最初に、Glu20からLeu455(574aa)までの残基をコードするマウスELTD1遺伝子の細胞外ドメインを、bEnd3(マウス脳内皮ポリオーマミドルT抗原形質転換細胞)由来のRNAを使用した逆転写ポリメラーゼ連鎖反応(RT-PCR)によって増幅することに成功した。ヒトELTDl遺伝子の細胞外ドメインを、HUVECから調製されたRNAを使用したRT-PCRによって増幅した。これら2つの遺伝子の配列を、サンガーシーケンシングによって確認した。本発明者らは、これらの2つの遺伝子を哺乳動物発現ベクターにクローン化して、ELTDl断片Glu20-Leu455(574aa)(Favara et al.,“A review of ELTD1,a pro-angiogenic adhesion GPCR”,Biochem Soc Trans(2014)42(6):1658-1664)及びCK(ヒト免疫グロブリンκ軽鎖定常ドメイン)と融合したヒトELTD1断片のマウス組換えタンパク質を発現させることに成功した。ヒト胎児腎臓(HEK)293F過剰発現系にトランスフェクトした後、組換えタンパク質を、KappaSelect樹脂(GE Healthcare)を使用した親和性クロマトグラフィーによって精製した。
CD133陰性細胞の両方の表面上に見られ、GBMでより効率的なGSCマーカーとして機能する可能性がある[55、56]。さらに、ネスチンは、GBM及び他のがんの増殖、移動、生存において重要な役割を果たすことが示されている[57~59]。興味深いことに、抗ELTD1処理によって下方制御された遺伝子からの遺伝子-遺伝子間のクラスター分析は、ネスチン関連経路の下方制御応答があることを示唆する。
本実施例では、抗ELTD1一本鎖可変断片(scFv)を生成し、G55神経膠腫異種移植前臨床モデルにおいてELTD1の430AA外部領域の結合を維持することを示した。本発明者らは、形態学的MRIを使用して腫瘍体積を評価し、灌流イメージングを使用して、抗ELTD1 mAb処理と比較した抗ELTD1 scFv処理後の腫瘍領域内の血管変化を測定した。動物の生存も、抗ELTD1 scFv処理または抗ELTD1 mAb処理後に決定した。mt-MRイメージングを使用することにより、本発明者らは、プローブとしての抗ELTD1 scFv断片の結合親和性及び特異性も評価した。

- 前記モノクローナル抗体またはその抗体断片が、in vitroで、増殖を阻害するか、細胞死を開始させるか、またはその両方を行う、請求項71~81のいずれか一項に記載の二機能性モノクローナル抗体。
- 前記抗体が、がん細胞に結合する場合に:ナトリウムチャネルタンパク質5型サブユニットα、またはL1細胞接着分子の少なくとも一方の発現を低下させる、請求項71~81のいずれか一項に記載の二機能性モノクローナル抗体。
- 前記抗体が、がん細胞に結合する場合に、ELTD1を発現するすべての腫瘍において:ADA、SCN5A、L1CAM、BMP2、アルカリホスファターゼ(ALPL)、TRPM8の少なくとも1つの発現を増加させる、請求項71~81のいずれか一項に記載の二機能性モノクローナル抗体。
- 前記抗体が、がん細胞に結合する場合に、SELENBP1の発現を減少させる、請求項71~81のいずれか一項に記載の二機能性モノクローナル抗体。
- 前記抗体が、肝細胞癌に結合する場合に:VWA1を減少させる、請求項71~81のいずれか一項に記載の二機能性モノクローナル抗体。
- 前記抗体が、肺癌細胞に結合する場合に、以下の少なくとも1つの発現、すなわち:SCUBE3を減少させるか、PLCH1を増加させるか、CHRNA1を増加させるか、またはCDH2を増加させる、請求項71~81のいずれか一項に記載の二機能性モノクローナル抗体。

- 前記抗体が、網膜細胞に結合する場合に:アデノシンデアミナーゼまたはアペリンの少なくとも一方の発現を減少させる、請求項71~81のいずれか一項に記載の二機能性モノクローナル抗体。
- 前記抗体が、網膜細胞に結合する場合に:骨γカルボキシグルタミン酸タンパク質、マトリリン2、またはフォンヴィルブランド因子Aドメイン含有1の発現を増加させる、請求項71~81のいずれか一項に記載の二機能性モノクローナル抗体。
- 前記抗体を、多発性硬化症の対象に提供する場合に:ナトリウムチャネルタンパク質5型サブユニットα、アデノシンデアミナーゼ、アペリン、spinsterホモログ2、または骨形成タンパク質2の少なくとも1つの発現を減少させる、請求項71~81のいずれか一項に記載の二機能性モノクローナル抗体。
- 前記抗体を、多発性硬化症の対象に提供する場合に:CD74またはIKAROSファミリージンクフィンガー1(Ikaros)の少なくとも一方の発現を増加させる、請求項71~81のいずれか一項に記載の二機能性モノクローナル抗体。
- 前記抗体を、組織再生が必要な部位で対象に提供する場合に:骨γ-カルボキシグルタメート(gla)タンパク質、マトリリン2、またはフォンヴィルブランド因子Aドメイン含有1の少なくとも1つの発現を増加させる、請求項71~81のいずれか一項に記載の二機能性モノクローナル抗体。
- 前記抗体を、転移性結腸直腸癌、第一選択(first-line)非扁平上皮非小細胞肺癌、再発性多形性膠芽腫、転移性腎細胞癌、持続性、再発性、または転移性子宮頸癌、及び上皮性卵巣癌、卵管癌、または原発性腹膜癌から選択されるがんを治療するのに十分な量で提供する、請求項71~81のいずれか一項に記載の二機能性モノクローナル抗体。
- 抗EGF、ラトロフィリン及び7回膜貫通ドメイン含有タンパク質1(ELTD1)抗体またはその断片による、対象における疾患または病態の治療方法であって:
前記疾患または病態を治療する必要がある対象を特定し;そして
前記疾患または病態に由来する症状を軽減するか、または前記疾患または病態を治療するのに十分な、有効量のELTD1特異的結合抗体またはその結合断片を提供することを含み、前記疾患または病態が、多発性硬化症、網膜症、がん、または組織再生から選択される、前記治療方法。 - 前記抗体が、ポリクローナル抗体もしくはモノクローナル抗体、またはその断片である、請求項97に記載の方法。
- 前記抗体が、組換え二価scFv Fc融合抗体である、請求項97または98に記載の方法。
- 前記モノクローナル抗体またはその結合断片が、組換えscFv(一本鎖可変領域断片)抗体、Fab断片、F(ab’)2断片、またはFv断片である、請求項97または98に記載の方法。
- 前記モノクローナル抗体または抗体断片が、キメラ、ヒト化、完全ヒト、または二重特異性である、請求項97または100に記載の方法。
