(1)麦芽エキス寒天培地における培養的・形態的性質
白色の菌糸が放射状に密に生育してコロニーを形成する。接種30日目のコロニー径は約4cmである。
上記いずれの培地においても、接種後1ヶ月経過しても菌糸の発育はほとんどみられない。
白色の光沢を有し、マット状に生育する。接種30日目の生育距離は約5mmである。

滅菌処理した液体培地(3%グルコース、0.3%酵母エキス、pH7.0)10mLの入った100mL容三角フラスコに、マツタケFERM BP−7304株の種菌約2mgを接種し、5〜35℃の種々の温度でそれぞれ培養し、28日目にフラスコから菌体を取り出し、蒸留水でよく洗浄した後に乾燥させ、質量を測定した。その結果、菌体質量は5〜15℃の範囲で直線的に増加し、15〜25℃の範囲で緩やかに増加した。27.5℃以上ではほとんど増殖しなかった。最適生育温度は15〜25℃である。
液体培地(3%グルコース、0.3%酵母エキス)のpHを1モル/L塩酸または1モル/L水酸化カリウムで調整し、pH3.0〜8.0の種々の培地を調製して菌体の生育pHを調べた。すなわち各培地をフィルター滅菌し、培地10mLを滅菌済100mL容三角フラスコに分注した。マツタケFERM BP−7304株の種菌約2mgを接種後、22℃で培養し、フラスコから菌体を取り出し、蒸留水でよく洗浄した後に乾燥させ、質量を測定した。その結果、菌体の生育限界はpH3.0〜7.0、最適生育pHは4.0〜6.0であった。
エビオス寒天平板培地に、マツタケFERM BP−7304株のブロック(約3mm×3mm×3mm)と、公知の13種類のマツタケ株(例えば、IFO 6915株;(財)発酵研究所)の各ブロック(約3mm×3mm×3mm)とを、約2cm間隔に対峙して植菌し、22℃で3週間培養した後、両コロニー境界部に帯線が生じるか否かを判定した。
滅菌処理した菌根菌用合成培地(「太田培地」。Ohtaら、"Trans. Mycol. Soc. Jpn.", 31, 323-334, 1990)10mLの入った100mL容三角フラスコに、マツタケFERM BP−7304株の種菌約2mgを接種し、22℃で培養し、42日目にフラスコから菌体を取り出し、蒸留水でよく洗浄した後に乾燥させ、質量を測定したところ、菌体441mgが得られた。
GC含量は49.9%である。
6種類の異なるPCR(Polymerase Chain Reaction)用プライマー(10mer)をそれぞれ単独で用いるRAPD(Random Amplified Polymorphic DNA)法により生成するDNAパターンについて、マツタケFERM BP−7304株と、公知の44種類のマツタケ株(例えば、IFO 6915株;(財)発酵研究所)とを比較したところ、マツタケFERM BP−7304株は、公知のマツタケ株(44種類)のいずれとも異なるDNAパターンを示した。
用いる培地としては、一般にマツタケ菌を培養する栄養源基質を有する培地であれば特に制限なく使用することができる。例えば、太田培地(Ohtaら、"Trans. Mycol. Soc. Jpn.", 31, 323-334, 1990)、MMN培地(Marx, D. H., "Phytopathology", 59:153-163, 1969)、浜田培地(浜田、"マツタケ", 97-100, 1964)等が挙げられるが、これら例示に限定されるものでない。
次に、マツタケ菌II(マツタケ菌Iを固形培地または液体培地で培養または保存したマツタケ菌)を静置液体培養してマツタケ菌IIIを製造する方法について記載する。
接種したマツタケ菌IIを含有する培養液と液体培地とを合せた混合物と、接種したマツタケ菌IIを含有する培養液との体積比(「接種時拡大倍率」)が好ましくは2〜50倍、より好ましくは3〜30倍となる量の液体培地を使用する。
次いで、マツタケ菌III(マツタケ菌IIを静置液体培養して得られたマツタケ菌)を振盪培養して、マツタケ菌IVを製造する方法について記載する。
接種したマツタケ菌IIIを含有する培養液と液体培地とを合せた混合物と、接種したマツタケ菌IIIを含有する培養液との体積比(「接種時拡大倍率」)が、好ましくは2〜50倍、より好ましくは3〜30倍、特に好ましくは5〜10倍となる量の液体培地を使用する。
次に、攪拌培養により、マツタケ菌V、マツタケ菌VI、マツタケ菌VII、マツタケ菌VIIIを製造する方法について記載する。
栄養源基質は、振盪培養で使用する、炭素源、窒素源、無機元素源、ビタミンB1などのビタミン類、アミノ酸類と同じものを使用することができる。
マツタケFERM BP−7304株の菌糸体の乾燥物粉末(以下「CM6271」とも記す)の調製
マツタケFERM BP−7304株菌糸体を、滅菌処理した培地(3%グルコース、0.3%酵母エキス、pH6.0)3.5tの入った7t容培養タンクに接種し、25℃で攪拌しながら4週間培養を行った。得られた培養物を濾布濾過し、菌糸体を分離した後、蒸留水で充分に洗浄した。
糖尿病モデル作成に用いる試薬としてのストレプトゾシン(STZ)は、シグマ・ケミカル社(Sigma Chemicals Co.,米国)から購入した。
日本クレア(株)から妊娠Wistarラットを購入し、呉羽化学工業(株)生物医学研究所で個別飼育した。すなわち、出産後48時間以内の新生児期ラットに、上記で調製したSTZ溶液を80mg/kg皮下注射して糖尿病モデルラットを作成した(以下、当該ラットを「nSTZラット」と記すこともある)。対照(コントロール)ラットには、STZ溶液の代わりに、クエン酸緩衝液を等量皮下注射した。
上記処置後のnSTZラット、対照(コントロール)ラットを親の元に戻して、母乳栄養下で4週間飼育した。
続いて9週齢のnSTZラットと対照(コントロール)ラットを、それぞれ2群ずつに分け、基本食で飼育を続ける群(=基本食摂取群)と、CM6271添加飼料(実験食)で飼育を続ける群(=実験食摂取群)とに分け、それぞれ8週間飼育した。CM6271添加飼料(実験食)は、下記表1に示す組成(「実験食組成」)欄に記載)の飼料で、基本食のセルロース成分の代わりにCM6271を添加したものである。
(2)対照(コントロール)ラットで、実験食摂取群(=「対象・実験食群」)
(3)nSTZラットで、基本食摂取群(=「nSTZ・基本食群」)
(4)nSTZラットで、実験食摂取群(=「nSTZ・実験食群」)。
上記各群のラットを用いて以下の実験および評価を行った。
一般状態は、1日1回観察し、体重を投与開始日および最終投与日を含み週1回測定した。実験食摂取期間の体重、飼料摂取量、および飲水量は毎週2回測定した。
実験食投与の直前(9週齢)、投与後3週目および6週目にラットを5時間絶食させた後、無麻酔下で尾静脈よりヘパリン含有ヘマトクリット毛細管を用いて採血した。次いで、ヘマトクリット遠心機で分離し、血漿中のグルコース濃度(血糖値)を測定した。
実験食投与後4週目(=13週齢)にラットを20時間絶食させた後、無麻酔下で1g/kgのグルコースを経口投与した。グルコース投与前、および投与60分後、120分間後に、ヘパリン含有ヘマトクリット毛細管を用いて尾静脈より採血した。ヘマトクリット遠心機で分離し、血漿中のグルコース濃度を測定した。結果を表4および図3に示す。なお表4中の数字は平均±SDを示す。
実験終了前に、ラットを20時間絶食させた後、2g/kgの糖質に相当するそれぞれの実験食を30分間摂取させ、実験終了まで再び絶食させた。実験食摂取前および摂取後30分間、60分間、120分間の時、ヘパリン含有ヘマトクリット毛細管を用いて尾静脈より採血した。ヘマトクリット遠心機で分離し、血漿中のグルコース濃度およびインスリン濃度を測定した。
上記実験結果から明らかなように、実験群では、対照群に比し、空腹時高血糖の低下、グルコース負荷時の耐糖能の改善、食後の血糖の上昇抑制、およびインスリン分泌量の増加が認められた。一方、体重増加量、飼料摂取量および飲水量は両群間で有意差はなかった。これによりCM6271添加飼料はnSTZラットの症状を改善することが示唆され

- マツタケ(Tricholoma matsutake)FERM BP−7304株またはその抽出物を含む、2型糖尿病治療剤。
- マツタケ(T. matsutake)FERM BP−7304株が菌糸体、培養物(Broth)または子実体(胞子を含む)である、請求項1記載の2型糖尿病治療剤。
- マツタケ(T. matsutake)FERM BP−7304株が菌糸体の乾燥粉末である、請求項1または2記載の2型糖尿病治療剤。
- マツタケFERM BP−7304株の抽出物が菌糸体の熱水抽出液またはアルカリ溶液抽出液である、請求項1〜3のいずれか1項に記載の2型糖尿病治療剤。
