Description
本実施例では、Pichia属、Candida属、Debaryomyces属又はWilliopusis属に属する46種類の酵母菌株を、セロビオースを含む培地で培養した後、培地中のセロビオース及びエタノール量を測定した。なお、供試した菌株はATCC又はNBRCから分譲を受けた菌株である。
移動層:0.005NH2SO4
流速:0.6ml/min
カラム温度:35℃
検出:RI
46種類の菌株についてエタノール収率を以下のように算出した。収率は理論収率に対する収率として以下のとおり求めた。
NL2:5'-CTCTCTTTTCAAAGTTCTTTTCATCT (配列番号7)
NL3:5'-AGATGAAAAGAACTTTGAAAAGAGAG(配列番号8)
NL4:5'-GGTCCGTGTT TCAAGACGG(配列番号9)
次に、増幅した26s rDNA-D1/D2領域について、ABI PRISM 3100 Genetic Analyzer System (Applied Biosystems社製)を用いて塩基配列を決定した。Pichia pini ATCC76276株の26s rDNA-D1/D2の塩基配列を配列番号1に示し、 Pichia pini ATCC28781株の26s rDNA-D1/D2の塩基配列を配列番号2に示し、Pichia pini NBRC1794株の26s rDNA-D1/D2の塩基配列を配列番号3に示し、Pichia glucozyma ATCC18938株の26s rDNA-D1/D2の塩基配列を配列番号4に示し、Pichia dorogensis NBRC101078株の26s rDNA-D1/D2の塩基配列を配列番号5に示した。得られた塩基配列情報に基づき、国際塩基配列データベース(GenBank/DDBJ/EMBL)を用いて相同性検索を行った。
Claims (5)
- Ogataea dorogensis NBRC101078株、Ogataea pini ATCC76276株、Ogataea neopini ATCC28781株及びOgataea corticis NBRC1794株からなる群から選ばれる少なくとも1種の微生物を、セロビオースを含有する培地にて培養する工程と、
培地からエタノールを回収する工程と
を含むエタノールの製造方法。 - 上記セロビオースは、木質バイオマスに含まれるセルロース由来であることを特徴とする請求項1記載のエタノールの製造方法。
- 上記木質バイオマスに含まれるセルロースをセロビオハイドロラーゼにより分解する工程を更に含むことを特徴とする請求項2記載のエタノールの製造方法。
- 上記微生物は、セロビオース資化性を有するものであることを特徴とする請求項1記載のエタノールの製造方法。
- 上記微生物は、キシロース資化性を更に有するものであることを特徴とする請求項4記載のエタノールの製造方法。
