Abstract

前記アッセイは、前記水性試料中に治療レベルのN−アセチルシステイン(NAC)が存在するか又は存在しない場合に信頼でき、前記キシレノール発色団は2,5−ジメチルフェノールであり、前記触媒はMnCl2水和物であり、前記キシレノール発色団は2,5−ジメチルフェノールであり、前記触媒は、MnCl2水和物であり、かつ、R2は、事前にDMSOに溶解された2,5−ジメチルフェノールを含む。

表1および2に開示されている例示的な酵素および発色団試薬を使用したNACの非存在下および存在下でのアッセイの性能
脱イオン水中において既知の濃度(すなわち、250μmol/L、1000μmol/L、1500μmol/L、2000μmol/L、および2500μmol/L)を有する一連のアセトアミノフェン標準液を用意した。各標準のアリコート10μLをHitachi 717(R)分析装置(Roche Diagnostics)のキュベットで100μLのRlに加え、その後、100μLの脱イオン水で板上希釈した。各混合物を37℃で5分間インキュベートし、それぞれの初期吸光度値を得た。200μLのR2アリコートを各キュベットに添加し、酸化カップリング反応を5分間行い最終吸光度値を得た。660nmでの最終吸光度と初期吸光度との差から、アセトアミノフェンの濃度を計算し、800nmでの吸光度を差し引くことより、バックグラウンドノイズを補正した。各既知濃度の結果を以下の表3に示す。
キシレノール発色団の比較
キシレノール発色団の比較は、実施例1のR2製剤中の前記発色団を置換することにより行った。それ以外のパラメータは同じであった。R2緩衝剤とグルタチオンのストック溶液を作り、その後、6つの組に分けた。7.5g/Lの各異性体をそれぞれ対応した組に溶解することにより、それぞれ異なる異性体を有する6種類の「異なる」発色団試薬を作成した。Rlは変更しないため、分析において唯一の変数はR2における発色団であった。酵素試薬(R1)は、25℃でpH8.6であったが、6つの異性のキシレノール試薬それぞれ25℃でpH11.5であった。既知の各異性体の結果を以下の表5に示す。
2,5−ジメチルフェノールの評価は、既知濃度のアセトアミノフェンの含むイントラリピッド(R)スパイク血清を用いて行った。その結果、治療レベル(<200μmol/L)のアセトアミノフェンで、前記2,5−ジメチルフェノール発色団は200mg/dLのイントラリピッド(R)で±10%の許容限界内で回収した。テストは、Siemens Advia(R) 1650で行った。
表1および2に示されるアセトアミノフェンアッセイの再配合(Reformulation)、アッセイの効率を向上させるように設計された。一態様において、表2に示したアセトアミノフェンアッセイのR2色試薬製造ステップは非常に遅く、その主な理由は発色団の溶解が遅かったためであった。したがって、表1および2に開示されたアセトアミノフェンアッセイに2つの変更を行った。まず、発色団である2,5−ジメチルフェノールを事前にDMSOに溶解した後、発色団試薬(R2)の他の成分に添加した。次に、発色団試薬(R2)組成物中の発色団である2,5−ジメチルフェノールの濃度を半分に低減した。表1および2に示された配合において、2,5−ジメチルフェノールは24x過剰で存在した。再配合アッセイでは、前記濃度が12x過剰に減少した。
Claims (41)
- 試料中のアセトアミノフェンの濃度を決定するためのキットであって、
アセトアミノフェンをp−アミノフェノールに加水分解するためのアリールアシルアミダーゼを含む第1試薬(Rl)と;
p−アミノフェノールと酸化カップリングするためのキシレノール発色団を含む第2試薬(R2)と;
前記キシレノール発色団と前記p−アミノフェノールとの酸化カップリングを触媒するのに適切な触媒とを含み、
前記キシレノール発色団は、2,5−ジメチルフェノールであり、前記触媒は、MnCl2水和物であり、
R2は、DMSOに溶解された2,5−ジメチルフェノールを含み、
前記キットは、1,000mg/lのイントラリピッド(R)の存在下で15.1μg/mLのアセトアミノフェンを検出することができることを特徴とする、キット。 - 前記試料が水性試料であることを特徴とする、請求項1に記載のキット。
- 前記水性試料が血清または血漿であることを特徴とする、請求項2に記載のキット。
- 前記試薬が液状で安定であることを特徴とする、請求項1に記載のキット。
- 前記キットは、アセトアミノフェン決定アッセイを実施するための説明をさらに含むことを特徴とする、請求項1に記載のキット。
- 前記説明には、
前記試料をRlと第1の適切な希釈剤と接触させ、加水分解溶液を形成するステップと;
前記試料中のアセトアミノフェンをp−アミノフェノールに転化する加水分解反応を可能にするように前記加水分解溶液をインキュベートするステップと;
前記加水分解溶液をR2と、必要に応じて第2の適切な希釈液と接触させ、酸化カップリング溶液を形成し、ここでR2が2,5−ジメチルフェノールおよびDMSOを含むステップと;
前記触媒の存在下で、前記キシレノール発色団が前記p−アミノフェノールと結合して着色生成物を形成する酸化カップリング反応を可能にするように前記酸化カップリング溶液をインキュベートするステップと;
形成された前記着色生成物の量を決定し、前記着色生成物の量は、前記試料中に最初に存在するアセトアミノフェンの量に比例するステップとを規定することを特徴とする、請求項5に記載のキット。 - 前記アッセイが、i)生体液中に存在する生体分子、またはii)治療レベルのN−アセチルシステイン(NAC)の存在下で信頼できることを特徴とする、請求項6に記載のキット。
- 前記生体分子が、基本的にビリルビンおよびヘモグロビンからなる群から選択されることを特徴とする、請求項7に記載のキット。
- 前記NACの治療レベルが800mg/Lを超えることを特徴とする、請求項7に記載のキット。
- Rlは、約10U/L〜約5000U/Lの濃度でアリールアシルアミダーゼを含むことを特徴とする、請求項1に記載のキット。
- R2は、約0.075g/L〜約115g/Lの濃度でキシレノール発色団を含むことを特徴とする、請求項1に記載のキット。
- 前記触媒が約0.0005g/L〜約1.000g/Lの濃度でRl中に存在することを特徴とする、請求項1に記載のキット。
- R2は、約0.075g/L〜約115g/Lの濃度で2,5−ジメチルフェノールを含み、
前記触媒が、MnCl2.4H20であり、約2.5g/L〜約20g/Lの濃度でRlに存在することを特徴とする、請求項1に記載のキット。 - R2は、約0.005g/L〜約5.000g/Lの濃度で還元グルタチオンをさらに含むことを特徴とする、請求項1に記載のキット。
- Rlは、タンパク質可溶化剤、タンパク質安定剤、酵素安定剤、金属キレート剤、緩衝剤、界面活性剤、pH調整剤、防腐剤、賦形剤、またはそれらの組み合わせをさらに含むことを特徴とする、請求項1に記載のキット。
- 前記酵素安定剤が、基本的にポリビニルピロリドン40,000MW、BSAフラクションV、トレハロース、p−ヒドロキシ安息香酸ナトリウム、p−ヒドロキシ安息香酸、及びそれらの組み合わせからなる群から選択される1つ以上であることを特徴とする、請求項15に記載のキット。
- R2が、緩衝剤、界面活性剤、pH調整剤、防腐剤、抗酸化剤および賦形剤の1つ以上をさらに含むことを特徴とする、請求項1に記載のキット。
- Rlが、約932.7U/Lのアリールアシルアミダーゼおよび約0.0525g/LのMnCl2.4H20を含み、
R2が、約3.75g/Lの2,5−ジメチルフェノールおよび約0.5g/Lの還元グルタチオンを含むことを特徴とする、請求項1に記載のキット。 - 水性試料中のアセトアミノフェンの濃度を決定する方法であって、
アセトアミノフェンを加水分解してp−アミノフェノールを形成するステップと;
前記p−アミノフェノールを適切な触媒の存在下で酸化カップリングして着色生成物を形成するステップと;
形成された前記着色生成物の量を決定し、形成された前記着色生成物の量は、前記水性試料中に存在するアセトアミノフェンの量に比例するステップとを含み、
前記方法は、前記水性試料に治療レベルのN−アセチルシステイン(NAC)が存在するか又は存在しない場合に信頼でき、
前記キシレノール発色団は、2,5−ジメチルフェノールであり、前記触媒は、MnCl2水和物であり、
かつ、R2は、事前にDMSOに溶解された2,5−ジメチルフェノールを含むことを特徴とする、方法。 - 前記アセトアミノフェンをp−アミノフェノールに加水分解するステップにおいて、前記アセトアミノフェンをアリールアシルアミダーゼと接触させることを含むことを特徴とする、請求項19に記載の方法。
- 前記キシレノール発色団が、2,5−ジメチルフェノール、2,6−ジメチルフェノールおよび2,3−ジメチルフェノールからなる群から選択されることを特徴とする、請求項20に記載の方法。
- 前記触媒が、弱酸化剤であることを特徴とする、請求項21に記載の方法。
- 前記キシレノール発色団が、2,5−ジメチルフェノールであり、前記触媒が、MnCl2無水物または水和物であることを特徴とする、請求項22に記載の方法。
- 前記水性試料が、血清または血漿であることを特徴とする、請求項23に記載の方法。
- 水性試料中のアセトアミノフェンの濃度を決定するためのアッセイであって、
アリールアシルアミダーゼ酵素および適切な希釈剤を含む第1試薬(Rl)と、前記水性試料とを接触させて、加水分解液を形成するステップと;
必要に応じて、前記加水分解溶液を希釈するステップと;
前記アセトアミノフェンをp−アミノフェノールに転化する加水分解反応を可能にするように前記加水分解溶液をインキュベートするステップと;
事前にDMSOに溶解されたキシレノール発色団を含む第2試薬(R2)と、前記加水分解液とを接触させ、酸化カップリング溶液を形成するステップと;
適切な触媒の存在下で、前記キシレノール発色団が前記p−アミノフェノールに結合して着色生成物を形成する酸化カップリング反応を可能にするように、前記酸化カップリング溶液をインキュベートするステップと;
形成された前記着色生成物の量を決定し、形成された前記着色生成物の量は、前記水性試料中に存在するアセトアミノフェンの量に比例するステップとを含み、
前記アッセイは、前記水性試料中に治療レベルのN−アセチルシステイン(NAC)が存在するか又は存在しない場合に信頼できることを特徴とする、アッセイ。 - Rlは、約10U/L〜約5000U/Lの濃度でアリールアシルアミダーゼを含むことを特徴とする、請求項25に記載のアッセイ。
- 前記キシレノール発色団が、2,5−ジメチルフェノール、2,6−ジメチルフェノールおよび2,3−ジメチルフェノールからなる群から選択されることを特徴とする、請求項26に記載のアッセイ。
- 前記触媒が弱酸化触媒であり、かつ前記触媒が約0.0005g/L〜約1.000g/Lの濃度でRl中に存在することを特徴とする、請求項27に記載のアッセイ。
- Rは、約0.075g/L〜約115g/Lの濃度で2,5−ジメチルフェノールを含み、
および/または、
前記触媒は、MnCl2.4H20であり、かつ約2.5g/L〜約20g/Lの濃度でRlに存在することを特徴とする、請求項28に記載のアッセイ。 - R2は、約0.005g/L〜約5.000g/Lの濃度で還元グルタチオンをさらに含むことを特徴とする、請求項28に記載のアッセイ。
- Rlが、タンパク質可溶化剤、タンパク質安定剤、酵素安定剤、金属キレート剤、緩衝剤、界面活性剤、pH調整剤、防腐剤、または賦形剤の1つ以上をさらに含むことを特徴とする、請求項30に記載のアッセイ。
- 前記酵素安定剤が、PVP−40、BSAフラクションV、トレハロース、p−ヒドロキシ安息香酸ナトリウム、p−ヒドロキシ安息香酸及びそれらの組合せからなるから群から選択されることを特徴とする、請求項31に記載のアッセイ。
- R2が、緩衝剤、界面活性剤、pH調整剤、防腐剤、抗酸化剤または賦形剤の1つ以上をさらに含むことを特徴とする、請求項32に記載のアッセイ。
- 前記希釈剤が脱イオン水であり、
加水分解反応前に、前記加水分解溶液が前記希釈液で約1:1希釈されたことを特徴とする、請求項33に記載のアッセイ。 - Rlが、約932.7U/Lのアリールアシルアミダーゼおよび約0.0525g/LのMnCl2.4H20を含み、
R2が、約3.75g/Lの2,5−ジメチルフェノールおよび約0.500g/Lの還元グルタチオンを含むことを特徴とする、請求項25に記載のアッセイ。 - 前記加水分解反応および前記酸化カップリング反応が、それぞれ約37℃の温度で約3〜10分間行われ、
前記加水分解反応が、約8.6のpHで行われ、酸化カップリング反応が、約10.8のpHで行われることを特徴とする、請求項28に記載のアッセイ。 - 前記アセトアミノフェンの濃度が、前記加水分解反応終了時と前記酸化カップリング反応終了時の吸光度の差を得、標準または一連の標準との差を比較することにより決定され、
前記吸光度が、約610nm〜665nmの波長で測定されることを特徴とする、請求項36に記載のアッセイ。 - 前記吸光度が、約660nmの波長で測定されることを特徴とする、請求項27に記載のアッセイ。
- 前記水性試料が、血清または血漿であることを特徴とする、請求項19に記載のアッセイ。
- 水性試料中のp−アミノフェノールの濃度を決定する方法であって、
弱酸化剤である触媒の存在下で、前記p−アミノフェノールと、事前にDMSOに溶解されたキシレノール発色団とを酸化カップリングして着色生成物を形成し、前記キシレノール発色団が2,5−ジメチルフェノール、2,6−ジメチルフェノールおよび2,3−ジメチルフェノールからなる群から選択されるステップと;
形成された前記着色生成物の量を決定し、形成された前記着色生成物の量は、前記水性試料中に最初に存在するp−アミノフェノールの量に比例しているステップとを含むことを特徴とする、方法。 - 前記触媒が、MnCl2水和物であり、前記キシレノール発色団が、2,5−ジメチルフェノールであることを特徴とする、請求項40に記載の方法。
