Description
本発明は、感染性疾患、並びに原発性及び転移性の腫瘍性疾患の予防及び治療のための方法及び組成物に関する。感染性疾患及び癌の予防及び治療の実施においては、抗原性細胞及び/又はその消化産物からのサイトゾル及び膜由来タンパク質を含む組成物を、熱ショックタンパク質及び/又はα−2−マクログロブリンと複合体化し、腫瘍及び感染因子に対する免疫応答を増強する。
2.1.熱ショックタンパク質
熱ショックタンパク質(HSP)は、ストレスタンパク質とも呼ばれており、初めは、細胞が熱ショックに応答して合成するタンパク質として同定されたものである。HSPは分子量に基づいて5つのファミリーに分類されており、HSP100、HSP90、HSP70、HSP60及びsmHSPがある。その後、これらのファミリーの多くのメンバーは、他のストレス刺激、例えば、栄養欠乏、代謝破壊、酸素ラジカル、細胞内病原体による感染などに応答して誘導されることが見出された(Welch, May 1993, Scientific American 56-64; Young, 1990, Annu. Rev. Immunol. 8:401-420; Craig, 1993, Science 260:1902-1903; Gethingら, 1992, Nature 355:33-45; 及びLindquistら, 1988, Annu. Rev. Genetics 22:631-677を参照のこと)。
αマクログロブリンは、補体成分であるC3、C4及びC5をも含む構造的に関連したタンパク質のタンパク質スーパーファミリーのメンバーである。ヒト血漿タンパク質α(2)マクログロブリン(α2M)は、主にプロテイナーゼインヒビター並びに血漿性及び炎症性液性プロテイナーゼスカベンジャー分子として知られている720kDaのホモ四量体タンパク質である(総説としては、Chu及びPizzo, 1994, Lab. Invest. 71: 792を参照されたい)。α2Mは1474アミノ酸残基を有する前駆体として合成される。シグナル配列として機能する最初の23個のアミノ酸が切り離されて1451アミノ酸残基の成熟タンパク質となる(Kanら, 1985, Proc. Natl. Acad. Sci. U.S.A. 82: 2282-2286)。
本発明は、癌及び感染性疾患の予防及び治療に有用な抗原タンパク質及びペプチドと、熱ショックタンパク質(HSP)又はα−2−マクログロブリン(α2M)との複合体の製造方法を包含する。
本発明は、癌及び感染性疾患の予防及び治療に有用な、熱ショックタンパク質(HSP)又はα−2−マクログロブリン(α2M)を含む組成物の製造方法を提供する。本発明の方法は、HSP又はα2Mと、抗原性細胞の抗原タンパク質及びペプチドとの複合体をin vitroで調製することを含む。一実施形態において、本方法は、抗原タンパク質の集団を含む抗原性細胞のタンパク質調製物を生成し、抗原タンパク質の集団とHSP又はα2Mとをin vitroで複合体化することを含む。別の実施形態において、本方法は、抗原性細胞のタンパク質調製物を、少なくとも1つのプロテアーゼで消化して抗原ペプチドの集団を生成した後、抗原ペプチドをHSP又はα2Mとin vitroで複合体化することを含む。本発明は、抗原性細胞の完全な抗原性の可能性を利用する。
本発明の抗原性細胞は、それに対する被験体の免疫応答が望まれる抗原決定基を含む。
本発明において本発明の組成物は、HSPと複合体化した抗原タンパク質を含み、抗原タンパク質は目的の抗原性細胞のタンパク質の調製物に由来する。本発明の組成物はまた、α2Mと複合体化した抗原タンパク質を含み、抗原タンパク質は目的の抗原性細胞のタンパク質の調製物に由来する。本発明の組成物はまた、HSPと抗原ペプチドの複合体、又はα2Mと抗原ペプチドの複合体(まず、目的の抗原性細胞のタンパク質の調製物からペプチドの集団を生成し、次にHSP又はα2Mとペプチドとを複合体化することにより調製される)を含む。
本発明の一実施形態において、癌細胞、感染細胞、又は病原体から得られるタンパク質調製物が提供される。例えば癌の治療のために、癌患者から得られた腫瘍細胞から、術後にタンパク質調製物が調製される。本発明の別の実施形態において、抗原をコードする組換え発現系の導入により改変された細胞株中で、目的の1つ以上の抗原タンパク質を合成し、このような細胞はタンパク質を調製するために使用される。タンパク質は1つ以上の細胞画分(例えば抗原性細胞のサイトゾル)に由来するか、又は抗原性細胞の膜若しくは細胞壁から抽出若しくは可溶化される。細胞溶解、細胞内容物の分画、及びタンパク質濃縮又は単離のために当該分野で公知の任意の方法が使用できる。例えば、Current Protocols in Immunology、第2巻、第8章、Coliganら(編)、John Wiley and Sons;Pathogenic and Clinical Microbiology: A Laboratory Manual、Rowlandら、Little Brown & Co.、1996年6月を参照されたい(これらは、参照することによりその全文が本明細書に組み込まれる)。細胞内容物を分画するために使用される方法に応じて、細胞画分は、少なくとも20、50、100、500、1,000、5,000、10,000又は20,000種の異なるタンパク質を含む。
細胞(患者の生検により得られた腫瘍細胞、又はin vitro培養した腫瘍細胞、又は病原因子に感染した細胞)を、3倍量の1×溶解バッファー(30mM重炭酸ナトリウム、pH7.5、1mM PMSFを含む)に懸濁し、氷上で20分間インキュベートし、次に低張膨潤させた細胞を、95%を超える細胞が溶解されるまでダウンスホモジナイザーでホモジナイズする。剪断の代わりに、細胞を、顕微鏡観察で測定した時99%を超える細胞が溶解するまで氷上で超音波処理してもよい。超音波処理を使用する場合には、超音波処理前に細胞を、1mM PMSFを含むリン酸緩衝化生理食塩水(PBS)のようなバッファーに懸濁する。

本発明において、抗原性細胞に由来するサイトゾル及び膜由来タンパク質は、場合により消化して抗原ペプチドを生成することができる。一実施形態において、サイトゾル又は膜由来タンパク質は、消化して使用される。別の実施形態において、サイトゾル及び膜由来タンパク質は消化反応で組合わされて、抗原ペプチドを生成する。好適な実施形態において、プロテアーゼ消化に使用されるタンパク質調製物は、抗原性細胞、又は抗原性細胞のサイトゾル若しくは膜中の他のタンパク質から、1つ以上の特定のタンパク質を選択的に取り出したり保持する調製方法を行なっていない。
本発明においては、抗原性細胞に由来する抗原ペプチドをHSP及び/又はα2Mと複合体化させる。本明細書には、本発明において用いるためのHSP及びα2Mの単離及び調製に使用しうる方法を例示する。
HSP70−ペプチド複合体の精製は、以前に記載されており、例えば、Udonoら、1993, J. Exp. Med. 178:1391-1396を参照されたい。使用できる手順を以下に記載するが、これは例として示すに過ぎず、本発明を何ら制限するものではない:
最初に、30mM炭酸水素ナトリウム(pH7.5)と1mM PMSFとからなる、3容量の1×溶解バッファーに腫瘍細胞を懸濁させる。次に、ペレットを氷上で音波処理することにより、99%以上の細胞を溶解させるが、これは顕微鏡検定により決定される。音波処理に代わり、細胞を機械的せん断により、95%以上の細胞が溶解するまで、ダウンス型(Dounce)ホモジナイザーで均質化することにより、細胞を溶解しうる。
MethA肉腫細胞(5億個の細胞)を低張性バッファー中で均質化させ、溶解物を4℃にて100,000gで90分遠心する。上清をATP−アガロースカラムにアプライする。カラムをバッファー中で洗浄し、5カラム容量の3mM ATPで溶離する。HSP70−ペプチド複合体は、溶離する合計15分画のうち2〜10分画に溶離する。溶離した分画をSDS−PAGEにより分析する。この手順を用いて、HSP70−ペプチド複合体を見かけ均質性まで精製することができる。
用いることができる手順を以下に記載するが、これは例として示すに過ぎず、本発明を制限するものではない:

α−2−マクログロブリンは、市販のものを購入してもよいし、又はヒトの血液から精製して調製してもよい。血液からα2Mを精製するために、例として以下のプロトコールを用い得るが、これに限定されるものではない。
本発明の特定の実施形態では、HSP及びα2Mは、組換え手法によりHSP及びα2Mを高レベルで発現する細胞から調製することができる。多くのHSP及びα2Mのアミノ酸配列及びヌクレオチド配列は、GenBankなどの配列データベースから一般に入手可能である。Entrezなどのコンピュータープログラムを用いて、データベースを閲覧することができ、目的とするアミノ酸配列及び遺伝子配列をアクセッション番号により検索することができる。また、これらのデータベースは、アラインメントスコアや統計によって類似の配列を分類する、FASTA及びBLASTのようなプログラムを用いて、問合せ配列に対し様々な程度の類似性を有する配列を同定するために検索することもできる。本発明の組成物、方法、並びに、HSPペプチド複合体の調製に用いることができるHSPのヌクレオチド配列としては、限定するものではないが下記のものが挙げられる:ヒトHSP70、GenBankアクセッション番号M24743(Huntら、1995, Proc. Natl. Acad, Sci. U.S.A. 85:3801-3811);及びマウスBiP、GenBankアクセッション番号U16277(Haasら、1988, Proc. Natl. Acad, Sci. U.S.A. 85:2250-2254)。HSPをコードする縮重配列もまた使用しうる。
HSP又はα2M分子をコードするヌクレオチド配列は、組換え細胞において増殖及び発現するために発現ベクターに挿入することができる。発現構築物とは、本明細書中で使用する場合、適切な宿主細胞中でのHSP又はα2M分子の発現を可能にする1以上の調節領域と機能的に連結されたHSP又はα2Mをコードするヌクレオチド配列を指す。「機能的に連結された」とは、調節領域と発現しようとするHSP又はα2Mのポリペプチド配列とが、HSP又はα2Mの配列が転写及び最終的には翻訳されるように連結され、配置されている関係を指す。HSP又はα2Mの発現のために、種々の発現ベクターを用いることが可能であり、例えば限定するものではないが、プラスミド、コスミド、ファージ、ファージミド、又は改変ウイルスが挙げられる。例えば、λ誘導体などのバクテリオファージ、pBR322若しくはpUCプラスミド誘導体又はBluescriptベクター(Stratagene)などのプラスミドなどが挙げられる。典型的には、そのような発現ベクターは、適当な宿主細胞において該ベクターが増殖するための機能的な複製起点、HSP又はα2Mの遺伝子配列を挿入するための1以上の制限エンドヌクレアーゼ部位、及び1以上の選択マーカーを含む。
HSP又はα2Mと、抗原性細胞、その細胞画分又はウイルス粒子から調製されたタンパク質及び/又はペプチド集団とをin vitroで複合体化するための方法の例を本明細書に記載する。タンパク質及び/又はペプチドの集団は、第4.2.1節に記載のような抗原性細胞のタンパク質調製物に由来する。特定の実施形態において、ペプチドは、抗原性細胞、その細胞画分又はウイルス粒子のタンパク質調製物の消化の結果物である。複合体化反応によって、HSPと抗原性細胞又はウイルス粒子のタンパク質又はペプチドとの共有結合の形成が生じうる。複合体化反応によって、α2Mと抗原性細胞又はウイルス粒子のタンパク質又はペプチドとの共有結合の形成が生じうる。複合体化反応によって、HSPとタンパク質及び/又はペプチドとの非共有結合、あるいはα2Mとタンパク質及び/又はペプチドとの非共有結合の形成が生じることもある。
本発明によれば、抗原性細胞又はウイルス粒子の消化したサイトゾル及び/又は膜由来タンパク質に由来する抗原ペプチドと、HSP及び/又はα2Mとの複合体を含む本発明の組成物を、癌又は感染性疾患に罹患した被験者に投与する。一実施形態では、「治療」又は「治療すること」とは、癌若しくは感染性疾患、又は少なくとも1つの認識可能なその症状の改善を意味する。別の実施形態では、「治療」又は「治療すること」とは、必ずしも被験者により認識可能ではない、癌若しくは感染性疾患に関連する少なくとも1つの測定可能な物理的パラメーターの改善を意味する。さらに別の実施形態では、「治療」又は「治療すること」とは、物理的に(例えば、認識可能な症状の安定化)、生理的に(例えば、物理的パラメーターの安定化)、又はその両方のいずれかにより、癌又は感染性疾患の進行を阻止することを意味する。
本発明の方法により治療又は予防することができる感染性疾患は、限定するものではないが、ウイルス、細菌、真菌、原生動物及び寄生生物などの感染因子により引き起こされる。本発明は、細胞内病原体により引き起こされる感染性疾患の治療又は予防に限定されない。
本発明の方法により治療又は予防することができる癌の種類には、肉腫及び癌腫、例えば、線維肉腫、粘液肉腫、脂肪肉腫、軟骨肉腫、骨原性肉腫、脊索腫、血管肉腫、内皮肉腫、リンパ管肉腫、リンパ管内皮肉腫、滑膜性腫瘍、中皮腫、ユーイング腫、平滑筋肉腫、横紋筋肉腫、大腸癌、膵臓癌、乳癌、卵巣癌、前立腺癌、扁平上皮癌、基底細胞癌、腺癌、汗腺癌、皮脂腺癌、乳頭癌、乳頭腺癌、嚢胞腺癌、髄様癌、気管支癌、腎細胞癌、肝細胞癌、胆管癌、絨毛癌、精上皮癌、胎生期癌、ウィルムス腫、子宮頸癌、精巣腫瘍、肺癌、小細胞肺癌、膀胱癌、上皮癌、神経膠腫、星状細胞腫、髄芽腫、頭蓋咽頭腫、上皮細胞腫、松果体腫、血管芽細胞腫、聴音神経系腫、希突起神経膠腫、髄膜腫、黒色腫、神経芽細胞腫、網膜芽腫、白血病(例えば急性リンパ性及び急性骨髄性白血病(骨髄芽球性白血病、前骨髄球性白血病、骨髄単球性白血病、単球性白血病、及び赤白血病);慢性白血病(慢性骨髄性(顆粒球性)白血病及び慢性リンパ性白血病));及び真性赤血球増加症、リンパ腫(ホジキン病及び非ホジキン病)、多発性骨髄腫、ヴァルデンストレームマクログロブリン血症、及び、H鎖病が含まれるがこれらに限定されない。
HSP及びα2Mの特異的免疫原性は、HSP又はα2M自体に由来するのではなく、それらに結合した抗原ペプチドに由来する。本発明の好ましい実施形態では、癌ワクチンとして用いる本発明の組成物における複合体は、自己由来の(autologous)複合体であるため、癌免疫療法における最も困難な問題の2つを回避することができる。1つは、実験動物の癌と同様、ヒト癌は、抗原が異なる可能性があることである。本発明の好ましい実施形態では、上記の問題を回避するため、HSP及び/又はα2Mは抗原ペプチド及びペプチドと複合体化し、該複合体をタンパク質又はペプチドが由来する同じ被験者の癌を治療するために用いることができる。2つめは、癌免疫療法の現在最も一般的な手法が、癌細胞系のCTL−認識エピトープの決定に焦点を絞っていることである。この手法では、癌に対する細胞系及びCTLが利用可能でなければならない。これらの試薬は、ヒト癌の大部分には利用不可能である。自己由来の抗原ペプチドの使用に関する本発明の実施形態では、癌免疫療法は、細胞系又はCTLの利用可能性に依存せず、また、癌細胞の抗原エピトープの決定も必要としない。これらの利点によって、自己由来の抗原ペプチドに結合したHSP及び/又はα2Mの複合体は、魅力的な対癌免疫原となる。
養子免疫療法とは、癌又は感染性疾患を治療するための治療上の手法であって、免疫細胞が、腫瘍細胞及び/若しくは抗原成分に対し特異的な免疫を直接的又は間接的に媒介する、あるいは場合によっては感染性疾患を治療するか又は腫瘍を退縮させることを目的として、免疫細胞を宿主に投与するものである(1999年11月16日に登録された米国特許第5,985,270号を参照されたい;この文献はその全体を参照により本明細書に組み入れるものとする)。
抗原提示細胞(マクロファージ、樹状細胞及びB細胞を含むがこれらに限定されない)は、好ましくは、Inaba, K.ら, 1992, J. Exp. Med. 176:1693-1702に記載のようにヒトの末梢血又は骨髄からの幹細胞及び前駆細胞からin vitroで調製することにより得る。樹状細胞は当該技術分野において既知の種々の任意の方法により得ることができる。例えば限定するものではないが、樹状細胞は、Sallustoら、1994, J Exp Med 179:1109-1118及びCauxら、1992, Nature 360, 258-261(参照によりその全体を本明細書に組み入れる)に記載の方法により得ることができる。このましい態様においては、ヒトの血球から得られたヒト樹状細胞を使用する。
単核細胞を患者(好ましくは治療対象の患者)の末梢血から、フィコール−ハイパーク勾配遠心法により単離し、患者自身の血清又は他のAB+ヒト血清によりコーティングした組織培養ディッシュ上に播種する。該細胞を37℃にて1時間インキュベートし、次いで付着しなかった細胞をピペッティングにより除く。該ディッシュに残された付着細胞に冷たい(4℃)リン酸緩衝化生理食塩水中1mMのEDTAを添加し、室温にて15分間静置する。細胞を回収し、RPMIバッファーで洗浄し、RPMIバッファー中に懸濁させた。マクロファージの数は、マクロファージ−コロニー刺激因子(M−CSF)と共に37℃にてインキュベートすることにより増加させる。
APCを、抗原分子と結合したHSP又はα2Mで、好ましくは該細胞をin vitroにて該複合体とインキュベートすることにより、感作させる。APCを、HSP又はα2Mと抗原分子の複合体で、in vitroにてHSP−複合体又はα2M−複合体と共に37℃にて15分間〜24時間インキュベートすることにより、感作させる。限定するものではないが、例えば、4×107個の樹状細胞を、1mlあたり10μgのgp96−ペプチド複合体と共に、又は、1mlあたり100μgのHSP90−ペプチド複合体と共に、37℃にて15分間〜24時間、1mlの無添加RPMI培地中でインキュベートすることができる。細胞を3回洗浄し、患者に注射するのに適切な濃度(例えば1×107/ml)で好ましくは無菌の生理学的培地中に再懸濁する。好ましくは、感作された樹状細胞を注入する患者は、もともと樹状細胞を単離した患者である(自己由来の実施形態)。
感作したAPCを、通常の臨床的な手順により、好ましくは皮内投与により、患者に全身的に再注入する。これらの活性化された細胞を、好ましくは、該細胞が由来する(自己由来)患者に全身投与することにより、再注入する。患者は、該患者の状態に応じて一般的に約106〜約1012個の感作樹状細胞を投与される。治療計画においては場合により、患者に対し、上記に加えて、限定するものではないが、サイトカインIFN−α、IFN−γ、IL−2、IL−4、IL−6、TNF又はその他のサイトカイン増殖因子など、適量の生物応答修飾物質を投与してもよい。
本発明の方法により調製されたHSP及び/又はα2Mに結合した抗原ペプチドの複合体は、細胞増殖性疾患又は感染性疾患を治療又は改善するために治療上有効な量で患者に投与しうる。治療上有効な量とは、かかる疾患の症状の改善をもたらすのに十分な量の複合体を意味する。
本発明の組成物は、抗原ペプチド集団とHSP及び/又はα2Mとの複合体の免疫原性の有効量を含み、これを癌又は感染性疾患に対する治療が必要な被験者に投与する。ここでこの方法は、癌又は感染性疾患に対する免疫応答を誘導するものである。上記複合体の毒性及び治療効力は、例えば、LD50(集団の50%が致死する量)及びED50(集団の50%に治療上有効な量)の決定について、細胞培養物又は実験動物における標準的な薬学的手法により決定しうる。毒性と治療効果との用量比は治療インデックスであり、比LD50/ED50で表される。高い治療インデックスを示す複合体が好ましい。毒性の副作用を示す複合体も用いることができるが、非感染細胞に対する損傷の可能性を最小限に抑えるように罹患組織部位に上記複合体をターゲティングする送達系を設計し、それにより副作用を低減するように注意を払う必要がある。
本発明に従って使用するための医薬組成物は、1以上の生理学的に許容される担体又は賦形剤を用いて通常の方法により製剤化しうる。
本発明はまた、本発明の方法及び/又は治療方法を行うためのキットをも提供する。一実施形態において、かかるキットは、1以上の容器内に、第2の容器に収容されるHSP及び/又はα2Mと組み合わせるための抗原タンパク質及びペプチドを含むタンパク質調製物を含む。別の実施形態において、かかるキットは、1以上の容器内に、第2の容器に収容されるHSP及び/又はα2Mと組み合わせるための抗原ペプチドを含む消化ペプチドを含有する。あるいは、1以上の容器に、自己由来の投与のために特定の患者から単離されるHSP及び/又はα2Mと複合体化させるためのタンパク質及び/又はペプチドを供給しうる。場合により、さらに、タンパク質及びペプチドと複合体化させるための精製HSPを第2容器中に提供してもよい。
場合により、当該技術分野で公知の方法を用いて、本発明のHSP−タンパク質複合体、HSP−ペプチド複合体、α2M−タンパク質複合体及びα2M−ペプチド複合体を免疫原性についてアッセイしてもよい。例として、限定するものではないが、以下に挙げる手順の1つを用いることができる。好ましい実施形態では、ELISPOTアッセイを用いる(後掲、第4.9.4節を参照)。
簡単に説明すると、任意の好適な投与経路を用いて、マウスに所定量のHSP−及び/又はα2M複合体を注射する。陰性対照として、例えば正常組織から調製されたタンパク質及び/又はペプチドと複合体化したHSPを他のマウスに注射する。特異的抗原を含むことがわかっている細胞、例えば、腫瘍細胞又は感染性疾患の因子に感染した細胞は、このアッセイの陽性対照として用いることができると考えられる。マウスには、7〜10日あけて、2回注射する。最後の免疫感作から10日後に、脾臓を取り出して、リンパ球を放出させる。放出されたリンパ球は、後に、目的とする抗原を発現した死細胞の添加によりin vitroで刺激することができる。
Kruse及びSebald, 1992, EMBO J. 11:3237-3244に記載されているのとほぼ同様に、一次T細胞を脾臓、新鮮な血液又はCSFから取得した後、FICOLL−PAQUE PLUS(Pharmacia、スエーデンUpsalla)を用いた遠心分離により精製する。末梢血単核細胞を、抗原分子を発現する細胞の溶解物と一緒に7〜10日インキュベートする。溶解物中の抗原を処理及び提示するために、任意によりアッセイの24〜48時間前に抗原提示細胞を培養物に添加してもよい。次に、遠心分離により細胞を回収し、RPMI 1640倍地中で洗浄する(GibcoBRL、メリーランド州Gaithersburg)。5×104個の活性化T細胞/ウェルを、96ウェルプレート上の10%ウシ胎児血清、10mM HEPES(pH7.5)、2mM L−グルタミン、100単位/mlペニシリンG、及び100μg/ml硫酸ストレプトマイシンを含むRPMI 1640倍地中で37℃で72時間培養し、次いで、1μlCi3H−チミジン(Dupont NEN、マサチューセッツ州ボストン)/ウェルで6時間パルスした後、回収し、TOPCOUNTシンチレーションカウンター(Packard Instrument Co.、コネティカット州メリデン)で放射能測定した。
本発明の特定の実施形態では、HSP−又はα2M−複合体のワクチン接種に応答して産生された抗体を測定することにより、該複合体の免疫原性を決定する。実施形態の一例では、PBS中のワクチンに用いたタンパク質/ペプチドの精製された非HSP−又はα2M−複合体化形態の0.75μg/ml溶液を50μl/ウェルで、マイクロプレート(96ウェルイムノプレートII、Nunc)を4℃で16時間、20℃で1時間かけてコーティングする。ウェルを空にし、1ウェルにつき、200μlのPBS−T−BSA(0.5%(v/v)TWEEN20及び1%(w/v)ウシ血清アルブミンを含むPBS)で20℃で1時間かけてブロッキングした後、PBS−Tで3回洗浄する。ワクチン接種した動物(モデルマウス又はヒト患者など)から得た50μl/ウェルの血漿又はCSFを20℃で1時間かけて添加・放置した後、プレートをPBS−Tで3回洗浄する。次に、必要に応じて、PBS−T−BSA中で1:1,500に希釈したセイヨウワサビペルオキシダーゼ(Amersham)とコンジュゲートさせた50μl/ウェルのヒツジ抗マウス又は抗ヒト免疫グロブリンと一緒に、次いで(前記と同様PBS−Tでさらに3回洗浄後)、50μlのo−フェニレンジアミン(OPD)−H2O2基質溶液と一緒に、20℃で1時間インキュベートした後、抗ペプチド抗体活性を熱量計により測定する。5分後150μlの2M H2SO4で反応を停止し、Kontron SLT−20光度計(SLT Lab-instr., スイスチューリッヒ)により492nm(基準620nm)で吸光度を測定する。
本発明のHSP−又はα2M−複合体に対するCD4+T細胞増殖応答は、特定のサイトカインのレベルの検出及び定量により測定することができる。一実施形態では、例えば、本発明の複合体の免疫原性を調べるIFN−γ検出アッセイを用いて、細胞内サイトカインを測定することができる。この方法の一例では、HSP−ペプチド複合体又はα2M−ペプチド複合体で治療する被験者からの末梢血単核細胞を、所与の腫瘍のペプチド抗原又は感染性疾患の因子のペプチド抗原で刺激する。次に、フローサイトメトリーにより検出可能なT細胞特異的標識抗体、例えば、FITC−コンジュゲート抗CD8やPerCP標識抗CD4抗体で、細胞を染色する。洗浄後、細胞を固定し、透過性にし、ヒトIFN−γと反応性の色素標識抗体(PE−抗IFN−γ)と反応させる。標準的方法を用いて、サンプルをフローサイトメトリーにより分析する。
別の実施形態では、「四量体染色」アッセイ(Altmanら、1996, Science 274:94-96)を用いて、抗原特異的T細胞を同定することができる。例えば、一実施形態では、腫瘍特異的抗原のような特異的ペプチド抗原を含むMHC分子を多量体化することにより、可溶性ペプチド四量体を作製してから、例えば、ストレプトアビジンと複合体化させることにより、標識する。次に、MHC−ペプチド抗原複合体を、HSP−又はα2M−複合体で治療する被験者から取得したT細胞集団と混合する。次に、ビオチンを用いて、目的とする抗原、すなわち、腫瘍特異的抗原を発現するT細胞を染色する。
HSP−又はα2M−複合体による免疫療法の、新生物疾患の発症及び進行に対する効果は、当業者にとって既知の任意の方法によりモニターすることができる。その方法には、限定するものではないが、a)細胞性免疫の評価としての遅延型過敏性の測定;b)細胞傷害性Tリンパ球のin vitroでの活性の測定;c)腫瘍特異的抗原、例えば胎児性癌抗原(CEA)のレベルの測定;d)コンピューター断層撮影(CT)スキャン等の技法を用いた腫瘍の形態の変化の測定;及び、e)リスクの高い個体における、特定の癌のリスクに関する推定バイオマーカーのレベルの変化の測定;並びに、f)ソノグラムを用いた腫瘍の形態の変化の測定が挙げられる。
遅延型過敏性に関する皮膚試験は、ある抗原に対する全般的な免疫能及び細胞性免疫において非常に有用である。一群の共通皮膚抗原に対して反応できなくなることはアネルギーと称される(Sato, T.ら, 1995, Clin. Immunol. Pathol. 74:35-43)。
フィコール−ハイパーク勾配遠心法により単離した8×106個の末梢血由来Tリンパ球を、10%ウシ胎仔血清を含む3mlのRPMI培地中にて、4×104個のマイトマイシンC処理腫瘍細胞により再刺激する。場合により、33%の二次混合リンパ球培養物の上清又はIL−2を、T細胞増殖因子の供与源として培地中に含ませる。
全ての腫瘍上に特有の腫瘍抗原を検出するのは不可能かもしれないが、多くの腫瘍が正常細胞とは異なる抗原を提示する。モノクローナル抗体試薬は、抗原の単離や生化学的分析を可能にするとともに、形質転換細胞と形質転換されていない細胞との区別のため、及び形質転換細胞の細胞系統を確定するために、診断上非常に有益である。最も解明が進んでいるヒト腫瘍関連抗原は腫瘍胎児性抗原である。かかる抗原は胚形成の際に発現されるが、正常な成人の組織には存在しないか又は検出することが非常に難しい。この標準的な抗原が胎児性癌抗原(CEA)である。この抗原は、胎児の腸及びヒトの大腸癌細胞に見られる糖タンパク質であるが正常な成人の大腸細胞には見られない。CEAは大腸癌細胞から流出して血清中に見られるので、元々は、血清中のこの抗原の存在は大腸癌の患者をスクリーニングするために用いることができると考えられていた。しかしながら、他の腫瘍(脾臓癌及び乳癌など)を有する患者も血清CEAレベルの上昇を示す。従って、治療を受ける癌患者においてCEAレベルの下降及び上昇をモニターすることは腫瘍の進行及び治療への応答を予測するのに有用であることが分かっている。
癌の正確なステージ分類のための技術の選択肢としてCTがある。CTは転移を検出するための他のいかなるイメージング技法よりも高感度かつ特異的であることが判っている。
特定の癌のリスクに対する推定バイオマーカーのレベルを、サイトゾル及び膜由来タンパク質を含む組成物の効果をモニターするために測定する。例えば、前立腺癌のリスクが高い個体において、血清前立腺特異的抗原(PSA)をBrawer, M.K.ら, 1992, J. Urol. 147:841-845及びCatalonaら, 1993, JAMA 270:948-958に記載の手順により測定する。あるいは、大腸癌のリスクがある個体において、第4.5.3節に記載したように、CEAを測定する。さらに、乳癌のリスクが高い個体において、エストラジオールの16−α−ヒドロキシル化を、Schneider, J.ら, 1982, Proc. Natl. Acad. Sci. USA 79:3047-3051に記載の手順により測定する。上記引用した参照文献は、参照によりその全文を本明細書に組み入れる。
癌の正確なステージ分類のための技術の他に採り得る選択肢としてソノグラムがある。
以下の実験では、(a)細胞画分に由来する抗原ペプチドと、(b)HSP又はα−2−マクログロブリン(α2M)のいずれかとの複合体が、癌細胞増殖から動物を予防的に防御する上で有効であることを実証する。
5.1.1.タンパク質精製
α2Mの精製のために、マウスの血清を0.04M Tris(pH7.6)、0.15M NaClで1:1希釈し、同じバッファーで平衡化した65mlのSephacrylS300R(Sigma)カラムにアプライし溶出を行なった。α2M陽性画分をドットブロットにより測定し、その画分のバッファーをPD−10カラムを使用して0.01Mリン酸ナトリウムバッファー(pH7.5)に交換した。タンパク質含有画分をコンカナバリンAセファロースカラムにアプライした。結合したタンパク質を0.2Mメチルマンノースピラノシドで溶出し、0.05M酢酸ナトリウムバッファーで平衡化したDEAEカラムにアプライした。SDS−PAGEと0.13M酢酸ナトリウムを用いる免疫ブロッティングで分析すると、α2Mは純粋な形で溶出された。
すべての免疫は、100μl容量のPBSで皮内投与により行った。1週間置いて2回の免疫を行った。1回の注射で複合体として又は単独で、7μgのα2M又は1μgのgp96を使用した。肝腫瘍細胞(100,000)から培地を洗い流し、PBSに再懸濁し、最後の免疫の1週間後に皮内注射した。2次元で腫瘍を測定した。平均の半分を腫瘍の半径として、腫瘍容量を計算した。一元分散分析(ANOVA)を使用してP値を決定した。
MethA生細胞からダウンスを用いたホモジナイズと次に超遠心分離により、細胞溶解物を得た。100,000gの上清を0.1%トリフルオロ酢酸(TFA)と3mM ATPで10時間処理し、次にカットオフ限界値10kDaのCENTRICON膜フィルター(Millipore)で遠心分離した。10kDaより小さいペプチド(「MethA10」と呼ぶ)を、C18逆相カラムに結合させ、ペプチドをメタノールで溶出し、ペプチドを真空下で乾燥し、複合体化に適したバッファー中でペプチドを再生することにより、さらに単離した。gp96、α2M、又はアルブミン(これは対照として使用した)を、50モル過剰のMethA10の存在下で、50℃に加熱した。得られる複合体を含有する反応物を室温に30分置き、次に氷上に置いた。遊離の複合体化していないペプチドを、CENTRICON50(Millipore)を使用して除去した。こうして作製した複合体を免疫に使用した。
この実験では、MethA腫瘍モデルを使用して、gp96−ペプチド複合体及びα2M−ペプチド複合体により誘発される抗腫瘍免疫を証明した。この腫瘍の抗原性MHCIエピトープは不明である。MethA細胞溶解物をATPとトリフルオロ酢酸(TFA)で処理し、10kDaより小さいペプチドの画分(MethA10)を採取し、上記したようにα2M又はgp96と複合体化させた。複合体化していないか又はMethA10と複合体化したα2M又はgp96で、BALB/cマウスを免疫した。BALB/cマウスはまた、アルブミン−MethA10又は陰性対照としてPBSで免疫した。免疫は1週間置いて2回行った。最後の免疫の1週間後にすべてのマウスに、100,000個のMethA生細胞を皮内投与することによりチャレンジした。腫瘍増殖を5日ごとにチャレンジの20日後までモニターした。
本明細書に記載の腫瘍に対する免疫についての実験は、腫瘍からの総細胞ペプチドのアレイ(自己及び抗原ペプチドを含む)をHSP又はα2Mと複合体化するという癌の免疫療法に対する新規手法を実証する。そのような複合体は、宿主の免疫系を有効に刺激して、本明細書に記載のように特異的に応答させた。データは、予防におけるこの手法の有用性が、既存の疾患の治療、ならびに病原性感染症の治療及び予防にも拡張できることを示す。
Claims (30)
- 抗原ペプチドと複合体化された1つ以上の熱ショックタンパク質を含む複合体を製造する方法であって、抗原ペプチドの集団を1つ以上の異なる熱ショックタンパク質と、該ペプチドが該熱ショックタンパク質と複合体化する条件下においてin vitroで接触させるステップを含み、ここで、該抗原ペプチドの集団は、少なくとも50種の異なるペプチドを含み、かつタンパク質調製物を複数の反応混合物に分割することによって2以上のプロテアーゼによる消化に供すること、および消化反応混合物をプールすることを含む方法により作製され、その際、各反応混合液は異なるプロテーゼによる消化に供されるものであり、該タンパク質調製物は、(a)抗原性細胞またはその細胞画分に由来し、かつ(b)該抗原性細胞または該細胞画分それぞれの、少なくとも50%の異なるタンパク質または少なくとも50種の異なるタンパク質を含む、上記方法。

- 請求項1〜5、7、8および10〜24のいずれか1項記載の方法によって作製された、抗原ペプチドと複合体化された1つ以上の熱ショックタンパク質を含む複合体の免疫原性量の、あるタイプの感染性疾患を治療または予防するための医薬の製造のための使用。
- 請求項1〜24のいずれか1項記載の方法によって作製された、抗原ペプチドと複合体化された1つ以上の熱ショックタンパク質を含む複合体の免疫原性量の、第2の抗原性細胞に対する免疫応答を被験体において誘導するための医薬の製造のための使用であって、該第2の抗原性細胞は、抗原ペプチドの集団の由来となる抗原性細胞に共通する少なくとも1つの抗原決定基を発現する、前記使用。
