核酸コンストラクトは、ヒトCMV最初期(IE)エンハンサー、プロモーター及びイントロンを含む親1012DNAワクチンプラスミドから得られる。CMV/R調節エレメントを構築するために、CMV IEイントロンの大多数を含有する1012プラスミドのSacII/HpaI断片を、HTLV−1 R領域の227bp EcoRV/HpaI断片で置換した(Seiki et al., Proc. Natl. Acad. Sci. USA 80: 3618-3622ページ, 1983年)。生じたCMV/Rプラスミドは、ヒトCMV IEエンハンサー/プロモーターを、その後にHTLV−1 R領域及びCMV IE3’イントロンの123bp断片を含有する。R領域におけるスプライス・ドナー及びCMV IE3’イントロンにおけるスプライス・アクセプターは、一対のスプライシング・シグナルとして働く。同様に、CMV/RプラスミドのCMVエンハンサー/プロモーター領域をラウス肉腫ウイルス(RSV)エンハンサー/プロモーターの381bp AflIII/HindIII断片又はマウスユビキチンB(mUB)エンハンサー/プロモーターの842bp SpeI/EcoRV断片でそれぞれ置換することにより、RSV/R及びmUB/Rプラスミドを構築した。mUBエンハンサー/プロモーターは、前に記載されている(Yew他、 Mol. Ther. 4: 75-82ページ, 2001年)。
図1に示したDNAプラスミドVRC−4401を構築するために、HXB2(GENBANK(登録商標)、寄託番号K03455)由来のgagポリタンパク質のアミノ酸配列(Pr55、アミノ酸1〜432)を、ヒト細胞中での発現のために最適化されたコドンを用いてgag遺伝子の合成バージョンを作製するために用いた。合成gag遺伝子のヌクレオチド配列はHXB2遺伝子との相同性をほとんど示さないが、しかしコードされるタンパク質は同一である。Gag(B)をコードする合成遺伝子を含むSalI/BamHI断片を、pVR1012主鎖中に同一インサートを含むプラスミドVRC3900から切り出し、上記のCMV/R主鎖のSalI/BamHIサイト中にクローニングした。予測されるVRC−4401ドメインの概要を、表2に示す。プラスミドは5886ヌクレオチド塩基対(bp)長であり、およそ3.9MDaの分子量を有する。VRC−4401の配列は、配列番号1で示される。
図2に示したDNAプラスミドVRC−4409を構築するために、NL4−3(GENBANK(登録商標)、寄託番号M19921)由来のPolポリタンパク質のアミノ酸配列(アミノ酸3〜1003)を、ヒト細胞中での発現のために最適化されたコドンを用いてpol遺伝子の合成バージョンを作製するために用いた。Polの冒頭部分で翻訳を開始させるために、メチオニンコドンを合成ポリメラーゼ遺伝子の5’末端に付加して、Pol/h遺伝子を作製した。更にプロテアーゼ(PR)突然変異をアミノ酸553に導入し(AGG→GGC又はアミノ酸R→G)、逆転写酵素(RT)突然変異をアミノ酸771に導入し(GAC→CAC又はアミノ酸D→H)、そしてインテグラーゼ(IN)突然変異をアミノ酸1209に導入した(ACT→CAT又はアミノ酸D→A)。Polを発現する遺伝子を、上記のCMV/R主鎖中に導入した。予測されるVRC−4409ドメインの概要を、表3に示す。プラスミドは7344ヌクレオチド塩基対(bp)長であり、およそ4.8MDaの分子量を有する。VRC−4409の配列は、配列番号2で示される。
図3に示したDNAプラスミドVRC−4404を構築するために、HIV−1 NY5/BRU(LAV−1)クローンpNL4−3(GENBANK(登録商標)、寄託番号M19921)由来のNefタンパク質のアミノ酸配列を、ヒト細胞中での発現のために最適化されたコドンを用いてNef遺伝子の合成バージョン(Nef/h)を作製するために用いた。ヌクレオチド配列Nef/hはウイルス遺伝子との相同性をほとんど示さないが、コードされるタンパク質は同一である。ミリストール部位(GGC−Gly、アミノ酸2〜3)を欠失させた。Nefをコードする断片を、それが元々挿入されていたpVR1012主鎖から、XbaI/BamHIで消化して、上記のCMV/R主鎖のXbaI/BamHIサイト中にクローニングした。予測されるVRC−4404ドメインの概要を、表4に示す。プラスミドは5039ヌクレオチド塩基対(bp)長であり、およそ3.3MDaの分子量を有する。VRC−4404の配列は、配列番号3で示される。
図4に示したDNAプラスミドVRC−5736を構築するために、92rw020(R5向性、GENBANK(登録商標)、寄託番号U08794)由来のエンベロープポリタンパク質(gp160)のアミノ酸配列を、ヒト細胞中での発現のために改変されたコドンを用いて遺伝子の合成バージョン(分岐群A gp145delCFI)を作製するために用いた。ヒト細胞中に典型的に見出されるコドンを用いることにより、タンパク質発現を制限するウイルスRNA構造を破壊するよう意図された配列を用いてHIV−1遺伝子を発現するプラスミドを合成的に製造した。ヌクレオチド配列R5gp145delCFIは92rw020遺伝子との相同性をほとんど示さないが、コードされるタンパク質は同一である。切頭化エンベロープポリタンパク質は、全SUタンパク質及びTMドメインを含有するが、融合ドメイン及び細胞質ドメインを欠く。Heptad(H)1、Heptad2及びそれらの間空(IS)は、オリゴマー形成に関与する。アミノ酸486〜519からなる融合及び切断(F/CL)ドメインは欠失している。Heptad(H)1及び2間のアミノ酸576〜604からなる間空(IS)は欠失している。XbaI(ATGから18ヌクレオチド上流)〜BamH1(ATGから1912ヌクレオチド下流)断片は、5’末端のポリリンカー、コザック配列及びATGを含有しており、これを上記のCMV/R主鎖のXbaI−BamHIサイト中にクローニングした。予測されるEnv AのVRC−5736ドメインの概要を、表5に示す。プラスミドは6305ヌクレオチド塩基対(bp)長であり、およそ4.2MDaの分子量を有する。VRC−5736の配列は、配列番号4で示される。
図5に示したDNAプラスミドVRC−5737を構築するために、HXB2(X4向性、GENBANK(登録商標)、寄託番号K03455)由来のエンベロープポリタンパク質(gp160)のアミノ酸配列を、ヒト細胞中での発現のために最適化されたコドンを用いて遺伝子の合成バージョン(X4gp160/h)を作製するために用いた。ヌクレオチド配列X4gp160/hはHXB2遺伝子との相同性をほとんど示さないが、コードされるタンパク質は同一で、以下のアミノ酸置換を有する:F53L、N94D、K192S、I215N、A224T、A346D及びP470L。エンベロープタンパク質(R5gp160/h)のR5向性バージョンを産生するために、X4gp160/h(VRC3300)由来のHIV−1エンベロープポリタンパク質のアミノ酸275〜361をコードする領域を、HIV−1のBaL系統(GENBANK(登録商標)、寄託番号M68893、この場合もヒトの好ましいコドンを用いる)由来の対応する領域で置換した。pR5gp160/h(VRC3000)由来のエンベロープタンパク質遺伝子の全長R5向性バージョンを、アミノ酸704に関するコドンの後で終結させた。切頭化エンベロープポリタンパク質(gp145)は、全SUタンパク質及び融合ドメインを含むTMタンパク質の一部、膜貫通ドメイン、及びオリゴマー形成のために重要な領域を含有する。Heptad(H)1、Heptad2及びそれらの間空(IS)は、オリゴマー形成に関与する。アミノ酸503〜536の融合及び切断(F/CL)ドメインは欠失している。Heptad(H)1及び2間のアミノ酸593〜620からなる間空(IS)は欠失している。発現ベクター主鎖は、上記のCMV/Rである。予測されるVRC−5737ドメインの概要を、表6に示す。プラスミドは6338ヌクレオチド塩基対(bp)長であり、およそ4.2MDaの分子量を有する。VRC−5737の配列は、配列番号5で示される。

CMV/R転写調節エレメントを含むプラスミド由来の抗原発現を評価するために、3T3細胞を上記の発現ベクターでトランスフェクトして、gp145ΔCFI発現をウエスタンブロットにより測定した。ネズミ繊維芽細胞3T3細胞を、リン酸カルシウムを用いて6−ウェルプレート中で、HIV−1 Env gp145ΔCFI(9)を発現する0.5μgの親1012(CMV)、CMV/R、RSV、RSV/R、mUB及びmUB/R DNAワクチンでトランスフェクトした。トランスフェクションの24時間後に、細胞を採取し、プロテアーゼ阻害剤を含む50mMのHEPES、150mMのNaCl、1%NP−40中に溶解した。10μgの総タンパク質をSDS−PAGEにより電気泳動し、gp145の発現をウエスタンブロット分析により評価した。ヒトHIV−IgGの5000倍希釈液を一次抗体として利用し、HRP標識ヤギ抗ヒトIgGの5000倍希釈液を二次抗体として利用した。ブロットを、ECLウエスタンブロット現像系(Amersham Biosciences, ピスカタウェイ, ニュージャージー州)を用いて現像した。
DNAプラスミドワクチンVRC−HIVDNA016−00−VPを含むプラスミドコンストラクト、並びに組換えアデノウイルスベクターワクチンVRC−HIVADV014−00−VPを用いたDNAプラスミド初回刺激/アデノウイルスベクター追加免疫投与で、マウス及び非ヒト霊長類において、非臨床免疫原性試験を実行した。免疫した動物からの末梢血単核球(PBMC)によるIFN−γの産生を定量的に測定するインターフェロンガンマ(IFN−γ)ELISPOTアッセイにより、これらの非臨床的免疫原性試験において細胞性免疫応答を試験した。HIV−1抗原(ワクチン中で発現されるタ
ンパク質の長さに及ぶ一連の短い重複ペプチド)にin vitroで細胞を曝露した。抗原刺激を受けたTリンパ球により産生されるIFN−γをアッセイプレートを被覆する抗体と結合させて、アルカリ性ホスファターゼ標識読出しシステムを用いてスポット形成細胞(SFC)として比色計で計数することができる。結果を、SFC/106PBMCとして表す。
gag/pol及び分岐群A、B及びC Envをコードする4つのアデノウイルスベクターから成るGMP等級VRC−HIVADV014−00−VP(ロット番号026−03017、PCT出願番号PCT/US2005/12291、2005年4月12日出願)で構成される。表8は、プラスミドの概要を示す。
抗原発現の増強がin vivoにおけるこれら新規のDNAワクチンの免疫原性を改善する可能性を調べるために、50μgの親1012DNAワクチン又はHIV−1 Env gp145ΔCFIを発現するCMV/R、RSV/R、mUB、又はmUB/R
DNAワクチンを用いてBalb/cマウス(N=5/群)を免疫した。0、2及び6週目に、マウスを3回免疫した。最終免疫の10日後に、IFN−γ及びTNF−α細胞内サイトカイン染色(ICS)アッセイにより、脾臓細胞のEnv特異的細胞性免疫応答を評価した。CMV/R DNAワクチンは、同一抗原を発現する親1012DNAワクチンと比較して、約2倍高いCD4+(p=0.15)及びCD8+(p=0.043)Tリンパ球応答を誘導した(図9)。逆に、RSV/R、mUB及びmUB/R DNAワクチンはCD8+免疫応答の増強を誘導しなかったが、これは、HTLV−1 RエレメントがCMVプロモーターに関連して、免疫原性を選択的に改善したことを示唆する。
ンパク質を発現するこれらのDNAワクチンを用いて、マウス(N=8/群)を免疫した。0及び6週目に、マウスを2回免疫し、初回免疫又は追加免疫の3週間後に採取した脾臓細胞を用いて、IFN−γ ELISPOTアッセイにより、細胞性免疫応答を評価した。標準生理食塩水中で希釈されるプラスミドを以下の投与計画を用いて、BALB/c雌マウス(8匹/群)の群を免疫した:
分岐群B g−p−n(1012):VRC−4306(50μg/動物);このプラスミドは融合タンパク質としてGag−Pol−Nefを発現し、4−プラスミドワクチンVRC−HIVDNA009−00−VP(BB−IND 10681)中に含入される;
分岐群B g−p−n(CMV/R):VRC−4400(50μg/動物);このプラスミドは融合タンパク質としてGag−Pol−Nefを発現する。
Gag、Pol及びNefペプチドプール(VRC, ベテスダ, ミズーリ州)を用いて、応答を測定した。18時間インキュベーションを行った後、プレートを、0.25%Tween−20を含むダルベッコPBSで9回、そして蒸留水で1回洗浄した。次に5μg/mlのビオチニル化ラット抗マウスIFN−γ(Pharmingen, サンディエゴ, カリフォルニア州)を75μl/ウェルとなるよう加え、プレートを2時間インキュベートし、コールター洗浄液(Coulter Corporation, マイアミ, フロリダ州)で6回洗浄し、ストレプトアビジン−AP(Southern Biotechnology Associates, バーミンガム, アラバマ州)の500倍希釈液を加えて2時間インキュベートした。コールター洗浄液で5回、PBSで1回洗浄後、プレートをNBT/BCIP色原体(Pierce, ロックフォード, イリノイ州)で現像して、水道水で洗浄することにより停止させ、乾燥し、ELISPOTリーダー(Hitech Instruments, エッジモント, ペンシルベニア州)を用いて読み取った。
1(GraphPad Software, Inc., 2004年)を用いて、統計的分析を実施した。両側ノンパラメトリック・マン・ホイットニー検定により、動物の群間の平均細胞性免疫応答を比較した。すべての場合に、0.05未満のp値は有意であるとみなされた。
カニクイザルにおいて、親1012DNAワクチンの免疫原性を、多HIV−1抗原を発現するCMV/R DNAワクチンと比較した。成体カニクイザルの2つの群(N=6/群)を、1012の4−プラスミド、又は分岐群A、B及びC由来のHIV−1 Env gp145ΔCFIを発現するCMV/R DNAワクチン並びに分岐群BからのGag−Pol−Nef融合タンパク質の1:1:1:3比の混合物を用いて免疫した。この多分岐群多価DNAワクチンは以前に記載されており、現在、臨床試験で評価されつつある(VRC−HIVDNA009−00−VP;PCT公開番号WO/05034992)。Gag−Pol−Nef融合タンパク質を、別個のプラスミド上でコードされる別個の遺伝子に分離するとこれらの抗原(VRC−HIVDNA016−00−VP)に対する免疫応答が更に増大されるか否かを調べるために、第3の群のサルを加えた。この第3の群のサルには、分岐群A、B及びC由来のHIV−1 Env gp145、並びに分岐群B由来の別個のGag、Pol及びNefタンパク質をコードするCMV/R DNAワクチンを1:1:1:1:1:1の比で含む6−プラスミド混合物を投与した。全サルには、0、4及び8週目に、8mgの総DNAワクチンで3回免疫した。

n−20を除去し、ペプチドプール及び反応容積100μl中の2×105PBMCと共にインキュベートし、三重反復試験を行った。37℃で18時間インキュベーションした後、プレートを、D−PBS/Tweenで9回、蒸留水で1回洗浄した。次に2μg/mlのビオチニル化ウサギ抗ヒトIFN−γ(Biosource)と共にプレートを室温で2時間インキュベートし、コールター洗浄液(Beckman-Coulter)で6回洗浄し、ストレプトアビジン−AP(Southern Biotechnology)の500倍希釈液で2.5時間インキュベートした。コールター洗浄液で5回、PBSで1回洗浄後、プレートをNBT/BCIP色原体(Pierce)で現像し、水道水で洗浄することにより停止させ、乾燥して、ELISPOTリーダー(Hitech Instruments)を用いて読み取った。スポット形成細胞(SFC)/106PBMCを算定した。培地バックグラウンドは、全て<15スポット形成細胞/106PBMCであった。

する唯一の有害事象は、等級3の汎発性蕁麻疹であった。該被験体は初回ワクチン接種後約2週間での抗ヒスタミン薬の開始を報告したが、しかしその時点で、理由はラテックスアレルギーと報告した。ロールオーバー追加免疫試験、VRC010に関してスクリーニングする間に、抗ヒスタミン薬の供給が尽きると、2回目のワクチン接種の時期辺りで被験体は汎発性蕁麻疹を発症することが判明した。被験体は、抗ヒスタミン薬により良好に制御される慢性蕁麻疹を有する。評価は進行中である。病因は不明であるが、この時点で、慢性蕁麻疹は試験ワクチンにおそらくは関連すると査定される。今日まで、おそらくはワクチンが関与する2つの中等度(等級2)の有害事象が認められている。これらは、1人の被験体(この被験体は症候の再発を伴わずに3回目のワクチン接種を受けた)において、2回目のワクチン接種後13日目に始まり、2日間持続する間欠性眩暈であり、そして別の被験体では無症候性低血糖症で、最初に気づいたのは3回目のワクチン接種後14日目の追跡調査来院時であった。追跡調査できなかった被験体の最終安全性評価は、2回目のワクチン接種後1日目の電話によるものであった;その時点で、被験体はワクチン接種による副作用を全く報告しなかった。
大多数がGagに対して検出可能応答を有し、Nefに対して応答するものも存在する。
中和抗体の誘導における異なる遺伝子配列の役割を示すために、2つの異なる分岐群由来のウイルスエンベロープの、異なる領域を有するキメラ抗原ポリペプチドを発現する核酸コンストラクトを産生した。分岐群C Envポリペプチド及び分岐群B Envポリペプチドの異なる部分をコードする核酸コンストラクトを分析し、分岐群C Envポリペプチドと比較した。分岐群B主鎖上への近位25%の分岐群Cの転位は、種々の分岐群B単離物に対する中和の潜在力及び幅の増大、並びに分岐群C単離物の中和の改善を示した。分岐群C Envによる分岐群B Envの遠位領域の置換は、分岐群B単離物に対する中和改善を生じたが、これは、分岐群B単離物中のV3を含有する領域が種々の多様なウイルス単離物を抑制する能力に寄与ことを示す。これらの核酸コンストラクトは、配列番号7〜15により表わされる。従って、開示される組成物の特定の実施形態は、多分岐群を併せ持つキメラEnvポリペプチドをコードするコンストラクトを含む。

Claims (5)
- ヒトにおいてHIVに対する免疫応答を誘導するための、第1の組成物および第2の組成物を含む医薬であって、
該第1の組成物は、
(a)分岐群A HIV Envタンパク質をコードする核酸を含むプラスミド、
(b)分岐群B HIV Envタンパク質をコードする核酸を含むプラスミド、
(c)分岐群C HIV Envタンパク質をコードする核酸を含むプラスミド、
(d)分岐群B HIV Gagタンパク質をコードする核酸を含むプラスミド、
(e)分岐群B HIV Polタンパク質をコードする核酸を含むプラスミド、
(f)分岐群B HIV Nefタンパク質をコードする核酸を含むプラスミド、および(g)薬学的に許容可能な担体を含み、
該第2の組成物は、
(a)分岐群A HIV Envタンパク質をコードする核酸を含むアデノウイルスベクター、
(b)分岐群B HIV Envタンパク質をコードする核酸を含むアデノウイルスベクター、
(c)分岐群C HIV Envタンパク質をコードする核酸を含むアデノウイルスベクター、
(d)分岐群B HIV Gag−Pol融合タンパク質をコードする核酸を含むアデノウイルスベクター、および
(e)薬学的に許容可能な担体を含み、
該第1の組成物は、該第2の組成物の投与の前に該ヒトに投与される、医薬。 - 前記第1の組成物に含まれるプラスミドによってコードされる分岐群A、BおよびCのEnvタンパク質の各々は、融合ドメインおよび切断ドメイン並びにHeptad(H)1及び2間の間空(IS)を欠くgp145タンパク質である、請求項1に記載の組成物。
- HIV Env、Gag、Polおよび/またはNefタンパク質をコードする核酸配列
