Description
(a)コレステリルエステル、トリグリセリド、リン脂質、コレステロール、及び陽イオン性脂質を有機溶媒に溶解する段階;
(b)前記溶解液から有機溶媒を除去して脂質膜を形成する段階;
(c)前記脂質膜に水溶液を加えて水和させる段階;
とを含む核酸伝達用低密度リポタンパク質類似陽イオン性ナノ粒子を製造する方法を提供する。
(a′)コレステリルエステル、トリグリセリド、リン脂質、コレステロール、及び陽イオン性脂質を溶解する段階;
(b′)前記溶解液に水を添加して混合する段階;
(c′)前記混合液を撹拌し、均一溶液を作成する段階;
とを含む核酸伝達用低密度リポタンパク質類似(LDL−like)陽イオン性ナノ粒子の製造方法を提供する。
陽イオン性脂質マイクロエマルション(CLM)を変形溶媒乳化法で調製した。すなわち、22.5mg(45重量%)のコレステリルオレート、1.5mg(3重量%)のグリセリルトリオレート、7mg(14重量%)のDOPE、5mg(10重量%)のエステル化されていないコレステロール、及び14mg(28重量%)のDC−コレステロールをガラス瓶内の2mLのクロロホルム:メタノール(2:1)溶液に溶解した。CLMでDOPE:コレステロール:DC−Cholの間のモル比は9.4:13:26であり、陽イオン性脂質/ヘルパー脂質のモル比は有効組成物に対してほぼ等モル比を提供する値である1.16であった。
前記siRNA−PEGは二硫化物結合を用いて共役処理した。すなわち、感知ストランド(20nmolのVEGFまたはGFP siRNA)の3′にあるヘキシルアミン基によって改質された300μgのsiRNAをPBS(pH7.5)に溶解した。
DC−Chol(CLMに含まれる)/siRNAの重量比がそれぞれ0、1.4、2.8、4.2、5.6、及び8.4となるようにsiRNA−PEGを取り、室温で15分間PBS(pH7.4及び150mMのNaCl)または脱塩水内でCLMを培養した。生成された錯体に対してゲル遅延化、大きさ、及びゼータ電位を測定した。
siRNA−PEG/CLM錯体(20μL)を上述したように相違した重量比で形成した後、2μLの積荷染料(loading dye)と混合した(10×)。22μLの懸濁液全体をトリス−アセテート(TAE)展開緩衝液(running buffer)と共に2%のアガロースゲル上の電気泳動分解用ウェルに積荷した後、100Vで15分間陰極から陽極に移動させた。
細胞培養はPC3(ヒト前立腺癌細胞株)を韓国細胞株銀行(ソウル、大韓民国)で求め、加熱非活性化した10%(v/v)のウシ胎児血清、ペニシリン(100UI/mL)、及びストレプトマイシン(100μg/mL)を含むRPM−1培地1640で成長させた。
実施例3で製造されたCLMの平均直径はレーザ光散乱法で測定した結果103.6±4.5nmであり、TEM観測の結果、図2に示すように前記CLMは球状であることが確認された。CLMのゼータ電位値は+41.76±2.63mVであった。天然LDLと比較すると、DC−Chol及びDOPEの結合によってこの数値が増加し、また表面電荷が陰の値から陽の値に変化した。CLMは調製後、室温で数週間凝集せずに安定性を維持した。
陽イオン性ナノ粒子の細胞毒性を測定するために、MDAMB435細胞を分析の24時間前に96ウェルプレート(ウェルあたり104個の細胞)に播種した。
siRNA−PEGとCLMの錯化過程を特性化するために、siRNA−PEGを室温の水溶液でDC−Chol(CLMに含まれる)/siRNA重量比との関数的関係で培養した。静電気相互作用による錯体形成度は、動的光散乱法(DLS)によってマイクロエマルションの大きさ及びゼータ電位値を観測して算出した。
PEG5Kが二硫化物結合による共役化によってsiRNA伝達系に導入された。siRNA−PEG/CLM錯体形成後、10%血清含有培地でDLSを用いてその大きさを測定した。
siRNA/CLM錯体の細胞吸収能力を評価するために、cy3−ラベル化されたsiRNA−PEGをCLMと錯化反応させ10%血清含有培地内のPC3細胞で2時間培養した。Cy3−siRNA−PEG/CLM錯体の細胞吸収相対量を流速細胞分析法で分析した。
siRNA−PEG/CLM錯体のsiRNA遺伝子抑制効果を血清含有培地に含まれたGFP過発現及びトランスフェクションされた安定したMDAMB435細胞株を用いて測定した。
Claims (6)
- コレステリルエステル及びトリグリセリドを含有するコア脂質部(lipid core part);
前記コア脂質部の上層面に疎水性相互反応によって結合されており、コレステロール、リン脂質、及び陽イオン性脂質を含有する陽イオン性の表面脂質部;及び
静電気的相互作用によって前記陽イオン性の表面脂質部に結合された核酸;
を含む核酸伝達用核酸−低密度リポタンパク質類似(LDL−like)陽イオン性ナノ粒子複合体。 - コレステリルエステル30〜60重量%、トリグリセリド0.1〜10重量%、コレステロール5〜20重量%、リン脂質5〜30重量%、及び陽イオン性脂質10〜50重量%を含有することを特徴とする、請求項1に記載の核酸伝達用核酸−低密度リポタンパク質類似(LDL−like)陽イオン性ナノ粒子複合体。
- コア脂質部と表面脂質部の重量比が30:70〜70:30であることを特徴とする、請求項1に記載の核酸伝達用核酸−低密度リポタンパク質類似(LDL−like)陽イオン性ナノ粒子複合体。
- 前記リン脂質が、ジオレオイルホスファチジルエタノールアミン(DOPE)、パルミトイルオレオイルホスファチジルコリン(POPC)、エッグホスファチジルコリン(EPC)、ジステアロイルホスファチジルコリン(DSPC)、ジオレオイルホスファチジルコリン(DOPC)、ジパルミトイルホスファチジルコリン(DPPC)、ジオレオイルホスファチジルグリセロール(DOPG)、及びジパルミトイルホスファチジルグリセロール(DPPG)からなる群から1つ以上選択されることを特徴とする、請求項1に記載の核酸伝達用核酸−低密度リポタンパク質類似(LDL−like)陽イオン性ナノ粒子複合体。
- 前記陽イオン性脂質が、3β−[N−(N′,N′,N′−トリメチルアミノエタン)カルバモイル]コレステロール(TC−コレステロール)、3β[N−(N′,N′−ジメチルアミノエタン)カルバモイル]コレステロール(DC−コレステロール)、3β[N−(N′−モノメチルアミノエタン)カルバモイル]コレステロール(MC−コレステロール)、3β[N−(アミノエタン)カルバモイル]コレステロール(AC−コレステロール)、N−(N′−アミノエタン)カルバモイルプロパン酸トコフェロール(AC−トコフェロール)、N−(N′−メチルアミノエタン)カルバモイルプロパン酸トコフェロール(MC−トコフェロール)、N,N−ジオレイル−N,N−ジメチルアンモニウムクロライド(DODAC)、N,N−ジステアリル−N,N−ジメチルアンモニウムブロマイド(DDAB)、N−(1−(2,3−ジオレオイルオキシ)プロピル−N,N,N−トリメチルアンモニウムクロライド(DOTAP)、N,N−ジメチル−(2,3−ジオレオイルオキシ)プロピルアミン(DODMA)、N−(1−(2,3−ジオレイルオキシ)プロピル)−N,N,N−トリメチルアンモニウムクロライド(DOTMA)、1,2−ジオレオイル−3−ジメチルアンモニウム−プロパン(DODAP)、1,2−ジオレオイルカルバミル−3−ジメチルアンモニウム−プロパン(DOCDAP)、1,2−ジリネオイル−3−ジメチルアンモニウム−プロパン(DLINDAP)、ジオレオイルオキシ−N−[2−スペルミンカルボキシアミド)エチル}−N,N−ジメチル−1−プロパンアミニウム−リフルオロアセテート(DOSPA)、ジオクタデシルアミドグリシルスペルミン(DOGS)、1,2−ジミリスチルオキシプロピル−3−ジメチル−ヒドロキシエチルアンモニウムブロマイド(DMRIE)、3−ジメチルアミノ−2−(コレスタ−5−エン−3−ベータ−オキシブタン−4−オキシ)−1−(シス,シス−9,12−オク−タデカジエンオキシ)プロパン(CLinDMA)、2−[5′−(コレスタ−5−エン−3.ベータ.−オキシ)−3′−オキサペントキシ)−3−ジメチル−1−(シス,シス−9′,12′−オクタデカジエンオキシ)プロパン(CpLinDMA)、N,N−ジメチル−3,4−ジオレイルオキシベンジルアミン(DMOBA)、1,2−N,N′−ジオレイルカルバミル−3−ジメチルアミノプロパン(DOcarbDAP)、1,2−ジアシル−3−トリメチルアンモニウム−プロパン(TAP)、及び1,2−ジアシル−3−ジメチルアンモニウム−プロパン(DAP)からなる群から1つ以上選択されることを特徴とする、請求項1に記載の核酸伝達用核酸−低密度リポタンパク質類似(LDL−like)陽イオン性ナノ粒子複合体。
- 前記核酸が小干渉リボ核酸(siRNA)、リボソームリボ核酸(rRNA)、リボ核酸(RNA)、ジオキシリボ核酸(DNA)、相補性ジオキシリボ核酸(cDNA)、アプタマー、伝令リボ核酸(mRNA)、運搬リボ核酸(tRNA)、及びアンチセンスオリゴジオキシヌクレオチド(AS−ODN)からなる群から選択されることを特徴とする、請求項1に記載の核酸伝達用核酸−低密度リポタンパク質類似(LDL−like)陽イオン性ナノ粒子複合体。
