Description
固相化酵素免疫スポットアッセイ(ELISPOT)とも呼ばれるところのフィルター免疫プラークアッセイは、元来は個々の抗体分泌B細胞を検出し定量するために開発されたものである。この技術が開発されたことによって、従来のプラーク形成細胞アッセイに代わる迅速かつ多目的の方法が提供された。最近の修正は、毎秒約100分子という少量の特定の蛋白質を生産する細胞が検出可能となるまでにELISPOTアッセイの感度を向上させた。これらのアッセイは、蛋白質分泌細胞をとりまく直接の環境での比較的高濃度の特定の蛋白質(たとえばサイトカイン)を利用している。これらの細胞生産物は捕捉され、そして高親和性抗体を用いて検出される。
ELISPOTアッセイは、同じサイトカイン分子上の異なるエピトープに対する2つの高親和性サイトカイン特異性の抗体を用いるもので、それは2つのモノクローナル抗体、または1つのモノクローナル抗体と1つの抗血清の組み合わせである。ELISPOTは、単一細胞によって分泌されたサイトカインを検出する呈色反応に基づくスポットを生成する。このスポットは、もとのサイトカイン生成細胞の「足跡」をあらわす。スポットは永久的なものであり、肉眼で、顕微鏡で、または電子的に定量することができる。
ELISPOTアッセイは5つの特異的なステップを含んでいる。すなわち(1)精製したサイトカイン特異的抗体を、ニトロセルロースで裏打ちしたマイクロタイタープレートに被覆すること;(2)そのプレートを、他の蛋白質の非特異的な吸収から防ぐためにブロックすること;(3)サイトカイン分泌細胞を、種々の異なる希釈度で培養すること;(4)標識された第二の抗サイトカイン抗体を加えること;および(5)抗体-サイトカイン複合体を検出すること。
本発明において、その手法は、特定のペプチドに対してインビボで前感作されたペプチド特異的T細胞のアッセイを開発するために用いられてきた。
すなわち本発明は、ペプチド特異的エフェクターT細胞が計数されるアッセイ方法であって、前記アッセイ方法が以下のことを含み、前記アッセイ方法がB型肝炎、C型肝炎、結核、マラリア、HIVまたはインフルエンザの如き細胞内病原体の感染の診断または監視に応用されるアッセイ方法を提供する:
インビトロで培養されていない新鮮なT細胞を含む液を準備し、サイトカインインターフェロン−γに対する固体化された抗体を担持する表面と前記T細胞を含む液を接触させ;
T細胞をT細胞活性化ペプチドに与え;
液を培養して前記サイトカインを放出させ、そして前記固定化された抗体に結合した、放出されたインターフェロン−γを検出し、ただし前記培養は、ペプチドに対してインビボで前感作されているT細胞であってペプチド存在下のインビトロ培養で分裂/分化を生じる必要のない即時型エフェクター機能の能力のあるT細胞だけによって、サイトカインの放出を可能にするような時間の間、続けられる。好ましくはこの方法は、サイトカインに対する固定化された第一の抗体を担持する表面とT細胞を含む液を接触させ、ペプチドをその液へ加え、得られた液をサイトカインを分泌するようにペプチドに対してインビボで前感作された(pre-sensitised)ペプチド特異的T細胞を生じる条件下で培養し、そして固定化された第一の抗体と結合した分泌サイトカインを検出することから成る。
細胞は、好ましくは末梢血液単核細胞(PBMC)である。それは、たとえばウイルスのような細胞内病原体による感染にかかっているか又はかかったことが知られている患者から適宜採取することができる。新鮮な細胞を用いるのが本発明の実施態様である。何故ならば、インビトロで培養した細胞は、変化した特性が進行することがあり、すなわちアッセイの診断的価値を減少させるからである。このアッセイの目的は、特定のペプチドに対してインビボで前感作するかまたは活性化されたペプチド特異的T細胞、たとえばCD8+またはCD4+細胞を同定し定量することである。これらは非再賦活性T細胞、すなわちインビトロ培養で分裂/分化を生じる必要のない即時型エフェクター機能の能力のある細胞である。問題のペプチドがそのような細胞に存在するときは、それらの細胞は種々のサイトカインを分泌し、それらのうち何れか1つが本アッセイの目的のために選択される。選択されるサイトカインは、インターフェロン-γ(IFN-γ)である。
分泌されたサイトカインは、文献で公知の種々の方法のいずれかによって検出することができる。好ましくはこのアッセイ方法は、IFN-γまたはその他のサイトカインに対する固定化された第一の抗体を担持する表面を提供することを含んでいる。PBMCまたはその他の新鮮な細胞を含む液を、固定化抗体と接触するところへおく。PBMCの約30%がCD8+細胞である。かつてインフルエンザウイルスに感染し回復した患者のPBMCの中では、105から106の中の約1コのCD8+細胞が、インフルエンザウイルスと関連する特異的なエピトープに対して前感作されたことのある記憶細胞である。
本発明の方法は、ペプチドを液に加えることを含む。ペプチドは、よく特徴づけられたウイルス感染の公知のエピトープであることができるし、またはあまりよく特徴づけられてないウイルス感染とたぶん関連するエピトープ候補であってもよい。得られた液の混合物は、インビボでその特定のウイルスから誘導されたペプチドに対して前感作されているようなペプチド特異的T細胞を賦活するような条件下で培養する。このペプチドは、CD8+細胞によって認識される長さ、たとえば7-15個か特に8-12個または8-10個のアミノ酸残基長である必要がある。CD8+細胞(およびその他のPBMC)の多くは、ペプチドに対して前感作されたであろう少数のCD8+細胞に対するペプチドを提供するものと考えられる。もしそのような活性化され又は前感作されたペプチド特異的T細胞が検査液の中に存在すれば、それらはIFN-γまたは他のサイトカインを分泌することによって応答し、それはついで固定化された抗体に結合する。
ペプチドは、非結合の形で新鮮な細胞へ加えられることが望ましい。ペプチドでパルス標識した培養細胞を加えることも可能ではあるが、これは定められたペプチドエピトープを使用したときには必要ではない。ペプチドは、観察可能なシグナルを生じるに充分な量だけ加えるべきである。液中の好ましい濃度範囲は0.01から100μM、とくに0.5-5.0μMである。
培養は、選ばれた特定のペプチドに対してインビボで前感作してあるCD8+細胞がIFN-γまたはその他のサイトカインを分泌できるに充分な時間だけ続けるべきである。培養は、非活動的なCD8+細胞が分化しそしてペプチドで活性化されサイトカインを分泌し始める程度の長時間にわたって続けるべきでない。これは、培養時間は4-24時間、さらに好ましくは6-16時間であることを示唆する。テストの中の培養の部分は、インビトロでの細胞培養に必要とされる無菌条件を用いずに1労働日または1晩で行なうことができるのが本発明の利点である。
培養中に、IFN-γやCD8+によって分泌される他のサイトカインの何れもが、表面で固定化した第一の抗体と結合するようになる。培養後、その表面を洗って非結合材料を除去する。検出のためには、サイトカインに対して標識した第二の抗体を用いるのが好ましい。この方法を表面に適用すると、該抗体はそこに存在する如何なるサイトカインとも結合するようになる。第二の抗体は好ましくは、第一の抗体とは異なるエピトープを認識すべきである。第一および第二の抗体の片方または両方は、好ましくはモノクローナルであるべきである。標識は、質量分析または屈折率(たとえば表面プラスモン共鳴)で検出するための蛍光基、色彩か化学ルミネッセンスを生じる酵素、放射性同位元素を含む領域でふつうに使用されるもののうち如何なるものであってもよい。標識された抗体を使用するのは便宜ではあるが必要ではない。サイトカインと特異的に結合する如何なる試薬も、標識され使用されることができる。標識の検出および多分定量は、この分野でよく知られており、用いた標識の性質に適応した手段で行われる。
アッセイは、マルチウエルのプレートで便宜に行なうことができる。プレートの各々のウエルは、結合した第一の抗体を担持した表面をもっている。各々のウエルへ、適当な数の、たとえば103から106個の細胞を含んだ液を加える。異なったペプチド及び/又は対照を、プレートの個々のウエルに加える。培養中にサイトカインを分泌する細胞は、スポット(スポット形成性細胞すなわちSFC)として現われ、そして各々のウエル中のこれらの数や密度は容易に測定することができる。
このアッセイ技術は、公知の技術にくらべ次のような多くの利点をもっている。
a)それは、一層迅速で便利である。アッセイの期間はわずか6時間であって、滅菌の条件や技術を必要としない。前駆物質エフェクターT細胞を数え上げる現在の方法は、限定希釈アッセイ(LDA)での自己支持細胞と特定の抗原でのインビトロ培養を必要とする。そのような方法は煩雑で時間がかかる。
b)それは、最小の技術装置を必要とするのみで、熱帯地域や開発途上国ならびに通常の診断ラボでの分野条件に適している。これと対照的にLDAは、細胞を1-2週間培養せねばならないので、多くの末梢血液リンパ球と、支持細胞を不活性化するためのガンマ線照射源と、滅菌条件を必要とする。
c)それは、安全で非放射性である。しかしLDAの場合は、培養された細胞は、放射性同位元素であるクロム-51を用いた細胞毒性T細胞アッセイ(CTL)でアッセイが行われる。
d)それは、直接のエキスビボ(ex vivo)アッセイである。したがってそれは、細胞が抗原と外因性のサイトカインと共にインビトロ培養を増殖させるときに生じる未知のバイアスの導入なしに天然の状態でのエフェクター細胞を測定する。
e)このアッセイはわずか6時間で行われる。すなわちペプチド特異的エフェクター細胞を直接測定するもので、これらの細胞のインビトロでの増殖を必要としない。アッセイが短時間であるということはまた、細胞がインビトロで活性となる可能性をなくすることである。したがって、インビボで存在するエフェクター機能を測定することになる。LDAは、何倍にも増殖する細胞を必要とする。しかし多くのエフェクター細胞はこういった条件では増殖せず、そのためLDAの結果は、循環するエフェクターの真の数よりしばしば低くなる。
このアッセイ技術は、いろんな違った方面で価値があると期待されている。
i)まず、直接に末梢血液からのHIVに感染した個人におけるペプチド特異的エフェクターの定量のためである。
ii)また、たとえば薬物または治療ワクチンへの応答のような、慢性の感染性疾患の進展またはそれに対する耐性の監視のためである。これは、HIV、B型肝炎、およびC型肝炎において特に有用であることが期待される。
iii)また、たとえばインフルエンザのような特定の病気に罹患している患者の、将来の感染に対する抵抗性の程度を監視するためである。
以下の例は、本発明を説明するものである。
例1.インフルエンザウイルスへの免疫学的記憶:記憶状態での循環ペプチド特異的CD8+活性化T細胞のエキスビボ計数と特性化
被験者は、補体仲介リンパ球による血清学的にHLAタイプの、健康な研究所の個人または健康な成人のボランティアであった。5つのMHCクラスのI-制限インフルエンザエピトープが用いられ、それらを表1に示した。
96ウエルの、PVDFで裏打ちしたプレートを、4℃で一晩または室温で3時間、15μg/mlの抗IFN-γ Mab 1-DIKの100μlで被覆した。プレートを洗浄し、次いで室温で1時間、R10(ウシ胎児血清10%を含む標準組織培養培地)でブロックした。被験者のPBMCを、ヘパリン化した全血から遠心分離により分離し、R10に再懸濁し、そして96ウエルのPVDF裏打ちマイクロタイタープレートへ、最終容量がウエルあたりのR10が100μlとなるように加えた。添加する細胞の数は通常はウエルあたり5×105であり、すべてのアッセイは2つのウエルで行なった。ペプチドは、M1 58-66ペプチド濃度を20nMに希釈したペプチド滴定実験1件を除き、通常は1-2μMの最終濃度となるように添加した。アッセイは通常12-14時間行われたが、あるアッセイは抗原特異性細胞が直接のエフェクター機能が可能であることの確認のために6時間行われた。培養は、5%のCO2を含む雰囲気中、37℃で行われた。培養は、ウエルの内容物を振盪し洗浄することによって停止させた。次いでウエルに、1μg/mlのビオチン化した第二の抗IFN-γ MAB 7-B6-1-ビオチン(スウェーデン、ストックホルム、Mabtech)を100μl加え、プレートを3時間培養した。ストレプトアビジン-アルカリホスファターゼ結合物の1:1000希釈物の100μlをウエルに加え、プレートを室温でさらに2時間培養した。ウエルを再び洗浄し、脱イオン水で1:25に希釈したクロモジェニックなアルカリホスファターゼ100μlをウエルに加えた。室温で更に30-60分培養した後、ウエルを洗浄して呈色反応を終了させた。スポットを、解剖用立体顕微鏡を用い20倍の倍率で計数した。
結果
5つの良く規定されたHLAクラスI-制限インフルエンザエピトープを用いて、遊離のペプチドを、ELISPOTアッセイにおいて新しく単離されたPBMCへ最終濃度2μmになるように添加した。この方法でテストした個体の殆どすべてにおいて、個体に存在するHLAクラスI対立遺伝子で制限されたエピトープを用い、IFN-γを分泌するペプチドに特異的なエフェクターT細胞が検出された。表1は、これら5個のエピトープに対する応答をまとめたものである。これらのアッセイの多くは、12-14時間おこなわれ、そして図1はウエルごとの異なる濃度のPBMCにおける応答を示す棒グラフである。しかしながら、記憶T細胞が14時間のアッセイの途中において増殖するか又はインビトロで活性化される可能性を排除するために、6時間のアッセイもまた行われた。ペプチド特異的SFCが、図2に定量的に同じM1 58-66エピトープについて示したようにして検出された。負の対照として、ドナーが感染してない感染源からの無関係のペプチドを、新鮮なPBMCに直接加えた。
ほとんどの実験は、最終ペプチド濃度2μmで行なわれた。しかしながら、インフルエンザHLA-A2.01-制限マトリックスエピトープについて図3に示したように、ペプチド濃度が0.02μmまで減少したときにも、まだ応答は容易に検出可能であった。
新鮮なPBMCからの、抗CD8抗体被覆した磁気ビーズによるCD8+T細胞の消耗は、ペプチド特異的応答を完全に終わらせるものであり、これは公知のクラスI-制限エピトープで引き出されたスポットを生じるエフェクターがCD8+Tリンパ球であることを示すものである。これと逆に、CD4+細胞の消耗はIFN-γ SFCの数を減少するものではなく、これはCD4+も又それらのサイトカイン生産物も、新しく単離したペプチド特異的CD8+T細胞によるエフェクター機能の獲得または配置には必要でないことを示す。直接のエフェクター応答は、テストした特定のドナーに存在するHLAクラスI対立遺伝子で制限されたインフルエンザエピトープに対してのみ検出された。ドナーに存在しないHLAクラスI分子で制限されたインフルエンザエピトープの添加は、まったくSFCを生じなかった(データは示してない)。
急性インフルエンザのときのペプチド特異的CD8+エフェクターCTLの増大は、新鮮な未培養のPBMCで行なった公知の51Cr放出細胞毒性アッセイという手段により細胞を検出可能とするけれども、急性の病気から回復した後は、そのような細胞はもはや検出不可能である。これは、それら細胞が存在しないのではなく、むしろ検出するには余りにも低い頻度で存在しているからであると思われる。急性の病気から回復した後は、そのような細胞は本発明のELISPOTアッセイ技術でまだ検出可能である。
倍率をかけてのスポットの計数、およびこの数と新鮮なPBMCの添加数との比較は、末梢血液で循環している活性化ペプチド特異的CD8+エフェクターの相対的頻度の尺度となる。ドナーWBのHLA-A2.1-制限エピトープM158-56に対するIFN-γ-分泌CD8+エフェクターの頻度は、本発明のアッセイ(1/15000)および従来の限度希釈法(LDA)(1/103000)で測定された。
上の実験は、J. Exp. Med., 186, 6, September 15, 1997, 859-865に詳しく記載されており、それは参照としてここに含まれる。
例2.末梢血液から直接行われる、HIV-感染個人におけるペプチド特異的エフェクターの定量への応用
患者868の末梢血液から新しく単離され冷凍保存された末梢血液リンパ球(PBL)を、抗インターフェロン-ガンマモノクローナル抗体であらかじめ被覆した、PVDF被覆の96ウエルのプレートに、1ウエルあたり50,000細胞の割合で平板培養した。各々の抗体について副のウエルを設けた。2種類の副の対照ウエルを用いた。すなわち、患者868にはなかったHLA対立遺伝子であるHLA-B8で制限された、ペプチドがなく無関係なHIVgagエピトープである。
ある範囲のペプチドおよびその対応する天然に存在する変種(患者868において既に同定されている)を、最終濃度が2μMとなるように細胞に直接添加した。これらのプレートを、5%のCO2中37℃で12時間培養し、既に述べたようにして展開させた(例1参照)。得られたスポットを、40倍の解剖顕微鏡で計数した。これらの結果は、図4に表の形で表わした。
例3.エム・ツベルクロシス(M. Tuberculosis)の分泌抗原でのCD4+およびCD8+エピトープの同定
結核菌(ミコバクテリウム・ツベルクロシス(Mycobacterium tuberculosis))感染においてCD8+細胞毒Tリンパ球が保護的役割を果たすことを示す証拠が増大しているものの、結核菌特異性のCD8+T細胞はこれまでヒトでは同定されていなかった。逆の免疫遺伝学的アプローチを使って本発明者等は、結核菌のESAT-6および抗原85という2つの免疫優勢(immunodominant)抗原からHLAクラスI制限ペプチド候補の配列を合成した。本発明者等は、結核菌感染の広範な臨床スペクトルを表わす75の症例をスクリーニングした。末梢血液リンパ球はペプチドによってインビボで賦活され、そして51Cr放出アッセイでの細胞溶解活性と、ELISPOTアッセイでの単一細胞インターフェロンγ放出について試験された。本発明者等は、患者や露出接触のESAT-6や抗原85から幾つものオクタマーやノナマーエピトープを同定した。ある種のエピトープは、MHCクラスI制限の態様でCD8+リンパ球によって認識され、その他のものはCD4+細胞によって優先的に認識された。
ESAT-6ならびに抗原85A、BおよびCの配列は、HLAクラスIのタイプA2、B7、B8、B35、B52およびB53(それらは全て研究個体群中に存在する)に関して、対立遺伝子特異性のペプチドモチーフを用いて調べた。
ESAT-6に関しては、HLA-A2、B8およびB52のペプチドモチーフに適合した配列が同定された。これらのペプチドは合成され表2に示してある。HLA-B7、-B35および-B53に適合する配列はESAT-6の中には存在しなかったので、これらHLAクラスI対立遺伝子のためのペプチドは合成されなかった。ペプチドは、ドナーPBMCのインビトロでの再賦活に用いられたプールへ分類された。CD8+エピトープであると判明したペプチドは、太字で示してある。同様にして、抗原85Aおよび85Cの配列に基づいて、42個のペプチドを合成した。これらの中にはCD8+エピトープは同定されず、該ペプチドは示されていない。
IFN-γのためのELISPOTアッセイ
抗IFN-γ mAb 1-DIK 15μg/mlで前被覆した96ウエルのPVDF裏打ちプレートを、RPMIで洗浄し、R10で1時間室温でブロックした。1つの実験では、1ウエルあたり500,000の新しく単離した未培養のPBMCを用いた。別の実験では、短期間細胞系(STCL)またはCD8+細胞毒Tリンパ球(CTL)またはクローンをRPMIで2回洗浄し、R10に再懸濁し、そしてウエルあたり既知の細胞添加数で2つのウエル中へ分散した。応答は、もし最低10個のSFCがウエルごとに存在し、更にこの数が対照のウエルのものより少なくとも2倍であれば、有意であるとみなした。ペプチドは、最終濃度が2μl(遊離ペプチド)となるように上澄液へ直接加えた。プレートを5%CO2、37℃で12時間培養した。0.05%のTween-20含有リン酸塩緩衝食塩水で6回洗浄して細胞を除去し、プレートを、1μg/mlの第二のビオチン化抗IFN-γ mAb 7-B6-1-ビオチンで3時間培養した。上記のような洗浄を行ない、ついで1:1000に希釈したストレプトアビジン-アルカリホスファターゼ結合物で2時間培養した。さらに洗浄してから、クロモジェニックなアルカリホスファターゼ基質をウエルに添加し、30分後に、プレートを水道水で洗浄した。乾燥後、スポット形成細胞(SFC)を20倍の倍率下で計数した。
STCLは、Nature, 346(1990)183-7に記載の方法で生成された。CD8+T細胞クローンは、標準的方法で生成された。
ESAT-6特異的エフェクターT細胞の、末梢血液からの直接同定
ESAT-6中の2つのCD8+エピトープを同定した。結核縦隔リンパ節炎のドナーNPH54からのT細胞は、これら双方のエピトープに対応するペプチドを認識した。HLA-B52とHLA-A2.01をもつNPH54からの診断時に単離した未培養のPBMCは、エクスビボELISPOTアッセイにおいてこれらのクラスI対立遺伝子のためのESAT-6誘導ペプチドプールに応答するIFN-γを分泌した。複製ウエルで5×105個のPBMCから計数したIFN-γスポット形成細胞(SFC)の平均数は、ペプチドのない対照ウエルの場合のESAT-6ペプチドが2個であるのに対して、19個であった。応答するプールの中での個々のペプチド各々に対して、新しく単離したPBMCの引き続いてのアッセイが行われた。IFN-γ SFCはES12とES13ペプチドの応答の中で検出され、それらの配列はHLA-B52とHLA-A2.01ペプチドモチーフとそれぞれ適合していた。ES12-とES13-特異性IFN-γ SFCの頻度は、HLA-A2.01制限インフルエンザマトリックスエピトープM1 58-66のSFCとしての大きさと同じ程度であった。結核性骨髄炎の第二のドナーNPH97からの非再賦活PBMCもまた、ES12エピトープを認識した。この患者はまたHLA-B52と-A2.01をもっており、ES12特異的応答の大きさはHLA-A2制限インフルエンザマトリックスエピトープに対する応答と類似していた。同種のペプチド以外の刺激を用いたこれら12時間の短いエクスビボアッセイで新しく単離したT細胞による単一細胞IFN-γ放出は、これらの細胞はほとんど循環活性エフェクターT細胞であろうことを示している。
Claims (10)
- ペプチド特異的エフェクターT細胞が計数されるアッセイ方法であって、前記アッセイ方法が以下のことを含み、前記アッセイ方法がB型肝炎、C型肝炎、結核、マラリア、HIVまたはインフルエンザの如き細胞内病原体の感染の診断または監視に応用されるアッセイ方法:
インビトロで培養されていない新鮮なT細胞を含む液を準備し、サイトカインインターフェロン−γに対する固定化された抗体を担持する表面と前記T細胞を含む液を接触させ;
T細胞をT細胞活性化ペプチドに与え;
液を培養して前記サイトカインを放出させ、そして前記固定化された抗体に結合した、放出されたインターフェロン−γを検出し、ただし前記培養は、ペプチドに対してインビボで前感作されているT細胞であってペプチド存在下のインビトロ培養で分裂/分化を生じる必要のない即時型エフェクター機能の能力のあるT細胞だけによって、サイトカインの放出を可能にするような時間の間、続けられる。 - エム.ツベルクロシス(M.tuberculosis)のESAT−6由来のペプチドがT細胞に対して与えられる、請求の範囲第1項の方法。
- T細胞が末梢血液単核細胞である、請求の範囲第1項または第2項の方法。
- 7-12個のアミノ酸残基の長さのペプチドがT細胞を含む液に加えられ、これはCD8+細胞によって認識される、請求の範囲第1項から第3項までのいずれかの方法。
- 得られた液混合物が非滅菌条件下で培養される、請求の範囲第1項から第4項までのいずれかの方法。
- ペプチドが公知のエピトープである、請求の範囲第1項から第5項までのいずれかの方法。
- 培養が4-24時間の間続けられる、請求の範囲第1項から第6項までのいずれかの方法。
- T細胞が、細胞内病原体による感染に罹患しているか又は罹患したことがわかっている患者から採られたものである、請求の範囲第1項から第7項までのいずれかの方法。
- HIV感染の進行を監視するために行われる、請求の範囲第1項及び第3項から第8項までのいずれかの方法。
- ワクチンの効果を監視するために行われる、請求の範囲第1項から第9項までのいずれかの方法。
