Abstract
Description
本願は、2016年6月21日に出願された米国特許仮出願第62/365,126号の恩典を主張する。参照する出願の全内容は本明細書において参照により組み入れられる。
本発明は全体として、バイオテクノロジーの分野に関する。より具体的には、本発明は、ゲノムDNAを改変するための新規の方法および組成物に関する。
標的ゲノム工学は、細胞内のDNAに沿った特定の部位において定方向の様式で内因性DNAを編集または変更することを含む。遺伝子修復および相同性定方向遺伝子改変の非常に大きな可能性にもかかわらず、現在のゲノム工学的アプローチでは、修復または編集の非常に低い効率が提供され、有害なまたは望ましくないDNA配列および結果を導入する可能性を有する。
(図1)Maxcyte STXの静的およびフローエレクトロポレーショントランスフェクション技術を用いた安定な細胞株の開発プロセス。図は、安定な細胞生成のワークフローを示す。エレクトロポレーション後、選択無しで細胞をある期間培養して、エレクトロポレーション法からの回復を可能にしてもよい(図中に示していない)。エレクトロポレーション後、選択物質の存在下で細胞を培養することにより選択する(選択段階)。選択段階の後、選択物質の存在下で細胞を低密度で培養して、限界希釈クローニングを可能にする(維持/クローン選択段階)。クローン集団の生成後、クローンを、外因性ポリペプチドの発現についてスクリーニングして、増殖させる(クローンのスクリーニングおよび増殖の段階)。スクリーニング後、所望の活性を有するクローンを、産生の目的のために大規模で成長させ(大規模スケールアップ段階)、凍結保存などの長期保存に供する。
(図2)図2A〜C:DNAトランスフェクションは、細胞に対する分化細胞毒性を有する。図2に、DNAおよびmRNAをトランスフェクトされた末梢血液リンパ球(PBL)およびK562細胞の生存率(図2A)、DNAおよびmRNAをトランスフェクトされたPBLおよびK562細胞のGFP発現(図2B)、ならびにDNAおよびmRNAをトランスフェクトされたPBLおよびK562細胞の細胞数(図2C)を示す。データは、DNAトランスフェクションによって、K562への細胞毒性は生じないが、休止PBLにおいて強い細胞毒性を誘導することを実証している。

(図6)K562のAAVS1部位におけるmRNA-CRISPRトランスフェクション誘導性Hind III配列組み込みに依存する一本鎖DNAオリゴサイズ。細胞は、トランスフェクトされなかった(-EP)か、mRNA-CRISPR単独(c+g)をトランスフェクトされたか、または、示すとおりの異なるサイズのmRNA-CRISPR+一本鎖DNAオリゴをトランスフェクトされた。試料は、マーカー、c+g、c+g+26mer、c+g+50mer、c+g+70mer、およびc+g+100merの順序でロードした。6個のヌクレオチドを認識するHindIIIが、HindIII消化部位を作製するAAVS1部位中にあった。組み込まれたオリゴドナー配列を伴うAAVS-1部位の切断生成物は、298および170塩基対であり、親バンドは468塩基対である。組み込み率は、(消化バンドの密度)/(消化バンドの密度+親バンドの密度)として算出した。核酸50、70、および100個のドナーオリゴは、それぞれ43%、35%、および34%の組み込みを示し、一方で、核酸20個は0%組み込みを示した。
(図7)mRNA-CRISPRオリゴトランスフェクションによって、増殖したT細胞のAAVS1部位におけるHind III配列の組み込みが誘導された。細胞は、mRNA-CRISPR単独またはmRNA-CRISPR+50mer一本鎖オリゴ(c+g+o)のいずれかをトランスフェクトされた。PCRアンプリコンは、HidIIIを用いて消化された(+H3)か、または消化されなかった(-H3)。試料を、以下のとおりにロードした:(1)マーカー;(2)c+g-H3;(3)c+g+H3;(4)c+g+o-H3;(5)c+g+o+H3。ドナーオリゴによって、HindIII消化部位を作製したAAVS1部位中に6個のヌクレオチドを組み込んだ。組み込んだオリゴドナー配列を伴うAAVS-1部位の切断生成物は、298および170塩基対であり、親バンドは468塩基対である。組み込み率は、(消化バンドの密度)/(消化バンドの密度+親バンドの密度)として算出した。ドナーオリゴをトランスフェクトされた増殖したT細胞は、15〜30%の組み込みを示した。
(図8)図8A〜C: MaxCyteシステムによるmRNAのトランスフェクションは、ヒトの増殖したT細胞に対して低細胞毒性を有する。図7と同じ増殖したt細胞についての生存率および細胞増殖(図8A)、トランスフェクション後の増殖したT細胞の増殖(図8B)、ならびにトランスフェクション後の増殖したT細胞のGFP発現(図8C)。データは、一本鎖オリゴDNAを伴うmRNAと同様にヌクレアーゼが、6個のヌクレオチドの組み込みを媒介するだけでなく(図7)、増殖したT細胞に対して低い細胞毒性を示したことを実証している。

B.オリゴ
態様は、DNAオリゴとDNA消化作用物質とを含む組成物を用いて細胞をエレクトロポレーションすることによる、標的ゲノムDNA配列の配列改変に関する。一部の態様において、DNAオリゴは一本鎖である。

遺伝的疾患は、我々の社会で満たされていない課題である。CGDまたは鎌状赤血球症などの多くの遺伝的疾患は、抗菌/抗カビ予防法、CGDのためのIFN-rおよび鎌状赤血球症のための輸血を使用するなどの、症状を制御する能力のみを用いて治療されてきた。疾患を治癒する有効な方法の欠如により、CGD患者のために抗生物質/抗菌治療において有意な進歩がなされたが、残念ながらCGDを有する患者は、重篤で致命的でさえある再発性感染症を発症する。幹細胞移植は疾患を治癒することができるが、それは、厳密に適合するドナーが必要であり、ドナーを見つけるのが困難であり、適用可能性が限定される。

を含む、該細胞における標的ゲノムDNA領域の部位特異的な配列改変のための方法であって、
該DNAオリゴが、
(i)該標的ゲノムDNA領域に相同なDNA配列を含む相同領域と、
(ii)配列改変領域と
を含み、かつ
該標的ゲノムDNA領域においてゲノムDNA配列が特異的に改変される、方法。 - エレクトロポレーションが、フローエレクトロポレーションデバイスを用いるフローエレクトロポレーションである、請求項1に記載の方法。
- 前記DNA消化作用物質がヌクレアーゼである、請求項1または2に記載の方法。
- 前記ヌクレアーゼがCas9を含む、請求項3に記載の方法。
- 前記ヌクレアーゼが部位特異的なヌクレアーゼである、請求項3または4に記載の方法。
- 前記ターゲティングRNAがガイドRNAを含む、請求項5に記載の方法。

該ゲノムDNA配列改変を含む安定な細胞株を産生するために、単離したトランスフェクトされた細胞を増殖させる工程
を含む、該標的ゲノムDNA配列の該ゲノムDNA配列改変を含む安定な細胞株を産生するための方法。 - 請求項77に記載の方法により産生された細胞株。
- 請求項1〜78のいずれか一項に記載の方法を使用して産生された、エレクトロポレーションされた細胞。
- 請求項79に記載のエレクトロポレーションされた細胞または請求項78に記載の細胞株の有効量を投与することにより、疾患もしくは状態を有するかまたは疾患もしくは状態を有することが疑われる対象を処置する方法。
- 請求項79に記載のエレクトロポレーションされた細胞または請求項78に記載の細胞株の有効量を投与する工程を含む、臨床研究方法。
