1,3−ジハロ−5,5−ジアルキルヒダントインは例えば工業用冷却水、再生水、および廃水のような水系に対する殺生物剤として有効である。
本発明には特に、1,3−ジブロモ−5,5−ジメチルヒダントインは水処理に使用した場合、N,N’−ブロモクロロ−5,5−ジアルキルヒダントインの1種または混合物を使用した場合に必要とされる僅か半分の投与レベルで米国環境保護局U.S.Environmental protection Agency)により義務づけられた要求を達成できるという驚くべき発見が含まれている。即ちこの発見により水を効果的に消毒するために極端に低い濃度の1,3−ジブロモ−5,5−ジメチルヒダントインを使用することができる、同時に優れた微生物の抑制を行うことができる。さらにこのような低濃度で効果的に使用できる能力のために、消毒による副成物を著しく減少させることができる。
本発明の特徴の中には、上記の圧縮された形は接合剤なしで製造し提供することができるという事実がある。この予測できない特徴は、先ず平均粒径が少なくとも約175μの1,3−ジブロモ−5,5−ジメチルヒダントインを製造できるという事実によって可能になる。実際平均粒径が少なくとも約200μ、少なくとも約300μおよび少なくとも約600μの1,3−ジブロモ−5,5−ジメチルヒダントインは、2000年1月18日付けの同じ出願人による同時出願の米国特許願09/484,844号に記載された方法を用いて製造することができる。接合剤を含まない圧縮された形の製造法は2000年1月18日付けの同じ出願人による同時出願の米国特許願09/484,687号に記載されている。
1,3−ジブロモ−5,5−ジメチルヒダントインの粒剤を使用して夏季の61日間水泳プールを消毒した。この粒剤を市販品の供給業者から得た3ポンドのプールの浮きの中に入れた。最初浮きに最大容量を装入し、必要に応じ再装入を行った。砂のフィルターを取り付けた地上にある46,935L(12,400ガロン)のビニルでライニングした水泳プールをこの試験に使用した。プールは全部が日光に露出している。1.5馬力のポンプを用いて午前7時から午後7時まで水を循環させた。1日に2回、即ち朝(午前8時)1回と午後(午後1時)1回ハロゲンのレベル、pHおよび温度を測定した。工業標準と比較し易くするために臭素のレベルは塩素として表した。このような塩素値から臭素値へ変換するためには、塩素値に2.25を乗じなければならない。アルカリのレベルは1日に1回測定した。5,5−ジメチルヒダントイン、臭素イオンのレベル、およびカルシウム硬度の濃度は1週間に1回測定した。
冷却塔用水中の微生物の抑制に対する1,3−ジブロモ−5,5−ジメチルヒダントインの効果を研究した。冷却塔は交叉流の設計の500トンのユニット2個から成っていた。全システムに含まれる容積は52,990L(14,000ガロン)であり、この塔は中程度の効率をもったフィルム充填材を含んでいた。塔から出た水が2個の300トンの空調機(冷却器)のコイルを冷却する。この塔は典型的にはpH約9.1において4サイクルの濃度で動作する。減速は伝導度によって制御される。調合用水は軟化した良質の水道水から成っていた。調合用水はカルシウム濃度が非常に低い(<10mg/L)が、pHは高い(8.7)。アルカリ度は145mg/L(CaCO3として)であり、シリカ・レベルは28mg/Lであった。この塔は腐蝕および沈澱の抑制に通常のポリ燐酸塩/モリブデン酸塩/フォスフォン酸塩プログラムを使用している。
235gのNaOH(5.85モル)を1800gの水に溶解し、375gの5,5−ジメチルヒダントイン(2.93モル)をこのNaOH溶液に加える。臭素の貯蔵器に935gのBr2(5.85モル)を入れる。1リットルのジャケット付きフラスコにBr2および5,5−ジメチルヒダントイン/NaOH溶液を供給し、冷却浴を用いて25℃に保つ。5,5−ジメチルヒダントイン/NaOH溶液はBr2と同時に、但し別々に反応フラスコに一緒に供給する。5,5−ジメチルヒダントイン/NaOH溶液の供給はBr2の供給を開始する少し前(例えば3〜4分前)に開始する。5,5−ジメチルヒダントイン/NaOH溶液の供給速度は10mL/分であり、Br2の供給速度は1.60〜1.70mL/分である。反応混合物を350〜400rpmの速度で機械的撹拌機を用いて撹拌した。反応中pHの範囲は7.4〜7.9である。反応フラスコ中の液面が一定に保たれるような速度で、反応の進行と共に生成するスラリを捕集する。生成物の500mLの部分を、各部分について平均30分の時間で反応フラスコを通して捕集した。5,5−ジメチルヒダントイン/NaOH溶液の供給が終わった時、臭素貯蔵器の中に86gのBr2(0.54モル)が残っていた。
44gのNaOH(1.1モル)を338gの水に溶解し、70.4gの5,5−ジメチルヒダントイン(0.55モル)をこのNaOH溶液に加える。臭素の貯蔵器に175.1gのBr2(1.1モル)を入れる。反応フラスコにBr2および5,5−ジメチルヒダントイン/NaOH溶液を供給し、加熱浴を用いてこれを35℃に保つ。反応フラスコに1,3−ジブロモ−5,5−ジメチルヒダントインの濾液(母液)の残りの部分約200ml(238g)を装入する。反応混合物を400rpmの速度で機械的撹拌機を用いて撹拌した。反応中pHの範囲は6.9〜8.2であった。0.5時間の添加時間の間反応温度は37℃で安定していた。試薬の供給が終わった時、橙色のスラリを35℃で濾過し、650mLの水で洗滌した。得られた白色の固体を一晩窒素気流の下で乾燥する。1,3−ジブロモ−5,5−ジメチルヒダントインの分離された収量は147.6gであり、収率は94%、1,3−ジブロモ−5,5−ジメチルヒダントインの活性臭素含量はヨード滴定法で決定して55.1重量%(理論値の98.6%)であった。
44gのNaOH(1.1モル)を338gの水に溶解し、70.4gの5,5−ジメチルヒダントイン(0.55モル)をこのNaOH溶液に加える。臭素の貯蔵器に172.0gのBr2(1.07モル)を入れる。反応フラスコにBr2および5,5−ジメチルヒダントイン/NaOH溶液を供給し、加熱浴を用いてこれを67℃に保つ。反応フラスコに1,3−ジブロモ−5,5−ジメチルヒダントインの濾液(母液)の残りの部分約200ml(238g)を装入する。5,5−ジメチルヒダントイン/NaOH溶液はBr2と同時に、但し別々に反応フラスコに一緒に供給する。臭素は窒素で希釈し、反応フラスコ中の溶液の表面より下に供給する。反応混合物を400rpmの速度で機械的撹拌機を用いて撹拌した。反応中pHの範囲は6.7〜7.1であった。0.5時間の添加時間の間反応温度は67℃で安定していた。試薬の供給が終わった時、橙色のスラリを反応フラスコからビーカーに取り出して徐々に冷却させる。このスラリを45℃で濾過し、各500mLの水で2回洗滌した。得られた白色の固体を一晩窒素気流の下で乾燥する。1,3−ジブロモ−5,5−ジメチルヒダントインの分離された収量は130.5gであり、5,5−ジメチルヒダントインに関する収率は83%、Br2に関する収率は85%であった。1,3−ジブロモ−5,5−ジメチルヒダントインの活性臭素含量はヨード滴定法で決定して55.9重量%(理論値の100%)であった。生成物の代表的な乾燥した試料に関してこの操作で生成した1,3−ジブロモ−5,5−ジメチルヒダントイン生成物の粒径のデータを表6にまとめる。
354gのNaOH(8.85モル)を2700gの水に溶解し、562gの5,5−ジメチルヒダントイン(2.93モル)をこのNaOH溶液に加える。反応フラスコに1,3−ジブロモ−5,5−ジメチルヒダントインの濾液(母液)の残りの部分約500mlを装入する。臭素の貯蔵器に935gのBr2(5.85モル)を入れる。ジャケット付きフラスコにBr2および5,5−ジメチルヒダントイン/NaOH溶液を供給し、加熱または冷却をせずに同時にしかし別々にBr2を供給する。5,5−ジメチルヒダントイン/NaOH溶液の供給速度は10mL/分であり、Br2の供給速度は最初1.70mL/分であるが、後で1.68mL/分に調節し、反応混合物のpHを約7.0に保つ。反応混合物を400rpmの速度で機械的撹拌機を用いて撹拌した。反応温度は約42℃で安定した。反応フラスコ中の液面が一定に保たれるような速度で、反応の進行と共に生成するスラリを捕集する。生成物の500mLの8個の部分を、各部分について平均30分の時間で反応フラスコを通して捕集した。反応中全部で1374.5g(8.59モル)のBr2を加えた。
89gのNaOH(2.2モル)を676gの水に溶解し、141gの5,5−ジメチルヒダントイン(1.1モル)をこのNaOH溶液に加える。臭素の貯蔵器に350gのBr2(2.2モル)を入れる。反応フラスコにBr2および5,5−ジメチルヒダントイン/NaOH溶液を供給し、加熱浴を用いてこれを67℃に保つ。反応フラスコに1,3−ジブロモ−5,5−ジメチルヒダントインの濾液(母液)の残りの部分約400mlを装入する。5,5−ジメチルヒダントイン/NaOH溶液はBr2と同時にしかしそれとは別々に供給する。反応混合物を400rpmの速度で機械的撹拌機を用いて撹拌した。反応中pHの範囲は6.8〜7.1であった。66分の添加時間の間反応温度は67℃で安定していた。試薬の供給が終わった時、橙色のスラリを43℃で濾過し、1000mL(2×500mL)の水で洗滌した。得られた白色の固体を一晩窒素気流の下で乾燥する。1,3−ジブロモ−5,5−ジメチルヒダントインの分離された収量は212.5gであり、Br2に関する収率は77%、5,5−ジメチルヒダントインに関する収率は68%であった。1,3−ジブロモ−5,5−ジメチルヒダントインの活性臭素含量はヨード滴定法で決定して55.9重量%(理論値の100%)であった。
88gのNaOH(2.2モル)を338gの水に溶解し、140.8gの5,5−ジメチルヒダントイン(1.1モル)をこのNaOH溶液に加える。臭素の貯蔵器に352gのBr2(2.2モル)を入れる。反応フラスコにBr2および5,5−ジメチルヒダントイン/NaOH溶液を供給し、加熱浴を用いてこれを69℃に保つ。反応フラスコに1,3−ジブロモ−5,5−ジメチルヒダントインの濾液(母液)の残りの部分約200ml(240g)を装入する。5,5−ジメチルヒダントイン/NaOH溶液はBr2と同時にしかしそれとは別々に供給する。反応混合物を400rpmの速度で機械的撹拌機を用いて撹拌した。反応中pHの範囲は6.8〜7.0であった。66分の添加時間の間反応温度は68〜69℃で安定していた。試薬の供給が終わった時、橙色のスラリを40℃で濾過し、500mLの水で洗滌した。得られた白色の固体を一晩窒素気流の下で乾燥する。285.5gのBr2(1.78モル)が反応フラスコに供給された。分離された1,3−ジブロモ−5,5−ジメチルヒダントインの収量は186.8gであり、Br2に関する収率は73%、5,5−ジメチルヒダントインに関する収率は60%であった。1,3−ジブロモ−5,5−ジメチルヒダントインの活性臭素含量はヨード滴定法で決定して53.4重量%(理論値の96%)であった
表7に実施例7および8の生成物に対する粒径のデータをまとめる。
44.2gのNaOH(1.1モル)を338gの水に溶解し、70.4gの5,5−ジメチルヒダントイン(0.55モル)をこのNaOH溶液に加える。臭素の貯蔵器に173gのBr2(1.08モル)を入れる。反応フラスコにBr2および5,5−ジメチルヒダントイン/NaOH溶液を供給し、加熱浴を用いてこれを57℃に保つ。反応フラスコに1,3−ジブロモ−5,5−ジメチルヒダントインの濾液(母液)の残りの部分約200ml(244g)を装入する。5,5−ジメチルヒダントイン/NaOH溶液はBr2と同時にしかしそれとは別々に供給する。反応混合物を400rpmの速度で機械的撹拌機を用いて撹拌した。反応中pHの範囲は6.8〜7.2であった。臭素の供給速度を調節して所望のpHを維持する。33分の添加時間の間反応温度は57℃で安定していた。試薬の供給が終わった時、橙色のスラリを38℃で濾過し、500mLの水で洗滌した。得られた白色の固体を一晩窒素気流の下で乾燥する。285.5gのBr2(1.78モル)が反応フラスコに供給された。分離された1,3−ジブロモ−5,5−ジメチルヒダントインの収量は139.8gであり、Br2に関する収率は97%、5,5−ジメチルヒダントインに関する収率は89%であった。1,3−ジブロモ−5,5−ジメチルヒダントインの活性臭素含量はヨード滴定法で決定して55.7重量%(理論値の99.7%)であった。
44.2gのNaOH(1.1モル)を338gの水に溶解し、70.3gの5,5−ジメチルヒダントイン(0.55モル)をこのNaOH溶液に加える。臭素の貯蔵器に172.5gのBr2(1.08モル)を入れる。反応フラスコにBr2および5,5−ジメチルヒダントイン/NaOH溶液を供給し、加熱浴を用いてこれを48℃に保つ。反応フラスコに1,3−ジブロモ−5,5−ジメチルヒダントインの濾液(母液)の残りの部分約200mlを装入する。5,5−ジメチルヒダントイン/NaOH溶液はBr2と同時にしかしそれとは別々に供給する。反応混合物を400rpmの速度で機械的撹拌機を用いて撹拌した。反応中pHの範囲は6.8〜7.2であった。臭素の供給速度を調節して所望のpHを維持した。34分の添加時間の間反応温度は48℃で安定していた。試薬の供給が終わった時、橙色のスラリを38℃で濾過し、500mLの水で洗滌した。得られた白色の固体を一晩窒素気流の下で乾燥する。分離された1,3−ジブロモ−5,5−ジメチルヒダントインの収量は144.8gであり、Br2に関する収率は94%、5,5−ジメチルヒダントインに関する収率は92%であった。1,3−ジブロモ−5,5−ジメチルヒダントインの活性臭素含量はヨード滴定法で決定して55.0重量%(理論値の98.4%)であった。
この実施例の方法は連続法で行われた。5,5−ジメチルヒダントイン/NaOHの供給溶液は5,5−ジメチルヒダントインを9重量%のNaOH溶液に加えることにより、例えば5,5−ジメチルヒダントインの濃度が約1.1Mになるようにして加えた。5,5−ジメチルヒダントイン/NaOH溶液はBr2と同時にしかしそれとは別々に供給する。フラスコを加熱浴の中に吊り下げる。反応混合物を500rpmの速度で機械的撹拌機を用いて撹拌した。反応混合物のpHを約7.0±0.2に維持し、反応温度を55℃に保つ。生成物の10個の部分を1部分当たり平均30分の時間で捕集する。分離された1,3−ジブロモ−5,5−ジメチルヒダントインの収率は収量は5,5−ジメチルヒダントインに関して90%であり、Br2に関して92%であった。白色結晶生成物である1,3−ジブロモ−5,5−ジメチルヒダントインの純度は理論的な臭素含量に関して99.8%であった。部分5〜10は定常的な操作条件の間に生じた生成物の粒径を表している。表9には、連続法の定常的な操作の間に採取された各部分の試料に関して部分5〜10に関する平均粒径および粒径分布データをまとめる。この決定によって生成物の双峰分布がつくられていることが示される。生成物の全体的な平均粒径は512.3μであった。
実施例11と同様にして355g(8.87モル)を3550gの水に溶解して供給溶液をつくった。これに500g(4.37モル)の5,5−ジメチルヒダントインを加えた。水性反応混合物のpHを7.0±0.2に保つように同時供給流の調節を行った。温度を55℃に保った。臭素(Br2)の全供給量は1359.4g(8.50モル)であった。実施例11と同様に反応混合物の10個の部分を捕集した。しかしこの操作においては平均滞在時間が部分1個当たり約1時間になるように添加速度を調節した。分離された1,3−ジブロモ−5,5−ジメチルヒダントインの収率は使用した5,5−ジメチルヒダンに関して88%であり、添加した臭素に関して90%であった。1,3−ジブロモ−5,5−ジメチルヒダントインは白色の結晶性固体として得られた。表10にはこの反応で生じた生成物に関する平均粒径のデータおよび生成物の粒度分布データをまとめる。部分5〜10は定常状態の操作条件で生じた生成物の粒径を表している。実施例11と同様に生成物は双峰分布をしている。表10において「n.d.」は大きな粒径部分の粒径分布の決定が行われなかったことを意味する。使用した装置は大きさが2000μよりも大きな粒径を測定することができなかった。生成物の全体的な平均粒径は少なくとも455.5μであった。
ガラスの反応器を用いて他の連続操作を行った。反応器の中に連続的に5,5−ジメチルヒダントインおよびNaOHからつくられた水溶液、およびこれとは別に臭素の供給流を同時に供給する。この水溶液はNaOHの9重量%水溶液に5,5−ジメチルヒダントインを加えてつくった。この溶液は約22.4重量%の5,5−ジメチルヒダントインと7重量%のNaOHを含んでいる。外径が72mmの錨形の撹拌機を備えた内径が82mm、容量が1リットルのジャケット付き反応器を使用し、流体のシリコーン(Rhodersil 4720V20 fluid; Rhone−Poulenc社製)をジャケットの中に循環させた。反応温度を38℃にコントロールした。両方の供給流はポンプでコントロールした。5,5−ジメチルヒダントイン/NaOH溶液の平均供給速度はProminent Gamma G/4A容積形ポンプを介して15.84/分であり、臭素の供給速度はMasterflex Easy−Load容積形ポンプを介して4.67/分であった。平均反応混合物を400rpmで撹拌する。pHメーターを用いて流出液のpHを測定することによって反応のpHを監視した。反応中のpHは6.06〜6.36の範囲であった。やはりポンプを用いて反応器からの生成物の取り出しを制御した。滞在時間は1部分毎に平均30分であった。各部分の容積は約500mLである。反応器に供給した5,5−ジメチルヒダントインに関し1,3−ジブロモ−5,5−ジメチルヒダントインの収率は90.5%であった。1,3−ジブロモ−5,5−ジメチルヒダントインの活性臭素含量は標準的なヨード滴定法で決定して55.3%より大であった。従ってこの生成物の純度は99.0%より高い。
上記の方法で製造された1,3−ジブロモ−5,5−ジメチルヒダントインの5gの試料を、接合剤を用いずに、Hastelloy(R)C合金からつくられたパンチおよびダイス型を備えたSintech(R)プレス(MTS Systems Corporation製、米国ミネソタ州、Edenprairie)の中で圧縮した。ダイス型の中に充填する前に、ダイス型の内面に微粒化したポリプロピレン・ワックス(MICROPRO 400 wax; Micro Powders,Incorporated製、米国ニューヨーク州、Tarrytown)を軽くまぶして潤滑剤の作用をさせた。持続時間なしで34,475kg(5000psi)の圧力をかけた。即ち34,475kg(5000psi)に達したら直ちに自動的に圧力を取り去った。得られた錠剤をダイス型から取り出した後に6日間室温で老化させた。次にMTS Systems Corporationから提供されたTestworksソフトウエアを備えたSintech(R)1/S装置(MTS Systems Corporation製、米国ミネソタ州、Edenprairie)を用いて破砕強度の試験を行った。この装置は、コンピュータに連結された水平の円の形をしたロードセル、やはりコンピュータに連結されたディジタル・マイクロメーター、および該ロードセルの上方に配置されロードセルに対して垂直な下向きの力をかけるスクリューで駆動される垂直方向のピストンを含んでいる。この破砕強度を測定する方法では、マイクロメーターで錠剤の厚さを測定してコンピュータに対するディジタル化された入力をつくる。次に錠剤をロードセルの縁に載せ、錠剤の上方の縁をピストンに接触させる。次にこの装置を作動させ、ピストンが錠剤に対して次第に増加する下向きの直径方向の力をかけるようにする。同時にロードセルによって錠剤にかけた下向きの力を連続的に測定し、この測定値の入力をコンピュータに伝える。かけた力がその直前にかけた力の10%に急激に減少する点に達したら錠剤が破壊された点に達したのであり、ソフトウエア・プログラムによって直ちに力をかけることを止める。コンピュータへの入力から二つの値、即ち錠剤の破壊点における力(ポンド)および破壊点における錠剤の厚さ当たりの力(ポンド)が与えられる。即ちこの力が大きいほど強度は大きい。このような二つの組の試験を行った。第1の組(A組)では実施例12に記載した連続法によってつくられた1,3−ジブロモ−5,5−ジメチルヒダントインのバッチから5個の錠剤を作って評価した。他の組(B組)の試験では実施例8に記載したバッチ法でつくられた1,3−ジブロモ−5,5−ジメチルヒダントインのバッチから3個の錠剤を作った。
接合剤を用いて組成物にした1,3−ジブロモ−5,5−ジメチルヒダントインからつくられた錠剤の破砕強度を、実施例14に記載したような方法で行われた一群の試験において例示する。錠剤の製造方法は次の通りである。実施例13記載の方法で製造された1,3−ジブロモ−5,5−ジメチルヒダントインを、米国ニューヨーク州、TarrytownのMicro Powders Incorporated製の微粒化したポリエチレン・ワックス3重量%と約30分間手で混合した。次に得られた組成物を実施例14記載の方法で錠剤に変えた。実施例14記載の方法で行われた破砕強度を表13にまとめる。
1,3−ジブロモ−5,5−ジメチルヒダントインの試料および市販の1,3−ジハロ−5,5−ジメチルヒダントイン製品を用いて比較流動性試験を行った。これらの試験では236.6cc(8オンス)のジャーの中にその容量の約1/3の所まで試験試料を充填する。ジャーを閉じた後、その側方において一方向にジャーをゆっくりと回転させながら内容物の特性を観察する。表14にこれらの流動性試験の観察結果をまとめる。表14では次のような略号を用いた。

BCDMH: N,N’−ジブロモクロロ−5,5−ジメチルヒダントイン
微粒化したポリエチレン・ワックス2.5g(米国ニューヨーク州、TarrytownのMicro Powders Incorporated製のMPP−611)を秤量して結晶皿の中に入れ、次いで1,3−ジブロモ−5,5−ジメチルヒダントイン(47.5g)を加える。広刃のスパチュラを使用し、料理人がバターを小麦粉の中に練り込むようにしてこの混合物を配合する。この方法で10分間手で混合した後、生成物をガラス瓶の中に入れ、これを転がして混合物の流動性を評価した。配合物をつくるのに使用した1,3−ジブロモ−5,5−ジメチルヒダントインに比べ流動性は改善されていた。
微粒化したポリエチレン・ワックス2.5g(米国ニューヨーク州、TarrytownのMicro Powders Incorporated製のMICROPRO 400)を秤量して結晶皿の中に入れ、次いで1,3−ジブロモ−5,5−ジメチルヒダントイン(47.5g)を加える。この混合物を実施例17記載の方法で配合し、ガラス瓶の中に移し、配合物の流動性を評価した。配合物をつくるのに使用した1,3−ジブロモ−5,5−ジメチルヒダントインに比べ流動性は改善されていた。
実施例17および18でつくられた1,3−ジブロモ−5,5−ジメチルヒダントイン配合物について圧縮試験を行った。各試料を秤量し、Hastelloy(R)C合金からつくられた直径1.80cm(0.71インチ)のダイス型の中に入れ、やはりHastelloy(R)C合金からつくられたスクリュー駆動式パンチを用いて予め定められた圧力まで圧縮する。ダイス型を充填する前に、ダイス型の内面に微粒化したポリプロピレン・ワックスを軽くまぶして潤滑剤の作用をさせた。圧縮圧力に達した後の加圧持続時間はない(直ちに圧力を緩める)。ダイス型から錠剤を抜き出し、マイクロメーターで錠剤の厚さを測定し、錠剤を目で観測した。
実施例18でつくられた1,3−ジブロモ−5,5−ジメチルヒダントイン/5重量%MPP−611の錠剤を水の入ったガラスのビーカーの中に入れる。この錠剤には何事も起こらなかったように思われた。錠剤は数ヶ月に亙りゆっくりと溶解するから、その物理的一体性はそのままの状態に保たれる。溶解したハロゲンを放出することを証明するために、錠剤を水から取り出し、脱イオン水で洗滌し、紙タオルを用いて乾燥した。次にプラスティックスの洗滌瓶を用い、N,N−ジエチルフェニレンジアミン(DPD)粉末を含む脱イオン水溶液の中に錠剤を洗滌して流し込む。洗滌水を導入するとこの溶液は直ちにピンク色に変り、可溶性のハロゲンが錠剤から洗い出されたことを示した。この点に関しDPDはppmレベルで可溶性のハロゲンの存在を検出するのに用いられる高感度の指示薬である。このような量の溶解したハロゲンが存在する場合、DPDはピンクに変わる。
1,3−ジブロモ−5,5−ジメチルヒダントインを微粒化したポリエチレン・ワックス(MPP−611)と、該ワックスを3重量%含むような割合で配合する。この配合物(5g)の試料をHastelloy(R)C合金からつくられたダイス型の中に導入し、34,475kPa(5000psi)の圧力まで圧縮する。さらに3個の試料(それぞれ5g)を同じ方法で圧縮し、それぞれ圧力を緩めた後にダイス型から一つの錠剤を取り出した。いずれの場合もダイス型を充填する前に、微粒化したポリプロピレン・ワックスをダイス型の内面に軽くまぶして潤滑剤の作用をさせた。錠剤を手で壊して二つの同じ大きさの片にした。各錠剤の半分の一つを乳鉢で破砕して粉末にし、この粉末を滴定して活性臭素の重量%を決定する。錠剤の他の半分を密封したガラス瓶の中に入れ、50℃の乾燥器の中に入れる。30日後試料を乾燥器から取り出し、摩砕し、滴定して活性臭素の重量%を決定する。比較の目的で平均粒径が約64.5μの工業的に製造された1,3−ジブロモ−5,5−ジメチルヒダントイン(微粒化したポリエチレン・ワックスを含まない)の対照試料について同じ操作を行う。この対照試料の場合、ダイス型から一つの錠剤を抜き取ることができず、粉砕された積層品についてだけ試験することができた。
実施例21記載の方法で種々の量の微粒化したポリエチレン・ワックスと一緒に錠剤にした1,3−ジブロモ−5,5−ジメチルヒダントインの強度を一連の破砕強度試験で測定した。各試験において、5gの配合した材料をHastelloy(R)C合金からつくられたダイス型に加え、やはりHastelloy(R)C合金からつくられたスクリュー駆動式のパンチで圧力34,475kPa(5000psi)まで圧縮する。いずれの場合もダイス型を充填する前に、微粒化したポリプロピレン・ワックスをダイス型の内面に軽くまぶして潤滑剤の作用をさせた。ダイス型から錠剤を抜き取った後錠剤を目で観測した。
種々の微粒化したワックス(米国ニューヨーク州、TarrytownのMicro Powders Incorporated製)を用いて一連の異なった配合物をつくった。各配合物は実施例17記載の方法で製造され、配合物が3重量%のワックスを含むようにした。これらの配合物をつくるのに使用したDBDMH原料は工業的に製造された粒径が約64.5μの1,3−ジブロモ−5,5−ジメチルヒダントインであった。錠剤の製造および破砕強度試験は実施例21および22記載の方法で行った。
大きな粒径の1,3−ジブロモ−5,5−ジメチルヒダントインと接合剤との組成物の破砕強度を測定した。この1,3−ジブロモ−5,5一ジメチルヒダントインの平均粒径は約189μであり、接合剤は微粒化したポリエチレン・ワックス(MPP−611)であり、配合物中の接合剤の含量は3重量%であった。測定にはTestworksソフトウエアを備えたSintech(R)圧縮機を使用した。これらの試験において錠剤の長手方向の軸に沿ってかける力を破砕が起こるまで次第に増加させた。表20に破砕強度の結果をまとめる。
以下に本発明の主な特徴と態様を列挙する。
1 微生物の抑制、および/または水性媒質の表面と接触した表面上のバイオフィルムを根絶または低減させるために水性媒質を処理する方法において、同じ程度の効果を得るのに必要なN,N'−ブロモクロロ−5,5−ジメチルヒダントインの量よりも少ない有効量の1,3−ジブロモ−5,5−ジメチルヒダントインを処理すべき水性媒質に導入することを特徴とする方法。
2 使用される1,3−ジブロモ−5,5−ジメチルヒダントインは少なくとも約175μの平均粒径をもっていることを特徴とする1記載の方法。
3 使用される1,3−ジブロモ−5,5−ジメチルヒダントインは少なくとも約200μの平均粒径をもっていることを特徴とする1記載の方法。
4 使用される1,3−ジブロモ−5,5−ジメチルヒダントインは少なくとも約300μの平均粒径をもっていることを特徴とする1記載の方法。
5 使用される1,3−ジブロモ−5,5−ジメチルヒダントインは、接合剤を用いることなく、また圧縮能(compactibility)を強化するために固体の予備処理を行うことなく、1,3−ジブロモ−5,5−ジメチルヒダントインの固体粒子を加圧して圧縮することによってつくられた形状保持性をもった加圧圧縮成形品(pressure compacted article)の形をしていることを特徴とする方法。
6 該成形品の形をした1,3−ジブロモ−5,5−ジメチルヒダントインの固体粒子は圧縮する前に少なくとも約175μの平均粒径をもっていることを特徴とする5記載の方法。
7 使用される1,3−ジブロモ−5,5−ジメチルヒダントインは、接合剤を用い、1,3−ジブロモ−5,5−ジメチルヒダントインの固体粒子を加圧して圧縮することによってつくられた形状保持性をもった加圧圧縮成形品の形をしていることを特徴とする方法。
8 該成型品の形をした1,3−ジブロモ−5,5−ジメチルヒダントインの固体粒子は圧縮する前に少なくとも約175μの平均粒径をもっていることを特徴とする7記載の方法。
9 使用される1,3−ジブロモ−5,5−ジメチルヒダントインは、接合剤として該1,3−ジブロモ−5,5−ジメチルヒダントインと相容性をもっている微粒化された合成ポリオレフィンをベースにした炭化水素ワックス、および/または微粒化された合成ポリフルオロ炭素ワックスを圧縮された製品をつくるのに有効な量で使用し、1,3−ジブロモ−5,5−ジメチルヒダントインの固体粒子を加圧して圧縮することによってつくられた形状保持性をもった加圧圧縮成形品の形をしていることを特徴とする方法。
10 該ワックスは圧縮の前に平均粒径が約15μ以下であり、最大粒径は約40μ以下であり、25℃における密度が0.9〜1.4g/ccのポリエチレン・ワックスであることを特徴とする9記載の方法。
11 該ワックスは109〜111℃の温度範囲で熔融するポリエチレン・ワックスであることを特徴とする9記載の方法。
12 該ワックスは圧縮の前における平均粒径が5.0〜7.0μの範囲であり、最大粒径が約22μであり、25℃における密度が0.9〜1.4g/ccのポリエチレン・ワックスであることを特徴とする9記載の方法。
13 該ワックスは140〜143℃の温度範囲で熔融し、圧縮の前における平均粒径が5.0〜7.0μの範囲であり、また圧縮の前における最大粒径が約22μのポリエチレン・ワックスであることを特徴とする9記載の方法。
14 使用される1,3−ジブロモ−5,5−ジメチルヒダントインは粒径が少なくとも175μの1,3−ジブロモ−5,5−ジメチルヒダントインからつくられた圧縮生成物の形をしており、該圧縮生成物は該圧縮製品をつくるのに有効な量の通常は固体の飽和脂肪酸アミドを接合剤として用いてつくられることを特徴とする1記載の方法。
15 使用される1,3−ジブロモ−5,5−ジメチルヒダントインは少なくとも約200μ以下の平均粒径をもっていることを特徴とする14記載の方法。
16 使用される1,3−ジブロモ−5,5−ジメチルヒダントインは少なくとも約300μ以下の平均粒径をもっていることを特徴とする14記載の方法。
17 水性媒質中において少なくとも大腸菌および/またはエンテロコックス・ファエキウムを駆除する方法において、該媒質に殺生物剤的に有効な量の1,3−ジブロモ−5,5−ジメチルヒダントインを導入することを特徴とする方法。
18 該量は同じ程度の効果を得るのに必要なN,N'−ブロモクロロ−5,5−ジメチルヒダントインの量よりも少ないことを特徴とする17記載の方法。
19 水性媒質または水と接触した表面上のバイオフィルムを根絶または少なくとも低減させる方法において、該水性媒質または水に該バイオフィルムを根絶または少なくとも低減させるのに有効な量の1,3−ジブロモ−5,5−ジメチルヒダントインを導入することを特徴とする方法。
20 該量は同じ程度のバイオフィルムの根絶または低減を達成するのに必要なN,N'−ブロモクロロ−5,5−ジメチルヒダントインの量よりも少ないことを特徴とする19記載の方法。
21 水性媒質または水と接触した表面上の緑膿菌のバイオフィルムを根絶または少なくとも低減させる方法において、該水性媒質または水に該バイオフィルムを根絶または少なくとも低減させるのに有効な量の1,3−ジブロモ−5,5−ジメチルヒダントインを導入することを特徴とする方法。
22米国環境保護局の規制に従って水中で使用される微生物抑制剤を調達する(purveying)方法において、該方法は1,3−ジブロモ−5,5−ジメチルヒダントインを含む水質制御剤の容器を調達し、該容器は米国環境保護局によって公布された要求に従った投与量レベルを示すラベルを貼付していることを特徴とする方法。
23 調達される抑制剤は接合剤を用いないでつくられた圧縮生成物の形をしていることを特徴とする22記載の方法。
24 調達される抑制剤は接合剤を用いてつくられた圧縮生成物の形をしていることを特徴とする22記載の方法。
25 圧縮生成物の中に使用される接合剤は、該1,3−ジブロモ−5,5−ジメチルヒダントインと相容性をもっている微粒化された合成ポリオレフィンをベースにした炭化水素ワックス、および/または微粒化された合成ポリフルオロ炭素ワックスであることを特徴とする21記載の方法。
26 該ワックスは圧縮の前において平均粒径が約15μ以下であり、最大粒径が約40μ以下であり、25℃における密度が0.9〜1.4g/ccのポリエチレン・ワックスであることを特徴とする25記載の方法。
27 該ワックスは109〜111℃の温度範囲で熔融するポリエチレン・ワックスであることを特徴とする25記載の方法。
28 該ワックスは圧縮の前における平均粒径が5.0〜7.0μの範囲であり、最大粒径は約22μであり、25℃における密度が0.9〜1.4g/ccのポリエチレン・ワックスであることを特徴とする25記載の方法。
29 該ワックスは140〜143℃の温度範囲で熔融し、圧縮の前において平均粒径が5.0〜7.0μの範囲であり、また圧縮の前において最大粒径が約22μであるポリエチレン・ワックスであることを特徴とする25記載の方法。
30 調達される該抑制剤は、平均粒径が少なくとも175μの1,3−ジブロモ−5,5−ジメチルヒダントイン固体粒子からつくられた圧縮生成物の形をしており、該圧縮生成物は該圧縮生成物をつくるのに有効な量の通常は固体の飽和脂肪酸アミドを接合剤として用いてつくられることを特徴とする22記載の方法。
31 該圧縮生成物をつくるのに使用される1,3−ジブロモ−5,5−ジメチルヒダントインは圧縮する前において少なくとも約200μの平均粒径をもっていることを特徴とする30記載の方法。
32 該生成物が調達される水は再生水(recreation water)であることを特徴とする22記載の方法。
33 該生成物が調達される水は工業用冷却水であることを特徴とする22記載の方法。
34 該生成物が調達される水は廃水または処理水、或いは両方であることを特徴とする22記載の方法。
35 該生成物を調達する使用目的は水に接触した表面上のバイオフィルムの根絶または低減であることを特徴とする22記載の方法。
36 該生成物を調達する使用目的は少なくとも大腸菌またはエンテロコックス・ファエキウムの抑制、または水に接触した表面上の少なくとも緑膿菌のバイオフィルムの根絶または低減であることを特徴とする22記載の方法。
37 表面上のバイオフィルムを根絶または低減させる方法において、該バイオフィルムを根絶または少なくとも低減させるのに有効な量の1,3−ジブロモ−5,5−ジメチルヒダントイン導入した水性媒質または水と接触させ、その接触時間を該バイオフィルムを根絶または少なくとも低減させるのに十分な長さの期間またはそれよりも長い期間にすることを特徴とする方法。
38 該量は同じ程度のバイオフィルムの根絶または低減を達成するのに必要なN,N'−ブロモクロロ−5,5−ジメチルヒダントインの量よりも少ないことを特徴とする37記載の方法。
39 表面上の少なくとも緑膿菌のバイオフィルムを根絶または低減する方法において、該バイオフィルムを根絶または少なくとも低減させるのに有効な量の1,3−ジブロモ−5,5−ジメチルヒダントイン導入した水性媒質または水と接触させ、その接触時間を該バイオフィルムを根絶または少なくとも低減させるのに十分な長さの期間またはそれよりも長い期間にすることを特徴とする方法。
40 水性媒質または水と接触した表面上のバイオフィルムを根絶または少なくとも低減させ、同時に該水性媒質または水の中に存在する微生物種に対して微生物の抑制を行うのに有効な量で水性媒質または水の中に1,3−ジブロモ−5,5−ジメチルヒダントインを連続的にまたは周期的に放出することを特徴とする方法。
41 水性媒質または水の処理は、水性媒質または水の側流の中に1,3−ジブロモ−5,5−ジメチルヒダントインを放出し、処理によって得られた側流を水性媒質または水の全体の中に流し込むことを特徴とする40記載の方法。
42 水性媒質または水の処理は、浮き装置から1,3−ジブロモ−5,5−ジメチルヒダントインを放出させ、これによって殺生物剤的に有効な量の1,3−ジブロモ−5,5−ジメチルヒダントインを水中に放出する方法であることを特徴とする40記載の方法。
43 処理すべき水は再生水、工業冷却水、廃水または処理水であることを特徴とする40記載の方法。
Claims (6)
- 微生物の抑制、および/または水性媒質と接触した表面上のバイオフィルムを根絶または低減させるために水性媒質を処理する方法であって、そのような媒質に1,3−ジブロモ−5,5−ジメチルヒダントインを単独でBr2として0.2から10mg/Lのハロゲン残留量を達成するように導入することを含んでなり、そのようなハロゲン残留量はN,N'−ブロモクロロ−5,5−ジメチルヒダントイン単独で同レベルの微生物的な効果を達成するのに必要とされるそれよりも少なく、ここで該微生物的な効果は、AOACの公式方法:水泳用プールの消毒、によって測定され、
用いられる1,3−ジブロモ−5,5−ジメチルヒダントインが;
a)非圧縮の形をしており且つ少なくとも200μの平均粒径を有するか、または
b)いかなる結合剤も有さない形状保持加圧圧縮成型品の形をしており且つ圧縮する前に少なくとも200μの平均粒径を有する圧縮された1,3-ジブロモ−5,5−ジメチルヒダントイン固体から本質的に成るか、または
c)1,3−ジブロモ−5,5−ジメチルヒダントイン粒子固体、および圧縮成型品を形成するのに有効である結合剤量の合成ポリオレフィンをベースにした炭化水素ワックスおよび/または微粒化された合成ポリフルオロ炭素ワックスであって1,3−ジブロモ−5,5−ジメチルヒダントインと相容性のある該ワックス、から本質的に成る形状保持加圧圧縮成型品の形をしているか、または、
d)圧縮する前に少なくとも200μの平均粒径を有する圧縮された1,3−ジブロモ−5,5−ジメチルヒダントイン粒子固体、および圧縮成型品を形成するのに有効である結合剤量の飽和且つ通常は固体である脂肪酸アミド、から本質的に成る形状保持加圧圧縮成型品の形をしている、
上記方法。
- a)およびd)の1,3−ジブロモ−5,5−ジメチルヒダントインの平均粒径が少なくとも300μである、請求項1記載の方法。
- 水性媒質が、有効量の1,3−ジブロモ−5,5−ジメチルヒダントインを導入する前に、少なくとも大腸菌、エンテロコックス・ファエキウムまたはシュードモナス・アエルギノサを含有する、請求項1記載の方法。
- 水性媒質または水の側流の中に1,3−ジブロモ−5,5−ジメチルヒダントインを連続的にまたは周期的に放出し、これにより処理によって得られた側流を水性媒質または水の全体の中に流し込む、請求項1記載の方法。
- 浮き装置から1,3−ジブロモ−5,5−ジメチルヒダントインを放出させ、これにより殺生物剤的に有効な量の1,3−ジブロモ−5,5−ジメチルヒダントインが水性媒質または水に送達される、請求項1記載の方法。
- 処理すべき水が再生水、工業冷却水、廃水または処理水である、請求項4又は5記載の方法。
