Description
実施例1では、ClpL及びClpPの合成に対する熱ショックの影響を研究し、重要な肺炎球菌毒性遺伝子の試験管内発現に対するclpL−及びclpP−突然変異の影響を評価した。また、肺炎球菌の毒性に対するclpL−及びclpP−突然変異の影響をマウス腹腔内試験感染モデルで評価した。
i)細菌菌株、成長条件及び形質転換
本研究に用いられた細菌菌株は表1の通りである。Rx−1の誘導体(莢膜のない非病原性肺炎球菌)である肺炎球菌CP1200(前記Choi, I. H. et al., 1999)を本研究に使用し、これを37℃でカシトン−トリプトン(Casitone-Tryptone::CAT)基本培地で指数成長期の中間段階まで培養した:CAT基本培地1Lは、10gの酵素カゼイン加水分解物(enzymatic casein hydrolysate)(Difco Laboratories, USA)、5gのトリプトファン (Difco Laboratories)、1gの酵母抽出物(yeast extract)(Difco Laboratories)、5gのNaCl、5mgのコリン(choline)(Sigma, USA)、0.2%のグルコース(glucose)(Sigma, USA)、16.6mMのリン酸2カリウム(dipotassium phosphate)(Sigma, USA)を含有した。CATブロスにリットル当り147mgのCaCl2及び2gの牛胎児血清(分画V;Sigma)を添加して完全な形質転換培地を製造した。コンピテンス(Competence)をコンピテンス特異的ペプチドの添加によって調節し、従来の文献(Havarstein, L. S. et al., 1995, An unmodified heptadecapeptide pheromone induces competence for genetic transformation in Streptococcus pneumoniae. Proc. Natl. Acad. Sci. U.S.A. 92:11140-11144)に記載されているように、培養培地で細胞をDNAに露出させた後、収得したノボビオシン抵抗性形質転換体として定量化した。
肺炎球菌Dnak及びGroELに対するHSP抗体の生成は、従来の文献(前記Choi et al., 1999)に記載されている。ClpL及びClpPに対する抗体を製造するために、肺炎球菌CP1200の指数成長期の培養物を42℃で30分間培養させ、細胞を超音波処理させ、蛋白質をドデシル硫酸ナトリウム10%ポリアクリルアミドゲル電気泳動(SDS−PAGE)によって分離し、クマシーブリリアントブルー(Commasie Brilliant Blue)で明るく染色させた。84kDa及び21kDaの蛋白質バンドを切り出し、電気溶離させた。食塩水1ml当り蛋白質100μgをフロイント不完全アジュバント1mlと混合した。その後、この混合物をウサギに筋肉内及び皮下注射した。2つの追加抗原投与量を2週間隔で投与し、抗血清を6週後に採取した。CbpA、肺炎球菌表面抗原A(PsaA)及びPlyに対する血清の製造は、従来の文献(前記Ogunniyi, A. D. et al., 2000)に記載の方法と本質的に同一であった。
蛋白質標識実験のために、細胞をCAT培地でA550(550nmにおける吸光度)=0.2となるまで成長させた後、2mlの分取液に分けた。その後、細胞を収集し、予め加温した新しい低含量メチオニン標識培地に再懸濁させ、30℃で10分間平衡化させた。10μCiの[35S]-メチオニン(1000Ci/mmol, Amersham)を添加し、培養物の温度を熱ショックのために42℃にした。細胞を集め、20μlの溶解緩衝液を(5mM Tris[pH8.0]、30mMのエチレンジアミンテトラ酢酸[EDTA]、0.1%のトリトンX−100、0.025%[W/v]のフェニルメタンスルホニルフルオリド[PMSF]、1mMのジチオスレイトール)に再懸濁させた後、従来の文献(前記Choi et al., 1999)に記載されているように、超音波(氷上で)によって完全溶解させた。SDS−PAGE(10または15%のポリアクリルアミドゲル)を文献(Laemmli et al., 1970, Cleavage of structural proteins during the assembly of the head of bacteriophage T4. Nature 227:680-685)に記載されているように行い、蛋白質をクマシーブリリアントブルー染色によって可視化した。ポリアクリルアミドゲルを放射線敏感性映像化プレートに数日間露出させて映像を得た。放射線写真映像データを映像化分析システム(Fujix Bio-imaging Analyzer BAS2500, Fuji Photo Film Co.)を用いて定量化した。
10%のSDS−PAGEによって分離された蛋白質をポリビニルリデンジフルオリド(PVDF)膜上にエレクトロブロットした後、1次抗体として肺炎球菌の熱ショック蛋白質に対して生成されたウサギ抗血清の1:100の希釈液、または肺炎球菌の毒性蛋白質(CbpA、PsaA及びPly)に対して生成されたマウス抗血清の1:5,000の希釈液と反応させた。2次抗体は、ホースラディシュ・ペルオキシダーゼ(Sigma)またはアルカリ性ホスファターゼ(Bio-Rad)にコンジュゲートされたヤギ抗−ウサギまたはヤギ抗−マウスIgGの1:2,000の希釈液であった。
従来の文献(Ogunniyi, A. D. et al., 2002, The genes encoding virulence-associated proteins and the capsule of Streptococcus pneumoniae are upregulated and differentially expressed in vivo. Microbiol. 148:2045-2053)に記載されているように、高温酸フェノール法を用いて全体RNAを抽出した。ply、psaA、cbpA及びcps2Aに対するmRNAの水準を、Promega Access RT-PCR System(Promega Biotech, Cat.# A1250)を用いる1段階の実時間逆転写法(RT−PCR)によって定量化した。様々なRT−PCR検定に用いられる特異的プライマーは、文献(前記Ogunniyi et al., 2002)に記載されており、反応当り50nMの最終濃度で使用された。内部対照として、16S rRNAに対して特異的なプライマー(正方向、5'-GGT GAG TAA CGC GTA GGT AA-3': SEQ ID NO. 1;逆方向、5'-ACG ATC CGA AAA CCT TCT TC: SEQ ID NO. 2, Bioneer Co.)を使用した。別個のRT−PCR反応(構成プライマーのみが異なる)を、Sybr®Green(Molecular Probes)が最終濃度1:50,000で添加されたマスター混合物から設定した(氷上で)。混合物を氷上で各上流及び下流プライマーを含有するチューブに分取し、徐々に攪拌して完全混合させた。
肺炎球菌のclpLの欠失−挿入突然変異(ΔclpL::ermB)を作るために、860−bp ermBカセット(フランスのトルーズCNRSのClaverys博士から入手、Vasseghi, H., and J. P. Claverys. 1983. Amplification of a chimeric plasmid carrying an erythromycin-resistance determinant introduced into the genome of Streptococcus pneumoniae. Gene 21:285-292)をエリスロマイシン抵抗性大腸菌染色体DNAからのprs3(5'-CCG GGC CCA AAA TTT GTT TGA T-3' : SEQ ID No. 3)及びprs4(5'-AGT CGG CAG CGA CTC ATA GAA T-3': SEQ ID No. 4)を用いて増幅させ、clpLを分解(disruption)させることに使用した。clpL及びermBの5′末端の一部を含有する410bp断片(clpL-up)をCP1200 DNAからのhlp3(5'-CGG TAC CAT GAA CAA TAA TTT TAA C-3': SEQ ID No. 5)及びhlp1 (5'-ATC AAA CAA ATT TTG GGC CCG GTC AGA TGT TTC TTG AAT TTC C-3': SEQ ID No. 6)を用いて増幅させた。下流clpL配列及びermBの3′末端の一部を含有する300bp断片(clpL-down)をCP1200 DNAからのhlp2(5'-ATT CTA TGA GTC GCT GCC GAC TGT TCT AGA TGA TGG TCG TTT G-3': SEQ ID No. 7) 及びhlp4(5'-GGC CGA GCT CTT AGA CTT TCT CAC GAA TAA C-3': SEQ ID No. 8)を用いて増幅させた。3つのPCR生成物を、hlp3及びhlp4を用いるPCRに対する混合テンプレートとして用い、ermB遺伝子によって置換されたclpLの1,301bp欠失部分(Genebank AE008411塩基配列6374番目〜7674番目)を有する1.6kb断片を生産した。
大腸菌でHis6−タグClpLを過発現させるために、clpL ORFをCP1200DNAのhlp3及びhlp4を用いて増幅させた。断片をKphI及びSacIで分解させ、pET30(a)(Novagen)KpnI及びSacIの部位にクローニングさせてプラスミドpKHY004(図2)を生成させた。His6−タグ蛋白質を大腸菌内で発現させ、DEAE-Sepharose fast flowTMクロマトグラフィ(Amersham Pharmacia)によって0.1〜0.4MのNaCl勾配で溶離させた。ClpLを含有する分画をプーリングし、製造者(Novagen)の指示をやや変形させた方法によってニッケル−ニトリロ酢酸カラム上で精製した。結合したHis6−タグ蛋白質を40mMのイミダゾール緩衝液で洗浄し、0.4Mのイミダゾール緩衝液(pH7.9)で溶離させ、20mMのTris−HCl(pH7.8)、5mMのMgCl2に対して透析させた。蛋白質の純度はSDS−PAGEで判定して>95%であり、前記蛋白質をクマシーブリリアントブルーR250で染色した(データは表示せず)。
従来の文献(Kudlicki, W. et al., 1997, Renaturation of rhodanese by translational elongation factor (EF) Tu. Protein refolding by EF-Tu flexing. J. Biol. Chem. 272:32206-32210)に記載の方法を次のように変形させた方法によってClpLのシャペロン活性を決定した。Rhodanese(Sigma, USA)(9μM)を1mMのβ−メルカプトエタノール及び8Mのウレアを含有する200mMのリン酸カリウム緩衝液(pH7.6)で1時間25℃で変性させた。8Mのウレア中の変性された酵素2.5μlを希釈させ、50mMのTris−HCl(pH7.8)、200mMのβ−メルカプトエタノール、5mMのチオ硫酸ナトリウム、10mMのMgCl2、10mMのKClを含有する最終容積250μlの溶液にして自発的なClpL補助リフォールディングを開始した。リフォールディング反応において、Rhodaneseの最終濃度は90nMであった。リフォールディング反応は25℃で30分間行った。Rhodaneseの天然形態へのリフォールディングによってClpLのシャペロン活性を測定した。Rhodaneseの酵素活性は、文献(Sorbo, B. H. et al., 1953, Crystalline rhodanese. I. Purification and physicochemical examination. Acta Chem. Scand. 7:1129-1136)に記載の方式で決定した。
毒性が非常に強い莢膜タイプ2菌株(D39)及びその同種遺伝子型clpP―及びclpL−突然変異体(それぞれHYK302及びHYK304)で腹腔内(i.p.)試験感染を行い、肺炎球菌の毒性に及ぼすclpLまたはclpPの突然変異の影響を評価した。細菌を、10%[vol/vol]の馬血清を加えた脳心臓注入寒天(Difco Laboratories, USA)またはTH(Todd Hewitt)寒天(Difco Laboratories, USA)(必要に応じてエリスロマイシン補充)上で一晩中37℃で培養した後、血清ブロス(10%[vol/vol]の馬血清を加えた脳心臓注入ブロス(Difco Laboratories, USA)またはTH (Todd Hewitt)ブロス(Difco Laboratories, USA))で3時間37℃で成長させて約108CFU/mlの細菌培養物を収得した(前記Ogunniyi, A. D. et al., 2000)。次に、各細菌培養物を血清ブロスで希釈させて約106CFU/mlとし、10匹のBALB/cマウス群を体積0.1mlのD39、HYK302またはHYK304で腹腔内感染させた。試験感染したマウスの生存を、最初5日間は1日4回、次の5日間は1日2回、試験感染後の21日目までは毎日モニタリングした。
従来の文献(前記Hanahan, D., 1983)に記載の方法をやや変形させた方法によって溶血活性を決定した。THYブロスで指数成長期の初期−中期段階(A600=0.05〜0.1)まで成長させた肺炎球菌(D39、HYK302、HYK304)を、4℃で10分間3900×gの遠心分離によって収集し、リン酸塩緩衝食塩水に再懸濁させた。デオキシコール酸ナトリウムを最終濃度0.1%で添加した後、37℃で10分間培養した。サンプルを遠心分離した後、上澄液を回収し、連続的に希釈させた。96ウェルマイクロタイタープレートに1.5%の洗浄されたヒト赤血球の等量と共に培養して溶血活性を決定した。溶血力価は540nmで赤血球50%が溶解される推定希釈度の逆数で決定した。
対応標本または独立標本スチューデントt検定(paired or unpaired Student's t test)を用いて統計的分析を行った。提示されたデータは2つ〜4つの独立実験に対する平均±標準偏差である。群間の平均生存時間の差はマン・ホイットニーのU検定(両側)(2-tailed)によって分析し、群間の全体的な生存率の差はフィッシャーの正確検定 (Fisher Exact test)によって分析した。
i)ClpLの特性究明
従来、84kDa HSPはN−末端アミノ酸配列化によってClpLと確認された(前記Choi et al., 1999)。Clpファミリーの一員は、2つの高度に保存されたATP−結合領域(ATP−1及びATP-2)を含有し、前記各領域はアデニンヌクレオチド結合に対するコンセンサス配列を含有する(Gottesman, S. et al., 1990, Conservation of the regulatory subunit for the Clp ATP-dependent protease in prokaryotes and eukaryotes. Proc. Natl. Acad. Sci. U.S.A. 87:3513-3517)。推定肺炎球菌ClpLが実際Clpファミリーの一員であることを確認するために、N−末端アミノ酸配列(5'-GAT GAA YAA YAA YTT YAA YAA YTT YAA-3': SEQ ID NO. 13)からのオリゴーヌクレオチドとClpファミリーの最もよく保存されたアミノ酸配列(PTGVGKT)であるClpの一員に対する第2ATP結合部位(5'-GTY TTN CCN CAN CCN GYN GG-3'、ここでY=TまたはC, N=A, C, Gまたは T: SEQ ID NO. 14)をCP1200染色体DNAのPCR増幅に使用した。
それぞれ分子量84kDa、73kDa及び65kDaの主なHSPであるClpL、DnaK及びGroELは、熱ショック後に相応する肺炎球菌蛋白質のN−末端アミノ酸配列化によって確認された。HSP発現の協同的または独立的な調節に対しては決定されたことがなかったため、本発明者は、[35S]−メチオニンでパルス標識化してHSP合成の動力学を調査した。熱ショック蛋白質の誘導動力学を決定するために、30℃の指数成長期のCP1200細胞(A550=0.2)に温度を42℃に変化させて熱ショックを加えた後、[35S]−メチオニンで10分間パルス標識した。培養物2mlを取って細胞を溶解緩衝液中で超音波によって溶解させた後、細胞溶解物をSDS−PAGEで分析し、蛋白質バンドを自動放射線写真法によって可視化させた。その結果、HSPの誘導は、温度上昇変化10分後に最高に達した後、基本水準に急激に減少した(図4a)。細胞を42℃で10分間培養した後、ClpL、DnaK及びGroELの合成は、対照に比べてそれぞれ11.3±0.8、5.0±0.3及び2.7±0.2倍に増加した。GroELバンドは、近い蛋白質バンドと非常に類似であったが、自動放射線写真を高倍率で拡大した結果、GroELが誘導されたことが明確であった(図4b)。
clpL−及びclpP−突然変異体を構成するために、ΔclpL::ermBまたはΔclpP::ermB挿入部分を含有するDNA断片をPCRによって増幅させ、材料及び方法編で記載されているように、形質転換によって染色体内に挿入させた。挿入突然変異をPCR及び免疫ブロット分析によって確認し、ClpLまたはClpPの不在をそれぞれ確認した。D39及びその同種遺伝子型clpL−(HYK304)及びclpP―(HYK302)突然変異体を550nmにおける吸光度0.1まで培養した。次に、温度を37℃から43℃に変化させ、培養物を表示時間で培養し、その結果を図6に示した。ΔclpL::ermBを保有するD39誘導体HYK304の成長率は、30℃で母菌株の成長率と類似であったが、37℃では倍増時間が母菌株で約40分であったことに比べて55分であって、さらに遅く成長した(図6a〜図6c)。したがって、ClpLは30℃及び37℃で肺炎球菌の成長に必須的でないものと判断される。
ClpL及びClpPが同一のCtsRによって調節されるものと判断され、ClpPはCtsRの分解と関連があるという従来の研究結果より、ClpLはClpPによって調節できるものと予測された。このような可能性を調査するために、本発明者は、CP1200またはそのclpLまたはclpP陰性突然変異体を用いてClpL及びClpPの量を決定した。指数成長期の肺炎球菌CP1200(A550=0.3)及びその同種遺伝子型clpL−及びclpP−誘導体に42℃で30分間熱ショックを加えた。培養物3mlから得た蛋白質に対して抗ClpLまたはClpPポリクロナール血清を用いて免疫ブロット分析を行った。野生型(CP1200)及びclpL−突然変異体(HYK1)において、ClpPは30℃で検出されたが、細胞に30分間熱ショックを加えた後、ClpPの量は図7a及び図7bに示すように限界的に増加した。ClpLが誘導されたが、誘導されていない培養物中のClpLの量は野生型よりclpP―突然変異体(HYK2)で一層多く、これはClpPがClpLの発現を抑制することを示唆する(図7a及び図7b)。
HSPが分泌を促進し且つ蛋白質の適当なフォールディング及び位置移動を助けるため(Craig, E. A. et al., 1993, Heat shock proteins: Molecular chaperones of protein biogenesis. Microbiol. Rev. 57:402-414)、肺炎球菌におけるClpLのシャペロン活性を調査した。シャペロン活性を定量的に測定するために、変性された蛋白質を天然の形態にリフォールディングさせる方法を使用した(Mendoza, J. A. et al., 1991, Unassisted refolding of urea unfolded rhodanese. J. Biol. Chem. 266:13587-13591)。Rhodaneseの触媒されていないRhodaneseリフォールディングは相対的に遅く起こり(前記Mendoza et al., 1991)、Rhodaneseの活性は簡単且つ敏感な検定によって決定できるため、変性されたRhodaneseのリフォールディングは蛋白質フォールディングの研究に幅広く用いられてきた。ヒスチジンタグClpL(pKHY004)(図2)を大腸菌で過発現させ、精製させ、リフォールディング活性の決定に使用した。試験条件の下で、変性されたRhodaneseは、従来の文献(前記Craig et al., 1993)に開示されているように、天然Rhodanese活性の只2.8〜7.7%のみを示した。これは、変性されたRhodaneseの自発的リフォールディングが8Mのウレア溶液から100倍希釈された場合に非効率的に起こることを意味する。

vi)熱ショックによる毒性関連因子の発現調節
熱ショック及び飢餓を含む環境ストレスは、毒性因子の発現に影響を及ぼす可能性がある(Mekalanos, J. J. 1992. Environmental signals controlling expression of virulence genes determinants in bacteria. J. Bacteriol. 174:1-7)。したがって、莢膜菌株D39及びそのclpP−突然変異体(HYK302)及びclpL−突然変異体(HYK304)の毒性関連因子の発現に対する熱ショックの効果を、コリン結合蛋白質(CbpA)、PsaA、肺炎球菌表面蛋白質A(PspA)、Ply及びオートリシン(LytA)に対する抗体を使用する免疫ブロット分析によって決定した。指数成長期の莢膜肺炎球菌D39(A600=0.1)及びその同種遺伝子型clpP―(HYK302)及びclpL−(HYK304)誘導体に42℃で20分間熱ショックを加えた。培養物0.6mlを遠心分離し、細胞ペレットを溶解緩衝液に再懸濁させた後、3分間沸かした。
D39の毒性に及ぼすclpL−及びclpP−突然変異の影響をさらに調査するために、肺炎球菌約105CFUを腹腔内注射した後、マウスの生存時間を測定した。約105CFUのD39またはそのclpP−(HYK302)またはclpL−(HYK304)誘導体で10匹のBALB/cマウス群を感染させた。図10にその結果を示した。図10において、各データポイントはマウス1匹を示し、水平線は各群に対する中間生存時間を示す。母菌株(D39)及びclpL−突然変異体で感染させた群のマウスに対する中間生存時間はそれぞれ55時間及び60時間であり、このような差異は統計的に有意性がなかった。
本研究において、本発明者は、肺炎球菌においてATP−依存性ClpプロテアーゼAAK74513がclpL相同物であることを確認した。ClpL相同物はいろいろのグラム陽性有機体で確認されたが(L. lactis X62333; S. aureus AP003365, AP003137; S. pyogenes AE006538, AE004092; Lactobacillus rhamnosus AF323526)、グラム陰性有機体では確認されなかったので(Derre, I. Et al., 1999, ClpE, a novel type of HSP100 ATPase, is part of the CtsR heat shock regulon of Bacillus subtilis. Mol. Microbiol. 32:581-593)、ClpLはClpEと同様にグラム陽性有機体に特異的なようである。
本実施例では、ClpPが毒性を弱化させる根本的なメカニズムを調査し、ClpPによる免疫化が毒性肺炎球菌による試験感染からマウスを保護することができるか否かを評価した。
i)細菌菌株、成長条件及び形質転換
細菌菌株及びプラスミドベクターは、本研究で新しく生成された組合体と共に表4に提示されている。Rx−1の誘導体である肺炎球菌CP1200をカシトン−トリプトン(Casitone-Tryptone:CAT)基本培地で成長させた(前記Choi et al., 1999)。肺炎球菌菌株D39(タイプ2)をTHY(Todd Hewitt Yeast)ブロスで培養した。肺炎球菌形質転換体の選別のために、エリスロマイシンを0.2μg/mlの濃度で成長培地に添加した。大腸菌菌株(表4のBL21(DE3)、DH5a、XL1−Blue)をLB(Luria-Bertani)ブロスまたはLB寒天で成長させた。プラスミドを、文献(前記Hanahan, D., 1983)によって記述されているように、形質転換によって大腸菌に導入した。大腸菌形質転換体の選別のために、カナマイシン(30μg/ml)を成長培地に添加した。
ヒト肺上皮癌種A549(ATCC CCL−185)及びマウスマクロファージRAW264.7細胞株(ATCC TIB−71)をアメリカンタイプカルチャーコレクション(American Type Culture Collection)から購入し、これらを37℃で5%のCO2下に培養させた。A549細胞をグルコース4.5μg/L、10%の牛胎児血清(FBS; Gibco BRL, Gaithersburg, Md.)及びペニシリンG100U/ml、及びストレプトマイシン100μg/mlを含有するダルベッコ変法イーグル培地(DMEM)(Gibco BRL, Gaithersburg, Md)で培養させた。RAW264.7細胞の培養のために、10mMのHEPES、2mMのL−グルタミン、ペニシリンG100U/ml及びストレプトマイシン100μg/ml、及び0.2%のNaHCO3が補充されたRPMI1640培地(Gibco BRL, Gaithersburg, Md)を基本培地として用い、FBS(Gibco BRL, Gaithersburg, Md)を10%濃度で添加した。
血液寒天プレート上で一晩中成長させた肺炎球菌(A600=0.5)を150mMのTris−HCl(pH7.0)、1mMのMgSO4で再懸濁させてD39及びその同種遺伝子型(isogenic)clpP―突然変異誘導体のCPS標本を製作した。これはml当りの肺炎球菌数5×109と等量である。その1mlの分取液を10,000×gでペレット化した。0.1%(w/v)のデオキシコール酸ナトリウム(Sigma, St. Louis, MD)を添加し、15分間37℃で培養して前記細菌を溶菌させた。次のサンプルを100Uムタノリシン(Sigma)、50μgのDNaseI(Roche Applied Science, Mannheim, Germany)及び50μgのRNaseA(Roche Applied Science)とともに37℃で18時間培養した。次に、サンプルを50μgのプロテイナーゼK(Sigma)とともに56℃で4時間培養した後、−20℃で貯蔵した。次に、ポリクロナールタイプ2多糖類特異的抗血清を使用する免疫ブロットによって細胞性物質を分析した。
PspA及びPdB(Plyのトキソイド誘導体)に対する血清は、本質的に従来の文献(前記Ogunniyi, A. D. et al., 2000)の記載と同一に製作した。免疫ブロットのために細菌をTHYでA600=0.3になるまで培養し、従来の文献(Kwon, H. Y. et al., 2003, Effect of Heat Shock and Mutations in ClpL and ClpP on Virulence Gene Expression in Streptococcus pneumoniae. Submitted to Infect. Immun)の記載と同一に製作した。ドデシル硫酸ナトリウム−ポリアクリルアミドゲル電気泳動(SDS−PAGE、10%または15%ポリアクリルアミドゲル)を文献(Laemmli, U. K. 1970. Cleavage of structural proteins during the assembly of the head of bacteriophage T4. Nature 227:680-685)の記載と同一に行った。蛋白質をニトロセルロース膜上にエレクトロブロット(electroblotting)した後、PspA及びPdBに対するポリクロナールマウス血清の1:5,000希釈液と反応させた。莢膜ブロットのために、サンプルをナイロン膜上にエレクトロブロットした後、抗血清タイプ2のポリクロナールマウス血清の1:5000希釈液と反応させた。2次抗体はアルカリ性ホスファターゼにコンジュゲートされたヤギ抗−マウスIgG(Bio-Rad)の1:2000希釈液であった。
従来の文献(Lock, R. A. et al., 1996, Sequence variation in the Streptococcus pneumoniae pneumolysin gene affecting haemolytic activity and electrophoretic mobility of the toxin. Microb. Pathog. 21:71-83)に記載の方法を若干修正した方法によって溶血活性を決定した。簡単には、THYブロスで指数成長期の初期−中期段階(early-mid log phase)(A600=0.05〜0.1)まで成長させた肺炎球菌を3900×gで4℃にて10分間遠心分離させて収集し、リン酸塩緩衝食塩に再懸濁させた。デオキシコール酸ナトリウムを0.1%の最終濃度で添加した後、37℃で10分間培養させた。サンプルを遠心分離させた後、上澄液を回収して連続的に希釈させた。96ウェルマイクロタイタープレートで同一体積の1.5%洗浄されたヒト赤血球(0.001%の2−メルカプトエタノール(Merck)含有)とともに37℃で30分間培養して溶血活性を決定した。溶血力価はA540で50%の赤血球が溶解される推定希釈度の逆数で決定した。
全体RNA抽出のために培養懸濁液の1.5ml分取液を周期的に採取した。mRNA半減期を測定するために、リファンピシン(100μg/ml)を添加した。全体RNAを、従来の文献(前記Ogunniyi, A. D. et al., 2002)に記載されているように、高温酸フェノール法によって抽出した。Promega Access RT−PCR System(Promega Biotech, Cat.# A1250)を用いて1段階の実時間逆転写(RT−PCR)によってcps2A及びplyに対するmRNA水準を定量した。これらの反応に対して特異的な内部対照(16S rRNA)プライマーは、従来の文献(前記Ogunniyi et. al., 2002)に記載されている。RT−PCR反応の準備、サイクリング条件及びデータの分析は、本質的に従来の文献(前記Kwon et. al., 2003)の記載と同一であった。全ての反応をRotor−Gene 2000Real−Time cycler(Corbett Research, Australia)で行った。mRNA半減期の分析をSigmaPlot曲線当てはめプログラム(指数値の総合に対して非線形最小二乗法(non-linear least squares fitting))によって行った。2つのモデル、すなわち1相分解または2相分解モデルが提案された。各場合に、最小偏差を有するデータを当てはめさせたモデルがより有効なものとして保有された。
clpP ORF(Genebank AE008443で塩基配列5416番目〜6006番目)をCP1200 DNAから前方向及び逆方向プライマー(5'-CGA ATT CAT GAT TCC TGT AGT TAT-3': SEQ ID NO. 9、及び5'-CGA GCT CTT AGT TCA ATG AAT TGT TG-3' : SEQ ID NO. 12、プライマーはEcoRI及びSacI部位にそれぞれ挿入される)を用いてPCR増幅させた。PCR断片を同一の酵素で分解させ、pET30(a)(Novagen)の該当制限部位内にクローニングさせてプラスミドpET30(a)−clpPを作った(図1)。大腸菌BL21(DE3)菌株内で0.1mMのIPTG(イソプロピル−β−D−チオガラクトピラノシド)を用いて3時間発現を誘導した。細胞を6,000×gで10分間遠心分離させて収集した後、プロテアーゼ抑制剤のフェニルメチル−スルホニルフルオリドが最終濃度1mMで添加された溶解緩衝液(50mMのリン酸ナトリウム、pH8.0、2MのNaCl、40mMのイミダゾール)に再懸濁させた。
指数成長期の細胞を遠心分離によって収集し、スクロースによって誘導された原形質体の形成を、従来の文献(Vijayakumar, M. N. et al., 1986, Localization of competence induced proteins in Streptococcus pneumoniae. J. Bacteriol. 165:689-695)に記載の方法によって行った。細胞を1Mのスクロース緩衝液(1Mのスクロース、100mMのTris HCl pH7.6、2mMのMgCl2、1mMのPMSF)とともに30℃で1時間培養させて原形質体に転換させた。13,000×gで20分間遠心分離させて細胞壁分画(上澄液)を原形質体(ペレット)から分離させた。原形質体を19体積の低張性(hypotonic)緩衝液(100 mM Tris HCl pH 7.6, 1 mM PMSF, 1 mM EDTA)で希釈させて浸透的に溶解させた。溶解物をまず5,000×gで5分間遠心分離させて未溶解細胞を除去した後、50,000×gで30分間遠心分離させて細胞質分画(上澄液)及び細胞膜分画(ペレット)を収得した。
リンゴ酸脱水素酵素(MDH)の酵素活性を、0.5mMのオキサル酢酸塩を含有する0.15Mのリン酸カリウム(pH7.6)の中で0.2mMのNADHの340nm及び25℃での吸光度(A340=6.22/mM/cm)の減少率をモニタリングして決定した。サンプルを添加した後、反応混合物を25℃で1分40秒間培養し、340nmで吸光度を測定した。反応を開始するために基質を添加した。最初1分40秒間の反応からNADHの酸化率の初期傾斜度を用いてMDHの活性を計算した。
ヒト肺A549細胞への肺炎球菌の浸透性検定は、従来の文献(前記Vijayakumar, M. N. et al., 1986)に記載の抗生剤保護検定を変形させて行った。A549細胞を24ウェル組織培養プレートでコンフルアンス(confluence)的に成長させ、リン酸塩緩衝食塩(PBS、pH7.2)で3回洗浄した後、ウェル当り培養培地(抗生剤非含有)1mlを添加した。RタイプCP1200及びその異種遺伝子型clpP−突然変異体菌株の指数期培養物(A550=0.3、108CFU/ml)を遠心分離によってペレット化し、PBSで1回洗浄し、DMEMに再懸濁させた。単層を107個の細菌で感染させ(細菌:細胞比、10:1)、細胞単層と細菌の初期接触を、800×gで4℃にて10分間遠心分離した後、37℃で2時間培養して促進させた。10μg/mlのペニシリン及び200μg/mlのゲンタマイシンを含有する新規培地を各ウェルに添加し、このような処理が全ての細胞外細菌を死滅させるに十分であるかを確認した。その上、1時間さらに培養した後、単層をPBSで3回洗浄し、100μlの0.25%トリプシン−0.02%のEDTAで処理してプレートから細胞を取り外した後、400μlのトリトンX−100(0.025%の水溶液)を添加して溶解させた。適切な希釈液を血液寒天にプレーティングさせて生存細菌数を決定した。付着性細胞内細菌の総数を決定するために、感染した単層を前述のように洗浄した後、トリプシンで処理し、溶解させ、抗生剤処理なしで定量的にプレーティングさせた。全てのサンプルを三重(triplicate)に検定し、各検定を3回以上繰り返し行った。
細胞単層を抗生剤非含有RPMI1640培養培地(Sigma)で107CFU肺炎球菌(細胞:細胞比、10:1)にて感染させた。細菌感染のために、培養物を37℃で2時間培養させた。前記培養後に、細胞をPBSで3回洗浄し、10μg/mlのペニシリン及び200μg/mlのゲンタマイシンを含有する新規培地を添加して細胞外細菌を死滅させた(検定の0時間)。感染後、様々な時期における細胞内肺炎球菌を定量化するために、上澄液を除去し、細胞をPBSで3回洗浄した後、前述のようにトリトンX−100で溶解させた。各ウェルの溶解物の一連の希釈液を血液寒天上にプレーティングさせた。37℃で24時間培養した後、CFUの数を決定した。各細菌菌株に対して3つの独立的な検定を行った(三重検定)。対応標本または独立標本ステューダントt検定(paired or unpaired Student's t test)を用いて統計的分析を行った。
本研究は、最近の文献(前記Ogunniyi et al., 2003, MS in preparation)に記載の方法と本質的に同一に行った。試験感染の実施前に、細菌を10%[vol/vol]の馬血清を加えたTH(Todd Hewitt)寒天(Difco Laboratories, USA)(必要な場合、エリスロマイシンを補充する)上で一晩中37℃で培養させた後、THYブロスで約4時間37℃で成長させて約4×107CFU/ml(A600=0.1)の細菌を得た。その後、各細菌培養物をTHYブロスで約109CFU/mlに調節し、細胞10μl(約107CFU)を5週齢のCD1マウスの外鼻腔に接種させた。感染後1、2及び4日目に、各群のマウス4匹をランダムに犠牲させて各菌株の保菌程度を評価した。鼻咽頭、血液及び肺のサンプルを適切に滅菌PBDで連続的に希釈させ、適した抗生剤を含有する血液寒天上に二重に(duplicate)プレーティングした。プレートを95%の空気/5%のCO2の雰囲気下に37℃で約16時間培養した後、コロニーを計数し、複製本間の平均を求めた。
マウスを従来の文献(前記Ogunniyi, A. D. et al, 2000)に記載されている通りに腹腔内免疫させた。5〜6週齢のCBA/Nマウスの雌からなる4個群(群当り12匹)をAlPO4単独、遺伝子変形されたPlyトキソイド(PdB)+AlPO4、PspA+AlPO4、またはClpP+AlPO4で腹腔内免疫させた。各マウスに12〜14日間隔で各蛋白質抗原10μgを3回投与し、3回免疫後1週目に眼窩後方の採血によってマウスから血清を採取した。血清を群別にプーリングし、酵素結合免疫吸着検定(ELISA)によってPly、PspAまたはClpP特異的抗体に対して分析した。また、肺炎球菌D39の全体細胞溶解物、または精製されたPly、PspAまたはClpPを抗原として用いるウェスタン免疫ブロットで前記血清を分析した。
最終免疫後2週目に、免疫されたマウスを猛毒性の莢膜性タイプ2菌株(D39)で腹腔内試験感染させた。試験感染の実施前に、免疫細菌を血液寒天上で一晩中37℃で成長させた後、肉類抽出ブロス(脳心臓注入ブロス、Difco Laboratories, USA)またはTH (Todd Hewitt)ブロス(Difco Laboratories, USA)に10%(v/v)の馬血清を添加した血清ブロスに接種させた。次に、細菌を37℃で3時間静的に成長させて約108CFU/mlの細菌を得、接種物を試験感染容量当たり7.5×105CFUに調整した。血清タイプ特異的莢膜の生成は、Statens Seruminstitut(Copenhagen, Denmark)から購入した抗血清を用いて膨脹反応(quellung reaction)によって確認した。試験感染の実施後に、マウスを最初7日間4時間毎にモニタリングした後、21日までは毎日モニタリングし、各マウスの生存時間を記録した。各群間の中間生存時間上の差異をマン・ホイットニーのU検定(片側)(Mann-Whitney U test, one-tailed)によって分析した。
i)clpP−突然変異体は、マウスで持続的な感染をもたらさない
本発明者は、clpP−突然変異体が著しく弱毒された毒性を示したことを立証したことがある。cps2A、すなわち莢膜生合成遺伝子座の第1遺伝子の発現水準は、この菌株で熱ショック後に減少しなかったが、母菌株におけるcps2Aの発現は熱ショック後に著しく減少した。これは、全体的な毒性が減少したとしても、clpP−突然変異体がストレス負荷時に宿主マクロファージに対して野生型水準の耐性を示さなければならないことを示唆する。これは慢性菌血症を引き起こすおそれがあり、この場合、細菌が宿主免疫系に浸透して宿主内で生存することができるが、D39のニューモリシン陰性突然変異体に対して従来立証されている現象である電撃性の疾患は誘発しない。
本発明者は、clpP−突然変異体において、熱ショック後にply mRNA発現は増加したが、同時にPly蛋白質及びPly溶血活性は増加しなかったことを最近立証した。ところが、野生型では、熱ショック後にply mRNA水準だけでなく、Ply蛋白質及び溶血活性が全て著しく増加した(前記Kwon, H. Y. et al., 2003)。
ClpPの脱安定化作用は、plyプロセス産物及びply1次転写体の低い安定性をもたらすことが可能である。ply mRNAが熱ショックによって安定化されたとしても、Ply蛋白質の量またはその溶血活性の水準で相応する増加はなかった。このような不一致は、Plyが直接ClpPプロテアーゼによって活性化され、溶血活性が母菌株では増加することができるが、clpP−突然変異体では増加することができないためである。
抗−hsp100抗体を使用する免疫金標識法(immunogold labeling)によってhsp100の位置を測定した結果、hsp100は、酵母サッカロマイセス・セレビシエ (Saccharomyces cerevisiae)で熱ショック前後に細胞質及び核に位置するものと明らかになった(Fujita, K. et al., 1998, Hsp104 responds to heat and oxidative stress with different intracellular localization in Saccharomyces cerevisiae. Biochem. Biophys. Res. Commun. 248: 542-547)。バシラス・サブティラス(B. subtilis)では、抗体を用いた免疫金標識結果、ClpC及びClpX ATPaseが細胞外皮(envelope)及び細胞内部から検出され(Kruger, E. et al., 2000, The clp proteases of Bacillus subtilis are directly involved in degradation of misfolded proteins. J. Bacteriol. 182: 3259-3265)、これはhsp100、ClpC及びClpX蛋白質が熱ショック処理中に蛋白質凝集体と密接に関連があることを示唆する。
従来では、clpP−突然変異体はマウス敗血症モデルで毒性が非常に弱化された(前記Kwon, H. Y. et al., 2003)。しかも、clpP−突然変異体は、器官内試験感染後にマウスの肺に著しい水準でコロニーを形成せず、48時間の感染中に、死亡は記録されなかった(前記Robertson, G. T. et al., 2002)。ところが、肺炎球菌の侵入及び伝播は、その自然適所である鼻咽頭を介して行われるようである。
肺胞マクロファージは、肺炎球菌の侵入に対する宿主防御における1次的な要素である。(Knapp, S. et al., 2003, Alveolar macrophages have a protective antiinflammatory role during murine pneumococcal pneumonia. Am. J. Respir. Crit. Care Med. 167:171-179)。本研究において、clpP−突然変異体は、その母菌株に比べて鼻粘膜のコロニー形成において欠陥があるものと観察されたため、これは、突然変異体がさらに遅く成長するうえ、肺炎球菌が迅速に除去(clearance)されるためであると推論された。
ストレプトコッカス・ミュータンス(Streptococcus mutans)及びシュードモナス・フルオレッセンス(Pseudomonas fluorescence)のclpP−突然変異体において、菌膜水準は著しく減少した(Lemos, J. A., and R. A. Burne. 2002. Regulation and physiological significance of ClpC and ClpP in Streptococcus mutans. J. Bacteriol. 184:6357-6366)。したがって、これらの突然変異体において、cpsの量は母菌株タイプの場合に比べてさらに低いものと考慮された。
HSPは一部病原体で抗原として機能し、これら蛋白質は感染性疾患に対する防御機能をするものと知られた。また、ClpPが熱ショック後に細胞壁へ移動するので、肺炎球菌試験感染に対する前記蛋白質の防御誘導能を評価した。
本研究の目的は、肺炎球菌性疾患の発病において熱ショック蛋白質ClpPの役割を評価しようとすることにある。本発明者は、熱ショック後にclpP−突然変異体においてply mRNA発現は増加するが、Plyの量または溶血活性が同時に増加しなかったことを立証した。このような矛盾は、clpP−突然変異体においてply及びcps2A mRNAが30℃及び42℃の両方ともで安定しているので、ClpPが転写体の分解に対する寄与因子であることにあると思われる。しかも、熱ショック後にclpP−突然変異体においてcps2A及びply mRNAの両方ともの半減期が野生型の場合に比べて2倍以上さらに長かった。これは、熱ショック後にmRNA種の半減期は、ClpPの不在下に増加することを明確に示す。Plyの溶血活性は、37℃で細胞溶解物の培養によってさらに増加されなかった。これはClpPがPlyの溶血活性の活性化に対する直接的な原因ではないことを示す。本発明者は、このような発見からcps2A及びply mRNAが転写後水準でClpPによって分解されるが、これに対する特定のメカニズムはまだ不明であると結論付けた。
i)菌株及び培養
多糖類カプセルのない非病原性肺炎球菌(Rタイプ)ストレプトコッカス・ニューモニエ(Streptococcus pneumoniae)CP1200(前記Choi等、1999)菌株をCAT培地(Casitone 1%、Tryptone 0.5%、NaCl 0.5%、Yeast Extract 0.1%、0.175 M K2HPO4、及び glucose 0.2%)で培養し、多糖類カプセルを含有する病原性肺炎球菌D39(type 2; Avery Avery, O.T. et al., 1944. Studies on the chemical nature of the substance inducing transformation of pneumococcal types. Induction of transformation by a desoxyribonucleic acid fraction isolated from pneumococcus type III. J. Exp. Med. 79:137-158)と臨床で分離したSpn1049菌株(韓国の三星医療院で患者から分離された肺炎球菌であって、Optochinと胆汁酸に敏感であり、血液寒天培地で不完全溶血反応を示す)は、0.5%の酵母抽出物(yeast extract)を添加したTH(Todd-Hewitt)ブロス培地で培養した。サッカロマイセス・セレビシエ(Saccharomyces cerevisiae)(ATCC 287)は、グルコースが2%含まれたYNB培地(Difco Laboratories, USA)で、ストレプトコッカス・サーモフィラス(Streptococcus thermophilus) (KCTC 3778)は、MRS培地(Difco Laboratories, USA)で、バシラス・サブティラス(Bacillus subtilis)Marburg菌株(Boylan SA, Chun KT, Edson BA, Price CW. Early-blocked sporulation mutations alter expression of enzymes under carbon control in Bacillus subtilis. Mol Gen Genet. 212(2):271-280 (1988))、シュードモナス・アエルギノーサ(Pseudomonas aeruginosa) (ATCC 15522)、チフス菌 (Salmonella typhi) (ATCC 27870)、E.coli DH5a(Bethesda Research Laboratory)はNutrient培地(Difco Laboratories, USA) で、ヒト肺癌細胞株A549(ATCC CCL 185)は10%のFBSと2%のペニシリンストレプトマイシン(penicillin streptomysin)含有のDMEM培地(Gibco BRL)でそれぞれ培養した。
肺炎球菌のClpPと他の生物体蛋白質との類似性を確認するために、抗ClpP抗体と免疫ブロットを行った。肺炎球菌といろいろの生物体の溶解液を10%のポリアクリルアミドゲル(polyacrylamide gel)で電気泳動し、ニトロセルロース膜に転移させた後、抗ClpP抗体と免疫ブロットした後、酵素で標識された2次抗体(secondary antibody)を用いて検索した。すなわち、ニトロセルロースを、2%のTween20を含有するトリス緩衝塩水(Tris-buffered saline) (TBS; 50 mM Tris, 150 mM NaCl [pH 7.2]) 溶液で処理して非特異的な抗原−抗体反応をブロッキングさせた後、0.05%のTween20を含有するTBS溶液でウサギ抗−ClpP抗血清と室温で1時間徐々に揺すりながら 反応させた。HRP(Horse radish peroxidase)−コンジュゲーションヤギ抗−ウサギ免疫グロブリンG(IgG)抗体を2次抗体(secondary antibody)として用いて、0.05%のTween20を含有するTBS溶液で1:1000にて希釈して反応させ、過酸化水素と95%のエタノールで溶解されたN’,N’,N’,N’−4メチルベンジジンをそれぞれ基質と発色剤で反応させた。
National Center of Biotechnology Information(NCBI, U.S.A.)から提供するBLAST検索を用いて肺炎球菌clpPと隣接した遺伝子を確認し、かつ他の生物体のClpP蛋白質との類似性を確認した。BLAST検索を行ったとき、肺炎球菌clpPの塩基配列から類推されたアミノ酸配列は、他の生物体のClpPと高い類似性を示した。特に、ストレプトコッカス・サリバリウス(Streptococcus salivarius)とストレプトコッカス・アガラクチェ(Streptococcus agalactiae)のClpP蛋白質とはそれぞれ88%及び87%一致し(identity) 、91%及び92%の類似性(similarity)を示した。また、ラクトコッカス・ラクチス(L. lactis)ClpPと89%、エンテロコッカス・フェカーリス(Enterococcus faecalis)ClpPと81%、スタフィロコッカス・アウレウス(Staphylococcus aureus)ClpPと79%、バシラス・サブティラス(B. subtilis)ClpPと75%の類似性をそれぞれ示す。特に、ホモサピエンス(Homo sapiens) とは70%の類似性を示した。ClpP蛋白質のアミノ酸配列を比較した結果、全体的に高い水準の類似性を示し、特にセリンプロテアーゼの活性部位として推定されるセリン−96、ヒスチジン−121、アスパテート−172残基部分が非常に高い水準の保存性を示した。
現在、肺炎球菌DnaKは、抗原性が高く(Hamel J, Martin D, Brodeur BB. Heat shock response of Streptococcus pneumoniae: identification of immunoreactive stress proteins. Microb Pathog. 23(1):11-21 (1997))、肺炎球菌DnaK抗体がヒト蛋白質と反応せず(Kim SW, Choi IH, Kim SN, Kim YH, Pyo SN, Rhee DK. Molecular cloning, expression, and characterization of dnaK in Streptococcus pneumoniae. FEMS Microbiol Lett. 161(2):217-224 (1998))、ワクチン候補物質として評価されているので、ClpPに対してもこのような可能性を調査した。まず、ClpP蛋白質が他の生物体のClpPファミリーと類似であるか否かを確認するために、他の細菌または高等生物体の細胞溶解液と免疫ブロットを行った。予想とは異なり、グラム陽性菌であるストレプトコッカス・サーモフィラス(Streptococcus thermophilus, Sth)と肺炎球菌D39(D39)及び臨床で分離された肺炎球菌(Spn1049)とは反応したが、E.coli(Eco)、サッカロマイセス・セレビシエ(Saccharomyces cerevisiae, Sce)、バシラス・サブティラス(B. subtilis, Bsu)、シュードモナス・アエルギノーサ(Psuedomonas aeruginosa, Pae)、チフス菌(Salmonella typhi, Sty)及びヒト肺癌細胞株A549の細胞蛋白質とは全く反応しなかった。その結果を図17a及び図17bに示した。このような結果は、肺炎球菌のClpP蛋白質がDnaKのようにワクチン候補物質として使用できることを提示する。
Claims (8)
- 組み換え肺炎球菌ClpP蛋白質を抗原として含むワクチン。
- アジュバントをさらに含む、請求項1記載のワクチン。
- アジュバントが明礬である、請求項2記載のワクチン。
- ヒトまたは動物を肺炎球菌感染症に対して免疫化させるための、請求項1〜3のいずれか1項記載のワクチン。
- 肺炎球菌感染症が細菌性肺炎、中耳炎、菌血症または髄膜炎である、請求項4記載のワクチン。
- 組み換え肺炎球菌ClpPを抗原として含むワクチンであって、当該組み換え肺炎球菌ClpPが、
i)肺炎球菌ClpPをコードするDNA塩基配列が作動的に結合した発現ベクターを提供する段階と、
ii)前記i)の発現ベクターを宿主細胞に導入する段階と、そして
iii)宿主細胞から組み換え肺炎球菌ClpPを分離精製する段階
を含む組み換え肺炎球菌ClpPを製造する方法によって製造される、
前記ワクチン。 - 組み換え肺炎球菌ClpPを製造する方法に使用される発現ベクターがpET30(a)−clpPである、請求項6記載のワクチン。
- 組み換え肺炎球菌ClpPを製造する方法に使用される宿主細胞が大腸菌である、請求項6または7記載のワクチン。
