregのサプレッサー機能を取り消すかもしくは減少することが報告されている受容体、GITR(Ji,et al.,2004,J.Immunol.,172:5823−5827)およびOX40(CD134)(Takeda,et al.,2004,J.Immunol.,172:3580−3589)に対するアゴニスト抗体を試験し、そして培養したサプレッサー細胞への最小効果を有することが見出された。OX40に対するアゴニスト抗体は、他のドナーよりあるドナーにおいてサプレッサー機能を損なうように思われた(平均32%の取り消し、n=6、範囲15〜75%)。しかしながら、OX40に対するアゴニスト抗体はまた、抑制の取り消しの大きさとほぼ相関して、コントロールMLRの大きさも増加した(図13C)。OX40Lに対する抗体はMLRを抑制したが(ドナー変動を有する)、抑制の見かけの大きさも増加した(コントロール培養物の90%に対して、平均96%の抑制、n=5、範囲90〜98%)(図7C)。
Claims (4)
- T細胞をTreg細胞と接触させることを含んでなるT細胞の増殖の抑制方法であって、該Treg細胞が、
表現型的にCD45RA+の血液細胞の集団から調節性T細胞(Treg細胞)を単離し、該細胞を培養活性化してTreg細胞の集団を生成せしめ、ここで、該Treg細胞は混合リンパ球反応においてT細胞増殖を95%より多いファクターだけ抑制する方法であって、
a)ヒト臍帯血サンプルから単核細胞の集団を単離すること;
b)単核細胞の該集団を単核細胞−抗体複合体の形成に適する条件下でCD25に特異的に結合する抗体と接触させること;
c)単核細胞の該集団から該単核細胞−抗体複合体を実質的に分離し;それによりCD25+細胞の集団を単離すること;
d)該単離されたCD25+細胞の集団を抗CD3/CD28抗体被覆ビーズおよびIL−2の存在下で少なくとも3週間増殖および活性化させること;
e)該増殖されたCD25+細胞の集団をIL−2の存在下で維持すること;
f)該活性化されたCD25+細胞の集団を実質的に分離し;それによってCD25、細胞内CTLA4、CD27およびCD62Lを均一に発現するTreg細胞を生成せしめること
を含んでなる方法により単離されるTreg細胞である

- 該T細胞がCD8 T細胞である請求項1の方法。
- 表現型的にCD45RA+の血液細胞の集団から調節性T細胞(Treg細胞)を単離し、該細胞を培養活性化してTreg細胞の集団を生成せしめ、ここで、該Treg細胞は混合リンパ球反応においてT細胞増殖を95%より多いファクターだけ抑制する方法であって、
a)ヒト臍帯血サンプルから単核細胞の集団を単離すること;
b)単核細胞の該集団を単核細胞−抗体複合体の形成に適する条件下でCD25に特異的に結合する抗体と接触させること;
c)単核細胞の該集団から該単核細胞−抗体複合体を実質的に分離し;それによりCD25+細胞の集団を単離すること;
d)該単離されたCD25+細胞の集団を抗CD3/CD28抗体被覆ビーズおよびIL−2の存在下で少なくとも3週間増殖および活性化させること;
e)該増殖されたCD25+細胞の集団をIL−2の存在下で維持すること;
f)該活性化されたCD25+細胞の集団を実質的に分離し;それによってCD25、細胞内CTLA4、CD27およびCD62Lを均一に発現するTreg細胞を生成せしめること
を含んでなる方法により単離されるTreg細胞。
