Abstract
Description
本出願は、2018年8月20日出願の米国仮出願第62/719,760号の優先権を主張するものであり、その全体が内容を参照することにより組み込まれる。
本明細書中で使用される場合、「微生物ベースの組成物」とは、微生物又は他の細胞培養物の成長の結果として生成された成分を含む組成物を意味する。このように、微生物ベースの組成物は、微生物自体及び/又は微生物成長副生成物を含む。微生物は、プランクトン性であっても、バイオフィルム形態であっても、又は両方の混合物であってもよい。成長副生成物は、例えば、代謝産物(例えば、バイオサーファクタント、酵素)、細胞膜成分、タンパク質、及び/又は他の細胞成分であり得る。微生物は、無傷であっても溶解されていてもよい。細胞は組成にないか、又は、例えば、組成物1ミリリットル当たり、少なくとも1×104、1×105、1×106、1×107、1×108、1×109、1×1010、1×1011、1×1012以上のCFUで存在していてもよい。
本発明は、有益な微生物、ならびにバイオサーファクタント、代謝産物、酸、溶剤及び/又は酵素等の1つ又は複数の微生物成長副生成物を含む微生物ベースの組成物を提供する。さらに、本発明は、微生物ベースの組成物を生成するための材料及び方法を提供する。
一実施形態において、組成物は、好ましくは土壌、種、植物全体、又は植物の一部(根、塊茎、茎、花及び葉が挙げられるが、これらに限定されない)への適用のために処方される。特定の実施形態において、組成物は、例えば、液体、粉塵、顆粒、微小顆粒、ペレット、湿潤性粉末、流動性粉末、エマルジョン、マイクロカプセル、油、又はエアロゾルとして処方される。
特定の実施形態において、組成物は、ヒト及び/又は動物の栄養補助食品又は消化補助食品として処方される。
本発明は、微生物を培養し、微生物代謝産物及び/又は成長副生成物を生成するための方法を利用する。より具体的には、本発明は、バイオマス、細胞外代謝産物、残留栄養素及び/又は細胞内成分の生成のための材料及び方法を提供する。
本発明の1つの微生物ベースの生成物は、単に、微生物及び/又は微生物によって生成される微生物代謝産物及び/又は任意の残留栄養素を含有する発酵ブロスである。発酵産物は、抽出又は精製することなく直接使用することができる。所望であれば、抽出及び精製は、文献に記載されている標準的な抽出及び/又は精製方法又は技術を用いて容易に行うことができる。
本発明のシステム及び方法に従って増殖する微生物は、例えば、細菌、酵母及び/又は真菌である。これらの微生物は、天然の微生物、又は遺伝的に改変された微生物であってもよい。例えば、微生物は、特定の特徴を示すために特定の遺伝子で形質転換することができる。微生物はまた、所望の株の突然変異体であってもよい。本明細書中で使用される、「突然変異体」は、参考微生物の株、遺伝的変異体又はサブタイプを意味し、突然変異体は、参考微生物と比較して、1つ以上の遺伝的変異(例えば、点突然変異、ミスセンス突然変異、ナンセンス突然変異、欠失、重複、フレームシフト突然変異又は反復拡大)を有する。突然変異体を作製するための手順は、微生物学的技術分野において周知されえている。例えば、UV突然変異誘発及びニトロソグアニジンは、この目的のために広く使用されている。
特定の実施形態において、本発明は、フィチン酸含有有機物からリン酸塩を遊離させるための、環境に優しく、費用効率の高い材料及び方法を提供する。一実施形態において、本発明の組成物を利用して作物成長及び収量を増大させるために、土壌を肥沃にするための方法も提供される。他の実施形態において、本発明の組成物を利用して、家畜及び/又は養殖魚に給餌するための方法が提供される。さらに他の実施形態において、本発明の組成物を利用するヒト対象の健康を増進するための方法が提供される。
本発明及び多くの利点は、例示としての以下の実施例により、理解されるであろう。以下の実施例は、本発明の方法、用途、実施形態及び変形を例示するものである。本発明を限定するものではない。本発明に関して、数多くの変更や修正を行うことができる。
酵母、例えば、Wickerhamomyces anomalus(Pichia anomala)の培養から生じる酵母発酵生成物は、フィチン酸又はフィチン酸塩含有有機物からリンを遊離させるために有用である。
Wickerhamomyces anomalusを、糖蜜(2.0〜5.0%又は3.0〜4.0%)、尿素(0.1〜1.3%又は0.15〜0.2%)、ペプトン(2.0〜3.0%又は2.2〜2.5%)の増殖培地を用いて、pHを5.0に調整した発酵反応器中で72時間増殖させた。他の微生物を使用する、及び/又は他の増殖培地処方を使用するフィターゼの典型的な微生物生成は、より低い濃度、約300〜500u/mlのフィターゼを生じる。

Claims (28)
- 有機物中に存在するフィチン酸からリン酸塩を遊離する組成物であって、Wickerhamomyces anomalus酵母及び/又は1つ以上の微生物成長副生成物と担体とを含む組成物。
- 前記成長副生成物は、フィターゼである請求項1に記載の組成物。
- 前記酵母の培養から得られる発酵ブロスを含む、請求項1に記載の組成物。
- 前記酵母は、
糖蜜2〜5%、
尿素0.1〜0.3%及び
ペプトン2.0〜3.0%
を含む培養培地中で培養される、請求項1に記載の組成物。 - 1つ以上のバイオサーファクタントをさらに含む、請求項1に記載の組成物。
- 前記1つ以上のバイオサーファクタントは、糖脂質リポペプチド、フラボリピド、リン脂質、脂肪酸エステル化合物、リポタンパク質、リポ多糖−タンパク質複合体、及び/又は多糖−タンパク質−脂肪酸複合体である、請求項5に記載の組成物。
- フィチン酸塩及び/又はフィチン酸の1つ以上の供給源をさらに含む、請求項1に記載の組成物。
- 1つ以上の栄養素及び/又はプレバイオティクスをさらに含む、請求項1に記載の組成物。
- 有機物中のフィチン酸からリンを遊離する方法であって、Wickerhamomyces anomalus酵母及び/又は1つ以上の微生物成長副生成物と担体とを含む組成物を、前記有機物に適用することを含む方法。
- 前記組成物と前記有機物とを混合することをさらに含む、請求項9に記載の方法。
- 前記有機物は、作物残渣、糞、堆肥、庭塵芥、コーヒー粉、おがくず、蒸留穀物、リグニン及び醸造者の使用済み穀物から選択される有機廃棄物である、請求項9に記載の方法。
- 前記有機廃棄物は、農作物畑の表面に広がる作物残渣である、請求項11に記載の方法。
- 前記有機物は、堆肥である、請求項9に記載の方法。
- 前記有機物は、ナッツ、シード、マメ科植物、ジャガイモ及び豆から選択される植物体である、請求項9に記載の方法。
- 前記有機物は、土壌である、請求項9に記載の方法。
- Wickerhamomyces anomalus酵母及び/又は1つ以上の微生物成長副生成物と担体とを含む組成物を土壌に適用して、前記土壌中に存在するフィチン酸からリン酸塩を遊離する、土壌を濃縮する方法。
- 前記組成物で前処理された有機廃棄物が土壌に適用される、請求項16に記載の方法。
- 植物が前記濃縮土壌中で生育されるか、又は生育されることになる、農業又は林業における生産を増進するためにさらに使用される、請求項16に記載の方法。
- 前記植物の成長及び/又は収量を増進するために使用される、請求項18に記載の方法。
- 前記植物におけるリン欠乏を処置及び/又は予防するために使用される、請求項18に記載の方法。
- 動物畜産における生産を増進する方法であって、有効量のWickerhamomyces anomalus酵母及び/又は1つ以上の微生物成長副生成物と、任意で、担体を含む組成物を、動物の飲料水及び/又は飼料に、栄養補助食品及び/又は消化補助食品として適用するか、又は前記組成物を処方することを含む、方法。
- 前記動物畜産は、家畜飼育である、請求項21に記載の方法。
- 前記動物畜産は、水産養殖である、請求項21に記載の方法。
- 前記動物におけるリン欠乏を処置及び/又は予防するために使用される、請求項21に記載の方法。
- ヒトの健康を増進する方法であって、有効量のWickerhamomyces anomalus酵母及び/又は1つ以上の微生物成長副生成物と担体とを含む組成物を、栄養補助食品及び/又は消化補助食品としてヒト対象に投与することを含む、方法。
- 前記組成物は、ヒト対象に経口投与される、請求項25に記載の方法。
- 前記対象におけるリン欠乏を処置及び/又は予防するために使用される、請求項25に記載の方法。
- フィチン酸が、対象の消化管のミネラル吸収を損なうことを防止するために使用される、請求項25に記載の方法。
