【発明の属する技術分野】本発明は、しみ、そばかすな
どの色素沈着症の原因となるメラニン抑制剤に関し、さ
らに詳しくはカモカモを水、親水性有機溶媒またはこれ
らの混合物で抽出処理することにより得られるカモカモ
の抽出物を有効成分として含有するメラニン抑制剤に係
るものである。

【従来の技術】しみ、そばかす、肝斑および日焼け後の
色素沈着は、加齢に伴い発生、増加あるいは消失しにく
くなり、中高年齢層の肌の悩みのひとつとなっている。
これらの色素沈着症の発症メカニズムは、いまだ明確に
はされていないが、紫外線、メラノサイト刺激ホルモン
(MSH)などの作用によりメラノサイトのメラニン合
成機能が亢進したためと考えられている。また、表皮角
化細胞(ケラチノサイト)の加齢に伴う角化遅延傾向
も、メラニンの表皮外への排泄速度を遅延させ、メラニ
ン合成機能の亢進と合わせて、表皮内のメラニン顆粒密
度の増加によるものと考えられている。さらにそれらの
色素沈着部は局部的に存在し、周囲の正常皮膚色と明ら
かな差異が生じることから、局部的なメラニン合成機能
の亢進、あるいはメラニン合成をコントロ−ルする機構
の変調の結果とも考えられている。従来から、これらの
後天的な色素、すなわちメラニンの沈着部を正常な皮膚
色にまで回復させる研究が行われ、これまでに数多くの
提案が報告されている。

スの防止及び改善効果をも有する皮膚外用剤(特開昭6
3−303919号公報参照)、また霊芝の子実体から
の抽出成分を配合した化粧料であって、メラニン生成抑
制作用、メラニン淡色化作用を有する製剤(特開平2−
124809号公報参照)等が提案されている。

【発明が解決しようとする課題】しかしながら、これら
のメラニン抑制剤の抑制効果は、必ずしも満足できるも
のではなく、更に優れた効果を有するメラニン抑制剤の
開発が強く求められている。

【課題を解決するための手段】そこで本発明者らは上記
課題を解決するため、安全性、安定性が高く、メラニン
抑制効果に優れた機能を有するメラニン抑制物質、特に
該機能を有する種々の植物の抽出物について鋭意研究の
結果、カモカモ(MYRCIARIA DUBIA)を
水、親水性有機溶媒またはこれらの混合物で抽出処理す
ることにより得られるカモカモの抽出物が、メラニンの
生成を抑制する機能を保有すること、美白作用、さらに
は保湿作用、皮膚の老化防止にも有効であることを見い
だし、本発明に至った。
【0006】カモカモはフトモモ科に属する食用植物で
あって、ペルー東部からブラジル西北部のアマゾン盆地
の河岸又は流水のある湖沼の岸に生える潅木で、その果
実のビタミンC含有量はレモンの約100倍に達すると
言われ、又、ミネラルの含有量も高く、その他有効成分
が多く含まれていることが知られている。現地ではその
果実をそのまま食用に、または果汁を絞ってジュースな
どとして飲用に供されている。カモカモは、上述のよう
にフトモモ科の果実であり、本発明においては、生、半
乾燥、乾燥品のいずれでも使用でき、これらの原料は粉
砕して用いるのが好適である。
【0007】すなわち、本発明は、カモカモを水、親水
性有機溶媒またはこれらの混合物で抽出処理することに
より得られるカモカモの抽出物を有効成分として含有す
るメラニン抑制剤を要旨とするものである。

【発明の実施の形態】本発明のカモカモエキスは、水、
親水性有機溶媒またはこれらの混合物で抽出処理するこ
とにより容易に得ることができる。かかる抽出処理に用
いる親水性有機溶媒としては、例えば、メタノール、エ
タノール、n−プロピルアルコール、アセトン、プロピ
レングリコール、グリセリンなどの溶媒を挙げることが
できる。これらの溶媒は、例えば水と任意の割合で混合
した含水溶剤の形で用いることもできるが、約10〜約
99.9%の含水溶媒で抽出を行うのが好ましい。
【0009】抽出操作および抽出条件は用いる抽出溶剤
の種類に応じて、種々変更することができるが、抽出溶
剤として前記の如き親水性有機溶媒またはそれと水との
混合液を用いて抽出を行う場合には、例えば、カモカモ
に約1.5倍〜約50倍量の溶媒を加え、室温ないし使
用溶媒の沸点温度で、約5分ないし約24時間、静置も
しくは撹拌して行うことができる。このようにして抽出
操作を行った後、例えば遠心分離、ろ過、圧搾その他の
固液分離手段によって不溶性残渣を除去することによ
り、カモカモエキスを得ることができる。さらに必要な
らば、不溶性抽出残渣に抽出溶剤を加えて、同様の操作
をくり返し、抽出することもできる。また更に、該抽出
操作の際に、ペクチナーゼなどの酵素を添加して、酵素
処理を行うことにより、抽出効率を上げることができ
る。
【0010】本発明においては、カモカモから上記のご
とき方法で得られる抽出液をそのまま本発明の抽出物と
して使用してもよく、或いは該抽出液から溶剤を除去す
ることにより得られる濃縮物、さらには、それを減圧乾
燥または凍結乾燥して得られる乾燥物を本発明の抽出物
として利用することもできる。
【0011】さらに、上記のような方法によって得られ
る抽出物を、シリカゲル、活性炭、活性アルミナ、ベン
トナイト、酸性白土、珪藻土などの吸着剤により、脱
色、脱臭を行ってもよい。吸着操作は、抽出物をそのま
まあるいは濃縮した濃縮物を水など適当な溶媒に溶解し
た溶液に、適当量の吸着剤を添加することにより行うこ
とができる。吸着処理液は、そのまま或いは減圧下に濃
縮して得られる濃縮液、さらにそれを、例えば減圧乾燥
または凍結乾燥して得られる乾燥物を抽出物として利用
することもできる。
【0012】さらにまた、上記の脱色、脱臭処理前また
は処理後の抽出物を、例えば適当量の水に溶解して、ス
チレンビニルベンゼン、メタクリル酸エステルなどの樹
脂よりなる合成吸着剤を用いて有効成分を吸着処理し、
次いで例えば、エタノール、メタノール、アセトン、酢
酸エチルのごとき親水性有機溶媒あるいはこれらと水と
の混合溶媒で脱着操作を行って精製し、カモカモの抽出
物中の有効成分を濃縮して、本発明の抽出物として利用
することもできる。吸着、脱着操作は、バッチ方式また
はカラムによる連続方式で行うことができ、好ましくは
カラムによる連続方式が採用される。この操作後の脱着
液は、そのまま本発明の抽出物として利用することがで
きるが、また減圧下に濃縮して得られる濃縮液、更にこ
れを、例えば減圧乾燥または凍結乾燥して得られる乾燥
物を本発明の抽出物として利用することもできる。
【0013】以上述べたようにして調製されるカモカモ
の抽出物は、優れたメラニン抑制能を有する。本発明の
メラニン抑制剤は、その他の任意成分を加えて種々の形
態にすることができるが、一般的には例えば、ロ−ショ
ン、乳液状、クリ−ム状、軟膏状、スティック状、有機
溶剤による溶液状、パック状、ゲル状等とすることが好
ましい。また、該カモカモ抽出物以外に配合することの
できるその他の任意成分としては、化粧品に通常配合し
て使用されている成分、例えば、精製水、エタノール、
油性物質、保湿剤、美白剤、増粘剤、防腐剤、乳化剤、
薬効成分、粉体、紫外線吸収剤、色素、香料、乳化安定
剤、pH調整剤等を挙げることができる。上記の保湿剤
としては、例えば、γ−リノレイン酸、スフィンゴ脂
質、キサンタンガム、ヒアルロン酸、牛骨髄抽出物、水
溶性コラーゲン、キチン、キトサン、ムチン、乳酸菌培
養液、ビフィズス菌代謝物、酵母発酵代謝物、酵母抽出
物、ピロリドンカルボン酸、プルラン、マルトース、ヘ
チマエキス;美白剤としては、例えば、プラセンターエ
キス、コウジ酸、桑白皮、当帰抽出物、ワレモコウ、イ
ブキトラノオ、ビタミンCおよびその誘導体などと本発
明のカモカモ抽出物を併用すれば、美白作用、保湿作
用、皮膚の老化防止の効果を増強することができる。こ
のようにして得られる本発明のメラニン抑制剤は、皮膚
のしみ、そばかす、日焼け後の色素沈着部などの患部に
局所的に適用される。また一般にその使用量は、特に限
定されないが、通常クリーム状、軟膏状製剤の場合は皮
膚面1cm2当たり約0. 5〜10mg、液状製剤の場
合は約1〜15mg程度の範囲で使用される。

実施例
(カモカモ抽出物試料の調製)カモカモ粉砕物50g
