材料及び方法

WO96/02555及び米国特許第6,239,116号の特許は、免疫賦活性であるためには、オリゴヌクレオチドが非メチル化CpGモチーフを含む配列を必要とすると教示している(WO96/02555、カラム13、19〜20行及び米国特許第6,239,116号、カラム6、1〜3行)。EP 0 468 520は、免疫賦活性であるために、オリゴヌクレオチドが、少なくとも6ヌクレオチド長の回文配列を満たすことを必要とすると教示している(EP 0 468 520、カラム11、34〜37行)。従って、少なくとも6ヌクレオチド長の回文配列を有さない、いくつかの非CpGオリゴヌクレオチドを、ヒト末梢血単核白血球(PBMC)に対して実施した増殖アッセイ、細胞分化アッセイ、サイトカインIL6分泌アッセイ及びIgM分泌アッセイを用いて調査した。陽性コントロールとして、Hartman及びKrieg(Hartmann,G.、Krieg,A.M.2000.Mechanism and function of a newly identified CpG DNA motif in human primary B cells.J.Immunol.164:944)に記載される、組成:

免疫賦活を、ヒトB細胞についての一般的なマーカーCD19及びヒトB細胞についての活性化マーカーCD86を用いるフローサイトメトリーによっても評価した。

ホスホロチオエートCpG及び非CpGオリゴヌクレオチドである2006及びIMT 021によるヒトIL−6分泌の誘導
このアッセイの結果も、1.5μg/mlで用いられた場合、非CpG ODN(S)IMT 021が有効な免疫賦活剤であることを示す。従って、2006オリゴヌクレオチドの他の非CpG配列改変体を調査した。第3表は、これらの改変体(ここで、このODNの全てのCpGのCまたはGが他のヌクレオチドによって置換されている)のうち6つの結果を示す。観察され得るように、ODN(S)2006におけるCpGのGは、B細胞増殖またはIL6分泌に必ずしも必要ではない。しかし、CpGのCの改変は、その置換がAまたはGである場合有害であるが、それがTである場合有害でない。これらの結果は、明らかに、ODN(S)2006によるB細胞増殖及びIL6分泌の刺激がCpGグループの完全性とは関連しないことを示す。

非CpGオリゴヌクレオチドの免疫賦活活性に対する構造改変の効果:活性モチーフの定義
非CpG ODNの免疫賦活活性に対する一次構造の影響を研究するために、2006及びIMT 021 オリゴヌクレオチドのいくつかの改変体を合成した。第4表は、いくつかのIMT 021改変体の一次構造及びその免疫賦活活性を評価するために実施した増殖及びIL−6アッセイの結果を示す。
免疫賦活IMT 021オリゴヌクレオチドから誘導されたホスホロチオエート非CpGオリゴヌクレオチドによる末梢白血球増殖及びIL−6分泌の誘導
非CpGオリゴヌクレオチドの活性中心(モチーフ)の組成を調査するために、いくつかのODNを0.375μg/mlでアッセイした(図3)。以下のことが見出された:
a)オリゴヌクレオチド中のTGグループの存在は、強力な免疫賦活のために十分でない(WO 01−22972との相違)。例えば、ODN IMT 028、IMT 046、IMT 546、IMT 547及びIMT 550(これらの全てが2つのTGを有する)は、非常に乏しい活性を有する。一方、TGジヌクレオチドを保有しないODN IMT 543は、同じ一般的組成を有するが2つのTGジヌクレオチドを有するODN IMT 544と同じ(低い)活性を有する。
ホスホロチオエートまたはホスホジエステルのPyNTTTTGT ODNによるIL6分泌の誘導
(S):ホスホロチオエート。(O)ホスホジエステル。ホスホロチオエートODNを0時間のインキュベーションで添加した(6μg/ml)。ホスホジエステルODNを以下のように添加した:インキュベーション0時間で30μg/ml、4時間後に30μg/ml、16時間後に30μg/ml。

第6表は、ODN IMT 504中に存在する2つの非CpGモチーフ(モチーフ:CATTTTGT)のヌクレオチドの各々における変化の効果を示す。このモチーフの第1位のCの、A(ODN IMT 531)またはG(ODN IMT 533)による置換は、活性の約50%の損失を生じる。しかし、このCのTによる置換(ODN IMT 532)は、活性を変化させなかった。従って、最大の活性を得るために、このモチーフの第1位は、ピリミジンヌクレオチド(CまたはT)により占められているべきである。このモチーフの第2位において、A(ODN IMT 504)またはT(ODN IMT 535)またはC(ODN IMT 534)は、等価な選択肢である。

免疫賦活モチーフの第2位に変化を有するホスホロチオエート非CpGオリゴヌクレオチドによる末梢白血球増殖及びIL−6分泌の誘導
ODN鎖内でのモチーフの位置の変化の効果を調査するために、ODN IMT504中に存在する非CpGモチーフCATTTTGTを、24ヌクレオチド長のポリT鎖における種々の位置に導入した(図5)。見出され得るように、ポリT ODN(S)自体が有意な有糸分裂活性を有していた。一方、たった1つのCATTTTGTモチーフの導入が、そのモチーフが配置された場所に依存して、ポリT鎖の免疫賦活活性の1.2〜1.8倍(増殖アッセイによる測定)または2.4〜3.5倍(IL−6分泌アッセイによる測定)の増加を生じた。5’末端から少なくとも2ヌクレオチドまたは3’末端から4の距離が、最大の活性を達成するために必要であるようである(ODN IMT174〜IMT 179)。最適な位置への2つのモチーフの導入(ODN IMT182)は、ポリT鎖の免疫賦活活性の1.9倍(増殖アッセイによる測定)、または2.9倍(IL−6分泌アッセイによる測定)の増加を生じた。このことは、第2のモチーフの寄与が無視できることを示す。一方、ODN IMT504のような最も有効なODNの活性は、増殖アッセイにより測定した場合でポリTの活性より3倍を超えて高く、これは、ODNの全体的な活性に対して、本発明のモチーフの周辺の少なくともいくつかのヌクレオチドの組成の有意な影響があることを示唆する事実である。
本発明の非CpGオリゴヌクレオチドの免疫賦活活性に対する構造改変の影響:活性モチーフの外側のヌクレオチド組成の影響
図5に示される結果は、非CpGコアモチーフの外側のオリゴヌクレオチドの組成が最適な免疫賦活活性を達成するために重要であることを示す。従って、これらのモチーフの周囲のヌクレオチド組成における変化を有する多数のオリゴヌクレオチドを合成した。第8表は、ODN IMT 504の免疫賦活活性における、5’末端の最初の4ヌクレオチド及び3’末端の最後の4ヌクレオチドの組成における変化の効果を示す。観察され得るように、最適な選択は:これら2つのモチーフに対して−1位にCまたはG、−2位にC、TまたはG、−3位及び−4位に任意のヌクレオチド、+1位にAまたはT、+2位にG、+3位に任意のヌクレオチド及び+4位にG、である。

本発明の非CpGオリゴヌクレオチドの免疫賦活活性に対する構造改変の効果:オリゴヌクレオチドのサイズの影響
第9表は、1つの免疫賦活モチーフを有するODNは、その鎖が16以上のヌクレオチド長である場合に活性であることを示す。そのODNが20〜25ヌクレオチド長である場合、活性が最大である。本研究の間にアッセイした、少なくとも1つの本発明の非CpGモチーフを保有するこの範囲内の全ての(100より多い)ODNは、非常に活性であった。
B細胞増殖及びIL6分泌に対するホスホロチオエート非CpGオリゴヌクレオチドのサイズの効果

本発明のホスホロチオエート非CpGオリゴヌクレオチドによる末梢白血球のIgM分泌の誘導
末梢白血球におけるIgM分泌の誘導は、オリゴヌクレオチドの免疫賦活活性の別の重要なマーカーである。第10表は、上記のいくつかのホスホロチオエート非CpGオリゴヌクレオチドによるIgM分泌の賦活を示す。観察され得るように、増殖及びIL6分泌の誘導において最も活性の高い非CpG ODN(S)は、IgM分泌の誘導においても最高であった。

実施例7
本発明のホスホロチオエート非CpGオリゴヌクレオチドによるBリンパ球におけるCD40、MHC I及びMHC IIの発現の刺激
ヒトPMBCをODN IMT 504、陽性コントロールとしてのODN 2006及び陰性コントロールとしてのODN IMT 022と共にインキュベートした(図6)。観察され得るように、ODN IMT 504は、Bリンパ球上でのCD40、MHC I及びMHC IIの発現の刺激についてCpG ODN 2006と同様に活性である。
本発明の非CpGオリゴヌクレオチドによる精製Bリンパ球の刺激
ヒトCD19+(B)細胞を、95%より高い純度まで精製した。第11表及び図7は、これらの精製細胞上での免疫賦活活性がヒトPMBCを用いて観察されるものに匹敵することを示す。これらの結果は、ヒト細胞に対する本発明の非CpGオリゴヌクレオチドによる刺激が直接的であることを示す。

実施例9
本発明のホスホジエステル非CpGオリゴヌクレオチドによる免疫賦活
図8及び第12表は、ヒトPMBCに対する本発明のホスホジエステル非CpGオリゴヌクレオチドの効果を示す。ホスホジエステルオリゴヌクレオチドはヌクレアーゼに対して非常に感受性であるので、これらを、培地に3回(0、4、及び16時間)、30μg/mlの最終濃度まで添加した。非常に強力なホスホジエステルCpG ODN(ODN 2080)を、サイトメトリックアッセイ(Hartmann G、Weeratna R、Ballas ZK、Payette P、Suparto I、Rasmussen WL、Wadschmidt M、Sajuthi D、Purcells RH、Davis HL、Krieg AM.Delineation of a CpG phosphorothioate oligodeoxynucleotide for activating primate immune responses in vitro and in vivo.J.Immunol.164、1617〜1624(2000))における陽性コントロールとして使用した。観察され得るように、これらの条件下では、本発明の非CpGモチーフを保有するホスホジエステルODNは、免疫賦活活性を有する。これに関し、これらの非CpG ODNと、WO 01/22972において特許請求される非CpG ODNとの差異は、注目に値する(第5表及び図4を参照のこと)。

実施例10
本発明の非CpG ODNによるCebus apella及びMacaca fascicularis種のサルの末梢白血球における細胞増殖の誘導
本発明の非CpG ODN(S)は、マウス、ブタ及びヒツジにおいてあまり効果的な免疫賦活剤ではない。しかし、これらは、サルにおいては有効である。例えば、図9は、Cebus apella及びMacaca fascicularis種のサルの末梢白血球に対する非CpG ODN(S)の免疫賦活活性を示す。これらの結果に従い、非ヒト霊長類において最も有効な非CpG ODNは、CATTTTGTモチーフを保有するものである。従って、サルは、これらの非CpG ODN(S)の臨床応用に対する調査のために動物モデルとして用いられ得る。
被験体のワクチン接種
本発明の非CpG ODNアジュバントを含むワクチン処方物が、被験体を種々の細菌性及びウイルス性疾患因子ならびに腫瘍細胞に対するワクチン接種をするために用いられ得る。第13表は、組換えB型肝炎表面抗原(rHBsAg)及びアジュバントとしてのODN IMT 504の存在下または非存在下でアルミナを含むワクチン処方物を用いた、Cebus apella種のサルの接種の効果を示す。
IMT ODNを有するHBsAgに対して免疫したCebus apellaにおける抗HB応答
ホスホロチオエート非CpGオリゴヌクレオチドによる、悪性Bリンパ球における共刺激分子の発現の刺激
本発明のホスホロチオエート非CpGオリゴヌクレオチドによる、慢性リンパ性白血病(CLL)に罹患する患者の血液から回収した悪性Bリンパ球の刺激を、フローサイトメトリック(FACS)分析によって試験した。代表的なFACS分析の結果を図10に示す。見出され得るように、本発明のホスホロチオエート非CpGオリゴヌクレオチドIMT 504は、CpG ODN 2006と同様に、悪性Bリンパ球上でのCD86、CD40及びMHCクラスI表面共刺激分子の発現を刺激することができる。
ホスホロチオエート非CpGオリゴヌクレオチドによる悪性B細胞アポトーシスの刺激
本発明のホスホロチオエート非CpGオリゴヌクレオチドによる、慢性リンパ性白血病(CLL)に罹患する患者の血液から回収した悪性Bリンパ球におけるアポトーシスの刺激を、アポトーシスマーカーとしてヨウ化プロピジウム及びアネキシンVを用いるフローサイトメトリック(FACS)分析によって試験した。代表的なFACS分析の結果を図11に示す。観察され得るように、本発明の非CpGオリゴヌクレオチドIMT 504は、CpG ODN 2006と同様に、白血病細胞のアポトーシスを刺激することができる。
本発明のホスホロチオエート非CpGオリゴヌクレオチドによる、形質細胞様樹状細胞上での共刺激分子の発現の刺激
本発明のホスホロチオエート非CpGオリゴヌクレオチドによる、正常なドナーの血液から回収した形質細胞様樹状細胞の刺激を、フローサイトメトリック(FACS)分析によって試験した。代表的なFACS分析の結果を、図12に示す。見出され得るように、本発明のホスホロチオエート非CpGオリゴヌクレオチドIMT 504は、CpG ODN 2006と同様に、形質細胞様樹状細胞上でのCD86、CD40及びMHCクラスI表面分子の発現を刺激することができる。
本発明のホスホロチオエート及びホスホジエステル非CpGオリゴヌクレオチドと共にインキュベートした精製形質細胞様樹状細胞によるインターフェロンα分泌
腫瘍性疾患を有する被験体の処置
1つ以上の本発明の免疫賦活オリゴヌクレオチドを含む薬学的処方物は、種々の腫瘍性疾患に対して被験体を処置するために用いられ得る。特に、黒色腫を有するヒトは、活性成分として本発明のオリゴヌクレオチドを含む薬学的処方物の投与によって処置され得る。各々の用量におけるオリゴヌクレオチドの量、及び投与スケジュールは、被験体の体重及び腫瘍進行のステージに従い変化し得る。例えば、進行した切除不可能な転移性の黒色腫を有する約70kgのヒトについては、約1mgの用量の本発明のオリゴヌクレオチドが、約10週間にわたり、1週間に3回投与され得る。
本発明のホスホロチオエート非CpGオリゴヌクレオチドによる、末梢白血球のIL−10分泌の誘導及びIL−5分泌の阻害
IL−10は、IgE産生の強力なサプレッサーであり、そしてエピトープ特異的T細胞アネルギーの誘導及び維持は、増加したIL−10に関連する。図13は、幾人かのドナーのPMBCにおける、ホスホロチオエート非CpGオリゴヌクレオチドIMT 504及びコントロールによるIL−10の分泌の刺激を示す。観察され得るように、IMT 504は、CpG陽性コントロールODN 2006と同様に活性である。アレルゲン特異的Th2細胞は、IL−5を産生し、このIL−5は、好酸球の分化、活性化及び生存を促進する。図14は、ホスホロチオエート非CpGオリゴヌクレオチドIMT 504及びCpG陽性コントロールODN 2006が幾つかのヒトPMBCにおいてIL−4及びCon Aにより誘導されるIL−5産生のリプレッサーであることを示す。
アレルギー性疾患を有する被験体の処置
1つ以上の本発明の免疫賦活オリゴヌクレオチドを含む薬学的処方物は、種々のアレルギー性疾患に対して被験体を処置するために用いられ得る。
