Raは、水素、又は置換若しくは非置換C1〜4アルキルであり;
Rbは、水素、又は置換若しくは非置換C1〜4アルキルであり;
Rc及びRdは、環Bの1つの炭素原子上の置換基であり、
Rcは、H、C1〜3アルキル、C1〜3アルキレンORa、ORa、又はハロであり;
Rdは、H、C1〜3アルキル、C1〜3アルキレンORa、ORa、又はハロであり;
Reは、−C(=O)ORa、−C(=O)NRaRb、又は−C(=O)NHSO2CH3である)である。
本発明の例示された化合物は、以下のとおりに調製される。以下の合成スキームは、化合物を合成するために使用される反応の代表例である。本発明の化合物を調製するための変更及び代替のスキームは、以下に示される合成における適切な試薬及び薬剤の置換によって、当業者の能力の範囲内で容易になされる。
[ビス(ジメチルアミノ)メチレン]−1H−1,2,3−トリアゾロ[4,5−b]ピリミジニウム3−オキシドヘキサフルオロホスファート(HATU)(616mg、1.62mmol)、N,N−ジイソプロピルエチルアミン(0.550mL、3.24mmol)を、ジクロロメタン(DCM)(15ml)中の酸化合物I(500mg、1.08mmol)の懸濁液に加え、撹拌した。10分後、4−アミノビシクロ[2.2.2]オクタン−1−カルボン酸メチル(396mg、2.16mmol)及び4−ジメチルアミノピリジン(132mg、1.08mmol)を反応物に加えた。一晩後、溶媒を真空中で除去し、粗製物を、カラムクロマトグラフィーにより精製して、549mgの中間体IIを得た。
10週齢のC57BL/6マウスが、試験のために使用された(各群に関してn=12)。前十字靭帯切除(ACLT)モデルを使用して、OAにおける病態を刺激した。ACLT手術を実施するために、マウスの右後肢の関節嚢を開き、前十字靭帯(ACL)を、手術用顕微鏡下にてマイクロ剪刀で切除した。組織細片を除去するための生理食塩水による洗浄の後、皮膚の切り込みを閉じた。
Claims (15)
- 変形性関節症を治療するか又は予防するための化合物であって、前記化合物が、式(I):
環Bは、
R1は、H、置換又は非置換C1〜4アルキルであり;
nは、0、1、又は2であり;
R2、R3、R4、R5、R7、R8、R9、及びR10は独立して、H、F、Cl、CH3、及びCF3からなる群から選択され;
R6は、
Raは、水素、又は置換若しくは非置換C1〜4アルキルであり;
Rbは、水素、又は置換若しくは非置換C1〜4アルキルであり;
Rc及びRdは、環Bの1つの炭素原子上の置換基であり、
Rcは、H、C1〜3アルキル、C1〜3アルキレンORa、ORa、又はハロであり;
Rdは、H、C1〜3アルキル、C1〜3アルキレンORa、ORa、又はハロであり;
Reは、−C(=O)ORa、−C(=O)NRaRb、又は−C(=O)NHSO2CH3である)
である、化合物又はその薬学的に許容される塩。 - nが、0又は1であり、且つR1が、H又はCH3であるか;又は−(CH2)n−R1が、H、CH3、又はCH2CH3であり;R4が、Hであり、R5が、Hであるか、又は両方のR4及びR5が、Hである、請求項1に記載の変形性関節症を治療するか又は予防するための化合物。
- Ra及びRbが、個別に、H、CH3、又はCH2CH3であり;Rc及びRdが、個別に、H、ハロ、OH、CH3、CH2CH3、又はCH2OHである、請求項1に記載の変形性関節症を治療するか又は予防するための化合物。
- Reが、−C(=O)OH、−C(=O)NH2、又は−C(=O)NHSO2CH3である、請求項1に記載の変形性関節症を治療するか又は予防するための化合物。
- 変形性関節症を治療するか又は予防するための医薬組成物であって、治療若しくは予防有効量の請求項1〜8のいずれか一項に記載の化合物又はその薬学的に許容される塩、及び薬学的に許容される担体、賦形剤、添加剤などを含む、医薬組成物。
- 局所送達のための剤形中にあり、これが、局所的に投与されることになるか又は前記変形性関節症の関節内注射を介して投与されることになる、請求項9に記載の医薬組成物。
- 原発性変形性関節症又は二次性変形性関節症の治療のためのものである、請求項9又は10に記載の医薬組成物。
- 好ましくは、膝関節における変形性膝関節症、股関節における変形性股関節症、足における変形性足関節症及び脊椎における変形性脊椎関節症から選択される体重負荷関節の治療のためのものである、請求項9又は10に記載の医薬組成物。
- 外傷後変形性関節症の治療のためのものである、請求項9又は10に記載の医薬組成物。
- 変形性関節症を治療するか又は予防するための方法であって、それを必要とする患者に請求項9〜13のいずれか一項に記載の医薬組成物を投与することを含む、方法。
- 変形性関節症を治療するか又は予防するための医薬の製造における請求項1〜8のいずれか一項に記載の式(I)の化合物又はその薬学的に許容される塩の使用。
