Description
被検出物質である抗原に対する抗体により感作させた微粒子を固相微粒子としてクロマトグラフ媒体(例えば、ニトロセルロースからなるメンブレン)に固定化することにより、あるいはこの抗体そのものをクロマトグラフ媒体に直接固定化することにより、検査領域を有するクロマトグラフ媒体を調製する。一方、標識微粒子に被検出物質と特異的に結合可能な抗体を感作させて感作標識微粒子を調製する。この感作標識微粒子を、試料と共に、クロマトグラフ媒体上でクロマトグラフ的に移動させる。
前記被検出物質と特異的に結合する第1の結合物質が付与された検査領域を一部に有するクロマトグラフ媒体の該検査領域に、前記被検出物質と特異的に結合する、視認可能に標識された第2の結合物質を含む前記検体液を浸透させ、
該検体液を前記検査領域に浸透させると共に、または浸透させた後に、前記クロマトグラフ媒体の屈折率との屈折率差Δnが−0.1≦Δn≦0.1の屈折率を有する視認用溶液を、該クロマトグラフ媒体に浸透させ、前記検査領域に前記視認用溶液が浸透している状態で前記検査領域を視認することを特徴とする。
前記検査領域における反応を視認する前に、前記検査領域に浸透させる、前記クロマトグラフ媒体の屈折率との屈折率差Δnが−0.1≦Δn≦0.1の屈折率を有する視認用溶液を備えたことを特徴とする。
図2はイムノクロマト測定方法の工程を模式的に示す図である。図2においては、メンブレン内における、抗原と標識2次抗体の移動、1次抗体、参照用抗体との結合状態等を視認しやすくするため、それぞれ1つもしくは数個だけを模式的に示している。

Claims (6)
- 検体液中における被検出物質の存在の有無を検出するイムノクロマト測定法であって、
前記被検出物質と特異的に結合する第1の結合物質が付与された検査領域を一部に有するクロマトグラフ媒体の該検査領域に、前記被検出物質と特異的に結合する、コロイド状金属粒子またはコロイド状金属酸化物粒子からなる標識物質により視認可能に標識された第2の結合物質を含む前記検体液を浸透させ、
該検体液を前記検査領域に浸透させた後に、前記クロマトグラフ媒体の屈折率との屈折率差Δnが−0.1≦Δn≦0.1の屈折率を有する視認用溶液を、該クロマトグラフ媒体に浸透させ、前記検査領域に前記視認用溶液が浸透している状態で前記検査領域を視認することを特徴とするイムノクロマト測定法。 - 前記屈折率差Δnが−0.07≦Δn≦0.07であることを特徴とする請求項1記載のイムノクロマト測定法。
- 前記視認用溶液がジメチルスルホキシド溶液であることを特徴とする請求項1または2記載のイムノクロマト測定法。
- クロマトグラフ媒体と、該クロマトグラフ媒体の一部に設けられた検査領域に付加された、被検出物質と特異的に結合する第1の結合物質と、該クロマトグラフ媒体を内包し、該クロマトグラフ媒体に検体液を注入するための注入口および前記検査領域を視認するための窓部を備えたケースと、前記クロマトグラフ媒体の、前記注入口から前記検査領域に至るまでの一部に付加された、前記被検出物質と特異的に結合する、コロイド状金属粒子またはコロイド状金属酸化物粒子からなる標識物質により視認可能に標識された第2の結合物質とを備えた検体保持具、および
前記検査領域における反応を視認する前に、前記検査領域に浸透させる、前記クロマトグラフ媒体の屈折率との屈折率差Δnが−0.1≦Δn≦0.1の屈折率を有する視認用溶液を備えたことを特徴とするイムノクロマト測定キット。 - 前記屈折率差Δnが−0.07≦Δn≦0.07であることを特徴とする請求項4記載のイムノクロマト測定キット。
- 前記視認用溶液がジメチルスルホキシド溶液であることを特徴とする請求項4または5記載のイムノクロマト測定キット。
