)、ならびにSEQ ID NO:81および82の標準3’プライマーを用いて、テールPCRを適用した。製造者のプロトコールに従って、pEXP5CT/TOPO(登録商標)発現ベクター(Invitrogen, Carlsbad, CA, USA)に、PCR産物をクローニングした。tRNAの枯渇を回避し、かつフレームシフトのリスクを減少させるため(利用可能なリジンのトリプレットはAAAおよびAAGのみの2つであり、長いA-ストレッチをもたらす)、リジントリプレットに加えて、アルギニントリプレットを組み入れた。さらなるポリカチオン性カセットを設計されたBamHI制限部位へ挿入することによって、追加のカチオン性残基を有するテールが伸びる。この挿入は、各接合部位に、アルギニントリプレットおよびセリントリプレットを生み出す(図1)。ポリカチオン性ペプチドストレッチを4つまで、phiKZgp144およびELgp188の双方に融合し、正に荷電された各々9個、19個、29個および39個のアミノ酸残基をN末端に含む(POLY)n-gp144、または(POLY)n-gp188(n=1、2、3、4)と称した。したがって、以下の構築物を大腸菌BL21(DE3)pLysS細胞中で発現させた(37℃で対数増殖中の細胞、1 mM IPTGを使用して誘導、37℃で4時間に渡って発現)。
対数増殖期の(約106/ml)P. エルジノーサPAO1p細胞(Pirnay JP et al. (2003), J Clin Microbiol., 41(3): 1192-1202)を100倍希釈し(最終密度は約106/ml)、緩衝剤(20 mM NaH2PO4-NaOH pH 7.4;0.5 M NaCl;0.5 Mイミダゾール)において100μg/mlの最終濃度で、10μgの各々の未透析タンパク質(未改変エンドリシンphiKZgp144(SEQ ID NO:1)およびELgp188(SEQ ID NO:2)、ならびに改変されたエンドリシン変異体、POLY-gp144(SEQ ID NO:35)、(POLY)2-gp144(SEQ ID NO:36)、POLY-gp188(SEQ ID NO:39)および(POLY)2-gp188(SEQ ID NO:40)と共に室温で保温した。1時間後、細胞懸濁液をPBS緩衝剤中で希釈し(10e-5、10e-4および10e-3)、プレーティングした(標準LB培地、37℃で一晩培養)。さらに、PBS緩衝剤中に細胞を含む陰性対照をプレーティングした。一晩保温後、残留コロニーを計数した。計数された細胞数に基づいて、対数ユニット(=log10N0/Ni)での相対的な不活性化(%)(=100-(Ni/N0)*100、N0=未処置細胞の数、およびNi=処置された細胞数)として、抗菌活性を算出した(表2)。全ての試料を6回反復した。平均値/標準偏差を表す。スチューデントのt検定を使用して、統計学的解析を行った。
POLY-gp144(SEQ ID NO:35)をコードするオープンリーディングフレームを、pPICZαAシャトルベクター(Invitrogen)にクローニングし、続いて相同組換えによってP. パストリスのゲノムに組み込んだ(製造者によって示される;P. パストリスX33細胞(Invitrogen))。BMMY培地中でメタノール(1%)を用いて遺伝子発現を誘導し、1日後、3日後、および4日後に、酵素活性の存在に関して上清を解析した。したがって、1日後、3日後、および4日後に、30μlの量のP. パストリスの発現培養液の上清を、270μlのクロロホルムで透過処理されたP. エルジノーサPAO1p細胞(Pirnay JP et al. (2003), J Clin Microbiol., 41(3): 1192-1202)に添加した(緩衝剤条件:KH2PO4/K2HPO4 I=120 mM pH 6.2)。続いて、分光光度学的に光学密度を記録した(図2)。光学密度の低下は、P. パストリスによる壁分解酵素の分泌を示す。陰性対照として、発現プラスミドを含まないP. パストリスX33を包含した。したがって、上清試料を添加した場合の基質の溶解は、POLY-gp144(SEQ ID NO:35)が、P. パストリスにより成功裡に組換え産生され、かつ分泌されたことに関する尺度となる。1日後に、限定的な酵素活性を検出することができた。4日目には上清中にいかなる有意な活性の増加も観察されなかったため、最大活性は3日後に観察された。P. パストリスの細胞密度に対して、いかなる毒性効果も観察されなかった。
phiKZgp144および他のエンドリシンの、ポリカチオン性ペプチド変異体の潜在能力を試験しかつ比較するため、タンパク質のN末端に異なるポリカチオン性ペプチドを有するようコード遺伝子を合成した。ペプチドストレッチの変動は、リンカー配列の長さ、組成および挿入に関わる。一方で、N末端の複合的KRKモチーフを有するさらなるポリカチオン性ペプチドストレッチを産生した。他方で、アルギニン(R)またはリジン(K)のみからなるポリカチオン性ペプチドストレッチを産生した。さらに、長いポリカチオン性ペプチドストレッチの翻訳を増強するために、リンカー配列を含むポリカチオン性ペプチドストレッチを産生した。
SEQ ID NO:8のPSP3gp10は、サルモネラ・チフィムリウムのファージPSP3に由来する165アミノ酸残基の球状エンドリシンであり、触媒λ様ムラミダーゼドメインを含む。BLASTpおよびPfam解析によって予測されるように、PSP3gp10エンドリシンは、アミノ酸残基約34位からアミノ酸残基約152位の範囲に、その触媒ドメインを含む。

およびSEQ ID NO:51の標準3’プライマーを用いたテールPCR(同一パラメータを用いたHot Start TaqポリメラーゼPCR)を適用した。元の未改変PSP3gp10のPCR断片およびPK伸長された断片の双方を、製造者のTAクローニングプロトコールに従って、pEXP5CT/TOPO(登録商標)発現ベクター(Invitrogen, Carlsbad, CA, USA)にライゲーションした。
1.流速0.5 ml/分で、10カラム容量の洗浄緩衝剤(60 mMイミダゾール、0.5 mM NaClおよび20 mM NaH2PO4-NaOH pH 7.4)を用いた、Histrap HP 1 mlカラム(GE Healthcare)の平衡化。
2.流速0.5 ml/分で、Histrap HP 1 mlカラムへの全溶解物(所望のエンドリシンを含む)のローディング。
3.流速1 ml/分で、15カラム容量の洗浄緩衝剤を用いたカラムの洗浄。
4.流速0.5 ml/分で、10カラム容量の溶出緩衝剤(500 mMイミダゾール、5 mM NaClおよび20 mM NaH2PO4-NaOH pH 7.4)を用いた、カラムからの結合エンドリシンの溶出。

未改変および改変されたエンドリシンの抗菌活性に対するEDTAの影響を判定するため、未改変OBPgpLYSエンドリシン(SEQ ID NO:7)、およびPKOBPgpLYSエンドリシン(SEQ ID NO:61)の抗菌活性を、異なる濃度のEDTAおよびエンドリシンを使用して、シュードモナス・エルジノーサPAO1p細胞に対して試験した(Pirnay JP et al. (2003), J Clin Microbiol., 41(3): 1192-1202(2003))。対数増殖中の細菌細胞(0.6のOD600nm)を、約106/mlの最終密度に100倍希釈し、未改変エンドリシンOBPgpLYS(SEQ ID NO:7)および改変されたエンドリシンPKOBPgpLYS(SEQ ID NO:61)と共に、撹拌せず室温で30分間に渡って保温した。保温に関して、0.013μM、0.131μMおよび1.315μMのエンドリシン最終濃度で、緩衝剤(20 mM NaH2PO4-NaOH pH 7.4;0.5 M NaCl;0.5 Mイミダゾール)中で各々のエンドリシンを使用した。それによって以下、0 mM、0.05 mM、0.5 mM、および10 mMの異なるEDTA濃度を使用した。対照として、一つの試料に、エンドリシンを含まず、代わりに緩衝剤(20 mM NaH2PO4-NaOH pH 7.4;0.5 M NaCl;0.5 Mイミダゾール)を添加して、30分間保温した。保温後、細胞懸濁液を3回希釈し(各々105-104-103細胞/ml)、100μlの各希釈液をLB培地上にプレーティングした。37℃で一晩保温後、残留コロニーを計数した。計数された細胞数に基づいて、対数ユニットでの相対的な不活性化として(=log10N0/Ni、N0=未処置細胞の数、およびNi=処置された細胞数、双方とも保温後に計数)、抗菌活性を算出した(表13)。全ての試料を3回反復した。平均値+/-標準偏差を表す。観察された最大の減少(5.69対数ユニット)は、10細胞/mlの検出レベル、および初期細胞密度に依存する。「Δ」は、各々OBPgpLYSとPKOBPgpLYS試料との活性の相違を示す。
phiKZgp144および他のエンドリシンのポリカチオン性ペプチド変異体の潜在能力を試験しかつ比較するため、タンパク質のN末端にポリカチオン性ペプチドを有するコード遺伝子を合成した。
Claims (26)
- LPS破壊活性を有するカチオン性ペプチドストレッチが融合されたエンドリシンを含むエンドリシン変異体であって、
該カチオン性ペプチドストレッチが、5〜100個のアミノ酸残基を含み、該ペプチドストレッチ中に含まれる少なくとも70%のアミノ酸残基が、アルギニン、および/またはリジンであり、かつ0%〜30%がセリン、および/またはグリシンである、前記エンドリシン変異体。 - ペプチドストレッチが、5個〜50個のアミノ酸残基を含む、請求項1記載のエンドリシン変異体。
- ペプチドストレッチが、5個〜30個のアミノ酸残基を含む、請求項1または2記載のエンドリシン変異体。
- ペプチドストレッチが、エンドリシンのN末端および/またはC末端に融合される、請求項1〜3のいずれか一項記載のエンドリシン変異体。
- ペプチドストレッチが、エンドリシンのN末端に融合される、請求項1〜4のいずれか一項記載のエンドリシン変異体。
- エンドリシンが、以下からなる群より選択されるグラム陰性細菌の細胞壁を分解する活性を有する、請求項1〜5のいずれか一項記載のエンドリシン変異体:
