Description
Claims (48)
- サンプル中のターゲットの検出方法であって:
a)(i)支持体と、(ii)固定化材料を介して上記支持体上にランダムに固定化された複数の微粒子とを含んでなるアレイであって、各微粒子が、その微粒子に結合した自己コーディング・マーカーとプローブとを含んでなり、各自己コーディング・マーカー及びプローブが、独自の自己コーディング・マーカー/プローブ対を含んでなり、当該プローブが、ターゲットに特異的に結合するよう形状化され、且つ配置されているアレイを用意するステップ;
b)上記ターゲットを含むと疑われるサンプルを上記アレイに適用するステップ;
c)上記ターゲットが上記プローブに結合しうる条件下に上記サンプルと上記アレイを維持するステップ;そして
d)上記微粒子に結合した自己コーディング・マーカーと、当該微粒子に会合したターゲット・マーカーとを検出するステップを含んでなるとともに、
固定化材料が反応性ポリマーを含んでなり、反応性ポリマーが支持体に結合され、微粒子が反応性ポリマーを介して支持体上に固定化され、
反応性ポリマーが少なくとも1つの光反応基を有する光反応性ポリマーを含んでなり、光反応性基が、アリール・ケトン、アリールアジド、アシル・アジド、スルホニル・アジド、ホスホリル・アジド、ジアゾアルカン、ジアゾケトン、ジアゾアセテート、及びケテンからなる群より選ばれ、且つ、
前記固定化材料が上記支持体上でパターン形成して上記固定化材料のパッチを形成し、前記微粒子が固定化材料の当該パッチ上に固定化される、方法。 - 前記反応性ポリマーが、官能性ポリアクリルアミド、ポリメタクリルアミド、ポリビニルピロリドン、ポリアクリル酸、ポリエチレン・グリコール、ポリビニル・アルコール、ポリ(HEMA)、これらのコポリマー、及びこれらの組み合わせから成る群から選ばれるポリマーを含む、請求項1に記載の方法。
- 前記反応性ポリマーが、ビニルピロリドンとN−[3−(4−ベンゾイルベンズアミド)プロピル]メタクリルアミド(BBA−APMA);及びアクリルアミドとBBA−APMAから成る光反応性コポリマーの群から選ばれる、請求項1に記載の方法。
- 前記反応性ポリマーが、官能性多糖、グリコサミノグリカン、ポリペプチド、及びこれらの組み合わせから成る群から選ばれるポリマーを含む、請求項1に記載の方法。
- 前記複数の微粒子が、独自の自己コーディング・マーカー/プローブ対を有する微粒子のサブセットを2つ以上含み、かつ、当該微粒子のサブセットを、前記支持体上の固定化材料の別々のパッチ上に別個に配置する、請求項1に記載の方法。
- 前記検出ステップが前記サブセットの位置を決定する操作をさらに含み、上記自己コーディング・マーカーを検出する操作と組み合わせることにより、前記プローブの同定を少なくとも可能にする、請求項5に記載の方法。
- 前記固定化材料が結合ペアを含み、ここで、上記結合ペアの一方は支持体に結合し、及び当該結合ペアの他方は微粒子に結合し、そして上記結合ペアのメンバー同士の相互作用によって上記支持体上に上記微粒子が固定化される、請求項1に記載の方法。
- 前記結合ペアが、ストレプトアビジン:ビオチン、プロテインA/G:IgG及び抗原:抗抗原抗体、レクチン:炭水化物、並びに化学的−親和性結合ペアから成る群から選ばれる、請求項5に記載の方法。
- 前記微粒子を反応性化合物を用いて官能化し、そして当該反応性化合物を用いて上記微粒子を前記固定化材料、前記自己コーディング・マーカー、前記プローブ、あるいはこれらの任意の組み合わせに結合させる、請求項1に記載の方法。
- 前記反応性化合物が、カルボン酸、スルホン酸、リン酸、ホスホン酸、アルデヒド基、アミン基、チオール基、チオール反応基、及びエポキシド基から成る群から選ばれる反応基を含む、請求項9に記載の方法。
- 前記サンプルを処理して、前記ターゲット・マーカーを前記ターゲットに結合させる、請求項1に記載の方法。
- 前記プローブが核酸プローブであり、かつ、前記ターゲットが、サンプル中に存在していて当該核酸プローブと特異的にハイブリダイズする核酸ターゲットである、請求項1に記載の方法。
- 前記プローブが抗体プローブであり、かつ、前記ターゲットが、サンプル中に存在していて当該抗体によって特異的に認識される分子である、請求項1に記載の方法。
- 前記自己コーディング・マーカーが、少なくとも1つの検出可能な粒子を含む、請求項1に記載の方法。
- 前記少なくとも1つの検出可能な粒子が、発蛍光団、量子ドット、放射性同位体、及び磁性粒子から成る群から選ばれる、請求項14に記載の方法。
- 前記自己コーディング・マーカーが、検出可能な粒子の組み合わせを含む、請求項15に記載の方法。
- アレイの製造方法であって:
a)複数の微粒子を含む混合物を調製するステップ;
b)支持体と、場合によっては上記微粒子を固定化材料でコーティングするステップ;そして
c)複数の微粒子を含む上記混合物を、当該微粒子が上記固定化材料を介して上記支持体上にランダムなパターンで固定化されるように当該支持体上に配置するステップを含んでなるとともに、

前記コーティング・ステップが、固定化材料を前記支持体上でパターン形成して上記固定化材料のパッチを形成する操作を含む、方法。 - 当該光反応性基が、アリール・ケトン、アリールアジド、アシル・アジド、スルホニル・アジド、ホスホリル・アジド、ジアゾアルカン、ジアゾケトン、ジアゾアセテート、及びケテンから成る群から選ばれる、請求項17に記載の方法。
- 前記反応性ポリマーが、官能性ポリアクリルアミド、ポリメタクリルアミド、ポリビニルピロリドン、ポリアクリル酸、ポリエチレン・グリコール、ポリビニル・アルコール、ポリ(HEMA)、これらのコポリマー、及びこれらの組み合わせから成る群から選ばれるポリマーを含む、請求項17に記載の方法。
- 前記反応性ポリマーが、ビニルピロリドンとN−[3−(4−ベンゾイルベンズアミド)プロピル]メタクリルアミド(BBA−APMA);及びアクリルアミドとBBA−APMAから成る光反応性コポリマーの群から選ばれる、請求項17に記載の方法。
- 前記反応性ポリマーが、官能性多糖、グリコサミノグリカン、ポリペプチド、及びこれらの組み合わせからなる群から選ばれるポリマーを含む、請求項17に記載の方法。
- 前記配置ステップが、前記の固定化材料のパッチを介して微粒子を前記支持体上に固定化する操作を含む、請求項17に記載の方法。
- 前記調製ステップが、ユニークな自己コーディング・マーカー/プローブ対を有する微粒子のサブセットを2つ以上調製する操作を含み、かつ、前記配置ステップが、前記支持体上の固定化材料の別々のパッチ上に微粒子のサブセットを別個に配置する操作を含む、請求項17に記載の方法。
- 前記固定化材料が結合ペアを含み、ここで、前記コーティング・ステップが、上記結合ペアの一方のメンバーを支持体と結合させ、及び当該結合ペアの他方のメンバーを微粒子と結合させる操作を含み、そして前記配置ステップが、上記結合ペアのメンバー同士を相互作用させて上記支持体上に上記微粒子を固定化する操作を含む、請求項17に記載の方法。
- 前記結合ペアが、ストレプトアビジン:ビオチン、プロテインA/G:IgG及び抗原:抗抗原抗体、レクチン:炭水化物、並びに化学的−親和性結合ペアから成る群から選ばれる、請求項24に記載の方法。
- 前記調製ステップが、前記微粒子を反応性化合物でコーティングする操作であって、当該反応性化合物を用いて上記微粒子を前記固定化材料、前記自己コーディング・マーカー、前記プローブ、あるいはこれらの組み合わせに結合させる操作をさらに含む、請求項17に記載の方法。
- 前記反応性化合物が、カルボン酸、スルホン酸、リン酸、ホスホン酸、アルデヒド基、アミン基、チオール基、チオール反応基、及びエポキシド基から成る群から選ばれる反応基を含む、請求項26に記載の方法。
- 前記プローブが核酸プローブである、請求項17に記載の方法。
- 前記プローブが抗体プローブである、請求項17に記載の方法。
- 前記自己コーディング・マーカーが、少なくとも1つの検出可能な粒子を含み、かつ、前記調製ステップが、前記微粒子を当該少なくとも1つの検出可能な粒子に結合させる操作を含む、請求項17に記載の方法。
- 前記検出可能な粒子が、発蛍光団、量子ドット、放射性同位体、及び磁性粒子から成る群から選ばれる、請求項30に記載の方法。
- 前記自己コーディング・マーカーが、検出可能な粒子の組み合わせを含み、かつ、前記調製ステップが、前記微粒子を当該検出可能な粒子の組み合わせに結合させる操作を含む、請求項31に記載の方法。
- a)支持体;
b)上記支持体上にランダムに固定化された複数の微粒子;及び
c)上記微粒子を上記支持体上に固定化する固定化材料を含んでなるアレイであって、
各微粒子が、その微粒子に結合した自己コーディング・マーカーとプローブとを含んでなり、各自己コーディング・マーカー及びプローブが、独自の自己コーディング・マーカー/プローブ対を含んでなり、プローブが、ターゲットに特異的に結合するよう形状化され、且つ配置されており、
固定化材料が反応性ポリマーを含んでなり、反応性ポリマーが支持体に結合され、微粒子が反応性ポリマーを介して支持体上に固定化され、
反応性ポリマーが少なくとも1つの光反応基を有する光反応性ポリマーを含んでなり、且つ、
前記固定化材料が上記支持体上でパターン形成されることで上記支持体上に固定化材料のパッチを形成している、アレイ。 - 当該光反応性基が、アリール・ケトン、アリールアジド、アシル・アジド、スルホニル・アジド、ホスホリル・アジド、ジアゾアルカン、ジアゾケトン、ジアゾアセテート、及びケテンから成る群から選ばれる、請求項33に記載のアレイ。
- 前記反応性ポリマーが、官能性ポリアクリルアミド、ポリメタクリルアミド、ポリビニルピロリドン、ポリアクリル酸、ポリエチレン・グリコール、ポリビニル・アルコール、ポリ(HEMA)、これらのコポリマー、及びこれらの組み合わせから成る群から選ばれるポリマーを含む、請求項33に記載のアレイ。
- 前記反応性ポリマーが、ビニルピロリドンとN−[3−(4−ベンゾイルベンズアミド)プロピル]メタクリルアミド(BBA−APMA);及びアクリルアミドとBBA−APMAから成る光反応性コポリマーの群から選ばれる、請求項33に記載のアレイ。
- 前記反応性ポリマーが、官能性多糖、グリコサミノグリカン、ポリペプチド、及びこれらの組み合わせから成る群から選ばれるポリマーを含む、請求項33に記載のアレイ。
- 前記微粒子が前記固定化材料のパッチを介して前記支持体上に固定化されている、請求項33に記載のアレイ。
- 前記複数の微粒子が、ユニークな自己コーディング・マーカー/プローブ対を有する微粒子のサブセットを2つ以上含み、かつ、当該微粒子のサブセットを、前記支持体上の固定化材料の別々のパッチ上に別個に配置する、請求項33に記載のアレイ。
- 前記固定化材料が結合ペアを含み、ここで上記結合ペアの一方は支持体に結合し、及び当該結合ペアの他方は微粒子に結合し、そして上記結合ペアのメンバー同士の相互作用によって上記支持体上に上記微粒子が固定化される、請求項33に記載のアレイ。
- 前記結合ペアが、ストレプトアビジン:ビオチン、プロテインA/G:IgG及び抗原:抗抗原抗体、レクチン:炭水化物、並びに化学的−親和性結合ペアから成る群から選ばれる、請求項40に記載のアレイ。
- 前記微粒子が、反応性化合物から成るコーティングをさらに含み、ここで、当該反応性化合物を用いて上記微粒子を前記固定化材料、前記自己コーディング・マーカー、前記プローブ、あるいはこれらの組み合わせに結合させる、請求項33に記載のアレイ。
- 前記反応性化合物が、カルボン酸、スルホン酸、リン酸、ホスホン酸、アルデヒド基、アミン基、チオール基、チオール反応基、及びエポキシド基から成る群から選ばれる反応基を含む、請求項42に記載のアレイ。
- 前記プローブが核酸プローブである、請求項33に記載のアレイ。
- 前記プローブが抗体プローブである、請求項33に記載のアレイ。
- 前記自己コーディング・マーカーが、少なくとも1つの検出可能な粒子を含み、その少なくとも1つの検出可能な粒子が前記微粒子に結合している、請求項33に記載のアレイ。
- 前記検出可能な粒子が、発蛍光団、量子ドット、放射性同位体、及び磁性粒子から成る群から選ばれる、請求項46に記載のアレイ。
