Description
幹細胞は、自己再生および成熟体組織への分化の両方の能力を有する前駆細胞である。
胚幹細胞は、原型幹細胞であり、それは成体動物に見られる全般の細胞型を形成するために分化可能である。このような幹細胞は、多くの細胞型に分化可能な多能性として記載されている。幹細胞の他の型、例えば、骨髄幹細胞および表皮幹細胞は、成体動物中に存続している。これらの幹細胞はより限定された分化能力を有する。
一般に、研究目的または医療使用に必要である場合、幹細胞は種々の分画手順により組織試料から単離されなければならないが、注意深い細胞型の分離の後でさえ、これらの幹細胞調製物は、幹細胞に富んでいるが、混合した細胞型からなり、幹細胞として分類されない高割合の分化した細胞を含有する。
さらに、大部分の幹細胞は培養液中で容易に培養され得ず、そして幹細胞を培養する試みがなされる場合、培養される細胞(通常、細胞型の混合集団を含む)は、異なる速度で成長し、そして幹細胞は容易に非幹細胞型により過剰成長する。マウスの2つの特異的な株(129およびBlack6)由来の胚幹細胞がインビトロで培養され得ることは除外する。従って、胚幹細胞の樹立株は、ネズミ129株およびBlack6株、またはそのハイブリッド由来の初期(3.5日)胚細胞の培養により得られ得る。
インビトロで胚幹細胞を、他のネズミ株から、およびさらに特別には、他の実験動物(例えば、ラット、ウサギ、およびモルモット)、家畜動物(例えば、ヒツジ、ヤギ、ウマ、ウシ、ブタ、鳥、魚など)、および霊長類を包含する他の種から、単離および維持を強いる必要性が出てきた。同様に、造血幹細胞のような他の多能性細胞についての多くの医療適応もまた、それらの単離およびインビトロでの培養を必要とする。
しかし、従来、十分に低い程度に不均一な細胞集団を生産する問題および非多能性細胞の培養での過剰成長の問題を包含する幹細胞培養物の生産に関する問題は、解決されていない。特定の分化細胞型の存在の存続に関する特別な問題は、それらの分化またはプログラムされた細胞死を誘導することにより培養物から幹細胞を除去し得ることである。
本発明者らは、現在、前記の問題を解決し得ることにより技術を開発した。
本発明の1つの局面によれば、動物幹細胞を単離するおよび/または富化するおよび/または選択的に増殖する方法を提供し、この方法は、細胞の生存の助けとなる培養条件下で上記細胞の供給源を維持することを包含し、細胞の供給源は、(a)幹細胞および(b)所望の幹細胞以外の細胞で特異な発現が可能な選択マーカーを含む細胞を包含することで特徴づけられ、それにより、上記選択マーカーの特異な発現は、所望の幹細胞の選択的な単離および/または生存および/または分割を生じる。本発明の文脈において、用語「動物細胞」は、全ての動物細胞、特にヒト細胞を含む哺乳動物種の細胞を包含することを意図する。
幹細胞の例には、単能性および多能性の両方の幹細胞、胚幹細胞、性腺幹細胞、体幹/前駆細胞、造血幹細胞、表皮幹細胞、および神経幹細胞を包含する。
本発明の方法の実施において、細胞の供給源は陽性選択マーカーを有する多能性細胞を包含し得、そして上記マーカーの発現は多能性細胞の単離および維持を可能にするために使用される。あるいは、細胞の供給源は、所望の多能性細胞以外の細胞で発現する陰性選択マーカーを包含し得、そして所望の多能性細胞以外の細胞の細胞供給源を枯渇するために選択的に用いられる。
選択マーカーは、例えば、外来遺伝子、細胞性遺伝子、または抗生物質耐性遺伝子(例えば、細菌性ネオマイシン耐性遺伝子)であり得る。
あるいは、選択マーカーは成長刺激遺伝子、例えば、不死化遺伝子、オンコジーン、またはポリオーマもしくはSV40T抗原またはその誘導体をコードする遺伝子であり得、あるいは選択マーカーは成長因子または成長因子レセプターまたはシグナル形質導入分子または細胞死を阻止する分子をコードする遺伝子であり得る。
1つの特定の実施態様では、所望の多能性細胞の単離および/または富化および/または選択的な増殖は、所望の多能性細胞以外の細胞の存在に依存し、そして両方の細胞型の同時の持続は、一方または他方の細胞集団での選択マーカーの発現に依存し、それはマーカーを発現しないがマーカーを自己発現する隣接の細胞を助ける能力を有する。この例では、選択マーカーは、例えば、ヒポキサンチンホスホリボシルトランスフェラーゼ(HPRT)遺伝子であり得る。
他の実施態様では、選択マーカーは、毒性そのものである産物をコードする遺伝子、または自殺基質との組合せで条件により活性である毒性遺伝子産物であり得る。このような遺伝子産物の例は、ガンシクロビル(ganciclovir)と組合せた単純ヘルペスウイルスチミジンキナーゼ(HSV-TK)である。
選択マーカーの発現は、細胞供給源に導入する前に、選択マーカーを発現構築物に作動可能に挿入することにより達成され得、この場合、選択マーカーの発現は、安定に組み込まれた構築物または一過的に組み込まれた構築物のいずれかの誘導から生じ得る。あるいは、選択マーカーの発現は、細胞供給源の内因性遺伝子に選択マーカーを作動可能に挿入することから生じる。
選択マーカーの種々の導入手段が使用され得、これにはトランスフェクション、リポフェクション、注入、衝撃ミサイル、ウイルスベクター、またはエレクトロポレーションによる細胞への導入を包含する。
細胞の供給源は稔性化卵母細胞のような単細胞であり得、あるいは、それは、正常に成長したトランスジェニック動物の胚、血液、または体組織、または正常に成長した細胞株由来の細胞のような細胞混合物を包含し得る。後者の場合では、選択マーカーをトランスジェニック動物のゲノムに取り込み得る。
最も好ましくは、本発明の実施で、選択マーカーが作動可能に結合し、そして選択マーカーの発現を調節する遺伝子または遺伝子フラグメントは、細胞発達の多能性段階に関連する。このような遺伝子または遺伝子フラグメントは、発達している胚の多能性細胞、特に内細胞塊および/または初期外胚葉で活性であり得、あるいは、成熟幹細胞で活性であり得る。
本発明に従って使用するための細胞供給源の調製において、以下のプロトコルの1つを有利に適応し得る:
−幹細胞を含有する細胞供給源に、選択マーカー構築物を導入すること、ここで、上記選択マーカー構築物は、上記特異な発現を提供する内因性遺伝子に作動可能に連結するために適応される、または
−幹細胞を含有する細胞供給源に、選択マーカー構築物を導入すること、ここで、上記選択マーカー構築物は、上記特異な発現を提供する1つまたはそれ以上の遺伝子フラグメントに作動可能に連結されている。
遺伝子マーカーは、好ましくは、所望の多能性細胞で特異に活性であるプロモーターに作動可能に連結した選択マーカーを包含する(例えば、初期外胚葉)。「選択マーカー」は、標的細胞集団で、外来遺伝子、または天然に発現する細胞性遺伝子、または天然に発現するが不適当なレベルであるような遺伝子であり得る選択遺伝子を意味する。用いるこの遺伝子は、標的細胞集団の表現型に適応することにより、選択マーカーとして働き、このように適応した表現型を有する細胞は、特定の培養条件下で富化または枯渇し得る。
幹細胞が胚細胞である場合では、選択マーカーが幹細胞(例えば、初期外胚葉、原基生殖細胞)および非幹細胞で特異に活性であるプロモーターに作動可能に連結されることが好ましい。プロモーターおよびその他のシス調節エレメントは、細胞に導入する前またはプロモーターがない構築物を部位特異的な組換えによる特異的な遺伝子に標的することにより、発現構築物に含まれ得る。
多様な遺伝子産物は、所望の幹細胞の選択的な単離および増殖に依存し得、培養液中に存在する阻害因子の影響から所望の細胞を防御するために設計されるマーカーを包含している。この例では、阻害因子は、例えば、培養細胞(すなわち、所望の細胞以外の細胞)の成長または再生を阻害し、遺伝子を発現しない抗生物質であり得る。選択マーカー(例えば、HPRT)はまた、それが発現される所望の細胞および代謝的救済によるその他の密接に関連した細胞の両方についての防御を提供し得る。
あるいは、選択マーカーは、幹細胞の成長を選択的に可能にし得る。この例では、マーカーは成長因子、成長因子レセプター、転写因子、不死化またはオンコジーン産物(例えば、温度感受性シミアンウイルス40T抗原)をコードし得る。
あるいは、選択マーカーは、例えば、パンニングまたは蛍光活性化細胞分類(FACS)による、発現細胞の精製または枯渇を可能にする細胞表面抗原またはその他の遺伝子産物であり得る。従って、本発明は、幹細胞集団を十分な程度の相同性を有しながら獲得/維持することを可能にする。

遺伝子マーカーは、注入、トランスフェクション、リポフェクション、エレクトロポレーションを含む種々の手段により、またはウイルスベクターの感染により、細胞供給源に導入され得る。
さらに、細胞供給源は、即席のトランスフェクションにより生産され得、あるいは細胞供給源は、トランスジェニック動物(例えば、創始トランスジェニック動物)、またはその遺伝子補足物に導入した前述の遺伝子マーカーを有する少なくとも1匹の祖先がいる動物に由来し得る。このようなトランスジェニック動物では、マーカーが、生殖系列を代々伝えそして必要な細胞供給源が由来し得る組織から(特に、胚組織から)、最終的に子孫を生産することが可能である。
従って、本発明のさらなる局面によれば、幹細胞の単離および/または富化および/または選択的増殖を可能にするように適切な選択培養条件下で培養され得る動物細胞を提供し、上記細胞が選択マーカーを包含することで特徴付けられ、ここで、(a)所望の幹細胞および(b)所望の幹細胞以外の細胞での選択マーカーの特異な発現が、所望の幹細胞の選択的な成長が生じることを可能にする。
本発明はさらに、幹細胞として成長するよう選択培養条件下で培養され得る動物細胞を提供し、上記細胞が、遺伝子マーカーを含む幹細胞を包含することで特徴付けられ、それにより遺伝子マーカーと関連した遺伝子産物が生産され、そして上記培養条件下で、所望の幹細胞の選択的生存および/または分割を生じさせる。
本発明のこの局面によれば、動物細胞は、好ましくは上記の好ましい特徴を有することにより特徴付けられる。
本発明はさらに、本発明の他の局面によれば、トランスジェニック動物を提供し、これはこの動物またはその子孫が、胚発達またはその後の寿命の間、上記の定義のように動物多能性細胞の供給源を構成するような遺伝的特徴を有する。
このようなトランスジェニック動物を、インビトロで稔性化卵母細胞または胚細胞、または胚幹細胞に遺伝子マーカーを導入することにより、ここでこの遺伝子マーカーは上記の特徴を有しており、そして所望のトランスジェニック動物の前駆細胞として得られる形質転換された卵母細胞または胚細胞を利用することにより、本発明に従って生産し得る。
上記の動物細胞の生産で用いるためのベクターは本発明のさらなる局面を形成する。
従って、本発明は、さらに、上記の方法のための細胞供給源としての使用に適切な形質転換細胞を生産するように、動物細胞の遺伝学的な改変に用いるためのベクターを提供し、上記ベクターは、上記選択マーカーに対応する第一の遺伝子成分および第二の遺伝子成分を包含し、該第二の遺伝子成分は遺伝学的に改変された動物細胞において、(1)一過的に組み込まれた構築物または安定に組み込まれた構築物のいずれかに由来の選択マーカーの上記特異な発現を生じ、あるいは(2)選択マーカーと標的化内因性遺伝子調節エレメントとの作動可能なカップリングを提供するような、特異的な遺伝子への選択マーカーの部位指向的組み込みを可能にする。
このようなベクターは、発現ベクターの形態であり得、ベクター内で上記第二の遺伝子成分は、制御配列を包含し、その配列は、(a)幹細胞でおよび(b)所望の幹細胞以外の細胞で特異に活性化される。
本発明は、動物細胞を形質転換するに用いる場合に動物ゲノムに組み込まれるベクターおよびそのように組み込まれないベクターを包含する。
上記の発現ベクターは、遺伝制御エレメントに作動可能に連結した上記の選択マーカーをコードするDNA配列、またはプロモーターのないマーカーの上記幹細胞および所望の幹細胞以外の細胞で特異に発現する内因性遺伝子に対する標的を可能にする配列を包含し得る。
多能性胚幹細胞を生じるために、発現構築物は、好ましくは、遺伝制御エレメントに作動可能に連結したまたは標的する前記選択マーカーをコードするDNA配列を包含し、この遺伝制御エレメントが多能性胚細胞の分化前に、胚発達の段階と関連する。最も好ましくは、遺伝制御エレメントは、マウス胚盤胞の内細胞塊で、初期外胚葉で、および初期胚の原基生殖細胞で特異的に活性な遺伝子に由来する。
さらに詳細には、本発明では、幹細胞で選択マーカーを特異的に発現しそして分化した細胞型で発現しない発現構築物の発達を得た。トランスフェクションによりまたはトランスジェニック動物の発生を介することにより発現構築物を導入すると、混合細胞集団内に存在する幹細胞は、インビトロで選択剤の存在下で培養することにより、または培養条件を他に操作することにより単離され得る。
適切に制限された幹細胞発現パターンを示し、そしてそのために、本発明による選択マーカーの発現のために適切な「幹細胞特異的」調節エレメントを提供し得る遺伝子の1例は、Oct4遺伝子である。
8量体結合転写因子4は、転写因子のPOUファミリーのメンバーである
Oct4転写は、発達中のマウス胚で4細胞期と8細胞期との間で活性化され、そして増大する胚盤胞でそして次いで卵円柱(egg cylinder)の多能性細胞で高発現する。転写は、初期外胚葉が分化して中胚葉を形成するようにダウンレギュレートされ
そして8.5d.p.c.(交尾(coitum)後の日数)までに原基生殖細胞の移動を制限する。高レベルのOct4遺伝子発現はまた、多能性胚癌腫および胚幹細胞株で観察され得、そしてこれらの細胞が分化するように誘導される場合、ダウンレギュレートされる
本発明によれば、Oct4プロモーターの制御下の選択マーカー遺伝子は、胚幹細胞系の単離に適用され得る。さらに、いくつかの成人組織での低レベルのOct4発現を記載する報告(Takedaら、1992)は、胚幹細胞を超えたこれらの発現構築物の有用性を、組織恒常性に不可欠なその他の幹細胞を含みそして造血系を包含するその他の系で修復するように拡張し得る。Oct4が体幹細胞で発現しない場合では、造血幹細胞特異抗原CD34と関連したような他の転写調節エレメントは、同様の方法で有用であり得る。
本発明によれば、2つの特異的なアプローチが、トランスジェニック発現で所望の空間的および時間的な制限を生じることについて提供される。第一のアプローチは、部分的に特徴付けられたOct4遺伝子プロモーターフラグメント(Okazawaら、1991)を使用して選択マーカーの幹細胞特異転写を行うトランスジェニック動物の発生を介する。適切な選択マーカーは、抗生物質G418に耐性を与えるネオマイシンホスホトランスフェラーゼ遺伝子である。あるいは、代謝物中の遺伝子欠失を妨げ得る遺伝子産物の生産に関する選択マーカー、例えば、ヒポキサンチン−グアニンホスホリボシルトランスフェラーゼ(HPRT)遺伝子欠失細胞中のHPRT遺伝子(Hooperら、1987)を利用することである。このアプローチは、幹細胞が密接関連した分化細胞からの連続支持を必要とする状況で有益である。この例では、直接的な細胞の接触は、支持細胞で選択マーカー遺伝子が発現しないにもかかわらず、幹細胞による隣接している支持細胞の代謝救済を可能にする。
第二のアプローチは、内因性Oct4遺伝子座を利用し、そしてそれゆえ、内因性Oct4遺伝子発現プロフィールとできるだけ密接に耐性マーカー遺伝子発現を連結するために、関連したOct4遺伝子調節エレメントを利用する。これは、胚幹細胞でOct4遺伝子の変異誘発を標的する高効率な遺伝子トラップにより達成され得る。このアプローチは、しばしば、ランダムに組み込まれた構築物の予測できない発現パターンを生じるランダムな組み込み部位効果を避けることにより、選択マーカー遺伝子発現のより厳しい調節制御を提供する。
本発明は、以下の実施例、特に添付の図面(図1は、プラスミドOct-4-Neo-βSの構造を示し、図2は、プラスミドOct-4-Neo-βfosの構造を示し、そして図3は、プラスミドOct-4-tgtvecの構造を示す)に関してさらに詳細に記載される。
実施例1
1.OCT4プロモーター配列の単離:
本発明者らは、330bpの5'Oct4 cDNAフラグメントを用いて、129株マウスゲノムラムダライブラリーをスクリーニングした。いくつかのクローンを単離し、制限分析およびサザンブロットハイブリダイゼーションによってスクリーニングした。Oct4プロモーターエレメントを含む1.4kbのBam HI-HindIIIフラグメント(Okazawaら、1991)をクローン1から単離し、pBlue script II KS(-)(Stratagene)に連結し、pOct4(5'ゲノム)を作成した。
2.プラスミドの構築:
Oct4-Neoプロモーター構築物を作成するために、翻訳開始コドンにNcoI制限部位を提供するように設計された遺伝子操作されたネオマイシン耐性遺伝子(neo)を、脳心筋炎(encephalomyocarditus)ウイルス内部リボソームエントリー部位配列(EMCV-IRES、Ghattasら、1991)および5'-Neoコーディング配列を含む1.1kbのXbaI-SphIフラグメントとしてpLZINから単離し(Ghattasら、1991)、そしてpSP72(Promega Biotech)にクローニングした。KpnI-NcoI EMCV-IRES配列を、KpnIおよび次に部分的なNcoI制限消化によってpOct4(5'ゲノム)から単離した1.3kbのOct4プロモーターフラグメントで置換した。Neo3'-コーディング配列、ウサギβ-グロビン遺伝子配列(イントロン)、およびSV40ポリアデニル化配列を、6P-IRESNeo-βS由来の1.7kbのSphIフラグメントとして単離し、そしてSphI部位に連結し、Oct4-Neo-βS(図1)を作成した。Oct4-Neo-βfos構築物(図2)を作成するために、Oct4プロモーター、neo遺伝子、およびウサギβ-グロビンイントロンを含むOct4-Neo-βS BamHIフラグメントをヒトc-fosゲノム配列の5'側に挿入した。
この1.7kbのゲノム配列(BalI-SphI)は、先にmRNA不安定化と関連のあったヒトc-fos mRNA3'コーディング配列および非コーディング配列(WilsonおよびTriesman, 1988)、ならびにc-fosポリアデニル化配列をコードする。
Oct4-neo構築物(Oct4-tgtvec)は、Oct4遺伝子に標的組み込みのために設計されている(図3)。Oct4標的構築物は、1.7kbの5'Oct4遺伝子配列および4.2kbの3'Oct4遺伝子配列を含む。以下の相同組換えでは、この構築物は、lacZ-ネオマイシン融合遺伝子(βgeo、2機能性タンパク質、FreidrichおよびSoriano、1991)をOct4遺伝子の最初のイントロンに組み込む。最初のエキソン-イントロンの境界のスプライスドナー配列から組み込まれたIRES-βgeo配列までのスプライシングは、IRES-βgeo配列のすぐ5'側のネズミのとがった波形になった(engrailed)-2スプライスアクセプター配列(Skarnesら、1992)の含有によって促進される。Oct4-βgeo融合転写物のβgeoシストロンの翻訳は、βgeoコーディング配列のすぐ5'側のEMCV-IRESの含有によって促進される。
3.ES細胞のトランスフェクションおよびコロニー選択:
マウス129ES細胞(CGR-8株)を、Smith(1991)によって記載されたように、分化阻害活性剤(DIA)または白血病阻害因子(LIF)の存在下で調製しそして維持した。トランスフェクションのためのプラスミドDNAをSalI消化によって直鎖状にし、エタノール沈澱し、そしてPBS中で10〜14mg/mlで再懸濁した。新鮮な培養液中で10時間培養した後、ほぼコンフルエントなES細胞をトリプシン処理によって分散し、続いて培養液およびPBSで洗浄し、そして直後のトランスフェクションのためにPBS中で1.4×108/mlに再懸濁した。日常的に、0.7mlの細胞懸濁液を、溶液を含む0.1mlのDNAと混合し、そしてBiorad Gene Pulserおよび0.4cmキュベットを用いて0.8kVおよび3.0μFDでエレクトロポレーションを行った。トランスフェクションは、ゼラチン化した組織培養皿上で増殖培地中5〜8×104/cm2で播いて16時間放置し、その後200μg/ml(活性)G418(Sigma)を含む選択培養液を加えた。単一なコロニーをトランスフェクションの8〜10日後に選択し、そして200μg/mlのG418を含む増殖培養液中でさらなる増加のために24ウエル組織培養プレートに2連で移した。
Oct4-Neo-βSおよびOct4-Neo-βfos構築物を有するクローン化細胞株(Oct4-Neo細胞株と称す)を2日間培養し、PBSで2回洗浄し、そして培養液をDIAを含むまたは含まないG418培養液に置き換えた。DIAの存在下で正常に成長するが、DIAの非存在下で生存しない細胞株を、拡張およびさらなる分析のために選択した。
Oct4-tgtvec標的構築物を有するクローン化細胞株(Oct4-標的細胞株と称す)を、2連の24ウエルプレートで増加させた。一旦コンフルエントになると、一連の細胞を保存のために冷凍するが、残存物をサザン分析で分析した。
4.Oct4-NeoおよびOct4-標的細胞株のさらなる特徴付け:
選択したOct4-Neo細胞株を、G418の種々の濃度でDIA追加した培養液または追加しない培養液中でのES細胞の成長および分化についてアッセイした。細胞を、種々の培養調製物中1×104/cm2で12ウエル組織培養プレートに播き、そして6日間培養した。新鮮な培養液を、先に記載のように(Smith, 1991)、細胞が固定しそして染色される6日目まで2日毎に加えた。ゲノムサザン分析によりポジティブなOct4-標的細胞株を、lacZ染色により、ならびに200μg/mlのG418中のDIA追加または非追加培養液における成長および分化を分析した。
5.Oct4-Neo細胞株由来の胚様体の生産:
胚様体を、懸滴培養法(hanging dropn method)(HoleおよびSmith,印刷中)により発生させ、そしてG418の存在下または非存在下で2、4、6、または8日間懸濁培養で維持した。コントロールの胚様体を、G418の非存在下で親細胞株CGR-8から発生させた。次いで、胚様体を回収しそしてゼラチン化組織培養皿に移し、寄与している細胞型の分析のための集合体の付着および増加を可能にする。全ての胚様体をDIAおよびG418の非存在下で4日間維持した後、視察した。
6.Oct4-NeoおよびOct-4標的細胞株由来のキメラの生産:
選択したOct4-Neo細胞株を、G418の非存在下で7日間培養した後、先に記載されたように(Nicholsら、1990)胚注入した。簡単に言えば、注入する胚盤胞を、C57BL/6ドナー由来の4d.p.c.を回収し、10〜20細胞に注入しそして培養で再増加することを可能にした後、偽妊娠レシピエントの子宮に移した。キメラを、C57BL/6の地色の薄茶の皮の色の斑点の存在により同定した。雄のキメラを、Oct4-Neoトランスジーンの伝達について試験飼育した。次いで、トランスジェニックマウスを異なった遺伝子背景上で掛け合わせた。
7.結果
Oct4-Neo-βS構築物は、約50コロニー/106のトランスフェクトした細胞を生産したが、Oct4-Neo-βfos構築物は約10倍少ないコロニーを生産した。3つのクローンを、DIAの存在下または非存在下でG418(200μg/ml)を含有する培養液中におけるそれらの特異な生存に基づき選択した。3つの細胞株は全て、DIA追加G418含有培養液で正常な成長速度を明らかに示し、そしてG418培養液中でDIAの非存在下で培養した場合に死んだ。DIA追加G418培養液で維持される培養物は、本質的に純粋なES細胞として成長したが、G418の非存在下でDIA追加培養液で維持される培養物は、ES細胞と親CGR-8株の細胞と非常によく似た分化した子孫との混合した培養物として成長した。従って、G418選択は、分化した細胞型を排除し、そして純粋な幹細胞集団の増殖を可能にする。選択した3つの細胞株を、Oct4-Neo-βS18、Oct4-Neo-βS21、およびOct4-Neo-βfos11を命名した。これらの細胞株は、宿主胚盤胞に導入され、そして生じたキメラは試験飼育され得る。同様な結果は、Oct4-tgtvec構築物で標的したESクローンで得られた。さらに、β-ガラクトシダーゼ活性についてのこれらの培養物の組織化学的染色は、βgeoの発現が未分化の幹細胞に制限されることを確認した(Mountfordら、1994)。
胚様体をOct4-Neo細胞株Oct4-Neo-βfos11から発生させて、混合細胞集合体でのG418選択の効果を試験し、そして混合細胞集団からES細胞を単離する選択系の有用性を試験した。実験用細胞株(Oct4-Neo-βfos11)および親細胞株(CGR-8)の両方で発生し、そしてG418の非存在下で培養される胚様体は、ES様細胞をほとんど有さないほぼ完全な分化細胞で構成された。対照的に、増加したコロニーの視覚分析は、G418の存在下で培養したOct4-Neo-βfos11胚様体が高い割合のES細胞を含有することを示した。従って、分化系からの幹細胞の単離の可能性を確認した。
8.要旨
生殖系伝達可能なES細胞は、マウスの2つの同系繁殖系、129およびC57BL/6のみから既に樹立されている。Oct4-ネオマイシン選択図とES細胞単離および増殖手順の確立(EvansおよびKaufman、1981;Martin、1981;Nicholsら、1990;Yoshidaら、1994)を組み合わせることは、Oct4が特異に発現される、以前の非生産性マウス株およびその他の哺乳動物種由来のES細胞株誘導を可能にすべきである。
混合細胞集団における非幹細胞表現型に対する選択は、いくつかの理由で有利であり得る。第一には、混合集団における分化細胞に対する選択は、大量な幹細胞富化物についての方法を提供する。第二には、分化細胞の選択的な除去は、培養物中でのそれらの過剰成長を防御する。第三には、分化細胞の排除は、分化細胞に関連する活性を誘導する分化の損失のために、幹細胞自身の再生を増強し得る。
実施例2
ES細胞胚様体由来の多能性幹細胞の救済および回収
方法
1.細胞培養
ES細胞を、Smith(1991)により記載されたように分化阻害活性剤(DIA)を追加した培養液中で日常的に維持した。胚様体をDIAの非存在下でES細胞の集合体により形成した。この集合体は、10μlまたは30μl滴の培養液中で100細胞/滴の濃度で分離したES細胞を播くことにより生産した。滴の配列をマルチピペッターを用いて10cm組織培養皿のフタの上に置いた。次いで、これを皿の底と逆転し、この皿は湿度を維持するために10mlの水を含有し、そして懸滴を7%CO2雰囲気中で37℃にて培養した。
2.組織学およびβ-ガラクトシダーゼ染色
胚様体をブーアン(Bouin)溶液中で固定しそして寒天中に包埋した。次いで、パラフィン切片を標準的手順により調製しそしてヘマトキシリン(haematoxylin)およびエオシンで染色した。胚様体派生物のアルカリホスファターゼ染色をSigma Kit 86-Rを用いて実施した。β-ガラクトシダーゼの組織化学的染色を記載(Beddingtonら、1986)のようにXgalを用いて行った。
結果
3.細胞株および選択系
Fos11は、Oct4neofos構築物でトランスフェクトしたES細胞株CGR8の誘導体である。Fos11細胞は、Oct4近接プロモーターエレメントの制御下で低レベルのG418耐性を発現するが、分化した子孫はトランスジーンの発現を示さず、そしてそれゆえG418に感受性である。OKO160およびOKO8は、それぞれES細胞株CGR8およびE14TG2aの誘導体であり、そこではIRES-βgeopAカセットが前記のような相同組換えによりOct4遺伝子の1つの対立遺伝子に導入されている。OKO細胞株は未分化状態で高レベルのβgeoを発現し、そしてそれゆえXgalで強力に染色しそしてG418耐性である。分化した子孫はβgeoの発現を喪失しそしてXgal染色について陰性およびG418に感受性となる。単層培養では、Fos11およびOKO細胞を、DIAの存在下での培養により純粋なES細胞集団として維持し、G418で選択する。しかし、DIA(Smith, 1991)の低濃度および非存在めような分化に有利な条件下では、G418選択の結果、3〜5日間にわたりこれらの培養物は完全に除去される。Rb40細胞は、ヒトβ-アクチンプロモーターにより指示されるneoRの発現のため、G418に対して構成的に耐性であるCGR8の誘導体である。
4.分化細胞に対する選択の存在および非存在下における胚様体の形成
細胞の凝集塊を分離し続いて大量の懸濁培養物中で集合する従来の手順(Doetschemanら、1986)による胚様体の生産は、サイズおよび分化状態の両方が不均一な集合体の混合集団を生じる。さらに単一および一貫した発達を得るために、最近の研究では、胚様体を、培養培地の個々の滴中で一定の数の細胞の集合体により形成した(方法の項を参照のこと)。懸滴培養の48時間後、集合体をG418の存在または非存在下の懸濁培養にひとまとめにして移した。
分化した子孫に対するG418選択下において、集合体はFos11細胞およびOKOクローンの両方からなお形成されていた。いくつかの死んだ細胞が集合体の周囲に現れたが、本体自身は、培養期間の間にサイズを増大した。試料を大量の培養物から定期的に採取し、そして組織学的試験を行った。数日後、選択の非存在下で形成した胚様体は、ほとんど嚢胞性でありそして種々の形態学的に分化した細胞型を包含していた。未分化細胞はまれに見られた。対照的に、選択下で維持される集合体は、細胞の特殊化の徴候を示さず、そして本体は、未分化細胞の固体球からなるようであった。これらの未分化集合体中の大部分の細胞は、正常および生存可能なようであり、そして時折アポトーシスを示唆する核濃縮核が見られたが、壊死の証拠はなかった。しかし、G418で形成した胚様体は、選択の非存在下で形成したそれらの対照物より顕著に小さかった。これは嚢胞発達の欠損および分化細胞の除去の組合せに特徴があり得る。
5.分化細胞に対する選択下で形成した胚様体中の多能性幹細胞の持続性。
コントロール培養物中において未分化集合体が少しも存在しないことは、おそらく、G418の効果が非分化集合体の亜集団の選択のためではないことを意味する。しかし、この可能性を最終的に除外するために、およびまたG418選択の効果の量的な決定を容易にするために、改変したプロトコルを用いて個々の集合体の行動(behavior)の評定を可能にした。培養を、G418の存在下または非存在下で30μlの懸滴中で開始し、そして7〜8日間滴培養で維持した。次いで、胚様体をゼラチンコートした96ウエル組織培養プレートに個々に移し、そしてこの培養液をG418を欠いた培養液で6倍希釈した。幹細胞維持因子DIAをこの段階で添加し、存在するあらゆる未分化ES細胞を増加させた。培養物を、付着させ、次いで48時間過剰成長させて固定し、アルカリホスファターゼまたはβ-ガラクトシダーゼで適切に染色した。
表1に要約したデータは、選択の全くの非存在下で、未分化幹細胞が、7日目の懸濁培養物内の胚様体からほぼ完全に除去されることを示す。過剰成長物は種々の形態学的に分化した細胞型を含むが、ESの細胞形態学を有する細胞領域は観察されなかった。OKO細胞におけるβ-ガラクトシダーゼの発現は、幹細胞特異的転写因子Oct4に結びつき(Mountfordら、1994)、そしてそれゆえ、未分化細胞のマーカーとして働く。単離したXgal-染色細胞は、時折OKO過剰成長物中に見られるが、染色細胞のクラスターはこれらの状態下で検出されなかった(しかし、論考の項を参照のこと)。
G418での胚様体形成の効率は、非選択培養中で本質的に100%に等しかった。しかし、未処理の胚様体との著しい対比では、継続的なG418選択下で樹立した胚様体は、主としてES細胞からなる過剰成長を生じた。これらの未分化な細胞の性質を、ES細胞コロニーの形態学的特徴によりおよびアルカリホスファターゼでの染色により同定し、そしてOKO過剰成長物のXgal染色により確認した。
選択下で形成した胚様体由来のいくつかの過剰成長物を摘出し、そして2cmウエルに移した。摘出した全てのコロニーを未分化細胞の大量培養物で容易に増加した。これらの培養物は、非追加培養液中に置いた場合、DIAに依存して残り、そして親ES細胞と同様の様式で分化した。さらに、これらの誘導体は、集合体で複数の細胞型に効率よく分化し、それらの多能性を確認した。
これらの発見は、分化した子孫の選択的除去がES細胞集合体での多能性幹細胞の持続性を生じることを証明する。
6.混合集合体での幹細胞の死滅
分化した子孫が胚様体での幹細胞死滅に直接的な原因であり得るという意味がさらに提示された。OKO細胞の行動を、G418の存在下で分化し得るRb40 ES細胞と混合した集合体の形成に続き評定した。Rb40細胞はネオマイシンホスホトランスフェラーゼを構成的に発現し、そしてG418選択は、単層培養または集合体のいずれかにおいてそれらの分化への認められる効果を有さない。懸滴培養物を、3:1の比のOKO細胞:Rb40細胞を用いて樹立した。混合胚様体のパラフィン切片は、それらがG418の存在または非存在下の両方で大量に分化をすることを明らかにした。両方の条件下での未分化幹細胞の効果的な除去を、過剰成長物のXgal染色により確認した(表1)。
この結果は、分化した子孫の存在が多能性幹細胞の除去を誘導する直接的な証拠を提供する。これは、特定の分化幹細胞の子孫が誘導シグナルの供給源であることを包含し、このシグナルは、残っている幹細胞のさらなる分化を指示するかまたはおそらくアポトーシスへ導入するよう誘導するかのいずれかである。
結論
集合体はES細胞を誘導して、胚様体として公知の分化構造物に発達する。多能性幹細胞は、効果的な分化の誘導およびおそらくまた選択的な細胞死のために、これらの胚様体で迅速に死滅する。しかし、分化した子孫が本発明の方法を用いて集合体から特異的に除去される場合、幹細胞は持続しそして増殖し得る。
上記の詳述した発見は、本発明の幹細胞特異的選択系の使用を通じて、分化または死のいずれかにより正常にそれらの除去をさせる条件から幹細胞を再生することが可能であることを明確に示す。
実施例3
マウス胚から胚幹細胞培養物を樹立する手順
トランスジェニックマウスの系を、G418耐性を与えるネオマイシンホスホトランスフェラーゼ遺伝子がOct4遺伝子の特異性を有して発現することで樹立した。βS21系はOct4neoβSトランスジーンを有するが、OKO系では、neo遺伝子がOct4-tgtvec構築物で標的する遺伝子を介して内因性Oct遺伝子に取り込まれている。これらのマウスを、MF1異系交配アルビノマウスおよび同系交配CBAマウスを用いて2世代について外部交配した。これらのマウスの系のいずれも標準手順を用いてES細胞を生産しない。
幹細胞を単離する4つの好ましい手順を記載する。全ての場合において、胚は、分化阻害活性剤(Smith, 1991)またはインターロイキン-6と可溶性インターロイキン-6レセプター(Yoshidaら、1994)のいずれかを追加した標準ES細胞培養液で培養される。G418を200μg/ml〜1mg/mlの濃度で添加する。
手順1
胚盤胞を妊娠4日目で洗い出す。4〜10のグループの胚盤胞を1cmの組織培養ウエルでG418選択下で培養する。過剰成長を個々に分離し、そして記載した(Nicholsら、1990)ように培養4〜6日後、トリプシンで分離し、そして1つのウエルに置き換える。G418選択を維持する。次の14日間にわたり培養物中に見られるES細胞の形態学的特徴を有するコロニーを摘出し、そして連続選択下で増加させる。
手順2
胚盤胞が、妊娠3日目で母体の卵巣切除により採取する前に着床遅延になることを除いては、手順1のように行った。胚盤胞を卵巣切除4日後に洗い出す。
手順3
交尾後5.5および7.5日の間の着床後の胚を単離し、そして初期外胚葉を顕微解剖および/またはプロテアーゼ消化より分離する。外胚葉は、穏やかに20〜50細胞の凝集塊に分離し、それは、次いで手順1のように培養される。
手順4
手順1、2、または3のように調製した胚は、組織培養ウエルに移しそして続いて手順1のように操作する前に、5〜7日の期間G418選択下の懸滴中で培養される。
Claims (24)
- 動物幹細胞を単離するおよび/または濃縮するおよび/または選択的に増殖する方法であって、該方法は、選択マーカーを有するあるいは選択マーカーが導入された所望の幹細胞を含む細胞の供給源を準備する工程;および、該所望の幹細胞の生存の助けとなる培養条件下で、該細胞の供給源を維持する工程を包含し、
該選択マーカーは、Oct4プロモーターに作動可能に連結され、そして(a)所望の幹細胞および(b)所望の幹細胞以外の細胞において差次的に発現することが可能であり、
該選択マーカーの差次的発現によって、該細胞の供給源中の該所望の幹細胞の選択的な単離および/または生存を生じさせ、
そして、該細胞の供給源が、非ヒト受精卵母細胞;非ヒトトランスジェニック動物から分離された細胞;非ヒト非トランスジェニック動物から分離された細胞;非ヒト胚から分離された細胞;血液から分離された細胞;および体組織から分離された細胞、ならびにこれらの細胞株から選択される、方法。 - 前記所望の幹細胞が、単能性幹細胞、多能性幹細胞、胚幹細胞、性腺幹細胞、体幹/前駆細胞、造血幹細胞、表皮幹細胞、および神経幹細胞から選択される、請求項1に記載の方法。
- 前記細胞供給源が陽性選択マーカーを有する幹細胞を包含し、そして前記マーカーの発現が幹細胞の単離および/または濃縮および/または維持を可能にするために用いられる、請求項1または2のいずれかに記載の方法。
- 前記選択マーカーが、外来遺伝子、細胞性遺伝子、および抗生物質耐性遺伝子から選択される、請求項1〜3のいずれかに記載の方法。
- 前記抗生物質耐性遺伝子が細菌性ネオマイシン耐性遺伝子である、請求項4に記載の方法。
- 前記選択マーカーが成長刺激遺伝子である、請求項1〜5のいずれかに記載の方法。
- 前記成長刺激遺伝子が、オンコジーンまたはその誘導体である、請求項6に記載の方法。
- 前記成長刺激遺伝子が、SV40ラージT抗原またはSV40ラージT抗原の誘導体である、請求項6に記載の方法。
- 前記成長刺激遺伝子が、成長因子をコードする遺伝子、成長因子レセプターをコードする遺伝子、シグナル伝達分子をコードする遺伝子、および転写因子をコードする遺伝子から選択される、請求項6に記載の方法。
- 前記選択マーカーが不死化遺伝子である、請求項1〜4のいずれかに記載の方法。
- 前記不死化遺伝子が、ポリオーマラージT遺伝子、細胞死を阻止する遺伝子、およびbcl-2遺伝子から選択される、請求項10に記載の方法。
- 前記選択マーカーが、HPRT、毒性産物をコードする遺伝子、自殺基質と組合せて条件により活性な毒性遺伝子産物、および単純ヘルペスウイルスチミジンキナーゼ(HSV−TK)遺伝子から選択される、請求項1〜4のいずれかに記載の方法。
- 前記細胞が2つの選択マーカーを含む、請求項1〜12のいずれかに記載の方法。
- 前記選択マーカーの発現が、細胞供給源に導入する前に、該選択マーカーを発現構築物に作動可能に挿入することにより達成される、請求項1〜13のいずれかに記載の方法。
- 前記選択マーカーの発現が、安定に組み込まれた構築物、エピソームにおいて維持された構築物、または一過的に維持された構築物の導入から生じる、請求項1〜14のいずれかに記載の方法。
- 前記選択マーカーの発現が、細胞供給源の内因性遺伝子に該選択マーカーを作動可能に挿入することから生じる、請求項1〜15のいずれかに記載の方法。
- 前記選択マーカーが、トランスフェクション、リポフェクション、注入、衝撃ミサイル、ウイルスベクター、エレクトロポレーション、または任意の他の手段により、前記細胞へ導入される、請求項1〜16のいずれかに記載の方法。
- 前記選択マーカーが非ヒトトランスジェニック動物に取り込まれる、請求項1〜17のいずれかに記載の方法。
- 前記選択マーカーがネオマイシンホスホトランスフェラーゼ遺伝子である、請求項1〜18のいずれかに記載の方法。
- 選択マーカー構築物を、幹細胞を含む細胞供給源に導入する工程を包含する、請求項1〜19のいずれかに記載の方法であって、該選択マーカー構築物が、前記差次的発現を提供する内因性遺伝子に作動可能に連結するために適応されている、方法。
- 選択マーカー構築物を、幹細胞を含む細胞供給源に導入する工程を包含する請求項1〜19のいずれかに記載の方法であって、前記選択マーカー構築物が前記差次的発現を提供する1つまたはそれ以上の遺伝子または遺伝子フラグメントに既に連結されている、方法。
- 動物幹細胞を選択的に単離および/または濃縮および/または増殖する方法であって、選択マーカー構築物を、幹細胞を含む細胞の供給源に導入する工程;および所望の幹細胞に適切な培養条件下で該細胞の供給源を培養する工程;を包含し、
該選択マーカー構築物がOct4プロモーターに作動可能に連結するかまたは既に連結されており、
該選択マーカー構築物は、所望の幹細胞と所望の幹細胞以外の細胞において該選択マーカーを差次的に発現することが可能であり、該選択マーカーの差次的発現によって、適切な培養条件下で該所望の幹細胞の選択的な単離および/または濃縮および/または増殖を可能にし、
そして、該細胞の供給源が、非ヒト受精卵母細胞;非ヒトトランスジェニック動物から分離された細胞;非ヒト非トランスジェニック動物から分離された細胞;非ヒト胚から分離された細胞;血液から分離された細胞;および体組織から分離された細胞、ならびにこれらの細胞株から選択される、方法。 - 前記選択マーカーがネオマイシンホスホトランスフェラーゼ遺伝子である、請求項22に記載の方法。
- 動物幹細胞を選択的に単離するおよび/または濃縮するおよび/または増殖する方法であって、所望の幹細胞の選択培養条件下で細胞の供給源を培養する工程を包含し、

そして、該細胞の供給源が、非ヒト受精卵母細胞;非ヒトトランスジェニック動物から分離された細胞;非ヒト非トランスジェニック動物から分離された細胞;非ヒト胚から分離された細胞;血液から分離された細胞;および体組織から分離された細胞、ならびにこれらの細胞株から選択される、方法。
