Description

新規なポリペプチド(これは、T細胞同時刺激経路におけるタンパク質に関する)が、同定されている。このポリペプチド(B7−様ポリペプチド(B7−L)と呼ばれる)は、CD80、CD86、B7RP−1、およびB7−H1に類似の構造を有し、そしてこれらと相同性を共有するB7関連ポリペプチドを表す。ヒトB7−Lポリペプチドは、B7ファミリーの他のメンバー(B7rp−1、20%;B7−1、12%)とよりも、ヒトB7−H1とより高いアミノ酸同一性(35%)を共有する。さらに、B7−LポリペプチドおよびB7−H1のマウスおよびヒトのオルソログは、B7−1、B7−2、またはB7rp−1のマウスおよびヒトのオルソログ(約40%)よりも、より高い程度のアミノ酸相同性を共有する(約70%)。さらに、B7−H1およびB7−Lの両方は、同じレセプター(PD−1)に結合する。

(A.細菌におけるB7−Lポリペプチドの発現)
PCRを用いて、B7−LポリペプチドをコードするテンプレートDNA配列を増幅する。このPCRには、この配列の5’および3’末端に対応するプライマーを用いる。増幅したDNA産物を改変して、発現ベクター内への挿入を可能にするために制限酵素部位を含むようにし得る。PCR産物をゲル精製し、そして標準的組み換えDNA方法論を用いて発現ベクター中に挿入する。luxプロモーターおよびカナマイシン耐性をコードする遺伝子を含むpAMG21(ATCC番号98113)のような代表的なベクターを、挿入したDNAの方向性クローニングのために、BamHIおよびNdeIを用いて消化する。連結した混合物を、エレクトロポレーションによってE.coli宿主株中に形質転換し、そして形質転換体をカナマイシン耐性について選択する。選択したコロニー由来のプラスミドDNAを、単離し、そしてDNA配列決定に供してインサート(挿入物)の存在を確認する。
PCRを用いて、B7−LポリペプチドをコードするテンプレートDNA配列を増幅する。このPCRには、この配列の5’および3’末端に対応するプライマーを用いる。増幅したDNA産物を改変して、発現ベクター内への挿入を可能にするために制限酵素部位を含むようにし得る。PCR産物をゲル精製し、そして標準的組み換えDNA方法論を用いて発現ベクター中に挿入する。エプスタイン−バーウイルス複製起点を含む代表的な発現ベクターであるpCEP4(Invitrogen,Carlsbad,CA)を、293−EBNA−1細胞におけるB7−Lポリペプチドの発現のために用い得る。増幅およびゲル精製したPCR産物を、pCEP4ベクターに連結し、そしてリポフェクチンによって293−EBNA細胞中に導入する。トランスフェクトした細胞を、100μg/mLのハイグロマイシン中で選択し、そして得られた薬物耐性培養物をコンフルエンスになるまで増殖する。次いで、この細胞を無血清培地中で72時間培養する。馴化培地を取り出し、B7−Lポリペプチド発現をSDS−PAGEによって分析する。
B7−Lポリペプチドのアミノ末端に220アミノ酸をコードするB7−L cDNAの660bpフラグメント(これは、B7−Lポリペプチドのシグナルペプチドおよび細胞外ドメインを含む)を、適切なプライマーおよび標準的な技術を使用して、PCRによって増幅した。実施例1に記載される特別に合成したライブラリーから単離したプラスミドDNAを、PCR増幅におけるテンプレートとして使用した。
Transfection Reagent(Roche Molecular Biochemicals,Indianapolis,IN)を使用して、293−EBNA−1細胞をトランスフェクトした。形質転換後、この細胞を、10%ウシ胎仔血清を補充したDMEM培地中、37℃で24時間培養し、次いでDMEM無血清培地に移し、そして37℃で5日間増殖させた。このB7−Lポリペプチド/Fc融合タンパク質を、製造者の指示に従ってHiTrap Protein A Column(Amersham Pharmacia Biotech,Piscataway,NJ)上で順化培地から精製し、次いでこの融合タンパク質を、PBS中で透析した。この精製したタンパク質の濃度を、標準としてCoomassie(登録商標)Plus Protein Assay Reagent(Pierce,Rockford,IL)およびBSAを使用して、製造者の指示に従って測定した。
既知のB7媒介同時刺激経路の1つにおいてB7−Lポリペプチドが機能するか否かを決定するために、B7−Lポリペプチド/Fc融合タンパク質での処理後に、CD28、CRP−1/ICOS、またはPD−1を発現するCHO D−細胞上でFACS分析を行った。
B7−Lポリペプチドが、T細胞増殖における役割を果たすか否かを決定するために、B7−Lポリペプチド/Fc融合タンパク質を使用してヒトT細胞増殖アッセイを行った。高度に精製されたヒトT細胞(>98% CD3+)を、Pan T細胞単離キット(Miltenyi Biotec,Auburn,CA)を使用して、新鮮であるかまたは解凍した、接着性が劣化した末梢血単核細胞(PBMC)のネガティブ選択によって単離した。丸底96ウェル細胞培養プレートを、0.5または1.5μg/mlの抗CD3抗体(PharMingen,San Diego,CA)、10μg/mlの抗ヒトIgG Fc(Sigma)、およびPBS(容量0.1ml)で、4℃で一晩プレコーティングした。このプレコーティング溶液を除去し、このプレートをPBSで1回洗浄し、そして10%のFCS培地を補充したRPMI 1640または培地単独中、20μg/mlまで希釈した0.1mlのB7−Lポリペプチド/Fc、B7−2/FcまたはB7rp−1/Fc融合タンパク質を、このウェルに加えた。次いで、このプレートを、37℃で4時間インキュベートした。インキュベーション後、培地を除去し、そして0.2mlの精製T細胞(10×105個の細胞を含む)をプレコーティングした各ウェルに加えた。次いで、このプレートを、37℃で48時間インキュベートした。インキュベーション後、1μCi/ウェルの[3H]チミジンを各ウェルに加え、そしてこの培養物を37℃でさらに18時間インキュベートした。次いで、この細胞を収集し、そして1分当たりのカウント(CPM)を測定した。
FACS分析を使用して、ヒトPBMCにおけるT細胞およびB細胞に対するB7−Lポリペプチドレセプターを同定した。Ficoll−Paque(Amersham Pharmacia Biotech)勾配遠心分離を使用して、健常なヒトのボランティアから得た血液からPBMCを精製し、そしてこの細胞を、10μg/mlのリポ多糖(LPS)中での3日間のインキュベーションによって刺激した。LPS処理後、0.2mlの容量中の5×105個の細胞を、氷上で10分間、100μg/mlのヒトIgG Fcでブロックした。次いで、この細胞を、10μg/mlの種々のビオチン化Fc融合タンパク質(または適切なコントロール)と共に、氷上で30分間インキュベートした。Fc融合タンパク質とのインキュベーション後、図11において示される細胞サンプルを、抗CD3抗体(PharMingen)または抗CD19抗体(Becton
Dickinson)と共に氷上で30分間インキュベートした。インキュベーション後、細胞を洗浄緩衝液(0.5%BSA含有PBS)中で2回洗浄し、次いで氷上で30分間、FITCアビジン(1:100希釈)で染色した。この細胞を、1mlの洗浄緩衝液中に再懸濁し、そしてFACS Starで分析した。
B7−Lポリペプチドに対する抗体は、精製したタンパク質での免疫、または生物学的合成もしくは化学的合成によって生成したB7−Lペプチドでの免疫によって得られ得る。抗体を産生するための適切な手順としては、HudsonおよびBay,Practical Immunology(2nd ed.,Blackwell Scientific Publications)に記載される手順が挙げられる。
B7−Lポリペプチドの生物学的活性を評価するために、肝臓特異的ApoEプロモーターの制御下でB7−Lポリペプチド/Fc融合タンパク質をコードする構築物を調製する。この構築物の送達は、B7−Lポリペプチドの機能についての参考となる病理学的変化を引き起こすと予想される。同様に、βアクチンプロモーターの制御下で全長B7−Lポリペプチドを含む構築物を調製する。この構築物の送達は、遍在的な発現を生じると予想される。
プロテイナーゼK)中、55℃で一晩消化する。次いで、このサンプルを200mLのTEで希釈し、そして2mLの耳サンプルを、適切なプライマーを使用して、PCR反応において使用する。
Preamplification System(Gibco−BRL)を使用してcDNAに変換する。発現ベクター配列およびB7−Lポリペプチド導入遺伝子に対して3’側に位置する適切なプライマーを使用して、導入遺伝子転写物からのcDNA合成をプライムする。トランスジェニック創始動物およびコントロール由来の10mgの総脾臓RNAを、1mMのプライマーと共に70℃で10分間インキュベートし、そして氷上に置く。次いで、この反応物に、10mM Tris−HCl(pH8.3)、50mM KCl、2.5mM MgCl2、10mMの各dNTP、0.1mM DTT、および200UのSuperScript II逆転写酵素を補充する。42℃で50分のインキュベーション後、この反応を72℃で15分間加熱することによって停止させ、そして37℃で20分間、2UのRNase Hで消化する。次いで、サンプルを、B7−Lポリペプチドに特異的なプライマーを使用するPCRによって増幅する。
安楽死の前に、トランスジェニック動物を秤量し、イソフルラン(isofluorane)によって麻酔し、そして心臓穿刺によって採血する。サンプルを、血液学および血清化学分析に供する。X線撮影を、最終的な放血後に行う。肉眼での解剖の際に、主な内臓器官を、重量分析に供する。
Claims (10)
- T細胞増殖を阻害するための組成物であって、該組成物は、以下:
(a)B7−Lポリペプチドのシグナルペプチドおよび細胞外ドメインを含む、B7−Lポリペプチドのアミノ末端における220アミノ酸
をコードする、B7−L cDNAの660bpフラグメントによってコードされるポリペプチド;および、
(b)B7−Lポリペプチドの細胞外ドメインのアミノ酸配列
からなるポリペプチド、
からなる群より選択されるポリペプチドを含み、ここで、該ポリペプチドは、抗CD3媒介性T細胞増殖の阻害活性を有する、組成物。 - 前記ポリペプチドは、水溶性ポリマーを用いて共有結合的に修飾されている、請求項1に記載の組成物。
- 前記水溶性ポリマーは、ポリエチレングリコール(PEG)、モノメトキシポリエチレングリコール、デキストラン、セルロース、ポリ−(N−ビニルピロリドン)ポリエチレングリコール、プロピレングリコールホモポリマー、ポリプロピレンオキシド/エチレンオキシドコポリマー、ポリオキシエチル化ポリオール、およびポリビニルアルコールからなる群より選択される、請求項2に記載の組成物。
- 異種アミノ酸配列に融合された請求項1に記載のポリペプチドを含む融合ポリペプチドを含む、T細胞増殖を阻害するための組成物。
- 前記異種アミノ酸配列が、IgG定常ドメインまたはそのフラグメントである、請求項4に記載の組成物。
- 請求項1に記載のポリペプチド、および薬学的に受容可能な処方剤を含む、T細胞増殖を阻害するための組成物。
- 前記薬学的に受容可能な処方剤が、キャリア、アジュバント、溶解剤、安定剤、または抗酸化剤である、請求項6に記載の組成物。
- 前記ポリペプチドが水溶性ポリマーを用いて共有結合的に修飾された、請求項6に記載の組成物。
- 請求項8に記載の組成物であって、ここで、前記水溶性ポリマーが、ポリエチレングリコール、モノメトキシポリエチレングリコール、デキストラン、セルロース、ポリ−(N−ビニルピロリドン)ポリエチレングリコール、プロピレングリコールホモポリマー、ポリプロピレンオキシド/エチレンオキシドコポリマー、ポリオキシエチル化ポリオール、およびポリビニルアルコールからなる群より選択される、組成物。
- B7−Lポリペプチド関連の疾患、状態、または障害を、処置、予防、または改善するための組成物であって、請求項1に組成物の有効量を含む、組成物。
