本発明の文脈において「オリゴマー化合物」という用語は、少なくとも核酸標的物の領域にハイブリダイズすることができるポリマー構造を指す。一実施形態において、核酸標的物はmiRNAである。「オリゴマー化合物」という用語は、オリゴヌクレオチド、オリゴヌクレオシド、オリゴヌクレオチド類似体、オリゴヌクレオチド摸倣体、及びそれらの組み合わせを含む化合物を含むが、これらに限定されない。オリゴマー化合物はまたアンチセンスオリゴマー化合物、アンチセンスオリゴヌクレオチド、siRNA、選択的スプライサー、プライマー、プローブ、及び標的核酸の少なくとも一部分をハイブリダイズする他の化合物を含むが、これらに限定されない。オリゴマー化合物又はオリゴヌクレオチドは、5'から3'方向に読み込まれたその核酸塩基配列が標的核酸の対応する領域の逆相補体を含む場合に、アンチセンスである。
本発明の文脈において「ハイブリダイゼーション」はオリゴマー化合物の相補鎖の対形成を意味する。本発明において、対形成のメカニズムは、オリゴマー化合物の鎖の相補的なヌクレオシド又はヌクレオチド基(核酸塩基)の間の、ワトソン−クリック、フーグスティーン、又は逆フーグスティーン水素結合である水素結合を含む。例えば、アデニンとチミンは、水素結合の形成を通して対形成する相補的な核酸塩基である。ハイブリダイゼーションは様々な状況下で生じる。

オリゴマー化合物の具体的例は、例えば、非天然ヌクレオシド間結合などの修飾を有するオリゴヌクレオチドを含む。そのような非天然ヌクレオシド間の結合は、例えば、増強した細胞の取り込み、他のオリゴヌクレオチドまたは核酸標的物に対する増強した親和性、及びヌクレアーゼ存在下での増強した安定性のような望ましい特性のために、しばしば天然形態より多く選択される。
本発明のオリゴマー化合物もまた、1もしくはそれ以上の修飾されたまたは置換された糖鎖を含むことが可能である。塩基部分は適切な核酸標的化合物とのハイブリダイゼーションのために保持されている。糖修飾はオリゴマー化合物に対して、ヌクレアーゼ安定性、結合親和性、他の有益な生物学的特性を与えることができる。代表的な修飾された糖は炭素環又は非環式糖、2'、3'、または4'位の1もしくはそれ以上位置において置換基を有する糖、糖の1もしくはそれ以上の水素基の位置に置換物を有する糖、糖のいずれの2つの他の原子の結合間の結合を有する糖を含む。多くの糖修飾は従来技術で周知であり、2'位置に修飾された糖及び糖の任意の2原子の間に架橋を有する糖(糖が2環式となるように)は特に本発明で有用である。本発明で有用な糖の修飾の例は、これらに限定されないが、OH、F、O−アルキル、S−アルキル、またはN−アルキル、或いはO−アルキル−O−アルキルから選択される糖置換基を含む化合物を含み、前記アルキル、アルケニル、及びアルキニルは置換または未置換C1〜C10アルキルまたはC2〜C10アルケニル及びアルキニルであっても良い。特に適切であるのは、2−メトキシエトキシ(2'−O−メトキシエチル、2'−MOE、又は2'−OCH2CH2OCH3としても知られている)、2'−O−メチル(2'−O−CH3)、2'−フルオロ(2'−F)、又は4'炭素原子から2'炭素原子に連結する架橋基を有する2環式糖修飾ヌクレオシドであり、架橋基の例は−CH2−O−、−(CH2)2−O−、又は−CH2−N(R3)−O−を含み、R3はH又はC1〜C12アルキルである。

本発明のオリゴマー化合物に付与される置換基の1つは、得られるオリゴマー化合物の活性、細胞内分布、又は細胞内取り込みを増強する1もしくはそれ以上の部分又は抱合の結合を含む。一実施形態において、そのような修飾オリゴマー化合物は、ヒドロキシル基又はアミノ基のような官能基に、共有結合的に抱合基が結合することによって調製される。本発明の抱合基は、オリゴマーの薬力学的特性を増強し、薬物動態学的特性を増強する挿入物、レポーター分子、ポリアミン、ポリアミド、ポリエチレングリコール、ポリエーテル、オリゴマー基、及びオリゴマー基を含む。典型的な抱合基はコレステロール、炭水化物、脂質、リン脂質、ビオチン、フェナジン、葉酸、フェナントリジン、アントラキノン,アクリジン、フルオレセイン、ローダミン、クマリン及び色素を含む。本発明の文脈において、薬力学的特性を増強する基はオリゴマー取り込みを改善し、分解に対するオリゴマー抵抗性を増強し、及び/又はRNAとのハイブリダイゼーションを強化する基を含む。本発明の文脈において、薬物動態学的な特長を増強する基はオリゴマー取り込み、分配、代謝、又は排出を改善する基を含む。代表的な抱合基は、1992年10月23日付出願の国際特許出願第PCT/US92/09196号で開示されている。その全体の開示はこの参照により本願明細書に組み込まれる。抱合部分はコレステロール部分及び従来技術で周知の様々な他の脂質部分を含むが、これらに限定されない。
オリゴマー化合物はその長さに沿って特定の方向性で配置された化学修飾サブユニットを有することが可能である。「化学的なモチーフ」はオリゴマー化合物全体の化学修飾の配置として定義される。
マイクロRNA等を含むスモールノンコーディングRNAに対する有効なモジュレーターを同定するスクリーニング方法もまた、本発明に包含され、それらの方法は、スモールノンコーディングRNA又はその一部を、1若しくはそれ以上のモジュレーター候補物質と接触させる工程と、スモールノンコーディングRNAの強度、発現量を減少または増加させる、又はその機能を変化させる1若しくはそれ以上の候補物質を選択する工程と、を有するものである。本願明細書に記すように、モジュレーター候補物質はマイクロRNA、またはその任意の一部を標的とするオリゴマー化合物である可能性がある。モジュレーターまたはその候補物質が、スモールノンコーディングRNAの強度、発現量を調節すること(例えば減少または増加させるなどの形で)、またはその機能を変化させることが一旦示されれば、そのモジュレーターは、本発明に従って、さらなる研究用途に使用するほか、標的のバリデーション、試験研究、検査または治療での試薬、薬剤として用いることができる。一実施形態において、特定のマイクロRNAの機能を調節する能力に対して、モジュレーター候補物質をスクリーニングする。
オリゴマー化合物およびホスホアミダイトは、当業者にとって周知の方法で合成される。修飾型または天然型のヌクレオシドの低重合化は、DNA様化合物の合成方法に関する文献(Protocols for Oligonucleotides and Analogs,Ed.Agrawal(1993),Humana Press)、および/またはRNA様化合物の合成方法に関する文献(Scaringe,Methods(2001),23,206〜217.Gait et al,Applications of Chemically synthesized RNA in RNA:Protein Interactions,Ed.Smith(1998),1〜36.Gallo et al.,Tetrahedron(2001),57,5707〜5713)に従って適切に行われる。あるいは、例えばDharmacon Research Inc.(コロラド州Lafayette)などの様々なオリゴヌクレオチド合成会社からオリゴマーを購入することもできる。
スクリーニングおよび標的のバリデーションの研究において、本発明のオリゴマー化合物は、そのそれぞれに対応する相補鎖のオリゴマー化合物と同時に用いて、安定した二本鎖(二重鎖)のオリゴヌクレオチドを形成させて使用することができる。本発明に従い、本発明のオリゴマー化合物およびその相補鎖を含む一連の二重鎖が、スモールノンコーディングRNAを標的とするように設計することができる。前記二重鎖の末端は、1若しくはそれ以上の天然型、または修飾型核酸塩基の付加によって、オーバーハングを形成するように修飾することができる。そうすると、二重鎖RNAのセンス鎖はアンチセンス鎖に対して相補的に設計および合成され、いずれの末端にも修飾または付加を持つようにすることもまたできる。例えばいくつかの実施形態では上述のように、二重鎖のうちの両方の鎖は中央領域の核酸塩基が相補的であり、それぞれの鎖が一方または両方の末端にオーバーハングを有するようになるであろう。
本発明のオリゴマー化合物および組成物はさらに、試験研究、薬剤探索、キットおよび診断法、および治療法にも用いることができる。
本発明はまた、オリゴマー化合物、スモールノンコーディングRNA、および本発明の組成物を含む製剤設計および医薬品組成物を含む。医薬品組成物を設計するための方法および組成物は、例えば、これに限定されるものではないが、投与経路、病気の程度、または投与量を含む数々の基準に依存する。当業者は、そういった検討事項を良く理解している。
標的の核酸の発現または機能に対するオリゴマー化合物の効果は、標的の核酸が検出可能な強度で存在しさえすれば、いかなるタイプの細胞を用いて測定しても良い。使用する細胞の決定は、例えば、ノザンブロット法、リボヌクレアーゼプロテクションアッセイ法、またはリアルタイムPCR法など、当該技術分野で通常に使用されている方法によって容易に行うことができる。このような分析に用いられる様々な細胞は、民間の供給メーカー(例えば、American Type Culture Collection,ヴァージニア州Manassus;Zen−Bio,Inc,ノースカロライナ州Research Triangle Park;Clonetics Corporation,メリーランド州Walkersville)から入手可能であり、市販の試薬(例えば、Invitrogen Life Technologies,カリフォルニア州Carlsbad)を用いて供給メーカーの指示に従って細胞を培養した。実例となる細胞の種類としては、これに限定されるものではないが:T−24細胞、A549細胞、正常なヒト乳腺上皮細胞(normal human mammary epithelial cells:HMECs)、MCF7細胞、T47D細胞、BJ細胞、B16−F10細胞、正常ヒト臍帯静脈内皮細胞(HUVECs)、正常ヒト皮膚繊維芽細胞(human neonatal dermal fibroblast:NHDF)、ヒト表皮角化細胞(Hhuman embryonic keratinocytes:HEK)、293T細胞、HepG2、ヒト前駆脂肪細胞、分化したヒト脂肪細胞(当該技術分野において既知の方法に従ってヒト前駆脂肪細胞を分化させたもの)、NT2細胞(NTERA−2 cl.D1としても知られる)、およびHela細胞が挙げられる。
本発明のオリゴマー化合物による細胞の処理は、細胞密度が約80%に達した時点で行われる。オリゴマー化合物の細胞への導入には、カチオン性脂質トランスフェクション試薬LIPOFECTIN(登録商標)(Invitrogen,カリフォルニア州Carlsbad)を用いる。オリゴマー化合物とLIPOFECTIN(登録商標)をOPTI−MEM(登録商標)1(Invitrogen,カリフォルニア州Carlsbad)中で混合し、オリゴマー化合物とLIPOFECTIN(登録商標)とを望みの最終濃度に到達させる。細胞に添加する前に、オリゴマー化合物、LIPOFECTIN(登録商標)、およびOPTI−MEM(登録商標)1を完全に混合させ、約0.5時間インキュベートした。細胞培地をプレートから除去し、滅菌したガーゼ上でプレートを軽くたたく。96穴プレートの各ウェルを、それぞれ150μLのリン酸緩衝生理食塩水またはハンクス液で洗浄する。24穴プレートの各ウェルを、それぞれ250μLのリン酸緩衝生理食塩水またはハンクス液で洗浄する。各ウェル中の洗浄緩衝液を、96穴プレートまたは24穴プレートの場合、それぞれ100μL、250μLのオリゴマー化合物/OPTI−MEM(登録商標)/LIPOFECTIN(登録商標)の混合液と置換する。未処理のコントロール細胞には、LIPOEFCTIN(登録商標)のみを与える。プレートは37℃で4〜7時間ほどインキュベートした後、細胞培地を除去し、滅菌したガーゼ上でプレートを軽くたたく。96穴プレートまたは24穴プレートの各ウェルに、それぞれ100μL、1mLの成長培地をそのまま添加する。オリゴヌクレオチド処理から16〜24時間後に細胞を採取し、RNAを単離して標的量の減少をリアルタイムPCRで計測するか、もしくは他の表現型のアッセイを行う。処理した細胞のデータ測定は3回繰り返して行い、3回の試行の平均値を結果とする。
標的の強度または発現の調節は、当該技術分野において既知である様々な方法で分析できる。例えば、標的の核酸の強度を定量化する方法としては、ノザンブロット法、競合ポリメラーゼ連鎖反応(PCR)、または定量的リアルタイムPCR法などがある。RNAの分析は、細胞のトータルRNA、またはpoly(A)+mRNAで行うことができる。RNA単離の方法は当該技術分野において周知である。ノザンブロット法もまた、当該技術分野において通常に用いられている。定量的リアルタイムPCR法は、市販のABI PRISM(登録商標)7600、7700、または7900配列検出システム(PE−Applied Biosystems,カリフォルニア州Foster Cityより入手可能)を、メーカーの指示に従って用いることで簡便に行うことができる。
Hela、T−24、およびA549などの細胞株からのRNA試料の調製は、当該技術分野において既知の方法、例えば、TRIZOL(登録商標)(Invitrogen,カリフォルニア州Carlsbad)を用いてメーカーの推奨するプロトコルに従って行う。簡単に説明すると、細胞の単層を冷却PBSで2回洗浄した後、培養皿の表面積10平方センチメートル当たり1mLのTRIZOL(登録商標)(Invitrogen,カリフォルニア州Carlsbad)を用いて細胞を溶解し、TRIZOL(登録商標)のプロトコルに従ってトータルRNAを調製する。
標的RNA強度の計量は、ABI PRISM(登録商標)7600,7700、または7900配列検出システム(PE−Applied Biosystems,カリフォルニア州Foster City)を用いた定量的リアルタイムPCR法を利用し、メーカーの指示に従って行う。定量的リアルタイムPCRの方法は当該技術分野において周知のものである。
ノザンブロット法は当該技術分野においてルーティン的な手順に従って行われる。15〜20μgのトータルRNAを、TBEバッファシステム(Invitrogen)を用いた10%アクリルアミドゲル(含尿素)電気泳動で分画する。RNAは、Xcell SURELOCK(商標)ミニセル電気泳動槽(Invitrogen,カリフォルニア州Carlsbad)中で、ゲルからHYBOND(商標)−N+ナイロン膜(Amersham Pharmacia Biotech,ニュージャージー州Picataway)へエレクトロブロットする。メンブレンはSTRATALINKER(登録商標)UVクロスリンカー2400(Stratagene,Inc,カリフォルニア州LaJolla)を用いたUV架橋を施し、その後RAPID−HYB(商標)バッファ溶液(Amersham)を用いて、メーカーがオリゴヌクレオチドプローブ向けに推奨している方法でプローブによる検出を行う。
スモールノンコーディングRNAによって調節または制御されている下流の標的タンパク質量の評価または定量化は、当該技術分野において周知の様々な方法によって行うことができる。そのようなものとしては、免疫沈降法、ウェスタンブロット法(イミュノブロッティング)、ELISA法(enzyme−linked immunosorbent assay:ELISA)、定量的タンパク質解析、タンパク質活性解析(例えば、カスパーゼ活性アッセイなど)、免疫組織化学法、免疫細胞化学、またはFACS法(fluorescence−activated cell sorting:FACS)が挙げられる。標的に対する抗体は様々な方法で同定、および入手できる。例えば、MSRS抗体カタログ(Aerie Corporation,ミシガン州Birmingham)、または当該技術分野において周知であるモノクローナル、またはポリクローナル抗体の標準的な作製方法によることもできる。
ひとたびここに開示される方法によってモジュレーターが設計、または同定できたならば、そのオリゴマー化合物は1若しくはそれ以上のさらなる表現型解析に供することができ、その解析はそれぞれが特定の病状または症状に関連する生理条件の治療、改善、または向上においての有効性を予想させる、または示唆する重要な結果を有するであろう。
マイクロRNAsを標的とするオリゴマー化合物の活性は、DUAL−LUCIFERASE(登録商標)レポーターアッセイ(Promega,ウィスコンシン州Madison)で、ルシフェラーゼ活性が正常なマイクロRNA活性(すなわち、相補的な配列への結合)によって阻害されることを利用してインビトロで評価することができる。オリゴマー化合物は、標的とするマイクロRNAがルシフェラーゼレポーター中の相補的な配列へ結合することを阻害し、それによってルシフェラーゼの活性が促進される。ルシフェラーゼレポーターは、目的のマイクロRNAの配列を用いて作製できる。
実験動物モデルは、マイクロRNAsを標的とするオリゴマー化合物の効果、効能、および治療指数の評価に用いられる。
(実施例)
マイクロRNAを標的とし、均一に修飾されたオリゴマー化合物が、ルシフェラーゼレポーターアッセイにおいて、マイクロRNAの活性を調節する能力を検定した。均一に修飾された化合物は、糖による均一な修飾、ヌクレオシド間の結合部の均一な修飾、またはそれらの組合せを含む。培養細胞のオリゴマー化合物による処理に続いて、ルシフェラーゼ活性が測定される。ここで、ルシフェラーゼ活性の上昇は、オリゴマー化合物がマイクロRNAの活性を阻害していることを示す。
miR−21(TAGCTTATCAGACTGATGTTGA;配列ID番号:113)に相補的な22塩基のヌクレオチドを、pGL3−Control(Promega)の3'−UTRへ挿入することで、miR−21ルシフェラーゼセンサーコンストラクトを作製した。ここで記述している方法で、DUAL−LUCIFERASE(登録商標)レポーターアッセイ(Promega,ウィスコンシン州Madison)を実行した。
キメラのオリゴマー化合物とは、化学的に修飾された領域を2若しくはそれ以上含むオリゴマー化合物である。キメラのオリゴマー化合物の一つの例は「ギャップマー」である。ギャップマーでは、オリゴマー化合物は中央領域と、それの両側に隣接する二つの「ウィング」と名付けられた領域を含むモチーフを有する。一態様では、中央領域のヌクレオチドは一種類の糖修飾を含み、一方でウィング領域のヌクレオチドは違う種類の糖修飾を含む。通常、2つのウィング領域の塩基数は同じであるが、しかし、ギャップマーについては、その条件は必ずしも必須ではない。
C57BL/6の雄マウスを民間業者から入手した。マウスは以下の治療群に分類された:ISIS 327895で処理;ISIS 393206で処理;および生理食塩水で処理されたもの、である。それぞれのオリゴマー化合物は、5'―ACAAACACCATTGTCACACTCCA―3'(配列ID番号:19)という塩基配列を有する。ISIS 327895は、均一な2'−MOE糖修飾、およびヌクレオシド間に均一にホスホロチオエート結合を含む。生理食塩水で処理されたマウスがコントロールとされた。25mg/kgの用量のオリゴマー化合物を週二回、三週間にわたってマウスへ腹腔内投与した。処理の終了段階において、マウスは健康で正常な様子であり、血漿中のASTおよびALT量も正常の範囲内であった。
均一に修飾された、またはキメラのオリゴマー化合物による処理後のmiR−122阻害活性の作用発現を比較するために、ISIS 327895およびISIS 393206を25mg/kgの用量で週二回、五週間までマウスへ投与した。そして、1,2,3,4,5,6,8、および10回目の投与のそれぞれ24時間後に、各治療グループから4匹ずつのマウスを検体とした。それぞれの時点でALDOA mRNA量および血漿中コレステロールを測定したところ、2'−MOE/2'−F/2'−MOEオリゴマー化合物が、より強いanti−miR−122活性、すなわちALDOA mRNA量の上昇および血漿中コレステロール量の減少をもたらすことが示唆された。さらに、2'−MOE/2'−F/2'−MOEオリゴマー化合物の方が、均一な2'−MOEオリゴマー化合物と比較して、より早い段階でALDOAの上昇および血漿中コレステロール量の減少が確認された。従って、2'−MOE/2'−F/2'−MOEオリゴマー化合物は、均一な2'−MOEオリゴマー化合物と比較して、より強い効果およびより早い作用発現を示すことが明らかとなった。
anti−miRNAオリゴマー化合物の用量依存性を評価するために、均一な2'−MOEおよび2'−MOE/2'−F/2'−MOEオリゴマー化合物を、6.25,12.5,25、または50mg/kgの用量で週二回、三週間までマウスへ投与した。2'−MOE/2'−F/2'−MOEオリゴマー化合物を6.25,12.5,25、および50mg/kgの用量で投与した結果、生理食塩水を投与した動物での測定値と比較してALDOA mRNAはそれぞれ約4,5,4.25、および5倍増加した。均一な2'−MOEオリゴマー化合物の同量を投与した結果、生理食塩水を投与した動物での測定値と比較してALDOA mRNAはそれぞれ約0.25,1.5,2および3倍増加した。血漿中コレステロール量もまた同様に、用量依存的に減少した。すなわち、2'−MOE/2'−F/2'−MOEオリゴマー化合物を6.25,12.5,25、および50mg/kgの用量で投与すると、生理食塩水を投与した動物での測定値と比較して血漿中コレステロール量はそれぞれ約40%、45%、45%、および48%減少した。2'−MOEオリゴマー化合物では、低いほうから三番目の用量までは血漿中コレステロール量を約5%減少させる結果となり、最高の用量では血漿中コレステロール量を少なくとも40%減少させた。従って、2'−MOE/2'−F/2'−MOEオリゴマー化合物は2'−MOEオリゴマー化合物と比較して、著しく優れた効果および効能を示すことが明らかとなった。
マイクロRNAsを標的とし、位置ごとに修飾されたモチーフを有するオリゴマー化合物が、インビボでマイクロRNA活性を阻害する能力を検定した。
マイクロRNAを標的とするオリゴマー化合物への2'−MOE/2'−F/2'−MOEモチーフの組み込みが、インビボでの高い効果および効能をもたらしたことから、このモチーフは様々な用途の中でも治療用途のためにanti−miRオリゴマー化合物へ組み込むことが望ましい。このモチーフの治療指数をさらに改善するために、キメラのモチーフの中央領域へ、2'−F以外の2'−修飾を有するように、位置ごとに修飾されたオリゴマー化合物を設計した。後述するように検定を行ったオリゴマー化合物の集合の一部を表Aに示した。付加的に検定された化合物はISIS 400129であり、これは以下のようにヌクレオシド間のホスホロチオエート結合によって結合された修飾ヌクレオシドを有するものである:以下のように配置されたものである:二つの2'−MOE、三つの2'−F、一つの2'−MOE、五つの2'−F、一つの2'−MOE、五つの2'−F、一つの2'−MOE、三つの2'−F、二つの2'−MOEである。
内部領域に少なくとも16箇所の2'−F修飾ヌクレオチドを有する、位置ごとに修飾されたオリゴマー化合物を用いて、インビボでの実験を行った。オリゴマー化合物は配列ID番号:19の配列を含む。検定されたオリゴマー化合物は、ISIS 393206,ISIS 400124,400125,400126,400127,400128,400129,および400130を含む。オリゴマー化合物を、25mg/kgの用量で週2回、3週間に渡ってマウスに腹腔内投与した。ISIS 393206も同様に投与した。オリゴマー化合物で処理したマウスの肝臓でのALDOA mRNA量を定量的PCRで測定し、生理食塩水で処理したマウスの肝臓での測定値と比較した。ISIS 393206では、ALDOA mRNAが約4倍に上昇し、コレステロールは約50%減少した。ISIS 400124,400125,400126,400127,400128,400129,および400130オリゴマー化合物のそれぞれによる処理では、生理食塩水処理したマウスの肝臓での測定値の少なくとも2倍以上にALDOA mRNAが上昇する結果となった。血漿中総コレステロールの減少は20%から40%の間であった。特に、ISIS 400124,400126,および400127は、ISIS 393206と同等の効率でALDOA mRNA量を上昇させることができ、生理食塩水処理した場合の肝臓ALDOA量と比較して約4倍となった。さらには、ISIS 393206処理では脾臓重量の増加が見られたのに対して、これらのオリゴマー化合物では脾臓重量の著しい増加は見られなかった。ISIS 400125,400129,および400128では、ALDOA mRNA量がそれぞれ約3倍、2.5倍、および2倍へ上昇し、脾臓重量の増加はなかった。一方、オリゴマー化合物の内部領域に2'−Fを含むISIS 400130では、ALDOA mRNA量が約3倍に増加したものの、ISIS 393206処理したマウスで見られたのと同程度の脾臓重量の増加ももたらす結果となった。
miR−122を標的とするオリゴマー化合物を一回だけ投与した場合の効果を評価するための実験を行った。オリゴマー化合物は以下のモチーフを含む:均一に修飾した2'−MOE;2'−MOE/2'−F/2'−MOE;および内部領域に少なくとも16箇所の2'−Fを有する位置ごとに修飾されたもの、である。マウス(Balb/c 雌、治療グループにつきn=4)の腹腔内へ、11,33、または100mg/kgの用量を一回だけ投与し、4日後に検体とした。共にmiR−122のアンチセンスによる阻害によって上昇することが知られているALDOA mRNA量およびGYS1 mRNA量を肝臓で測定し、生理食塩水処理したマウスの肝臓中でのそれぞれのmRNA量と比較した。表Bはこの実験のデータを要約したものである;ここでALDOAおよびGYS1 mRNA量は、生理食塩水処理のコントロールに対するパーセンテージで示した;CHOLはコレステロールの基準値(実験開始時の値)に対するパーセンテージである;SDは標準偏差である。

- 以下の化学式Iを有する結合したヌクレオシドの連続配列を含むオリゴマー化合物:
