腫瘍組織が転移する能力は、悪性疾患の生命を脅かす局面の主要な部分を構成することが長く認識されてきた。転移は、原発性腫瘍とは異なる部位での続発性腫瘍の増殖である。従って、原発性腫瘍の外科的除去にもかかわらず、この状態の進行を停止させることは可能ではないかもしれない。転移が生じる機構の理解は、続発性腫瘍の増殖が制御され得るプロトコルの開発に重要である。転移の機構を理解するために、多くの宿主細胞のバックグランドに対する少数の腫瘍細胞の同定を可能にし、その結果、続発性腫瘍塞栓および微小転移が、リアルタイムの経過にわたって観察され得る、モデルを提供することが必要である。

Zolotukhin、S.ら、J Virol.(1996)70:4646−4654に記載されるヒト化hGFP−S65Tクローンを、緑色蛍光タンパク質をコードする配列として使用した。このコドン最適化された遺伝子を、Clontech Laboratories,Inc.(Palo Alto,CA)から購入し、そしてGenetics Institute,Cambridge,MAから購入し、そしてKaufman,R.J.ら,Nucleic Acids Res(1991)19:4485−4490に記載されるジシストロン性発現ベクター(pED−mtx’)へと連結した;hGFP−S65TをHindIIIで消化し、そして平滑末端化した。hGFPコード領域全体を、XbaIで切り出し、次いで、PstIで消化し、平滑末端化し次いでXbaIでさらに消化したpED−mtxl中に単一の方向にサブクローニングした。

図1aおよび1bは、SV40プロモーターの制御下にあるGFPのための発現ベクターの構築を示す。この構築物は、Clontechから入手可能な市販のpEGFPシリーズのベクターを使用する。細菌発現ベクターおよび哺乳動物発現ベクターの両方は、さらなるタンパク質およびGFPの産生を、融合タンパク質としてかまたはジシストロン性系においてのいずれかで産生し得るものが利用可能である。図1aは、メチオニナーゼとのGFPの融合物についての、発現ベクターpGFP/Metの構築を示す。図1bは、SV40T抗原とGFPとの融合タンパク質の産生のためのpGFP/SV40の構築を示す。

(実施例4:新脈管形成の観察)
1×107のクローン38細胞を含む懸濁物(実施例1に記載される)を、マウスの腹腔に注射した。5日後、マウスの尻尾にローダミンを注射し、次いでそのマウスを麻酔し、そして腹腔を充分に開口して腫瘍を可視化させた。この手術からの回復はすぐであった。いくつかの場合において、開腹は、不要である。なぜなら、腹腔内の腫瘍が生の皮膚を通して可視化され得るからである。腫瘍は、腹腔において可視化され、そして新脈管形成が、ローダミン蛍光によって同定されるように明らかになった。類似の結果が、小腸壁における網および腸間膜において増殖する腫瘍中に見られた。

類似の実験において、1×107細胞のクローン26を含む懸濁物(実施例1に記載される)を、マウスの腹腔中に注射した。1日後、腫瘍が腸間膜中および結腸壁に出現した。これらを、マウスを麻酔することおよび腹を最小限開口することによって観察した。類似の処置をしたマウスの3日目の観察によって、小腸の壁および網ならびに結腸壁および腸間膜における腫瘍を示した。5日目に、類似の処置をしたマウスの尻尾に、100μlの2×10-3Mのローダミンを注射し、そして数本の血管が、腸間膜中に成長する腫瘍においてみることができた。60日後、多数の血管が、結腸壁において成長する腫瘍において見られた。

(実施例5:転移モデルの構築)
実施例2に記載される標識されたヒト癌細胞株を用いて、種々の型の癌についてのマウスモデルを樹立した。この細胞株を、107GFP発現ヒト腫瘍細胞を単回用量で6週齢のnu/nu雌性マウスに移植し、これを、トリプシン処置によって採集し、そして冷血清含有培地で3回洗浄し、次いで氷上に維持した。採集の40分以内に0.4mlの総容量で動物の側腹部中の皮下空間に注射する。このヌードマウスを腫瘍細胞注射から3週間後に屠殺して、腫瘍断片を採集する。次いで、これらの腫瘍断片を、レシピエントとしてのヌードマウスにおいて、対応する組織(外科的歯科矯正術移植(SOI))中への外科的移植のために使用する。

Claims (9)
- 転移能を有する移植した原発性腫瘍の転移を阻害するための候補となるプロトコルまたは薬物を評価するための方法であって、ここで該原発性腫瘍は、肺、肝臓、膵臓、胃、乳房、卵巣、前立腺、骨髄、脳および悪性腫瘍に感受性の他の組織を含む部位に移植されており、該方法は、以下:
該原発性腫瘍がもとの移植部位から遠位の部位に転移した場合に、励起により緑色光を維持された高レベルで安定に放出する緑色蛍光タンパク質(GFP)を発現するように改変されている該原発性腫瘍を含む、実験のためのヒト以外の動物に該プロトコルまたは薬物を投与する工程であって、該動物が、該腫瘍に対して免疫無防備であるかまたは同系である、工程;
該動物を、D 425 / 60 帯域のフィルタを用いて励起することができ、かつGFPによって吸収される光で照射して、 GC475 長径フィルタを通過できる該GFPからの緑色の蛍光を生じさせる工程;
該処置された動物における種々の位置にて、該転移による該蛍光の存在、非存在または強度を観察することによって該転移の進行をモニターする工程;
緑色蛍光タンパク質を発現する細胞を含む同様な腫瘍を含むヒト以外のコントロール動物において該処置された動物における種々の位置にて、転移による蛍光の存在、非存在または強度を観察することによって該転移の進行を同様にモニターする工程;
該コントロール動物における該転移の進行と、該処置された動物における該転移の進行とを比較する工程;
該観察はインタクトで生きているヒト以外の動物全体においてリアルタイムで行われ、該動物の画像はコンピュターによって捕捉されるか、またはビデオ出力を通じて連続的に捕捉される工程;
を包含しそれによって、該コントロール動物と比較したときの、該処置された動物における該転移の進行の減少が、転移の阻害において有効であるとして該プロトコルまたは薬物を同定する、方法。 - 該原発性腫瘍が、外科同所移植により移殖され、そして該原発性腫瘍の細胞が該GFPの発現系を含有する請求項1記載の方法。
- 該動物がウサギ、ラットまたはマウスである、請求項1または2に記載の方法。
- 転移能を有する移植した原発性腫瘍の転移の阻害のための候補となるプロトコルまたは薬物を評価するための方法であって、ここで該原発性腫瘍は、肺、肝臓、膵臓、胃、乳房、卵巣、前立腺、骨髄、脳および悪性腫瘍に感受性の他の組織を含む部位に移植されており、該方法が、以下:

該動物を、D 425 / 60 帯域のフィルタを用いて励起することができ、かつGFPによって吸収される光で照射して、 GC475 長径フィルタを通過できる該GFPから緑色の蛍光を生じさせる工程;
該処置された動物における種々の位置にて、転移による該蛍光の存在、非存在または強度を観察することによって該転移の進行をモニターする工程;
を包含し、それによって、該処置された動物における該転移の進行の減少が、転移の阻害において有効であるとして該プロトコルまたは薬物を同定し、かつ該観察はインタクトで生きているヒト以外の動物全体においてリアルタイムで行われ、かつ該動物の画像はコンピュターによって捕捉されるか、またはビデオ出力を通じて連続的に捕捉される、方法。 - 該原発性腫瘍が、外科同所移植により移殖され、そして該原発性腫瘍の細胞が該GFPの発現系を含有する請求項4記載の方法。

原発性腫瘍とは遠位の部位への転移の進行をモニターする工程;
該動物を、D 425 / 60 帯域のフィルタを用いて励起することができ、かつGFPによって吸収される光で照射して、 GC475 長径フィルタを通過できる該GFPから緑色の蛍光を生じさせる工程;
該動物における該転移の進行は、該インタクトで生きている、動物全体における種々の位置にて、時間の関数として、転移による蛍光の存在、非存在または強度を観察することによってモニターされ、該観察はインタクトなヒト以外の動物全体においてリアルタイムで行われ、該動物の画像はコンピュターによって捕捉されるか、またはビデオ出力を通じて連続的に捕捉される工程;
を包含する、方法。 - 該原発性腫瘍が、外科同所移植により移殖され、そして該原発性腫瘍の細胞が該GFPの発現系を含有する請求項7記載の方法。
