Description
又、本発明の非経口的投与用製剤は薬物含有徐放性微粒子を有効成分とすることからなる。
又、非経口的投与用製剤が皮下注射剤または筋肉注射剤であることが好適である。
又、多孔性アパタイト誘導体がヒドロキシアパタイトの構成成分であるカルシウムの一部を組成として製造時に亜鉛に置換した多孔性アパタイト誘導体であることが好適である。
又、多孔性アパタイト誘導体の亜鉛置換率または含有率が0.1〜2.0であることが好適である。
さらに、その製造法において、水溶性2価金属化合物を含む水溶液を添加せず、したがって水溶性2価金属化合物を該多孔性アパタイト誘導体に浸潤させない場合もある。
又、同様に、多孔性アパタイト誘導体がヒドロキシアパタイトの構成部分であるカルシウムの一部を製造時に亜鉛に置換した多孔性アパタイト誘導体であること、多孔性アパタイト誘導体の亜鉛の置換または含有率が0.1〜2.0であること、水溶性2価金属化合物が塩化亜鉛または酢酸亜鉛であることが好適である。
表1中Betaは、リン酸ベタメサゾンを意味し、Hydは、リン酸ヒドロコルチゾンを意味する。また、ステロイドとは、リン酸ベタメタゾンまたはリン酸ヒドロキシコーチゾンを意味する。
いずれのシステムでも酢酸亜鉛の添加によって溶出は抑制され、無添加に比較してより長時間にわたる徐放性をしめした。HAPとHAP−Zn−0.5とを比較すると、明らかにHAP−Zn−0.5の方が溶出は遅延され、より長時間の徐放性を示した。
Claims (7)
- ヒト成長ホルモンを除く薬物が、
ヒドロキシアパタイトのカルシウム原子の一部を製造時に亜鉛に置換した、亜鉛含有多孔性ヒドロキシアパタイトに吸着されていることを特徴とする薬物含有徐放性微粒子。 - 請求項1に記載の薬物含有徐放性微粒子、および塩化亜鉛、酢酸亜鉛又は塩化カルシウムからなることを特徴とする薬物含有徐放性微粒子。
- 亜鉛含有多孔性ヒドロキシアパタイトが含有する亜鉛の原子数は、ヒドロキシアパタイトのカルシウム10原子に対して0.1〜2.0であることを特徴とする請求項1又は2に記載の薬物含有徐放性微粒子。
- 請求の範囲1ないし3に記載された薬物含有徐放性微粒子を有効成分として含有することを特徴とする非経口的投与用製剤。
- 非経口的投与用製剤が皮下注射剤または筋肉注射剤のいずれかであることを特徴とする請求項4に記載の製剤。
- ヒドロキシアパタイトのカルシウム原子の一部を製造時に亜鉛に置換した、亜鉛含有多孔性ヒドロキシアパタイトの微粒子を、ヒト成長ホルモンを除く薬物含有の水溶液中に攪拌・分散し、水溶液が該亜鉛含有多孔性ヒドロキシアパタイト中に浸潤したのち、塩化亜鉛、酢酸亜鉛又は塩化カルシウムを含む水溶液を添加して該亜鉛含有多孔性ヒドロキシアパタイトに浸潤させ、その後安定剤などの添加物を加えて、凍結乾燥または真空乾燥することにより製造されることを特徴とする薬物含有徐放性微粒子の製造法。
- 亜鉛含有多孔性ヒドロキシアパタイトの亜鉛の置換または含有率が0.1〜2.0であることを特徴とする請求の範囲第6項記載の製造法。
