【0012】
Bidens pilosa L.(コセンダングサ、コシロノセンダングサ、咸豊草)
Bidens pilosa L.var.minor(Blume)Sherff(シロバナセンダングサ、シロノセンダングサ、コシロノセンダングサ、コセンダングサ、咸豊草)

Bidens pilosa L.f.decumbens Scherff (ハイアワユキセンダングサ)
Bidens pilosa L.var.radiata Scherff (タチアワユキセンダングサ、ハイアワユキセンダングサを含むこともある)
Bidens pilosa L.var.radiata Schultz Bipontinus (シロノセンダングサ、オオバナノセンダングサ)

Bidens frondosa L.(アメリカセンダングサ、セイタカタウコギ)
Bidens maximowicziana Oett(羽叶鬼針草)
Bidens parviflora Willd (ホソバノセンダングサ)
Bidens radiata Thuill.var. pinnatifida (Turcz.)Kitamura(エゾノタウコギ)
Bidens tripartita L.(タウコギ)
【0013】

【0024】
Figure 0005033292
【0025】

[Industrial application fields]
The present invention relates to a tissue repair promoting composition containing a group of plants belonging to the genus Sendangusa, in particular, a plant known as Bidence Pilosa, or an extract thereof, or a component thereof. The present invention relates to a health food or drink in daily life as a drug used in light medical treatment for minor trauma such as scratches, cuts, rough skin, etc., and for promoting the repair of bilane etc. on the inside of the digestive tract such as the esophagus, stomach, and duodenum. As such, it is highly useful.
[Prior art and problems to be solved by the invention]
[0002]
Living organisms are always damaged by various stimuli and always maintain their homeostasis while repairing them. The ability to maintain healthy homeostasis is called healing power or resistance in a broad sense, and is a healthy diet that includes unbiased diet, adequate sleep, regular life, moderate exercise, and sufficient hydration. Nourished by essential lifestyle habits.
[0003]
Various biological functions are involved in repairing damaged tissues. Enzymes that repair the extracellular structure (ECM) that makes up the tissue, various biomaterials that the enzymes need, the ability to locally mobilize such enzymes and materials, and the cells that are destroyed and lost The functions involved in the growth of the body, etc. are reconstituted with a cleverly cluttered organization.
[0004]
In other words, ECM in damaged tissues is a fibrous protein such as collagen or elastin, a complex polysaccharide glycosaminoglycan (GAG) containing hyaluronic acid (HA) or chondroitin sulfate, glycoproteins such as fibronectin and laminin. Etc. As a result of the infiltration of blood components in the damaged tissue, the collection of cell groups involved in tissue-derived fibroblasts and inflammatory reactions, the old ECM is decomposed and the reconstruction of a new ECM proceeds, cell division and The tissue is repaired with angiogenesis. In the process, a granulation tissue is temporarily formed, and with this as a support, cells having the original function are induced to be differentiated and the repair of the tissue is completed.
[0005]
In order to artificially supplement the natural healing power of such a living body, it is also necessary to promote supplementation of necessary materials and elimination of waste products by improving blood circulation in the affected area. For example, ginger, capsicum, or garlic, which has been known to increase circulation from the old days, also makes you feel hot and warm after ingestion. On the other hand, many plants are known to keep the affected area clean and prevent infection and promote natural healing. For example, it is Sikon, Awaku, Daio, etc. These plants and their components have long been used as folk medicines and lore, and many of them have been isolated from medicinal ingredients and used as new drugs.
[0006]
The present invention aims to provide a method for artificially compensating for a biological reaction mobilized to repair a damaged tissue from a new viewpoint.
[0007]
Sendungusa plants, especially Bidence pirosa, do not cause allergic reactions to people of any constitution, tend to inhibit platelet aggregation, and promote peripheral blood flow by increasing the degree of blood smoothness, so every day There is a real feeling that decocting and drinking warms the limbs and moisturizes the skin. It also has anti-oxidation / anti-peroxidation, blood glucose-lowering, blood lipid-lowering, anti-inflammatory, cell growth-promoting, collagen synthesis-promoting, etc. It can be expected to maintain health at a high level.
[0008]
In addition, the present invention is a true health that can be used by healthy people to maintain their health, so that they can quickly recover from disability and achieve a healthy life without losing their homeostasis. It is intended to provide food and health supplements, and to realize a bright and long-lived society with an extended healthy life expectancy.
[Means for Solving the Problems]
[0009]
Bidence Pilosa is an Asteraceae wild grass widely distributed in the tropics and subtropics, and its fertility is strong and research on extermination is harmful as a harmful plant in agriculture. On the other hand, mainly in the African continent, Central and South America, China and Taiwan, etc. In antibacterial and anti-inflammatory analgesic, gastrointestinal diseases such as stomach pain, abdominal pain, diarrhea, constipation, intestinal parasites, infections such as pharyngitis, tuberculosis, dysentery, malaria, antipyretic such as rubella, fever, It has been empirically used as a folk medicine for inflammatory diseases such as conjunctivitis, otitis, and enteritis, rheumatism, diabetes, liver disease, or bites of burns and poisonous snakes. It is also used universally for purposes such as cough, chickenpox, wheezing, drenched eyes, toothache, severe emesis (cancer), cancer, adenoma, and diuresis. In other words, it seems to be used as a convenient wild grass nearby in low-developed areas. However, there are no findings suggesting the promotion and improvement of tissue repair, as researchers from all over the world are conducting various research reports and component studies.
[0010]
The inventors focused on ECM reconstitution as shown in the experimental example, and Bidens pilosa L. promotes the synthesis of collagen and glycosaminoglycan in in vivo tests using guinea pigs and promotes wound healing I discovered that. It was also found in in vitro tests using human fibroblasts that it contributes to wound healing by promoting cell migration and proliferation.
[0011]
The Asteraceae plant belonging to the asteraceae used in the present invention has already been described in detail in Japanese Patent Application No. 2000-105559, but it is a group of plants referred to by the genus Bidens in scientific name, and since there are many types of crosses, there are many varieties. However, there is confusion in botany, and it is extremely difficult to identify because the correspondence between scientific names, Japanese names, and Han names is mixed, but the Sendangsa plants used in the present invention are listed below including hybrids. Including things.

This Sendangsa plant has needles with reverse spines on top of dark brown seeds carried on animals and human clothes, and the number of needles is often uneven due to mating, but in China and Taiwan The genus, mainly called 咸 豊 草. There are also many synonyms, Doji, Oni, Tsubasa, Tsubasa, Tsubasa, Tsubaki, Aoi, Hakuhana, Aoi, Nobushi, Noboshi, Redhead, Gold There are names such as “Ginban”, “Wind Grass”, “South Wind Grass”, “King Park”, “Study”, etc. It is not clear which scientific name each corresponds to. In Japan, it is found in the south of Honshu, where the plant height ranges from 25cm to about 1m. Usually, it is an annual plant, but it overwinters when it is blessed with warm climatic conditions, and the flowers bloom one after another. The flower has a unique shape of the Asteraceae, with several tongue-like flowers like white or yellow round petals and many yellow-brown tubular flowers in the center. Some lack tongue-like flowers. The stem has square and light purple colored nodes. The leaves that are divided into three or five feathers have a pattern, and the edges have jagged edges, often facing each other.
[0014]
[Experiment 1]
A Hartley male guinea pig weighing about 300 g was shaved on an area of the back under anesthesia, and further depilated using a hair removal mousse. After alcohol disinfection, the wound was cut about 15 mm to the depth reaching the muscle layer with scissors on the back, and the wound was sutured using an automatic suturing device. To prevent infection, 5 mg of chloramphenicol was subcutaneously injected every other day. The day of cut was defined as day 0, and the skin 10 mm long and 5 mm wide was peeled around the wound after 3, 5, 7, and 10 days.
[0015]
About this exfoliated section, collagen was measured using hydroxyproline (Hypr) as an index, and glycosaminoglycan was measured using uronic acid (UA) as an index, and converted to a unit weight of delaminated dehydrated dehydrated skin sample. The numerical values in Table 1 are expressed as% relative to the normal group.
[0016]
[Table 1] Hypr and UA relative changes over time

* 3 : p <0.001, * 2 : p <0.01, * 1 : p <0.05 (for normal group)
# 3 : p <0.001, # 1 : p <0.05 (vs. control group), Student's t-test
[0017]
There are 4 animals (10 animals per group), the normal group is the group that is not cut, the control group is the group that is not applied with the drug after the cut, and about 30 mg of white petrolatum containing 10% Tachiawayukisendangsa (Bidence Pilosa) is used every day. A group applied to the wound was a test group, and a group applied with white petrolatum containing shikonin 0.2% was a shikonin group. The ointment of the test group used for this is a fine powder obtained by repairing and drying the above-ground part of Tachiwa Yusen Sendanga by the method of Japanese Patent Application No. 2000-105559, and kneading it in white petrolatum at a rate of 10%. Vaseline base was used because it was more effective than water-soluble base in the preliminary test. Shikonin used as a control was pure and was used as an ointment with a concentration of 0.2% that had been recognized as appropriate in other experiments.
[0018]
As a result, the collagen in the wound decreased in the control group as 71% on the 3rd day, 83% on the 5th day, 88% on the 7th day, and 90% on the 10th day in the control group. However, in the test group, the decrease was significantly suppressed to 99% on the 3rd day, and on the 5th, 7th and 10th days, it was not significant, but continued to show a tendency of suppression. Shikonin was unfortunate because there was no data on the third, seventh and tenth days, but it was 76% on the fifth day, a decrease from the control group. That is, it was discovered that Bidens pilosa L. greatly contributed to tissue repair from the rise and fall of hydroxyproline, which is a characteristic protein of collagen.
[0019]
Although uronic acid was not significantly different on the 7th day, the increase was significantly observed on the 3rd, 5th and 10th days of the test group. In contrast, the shikonin group was only on the fifth day, but no effect of increasing glycosaminoglycan was observed at the same level as the control group. In other words, Bidens pilosa L. discovered that it contributed to the construction of glycosaminoglycans far more than shikonin and achieved tissue repair.

[Experimental example 2]
Using Dulbecco's Modified Eagle Medium (DMEM) containing 10% calf serum (FBS), seed 1x10 4 cells / dish of human fibroblasts in a 35mm Petri dish at 37 ° C and 5% CO 2 Cultured. Connfluent, washed twice with PBS, replaced with DMEM containing 0.5% dialyzed FBS, cultured for 24 hours under the same conditions, then replaced with PBS, and in the middle of the petri dish with a large yellow tip × according to the method of Meyer et al. The cells were peeled off (peeling width about 0.5 mm). After thoroughly washing with PBS, DMEM containing 0.5% dialyzed FBS was treated with a drug having the concentration shown in Table 2 as a control, and the state of the peeling groove was examined after 24 hours.
[0021]
As a result, the state of the peeling groove showed the most cell migration / extension in the fraction D, and the similar situation was also observed in shikonin, fraction C, and hyaluronic acid.

[Experiment 3]
Human skin fibroblasts are seeded at 1 x 10 3 cells (100 µL) in each well of a 96-well microtiter plate and used for 24 hours under conditions of 37 ° C and 5% CO 2 using DMEM containing 10% calf serum. After incubation, wash twice with PBS, treat with DHEM containing 0.5% dialyzed FBS as a control, and use the treatment solution shown in Table 2. After 24 hours, add 10 μL of Cell Counting Kit 8 solution to each well and measure the absorbance at 450 nm. It was used as an index of cell number.
[0024]
Figure 0005033292
[0025]
As a result, as shown in Table 3, Bidens pilosa L., fraction D, and hyaluronic acid showed significant cell growth promotion, but shikonin did not promote growth.
[0026]
[Experimental Example 4]
Three groups of 5-8 ICR male 7-week-old mice were prepared per group, suspended in water for the control group, 200 mg cimetidine per kg body weight for the positive control group, and 0.25% carboxymethylcellulose solution for the test group. Bidens pilosa L. (same as Experimental Example 2) 0.5 g per kg body weight was forcibly administered with a gastric sonde, and 1 hour later, 0.1% M-hydrochloric acid 200% containing 60% ethanol was injected with the gastric sonde. A stomach ulcer was caused, and after 1 hour, the abdomen was opened, and the stomach was washed to obtain a constant amount, and the absorbance at 540 nm was measured as an indicator of the amount of bleeding. Moreover, the ulcer area of the stomach inner surface was measured by image analysis. As shown in Table 4, the ulcer prevention effect was not inferior to that of cimetidine, and all were highly significant compared to the control group (p <0.001).

DETAILED DESCRIPTION OF THE INVENTION
Embodiments of the present invention will be specifically described below with reference to examples, but the present invention is not limited to these examples.
[0029]
[Example 1]
(Preparation of health tea and effects on upper abdominal complaints)
The above-ground part of Bidence Pilosa (Tachiawayukisendangusa) was cut, washed with running water, cut and digested, then dried with hot air and finely chopped into raw materials for healthy tea. By itself, wild grass taste and wild grass odor were strong, so 2 g of dried ginger and 28 g of roasted barley (both coarsely crushed) were mixed with 70 g of the ginger to correct the taste, and 3 g each was packaged in the shape of a tea bag. This was poured into 1 liter of boiling water, extracted for about 5 minutes, kept warm in a pot and used for drinking as needed.
Ten people with complaints of suspected gastritis / gastric ulcers such as upper abdominal pain, stomach sag, nausea, vomiting (blood mixing), loss of appetite, stomach pain during fasting, etc. Within a month, the eight were completely relieved of their complaints.
[0030]
[Example 2]
(Preparation of extract powder to ointment and effect on light trauma)
Add 2 liters of water to 100 g dry product of Bidence pilosa (Shirobanasendangsa), extract for 5 hours with occasional stirring at 90-100 ° C., filter, concentrate, freeze-dry, and freeze Bidence pilosa hot water 23.1 g of extract powder was obtained. 1 g of this product is equivalent to about 4.3 g of dried evidence and pillows. This powder was kneaded into Carbowax 5000 / white petrolatum / olive oil (4/4/2) at a ratio of 0.1 g / 100 g to prepare an ointment.
The ointment was tried with the consent of 10 parents who were constantly injured with small injuries. When each schoolchild fell and scratched his knees and other shins, they were sterilized with rivanol after washing with water and tried this ointment. It was well-received that it healed quickly.
[0031]
[Example 3]
(Preparation of granule and alternative effect of cimetidine)
The processed dried product of Bidence Pilosa used in Experimental Example 1 was made into a 60-mesh-pass powder, and 85 g of this powder was mixed with 15 g of crystalline cellulose (Avicel) as an excipient and granulated wet. When administered to a subject, about 3 g was usually given as a daily dose in 3 doses.
A patient who was diagnosed with gastritis, gastric ulcer, or duodenal ulcer and explained to a patient receiving cimetidine and obtained consent, and 8 people took 1g of this granule instead of cimetidine for 1 month between meals. All reported that they were doing better than cimetidine.
[0032]
[Example 4]
(Preparation of tablets and effects on dermatitis)
Trehalose 70 g and crystalline cellulose (Avicel) 30 g were added as excipients to 100 g of the freeze-dried powder of the Bidence pirosa extract obtained in Experimental Example 2, and 1 tablet 350 mg was prepared by a conventional method. When administered to a subject, one tablet was orally administered usually three times a day.
For 5 people who have been administered topical steroids by doctors with severe atopic dermatitis that is always accompanied by bleeding in a significant part of the skin, but do not want to use it because of refusal to steroids. When they were used together with the health tea of Example 1 three times a day, all were released from dermatitis in 2 to 6 months.
[0033]
[Example 5]
(Effective against summer ulcers)
Figure 0005033292
[0034]
The above is a part of the 22 cases listed as Example 5 in the patent application “Blood Circulation Improving Agent” (Japanese Patent Application 2001-001720), but the healthy tea of Example 1 was boiled to about 1 liter of hot water. This is an example in which one pack is placed, kept warm, and divided into several times a day for drinking. Seven of them, Livedo reticularis with summer ulceration, are intractable blood circulation disorders that cause painful ulcers in the lower limbs during the summer. This is due to improvement of blood flow and newly discovered tissue repair. The promotion effect is also considered to have contributed greatly.
