Description
本発明は一般的に心臓学の分野に関するものであり、より詳細には安定な心筋移植体ならびにその移植を達成するために有用な方法および細胞組成物に関するものである。
さらに背景として、罹患した機能不全の組織と交換するために器官移植が広く採用されている。比較的最近になって、宿主の組織機能の欠陥を補うために細胞移植が有効な治療例として出現した。この方法の一例は、パーキンソン症候群を伴う個体においてよく知られている胎児組織の使用であり((1)に概説、後記の参考文献リストを参照されたい)、この場合は移植された細胞からのドーパミン分泌が患者の欠陥を軽減する。他の研究においては、デュジェンヌ症(Duchenne's)の患者において罹患していない同胞から移植された筋芽細胞が内在性筋管と融合した;重要なことは、移植した筋管が野生型ジストロフィンを発現したことである(2)。
これらの初期の研究は、他の領域においてそれらが適性を示すにもかかわらず、心臓に有効に適用しうる細胞移植技術については何ら述べていない。心臓において損傷を受けた心筋を交換することができれば、明らかに臨床適性があるであろう。さらに血管形成誘導因子およびイオノトロピックペプチドの長期発現を目標として心臓内移植体を使用すれば、それぞれ心筋虚血またはうっ血性心不全を伴う個体にとって治療上有用であろう。
この背景からみて、心臓における細胞移植技術の開発が要望されている。望ましくはそのような技術は、心臓における安定な移植体を提供するだけでなく、有用な組換え蛋白質または他の分子を心臓に直接に運搬することも可能にする。本発明はこれらの要望に対処するものである。
発明の概要
本発明者らは、心筋において長期間生存しうる細胞移植体を確立した。心筋細胞および骨格筋芽細胞と同系動物の心筋内へ直接に移植した。移植後少なくとも半年は、生存可能な移植体が検出された(アッセイを行った最後の時点)。移植体の存在に伴って明瞭な不整脈が生じることはなく、大部分の移植体は宿主の心筋に直接に近接しており、被包されていなかった。こうして、心筋を細胞移植体のための安定な足場として使用しうることが見出された。これらの移植体は心臓へ組換え分子を局所運搬するために、および/または罹患組織に代わって心筋の機能を補足するために使用しうる。
従って本発明の好ましい1態様は、動物の心筋組織に取り込まれた骨格筋芽細胞および心筋細胞の安定な移植体を含む、動物における心筋移植体を提供する。
本発明の他の好ましい態様によれば、動物において安定な心筋移植体を形成する方法が提供される。本発明方法には、動物の心筋組織に骨格筋芽細胞または心筋細胞を導入し、これによって安定な心筋移植体を形成する工程が含まれる。細胞は常法により、たとえば注射により導入することができる。
本発明の他の好ましい態様によれば、動物の心筋組織に組換え分子を運搬する方法が提供される。この方法には、動物の心筋組織に取り込まれた骨格筋芽細胞または心筋細胞の安定な移植体を確立する工程が含まれ、その際筋芽細胞または心筋細胞は心筋組織に組換え分子を運搬する。この態様においては、筋芽細胞または心筋細胞は組換え分子をコードするトランスジーンを保有するであろう。
本発明の他の好ましい態様によれば、不死化していない心筋細胞の実質的に均一な集団を含む細胞組成物が提供される。この、および他の細胞集団は、(i)胚幹細胞をトランスフェクションして、幹細胞の分化により生じる1細胞系列を他の細胞系列から選択しうるマーカー遺伝子を導入し、(iii)幹細胞を分化させ、そして(iv)この1細胞系列をマーカー遺伝子に基づいて選択することを含む、好ましい本発明方法により得られる。これらの方法に使用される細胞およびこれらの方法により得られる細胞も本発明の一部を構成する。
本発明のさらに他の好ましい態様によれば、動物の心筋組織に取り込まれた骨格筋芽細胞および心筋細胞の安定な移植体を有する、非ヒト動物が提供される。
従って本発明は、心筋移植体、心筋移植体を形成するのに有用な方法および細胞組成物、ならびに心筋移植体を有する動物を提供する。移植体は、治療剤、たとえば組換え蛋白質および他の分子を運搬するためのベヒクルとして、かつ罹患組織に代わって心筋の機能を補足するための手段として有用であろう。本発明の細胞組成物は移植体を調製するために直接に用いることができ、かつ心筋細胞に対する薬物の作用をスクリーニングする際に、または組換え蛋白質を発現および採取するためにも有用であろう。移植した動物は、たとえば心臓に対する組換え分子の作用をスクリーニングするために利用しうる。

好ましい態様の説明
本発明の原理をより良く理解するために、その特定の態様について述べ、それを説明するために具体的な用語を使用するであろう。それにもかかわらず、これは本発明の範囲を限定することを意図したものでないことは理解されるであろう。本明細書に示す本発明の原理をさらに変更および応用することは、本発明に関連する技術分野の当業者が容易になしうるものと考えられる。
前記のように、本発明は骨格筋芽細胞および/または心筋細胞の安定な心筋移植体を提供する。これに関して本明細書において使用する用語“安定な心筋移植体”は、その細胞が少なくとも約2週間は生存可能である心筋移植体を意味するものとする。意外にもこのような安定な移植体は本発明によって容易に得られ、好ましい移植体は6カ月以上生存可能な細胞を有する。従って本発明の心筋移植体は組換え蛋白質もしくは他の分子を心臓に長期間運搬し、および/または心筋組織を長期補足することができる。
本発明に用いられる骨格筋芽細胞および心筋細胞は、任意の適切な供給源から採取または単離することができる。たとえばC2C12骨格筋芽細胞を含めた骨格筋芽細胞を公的寄託機関、たとえばアメリカン・タイプ・カルチャー・コレクションズ(ATCC)(マリーランド州ロックビル)から入手しうる。骨格筋芽細胞は骨格筋から、たとえば当技術分野および文献で周知の技術により単離することもできる。本発明に用いられる心筋細胞は文献(3)に記載の技術により、またはより詳細に後記実施例に記載した方法により得られる。簡潔に言えばこのような方法の1つは心臓組織を消化して心筋細胞を得ることによる。
他の方法は、適宜なマーカーを用いて、特定の細胞系列、たとえば心筋細胞を胚幹細胞((22)に記載される胚盤胞の内部細胞塊に由来する全能性細胞系)の分化により生じる他の細胞系列から選択することによる。好ましい方法は陽性選択方式によるものである。たとえばマーカー遺伝子、たとえば抗生物質(たとえばネオマイシンまたはハイグロマイシン)耐性を付与する遺伝子を、遺伝子の発現が目的細胞系列においてのみ起こるような適宜な制御下で幹細胞に導入する。たとえば、マーカー遺伝子を目的細胞系列においてのみ活性であるプロモーターの制御下に置くことができる。次いで幹細胞が分化した時点で、マーカーに基づいて、たてえば分化した細胞の混合物を目的系列がそれに対する耐性を付与されている適宜な抗生物質と接触させることにより、目的系列を選択する。目的系列以外の細胞系はこうして死滅し、実質的に純粋かつ均一な目的系列の集団を回収することができる。より好ましい方法においては、2種類のマーカー、すなわちトランスフェクションした幹細胞を非トランスフェクション細胞から選択しうるマーカー、および目的細胞系列を他の系列から選択しうるマーカーを、母体である幹細胞に導入する。こうして各選択性マーカーが抗生物質耐性を付与する二重陽性選択方式を採用しうる。この選択法を用いると、後記実施例において証明されるように、約90%、さらに約95−100%もの目的細胞系列を含む集団を得ることができる。
本発明の移植体を得るためには、骨格筋芽細胞または心筋細胞を生存動物、たとえば哺乳動物の心筋組織に導入する。細胞は任意の適切な方法で導入することができるが、導入様式は可能な限り非侵襲性であることが好ましい。たとえば細胞を注射、カテーテル導入、またはこれに類する手段で運搬することがより望ましいであろう。
得られた移植体保有動物は正常な洞調律を示し、これは移植体自体および移植体−宿主心筋の境界帯域が不整脈を誘発しないことを示している。これはヒトにおいて梗塞後にしばしば起こるリモデリングとは著しく対照的である;梗塞の境界帯域は輪回ループ(circus loop)を形成し、その結果臨床的に重大な不整脈が起こる可能性がある(4,5)。
本発明の移植体は増殖性または非増殖性のいずれであってもよい。たとえば後記の具体例において確立されたAT−1移植体は増殖性である。これに対し実施例において形成された骨格筋芽細胞由来の移植体は非増殖性であり、トリチウム化チミジン取り込みがないことは安定な心臓内移植体の形成が持続的な細胞増殖に依存するものでないことを示す。
好ましい移植体は、直接的な細胞内結合の存在、および宿主と移植細胞との間のギャップ結合の形成を特色とするであろう。さらにそれらの移植体は宿主において免疫反応を引き起こさず、移植細胞の最終分化および非腫瘍化性を示すであろう。
本発明の移植体は、治療用蛋白質などを移植細胞からの分泌により運搬し、罹患または損傷した組織に代わって心筋機能を補足するのに特に有用である。治療用蛋白質の運搬の例として、移植体は血管形成誘導因子(たとえば塩基性および酸性繊維芽細胞増殖因子;トランスフォーミング増殖因子−β、血管内皮増殖因子ならびに肝細胞増殖因子)を発現して新生血管形成を誘発しうる。同様に梗塞領域付近で神経栄養物質(neurotrophic agent)を発現する移植体を用いて、境界帯域に付随する不整脈惹起性を改善することができる。心臓を標的とする運搬についてのこれらおよび他の多数の候補物質は、当業者には自明であろう。
本発明ならびにその原理および利点の理解をいっそう促進するために、以下の具体例を提示する。これらの例は説明のためのものであって限定的な性質のものでないことは理解されるであろう。
実施例1
安定なAT−1心筋細胞移植体の形成
A.方法
AT−1細胞の培養および心筋移植プロトコル AT−1心筋細胞を連続コラゲナーゼ消化により皮下腫瘍から分離し、10%ウシ胎児血清を含有するPC−1培地(ベントレックス、ミネソタ州クーン・ラピッズ)中で培養した:先に(6)に記載されたものに従う。注射位置の確認を容易にするために、細胞を10μMの8−クロロメチル−4,4−ジフルオロ−1,3,5,7−テトラメチル−4−ボラ−3a,4a−ジアザインデセン(BODIPY,モレキュラー・プローブズ、オレゴン州ユージーン)で37℃において30分間標識した。注射直前にトリプシンおよびコラゲナーゼを用いて細胞を採取し、無血清PC−1培地で3回洗浄し、開胸手術した同系B6D2/F1マウス(ジャクソン・ラボラトリーズ、メイン州バー・ハーバー)の心室心筋に直接注射した:(7)の記載に従う。細胞(4−10×104)は2−3μlの容量で30ゲージの注射針を備えたプラスチック注射器により注射された。
組織学的所見 頸部脱臼後に心臓を摘出し、30%スクロース中で凍結保護し、包埋し、クリオマイクロトームで10μmの切片に切断した:(8)の記載に従う。ヘマトキシリンおよびエオシン(HおよびE)染色のために切片をアセトン:メタノール(1:1)中で後固定し、製造業者の説明書(シグマ・ダイアグノスティックス、ミズーリ州セントルイス)に従って染色した。免疫組織学的分析のために、固定していない切片をポリクローナルウサギ抗−T−Ag抗体(161−T((3)参照)または162−T)と反応させ、次いで西洋ワサビペルオキシダーゼ共役−ヤギ抗−ウサギ抗血清(ベーリンガー・マンハイム、インディアナ州インディアナポリス)と反応させ、ニッケル増強を伴うジアミノベンジジン反応により視覚化した:(9)の記載に従う。共通白血球抗原(CD45;抗体M1/9.3HL、ベーリンガー・マンハイム)に対する、およびマクロファージMac−1抗原(CD11b;抗体M1/70HL、ベーリンガー・マンハイム)に対するモノクローナル抗体を用いて、心臓内移植体拒絶を監視した。Mac−1抗体はリンパ球と75−90%の交差反応性を有する。一次抗体で処理したのち、切片を西洋ワサビペルオキシダーゼ共役−ウサギ抗−ラット抗血清(ベーリンガー・マンハイム)と共にインキュベートし、ニッケル増強を伴うジアミノベンジジン反応により視覚化した。[3H]−チミジン取り込みのために、マウスに同位体(280Ci/mMにおいて400μCi、アメルシャム、イリノイ州アーリントン・ハイツ)を1回ボーラス注射し、18時間後に頸部脱臼により屠殺した。心臓を摘出し、30%スクロース中で凍結保護し、包埋し、クリオマイクロトームで切断した。切片をメタノール:アセトン(1:1)中で後固定し、HおよびEで染色し、蒸留水で1:1に希釈した写真乳剤(イルフォード(Ilford)L.4、ポリサイエンシズ、ペンシルベニア州ウォーリントン)の薄層を付与した。切片を4℃で5−7日間露光し、コダックD−19中で20℃において4分間現像し、蒸留水で1分間洗浄し、30%チオ硫酸ナトリウム中で10分間固定し、蒸留水中で洗浄した。
電子顕微鏡検査(EM) 組織塊を0.1Mカコジル酸緩衝液(pH7.4)中の2%グルタルアルデヒド中において固定し、2%五酸化オスミウム(スティーブンス・メタルルジカル・コーポレーション、ニューヨーク州ニューヨーク)中で後固定した。他のEM化学薬品はすべてラッド・リサーチ・インダストリーズ・インコーポレーテッド(バーモンド州バーリントン)から入手された。組織を塊のままpH5.2マレイン酸緩衝液(0.05M)中の2%酢酸ウラニルで染色し、脱水し、ラッドLX−112に包埋した。トルイジンブルーで染色した1μmの切片を用いて移植体の位置を調べた。トリミング後にブロックを薄く切断し、酢酸ウラニルおよびクエン酸鉛で染色した。検体をフィリップス400透過型電子顕微鏡により検査した。
心電図(ECG)分析 表面ECG記録のために、マウスを麻酔し(2.5%アベルチン、0.015ml/体重g、腹腔内、フルカ・ケミカルズ、ニューヨーク州レーク・ロンコムコマ)、標準第I導出位置に表面電極を配置し、A/D変換器(クールボーン・インスツルメンツ、ペンシルベニア州リーハイ・バレー)に連結したナルコ・バイオシステムズ(テキサス州ヒューストン)高利得増幅器によりECGを記録した。
血漿酵素アッセイ(PEA) 乳酸デヒドロゲナーゼ(LDH)イソ型アッセイのために、麻酔(2.5%アベルチン、0.015ml/体重g、腹腔内)下での眼窩後洞採血により血漿を分離した。血漿を1%アガロースゲル上で分画し(CKイソ酵素電気泳動システム、シグマ・ダイアグノスティックス、ミズーリ州セントルイス)により視覚化した。ニューヨーク州コーニング)、LDHイソ型をTNBT−ホルマザン組織化学的アッセイ(LDHアッセイキット、シグマ・ダイアグノスティックス、ミズーリ州セントルイス)により視覚化した。
B.結果
これらの研究においては、AT−1心筋細胞(心臓内でT−Ag癌蛋白質を発現するトランスジェニック動物に由来する)を同系マウスの心筋に直接に注射し、移植した材料の生存度を評価した。予備実験における注射部位の位置決定を容易にするために、AT−1心筋細胞を移植前に短期間、BODIPYと共にインキュベートした。BODIPYは生細胞の蛍光追跡を可能にする無毒性ダルタチオン反応性染料である。移植部位はFITCキューブ(cube)を用いる蛍光顕微鏡検査によって容易に視覚化された。以後の実験にはBODIPYを用いなかった。
AT−1心筋細胞注射を受けた動物の50%(14/28)が心臓内移植体を発達させた。大部分の場合、移植体は被包されておらず、また瘢痕を形成した心筋で囲まれてもいなかった。光学顕微鏡のレベルで、移植AT−1心筋細胞が宿主心筋細胞と直接に近接しているのが観察された。抗−T−Ag一次抗体(162−T)を用い、次いで西洋ワサビペルオキシダーゼ結合した二次抗体を用いる免疫−ペルオキシダーゼアッセイにより、AT−1心筋細胞の同一性が確認された。抗−T−Ag抗体の特異性は先に確立されている(10,11)。黒色沈着物が移植体の心筋細胞核上に見られたが、宿主心筋には見られず、移植体がAT−1心筋細胞からなることが確認された。同様な結果が他の抗−T−Ag抗体を用いて得られ、一次抗体の不在下ではシグナルが見られなかった。
生存AT−1心筋細胞が移植後少なくとも4カ月間は観察された。この期間中、ある程度の移植体増殖が行った;3[H]−チミジン取り込み分析により移植細胞におけるDNA合成が検出された。核への同位体取り込みにより証明されるように、10%のAT−1心筋細胞核がDNAを合成していた。しかしその率は培養AT−1心筋細胞につき観察されたものよりかなり低く、この場合は同様な3[H]−チミジンパルス後に50%の細胞がDNAを合成した。数例において、移植AT−1心筋細胞が心臓下腔内に位置していた。
心膜移植体が慢性拒絶を受けるか否かを判定するために、免疫組織学的実験を採用した。1カ月より古い移植体はマウス白血球に特異的な抗体と反応しなかった;同一切片上に位置する血管において観察されたシグナルはその実験に対する陽性対照を与えた。同様にマウスのマクロファージおよびリンパ球を検知する抗体は心臓内移植体と反応しなかった;この場合も同一切片上に位置する血管において陽性シグナルが観察された。総括すると、これらの結果は同系宿主による慢性移植体拒絶がないことを示す。この結果は、サイクロスポリン処置(50mg/体重kg、毎日腹腔内投与)が心臓内移植の成立頻度に有意に影響しなかったという所見により支持される(50%の成功率、n=6)。宿主動物の性別も移植体形成率に有意に影響しないと思われた(雄においては46%の成功率、n=13;雌においては53%の成功率、n=15)。移植の成立頻度は初期の時点で検査した動物の場合(移植の1−40日後、47%、n=15)は、より後の時点で検査した場合(移植の40−120日後、54%、n=13)と比較して類似していた。最後に、細胞を左心室の遊離壁(free wall)または心尖のいずれに運搬した場合でも、同様な成立頻度の心臓内移植が観察された。
AT−1心筋細胞移植体の電子顕微鏡分析により、被包のないことが確認された。高解像力の画像は、移植体内の隣接細胞間に結合複合体が良好に発達していることを明示した。移植心臓細胞は、インビボにおけるAT−1腫瘍に典型的な多数のポリリボソームおよび脱分化した筋原繊維性の超微細構造(6)を含んでいた。心房由来の筋細胞に予想されるように、AT−1心筋細胞移植体内に高電子密度の分泌性顆粒も見られた。移植体周縁の宿主心筋細胞は、良好に形成されたサルコメアを伴う正常な超微細構造を示した。薄い基底膜のみがAT−1心筋細胞と宿主心筋細胞を分離していたが、これら2細胞タイプ間には結合連結部は見られなかった。
AT−1心筋細胞移植体の存在が自己調律(autonomic rhyThm)に影響を及ぼすか否かを判定するために、表面心電図測定を行った。虚偽動物からの記録と移植体を含む動物からのものとの間に認めうるほどの相異は見られなかった。それぞれの場合、被験動物は正常な洞調律を示し、麻酔心拍数は約400回/分であった。正常なP−QRS連結が維持され、これは移植AT−1心筋細胞が異所性ペースメーカーとして作用しなかったことを示す。この後者の結果は、AT−1心筋細胞がインビボ(12)および培養(3)の双方において自発性電気的活性を示すという所見からみて重要である。明白な不整脈がないことも、移植体誘発性の心筋リモデリングに伴って有意の輪回リズムが発生しなかったことを示す。
表面ECGのほかに、AT−1心筋細胞移植体を保有するマウスにおいて血漿LDH水準を評価した。循環中に心臓LDHイソ型が存在することは、十分に成立した心筋梗塞の特徴である。移植前にはマウス血漿中に心臓LDH(イソ型−1)は見られなかった。AT−1心筋細胞の導入後に血漿中に心臓イソ型の一過性出現が生じたが、これは宿主心筋に対する損傷および損傷を受けたAT−1心筋細胞を反映している可能性が最も高い。移植のための外科処置後に血漿骨格LDHイソ型の一過性増加も見られたが、これは恐らく経胸腔切開により生じた損傷を反映していると思われる。血漿LDHプロフィルは移植の7日後までには正常に戻った。その後は血漿LDHプロフィルは移植体の存在にもかかわらず正常に維持された。
実施例2
安定なC2C12筋芽細胞移植体の形成
A.方法
C2C12細胞培養および心筋移植プロトコル C2C12筋芽細胞はATCCから得た。20%ウシ胎児血清、1%ニワトリ胚エキス、100単位/mlペニシリン、および100μg/mlストレプトマイシンを補充した高グルコースのダルベッコ改良イーグル培地(DMEM)中で低密度において培養することにより、細胞を未分化状態に維持した。ある研究のために、2%ウマ血清および抗生物質を補充したDMEM中で培養することにより、筋発生性の分化を誘発した。注射の直前に筋芽細胞をトリプシンで採取し、無血清DMEMで3回洗浄し、開胸手術した同系C3Heb/FeJマウス成体(ジャクソン・ラボラトリーズ)の心室心筋内に直接に注射した:(7)の記載に従う。細胞(4−10×104)は2−3μlの容量で30ゲージの注射針を備えたプラスチック注射器により注射された。
組織学的分析 実施例1の場合と同様に心臓を摘出し、凍結保護し、包埋し、切断した。HおよびE染色、ならびに[3H]−チミジン取り込みアッセイも実施例1の場合と同様に実施した。免疫組織学的分析のために、メタノール固定した(−20℃、10分)切片をモノクローナル抗−骨格ミオシンH鎖抗体(MY−32、シグマ・ケミカル・コーポレーション)と反応させ、次いでローダミン共役−ヒツジ抗−マウスIgG f(ab′)2フラグメント(ベーリンガー・マンハイム)と反応させ、エピフルオレッセンスにより視覚化した。
電子顕微鏡検査 EMを実施例1の場合と同様に実施した。
心電図分析 ECGを実施例1の場合と同様に実施した。
血漿酵素アッセイ PEAを実施例1の場合と同様に実施した。
B.結果
C2C12に例示されるように、培養において分化して筋管となる能力をもつ数種類の筋芽細胞が知られている(13)。C2C12筋芽細胞は、C3Hマウスの損傷した大腿筋の体外培養物に由来する。富血清培地に維持した場合、筋芽細胞は急速に増殖し、未分化表現型を保持する。しかし貧血清培地で培養した場合、筋発生性分化が誘発される。C2C12細胞は細胞周期から離れて融合し、これにより多核筋管を形成する。多数の筋特異性遺伝子産物の出現により証明されるように、筋発生性分化も誘発される。このようにこのモデルにおいては増殖と筋発生性分化は互いに排他的である(14)。インビトロでの筋芽細胞の分化はインビボでの衛星細胞仲介による神経繊維再生に類似すると考えられる。
筋芽細胞を同系C3Heb/FeJマウスの心筋に直接に注射し、この移植材料の生存度を評価した。CC12筋芽細胞の心臓内移植を受けたマウスの100%(13/13)が心臓内で移植体を発達させた。生存移植体が移植後6カ月間観察された(これがアッセイした最終時点であった)。すべての例において、移植された材料は被包されなかった。分化した状態の移植CC12細胞を、抗−ミオシンH鎖抗体(MY−32)を用いる免疫組織学的アッセイにより測定した。この抗体は筋芽細胞とも心筋ミオシンH鎖とも反応しない。分化したC2C12細胞は筋芽細胞注射を受けたすべての心臓において観察されたが、個々の細胞の移植効率は測定されなかった。追加の対照として、AT−1心臓内移植体を保有する心臓(実施例1参照)をMY−32抗体で検査した。染色が見られず、これによりC2C12移植体において見られたシグナルは宿主心筋細胞における骨格ミオシンH鎖誘導によるものであるという可能性は除かれる。
前記の実施例1はAT−1心筋細胞が同系心筋において安定な移植体を形成することを証明する。しかしこれらの細胞がインビボで増殖する能力を保持したという所見は、心臓内移植の成功には持続的な細胞分裂が必要であるかも知れないという可能性をもたらす。従ってC2C12移植体の増殖状態を検査した。DNA合成は実質的に観察されず(トリチウム化チミジンの取り込みにより評価)、これは移植したC2C12細胞の大部分が実際に細胞周期からはずれたことを示す。連続切片の検査により、DNA合成を行っているのは移植体内またはその付近の細胞の0.1%未満であることが示される。この結果はリモデリング過程での繊維芽細胞の増殖を反映している可能性が最も高い。AT−1移植体の場合と同様に、移後値2カ月の時点でC2C12移植体の免疫組織学的分析によりマクロファージ、炎症性白血球またはリンパ球浸潤を検出することができず、これは同系宿主による慢性移植拒絶がないことを示す。
光学顕微鏡の水準でC2C12心臓内移植体はHおよびEならびにMY−32免疫蛍光染色の双方により細胞不均一性を示した。C2C12移植体の細胞構成をさらに解明するために、電子顕微鏡分析を採用した。トルイジン染色した1μmの切片を100μm間隔で探査して、EM分析用の移植体部位の位置を調べた。位置が確認されると、そのブロックから薄い切片を調製した。骨格筋細胞に典型的な形態をもつ細胞が移植体全体に観察された。良好に発達したサルコメア間に位置する豊富なミトコンドリアが容易に検出された。顕著なZ膜ならびに太いフィラメントおよび細いフィラメントが観察された。場合により、伸長したt−管および波打った細胞膜が移植筋細胞中に観察された。良好に発達した筋細胞のほかに、より分化の低い第2の細胞タイプがC2C12移植体中に観察された。最も注目すべきことは、これらの細胞は大きな核:細胞質比を示し、核周辺に顕著なヘテロクロマチンの帯を伴うことであった。中心部に位置する中程度の量のヘテロクロマチンも検出された。これらの細胞には、限られた粗面小胞体およびわずかなミトコンドリアしか観察されなかった。類似の超微細構造特性がインビボおよび培養における衛星細胞の属性であると考えられている(15,16)。
宿主の心臓機能に対するC2C12心臓内移植体の有害な作用を評価するために、2つの研究を開始した。第1の研究において、表面心電図分析は対照マウスと被験マウスから得た記録間に認めうるほどの相異を検出し得なかった。すべての被験動物が正常なP−QRS連結を有し、麻酔心拍数約400回/分を伴う正常な洞調律を示した。これらのデータは、心臓内筋芽細胞移植体が明白な不整脈を誘発しなかったことを示す。第2の研究においては、移植体を保有する動物において血漿LDH水準を監視した。循環中に心臓LDHイソ型が存在することは、十分に成立した心筋梗塞の特徴である。移植前の血漿中には心臓特異性LDHイソ型(イソ型1,2および3)は観察されなかった。移植直後に血漿中に心臓イソ型の増加が観察されたが、これは宿主心筋に対する損傷を反映している可能性が最も高い。血漿骨格LDHイソ型(イソ型5)の一過性増加も見られたが、これは恐らく経胸腔切開により生じた損傷を反映していると思われる。血漿LDHプロフィルは移植の7日後までには正常に戻った。
実施例3
安定な胎児心筋移植体の形成
A.方法
心筋細胞培養および心臓移植プロトコル α−心臓ミオシンH鎖(MHC)プロモーターおよび修飾したβカラクトシダーゼ(nLAC)リポーターを含む融合遺伝子を保有するトランスジェニックマウスを形成した。MHC−nLACトランスジェニックマウスを形成するために、MHC−nLAC挿入DNA(図1)をガラスビーズへの吸着により精製し、5μg/mlの濃度で溶解し、近交系C3H3B/FeJの胚の1細胞の核にマイクロインジェクションした:確立したプロトコルに従う(17)。ポリメラーゼ連鎖反応(PCR)分析を用いて創始動物を同定し、トランスジーンの分離を監視した。センス鎖プライマー5′−GGTGGGGGCTCTTCACCCCCAGACCTCTCC−3′をMHCプロモーターに配置し、アンチセンス鎖プライマー5′−GCCAGGGTTTTCCCAGTCACGACGTTGT−3′をnLACリポーターに配置した。PCR分析は(18)の記載に従った。MHCプロモーターは4.5kbの5′側フランキング配列、およびエキソン1−3を包含する1kbの遺伝子を含むが、開始コドンは含まなかった。nLACリポーターは真核細胞翻訳開始部位およびSV40核局在化シグナルの両方を保有するように修飾された(19)。mP1配列はマウスプロタミン1遺伝子由来のイントロン、ならびに転写終止シグナルおよびポリアデニル化シグナルを保有していた。
βガラクトシダーゼ(βGAL)活性およびDAPIエピフルオレッセンスの検査に用いる調製物を得るために、トランスジェニック動物をヘパリン処理(10,000U/kg、腹腔内)したのち頸部脱臼により屠殺した。通気した(95%O2,5%CO2)KHB緩衝液(105mM NaCl,20mM NaHCO3,3.8mM KCl,1mM KH2PO4,1.2mM MgSO4,0.01mM CaCl2,1mMマンニトール,10mMタウリン,10mMデキストロース,5mM ピルビン酸Na)のビーカーに心臓を入れた。次いで心臓を大動脈により懸垂し、2.5mM EGTAを含有する通気したKHB(37℃で0.5ml/分)で灌流したのち、KHB中の0.17%コラゲナーゼ(タイプI、ワースィントン・バイオケミカル、ニュージャージー州フリーホールド)で灌流した。心臓を弛緩するまで灌流し、心室をカミソリで刻み、パスツールピペットで摩砕処理することにより、分離した細胞を得た。少なくとも1時間のホルマリン固定後に懸濁液を濾過し、正に帯電したスライド(スーパーフロスト・プラス(Superfrost Plus)、フィッシャー、ペンシルバニア州ピッツバーグ)上に塗抹し、乾燥させた。
注射用の単一細胞を分離するために、15日目の胚(膣栓により妊娠の開始を判定)を有する雌を頸部脱臼により屠殺した。胚を摘出し、断頭し、心臓をPBS下に採取し、心室と心房を分離した。トランスジェニック心房(心臓βGAL活性により確認)をDPBS(ダルベッコのリン酸緩衝食塩水、シグマ)中の0.1%コラゲナーゼ(ワースィントン)中で45分間消化し、10%FBSを含むPC−1培地(ベントレックス、ミネソタ州クーンズ・ラピッヅ)中においてバスツールピペットで摩砕処理して、単一細胞懸濁液を得た。
分離直後に胚性心筋細胞をDPBSで3回洗浄し、実施例1に従って開胸処置した同系マウス(ジャクソン・ラボラトリーズ)の心室心筋内に直接に注射した。1−10×104細胞を2−3μlの容量で、30ゲージの注射を備えたプラスック注射器により注射した。
組織学的分析 HおよびE、X−GAL、免疫組織学的分析およびチミジン分析のために、実施例1の場合と同様に頸部脱臼後に心臓を摘出し、30%スクロース中で凍結保護し、包埋し、クリオミクロトームにより10μmに切断した。HおよびE染色、心臓内移植体拒絶に関する監視、ならびに[3H]−チミジン取り込みアッセイも実施例1の場合と同様に実施した。βGAL活性をアッセイするために、切片をPBS中で中和し、アセトン:メタノール(1:1)中で後固定し、次いで1mg/mlのX−GAL(5−ブロモ−4−クロロ−3−インドリル−β−D−ガラクトシド)、5mMフェリシアン化カリウム、5mMフェロシアン化カリウムおよび52M塩化マグネシウムをPBS中に含有する混合物を積層した。青色発色団の出現により陽性染色が指示される。一次抗体で処理したのち、アビジン−ビオチン(ABC)キット(ベクター、ラボズ・カリフォルニア州バーリンゲーム)によりシグナルを視覚化した。心臓を前記により処理し、切片をメタノール:アセトン(1:1)中で後固定し、HおよびEで染色し、蒸留水で1:1に希釈した写真乳剤(イルフォードL.4、ポリサイエンシズ)の薄い層で被覆した。実施例1の場合と同様に切片を露光、現像、洗浄、固定および洗浄した。単一細胞調製物のX−GAL染色を実施例1の場合と同様に実施した。単一細胞調製物中の核の視覚化のために、スライドをPBS中のDAPIで染色し(0.28μM、室温で3分、ベーリンガー・マンハイム)、PBS中で3回洗浄し、グリセリンに溶解した2%没食子酸プロピル中で湿式マウントした。冠心臓切片を得るために、マウスを頸部脱臼により屠殺し、心臓を採取し、ランゲンドルフ装置により0.1Mカコジル酸緩衝液(pH7.4)中の2%グルタルアルデヒドで灌流した。同緩衝液中で一夜浸漬固定したのち、ビブラトーム(カムデン、英国ロンドン)で200μmの冠心臓切片を作成した。移植体の位置を調べるために、切片をプールし、前記に従ってX−GALによりβGAL活性用として染色した。
電子顕微鏡検査 MHC−nLAC胚移植体の位置を前記に従って冠心臓切片中において調べた。トリミング後に組織を2%五酸化オスミウム(スティーブンス・メタルルジカル・コーポレーション、ニューヨーク州ニューヨーク)中で後固定した。次いで組織を脱水し、ラッドLX−112(ラッド・リサーチ・インダストリーズ)に包埋した。移植領域をさらにトリミングし、薄く切断し、酢酸ウラニルおよびクエン酸鉛で染色した。検体を実施例1の場合と同様にフィリップス400透過型電子顕微鏡により検査した。
心電図分析 ECG分析を実施例1の場合と同様に実施した。
B.結果
前記に従って形成したトランスジェニックマウスは、MHCプロモーターおよびnLACリポーターを含む融合遺伝子を保有していた。nLACはSV40核輸送シグナルを保有し、その結果βガラクトシダーゼ活性が標的細胞の核内に蓄積する。4つのトランスジェニック系列が形成され、2つ(MHC−nLAC−2およびMHC−nLAC−4)を以後の分析用として選択した。MHC−nLACトランスジーンが適切な細胞系列マーカーを提供することを確認するために、トランスジェニック心筋細胞においてβガラクトシダーゼ活性(βGAL)を評価した。逆行(retrograde)コラゲナーゼ灌流により調製された単一細胞調製物をβGAL活性およびDAPIニピフルオレッセンスにつき同時に検査したところ、99.0±0.45%(n=400)のトランスジェニック心筋細胞核がβGALを発現し、これに対し非心筋細胞中にはβGAL活性は検出されなかった。さらに非トランスジェニック対照心筋細胞中に該βGAL活性は検出されなかった。
胚15日目のトランスジェニックマウスから心臓のコラゲナーゼ消化により単一細胞懸濁液を調製した。染料排除アッセイにより証明されるように、この方法で単離された心筋細胞の95%以上が生存性であった。心筋細胞は同系非トランスジェニック動物の左心室自由壁へ運搬された。移植心筋細胞はMHC−nLACトランスジーンによりコードされる核βGAL活性によって容易に、かつ確実に同定された。移植心筋細胞は運搬地点から遠位にしばしば観察された;移植細胞のこの分散が心筋細胞の移動を反映するのか、または注射過程で切断平面に沿って生じる受動的拡散を反映するのかは、現在のところ明らかでない。胚心筋細胞の心臓内注射を受けた動物の約50%(7/13)が移植体を発達させた。この移植体形成の成功頻度は、細胞の調製および移植のプロトコルが最適化されるのに伴って高まると思われる。
βGAL活性用に処理されたHおよびE染色切片の光学顕微鏡分析は、移植心筋細胞(青色の核)が宿主心筋細胞(紫色の核)と直接に近接していることを示した。他のHおよびE分析によって有意の移植体被包を検出することはできなかった。この観察された移植心筋細胞と宿主心筋細胞の近接、および被包がないことは、2細胞タイプ間の連結が成功するための前提条件である。
19日令の心臓内移植体の連続切片をHおよびE、βGAL活性、ならびにマクロファージおよび白血球免疫反応性に関して処理した。動物は免疫抑制されていないという事実にもかかわらず、移植体拒絶の徴候は見られなかった。免疫組織学的分析のための陽性対照として、不適合MHCハプロタイプの移植体を形成した;これらの心臓においては移植体拒絶が明瞭であった。トリチウム化チミジン取り込み分析により、非心筋細胞のDNA合成は明らかであるが、0.6%(n=156)のβGAL陽性核がDNAを合成しているにすぎないことが示された。胚15日目の供与体細胞は移植した時点ではなお有糸分裂活性を備えていた(標識指数約29%)ので、移植後19日目のβGAL陽性細胞において観察されたDNA合成水準が著しく低いことは、MHC−nLAC胚心筋細胞が最終分化を行ったことを示唆する。
光学顕微鏡分析により観察された移植心筋細胞と宿主心筋細胞の近接は、これら2細胞タイプ間に細胞間直接連結を検出しうるか否かの判定を容易にした。X−GAL反応生成物は、透過型電子顕微鏡(TEM、19参照)により検出しうる高電子密度沈殿である。グルタルアルデヒド灌流−固定心臓からのビブラトール切片をβGAL活性用に染色し、こうして確認された移植領域をトリミングし、TEM用に包埋した。βGAL陽性核は1μm切片の光学顕微鏡分析によって容易に観察された。X−GAL反応生成物は、包埋過程でわずかな程度の核浸出が起こったため核周辺外観を示した。同時の心筋細胞を連続した薄い切片のTEM分析により同定した。該周辺βGAL活性が存在しないため光学顕微鏡においては容易に同定されなかった宿主心筋細胞が、電子顕微鏡により移植細胞と近接しているのが観察された。多数の結合複合体が宿主心筋細胞と移植心筋細胞の間に存在し、これは高度の細胞間連結を示している。宿主心筋細胞と職種体心筋細胞の間の細胞間連結の多数の例が、移植領域全体に観察された。重要なことは、細胞間連結は多数の宿主細胞にわたるβGAL陽性心筋細胞から追跡することができ、従って移植心筋細胞が機能性融合細胞に関与している可能性が立証されることである。
TEM分析は豊富な細胞間連結が移植心筋細胞と宿主心筋細胞の間に存在することを証明するほか、移植心筋細胞が高度に分化していることを解明した。完全なサルコメアを形成する筋原繊維、細胞間の多数の結合複合体、および豊富なネトコンドリアを含めて、成体心筋細胞の正常な特徴が観察された。事実、X−GAL反応生成物の存在を別として、移植心筋細胞は宿主細胞と区別できなかった。さらに二核βGAL陽性細胞を心臓内移植体中に検出することができた。二核形成は成体げっ歯類の心筋細胞の特徴であるから、この所見は移植心筋細胞が最終分化を行ったことをさらに支持する。
結合した胚心筋細胞移植体の存在が宿主心臓の自動性に負の作用を及ぼすか否かを判定するために、表面ECG記録を採用した。移植体保有動物から得たECGトレースは虚偽手術した対照と区別することができず、マウスに特徴的なPおよびQRSコンプレックスを示した。高度の細胞間連結が移植心筋細胞と宿主心筋細胞の間に存在するにもかかわらず、移植体保有動物に不整脈の徴候はなかった。
実施例4
実質的に純粋な心筋細胞培養物の調製
胚幹細胞を、不死化していない心筋細胞の実施的に均一な集団を生成しうるように遺伝的に修飾した。母体ES細胞系(D3)をpGK−HYG(ハイグロマイシン)プラスミドおよびMHC−neor遺伝子を含むプラスミドで同時トランスフェクションした。pGK−HYGプラスミドはトランスフェクションしたES細胞に対する選択を与え、一方mMHC−neor遺伝子は分化した細胞に対する第2ラウンドの選択を容易にする:G418の存在下でのインキュベーションにより非心筋細胞(すなわちMHCプロモーターが活性でない細胞)が排除される。
ハイグロマイシンの存在下での培養によって、安定にトランスフェクションしたES細胞が選択された。プラスミドを線状にし、1180μF、220Vにおけるエレクトロポレーションにより幹細胞に導入した。トランスフェクションした細胞を、10%の予備選択されたFBS、0.1mM β−メルカプトエタノール、非必須アミノ酸、PenStrepおよびLIFを補充したDMEM中において維持し、培地にハイグロマイシンを添加することにより形質転換体を選択した。次いで同時トランスフェクション体を両方のトランスジーンに特異的なPCR分析により同定した。このトランスフェクションにより両方のトランスジーンを保有する9Aと表示される細胞系が形成された。
9A ES細胞において、2×106個の細胞をコーティングされていない100mmの細菌用ペトリ皿にLIFの不在下で接種することにより、心臓発生を誘発した。培養の8日後に自発収縮活性を示す細胞(すなわち心筋細胞)のパッチが多数観察された。この時点で培地にG418を添加し、細胞をさらに9日間インキュベートした。この処理に際して、多数の非心筋細胞がG418により死滅するのが認められた。重要なことは、G418が心筋細胞には認めうるほどの影響を与えず、心筋細胞は実験期間中それらの自発拍動活性を維持したことである。
合計9日間のG418選択後に、生存細胞をコラゲナーゼおよびトリプシンで解離し、次いでフィブロネクチンコーティングした顕微鏡スライドガラスに再び接種した。細胞をさらに24時間培養して、それらを継代培養から回復させ、次いで免疫細胞学的分析のために固定した。細胞をMF20、すなわちサルコメア性ミオシンを認識するモノクローナル抗体と反応させた。次いで細胞を個々に、心筋細胞に対するマーカーであるサルコメア性ミオシンの存否につき計数した。結果は以下のとおりであった:
計数した全細胞数: 794
MF20+の細胞数: 791
MF20−の細胞数: 3*
心筋細胞の割合: 99.6%
*MF20で染色されなかったこれら3細胞もやはり心筋細胞であった可能性はある。この抗体は単体ミオシンとは反応しないからである。
従って心臓特異性プロモーター下での薬物(ネオマイシン)耐性の発現により培養におけるES由来の心筋細胞の実質的に純粋な集団を選択しうることが証明された。このような集団を用いて、前記の各実施例において述べた方法で心筋移植体を形成することができる。
実施例5
移植体による蛋白質の運搬
A.方法
C2C12細胞培養およびトランスフェクション C2C12筋芽細胞(ATCC)を、20%ウシ胎児血清、1%ニワトリ胚エキス、100単位/mlペニシリン、および100μg/mlストレプトマイシンを補充した高グルコースのダルベッコ改良イーグル培地(DMEM)1中で低密度において培養することにより、未分化状態に維持した。ある研究のために、2%ウマ血清および抗生物質を補充したDMEM中で培養することにより、筋発生性の分化を誘発した。
修飾したトランスフォーミング増殖因子−ベータ1(TGF−β1)cDNAを駆動するメタロチオネイン(MT)プロモーターを含む融合遺伝子をサミュエルら(20)から得た。メタロチオネインプロモーターの転写活性は、細胞培養培地の貴金属含量を調整することにより調節しうる。TGF−β1 cDNAは部位特異的突然変異を保有し、その結果Cys223およびCys225がセリンに変換された。この修飾((21)に記述される)の結果、二量体を形成し得ず、結果的に正常な転写後調節を受けないTGF−β1分子が作り出される。この修飾されたcDNAを発現する細胞は、プロセシングされた活性TGF−β1を構成的に分泌する(20)。MT−TGF融合遺伝子をリン酸カルシウムトランスフェクションによりC2C12筋芽細胞に導入し;安定なトランスフェクション体をSV40−neorトランスジーンとの同時トランスフェクションにより選択した。4種類の独立したクローンを単離し、サザーンブロット分析によりトランスジーンの存在を確認した。まず種々のクローンの細胞系におけるTGF−β1発現の相対水準をノーザンブロット分析により評価し、C2(280)と表示される1つの系を以後の実験に用いた。
心臓移植プロトコル 移植プロトコルは実施例1に記載したものと同様である。手術の14日後に、移植体を保有する動物に貴金属を投与した(飲料水中に25mMのZnSO4)。亜鉛処理を実験が終了するまで継続した(1−4週間)。
組織学的分析 パラフィン切片を得るために、10%中性緩衝ホルマリン中で心臓を固定し、濃度勾配(graded)アルコール類により脱水し、パラフィンで浸潤した。次いで組織塊を6μmに切断した。切断直後に製造業者の説明書(シグマ・ダイアグノスティックス)に従ってHおよびE染色を直接に実施した。
[3H]−チミジン取り込みのために、実施例1の場合と同様にマウスにボーラス投与し、屠殺した。前記に従って心臓を摘出し、パラフィン包埋のために処理した。実施例1の場合と同様にオートラジオグラフィーを実施した。
B.結果
貴金属誘導に応答した組換えTGF−β1の発現を、C2(280)筋芽細胞および筋管において検査した。トランスジーン転写体(1.8kb)は、ノーザンブロット分析により内在性TGF−β1遺伝子(2.5kb)と容易に区別された。培地に貴金属を添加すると、C2(280)筋芽細胞および筋管において組換えTGF−β1転写体が顕著に増加した。前記に示したように、C2(280)細胞により発現する修飾TGF−β1は構成性の生物学的活性をもつはずである。これを直接に試験するために、C2(280)筋芽細胞および筋管から得たならし培地を増殖阻害アッセイ法により検査した。
C2(280)筋芽細胞を用いて同系C3Heb/FeJマウスにおいて心臓内移植体を形成した。移植体の存在はHおよびE染色切片において容易に検出された。C2(280)細胞の心臓内注射を受けた動物の100%(N>50)が移植体を発達させ続けた。興味深いことに、HおよびE分析はC2(280)移植体が非修飾C2C12細胞により形成されたものと比較して分化が若干低いことを示した。この結果は、骨格ミオシンH鎖を認識するモノクローナル抗体を用いた免疫組織学的分析によって確認された。
C2(280)移植体トランスジーン発現を、抗−TGF−β1抗体を用いた免疫組織学的分析によって評価した;TGF−β1発現はC2(280)移植体において容易に検出された。陰性対照として、C2C12筋芽細胞により形成される移植体においてTGF−β1発現を評価した。予想どおりTGF−β1発現の相対水準はC2(280)移植体と比較して著しく低かった。
TGF−β1は周知の血管形成因子である。従って、MT−TGFトランスジーンを発現する移植体において血管形成応答の増大が起こるか否かを判定するために、血管内皮細胞において3H−チミジン取り込み分析を評価した。血管内皮細胞におけるDNA合成は、3H−チミジン(H−THY)の一回ボーラス注射投与下のC2(280)移植体において直ちに認められた。これに対し非トランスフェクションC2C12移植体においては血管内皮細胞のDNA合成は著しく低かった(表1)。血管形成応答が移植体材料のみによるものであるという可能性を除くために、同様な大きさおよび日令のC2C12移植体とC2(280)移植体間で3H−チミジン取り込みを比較した(表1)。すべての分析において、DNAを合成している血管内皮細胞の数が著しく増加した。最後に、チミジン取り込みにより、ある割合の移植筋芽細胞が増殖し続けていることも明らかになった。この所見はTGF−β1が筋分化に及ぼす既知の阻害作用と一致し、C2(280)移植体が分化していない外観をもつことに関与している可能性が極めて高い。
以上の記載において本発明を詳細に説明および記述したが、これらは説明のためのものであって限定的性質のものではないとみなすべきであり、好ましい態様を記載したにすぎず、本発明の精神に包含されるすべての変更が保護されるべきであると解される。

- 動物の心筋組織中への骨格筋芽細胞又は心筋細胞の導入による、動物における安定な心筋細胞性移植体の形成において使用するための、生存可能な骨格筋芽細胞又は生存可能な心筋細胞を含む細胞組成物。
- 細胞が腫瘍形成性でない、請求項1の細胞組成物。
- 細胞がインビトロでの培養によって得られる、請求項1又は2の細胞組成物。
- 細胞が導入された選択マーカーを含む、請求項1−3のいずれか1項の細胞組成物。
- 細胞が治療用タンパク質を心筋組織に運搬するためのものである、請求項1−4のいずれか1項の細胞組成物。
- 治療用タンパク質が組換えである、請求項5の細胞組成物。
- 細胞が、組換え治療用タンパク質をコードする導入されたDNAを保持する、請求項6の細胞組成物。
