Description
本発明は、痒みを抑制するための薬剤に関するものである。
皮膚疾患の殆どは、痒みを伴うことが一般的である。その痒みには、湿疹や蕁麻疹などのように、抗ヒスタミン剤で代表される市販の鎮痒剤で抑制し得る痒み（一般掻痒）と、血液透析患者の皮膚掻痒症、老人性皮膚掻痒症、並びにアトピー性皮膚炎などのように、抗ヒスタミン剤やステロイド製剤では抑制し得ない痒み（難治性掻痒）とがある（特許文献１を参照されたい）。
特開平８−２６９９３号公報
しかるに、現在一般に知られている鎮痒剤では抑制し難い上記難治性掻痒の治療に関しては、今日の医療現場においても大きな問題の一つとなっている。また、抗ヒスタミン剤などが主体の鎮痒剤は、難治性掻痒には無力であるばかりでなく、内服薬として用いると、眠気など（中枢抑制）の副作用があるといった欠点がある。
本発明は、このような従来技術の問題点を解消すべく案出されたものであり、その主な目的は、一般掻痒は勿論のこと、難治性掻痒にも有効であり、且つ副作用の少ない薬剤を提供することにある。
このような目的を果たすために、本発明においては、痒みを抑制するための薬剤として、パン酵母由来の酵母亜鉛を含有することを特徴とするものとした（請求項１）。特に、この酵母亜鉛は、パン酵母（Saccharomyces Cerevisiae）に亜鉛ミネラルを富化培養して当該パン酵母に亜鉛を取り込ませた後に、菌体を洗浄、分離した上澄みのスラリーから採取すると良い（請求項２）。
本発明の薬剤、即ち、酵母亜鉛を含有する薬剤を内服または外用塗布することにより、一般的な皮膚疾患の湿疹、蕁麻疹などに伴う痒み（一般掻痒）のみならず、従来の鎮痒剤（抗ヒスタミン剤や抗アレルギー剤）で抑制し難い血液透析患者の皮膚掻痒症、老人性皮膚掻痒症、並びにアトピー性皮膚炎などの痒み（難治性掻痒）を、眠気などの副作用を生ずることなく安全に抑制することができ、皮膚症状を有効に予防、改善或いは治療する上に多大な効果を奏することができる。
以下に本発明について詳細に説明する。
一般掻痒および難治性掻痒に対する抑制作用を示す薬剤として、本発明においては、酵母亜鉛を含有するものとした。より具体的に言うと、先ず、パン酵母（Saccharomyces Cerevisiae）に亜鉛ミネラルを富化培養してパン酵母に亜鉛を十分に取り込ませた後、菌体を洗浄、分離した上澄みのスラリーを得る。次に、そのスラリーを加熱殺菌処理した濃縮液を噴霧乾燥もしくは凍結乾燥することにより、酵母亜鉛を含有した粉末製剤を得ることができる。この乾燥粉末を適宜な液剤に分散させることにより、液体製剤を得ることができる。
次に、酵母亜鉛を含有した薬剤の鎮痒効果の検証結果について説明する。
先ず、一般掻痒並びに難治性掻痒を誘発する発痒動物モデルを、マウス、ラット、並びにモルモット等の小動物で作成した。特にモルモットは、掻痒行動や皮膚の掻破痕の観察に最適であった。
より具体的に言うと、健康なハートレー系雄性モルモット（８週齢）の右脇腹の体毛を、バリカン及びシェーバーで実験前日に刈り取り、０．０５ｍｌの発痒物質（一般掻痒は塩酸ヒスタミン、難治性掻痒はブタ脾臓由来カリクレイン）をその部位に皮内投与して発痒させ、それを発痒動物モデルとした。
これにより、市販の鎮痒剤で抑えられる一般掻痒と、市販の鎮痒剤では抑えられない難治性掻痒とに対する鎮痒効果を同様に判定できる発痒動物モデルが得られる。
このモデルを適宜な頭数（例えば１２頭）で群分けし、時間表示付きのビデオカメラにて投与直後より所定時間（２時間）について各群の掻痒行動を真上から撮影しつつ観察し、掻痒行動（発痒部位を口または後肢で掻く）を起こしていた時間を累積した（秒／１２０分）。この際、ビデオカメラの映像から掻痒行動が見られる部分だけを抜き出すように編集すると、データの採取が楽に行える。
その結果、図１に示す通り、ヒスタミン発痒させた直後の累積掻痒行動時間が１８．０±５．１（秒／１２０分）であった第１群に対して１％濃度の酵母亜鉛溶液を塗布したところ、累積掻痒行動時間が１０．４±５．３（秒／１２０分）と、４２．２％の抑制率を示し、酵母亜鉛には顕著な鎮痒効果があることが分かった。
また、図２に示す通り、カリクレイン発痒させた直後の累積掻痒行動時間が４２．２±４．５（秒／１２０分）であった第２群に対して１％濃度の酵母亜鉛溶液を塗布したところ、累積掻痒行動時間が３７．８±４．２（秒／１２０分）と、１０．４％の抑制率を示し、更にカリクレイン発痒させた直後の累積掻痒行動時間が３９．８±５．６（秒／１２０分）であった第３群に対して３％濃度の酵母亜鉛溶液を塗布したところ、累積掻痒行動時間が３１．５±５．３（秒／１２０分）と、２０．９％の抑制率を示した。即ち、抗ヒスタミン剤では全く抑制し得ないカリクレイン発痒に対しても、少なくとも１％以上の濃度の酵母亜鉛溶液により、ヒスタミン掻痒に対するのと略同等の鎮痒効果が得られることが実証された。
本発明による酵母亜鉛を用いる際には、予防や治療に有効な量の酵母亜鉛が製薬学的に許容できる担体または希釈剤と共に製剤化されると良い。その他にも、結合剤、吸収促進剤、滑沢剤、乳化剤、界面活性剤、酸化防止剤、防腐剤、着色剤、香料、甘味料などを添加しても良い。
このような製剤において、有効成分である酵母亜鉛の担体成分に対する配合割合は、０．１〜３０．０重量％の範囲であり、特に０．５〜５．０重量％の範囲が好ましい。
剤形としては、症状に応じて巴布剤、噴霧剤、溶液剤、懸濁液剤、軟膏剤、ゲル剤、ペースト剤、クリーム剤、顆粒剤、細粒剤、錠剤、丸剤、カプセル剤などを挙げることができ、その投与経路としては、貼付、塗布、経口、静脈内、筋肉内、皮下、関節腔など、種々の投与経路を挙げることができるが、特に外用剤が良好である。本発明の物質を皮膚外用剤として用いる場合は、一般の皮膚外用剤に配合される通常の成分を必要に応じて適宜に配合すれば良い。また、有効成分の投与量および投与頻度は、病状、年齢、性別、投与経路などに応じて適宜に変更することができる。
本発明による酵母亜鉛を含有する薬剤は、栄養素を一種以上含む天然物およびその加工物からなるパン或いは菓子類、清涼飲料など、あらゆる飲食物に混入し、機能性食品としても適用可能である。
本発明に係る酵母亜鉛を含有する薬剤は、一般掻痒は勿論のこと、難治性掻痒の抑制が要求される医療現場における治療に適用できる。
Claims (2)
- パン酵母由来の酵母亜鉛を含有することを特徴とする痒みを抑制するための薬剤。
- 前記酵母亜鉛は、パン酵母（Saccharomyces Cerevisiae）に亜鉛ミネラルを富化培養して当該パン酵母に亜鉛を取り込ませた後に、菌体を洗浄、分離した上澄みのスラリーから採取したものであることを特徴とする請求項１に記載の痒みを抑制するための薬剤。
