Abstract
Description
本明細書には、金属-有機骨格体(metal-organic framework,MOF)を有効成分として含む傷治癒用組成物および前記金属-有機骨格体と親水性高吸水性高分子とを含む創傷被覆材が開示される。
本研究は、中小ベンチャー企業部の技術開発事業[S2737226]の支援による研究である。
皮膚は、表皮、真皮、皮下脂肪など大きく3つの層で構成されていて、各層の皮膚組織は、皮膚の色々な機能を担当する多様な細胞とこれを取り囲む物質で構成されている。皮膚が損傷を受けた場合、損傷部位が細菌と外部の毒素により侵入を受けることになり、これに対する皮膚の防御機能として炎症が誘発されることがある。

〔発明の概要〕
〔発明が解決しようとする課題〕
一態様において、本発明の目的は、金属-有機骨格体(metal-organic framework,MOF)を有効成分として含む傷治癒用組成物を提供することにある。
一態様において、本発明は、金属-有機骨格体(metal-organic framework,MOF)を有効成分として含む傷治癒用組成物を提供する。
本発明は、金属-有機骨格体(MOF)を含む組成物に関し、傷治癒能、具体的に線維芽細胞の移動を促進させて創傷を治癒する優れた効果があり、細胞毒性において対照群と有意差がないため、創傷部位への適用時に優れた安全性を有し、前記金属-有機骨格体と親水性高吸水性高分子とを含む創傷被覆材の場合、時間が経過するにつれて目視で確認できるほどに優れた創傷回復率を有する効果がある。
図1は、本発明の一実施例による、金属-有機骨格体が処理された線維芽細胞の細胞毒性実験結果を示すグラフである。x軸は、24、48、72時間の時間帯別に処理された金属-有機骨格体の濃度(対照群(Control)、0.1、1および10μg/ml)を、y軸は、線維芽細胞の生存率を示す。
以下、本発明を詳細に説明する。
創傷治癒用組成物および創傷被覆材の有効成分として含まれる金属-有機骨格体(metal-organic framework,MOF)としてMOF-808を製造するために、下記のような過程を行った。
前記実施例1で製造された金属-有機骨格体であるMOF-808を含む創傷被覆材を製造するために、下記のような過程を行った。
前記実施例1で製造された金属-有機骨格体であるMOF-808の細胞に及ぼす毒性程度と傷治癒用組成物の有効成分としての適合性を確認するために、下記のような実験を行った。実験方法は、ISO 10993 biological evaluation of medical devices-Part 5:test for in vitro cytotoxicity,test on extractsに提示された方法を活用し、この際に使用された細胞株は、マウス線維芽細胞(NIH3T3,ATCC)である。
前記実施例1で製造された金属-有機骨格体であるMOF-808の傷治癒能の評価のために、生体外スクラッチ傷分析(in vitro scratch wound assay)実験を下記のように行った。
前記実施例1で製造された金属-有機骨格体であるMOF-808の動物モデルでの傷治癒能の評価および創傷被覆材としての効能を確認するために、前記実施例2で製造された創傷被覆材、すなわちMOF-808を含む創傷被覆材を用いて下記のような動物実験を行った。本動物実験は、4週齢の雌(240g~270g)Sprague-Dawley種の白鼠を動物モデルとして利用した。
Claims (16)
- 金属-有機骨格体(metal-organic framework,MOF)を有効成分として含む傷治癒用組成物。
- 前記金属-有機骨格体は、AlおよびZrからなる群から選ばれる1種以上の金属イオンと、4,4’-ビフェニルジカルボン酸(4,4’-biphenyldicarboxilic acid)、ベンゼン-1,4-ジカルボン酸(benzene-1,4-dicarboxylic acid)、9,10-アントラセンジカルボン酸(9,10-anthracenedicarboxylic acid)、ビフェニル-3,3,5,5’-テトラカルボン酸(biphenyl-3,3,5,5’-tetracarboxylic acid)、ビフェニル-3,4’,5-トリカルボン酸(biphenyl-3,4’,5-tricarboxylic acid)、5-ブロモイソフタル酸(5-bromoisophthalic acid)、5-シアノ-1,3-ベンゼンジカルボン酸(5-cyano-1,3-benzenedicarboxylic acid)、2,2-ジアミノ-4,4’-スチルベンジカルボン酸(2,2-diamino-4,4’-stilbenedicarboxylic acid)、2,5-ジアミノテレフタル酸(2,5-diaminoterephthalic acid)、1,1,2,2-テトラ(4-カルボキシルフェニル)エチレン(1,1,2,2-tetra(4-carboxylphenyl)ethylene)、2,5-ジヒドロキシテレフタル酸(2,5-dihydroxyterephthalic acid)、2,2-ジニトロ-4,4-スチルベンジカルボン酸(2,2-dinitro-4,4-stilbenedicarboxylic acid)、5-エチニル-1,3-ベンゼンジカルボン酸(5-ethynyl-1,3-benzenedicarboxylic acid)、2-ヒドロキシテレフタル酸(2-hydroxyterephthalic acid)、2,6-ナフタレンジカルボン酸(2,6-naphthalenedicarboxylic acid)、1,2,4,5-テトラキス(4-カルボキシフェニル)ベンゼン(1,2,4,5-tetrakis(4-carboxyphenyl)benzene)、4,4,4’’-s-トリアジン-2,4,6-トリイル-三安息香酸(4,4,4’’-s-triazine-2,4,6-triyl-tribenzoic acid)、1,3,5-トリカルボキシベンゼン(1,3,5-tricarboxybenzene)、1,4,7,10-テトラアザシクロドデカン-N,N’,N’’,N’’’-テトラ酢酸(1,4,7,10-tetraazacyclododecane-N,N’,N’’,N’’’-tetraacetic acid)、1,3,5-トリス(4-カルボキシ[1,1’-ビフェニル]-4-イル)ベンゼン(1,3,5-tris(4-carboxy[1,1’-ビフェニル]-4-yl)benzene)、1,3,5-トリス(4-カルボキシフェニル)ベンゼン(1,3,5-tris(4-carboxyphenyl)benzene)、1,3,5-トリスカルボキシフェニルエチニルベンゼン(1,3,5-triscarboxyphenylethynylbenzene)からなる群から選ばれる1種以上の有機リガンドを含む、請求項1に記載の傷治癒用組成物。
- 前記金属-有機骨格体は、MOF-808(Zr6O4(OH)4(BTC)2(HCOO)6)である、請求項1に記載の傷治癒用組成物。
- 前記金属-有機骨格体は、皮膚細胞移動を促進させる、請求項1に記載の傷治癒用組成物。
- 前記金属-有機骨格体は、0.001~10μg/mlである、請求項1に記載の傷治癒用組成物。
- 前記金属-有機骨格体は、組成物の総重量を基準として0.01~10重量%で含まれる、請求項1に記載の傷治癒用組成物。
- 前記傷は、創傷、非治癒外傷性傷、放射線照射による組織の破壊、擦過傷、裂傷、引き抜き損傷、貫通傷、銃傷、切創、火傷、凍傷、皮膚潰瘍、皮膚乾燥、皮膚角化症、割れ、破裂、皮膚炎、手術の傷または血管疾患の傷、打撲傷、角膜創傷、床擦れ、臥瘡、慢性潰瘍、手術後の縫合部位、脊椎傷害性傷、婦人科的傷、化学損傷およびにきびからなる群から選ばれるいずれか一つ以上である、請求項1に記載の傷治癒用組成物。
- 前記組成物は、皮膚外用剤組成物である、請求項1に記載の傷治癒用組成物。
- 前記組成物は、薬学的組成物である、請求項1に記載の傷治癒用組成物。
- 前記組成物は、化粧料組成物である、請求項1に記載の傷治癒用組成物。
- 前記組成物は、医薬部外品組成物である、請求項1に記載の傷治癒用組成物。
- 金属-有機骨格体(metal-organic framework,MOF)と親水性高吸水性高分子とを含む創傷被覆材。

- 前記金属-有機骨格体は、MOF-808(Zr6O4(OH)4(BTC)2(HCOO)6)である、請求項12に記載の創傷被覆材。
- 前記親水性高吸水性高分子は、ヒドロゲル(hydrogel)である、請求項12に記載の創傷被覆材。
- 前記金属-有機骨格体は、親水性高吸水性高分子の総重量を基準として0.01~10重量%で含まれる、請求項12に記載の創傷被覆材。
