Description
本発明のワクチン組成物は、担体タンパク質、好ましくは髄膜炎菌P64kの外膜複合体由来のタンパク質に結合させた、ヒト組換え型インターロイキン−2(hIL−2r)を有効成分として含む(0474313 EP A2及びU.S.5,286,484)。さらに、このワクチン組成物は、適切なアジュバントを含む。本発明のワクチン組成物は、アジュバントとしてモンタニドISA 51を用いるのが好ましい。
hIL−2r及び担体タンパク質間の結合は、化学的結合又は遺伝子工学の技術により得られる融合タンパク質の作製により可能である。
hIL−2r及びP64kタンパク質間にタンパク質結合を達成するために、両成分を様々な割合、20:1から5:1(P64kタンパク質のモルに対するhIL−2rのモル)まで、好ましくは10:1(P64kタンパク質のモルに対するhIL−2rのモル)で混合する。グルタルアルデヒドを、0.02%及び0.1%間にある、好ましくは0.5%の最終濃度でこの混合物に添加し、室温(RT)で1〜5時間インキュベートする。最後に、これをリン酸塩緩衝食塩水(PBS)溶液中で充分に透析した。
結合反応は、10%SDSポリアクリルアミドゲル電気泳動により確認した(Laemmli UK (1970)Nature 277,680〜685)。
ヒトIL−2(447pb)をコードする遺伝子を、特異的プライマーを用いてポリメラーゼ連鎖反応法(PCR)により増幅する。得られたDNAフラグメントを消化し、生じたタンパク質が両分子の1コピー又はそれ以上を含むように、担体タンパク質をコードする遺伝子をクローニングする発現ベクターの特異的結合部位に結合させる。哺乳動物細胞及び細菌又は酵母菌の任意の発現ベクターを用いることができる。またベクターは、担体タンパク質のN末端に、6つのヒスチジンを含むことができる。得られたプラスミドは、アガロースゲル電気泳動での制限酵素分析、酵素シークエンス2,0(the enzyme Sequenace 2,0)(Amersham−USB)を用いるDNA塩基配列分析、及び最後に特異的抗hIL−2モノクローナル抗体を用いる「ウエスタンブロット」法で、大腸菌の任意の発現株で融合タンパク質産物の分析により確認する。タンパク質を得るために、細菌壁を強い破断方法を用いて破壊後、タンパク質を硫酸アンモニウムによる差次的な沈殿法とクロマトグラフィー法の組合せにより精製する。最後に、タンパク質を無菌条件下で濾過し、後に使用するまで−20℃で保存するか又は凍結乾燥して4℃で保存する。
IL−2のアミノ酸配列由来の化学合成により得たペプチドを、P64kタンパク質に、U.S.5,984,018で記載されているように化学的結合により結合できる。或いは、hIL−2r及びP64kタンパク質由来のペプチドの融合タンパク質は、前節に記載のものと同じ方法により本質的に得られる。実施例は下記の領域からのペプチドを含むことができる。すなわち、

この電気泳動は、10%SDSポリアクリルアミドゲル電気泳動により実施できる(Laemmli U.K.(1970)Nature 277,680〜685)。電気泳動では、1ウェル当たり15μgの試料を適用し、クーマシーで染色する。
− ワクチン組成物の免疫原性
動物で本発明のワクチン製剤の免疫原性を試験するために、生後8〜12週目の体重18〜20gの雌BALB/cマウスを使用した。実験の間、マウスを実験動物優良飼育及び使用基準に従い標準作業手順書(SOP)により定められた給餌及び処置の標準条件で飼育した。
− スケジュールA、hIL−2r−P64k/モンタニドISA 51ワクチン、0.1mL中に結合したhIL−2rの4μg当量の4用量を、肢の免疫部位を交互に換えて、2週間ごとに筋肉内投与する。
− スケジュールB、hIL−2r−P64k/モンタニドISA 51ワクチン、0.1mL中に結合したhIL−2rの10μg当量の4用量を、筋肉内投与する。最初の2用量は、2つの肢の独立した免疫部位に同時に投与し、2週間後に2用量を別の2つの肢に投与する。
特異的抗体又はB細胞を測定することにより、遮断抗体を測定するか又は末梢血液で特異的免疫応答を評価する最高水準の技術で利用可能な方法の1つにより、自己抗体力価を血清で検出できる。
hIL−2r−P64k/モンタニドISA 51ワクチンにより誘導される抗hIL−2r抗体は、免疫した動物の血清を用いる、以下に記載の間接ELISA(酵素結合抗体免疫吸着アッセイ)により評価できる。
96穴平底マイクロタイタープレート(Costar,High Binding,USA)は、0.1M炭酸塩−重炭酸塩pH9.6溶液中で10μg/mLの濃度に調製したhIL−2r(50μL/ウェル)を含む。プレートを4℃で終夜インキュベートし、0.05%のTween 20を含むPBS(PBS−Tween 20)を用いて200μL/ウェルで2回洗浄した。100μL(1%ウシ血清アルブミン(PBS−BSA 1%)を含む)/ウェルで37℃で1時間インキュベート後、プレートをPBS−Tween 20(200μL/ウェル)で洗浄し、PBS−BSA1%中、異なる希釈の血清試料50μL/ウェルを添加する。37℃で1時間インキュベート後、プレートをPBS−Tween 20で洗浄し、PBS−BSAで1/5000に希釈したアルカリホスファターゼ結合ヒツジ抗マウス血清(Jackson Immunoresearch Laboratories)を50μL/ウェルで添加し、37℃で1時間インキュベートした。プレートを洗浄し、pH9.8、ジエタノールアミン緩衝液中にp−ニトロフェニルリン酸(Sigma)1ng/mLを含む基質溶液を50μL/ウェルで添加する。室温(RT)で30分プレートをインキュベートした後に、反応産物の吸光度を405nmで、ELISAリーダー(Organon Teknika,Austria)で測定する。
B細胞又は特異的抗体を測定することにより、遮断抗体を測定するか又は末梢血液で特異的免疫応答を評価する最高水準の技術で利用可能な方法の1つにより、自己抗体力価は血清で検出できる。
Gonzalez、G.、ら、(2002)Vaccine Research 5,233〜244で記載されているように、EGFに基づくワクチンにより誘導される抗hEGF抗体を、免疫した動物の血清を用いる間接ELISA(酵素結合免疫吸着アッセイ)により評価できる。
IL−2依存性T細胞系CTLL−2を、連続的増殖下で、hIL−2hの1U/mLを含むRPMI−1640培地に保持する。CTLL−2の培養を、細胞懸濁液中0.5×105〜106細胞/mLと共に8〜20%のウシ胎児血清(FCS)を含むRPMI−1640が25mL入っている75cm2細胞培養フラスコで実施する。細胞はin vitroでの増殖の2日後に使用する。
アッセイを実施するため、細胞をin vitro培養から採取し、RPMI−1640又はPBSで少なくとも4回洗浄する。96穴平底培養プレート(Costar,High Binding,USA)へ、5×103細胞を播種する。これらの細胞を、hIL−2−P64k/モンタニドISA 51ワクチンで免疫した動物からのIL−2に対するELISA力価が1:10000の希釈血清又は抗IL−2特異的mAbで処理し、hIL−2r(1U/mL)を添加する。培養を5%CO2インキュベーターで、37℃の湿潤な空気中で24時間実施する。培養の最後の18〜24時間に[3H]チミジン(1μCi/ウェル)をパルスすることにより増殖を測定した。チミジン取り込みは、液体シンチレーションカウンターで測定する。全ての操作は、無菌状態で実施する。
本発明のワクチンの製剤の抗腫瘍効果を動物で試験するために、生後8〜12週間目の体重18〜20gの雌BALB/cマウスを使用した。動物はすでに詳述したスケジュールA及びBに従って、hIL−2−P64k/モンタニドISA 51ワクチンで免疫可能であり、1週後に乳癌F3IIのような腫瘍細胞系を、5×104細胞/動物で、同所性に皮下投与経路で接種する。触知可能な腫瘍を有するマウスを陽性と記録し、腫瘍増殖はカリパーを用いて測定し、最長の表面距離(a)及びこれに対して垂直な幅(b)を規定し、腫瘍サイズはa×bとして記録した。腫瘍は定期的に調べる。
驚くべきことに、予想外にも、本発明の著者らは、悪性腫瘍を有する対象で、IL−2のIL−2レセプターへの結合中和、この腫瘍に対する免疫応答が増強され、腫瘍サイズの減少を誘導することを見出した。以前の技術の状態は、そのような処置は腫瘍に対する個体の免疫系の応答の抑制を生じるであろうことを示していた。
減少が抗IL−2モノクローナル抗体で受動的治療を受けて得られる場合と同様に、対象が本発明のワクチンの組成物による能動的治療を受けている場合、循環血中のIL−2濃度が実質的に低下する場合、本著者らは腫瘍増殖に及ぼすこの効果が得られることを見出した。
その理由のために、本発明は悪性腫瘍患者の治療のために結果として否定できない利益になり、且つ、これらの場合用いられる通常の治療よりも患者にとってはるかに不快でなく積極的で有効で簡単なIL−2によるワクチン投与法を提供する。
下記の実施例は、本発明の対象のワクチン組成物の免疫学的有効性を例証する実験の詳しい説明を含む。
ワクチンで用いた髄膜炎菌由来のタンパク質P64kは、EP 0474313A2及びU.S.5,286,484で記載される組換えDNA技術により得た。
hIL−2r−P64k/モンタニドISA 51ワクチンにより誘導された抗体応答
ヒトインターロイキン−2に対する免疫原性を増進するために、ヒトインターロイキン−2を、髄膜炎菌由来の担体タンパク質P64kタンパク質に化学的に結合した。化学的結合は、グルタルアルデヒド法により行うことができる(U.S.5,984,018)。10%SDSポリアクリルアミドゲル電気泳動により、各成分の試料(hIL−2r、P64k)を別々に添加し、化学的結合体hIL−2r−P64kと分子量の標準パターンを比較して結合効率が確認される。hIL−2r−P64kを添加したレーンで連続的に存在するため、得られた結合型を確認することは可能である(図1)。
hIL−2r−P64k/モンタニドISA 51ワクチンにより誘導されたhIL−2rに対する抗体応答を評価するために、BALB/cマウスを、スケジュールA及びBで免疫した。hIL−2r−P64k/モンタニドISA 51ワクチンに応答した動物からの高度免疫血清を陽性対照として用い、免疫前血清を陰性対照として用いた。血清の吸光度が、免疫前血清の吸光度の平均値プラス5倍の標準偏差より大きい最大希釈を、免疫した動物の抗体力価と定義した。対照動物で力価を定義するため、陰性対照として免疫前血清の代わりにPBS−BSA1%を用いた場合を除き、同じ先の判定基準を用いた。
1:100から1:50000まで力価が達する抗体応答が誘導された。この免疫プロトコールは約52日間続く、その理由は短期間に同程度であるか又はより高い抗体力価を得るためにプロトコールを改変する必要があり、したがって我々はスケジュールBを用いて図2に示すように同程度の結果を得た。
hIL−2−P64k/モンタニドISA 51ワクチンで免疫した動物由来血清で処理したCTLL−2細胞系の増殖試験
hIL−2r−P64k/モンタニドISA 51ワクチンで免疫した動物で産生される血清抗体のin vitroでのIL−2結合能は、血清抗体と共にIL−2依存性T細胞系CTLL−2を培養することにより評価した。CTLL−2細胞をIL−2の存在下で培養プレートに播種して増殖させ、約1:10000抗体力価を有する動物からの種々の希釈血清を添加した。血清濃度とCTLL−2細胞系増殖の阻害の間に、直接的相関が認められた(図3)。この血清は、ワクチン接種した動物由来の血清のin vitroでのIL−2中和能力を実証する。
hIL−2−P64k/モンタニドISA 51ワクチンで処置した動物における抗腫瘍性の実験
スケジュールBに従い、動物をhIL−2r−P64k/モンタニドISA 51ワクチンで免疫した。1週間後に、動物を5×104 F3II腫瘍細胞に曝露した。腫瘍増殖カイネティクスは、PBS−P64k/モンタニドISA 51で免疫した対照群と比較して、hIL−2r−P64k/モンタニドISA51で免疫した動物でより緩慢であった(図4a)。これら2群間の腫瘍サイズに統計有意差が認められた。
抗IL−2特異的mAbで処理したCTLL−2細胞系の増殖試験
S4B6(ATCC # HB−8794)ハイブリドーマにより産生されたhIL−2rに対する特異的抗体のin vitroでのIL−2結合能を、IL−2依存性T細胞系CTLL−2と共にそれらを培養することにより評価した。CTLL−2細胞を、IL−2存在下で培養プレートに播種して増殖させ、種々の希釈抗体を添加した。抗体濃度とCTLL−2細胞系増殖の阻害の間に、直接的な相関が認められた(図5)。モノクローナル抗体のin vitroでのIL−2中和能力を実証する、1:100希釈抗体によりCTLL−2細胞系増殖を80%以上阻害できることが見出された。
抗IL−2 mAb及び抗CD25 mAb(ATCC−PC61)の抗腫瘍効果、F3II実験モデル(乳癌)
マウスを、日用量1mgの特異的モノクローナル抗体(抗CD25 mAb又は抗IL−2 mAb)を、連続して5日間投与した。2日後に皮下同所性注射により、マウスを実験的乳癌F3IIに5×104細胞/マウスで曝露した。最長の表面距離(a)及びこれに対して垂直な幅(b)を定期的に測定した。対照群の腫瘍サイズ(PBSを投与した)は、抗IL−2 mAbを投与した群よりも大きかった(図6)。2つの実験群間の腫瘍サイズは統計的に相異した。しかしながら抗CD25 mAb(ATCC―PC61)を投与した動物の腫瘍サイズは、対照群と同程度であった。この結果はCD25制御細胞の除去が影響を与えない腫瘍においてさえも、IL−2中和抗体は強い抗腫瘍効果を有することを示す。
抗IL−2 mAb及び抗CD25 mAb(ATCC−PC61)の抗腫瘍効果。EL4実験モデル(リンパ腫)
C57BL/6動物を、EL4リンパ腫(5×104細胞/マウス)に皮下で曝露した。動物に抗CD25 mAb(PC61)の1mgの1用量を、腫瘍への曝露の2日前に静脈内投与するか、又は抗IL−2 mAb(S4B6)の1mgの日用量を、腫瘍接種の6日前に開始して連続5日間静脈内投与した。別の群は、以前に記載した同様のスケジュールに基づき抗CD25及び抗IL−2 mAbsを同時投与した。
マウスの各群の腫瘍増殖を記録した。腫瘍へ曝露後毎週2回、各マウスにおける腫瘍サイズを、2つの垂直な大きさで測定した。EL4腫瘍に関する統計的差異は、*p=0.0232(**p=0.0039)、***p<0.0001である。
図7は、2つのモノクローナル抗体の組合せが、腫瘍増殖に及ぼす最も強い効果を有することを示す。
自己抗体抗IL−2の誘導は、EGF癌ワクチンに対する応答に影響を及ぼさない
hEGF−P64k/モンタニドISA 51ワクチンにより誘導されるhEGFに対する抗体応答が、自己抗体の誘導を伴うhIL−2rによるその前のワクチン投与により影響を受けるかどうかを評価するために、BALB/c動物をスケジュールAに従いhIL−2r−P64k/モンタニドISA 51で調製したワクチンで免疫し、19日後にhEGF−P64k/モンタニドISA 51又はhEGF/モンタニドISA 51で調製したワクチンで免疫した。hEGFのワクチンで免疫した全ての実験で、0.1mLの量で調製したワクチンに結合したhEGFの4μg当量を筋肉内経路で投与した。hEGF−P64k/モンタニドISA 51ワクチンに応答した動物の高度免疫血清を、陽性対照として用い、陰性対照には免疫前血清を用いた。血清の吸光度が、免疫前血清の吸光度の平均値よりさらに5倍標準偏差より大きい最大希釈を、免疫した動物の抗体力価と定義した。対照動物で力価を定義するため、陰性対照として免疫前血清の代わりにPBS−BSA1%を用いた場合を除き、同じ先の判定基準を用いた。EGF/モンタニドISA 51又はhEGF−P64k/モンタニドISA 51によるワクチン接種により誘導されるEGFに対する抗体の力価は、図8で示すように、hIL−2rに対する自己抗体の誘導によっては影響を受けないことが得られた。
in vivoでインターロイキン−2の中和により、腫瘍保持宿主のリンパ節細胞で評価した細胞溶解活性を回復する
腫瘍保持宿主で、名目抗原に対する免疫応答に及ぼすIL−2の中和効果を評価した。雌C57BL/6Jマウス(H−2b)は、標準飼育条件下で維持した。全ての実験で、6及び12週齢の間のマウスを用いた。卵白アルブミン(OVA)及びペプチド:OVAグレードVII(Sigma,St.Louis,MO)は、これらの実験の全体を通じて用いたモデルタンパク質Agであった。優占するペプチドOVA275−264(SIINFEKL)は、>90%の純度で用いた。増殖反応測定法:C57BL/6マウスを、左側腹の皮下経路でMB16F10腫瘍の104細胞又はPBSに曝露した。腫瘍直径は定期的に測定する。3週間後にこれらのマウスを、試験開始時に100μg/マウスのポリイノシン酸ポリシチジル酸[poly I:C](PIC)(Sigma,St.Louis,MO)と共に1mgのOVAで皮下に免疫し、その後の2日はPICのみを投与した。同時に動物に、hIL−2r(αlL−2)対する特異的モノクローナル抗体又はPBSを5日連続して投与した。差次的に蛍光色素CFSE(Molecular Probes,Paisley,UK)で標識した未処置のマウスからの脾臓細胞を用いて、in vivoでの細胞溶解活性を測定した。CFSEhighで標識した細胞を標的として用い、SIINFEKL(1μM;37℃で90分、5%CO2)でパルスしたが、CFSElowで標識した細胞は内部対照としてパルスしなかった。ペプチドでパルスした標的細胞を、遊離のペプチドを除去するために充分に洗浄後、前もって免疫したマウスに1:1の比率で同時に静注した。16時間後リンパ節及び脾臓を摘出し、両方の蛍光強度(CFSElow及びCFSEhigh)に対応する全事象を、フローサイトメトリーにより測定した。非コーティング対SIINFEKLコーティング(CFSEint/CFSEhigh)のパーセンテージ間の比率を算出して、細胞毒性の数値を得る。OVAペプチドに対するリンパ節細胞の細胞溶解活性を、最大の応答を与えたOVAプラスPICで免疫したC57BL/6マウスでin vivoで評価した。日用量1mgの抗IL−2モノクローナル抗体の連続5日間の静脈内注射は、これらの動物で細胞溶解性応答に影響を及ぼさない。MB16F10腫瘍に曝露されたC57BL/6マウスは、OVAに対する細胞溶解活性低下を伴う免疫抑制になる。しかしIL−2中和能を有するモノクローナル抗体をin vivoで投与すると、リンパ節細胞の免疫応答が回復する(図9)。
in vivoでのインターロイキン−2の中和により、腫瘍保持宿主の脾臓細胞で評価した細胞溶解活性が回復する
腫瘍保持宿主での名目抗原に対する免疫応答に及ぼすIL−2中和効果を評価した。C57BL/6マウスを、OVAプラスPICで免疫してOVAペプチドに対する脾臓細胞の細胞溶解活性をin vivo評価し、最大の応答を得た。日用量1mgの抗IL−2モノクローナル抗体の連続5日間の静注投与は、これらの動物で細胞溶解性応答に影響を及ぼさない。
MB16F10腫瘍に曝露されたC57BL/6マウスは、OVAに対する細胞溶解活性低下を伴う免疫抑制になる。しかし、IL−2中和能を有するモノクローナル抗体をin vivoで投与すると、脾臓細胞の免疫応答が回復する(図10)。
調製したワクチンhIL−2r−P64k/モンタニドISA 51で免疫した動物での血液細胞の計数
調製したhIL−2r−P64k/モンタニドISA 51ワクチンでの免疫が、最初の免疫後の100日まで、(スケジュールAに従い)調製したワクチンで免疫した動物又はワクチン接種していない動物と同程度に、赤血球、白血球及び血小板を計数した血液細胞数の変化を引き起こすどうかを評価する目的。図11の1つのように、1つは分析した群間に細胞数値に差異を得なかった。
Claims (3)
- 上皮腫瘍の治療のための、抗IL−2モノクローナル抗体を含む治療用製剤であって、該抗体はIL−2がその受容体へ結合することを遮断するものであり、及び該抗体は対象における循環IL−2濃度を減少させるものである、上記治療用製剤。
- 前記抗IL−2モノクローナル抗体がヒトIL−2に対して作成されるものである、請求項1に記載の治療用製剤。
- 抗IL−2モノクローナル抗体を腫瘍増殖因子に基づく癌ワクチンと組み合わせることを含む、請求項1又は2に記載の治療用製剤。
