Description
本明細書において、非造血組織を細胞源として(例えば、非造血組織由来γδ T細胞、例えば、非造血組織由来Vδ1 T細胞)、γδ T細胞(例えば、皮膚由来γδ T細胞および/または非Vδ2 T細胞、例えば、Vδ1 T細胞、および/またはダブルネガティブT細胞)を増幅する方法が提供される。増幅方法は、実質的にTCRの刺激のない条件、および/または、インターロイキン-4(IL-4)、インターロイキン-15(IL-15)、インターロイキン-21(IL-21)および/若しくはインターロイキン-2(IL-2)が存在する条件下での、γδ T細胞(例えば、非造血組織のストロマ細胞から分離されたγδ T細胞)の培養を含む。さらに、増幅されたγδ T細胞(例えば、皮膚由来γδ T細胞および/または非Vδ2 T細胞、例えば、Vδ1 T細胞、および/またはDN T細胞)の組成物、および増幅されたγδ T細胞を用いる方法(例えば、養子T細胞療法、例えば、癌の治療用)を提供する。
本明細書に記載される本発明の態様および実施形態は、「含む(comprising)」、「なる(consisting)」および「基本的になる(consisting essentially of)」態様および実施形態を含む。本明細書で使用される場合、「a」「an」および「the」の単数形は、特に別途の指示がない限り、複数形への適用も含む。
本発明は、患者から取り出せる、または取り出された、ヒトまたは非ヒト動物の非造血組織からのγδ T細胞(例えば、皮膚由来のγδ T細胞および/または非Vδ2 T細胞、例えばVδ1 T細胞および/またはDN T細胞)を単離および増幅する方法を提供する。一部の実施形態において、γδ T細胞が取り出され、増幅される元となる非造血組織は皮膚(例えば、ヒトの皮膚)であり、当技術分野で知られる方法により取得される。一部の実施形態において、皮膚はパンチ生検により取得される。あるいは、本明細書で提供される、γδ T細胞を単離および増幅する方法は、消化管(例えば、結腸)、乳腺、肺、前立腺、肝臓、脾臓、膵臓に適用することができる。γδ T細胞は、ヒト癌組織、例えば、乳房または前立腺の腫瘍に常在するものでもよい。一部の実施形態において、γδ T細胞は、ヒト癌組織(例えば、固形腫瘍組織)由来であってもよい。他の実施形態において、γδ T細胞は、ヒト癌組織以外の非造血組織(例えば、実質的な数の腫瘍細胞を有しない組織)であってもよい。例えば、γδ T細胞は、癌の周辺または癌の隣接部位から離間した、皮膚領域(例えば、健康な皮膚)由来であってもよい。
一部の実施形態において、重要なステップは、例えば、培養数日または数週間後の、非造血組織常在性T細胞(例えば、αβ細胞、ナチュラルキラー(NK)細胞、B細胞およびγδ2T細胞、および非γδ2T細胞等を含む混合リンパ球の集団の内部)を、T細胞を取得した組織の非造血細胞(例えば、ストロマ細胞、特に線維芽細胞)から、目的に沿って分離することである。これにより、非造血組織由来のVδ1 T細胞およびDN γδ T細胞の数日および数週間の優先的かつ迅速な増幅が可能になる。
本発明は、非造血組織常在性γδ T細胞(例えば、皮膚由来のγδ T細胞および/または非Vδ2 T細胞、例えばVδ1 T細胞および/またはDN T細胞)を増幅する方法を特徴とする。これらの方法は、in vitro で実行され得る。一部の実施形態において、非造血組織常在性のγδ T細胞は、上記の方法によって分離された非造組織から分離されたγδ T細胞の集団から増幅される。一般的に、非造血組織常在性γδ T細胞は、ストロマ細胞(例えば、皮膚線維芽細胞)との物理的な接触を除去すれば、自動的に増幅可能である。したがって、上記のスキャホールドをベースとした培養方法を使用して、このような分離を誘導し、γδ T細胞の抑制を解除して、増幅を引き起こすことができる。したがって、一部の実施形態においては、増幅ステップにおいて、実質的にTCR経路の活性化は存在しない(例えば、外因性TCR経路活性化剤は、培養液中に含まれない)。さらに、本発明は、非造血由来常在性γδ TCR細胞を増幅する方法であって、支持細胞、腫瘍細胞および/または抗原提示細胞との接触を含まない方法を提供する。

本明細書、例えば後述の実施例3の情報によって、当業者は、本発明が、細胞数の倍増時間が5日間未満(例えば、4.5日未満、4.0日未満、3.9日未満、3.8日未満、3.7日未満、3.6日未満、3.5日未満、3.4日未満、3.3日未満、3.2日未満、3.1日未満、3.0日未満、2.9日未満、2.8日未満、2.7日未満、2.6日未満、2.5日未満、2.4日未満、2.3日未満、2.2日未満、2.1日未満、2.0日未満、46時間未満、42時間未満、38時間未満、35時間未満、32時間未満)の非造血組織由来のγδ T細胞(例えば、皮膚由来のγδ T細胞および/または非Vδ2 T細胞、例えば、Vδ1 T細胞および/またはDN T細胞)の増幅方法を提供することを認識するであろう。
本発明の方法により得られたγδ T細胞は、薬剤として、例えば養子T細胞療法のために使用され得る。これは、本発明の方法で得られたγδ T細胞を患者へ移植することを含む。前記治療は、自己移植、つまり、γδ T細胞を、それを取り出した同じ患者に戻す移植を行ってもよく、同種異系移植、つまり、あるヒトからのγδ T細胞を異なる患者に移植してもよい。同種異系移植を含む例において、γδ T細胞は、実質的にαβ T細胞を含まない可能性がある。例えば、αβ T細胞は、例えば増幅後に、当技術分野で周知の任意の適した手段(例えば、負の選択による、例えば、磁性ビーズの使用)を用いて、γδ T細胞の集団から枯渇させることができる。治療方法は、ドナーから得られた非造血細胞のサンプルを提供すること、前記サンプルからのγδ T細胞を培養して増幅された集団を生成すること、増幅されたγδ T細胞の集団を個別のレシピエントに投与すること、を含み得る。
特に他の指定がない限り、以下の実施例の結果を得るために、下記の方法が利用された。
フローサイトメトリーは、以下の抗体-蛍光色素コンジュゲートを用いて行った:Ki-67-BV421、CD3-BV510、Vδ 1-PeVio770、TIM-3-PE、CD9-PE、CCR3-BV421、およびCD39-BV421。サンプルを、eFluor770NIRを用いて生存率に関しても染色した。市販の抗体は、Biolegend社またはMiltenyi社から購入した。生存率色素(近赤外)は、eBioscience社から入手した。Ki-67染色は、Foxp3染色バッファーセット(eBioscience社)を用いて固定および透過化した細胞に対して行った。各実験が完了した時点で、細胞集団をPBS中で洗浄し、半分に分けた。細胞を、生存率に関してeFluor770 NIRを用いて染色し、洗浄し、染色抗体の非特異的結合を避けるために続けてTrueStain(Biolegend社)での染色を行った。サンプルの半分を、記載された表面マーカーについて染色し、一方で、他方の半分は細胞系統マーカーのみ(CD3、Vδ 1)について、および用いた表面マーカーに相当するアイソタイプ対照と共に染色した。同じ蛍光色素にコンジュゲート化した適合するマウスアイソタイプ抗体を、同じ濃度で用いた。アイソタイプ対照は、既知のヒト抗原には結合せず、したがって非特異的結合または偽陽性を示す。ヒストグラムは、その対応するアイソタイプ対照と比較して示されるか、または、FMOで示される(図1D、2A、3B、4B、6B、7A、および11-13)。データのまとめは、比較された記載のマーカーについて陽性で、つまりアイソタイプよりも高いレベルで染色された細胞の割合(%)を示す。フローサイトメトリーデータ分析は、FlowJo(バージョン10.1)を用いて行なった。
ヒト皮膚由来のVδ1 T細胞およびヒト血液Vδ1 T細胞(T細胞受容体誘導型増幅後)をソーティングし(FACS)、遠心分離し、細胞ペレットをRLTバッファー中に再懸濁した。RNAは、RNA-Micro-plus kit(QIAGEN社)を用いて調製した。RNAライブラリーを、KAPA Stranded RNA-seq Kit with RiboErase(HMR)(KAPA BIOSYSTEMS社)を用いて作製した。HiSeq 2500(illumina社)でのペアエンドシーケンシングには、迅速ラン化学(リード長:100bp)を用いた。101塩基対のペアエンドリードをアライメントし、RSEM(v1.2.11)をBowtie2と共に用いて定量化した。リードを、ヒトトランスクリプトームに対してアライメントし、カウント値はlog2変換および分位数(quantile)正規化されている。
ヒト皮膚由来のVδ1 T細胞を、PMAおよびイオノマイシンまたはプレート結合型抗CD3 mAb(OKT3、5μg/mL)を用いて24時間刺激した。その後に、上清を取得し、ProcartaPlex Human Cytokine & Chemokine Panel 1A(34 plex)(eBioscience社)を用いて分析した。アッセイは、Luminex FlexMap3D(Luminex社)を用いて分析した。データを、Microsoft Excelで分析し、3名のドナー(二重反復実験)の平均を示す。エラーバーは、標準偏差を示す。
各グリッド培養設定に関して、本発明者らは、外科用メスを用いて数箇所を擦った2枚のペトリ皿(100× 25mm、Corning社)を準備した。切り刻んだ皮膚片を、擦り跡に配置した。空気中で5~10分間乾燥させた後、皮膚片をディッシュに正常に貼り付かせ、10mLのSkin-T培地を加えた。培地を週1回交換し、3週間の生育後に、ACCUTASE(登録商標)(LifeTechnologies社)を用いた処理に続いて、初代線維芽細胞を回収した。線維芽細胞を1× 104で48ウェルプレートに、またはトランスウェル実験の場合には2×104で24ウェルプレートのボトムチャンバー中に播種した。2~3日間後、線維芽細胞はコンフルエンスに達し、RPMIおよび記載されたサイトカインを用いて、48ウェルプレートの場合には2×105個の混合型皮膚リンパ球、または24ウェルプレート、ボトムウェルならびにトランスウェルの場合には3×105個のリンパ球を加えて、共培養実験を開始させた。
PBMC内の血液由来γδ T細胞は、TCRリガンド(Vδ 2の場合、例えば、IPP、HMBPP、ビスホスホネート)またはTCR受容体(mAb)もしくはTCR関連キナーゼCD3を架橋するための抗体添加を用いて刺激された場合にのみ、増幅することができる。TCR架橋の同じ効果は、PHAなどのレクチンを用いても達成することができる。そのようなTCR刺激物質の添加の非存在下では、PBMC中のγδ T細胞は、数日間は生存するが、増幅できず、わずかな多様性を有するT細胞サブセットの当初の組成にとどまる。
三次元皮膚外植片プロトコールを、Clarkプロトコールを利用して構築した。9mm×9mm×1.5mmの寸法のCellfoamマトリックス(Cytomatrix Pty Ltd, Victoria, Australia)または同等品をオートクレーブし、続いて100mg/mLラット尾I型コラーゲン(BD Biosciences社)溶液(PBS中)中で室温にて30分間インキュベートし、その後、PBS中で1回リンスした。成人ヒト皮膚のサンプルは、皮膚手術から3~6時間以内に取得した。皮下脂肪を除去し、残りの皮膚組織を、約1mm×1mmの大きさの断片へと切り刻んだ。約5個の皮膚断片/外植片を配置し、各マトリックスの表面へと押し当てた。各マトリックスを、2mLの「Skin-T」培地(10% 加熱不活性化胎児ウシ血清(Life Technologies社)、L-グルタミン(292μg/mL;Life Technologies社)、ペニシリン(100単位/mL;Life Technologies社)、ストレプトマイシン(100μg/mL; Life Technologies社)、N-2-ヒドロキシエチルピペラジン-N-2-エタンスルホン酸(HEPES; 0.01M;Life Technologies社)、ピルビン酸ナトリウム(1mM;Life Technologies社)、最小必須培地(MEM)非必須アミノ酸(1×;Life Technologies社)および3.5μL/L 2-メルカプトエタノール(Life Technologies社)を含むイスコフ改変ダルベッコ培地(IMDM; Life Technologies社)が入った24ウェルプレート(Corning社)の別々のウェル中に配置した。培養の最初の7日間、アンホテリシン(2.5μg/mL;Life Technologies社)を培地に加えた。上部の1mLの培地を各ウェルから吸引除去し、新鮮な培地と置き換えることにより、培地を週3回交換した。ヒト組み換えIL-2(PROLEUKIN(登録商標);Novartis Pharmaceutical UK Ltd)100IU/mLと、ヒト組み換えIL-15(Biolegend社)20ng/mLとを、培養初期に加え、表1に示す通り、21~35日後にリンパ球を単離するまでの間、培地に追加した。最大で96ウェル(4枚の24ウェルプレート)を、各ドナーについて培養中に設置した。
ヒトγδ T細胞は皮膚に豊富に存在し、大部分がVδ2-であり、ヒトリンパ系ストレス監視応答に従事する
図1A~1Dは、ヒトの皮膚が常在性γδT細胞の顕著な集団を含むことを示す。Clarkプロトコールを用いて、本発明者らは、IL-2およびIL-15を添加したヒト余剰皮膚サンプルを用いることで、3週間の期間にわたって、組織常在性リンパ球を増殖した。平均収量は、1スキャホールド当たり240,000リンパ球であった。以前の報告と合致して、本発明者らは、大部分が組織常在性「TRM」タイプの、従来のα β TCRを発現する大多数の細胞と共に、明らかに異なる皮膚常在性リンパ球サブセットを特定することができた。全体では、CD45+細胞のうちの59.9%(±8.6%)はCD4+であり、かつ18.3%(±2.8%)がCD8+ αβ T細胞であり、8.7%(±3.6%)のNK細胞画分が含まれた。加えて、本発明者らは、γδ T細胞の相当な集団(CD45+細胞のうちの平均8.513%、±6.564%)を、本発明者らのドナーで見出した(図1Aおよび図3D)。このリンパ球の特性は、器官型培養後、約100名のドナーで高度に再現可能であり、かつ新鮮消化皮膚サンプルと同等であり、γ δ 集団のわずかな増大でのみ異なったが、実用的に有用であり、標準的な組織消化プロトコールと比較して、はるかに大きくかつ純粋なリンパ球集団を提供した。それらのTCRデルタ鎖に基づくヒトγδ T細胞の組織限局化に関する文献と合致して、大多数のヒト皮膚γδ T細胞は、フローサイトメトリーにより特定される、種々のγ 鎖と対になったVδ1 TCR鎖を発現した。このことは、Vγ 9に連結されたVδ 2鎖の単一特異的TCRヘテロ二量体を示し、ヒト皮膚サンプル中にはほとんど存在しなかった、末梢血γδ T細胞のうちの大部分とは対照的であった。しかしながら、Vδ1 TCRまたはVδ2 TCRのいずれも発現しなかったサブセット(本発明では「ダブルネガティブ」γδ T細胞(DN γδ T細胞)として示す)に留意することが重要である(図1C)。
ヒト組織γδ T細胞をさらに研究するために、混合型皮膚リンパ球を細胞培養ウェルへと移し、長時間の生存率を維持するために、IL-2を添加した。興味深いことに、器官型培養中に存在するストロマ細胞および上皮細胞から分離することにより、Vδ1 T細胞が独特に、いかなる刺激も加えることなく、活性化および増殖の兆候を示した。7日間の間に、Vδ 1+ T細胞およびDN γδ T細胞は独特かつ大幅に、核因子Ki-67をアップレギュレーションし、かつIL-2受容体α(CD25)の表面発現を増加させた(図3Bおよび図4B)。際立ったことに、3週間の期間にわたって、およびIL-2のみの存在下で、組織由来Vδ 1+ T細胞およびDN γδ T細胞は、すべての他のT細胞サブセットよりも増殖し、それにより全皮膚リンパ球のうちの最大65%となり、個数は平均で127.18倍まで増加したが、一方で、αβ T細胞は、細胞絶対数により測定した場合に、5.21倍(p= 0.0124)しか増えなかった(図3A)。細胞周期関連核因子Ki-67のMFIは、Vδ 1+ T細胞およびDN γδ T細胞では14日間で2,664.5(±1,876.1)から8,457.7(±4,574.2)まで増加し、一方で、αβ T細胞では、MFIは592.8(±390.5)から284.7(±140.1)へと同じ期間で減少した(図3C)。選択的な皮膚常在性γδ T細胞増殖のこの現象はさらに、追加の組み換えIL-15を用いて補助することができ、組み換えIL-15は、リンパ球の生存率および総数を増加させた。
上記のVδ 1+ T細胞およびDN γδ T細胞の際立った増幅は、多量の線維芽細胞増殖が存在する器官型培養システム中では一切生じなかった。Vδ1+ T細胞およびDN γδ T細胞とのそれらの共培養がT細胞の増幅を阻害するか否かを直接的に調べるために、自家線維芽細胞を生育させた。3週間のスキャホールド培養後、空であるかまたは事前に達成された線維芽細胞のコンフルエントな単層を含有したウェルへと混合型皮膚リンパ球を播種し、それぞれの場合で、培地に外因的にIL-2を添加して、T細胞生育を維持させた。加えて、同じウェル中でTリンパ球が線維芽細胞と直接的に接触することを防止するが、線維芽細胞により分泌される可溶性因子によりT細胞が影響を受けることを可能にする、トランスウェル(transwell)を用いた。14日間の共培養中に、線維芽細胞がないウェル中およびT細胞が線維芽細胞と直接的に接触することを妨げられたウェル中では、Vδ 1+ T細胞およびDN γδ T細胞は増殖し始めた。以前の通り、αβ T細胞増殖は、すべての条件下で低かった。T細胞が線維芽細胞と直接接触させられた場合、Vδ 1+ T細胞およびDN γδ T細胞の2週間にわたる生育率は、線維芽細胞接触のないウェルの22.6(±8.07)倍から3.3(±0.17)倍へと著しく低下した(図4A)。この接触媒介型阻害は、単独で生育させたリンパ球と比較した場合の、7日間の期間にわたるVδ1 T細胞でのCD25、Ki-67および転写因子T-betのアップレギュレーションの非存在によりさらに確認された(図4B)。免疫系の組織媒介型制御の一部の形態が、組織ホメオスタシスの維持に対して基本的なものであることが明らかであろうが、これは、この制御なしでは、永続的な炎症の可能性が生じるであろうからである。間質線維芽細胞によるVδ 1+ T細胞およびDN γδ T細胞の抑制的調節は、そのような制御の一例であると思われる。
混合型皮膚由来リンパ球を14日間増幅させ、γδ T細胞からα β T細胞を除去するために、蛍光結合型細胞ソーティング(fluorescence associated cell sorting)を用い、最大90%の純度を可能とした。それらの非常に富化された細胞を、10% FCSを含有するRPMI培地中に150,000細胞/ウェルの濃度で細胞培養ウェルへと配置した。24時間後に上清を回収し、LUMINEX(登録商標)に基づくアレイを用いて、広範囲のエフェクターサイトカインに関して評価した。完全に予期せぬことに、増幅中のγδ T細胞(線維芽細胞からのその分離のみによって誘発された)は、多量のIFN-γ(12,383.46±16,618.90pg/mL)、GM-CSF(4,316.73±4,534.96pg/mL)などのTH1関連サイトカインならびに炎症促進性ケモカインCCL4(14,877.34±10,935.64pg/mL)およびCCL3(1,303.07± 757.23pg/mL)を自発的に産生した(図5A)。
Vδ1 T細胞受容体を発現する、ヒト結腸由来の非造血組織常在性γδ T細胞の集団も同定された(図6)。3名のドナーで、4~ 5週間の期間にわたって、皮膚細胞に関して用いたのと同じ方法を用いて、これらの細胞を増幅することができた。増幅中、結腸由来Vδ 1+ T細胞およびDN γδ T細胞は、線維芽細胞豊富な器官型細胞培養からのそれらの単離後に、類似のKi-67アップレギュレーションパターンを示した。同様に、結腸常在性Vδ 1+ T細胞およびDN γδ T細胞は、NKG2D受容体に対するリガンドの供給により、強く刺激された。血液由来γδ T細胞は、CD16発現を介して抗体依存性細胞媒介性細胞傷害を実行する能力を十分に備えており、このことは、リツキシマブと組み合わせた場合の、CD20陽性B細胞系統リンパ腫に対して標的化された強化された細胞傷害性を証明する。同様に、慢性リンパ性白血病(CLL)およびHER2陽性乳癌細胞が、モノクローナル抗体を用いて標的化された場合に、より効果的に殺傷される。皮膚由来Vδ1 T細胞が抗体オプソニン化型標的細胞を標的化する能力を評価するために、3種類のIgG1関連Fc受容体CD16、CD32およびCD64の発現レベルを定量した。皮膚由来Vδ1 T細胞は、わずかなレベルのFc受容体CD16を発現するが、高親和性IgG受容体CD64に関して良好な発現レベルを示す。したがって、組織由来Vδ1 T細胞は、それらが抗体により悪性腫瘍および転移の側に誘導され、オプソニン化された腫瘍細胞を認識し、かつADCCを介して標的を殺傷するであろうから、CD20療法またはHer2療法などのモノクローナル抗体療法に対するアジュバントとして用いられる能力を十分に備えている可能性がある。
皮膚由来γδ T細胞の増幅
3~4週間のスキャホールド培養の後、混合リンパ球を取り出し、PBSで洗浄し、遠心分離し、ろ過済みの熱で不活性化したウシ胎児血清(Life Technologies社)10%、L-グルタミン(292μg/mL;Life Technologies社)、ペニシリン(100単位/mL;Life Technologies社)、ストレプトマイシン(100μg/mL;Life Technologies社)、N-2-ヒドロキシエチルピペラジンN-2-エタン硫酸(HEPES; 0.01M;Life Technologies社)、ピルビン酸ナトリウム(1 mM; Life Technologies社)、最小必須培地(MEM)非必須アミノ酸溶液(1×; Life Technologies社)、および50μM 2-メルカプトエタノール(Life Technologies社)を有するRoswell Park Memorial Institute 1640培地(RPMI-1640; Life Technologies社)で細胞濃度が1×106細胞/mLとなるように再懸濁した。当初の集団のVδ1+細胞は、リンパ球の1.12%であった。96ウェル平底プレート(Corning社)に2×105細胞/ウェル、または24ウェルプレートに2×106細胞/ウェルを播種し、表2に示す濃度の因子を追加して、増幅させた。
従来、組織または腫瘍に由来するT細胞を製造する際に、複数の血液由来の血清および血漿を培地に添加することは一般的である。しかし、これらの血清または血漿成分の使用は、これらの成分のバッチ毎の差異、これらの成分の高いコスト、先進療法医薬品(advanced therapy medicinal products(ATMP))業界全体の高い需要を考慮した供給の制限、および、これらの成分からの外来性の物質による二次汚染のリスクの増加により、好ましくない状況を生じ得る。そのため、本明細書に記載するように、所望のγδ T細胞の増殖と富化を支持するサイトカインを首尾よく特定した後、そのようなサイトカインの使用により、組織由来γδ T細胞の増殖に通常使用される複数の血清/血漿成分の必要性をなくせるかどうかを試験した。これをさらに試験するため、免疫細胞を皮膚サンプルから放出させ、γδ T細胞+/-血清の増殖と富化を支持するために、血漿/血清が依然として必要かどうかを試験した。そのため、放出された細胞を取り出し、2つの培地に「0日目」に播種した。第一の培地は、10%のヒト由来血清およびサイトカインを加えた、動物由来因子非含有(animal derived component free (ADCF))培地(TexMACS, Miltenyi社)を含むものとした。第二の培地は、同様のADCF培地/サイトカインの混合物、標準定義追加成分(standard defined supplement、CTS(商標)、精製されたヒト血清アルブミン、組み換えインスリンおよびトランスフェリンを含む)を含むが、血清を含まないものとした。この研究の結果を図21に示す。驚くべきことに、血清の有無にかかわらず、同等の富化と同等の増幅倍率がみられた。これは、動物由来の因子またはヒト血清なしで、組織由来のγδ T細胞の増幅および富化が可能であることを示す。
3週間のスキャホールド培養後、上記のClarkプロトコールおよび実施例2で説明した同等の手法を使用して、混合リンパ球を組織から取得した。取得した細胞の特性は、実施例3に記載されたものと同等であった。これらの取得した細胞は、HBSS + HEPESで洗浄し、遠心分離し、(i) IL-2(100IU/L)、IL-4(Biolegend社、5ng/mL)、IL-15(Biolegend社、20ng/ml)およびIL-21(Biolegend社、5ng/ml)に加えて10% ヒト AB 血清(Life Science Production社)を含む、または (ii)IL-2(100IU/L)、IL4(Biolegend社、5ng/mL)、IL-15(Biolegend社、20ng/ml)およびIL-21(Biolegend社、5ng/ml)に加えて5% CTS(商標)(Thermo Fisher Scientific社)を含む、TexMACS培地(Miltenyi社)に再懸濁した。血清を含む培地および血清を含まない培地の両方に、ペニシリン/ストレプトマイシン抗生物質(それぞれ100単位/mL、100μg/mL、Life Technologies)、も加えた。次いで、細胞を、24ウェルプレート(Corning社)に2百万細胞/ウェルとなるように播種した。細胞がコンフルエント化したら、同じ培地を含む新しいウェルに1/2~1/4の間で分割して、増幅/継代した。21日目に、ACCUTASE(登録商標)細胞分離手段(Thermo-Fisher社)を用いて細胞を取り出し、フローサイトメトリーで分析して、図21および図22A~22Dに示されるように最終的な細胞特性を決定した。
図23に示す通り、TIGITは構成的に消化管常在性Vδ1細胞上に発現する。従来の細胞消化により消化管から単離した、Vδ1細胞を使用してデータを構築した。組織常在性γδ T細胞の構成的なTIGITの発現は、皮膚由来のVδ1細胞にのみ関連するわけではなく、TIGITの発現は、Clarkプロトコールのアーティファクト(合成物)でははない(グリッドに基づいた単離手順)。
3人のドナー(TS052、TS056およびSK073)からの皮膚組織を9mmグリッドに配置し、IL-2およびIL-15を加えた培地で3週間培養した。単離されたリンパ球を、IL-2、IL-4、IL-15およびIL-21(図26Aおよび26B、左のバー)またはIL-4、IL-9、IL-15およびIL-21(図26Aおよび26B、右のバー)を加えた培地で培養した。3週間の増幅後のγδ T細胞/グリッド(図26A)およびVδ1細胞/グリッド(図26B)の最終収量を計算した。
3週間のスキャホールド培養の後、混合リンパ球を取り出し、リン酸バッファー生理食塩水(PBS)で洗浄し、遠心分離し、ろ過済みの熱で不活性化したウシ胎児血清(Life Technologies社)10%、L-グルタミン(292μg/mL;Life Technologies社)、ペニシリン(100単位/mL; Life Technologies社)、ストレプトマイシン(100 μg/ml; Life Technologies社)、N-2-ヒドロキシエチルピペラジンN-2-エタン硫酸(HEPES; 0.01M; Life Technologies社)、ピルビン酸ナトリウム(1mM; Life Technologies社)、最小必須培地(MEM)非必須アミノ酸溶液(1×; Life Technologies社)、および50mM 2-メルカプトエタノール(Life Technologies社)を有するRPMI-1640で細胞濃度が1×106細胞/mLとなるように再懸濁した。24ウェルプレート(Corning社)に2×106細胞/ウェルを播種し、サイトカインを下記の最終濃度となるように加えて増幅させた:IL-2: 100 U/mL、IL-4: 5 ng/mL、IL-9: 10 ng/mL、IL-15: 20 ng/mLおよびIL-21: 10 ng/mL。
本明細書で言及されるすべての刊行物、特許、および特許出願は、それぞれの独立した刊行物または特許出願が参照により組み込まれることが具体的かつ個別に示されるのと同じ程度に、参照により組み込まれる。
本発明は以下の実施形態を包含する。
(1)以下のステップ:
(i) 非造血組織から得られたγδ T細胞の集団を提供するステップ;
(ii) 前記γδ T細胞を、効果的な量の
(a) IL-2またはIL-9;
(b) IL-15;および
(c) IL-21
の存在下で少なくとも5日間培養して増幅されたγδ T細胞の集団を生成するステップ;
を含む、γδ T細胞を増幅させる方法。
(2)ステップ(ii)が、さらにIL-4の存在下でのγδ T細胞の培養を含む、(1)に記載の方法。
(3)以下のステップ:
(i) 非造血組織から得られたγδ T細胞の集団を提供するステップ;
(ii) 前記γδ T細胞を、IL-2と、IL-15と、IL-21、ストロマ細胞由来因子(SDF)、IL-1β、IL-12、IL-18およびIL-33からなる群より選択される少なくとも1つの因子と、の存在下で少なくとも5日間培養して、増幅されたγδ T細胞の集団を生成するステップ; を含む、γδ T細胞を増幅させる方法。
(4)ステップ(ii)が、外因性TCR経路アゴニストの不存在下でのγδ T細胞の培養を含む、(1)~(3)のいずれか1項に記載の方法。
(5)ステップ(ii)が、無血清培地でのγδ T細胞の培養を含む、(1)~(4)のいずれか1項に記載の方法。
(6)ステップ(i)の後、分離されたγδ T細胞の集団を生成するために、前記γδ T細胞を非造血細胞から分離する工程をさらに含み、かつ、
ステップ(ii)が以下を含む:
(a) 実質的にストロマ細胞との接触がない、γδ T細胞の培養;
(b) 実質的に腫瘍細胞との接触がない、γδ T細胞の培養;および/または
(c) 実質的に支持細胞との接触がない、γδ T細胞の培養;
(1)~(5)のいずれか1項に記載の方法。
(7)以下のステップ:
(i) 非造血細胞およびγδ T細胞を含む非造血組織を提供するステップ;
(ii) γδ T細胞を非造血細胞から分離して、分離されたγδ T細胞の集団を得るステップ;
(iii) 前記γδ T細胞を、IL-2と、IL-15と、IL-21、SDF、IL-1β、IL-12、IL-18およびIL-33からなる群より選択される因子と、の存在下で少なくとも5日間培養して、増幅されたγδ T細胞の集団を生成するステップ;
を含む、γδ T細胞を増幅させる方法。
(8)前記非造血組織は、ヒトまたは非ヒト動物の対象から得られたものである、(7)に記載の方法。
(9)ステップ(ii)が、IL-2、IL-15およびIL-21の存在下でのγδ T細胞の培養を含む、(7)または(8)に記載の方法。
(10)ステップ(ii)が無血清培地でのγδ T細胞の培養を含む、(7)~(9)のいずれか1項に記載の方法。
(11)ステップ(iii)が、実質的にストロマ細胞と前記γδ T細胞との接触がない条件下でのγδ T細胞の培養を含む、(7)~(10)のいずれか1項に記載の方法。
(12)ステップ(iii)が、外因性TCR経路アゴニストが存在しない条件下でのγδ T細胞の培養を含む、(7)~(11)のいずれか1項に記載の方法。
(13)非造血細胞からγδ T細胞を分離するステップが、非造血組織から細胞を放出させるように構築された合成スキャホールド上でのγδ T細胞の培養を含む、(6)~(12)のいずれか1項に記載の方法。
(14)非造血細胞からγδ T細胞を分離するステップが、IL-2および/またはIL-15の存在下でのγδ T細胞と非造血細胞との培養を含む、(6)~(13)のいずれか1項に記載の方法。
(15)分離されたリンパ球の集団が分離されたγδ T細胞の集団を含み、前記分離されたγδ T細胞の集団が分離されたVδ1 T細胞の集団を含む、(6)~(14)のいずれか1項に記載の方法。
(16)前記分離されたリンパ球の集団の1~10%が、増幅前のγδ T細胞である、(15)に記載の方法。
(17)前記分離されたリンパ球の集団の1~10%が、増幅前のVδ1 T細胞である、(15)または(16)に記載の方法。
(18)前記分離されたγδ T細胞の集団の少なくとも80%が、増幅前のVδ1 T細胞である、(6)~(17)のいずれか1項に記載の方法。
(19)前記分離されたγδ T細胞の集団の10%未満が、増幅前のVδ2 T細胞である、(6)~(18)のいずれか1項に記載の方法。
(20)αβ T細胞および/またはNK細胞は、前記分離されたγδ T細胞の集団から除去される、(6)~(19)のいずれか1項に記載の方法。
(21)増幅前に、前記分離されたγδ T細胞の集団が、少なくとも10%のCCR3+細胞、少なくとも10%のCCR4+細胞、少なくとも10%のCCR7+細胞、少なくとも10%のCCR8+細胞、または少なくとも10%のCD103+細胞を含む、(6)~(20)のいずれか1項に記載の方法。
(22)増幅前に、前記分離されたγδ T細胞の集団が、参照となる血液常在性Vδ2 T細胞の集団と比較して、高頻度のCCR3+細胞、CCR4+細胞、CCR7+細胞および/またはCCR8+細胞を含む、(6)~(21)のいずれか1項に記載の方法。
(23)前記分離されたVδ1 T細胞の集団が、参照となる血液常在性Vδ1 T細胞の集団と比較して、高頻度のNKG2D+細胞、CD56+細胞、CD69+細胞および/またはTIM3+細胞を含む、(14)~(22)のいずれか1項に記載の方法。
(24)14日間以内の培養で、増幅されたγδ T細胞の集団が、増幅前の分離されたγδ T細胞の集団と比較して、少なくとも20倍の数のγδ T細胞を含む、(1)~(23)のいずれか1項に記載の方法。
(25)21日間以内の培養で、増幅されたγδ T細胞の集団が、増幅前の分離されたγδ T細胞の集団と比較して、少なくとも50倍の数のγδ T細胞を含む、(1)~(24)のいずれか1項に記載の方法。
(26)前記増幅されたγδ T細胞の集団が、増幅されたVδ1 T細胞の集団を含む、(1)~(25)のいずれか1項に記載の方法。
(27)14日間以内の培養で、増幅されたVδ1 T細胞の集団が、増幅前の分離されたVδ1 T細胞の集団と比較して、少なくとも20倍の数のVδ1 T細胞を含む、(26)に記載の方法。
(28)21日間以内の培養で、増幅されたVδ1 T細胞の集団が、増幅前の分離されたVδ1 T細胞の集団と比較して、少なくとも50倍の数のVδ1 T細胞を含む、(26)または(27)に記載の方法。
(29)増幅されたγδ T細胞の集団がCD27を発現する、(1)~(28)のいずれか1項に記載の方法。
(30)増幅されたγδ T細胞の集団が、分離されたγδ T細胞の集団よりも、高いCD27発現量の中央値を示す、(29)に記載の方法。
(31)増幅されたγδ T細胞の集団が、分離されたγδ T細胞の集団と比較して、少なくとも2倍のCD27発現量の中央値を示す、(30)に記載の方法。
