Abstract
Description
組換え酵素を製造するために、実施例1で製造した各形質転換微生物をLB液体培地5mlを含む培養チューブに接種し、600nmで吸光度が2.0になるまで37℃の振盪インキュベーターで種菌培養をした。本種菌培養された培養液をLB液体培地を含む培養フラスコに接種して本培養を進行した。600nmでの吸光度が2.0になる時1mM IPTGを添加して組換え酵素の発現生産を誘導した。上記培養過程中の攪拌速度は180rpmであり、培養温度は37℃が維持されるようにした。培養液は8,000×gで4℃で20分間遠心分離後に菌体を回収した。回収された菌体は50mM Tris-HC1(pH8.0)緩衝溶液で2回洗浄し、同一の緩衝溶液で懸濁後に超音波細胞破砕機を用いて細胞を破砕した。細胞破砕物は13,000×gで4℃で20分間遠心分離後に上澄液のみを取った。上記上澄液からHis-tag親和クロマトグラフィを用いて組換え酵素を精製し、50mM Tris-HC1(pH8.0)緩衝溶液で透析した後、反応に用いた。
果糖-6-リン酸からプシコース-6-リン酸を製造した後、プシコース-6-リン酸の脱リン酸化酵素よるプシコース生産活性を確認またはブドウ糖-1-リン酸を最初基質として3個の酵素を追加で入れてプシコースを検出して活性測定した。ここで、酵素3個はブドウ糖-1-リン酸をブドウ糖-6-リン酸(glucose-6-phosphate)に転換するホスホグルコムターゼ(phosphoglucomutase, EC 2.7.5.1)またはホスホマンノムターゼ(phosphomannomutase, EC 5.4.2.8)とブドウ糖-6-リン酸を果糖-6-リン酸(fructose-6-phosphate)に転換するブドウ糖-6-リン酸異性化酵素(glucose-6-phosphate isomerase, EC 5.3.1.9)そして果糖-6-リン酸からプシコース-6-リン酸に転換する酵素であるリブロース-5-リン酸-3-エピマー化酵素またはプシコース-6-リン酸-3-エピマー化酵素を本出願の脱リン酸酵素222種それぞれと混合して生成される一般糖であるブドウ糖、果糖、プシコースを定性及び定量評価して確認した。
B=高活性(リン酸糖から単糖への脱リン酸転換率が30%以上)
C=無活性(リン酸糖から単糖への脱リン酸転換率が1%未満)
マルトデキストリンからプシコース生産のために及び7酵素を選別して同時に(one-pot)反応させた。7酵素は、それぞれ澱粉を分解してブドウ糖-1-リン酸に転換するグルカンホスホリラーゼと澱粉を脱分枝化するプルラナーゼ、澱粉基質活用度を高める4-アルファーグルカノトランスフェラーゼ、ブドウ糖-1リン酸をブドウ糖-6-リン酸に転換するホスホグルコムターゼ、ブドウ糖-6-リン酸を果糖-6-リン酸に転換するグルコース-6-ホスフェート・イソメラーゼ、果糖-6-リン酸をプシュース-6-リン酸に転換するプシコース-6-ホスフェート-3-エピメラーゼとプシコース-6-リン酸からプシコースを生産することができる配列1~222酵素をそれぞれ準備して0.1unit/mlを用い、1~5mM MgCl2、10~50mMのリン酸ナトリウム(sodium phosphate pH 7.0)に5%(w/v)マルトデキストリンを添加して温度50℃で12時間反応させた後にHPLCを用いて反応結果物でプシコースをHPLCを用いて分析した。HPLC分析は、Aminex HPX-87C(Bio-RAD社)カラムを用いて80℃で移動相に0.6ml/min流速で流しながら行い、Refractive Index Detectorで検出した。

- 上記酵素は、配列番号1~222のいずれか一つのアミノ酸配列と70%以上の同一性を有するアミノ酸配列を含む、請求項1に記載の酵素。
- 上記酵素は、配列番号1、6、9、12、26、29、38~43、45~53、56、57、59、60、64~66、69、70、72、76、80、81、91~93、95、99~103、113、114、116、117、131、134、136、142、145、146、148、164、167、169、172、177、184~187、189、191、192、211、217、及び221のいずれか一つのアミノ酸配列と70%以上の同一性を有するアミノ酸配列を含む、請求項3に記載の酵素。
- 上記酵素は、プシコース-6-リン酸の選択的活性を示す、請求項1に記載の酵素。
- 請求項1~5のいずれか一項に記載のプシコース-6-リン酸の脱リン酸化酵素をコードする核酸。
- 請求項6に記載の核酸を含む形質転換体。
- 請求項1~5のいずれか一項に記載のプシコース-6-リン酸の脱リン酸化酵素、これを発現する微生物、または上記微生物の培養物を含むプシコース生産用組成物。
- 上記組成物は、リン酸糖をさらに含む、請求項8に記載の組成物。
- 請求項1~5のいずれか一項に記載のプシコース-6-リン酸の脱リン酸化酵素、これを発現する微生物、または上記微生物の培養物をプシコース-6-リン酸と接触させてプシコース-6-リン酸をプシコースに転換する段階を含む、プシコース製造方法。
- 上記方法は、プシコース-6-リン酸をプシコースに転換する段階以前、果糖-6-リン酸(fructose-6-phosphate)に果糖-6-リン酸-3-エピマー化酵素、これを発現する微生物または上記微生物の培養物を接触させ、上記果糖-6-リン酸をプシコース-6-リン酸に転換する段階をさらに含む、請求項10に記載のプシコース製造方法。
- 上記方法は、上記果糖-6-リン酸をプシコース-6-リン酸に転換する段階以前、ブドウ糖-6-リン酸(Glucose-6-phosphate)にブドウ糖-6-リン酸-異性化酵素、これを発現する微生物または上記微生物の培養物を接触させ、上記ブドウ糖-6-リン酸を果糖-6-リン酸に転換する段階をさらに含む、請求項11に記載のプシコース製造方法。
- 上記方法は、上記ブドウ糖-6-リン酸を果糖-6-リン酸に転換する段階以前、ブドウ糖-1-リン酸(Glucose-1-phosphate)にホスホグルコムターゼ、これを発現する微生物または上記微生物の培養物を接触させ、上記ブドウ糖-1-リン酸をブドウ糖-6-リン酸に転換する段階をさらに含む、請求項12に記載のプシコース製造方法。
- 上記方法は、上記ブドウ糖-6-リン酸を果糖-6-リン酸に転換する段階以前、ブドウ糖(Glucose)にポリホスフェートブドウ糖リン酸化酵素、これを発現する微生物または上記微生物の培養物、及びポリホスフェートを接触させ、上記ブドウ糖をブドウ糖-6-リン酸に転換する段階をさらに含む、請求項12に記載のプシコース製造方法。
- 上記方法は、上記ブドウ糖-1-リン酸をブドウ糖-6-リン酸に転換する段階以前、澱粉、マルトデキストリン、スクロースまたはその組合わせにα-グルカンホスホリラーゼ、澱粉ホスホリラーゼ、マルトデキストリンホスホリラーゼまたはスクロースホスホリラーゼ; これを発現する微生物;または上記微生物の培養物、及びホスフェートを接触させ、上記澱粉、マルトデキストリン、スクロースまたはその組合わせをブドウ糖-1 リン酸に転換する段階をさらに含む、請求項13に記載のプシコース製造方法。
- 上記澱粉、マルトデキストリン、スクロースまたはその組合わせをブドウ糖-1-リン酸に転換する段階はα-アミラーゼ、プルラナーゼ、イソアミラーゼ、α-グルカノトランスフェラーゼ、グルコアミラーゼまたはスクラーゼ;これを発現する微生物;または上記微生物の培養物をさらに含み、上記澱粉、マルトデキストリン、スクロースまたはその組合わせをマルトオリゴ糖またはブドウ糖に転換する段階をさらに含む、請求項15に記載のプシコース製造方法。
- 澱粉、マルトデキストリン、スクロースまたはその組合わせ、及びホスフェートに(a)請求項1に記載のプシコース-6-リン酸の脱リン酸化酵素;果糖-6-リン酸-3-エピマー化酵素;ブドウ糖-6-リン酸-異性化酵素; ホスホグルコムターゼまたはブドウ糖リン酸化酵素;及びα-グルカンホスホリラーゼ、澱粉ホスホリラーゼ、マルトデキストリンホスホリラーゼ、スクロースホスホリラーゼ、α-アミラーゼ、プルラナーゼ、イソアミラーゼ、グルコアミラーゼ、α-グルカノトランスフェラーゼ、ポリホスフェートグルコキナーゼまたはスクラーゼ;または(b)上記項目(a)の酵素を発現する微生物または上記微生物の培養物を接触させる段階を含む、プシコース製造方法。
- 上記接触は、pH5.0~9.0、40℃~80℃の温度、及び/又は2時間ないし24時間または120時間実施する、請求項11~17のいずれか一項に記載のプシコース製造方法。
