Description
多孔質フィルタは有意の数の白血球を除去するには有効であるが、欠点がある。例えば、遠心分離の後、多孔質フィルタの前に、ある期間をあけて、活性化した血小板を非活性状態に変換しなければならない。そうしないと、活性化血小板がフィルタに詰まる可能性が高い。したがって、多孔質フィルタの使用はオンライン・プロセスでは実行可能ではない。
ここでrは粒子の半径、ρp は粒子の密度、ρm は液体媒質の密度、ηは媒質の粘度、gは重力または遠心加速度である。粒子の半径はストークスの法則では2乗になり、粒子の密度はならないので、血球の密度ではなくそのサイズが、沈殿速度に大きい影響を及ぼす。これは、遠心水簸法の間に、粒子が同様の密度である場合、なぜ大きい方の粒子が概ねチャンバ内に保持され、小さい方の粒子が放出されるかを説明する。
以上およびその他の理由から、粒子の分離を改良する必要がある。

- 回転軸の周りに回転可能な回転体(12)を有する遠心分離機(10)にして、回転体(12)は、回転軸の半径方向内方の内壁(20)と半径方向外方の外壁(22)との間に画成される環状溝(18)を内部に有する保持器(16)と、管セット(90)とを有し、管セット(90)は、保持器(16)の環状溝(18)内に配置されて前記環状溝(18)の内壁(20)と協働する内壁(96)と、溝(18)の外壁(22)と協働する外壁(98)とを有し、管セット(90)の内壁(96)と外壁(98)との間で環状の流路(64)を画成する分離容器(28)を含み、環状流路(64)は、隔離壁(52)により隔離された入口端部分(48)と出口端部分(50)とを有し、
分離容器(28)は、
成分に分離する流体を分離容器に供給する入口(54)と、
流体の分離した成分を分離容器(28)から取り出すための少なくとも3つの第1収集口(56)、第2収集口(58)ならびに第3収集口(60)と、
入口(54)と流体結合した入口ライン(36)と、
第1収集口(56)と流体結合した第1収集ライン(38)と、
第2収集口(58)と流体結合した第2収集ライン(40)と、
第3収集口(60)と流体結合した第3収集ライン(42)と、
分離容器(28)内での分離後、流体の成分を分離する流体チャンバ(30)にして、該流体チャンバ(30)は、回転体(12)に装着され、かつ第1収集ライン(38)と流体結合する流体チャンバ入口(34)、流体チャンバ出口(32)および流体チャンバ入口(34)と流体チャンバ出口(32)との間に延在して流体チャンバ(30)を画成する流体チャンバ壁を含み、流体チャンバの壁は、流体チャンバ入口(34)と流体チャンバ出口(32)との間の位置で最大断面積を有する流体チャンバ内部を画成する内面を有し、流体チャンバ内部が最大断面積の位置から流体チャンバ入口(34)に向かって収束し、かつ最大断面積の位置から流体チャンバ出口(32)に向かって収束している流体チャンバ(30)と、
流体チャンバ出口(32)と流体結合した流出ラインと
を有している遠心分離機において、
入口(54)は、分離容器(28)の入口端部分(48)に設けられ、第1収集口(56)、第2収集口(58)ならびに第3収集口(60)は、分離容器(28)の出口端部分(50)に設けられ、
分離容器(28)は、分離容器の出口端部分(50)に設けられ、少なくとも流体の比較的中間密度の成分の流れをほぼ堰き止めるバリア(62)も有し、
第1収集口(56)は、バリア(62)と分離容器(28)の入口端部分(48)との間に設けられて流体の比較的中間密度の成分を抽出し、
分離容器(28)の内壁(96)は、可撓性であり、保持器(16)の溝(18)の内壁(20)は、保持器(16)の溝(18)の外壁(22)の方へ延在する隆起(68)を有し、これにより分離容器(28)の内壁(96)を変形させて分離容器(28)の外壁(98)へ向かう捕捉堰(70)を形成させて比較的低密度の流体の成分を捕捉し、比較的中間密度の流体成分の第1収集口(56)への移送を促進させ、
捕捉堰(70)が分離容器の入口端部分(48)と第1収集口(56)との間に設けられていることを特徴とする遠心分離機。 - さらに、分離容器(28)の出口端部分(50)に設けた第4収集口(61)および第4収集口(61)と流体結合した第4収集ライン(44)を包含し、第4収集口(61)が分離容器(28)中で流体の分離された成分の境界を調節する、請求項1に記載の遠心分離機。
- バリア(62)が分離容器(28)を横切って延在するスキマー堰(62a)であり、スキマー堰が、分離容器(28)の出口端部(50a)に、流体の少なくとも比較的低密度の成分のための第1流路(64a)と、流体の少なくとも比較的高密度の成分のための第2流路(66a)とを画成し、分離容器(28)を保持器(16)内に配置すると、第1流路(64a)が第2流路(66a)より回転軸に近くなっている請求項1あるいは2に記載の遠心分離機。
- 分離容器(28)の出口端部分(50c、50e)において、バリア(62)の上流で分離容器(28)の内壁(96)および外壁(98)が流体蜜に合わされた二つの密封部分(100、100e、102、102e)が設けられ、密封部分(100、100e、102、102e)間に流路(66c、66e)が画成される、請求項1から3のいずれか一項に記載の遠心分離機。
- 分離容器(28)の出口端部分(50d)において、分離容器(28)の内壁(96)と外壁(98)とは、分離容器(28)の頂部と底部との間の位置で流体蜜に合わされて少なくとも一つの密封部分(100d)を形成し、これにより出口端部分(50d)に少なくとも第1および第2の流路(64d,66d)を画成している、請求項1から3のいずれか一項に記載の遠心分離機。
- 分離容器(28)の出口端部分(50e)において、第1流路(64e)に隣接して外壁(98)上に少なくとも第1支持部(126)が設けられ、さらに第2流路(66e)に隣接して内壁(96)上に第2支持部(128)が設けられ、第1支持部(126)が外壁(98)から内壁(96)へ向く第1堰を形成し、第2支持部(128)が内壁(96)から外壁(98)へ向く第2堰を形成するようになし、分離容器(28)が保持器(16)内に配置された際に、第1堰と第2堰が少なくとも比較的中間密度の成分の流れを遮断し、さらに第1流路(64e)が第2流路(66e)よりも遠心分離機の回転軸の近くに配置されている、請求項4に記載の遠心分離機。
- さらに、密封部分を貫通する細長い部材(124f)を包含し、細長い部材(124f)の第1部分が第1支持部(126)を提供し、細長い部材(124f)の第2部分が第2支持部(128)を提供している、請求項6に記載の遠心分離機。
- 第2収集口(58)が第1および第3収集口(56,60)よりも分離容器(28)外壁(98)に近くに位置して流体の少なくとも比較的高密度の成分を収集し、第3収集口(60)が第1および第2収集口(56,58)よりも分離容器(28)の外壁(98)から遠くに位置して流体の少なくとも比較的低密度の成分を収集する、請求項1から7のいずれか一項に記載の遠心分離機。
- 流体チャンバ(30)が、流体チャンバ(30)の内部に形成された少なくとも1本の溝(72)を含み、溝(72)が、流体チャンバ入口(34)を通って流体チャンバ(30)の内部に入る流体の成分のコリオリ噴射を減少させる、請求項1から8のいずれか一項に記載の遠心分離機。
- 流体チャンバ(30)が、流体チャンバ(30)の内部に形成された少なくとも一つの段部(76)を含み、段部(76)が、流体チャンバ入口(34)を通って流体チャンバ(30)の内部に入る流体の成分のコリオリ噴射を減少させる、請求項1から8のいずれか一項に記載の遠心分離機。
