腫瘍免疫治療は腫瘍治療分野で近年得られた重大な突破である。近年、免疫チェックポイントPD−1/PD−L1に対する研究は盛んで、最近の1年もない時間で、国内では既に5つのPD−1抗体薬物が市販された。PD−1/PD−L1以外の免疫チェックポイントでは、T細胞免疫グロブリンドメイン及びムチンドメイン3(TIM−3)はPD−1/PD−L1とともにT細胞の活性を調節する免疫チェックポイントの一つである可能性がある。TIM−3はIFN−γを分泌するTh1(Tヘルパー1)CD4+細胞及び細胞傷害性のCD8+T(Tc1)細胞において発現される膜貫通受容体タンパク質である。INF−γを分泌するTh1、Tc1以外、制御性T細胞(Treg)、先天性免疫細胞、樹状細胞DC、ナチュラルキラー細胞NK、単核球monocytesなどにおいても発現される(Anderson A.C.ら,Immunity 44,2016,p989−1004)。TIM−3は複数のリガンドを有し、ガレクチン9(galectin−9)、ホスファチジルセリンphosphatidylserine、HMGB1及びCEACM−1などを含む。TIM−3は、一般的に、ナイーブT細胞において発現されないが、活性化したエフェクターT細胞において上方調節され、体内における免疫性及び耐性の調節作用がある。ほかの免疫チェックポイントと異なるのは、TIM−3は活性化したT細胞、例えばTh1、Tc1において高発現され、共抑制機能に関与し、エフェクターT細胞の活性を抑制し、耐性につながるのみならず、疲弊(exhausted)T細胞においても高発現され、T細胞の機能を抑制する。TIM−3はPD−1抗体によって治療される動物モデルにおいて高発現され、かつTIM−3の抗体と併用すると、顕著に治療効果が向上する(Nigiow SFら、Cancer Res. 71(10):3540−51,2011)。また、最近の研究では、PD−1抗体によって治療される患者に薬剤耐性が現れ、そのCD4+、CD8+細胞においてTIM−3の発現が顕著に向上することが見出され、これは動物モデルで見られた様子と一致する(Koyama Sら Nat Communication. 2016 Feb 17)。そのため、TIM−3抗体とPD−1抗体の併用治療は、PD−1抗体による治療の効率を向上させる方法の一つのみならず、PD−1抗体による治療に薬剤耐性が現れた患者の有効な選択である可能性がある。現在、臨床試験中のTIM−3抗体として、単独で又はPD−1抗体と併用して末期又は転移固形腫瘍を治療するTesaroのTSR−022、及び単独で又はPD−1抗体と併用して末期悪性腫瘍を治療するノバルティス社のMGB−453がある。
本発明の解決しようとする技術的課題は、既存技術では二重特異性抗体におけるTIM−3抗体の部分及びその相応する単独の完全な抗体のヒトTIM−3との結合活性が低く、かつPBMC(NK)細胞を活性化させて腫瘍細胞を殺傷する活性が低いなどの欠陥を克服し、二重特異性抗体及びその使用を提供する。前記二重特異性抗体はTIM−3を標的とするタンパク質機能領域及びもう一つのタンパク質機能領域を含み、単独のTIM−3抗体が有するPBMC(NK)を活性化させて腫瘍細胞を殺傷する活性を残しながら、もう一つのタンパク質機能領域に相応する全長抗体の元の活性を維持し、そして二つの分子の併用に相当するか、それ以上(相乗効果)のT細胞を活性化させる活性に達する。具体的に、発明者は、意外に、アカゲザルのTIM−3と弱い結合活性を有するが、キヌザル及びヒトのTIM−3と強い結合活性を有し、かつヒトNK細胞の腫瘍に対する殺傷作用を活性化させる、TIM−3抗体を見出したが、本発明のTim−3抗体の配列に基づいて特異的に設計された二重特異性抗体(Sbody)は構造が安定で、二つの標的に対する特異的な結合活性が良く、発現量が高く、精製プロセスが簡単である。そして、得られる二重特異分子(二重特異性抗体)は細胞機能活性が二つの単独のモノクローナル抗体の併用と同等かつ/又はそれ以上で、より意外なことに、本発明で設計される二重特異性抗体分子は動物薬効、生存優位性などがいずれも相乗効果を示し、二つの単独のモノクローナル抗体の併用よりも良い。また、二重特異性抗体は、さらに、単剤の低コスト、投与の便宜といった明らかな優勢がある。
本発明の第一の側面では、二重特異性抗体を提供し、第一のタンパク質機能領域及び第二のタンパク質機能領域を含み、前記第一のタンパク質機能領域はTIM−3を標的とする蛋白機能領域であり、前記第一のタンパク質機能領域に相応するTIM−3を標的とするTIM−3全長抗体はアカゲザル(Macaca)TIM−3と弱い結合活性を有し、前記弱い結合活性はELISAによって測定されるEC50値が1nM超であり、好ましくは10nM超であり、より好ましくはEC50値が検出されないものであり、かつキヌザル(Marmoset)及びヒトTIM−3と強い結合活性を有して腫瘍細胞に対するヒトNK細胞の殺傷作用の活性化ができ、前記強い結合活性はELISAによって測定されるEC50値が1nM未満であり、好ましくは0.5nM未満であり、より好ましくは0.2nM未満であり、腫瘍細胞に対する前記ヒトNK細胞の殺傷作用の活性化は、バックグラウンド、即ち、抗体濃度が0μg/mLの場合と比べ、殺される腫瘍細胞が3%以上、好ましくは5%以上、より好ましくは10%以上増加することである。本分野において、抗体と抗原の強い結合活性と弱い結合活性は相対的であり、本願では、上記のように、1nMのELISAによって測定されるEC50値を境に、1nM超の場合は弱い結合であり、1nM未満の場合は強い結合である。
本発明の第三の側面では、さらに、上記のようなDNA配列を含む発現ベクターを提供する。
本発明の第五の側面では、さらに、前記二重特異性抗体の製造方法であって、上記のような宿主細胞を培養し、培養物から二重特異性抗体を得る工程を含む方法を提供する。
サンプルにおけるTIM−3レベルを検出する医薬、TIM−3活性又はレベルを調節する医薬、生体に対するTIM−3の免疫抑制を解消する医薬、末梢血の単核球及び/又はNKリンパ球を活性化させる医薬。
サンプルにおけるPD−1及び/又はTIM−3レベルを検出する医薬、PD−1とPD−L1又はPD−L2の結合を遮断する医薬、TIM−3活性又はレベルを調節する医薬、PD−1活性又はレベルを調節する医薬、生体に対するPD−1及び/又はTIM−3の免疫抑制を解消する医薬、T細胞を活性化させる医薬、T細胞におけるIL−2の発現を向上させる医薬並びに/或いはT細胞におけるIFN−γの発現を向上させる医薬、或いは腫瘍細胞に対するNK細胞の殺傷作用を活性化させる医薬。ここで、前記二重特異性抗体の第二の機能領域は、腫瘍抗体、例えばPD−1を標的とするタンパク質機能領域である。
本明細書で使用されるように、用語「分離された」とは天然の状態で人工的手段によって得られることである。自然界である「分離された」物質又は成分が現れる場合、それが存在する天然の環境が変化したか、或いは天然の環境で当該物質が分離するか、或いは両者の場合いずれも存在する。例えば、ある動物の生体内に天然的にある種の分離されていないポリヌクレオチド又はポリペプチドが存在し、このような天然の状態で分離された高純度の同様のポリヌクレオチド又はポリペプチドは「分離された」という。用語「分離された」は、人工又は合成の物質の混雑も、物質の活性に影響のないほかの不純物の存在もありうる。
本発明で用いられる試薬及び原料はいずれも市販品として得られる。
本発明における二重特異性抗体では、TIM−3を標的とするタンパク質機能領域が相応するTIM−3を標的とするTIM−3全長抗体は、キヌザル、及びヒトのTIM−3との結合活性が良い。キヌザルTIM−3との結合活性は好適に0.05nMでもよい。ヒトTIM−3との結合活性は好適に0.11nMでもよく、US2017114135Aに係るTim−3−0028、Tim−3−0038抗体よりもヒトTIM−3との結合活性が3〜100倍以上強い。アカゲザルTim−3とはほとんど結合しないか、非常に弱く結合する。そのため、既存技術におけるTIM−3抗体の1種類の分子と異なり、本発明のTIM−3抗体は強いヒトPBMCの腫瘍細胞に対する殺傷を活性化させる活性を有し、最高で5.22に達する(乳酸脱水素酵素放出量の増加の百分率)。
以下、実施例の形によってさらに本発明を説明するが、これによって本発明を記載された実施例の範囲内に限定されるわけではない。以下の実施例において、具体的な条件が記載されていない実験方法は、通常の方法及び条件、或いは商品の説明書に従って選ばれる。
本発明で使用されるヒトTIM−3、PD−1、PD−L1細胞外領域−ヒトIgG1 Fc融合タンパク質、−hisタグタンパク質は本発明によってクローニング、発現及び精製して得られた。一部は以下の異なる会社から購入された:TIM−3−his(カタログ番号:TM3−H5229)、TIM−3−hFc(カタログ番号:TM3−H5258)は北京ACROBiosystems社から購入された。TIM−3−his−hFcは北京Sino biological社から購入され、カタログ番号:10390−H03Hである。
ヒトTIM−3配列の詳細はGenBank:Q8TDQ0.3の22〜199位のアミノ酸を参照し、−hIgG1 Fc又は−hisタグと融合した。本明細書におけるGenBank登録番号とは、通常、NCBI Reference Sequenceである。
キヌザル(Marmoset)TIM−3タンパク質配列の登録番号の詳細はGenBank: XP_008982203.1を参照する。
pHが7.4のPBS緩衝液でヤギ抗hFc(Jackson,109−005−008)を1μg/mlの濃度に希釈し、50μl/ウェルの体積で96ウェルマイクロプレート(Corning,CLS3590−100EA)に入れ、37℃インキュベーターで2時間置いた。(或いは、直接に抗原TIM−3で、0.5μg/mlでプレートを被覆した後、直接被験抗体を入れた)。液を捨てた後、PBSで希釈された5%脱脂牛乳(光明脱脂粉乳)のブロッキング液を200μl/ウェルで入れ、37℃インキュベーターで2.5時間インキュベートするか、4℃で一晩(16〜18時間)置くことでブロッキングした。ブロッキング液を捨て、そしてPBST緩衝液(pHが7.4のPBS含有0.05%ツイン−20)でプレートを5回洗浄した後、50μl/ウェルで0.5μg/mlのTIM−3−hFc(実施例1)を入れ、37℃インキュベーターで2時間インキュベートした。インキュベート終了後、PBSTでプレートを6回洗浄した後、50μl/ウェルで上清(被験抗体を含む)又は異なる濃度の被験抗体を入れ、37℃で2時間インキュベートし、PBSTでプレートを5回洗浄し、50μl/ウェルで1:2500で希釈されたHRPで標識された二次抗体(Jackson Immuno Research,115−035−003)を入れ、37℃で1時間インキュベートした。PBSTでプレートを5回洗浄した後、50μl/ウェルのTMB呈色基質(KPL,52−00−03)を入れ、室温で10〜15minインキュベートし、50μl/ウェルの1M H2SO4を入れて反応を中止させ、MMLTISKAN Goマイクロプレートリーダー(ThermoFisher,51119200)によって450nmにおいて吸収値を読み取り、OD値からEC50を計算し、或いは結合活性の高いクローンを選択した。
ヒトNK細胞はヒト末梢血の単核球(PBMC)由来のものであり、PBMCは健常者によって献血された末梢血由来のものであり、本発明によって分離・抽出された。PBMCは2.5×105個細胞/ウェルで、K562(ATCCカタログ番号:CCL−243(商標)、代理:上海素爾生物科技有限公司)は5×104個細胞/ウェルで96ウェルプレート(Corning 3599)に敷いた。被験ハイブリドーマ上清抗体、又は精製抗体を96ウェル細胞プレートに入れ、37℃インキュベーターで6時間インキュベートした後、LDH検出キット(上海同仁生物科技有限公司、カタログ番号:CK12)を使用し、説明書に従って検出し、MMLTISKAN Goマイクロプレートリーダーによって490nmにおける吸収値(OD)を読み取り、LDHの放出量変化の百分率を計算し、被験サンプルのヒトNK細胞(単なるNK細胞の代わりに高投与量のヒトPBMCを使用した)の腫瘍細胞に対する殺傷を活性化させる活性の強さを比較した。
Biacore T200(GE Healthcare)装置によって本発明の抗体と抗原(ヒトTIM−3)の親和力を測定した。pH 7.4の稼働緩衝液HBS−EP+(10mM HEPES、150mM NaCl、3mM EDTA及び0.05%のP20)を使用した(前記百分率は体積比である)。まず、プロテインA(Thermo Pierce,Cat# 21181)をバイオセンサーチップCM5(Cat. # BR−1005 −30,GE)にカップリングし、チップを新しく調製された50mM NHS(N−ヒドロキシコハク酸イミド、N−hydroxysuccinimide)及び200mM EDC[1−エチル−3−(3−ジメチルアミノプロピル)カルボジイミド塩酸塩、1−ethyl−3−(3−dimethylaminopropyl)carbodiimide hydrochloride]で活性化させた後、pH4.0の10 mM NaACで調製された10μg/mlのプロテインAを注いだ。被験抗体の濃度は5μg/mlであり、抗原TIM−3−his(又はPD−1−his)の濃度勾配は0nM、1.875nM、3.75nM、7.5nM、15nM及び30nMであり、流速が30μl/分であり、結合時間が180秒であり、溶離時間が300秒であった。実験後、10mM グリシン−HCl、pH 1.5、30μl/min、30sでチップを洗浄した。実験データはBiacore T200 evaluation version 3.0(GE)ソフトによって1:1のラングミュア(Langmuir)モデルでフィッティングし、親和力の数値KDを得た。
pHが7.4のPBS緩衝液で実施例1で発現されたPD−1を1μg/mlの濃度に希釈し、50μl/ウェルの体積で96ウェルマイクロプレート(Corning,CLS3590−100EA)に入れ、37℃インキュベーターで2時間置いた。液を捨てた後、PBSで希釈された5%脱脂牛乳(上海生工生物工程有限公司,A600669−0250)のブロッキング液を200μl/ウェルで入れ、37℃インキュベーターで3時間インキュベートするか、4℃で一晩(16〜18時間)置くことによりブロッキングした。ブロッキング液を捨て、そしてPBST緩衝液(pH7.4のPBS0.05% ツイン−20を含有)で5回プレートを洗浄した後、50μl/ウェルで1% BSAで5倍連続希釈されたPD−1抗体又は被験抗体を入れ、37℃で1時間インキュベートし、PBSTでプレートを5回洗浄し、50μl/ウェルで1:2500で希釈されたHRPで標識された二次抗体(Jackson Immuno Research,115−035−003)を入れ、37℃で1時間インキュベートした。PBSTでプレートを5回洗浄した後、50μl/ウェルのTMB呈色基質(KPL,52−00−03)を入れ、室温で5〜10minインキュベートし、50μl/ウェルの1M H2SO4を入れて反応を中止させ、MMLTISKAN Goマイクロプレートリーダー(ThermoFisher,51119200)によって450nmにおいて吸収値を読み取り、OD値からEC50を計算した。
pHが7.4のPBS緩衝液で実施例1で発現されたPD−1を2μg/mlの濃度に希釈し、50μl/ウェルの体積で96ウェルマイクロプレート(Corning,CLS3590−100EA)に入れ、37℃インキュベーターで2時間置いた。液を捨てた後、PBSで希釈された5%脱脂牛乳(上海生工生物工程有限公司,A600669−0250)のブロッキング液を200μl/ウェルで入れ、37℃インキュベーターで3時間インキュベートするか、4℃で一晩(16〜18時間)置くことでブロッキングした。ブロッキング液を捨て、そしてPBST緩衝液(pH7.4のPBS含有る0.05% ツイン−20)で5回プレートを洗浄した後、各ウェルに25μlの1% BSAで5倍連続希釈された被験PD−1抗体又は被験抗体及び25μlの最終濃度が10μg/mlのビオチンで標識されたPD−L1(本発明によって発現・精製された)を入れ、37℃で1時間インキュベートし、PBSTでプレートを5回洗浄し、50μl/ウェルで1:1000で希釈されたHRPで標識された二次抗体(genscript社,M00091)を入れ、37℃で1時間インキュベートした。PBSTでプレートを5回洗浄した後、50μl/ウェルのTMB呈色基質(KPL,52−00−03)を入れ、室温で5〜10minインキュベートし、50μl/ウェルの1M H2SO4を入れて反応を中止させ、MMLTISKAN Goマイクロプレートリーダー(ThermoFisher,51119200)によって450nmにおいて吸収値を読み取り、OD値からIC50を計算した。
本発明では、ヒトTIM−3を抗原とし、マウスを免疫させ、異なる融合をスクリーニングし(mab5、mab15、mab35及びmab50など)、数十万のハイブリドーマを得、これらのハイブリドーマからさらにスクリーニングして好適なクローンを得た。非常に意外なことに、先後に多くの異なるハイブリドーマ融合から、いくつかのモノクローナル細胞株をスクリーニングし、それぞれ融合番号はmab5、mab15、mab35及びmab50などで、中国特許出願CN201710348699.4及びCN201810197885.7を参照する。
ハイブリドーマから好適に得られたモノクローナル細胞株から抗体配列を抽出する過程は当業者によく使用される方法である。具体的に、上記モノクローナル細胞株を収集し、増幅培養した後、1×106個の細胞を取り、Trizol(Invitrogen,15596−018)でRNAを抽出し(キットの説明書の手順に従った)、抽出されたRNAをcDNAに逆転写し、逆転写キットは生工生物技術(上海)股フン有限公司から購入され、Cat # B532435であった。逆転写して得られたcDNAを鋳型とし、PCR増幅を行った。増幅産物をシークエンシングすることにより、ハイブリドーマのモノクローナル細胞株における抗体の軽・重鎖可変領域の塩基(コード)配列及びコードされる軽・重鎖のタンパク質配列を得た。使用されたプライマーはNovagenによって発表されたマニュアルTB326 Rev. C0308を参照する。本分野のCDR定義方法は下記表及びCCG定義により本発明の抗体のCDR配列を確認することを含む。
本実施例には、本発明の抗体のヒト化の過程が記載され、当該過程及び方法は本分野の多くの文献で開示された方法によって行われた。
上記実施例7〜9の方法によってハイブリドーマmab5から得られた単一クローンから抽出されたマウス由来抗体をヒト化及び最適化して好適なヒト化抗体ab6を得た。

上記実施例7〜9の方法によってハイブリドーマmab15から得られた単一クローンから抽出されたマウス由来抗体をヒト化した後、最適化し(例えば異なる復帰突然変異部位の組み合わせであり、下記表を参照する)、好適なヒト化抗体ab32を得た。

上記抗体は上記表1及びCCGの方法によってそのCDR領域を下記表のように定義した。
上記実施例9で得られたヒト化抗体の軽・重鎖可変領域に対して実施例1の方法によってクローニング、発現・精製を行って完全な抗体を得た。これらの可変領域は異なる軽・重鎖定常領域と組み合わせることができ、本発明はIgG4−κ鎖(hIgG4)の定常領域が好ましい。発現された好適なヒト化抗体ab6及びab32の活性を前記実施例によってそれぞれ検出した評価結果は以下の通りである。
本発明の抗TIM−3抗体はPD−1を含むほかの様々な標的の抗体と様々な形で二重特異性抗体に設計することができる。設計される二重特異性抗体は、二重機能抗体とも呼ばれ、同時にTIM−3及びもう1つの標的をターゲティングすることができる。本発明では、TIM−3標的及びPD−1標的を例とし、いくつかの好適な設計プランで様々な好適な設計を得た。具体的な設計プランの一つは以下の通りである。
本発明によって設計される上記TIM−3及びPD−1の二重特異性抗体は実施例1の方法によってクローニング、発現・精製された。精製サンプルはPBS(pH7.4)に保存され、3μgのサンプルを取り、順に6μlの5×タンパク質仕込み緩衝液(生工生物工程(上海)股フン有限公司、Cat#C508320−0001)を入れ、非還元サンプルに6μlのDTTのない5×タンパク質仕込み緩衝液(生工生物工程(上海)股フン有限公司、Cat#C516030−0005)を入れ、水で30μlまで追加し、95℃以上の水浴で5min処理した。仕込み、ポリアクリルアミドゲル電気泳動(PAGE)を140vで70min行い、クマシーブリリアントブルーで室温で染色した。結果は図2に示した。
上記本発明のTIM−3及びPD−1の二重特異性抗体に対し、実施例3及び実施例5の方法により、それぞれそのTIM−3及びPD−1との結合活性を検出したが、結果は表15及び図3に示す。
上記結果から、特に上記二重特異性の設計プラン1及び2(表12及び表13)によって設計される二重特異性抗体に生じる重鎖の断裂(図2を参照する)に対し、本発明の最適化設計は下記表16に示す。

上記本発明のTIM−3及びPD−1の二重特異性抗体の最適化設計に対し、実施例2及び実施例5の方法により、それぞれそのTIM−3及びPD−1との結合活性を検出し、結果は表17に示した。
本発明のTIM−3及びPD−1の二重特異性抗体の最適化設計分子は実施例3の方法によってそのヒトPBMCの腫瘍細胞に対する殺傷を活性化させる活性(TIM−3抗体活性)を評価し、結果は表18に示した。
本発明の最適化設計の二重特異性抗体のPD−1抗体活性を評価するため、上記実施例6の方法によってそのPD−1とPD−1リガンド(PD−L1)の結合を阻害する活性を評価した。結果を下記表に示した。
混合リンパ球反応(mixed lymphocyte reaction、MLRアッセイ)でINF−γ分泌を検出する方法によって、本発明の一連の二重特異性抗体のヒト血細胞を活性化させる活性を評価した。即ち、本発明の分離されたヒト血細胞PBMC(健常なボランティアによって献血された末梢血から分離されたもの)で誘導して樹状細胞(DC)を得た後、さらに別のボランティアからのT細胞を刺激した。
(1)取り出された150μlの細胞培養上清を適切な希釈倍数(実験ごとに希釈倍数が異なり、予備実験で希釈倍数が決定され、本実施例における希釈倍数は25倍であった)で希釈した後、マイクロプレートに入れた(100μl/ウェル)。標準品はサンプル共通希釈液で異なる濃度の勾配:1000pg/ml、500pg/ml、250pg/ml、125pg/ml、62.5pg/ml、31.25pg/ml、15.625pg/mlにし、各ウェルに100μl入れた。ブランクウェルにサンプル共通希釈液を入れた。
(3)プレートを5回洗浄し、毎回3分間で、各ウェルに100μlのビオチン抗体作業液を入れ、ブランクウェルにビオチン化抗体希釈液を入れ、新たなシールで反応ウェルを封じ、37℃で60分間インキュベートした。
(6)停止液を100μl/ウェルで入れ、均一に混合した後、3分間内でマイクロプレートリーダーによってOD450を読み取った。
本発明のTim−3抗体配列及びPD−1に対してさらに二重特異性抗体を設計したが、下記表に示す。
LB147 軽鎖:配列番号34;LB147重鎖(LB1413重鎖と同じであり、配列番号33)。
ヒトhPD−1/hTIM3二重遺伝子組み換えBalb/c系マウスBalb/c−hPD−1/hTIM3(江蘇集萃薬康生物科技有限公司から購入、生産許可証番号:SCXK(蘇)2018−0008)で動物薬効モデルを構築し、本発明の二重特異性抗体LB141に対して体内における薬効の評価を行った。
実施例23と同様のヒトTIM−3及びPD−1二重遺伝子組み換えマウスを使用し、同様の飼育条件において本発明の二重特異性抗体のPKの評価を行った。マウスを無作為にA、B群に3匹ずつ分けた。マウスの尾静脈にLB141を20mg/kg/匹/200μlで注射した。注射前0時間、注射後5分、15分、45分、2、6、23、30、47、53、97、122、143、166、196、214、232、238時間で目縁から採血した。採取された血液サンプルを遠心分離し、上清を取り、−20℃で保存し、測定に備えた。血液サンプルを全部収集した後、二重サンドイッチELISA及びPD−1のELISAによってそれぞれLB141のTIM3及びPD−1との結合を検出して(二重特異性抗体はTIM3とも、PD−1とも結合できる)、LB141のPKの特徴を評価した。EXCELソフトによってPKデータを分析し、LB141のT1/2を計算したが、結果は下記表に示した。
実施例25 本発明のTIM−3及びPD−1の二重特異性抗体の製剤配合の安定性の評価
本発明の二重特異性抗体LB141をそれぞれ製剤緩衝液及びpH7.4のPBS(上海源培生物科技股フン有限公司,Cat#B320KJ)で15mg/mLになるように調製し、均等に分けた。−80℃で凍結保存し、37℃で7日、14日置いた後、各サンプルに対して結合活性の検出(ELISA、方法は前記実施例と同様)、及びゲル電気泳動(PAGE)分析を行った。結果を下記表に示した。
本発明の二重特異性抗体LB141を製剤緩衝液で15mg/mLになるように調製し、pH=5.5又はpH=6であり、均等に分けた。−80℃で凍結保存したか、37℃で7日置いた後、各サンプルに対して結合活性の検出(ELISA、方法は前記実施例と同様)、及びゲル電気泳動(PAGE)分析を行った。結果を下記表に示した。
本発明の二重特異性抗体LB141を製剤緩衝液で28.5mg/mLになるように調製し(pH=5.5の緩衝液)、均等に分けた。−80℃で凍結保存したか、37℃で5日及び10日置いたか、又は40℃で5日及び10日置いた後、各サンプルに対して結合活性の検出(ELISA、方法は前記実施例と同様)、及びゲル電気泳動(PAGE)分析を行った。結果を下記表に示した。
前記実施例4の方法によって本発明の二重特異性抗体の親和力を検出し、結果は下記表に示した。
pH7.4のPBS緩衝液でPD−1(実施例1で用意された)を1μg/mlの濃度に希釈し、50μl/ウェルの体積で96ウェルマイクロプレートに入れ、37℃インキュベーターで2時間置いた。液を捨てた後、PBSで希釈された5%脱脂牛乳(上海生工生物工程有限公司,A600669−0250)のブロッキング液を200μl/ウェルで入れ、4℃で一晩(16〜18時間)置くことでブロッキングした。ブロッキング液を捨て、PBST緩衝液(pH7.4のPBSは0.05% ツイン−20を含有する)でプレートを5回洗浄した後、10μg/mlから、1% BSAで5倍連続希釈されたLB141を50μl/ウェルで入れ、37℃で1時間インキュベートし、PBSTでプレートを5回洗浄し、50μl/ウェルで1μg/mlのBio−TIM−3−his(ACROBiosystems,TM−H5229)を入れ、37℃で1時間インキュベートし、PBSTでプレートを5回洗浄し、50μl/ウェルで1:1000で希釈されたストレプトアビジン−HRP二次抗体(南京genscript社,M00091)を入れ、37℃で1時間インキュベートした。PBSTでプレートを5回洗浄した後、50μl/ウェルのTMB呈色基質(KPL,52−00−03)を入れ、室温で5〜10minインキュベートし、50μl/ウェルの1M H2SO4を入れて反応を中止させ、MULTISKAN Goマイクロプレートリーダー(ThermoFisher,51119200)によって450nmにおいて吸収値を読み取り、OD値からEC50を計算した。
LB141を製剤配合(実施例27を参照する)で19mg/mLになるように調製した(pH=5.5)。10μlのLB141を1.5ml遠心管に取り、無菌水で1μg/μLに希釈した。1μLのPNGaseF(Biolabs,P0704L)を入れ、均一に混合し、37℃で16h反応させた後、1μLの1M DTTを入れ、均一に混合した後、37℃で1hインキュベートした。Dionex Ultimate 3000 UHPLC/Thermo Scientific Q Exactive(thermo,MS−B20−03),(HPLC,Agilent,5188−2788)によって質量分析を行った。結果を下記表に示した。
Claims (16)
- 第一のタンパク質機能領域及び第二のタンパク質機能領域を含み、前記第一のタンパク質機能領域はTIM−3を標的とする蛋白機能領域である二重特異性抗体であって、前記第一のタンパク質機能領域に相応するTIM−3を標的とするTIM−3全長抗体はアカゲザル(Macaca)TIM−3と弱い結合活性を有し、前記弱い結合活性はELISAによって測定されるEC50値が1 nM超であり、好ましくは10nM超であり、より好ましくはEC50値が検出されないものであり、かつキヌザル(Marmoset)及びヒトTIM−3と強い結合活性を有して腫瘍細胞に対するヒトNK細胞の殺傷作用の活性化ができ、前記強い結合活性はELISAによって測定されるEC50値が1nM未満であり、好ましくは0.5nM未満であり、より好ましくは0.2nM未満であり、腫瘍細胞に対する前記ヒトNK細胞の殺傷作用の活性化は、バックグラウンド、即ち、抗体濃度が0μg/mLの場合と比べ、殺される腫瘍細胞が3%以上、好ましくは5%以上、より好ましくは10%以上増加することであることを特徴とする抗体。
- 前記第一のタンパク質機能領域は重鎖可変領域及び軽鎖可変領域を含み、前記重鎖可変領域はアミノ酸配列が配列表における配列番号1〜3で示されるCDR、又はアミノ酸配列が配列表における配列番号4〜6で示されるCDRを含み、並びに/或いは、前記軽鎖可変領域はアミノ酸配列が配列表における配列番号7〜9又は配列番号10〜12で示されるCDRを含み、
好ましくは、前記第一のタンパク質機能領域は、重鎖可変領域がアミノ酸配列が配列表における配列番号1〜3で示されるCDRを含み、軽鎖可変領域がアミノ酸配列が配列表における配列番号7〜9で示されるCDRを含むか、或いは重鎖可変領域がアミノ酸配列が配列表における配列番号4〜6で示されるCDRを含み、軽鎖可変領域がアミノ酸配列が配列表における配列番号10〜12で示されるCDRを含み、より好ましくは、配列表における配列番号1〜3で示される配列は順にそれぞれ重鎖可変領域のCDR1、CDR2及びCDR3であり、配列表における配列番号7〜9で示される配列は順にそれぞれ軽鎖可変領域のCDR1、CDR2及びCDR3であり、配列表における配列番号4〜6で示される配列は順にそれぞれ重鎖可変領域のCDR1、CDR2及びCDR3であり、配列表における配列番号10〜12で示される配列は順にそれぞれ軽鎖可変領域のCDR1、CDR2及びCDR3であることを特徴とする請求項1に記載の二重特異性抗体。 - 前記第一のタンパク質機能領域は、重鎖可変領域のアミノ酸配列が配列表における配列番号13又は15で示され、並びに/或いは、軽鎖可変領域のアミノ酸配列が配列表における配列番号14又は16で示され、
好ましくは、前記第一のタンパク質機能領域は、重鎖可変領域のアミノ酸配列が配列表における配列番号13で示され、かつ軽鎖可変領域のアミノ酸配列が配列表における配列番号14で示されるか、或いは重鎖可変領域のアミノ酸配列が配列表における配列番号15で示され、かつ軽鎖可変領域のアミノ酸配列が配列表における配列番号16で示されることを特徴とする請求項1又は2に記載の二重特異性抗体。 - 前記第一のタンパク質機能領域又は第二のタンパク質機能領域は免疫グロブリン、scFv、Fab、Fab’又はF(ab’)2であり、好ましくは、前記第一のタンパク質機能領域は免疫グロブリンであり、前記第二のタンパク質機能領域はscFvであるか、或いは前記第一のタンパク質機能領域はscFvであり、前記第二のタンパク質機能領域は免疫グロブリンであり、前記scFvにおける重鎖可変領域と軽鎖可変領域はリンカー1を介して連結し、前記リンカー1は好ましくは(G4S)nであり、前記nは好ましくは0〜10の間の整数であり、より好ましくは1、2、3又は4であり、前記免疫グロブリンの定常領域は好ましくはヒト抗体の定常領域であり、前記ヒト抗体の定常領域は好ましくはヒト抗体の軽鎖定常領域及びヒト抗体の重鎖定常領域を含み、前記ヒト抗体の軽鎖定常領域は好ましくはκ鎖又はλ鎖であり、より好ましくはκ鎖であり、前記ヒト抗体の重鎖定常領域は好ましくはヒトIgG1、IgG2又はIgG4であり、より好ましくはIgG4であることを特徴とする請求項1〜3のいずれかに記載の二重特異性抗体。
- 前記scFvは軽鎖可変領域−リンカー2−重鎖可変領域であり、軽鎖可変領域のN末端又は重鎖可変領域のC末端がリンカー2を介して相応的に前記免疫グロブリンの軽鎖及び/又は重鎖のC末端又はN末端に連結しているか、或いは前記scFvは重鎖可変領域−リンカー2−軽鎖可変領域であり、重鎖可変領域のN末端又は軽鎖可変領域のC末端がリンカー2を介して相応的に前記免疫グロブリンの軽鎖及び/又は重鎖のC末端又はN末端に連結しており、前記リンカー2は好ましくは(G4S)nであり、前記nは好ましくは0〜10の間の整数であり、より好ましくは1、2、3又は4であり、
好ましくは、前記リンカーは(G4S)3であり、並びに/或いは、前記scFvの数は2つ又は4つ又は6つ又は8つであり、それぞれ対称的に前記免疫グロブリンの軽鎖及び/又は重鎖のC末端及び/又はN末端に連結しており、より好ましくは、前記scFvの数が2つの場合、前記scFvは軽鎖可変領域−リンカー−重鎖可変領域の構造であり、2つのscFvの重鎖可変領域のC末端がそれぞれ(G4S)3を介して対称的に前記免疫グロブリンの2本の軽鎖可変領域又は重鎖可変領域のN末端に連結しているか、或いは前記scFvは重鎖可変領域−リンカー−軽鎖可変領域の構造であり、2つのscFvの重鎖可変領域のN末端がそれぞれ(G4S)3を介して対称的に前記免疫グロブリンの2本の軽鎖可変領域又は重鎖可変領域のC末端に連結しており、かつscFvが重鎖のC末端に連結している場合、前記重鎖のC末端のアミノ酸がKからAに突然変異していることを特徴とする請求項4に記載の二重特異性抗体。 - 前記二重特異性抗体はDVD−Ig(Dual−variable domain Ig、二重可変領域型免疫グロブリン)の二重特異性抗体であり、好ましくは、前記第二のタンパク質機能領域は正常の抗体の軽鎖及び重鎖を含み、前記第一のタンパク質機能領域は軽鎖可変領域及び重鎖可変領域を含み、前記第一のタンパク質機能領域は正常の抗体の軽鎖及び重鎖を含み、前記第二のタンパク質機能領域は軽鎖可変領域及び重鎖可変領域を含むことを特徴とする請求項4に記載の二重特異性抗体。
- 前記第二のタンパク質機能領域は腫瘍抗原、例えばPD−1を標的とするタンパク質機能領域であり、好ましくはPD−1抗体であり、より好ましくは抗PD−1抗体のニボルマブ(Nivolumab)、ペムブロリズマブ(Pembrolizumab)又はBa08であることを特徴とする請求項1〜6のいずれかに記載の二重特異性抗体。
- 前記二重特異性抗体の軽鎖のアミノ酸配列は配列表における配列番号17で示され、かつ前記二重特異性抗体の重鎖のアミノ酸配列は配列表における配列番号18で示されるか、或いは前記二重特異性抗体の軽鎖のアミノ酸配列は配列表における配列番号17で示され、かつ前記二重特異性抗体の重鎖のアミノ酸配列は配列表における配列番号19で示されるか、或いは前記二重特異性抗体の軽鎖のアミノ酸配列は配列表における配列番号20で示され、かつ前記二重特異性抗体の重鎖のアミノ酸配列は配列表における配列番号21で示されるか、或いは前記二重特異性抗体の軽鎖のアミノ酸配列は配列表における配列番号20で示され、かつ前記二重特異性抗体の重鎖のアミノ酸配列は配列表における配列番号22で示されるか、或いは前記二重特異性抗体の軽鎖のアミノ酸配列は配列表における配列番号23で示され、かつ前記二重特異性抗体の重鎖のアミノ酸配列は配列表における配列番号24で示されるか、或いは前記二重特異性抗体の軽鎖のアミノ酸配列は配列表における配列番号23で示され、かつ前記二重特異性抗体の重鎖のアミノ酸配列は配列表における配列番号25で示されるか、或いは前記二重特異性抗体の軽鎖のアミノ酸配列は配列表における配列番号32で示され、かつ前記二重特異性抗体の重鎖のアミノ酸配列は配列表における配列番号33で示されるか、或いは前記二重特異性抗体の軽鎖のアミノ酸配列は配列表における配列番号34で示され、かつ前記二重特異性抗体の重鎖のアミノ酸配列は配列表における配列番号33で示されることを特徴とする請求項7に記載の抗体。
- 請求項1〜8のいずれかに記載の二重特異性抗体をコードするDNA配列。
- 請求項9に記載のDNA配列を含む発現ベクター。
- 請求項10に記載のベクターを含む宿主細胞。
- 請求項1〜8のいずれかに記載の二重特異性抗体の製造方法であって、請求項11に記載の宿主細胞を培養し、培養物から二重特異性抗体を得る工程を含むことを特徴とする方法。
- 請求項1〜8のいずれかに記載の二重特異性抗体を含み、好ましくは、さらに、ほかの抗腫瘍の医薬、及び/又は、緩衝液を含み、より好ましくは、前記緩衝液はヒスチジン又はPBS緩衝液であり、pH5.5〜6.0であり、さらに好ましくは、前記ヒスチジン緩衝液は、10〜20mMのL−ヒスチジン、50〜70mg/mLのショ糖、及び0.1〜1.0%のツイン80又は0.01〜0.05%のツイン20を含み、例えば、前記ヒスチジン緩衝液は、10mMのL−ヒスチジン、70mg/mLのショ糖及び0.2%のツイン80を含む医薬組成物。
- 医薬キットA及び医薬キットBを含む医薬キットの組み合わせであって、前記医薬キットAは請求項1〜8のいずれかに記載の二重特異性抗体を含み、前記医薬キットBは抗腫瘍の医薬を含み、好ましくは、前記医薬キットAを前記医薬キットBと同時に施用するか、或いは前記医薬キットAを前記医薬キットBの前又は後に施用する医薬キットの組み合わせ。
- 癌を治療及び/又は予防する医薬の製造における請求項1〜8のいずれかに記載の二重特異性抗体の使用であって、前記癌は好ましくは肺癌、メラノーマ、腎臓癌、乳癌、結腸直腸癌、肝臓癌、膵臓腺癌、膀胱癌又は白血病である使用。
- 以下のような医薬の製造における請求項1〜6のいずれかに記載の二重特異性抗体の使用:
サンプルにおけるTIM−3レベルを検出する医薬、TIM−3活性又はレベルを調節する医薬、生体に対するTIM−3の免疫抑制を解消する医薬、末梢血の単核球及び/又はNKリンパ球を活性化させる医薬;
或いは以下のような医薬の製造における請求項7又は8に記載の二重特異性抗体の使用:
サンプルにおけるPD−1及び/又はTIM−3レベルを検出する医薬、PD−1とPD−L1又はPD−L2の結合を遮断する医薬、TIM−3活性又はレベルを調節する医薬、PD−1活性又はレベルを調節する医薬、生体に対するPD−1及び/又はTIM−3の免疫抑制を解消する医薬、T細胞を活性化させる医薬、T細胞におけるIL−2の発現を向上させる医薬、T細胞におけるIFN−γの発現を向上させる医薬、並びに/或いは腫瘍細胞に対するNK細胞の殺傷作用を活性化させる医薬。
