Description
本発明は、流体試料を分析して試料中の分析物の有無を測定する装置と方法を供給する。この装置は、試料から所望の分析物を分離し、その分析物を化学反応と光学的検出のために入れるカートリッジを有する。この装置は、試料処理のためにカートリッジを受ける装置を備える。所望の分析物には例えば、有機体、細胞、タンパク質、核酸、炭水化物、ウイルス分子、バクテリア、化学物質や生化学物質が含まれる。好ましくは、所望の分析物は核酸を含み、行われる化学反応は、例えば、ポリメラーゼ・チェーン・リアクション(PCR)を使う核酸の増幅がよい。

ここで、I0はボルトで表示したフォトンなどの放射光の出力照度、Cは検出されるべき分析対象物の濃度、Iiは入力照度、Lは光路長、Aは分析対象物の標識に用いた染料の固有吸光率である。
Claims (26)
- カートリッジ内で化学反応を行わせる方法であって、このカートリッジは、装置に載置され、
(1) 内部に形成された少なくとも第1と第2の流路を有する本体と、
(2) 上記本体から延び出す反応容器とを備え、上記装置は、熱面を有し、上記反応容器を収容する収容部を備え、上記反応容器は、
i) 反応室の複数の側壁を区画し、入口流路と出口流路を有する剛な枠と、
ii) 上記反応室に、十分柔軟で熱面に一致する主壁を形成すべく、上記剛な枠に取り付けられる少なくとも1つのシートまたは膜と、
iii) 上記反応室に上記入口流路を介して接続され、反応室に流体を供給する入口ポートと、
iv) 上記反応室に上記出口流路を介して接続され、反応室から流体を排出する出口ポートとを有し、
上記反応容器の入口ポートは、上記本体内の第1の流路に接続され、上記反応容器の出口ポートは、上記本体内の第2の流路に接続されており、
上記方法は、上記第1の流路と入口ポートを経て上記反応室に試料を導入し、この反応室内で化学反応を行わせる方法。 - 請求項1に記載の方法において、上記本体が、上記第2の流路に連通してガスを逃がすベントを更に有する方法。
- 請求項1または2に記載の方法において、上記カートリッジが、上記本体の第1の流路内の流体を上記反応容器の入口ポートを経て上記反応室に送る差圧源を更に有する方法。
- 請求項1乃至3のいずれか1つに記載の方法において、上記反応容器が、上記反応室の対向する2つの主壁を形成すべく上記剛な枠に取り付けられる第1と第2の柔軟なシートまたは膜を有する方法。
- 請求項4に記載の方法において、上記第1と第2の柔軟なシートまたは膜が、夫々ポリマー膜からなる方法。
- 請求項4または5に記載の方法において、上記側壁の少なくとも2つは、上記反応室内に光を伝えるためおよび反応室から出る光を検出するため透光性であるとともに、所定の角度をなして互いに隣接する方法。
- 請求項6に記載の方法において、上記角度が、90°である方法。
- 請求項1乃至7のいずれか1つに記載の方法において、上記反応室の厚さに対する幅の比が、少なくとも2:1である方法。
- 請求項1乃至8のいずれか1つに記載の方法において、上記反応室が、5mm以下の厚さを有する方法。
- 請求項1乃至9のいずれか1つに記載の方法において、上記反応室が、0.5〜5mmの範囲の厚さを有する方法。
- 請求項1乃至7のいずれか1つに記載の方法において、上記反応室の厚さに対する幅の比が、少なくとも4:1である方法。
- 請求項1乃至7または11のいずれか1つに記載の方法において、上記反応室が、2mm以下の範囲の厚さを有する方法。
- 請求項1乃至7または請求項11乃至12のいずれか1つに記載の方法において、上記反応室が、0.5〜2mmの範囲の厚さを有する方法。
- 請求項1乃至13のいずれか1つに記載の方法において、この方法は、流体試料から核酸を分離し、この核酸を増幅および検出のために保持するステップを更に備え、上記本体が、上記核酸を増幅試薬と混合するための混合室を更に有し、この混合室が、上記反応容器の入口ポートに上記第1の流路を経て接続されている方法。
- 請求項1乃至14のいずれか1つに記載の方法において、上記本体が、内部に形成された
i) 試料流路と、
ii) 試料流体から所望の分析対象物を分離すべく、上記第1の流路を経て上記入口ポートに接続される上記試料流路内の分離領域とを有する方法。 - 請求項15に記載の方法において、上記本体内の分離領域が、
a) 上記試料流路に設けられ、上記試料中の細胞またはウイルスを溶解させて、これらの細胞またはウイルスから物質を解き放たせる溶解室と、
b) 上記溶解させられるべき細胞またはウイルスを捕捉すべく、上記溶解室内に設けられた少なくとも1つの固体の担体とを有する方法。 - 請求項1に記載の方法において、上記カートリッジが上記装置の一部であって、この装置が、
a) 上記シートまたは膜に接触する少なくとも1つの熱面と、
b) 上記反応室内の圧力の増加が、上記シートまたは膜を上記熱面に一致するように接触させるに十分であるように、上記反応室内の圧力を増大させる手段と、
c) 上記反応室内の温度変化を引き起こさせるべく上記熱面を加熱または冷却する少なくとも1つの熱要素とを更に有する方法。 - 請求項6に記載の方法において、上記カートリッジが上記装置の一部であって、この装置が、上記透光性の側壁のうちの第1の側壁を介して上記反応室内に光を送る少なくとも1つの光源と、上記反応室から上記透光性の側壁のうちの第2の側壁を介して放出される光を検出する少なくとも1つの検出器とを有する光学システムを更に備えている方法。
- 請求項1乃至16のいずれか1つに記載の方法において、上記反応室の各側壁が、1〜15mmの長さを持つ方法。
- 請求項6に記載の方法において、上記反応容器は、上記反応室を通る光路長が、1〜15mmであるカートリッジ。
- 請求項6に記載の方法において、上記反応容器が、上記反応室を通る5〜12mmの光路長を有する方法。
- 請求項1乃至16または19乃至21のいずれか1つに記載の方法において、上記反応室が、100μリッター以下の体積容量を有する方法。
- 請求項1乃至16または19乃至21のいずれか1つに記載のカートリッジにおいて、上記反応室は、体積容量が12〜100μリッターであるカートリッジ。
- 請求項1乃至16または19乃至23のいずれか1つに記載の方法において、上記反応室の壁の少なくとも1つが、0.5mm以下の厚さを有する方法。
- 請求項1乃至16または19乃至24のいずれか1つに記載の方法において、上記反応室の壁の少なくとも1つが、0.003〜0.5mmの厚さを有する方法。
- 請求項1乃至16または19乃至24のいずれか1つに記載の方法において、上記本体内の第1と第2の流路が、0.4〜3.2mmの幅または直径を有する方法。
