Description

gは重力加速度又は遠心加速度である。
ストークスの方程式では、粒子の半径が2乗に累乗されるが、粒子の密度は累乗されないので、その密度ではなく、細胞の粒径がその沈降速度に大きく影響する。このように、粒子が同じような密度を有する場合、遠心水簸時には大きい方の粒子がチャンバ内に残り、他方で小さい方が放出される理由が説明される。
1.アフェレーシス処理から残留物を入手し、遠心分離によって白血球(WBC)を分離及び回収し、さらにヘマトクリット値を5〜20%に調整する。
2.この産物の処理前産物試料を入手し、それに「前」と標示を付け、フロリダ州マイアミ市のCoulter Corporation社から入手したCoulter Automated Cell Counterを使用して、この試料を(3重反復試験で)分析する。
3.3.51mLの分離チャンバ内部で細胞負荷(load)を実現するのに必要な産物の容積を計算する。
4.産物の適正容積を150mLの移変え容器に移し換える。
5.ヒト血清アルブミン又はヒドロキシエチルデンプンを含有する希釈液を調製し、50mLを150mLの移換え容器に移し換え、残部を第2移換え容器に移し換える。
6.入口ポンプループ、除去ポンプループ(内径0.112センチメートル(0.044インチ)管を使用)、出口管路、及び分離チャンバからなる管使い捨てセットを準備する。これらの要素は、ポンプループ44、赤血球ループ46、入口管路28、除去された赤血球用の出口管路30、及び図1のチャンバ18に対応する。
7.除去ポンプの出口を画分回収器に取り付け、5mLのポリプロピレン試験管中に400〜500μLの画分を回収するように設定する。
8.「出口」と標示された600mLの移換え容器に分離チャンバ出口からの出口管路を取り付ける。
9.管セットに調製した希釈液を注入する。
10.4mL/分の流量及び2400rpmの遠心器速度で細胞産物を分離チャンバ中に充填する。
11.細胞を分離チャンバ中に充填している間に、チャンバ出口を介して流出する液体及び細胞を移換え容器中に回収する。
12.50mLの希釈液で管と分離チャンバ内部の細胞とを洗浄する。
13.入口ポンプを停止し、事前に画定した時間の長さの間、分離チャンバ内部の細胞を沈降させる。
14.除去ポンプを使用して1mL/分の流量で細胞を分離チャンバの底から除去し、画分回収器を使用して小量のアリコートの細胞を回収する。
15.一旦、すべての細胞を分離チャンバから取り除いたら、画分回収を中断する。
16.「出口」と標示した移換え容器中及び試験管中の液体と細胞の重さを記録する。
17.CELL−DYN 4000 Systemを使用して「前」及び「出口」試料を分析すると共に、すべての試験管のアリコートも分析する。
Claims (9)
- 少なくとも白血球及び赤血球を含有する血液産物を、入口と出口を有し遠心器ロータに取り付けられた流体チャンバに、流体チャンバの入口を介して充填する段階と、
赤血球の連銭形成を助長し、かつそれらの沈降速度を増大させるために充填された血液産物にヒドロキシエチル澱粉を含む低密度の流体を追加する段階と、
遠心力下で赤血球を白血球から沈降させる段階と、
沈降させた赤血球を白血球から流体チャンバの入口を介して除去する段階と、
を含む赤血球を白血球から除去する方法。 - 供給源のすべての流体が空になるまで、充填する段階と、回転する段階と、追加する段階と、沈降させる段階と、除去する段階とを反復する段階をさらに含む、請求項1に記載の方法。
- 少なくとも白血球及び赤血球を含有する血液産物を有する流体供給源を、流体チャンバの入口に第1導管を介して流体連結する段階と、
流体チャンバから除去された赤血球を収容するため流体チャンバの入口に除去導管を流体連結する段階と、
除去された赤血球を収容するため除去容器に除去導管を連結する段階と、
をさらに含む、請求項1に記載の方法。 - 白血球を分離して、流体チャンバ出口を介して流出する流体から回収するため、流体チャンバの出口に分離容器を流体連結する段階をさらに含む、請求項3に記載の方法。
- 流体チャンバが、
入口と出口の間に延在する平滑な内部面を有する壁と、
壁中の最大面積であって、該壁は最大断面積から入口に向かって、かつ最大断面積から出口に向かって先細りする最大断面積と、
をさらに含む、請求項1〜4のいずれか一項に記載の方法。 - 流体チャンバの入口に粒子を収容するため除去導管を流体連結することをさらに含む、請求項5に記載の方法。
- 流体チャンバの入口に3方向コネクタを連結する段階と、
3方向コネクタを介して、流体チャンバの入口に入口導管を連結する段階と、
3方向コネクタを介して、流体チャンバの入口に除去導管を流体連結する段階と、
をさらに含む、請求項6に記載の方法。 - 除去する段階の後で、水簸によって白血球を白血球サブセットに分離する段階をさらに含む、請求項1に記載の方法。
- 流体チャンバの出口を介して、分離された白血球サブセットを除去する段階と、
除去された分離した白血球サブセットを粒子濃縮器に供給する段階と、
粒子濃縮器中で白血球サブセットから流体を分離する段階と、
望ましい白血球サブセットを回収する段階と、
をさらに含む、請求項8に記載の方法。
