本発明はセンダングサ属植物特にビデンス・ピローサと呼ばれる一群の植物またはその抽出物、またはその成分を含有する組織修復促進性組成物に関するものである。本発明は、擦り傷・切り傷・荒れ肌等の軽度の外傷に対する軽医療に用いる薬剤として、また食道・胃・十二指腸等消化管内面のビラン等の修復促進のための、日常生活における健康食品や健康飲料としても利用価値の高いものである。

センダングサ属植物、特にビデンス・ピローサはどんな体質の人にもアレルギー反応を起こすこともなく、血小板凝集を阻害する傾向があり、血液のサラサラ度を高めることによって末梢血液の流動を促進するので、毎日煎じて飲むことにより手足が温まり皮膚に潤いをもたらすという実感がある。また抗酸化・抗過酸化作用、血糖低下作用、血中脂質低下作用、抗炎症作用、さらに細胞増殖促進作用、コラーゲン合成促進作用、等々を併せ持っているので、これを常用することによって積極的に高いレベルで健康を保持することが期待できるものである。

ビデンス・ピローサは熱帯〜亜熱帯に広く分布するキク科の野草で、繁殖力が強く農業上は有害植物として駆除の研究が盛んである一方、主にアフリカ大陸や中南米地方、中国・台湾などの各地では抗菌作用、消炎鎮痛作用があるとして、胃痛・腹痛・下痢・便秘・腸内寄生虫等の胃腸疾患、咽頭炎・結核・赤痢・マラリア等の感染症、風疹・猩猴熱等の解熱、結膜炎・耳炎・腸炎等の炎症性疾患、リウマチ、糖尿病、肝臓病、あるいは火傷・毒蛇の咬傷などに経験的に民間薬として用いられてきた。また日常的に咳、水洟、喘鳴、ただれ目、歯痛、重症悪阻(おそ)、癌、腺腫、利尿などの目的にも万能薬的に用いられている。すなわち低開発地域においては手近にある便利な野草として重用されているらしい。しかし世界各地の研究者によって多岐に亘る調査報告や成分研究がなされている中に、組織修復の促進改善を示唆する知見は全く見られない。

発明者は、実験例に示すようにECMの再構成に着目し、モルモットを用いるin vivo試験においてBidens pilosa L.がコラーゲンやグリコサミノグリカンの合成構築を促進し創傷の治癒を促進していることを発見した。またヒト繊維芽細胞を使用するin vitro試験においても、細胞の遊走伸展及び増殖を促進して創傷の治癒に貢献することが見いだされた。

本発明に使用されるキク科のセンダングサ属植物は、既に特願2000−105559に詳述したが、学名ではビデンス属と言われる一群の植物で、種類も多岐に亘り互いに交配するので変種も多く、植物学上も混乱が見られ、学名、和名、漢名、の対応も交錯していて同定することは極めて困難であるが、本発明で用いられるセンダングサ属植物は雑種を含め以下に掲げるものを包含する。

このセンダングサ属植物は動物や人の衣服に付いて運ばれる黒褐色の種子の上部に逆棘のある針があり、針の数は交配のため一定しないものが多いが、中国・台湾では鬼針草属、主に咸豊草と呼ばれる。異名も多く、同治草、鬼針草、三葉鬼針草、三葉刺針草、刺針草、婆婆針草、白花婆婆針、蝦箝草、符因草、符因頭、赤査某、金盞銀盤、含風草、南風草、蝦公鋏、羞査某仔等の名があり、それぞれがどの学名に相当するのかは明らかでない。日本では本州の暖地以南で見られ、草丈は25cm〜約1mにもなり、通常一年草であるが温暖な気候条件に恵まれると越冬し、花は一年中次々と咲く。花はキク科特有の形で、白色または黄色の丸みのある花弁のような舌状花が数個、中央には黄褐色の管状花が多数集合している。中には舌状花を欠くものもある。茎は四角で薄紫に着色した節がある。3つ又は5つに羽状に分れた葉には柄があり、縁にはぎざぎざがあって対生するものが多い。

体重300g前後のHartley系雄モルモットを麻酔下に背中の一定範囲を剃毛し、さらに脱毛ムースを使用して脱毛した。アルコール消毒後、背中に鋏で筋層に達する深さまで約15mm切傷し、自動縫合器を用いて傷口を縫合した。感染防止のために隔日にクロラムフェニコール5mgを皮下注射した。切傷した日を0日とし、3、5、7、10日後に傷口を中心に縦10mm、横5mmの皮膚を剥離した。

動物は4群(各群10匹)とし、切傷しない群を正常群、切傷後薬物を塗布しない群を対照群、切傷後10%タチアワユキセンダングサ(ビデンス・ピローサ)含有白色ワセリン約30mgを毎日傷口に塗布した群を試験群、シコニン0.2%含有白色ワセリンを塗布した群をシコニン群とした。これに使用した試験群の軟膏はタチアワユキセンダングサの地上部を特願2000−105559の方法によって修治加工乾燥したものを微粉末とし白色ワセリンに10%の割合で練り込んだものである。ワセリン基剤を用いたのは予備試験において水溶性基剤よりも効きが良かったからである。対照に用いたシコニンは純品で、かねて別の実験で適当と認められた濃度の0.2%の軟膏として使用した。

その結果、創傷の治癒過程において傷口のコラーゲンは対照群では3日目71%、5日目83%、7日目88%、10日目90%と低下し、分解除去されていることを示しているが、試験群では3日目99%と低下が有意に抑制され、5、7、10日目は有意ではないが引続き抑制の傾向を示した。シコニンは3、7、10日目のデータがなく残念であるが5日目に76%と対照群よりも減少していた。すなわち、コラーゲンの特徴蛋白であるヒドロキシプロリンの消長からBidens pilosa L.が組織修復に大きく貢献していることを発見した。

10%子牛血清(FBS)を含むDulbecco’s Modified Eagle Medium(DMEM)を用い、35mmペトリ皿で1×10cell/dishのヒト繊維芽細胞を播種し、37℃、5%COの条件で培養した。connfluentになってPBSで2回洗浄、0.5%透析FBSを含むDMEMに交換し、同条件で24時間培養後PBSに交換し、Meyerらの方法に準じてシャーレの中央に大きくイエローチップで×形に細胞を剥離した(剥離幅約0.5mm)。PBSで十分洗浄後0.5%透析FBSを含むDMEMを対照に表2の濃度の薬物で処理し、24時間後に剥離溝の状態を検鏡した。

1群5〜8匹のICR雄性7週令マウスを3群用意し、対照群には水、陽性対照群には体重kg当り200mgのシメチジン、試験群には0.25%カルボキシルメチルセルロース溶液に懸濁したBidens pilosa L.(実験例2と同じ)を体重kg当り0.5g、をそれぞれ胃ゾンデで強制投与して1時間後に、60%エタノール含有0.15M−塩酸200μLを胃ゾンデで注入して実験的に胃潰瘍を起こさせ、1時間後に開腹、胃を洗浄して一定量とし540nmの吸光度を測定して出血量の指標とした。また胃内面の潰瘍面積を画像解析にて測定した。その結果は表4に見られるようにシメチジンに勝るとも劣らぬ潰瘍予防効果を認め、いずれも対照群に対して高度に有意差があった(p<0.001)。

【発明の実施の形態】
以下実施例をもって本発明の実施態様を具体的に説明するが、本発明はこれらの実施例に限定されるものではない。

【実施例1】(健康茶の調製と上腹部愁訴に対する効果)
ビデンス・ピローサ(タチアワユキセンダングサ)の地上部を刈り取り、流水で洗浄して裁断したものを蒸煮・揉解した後、熱風乾燥して細かく刻み、健康茶の原料とした。単独では野草味・野草臭が強いので、その70gに乾燥ショウガ2gと焙煎した大麦28g(いずれも粗砕物)を混合して味を矯正し、3gずつティーバッグの形に分包した。このものは沸騰水1Lに1包を投入して約5分間抽出し、ポットに保温して随時飲用に供した。
上腹部痛、胃もたれ、吐き気、嘔吐(血が混じる)、食欲不振、空腹時の胃痛など、胃炎・胃潰瘍を疑わせる愁訴のある人10人に、この健康茶を常用してもらったところ1カ月以内に8人は愁訴が全く消失して元気になった。

小さな怪我の絶えない学童10名の保護者の了解を得てこの軟膏を試用してもらった。各学童とも転んで膝や向こう脛に擦り傷を負った時に、水洗後リバノール液で消毒してこの軟膏を試用してもらい、保護者の所感を求めた所、全員が非常によく効く、傷の治りが速い、と好評であった。
