Abstract
Description
プラスミドの作製及び2’−フコシルラクトース(fucosyllactose,2’−FL)の生産のために、それぞれ、大腸菌(Escherichia coli)K−12 MG1655及びコリネバクテリウムグルタミクム(Corynebacterium glutamicum)ATCC 13032を使用した。
pFGW(Ps、図1)及びpXIL(図2)が挿入されたコリネバクテリウムグルタミクム(C.glutamicum)ATCC 13032を、カナマイシン(kanamycin)25μg/mL及びクロラムフェニコール(chloramphenicol)5μg/mLを含有する5mLのBHI(Brain Heart Infusion)培地が入れられた実験において、30℃、250rpmで12時間、種菌培養した。図1は、pFGW(Ps)プラスミドの模式図であり、図2は、pXILプラスミドの模式図である。
実施例1においてpFGW(Ps、図1)をpFGW(Hp、図3)に変更した以外は同一の培養方法を用いた。図3は、pFGW(Hp)プラスミドの模式図である。
実施例1及び比較例1で製造された組換え菌株における2’−FL(2’−フコシルラクトース)の生産量を比較した(図4、図5、表5)。図4は、ヘリコバクターピロリ(Helicobacter pylori)由来のフコース転移酵素(fucosyltransferase)が挿入された組換え菌株の培養結果であり、図5は、シュードペドバクターサルタンス(Pseudopedobacter saltans)由来のフコース転移酵素(fucosyltransferase)が挿入された組換え菌株の培養結果である(●:乾燥細胞重量、■:ラクトース、▼:グルコース、◆:2’−FL、◇:ラクテート、○:アセテート)。
Claims (6)
- シュードペドバクターサルタンス(Pseudopedobacter saltans)から由来した配列番号5に該当するアミノ酸配列を有するα−1,2−フコース転移酵素(α−1,2−fucosyltransferase)が発現するように形質転換され、GDP−D−マンノース−4,6−デヒドラターゼ(GDP−D−mannose−4,6−dehydratase)が発現するように形質転換され、GDP−L−フコースシンターゼ(GDP−L−fucose synthase)が発現するように形質転換され、ラクトースパーミアーゼ(lactose permease)が発現するように形質転換され、ホスホマンノムターゼ(phosphomannomutase)及びGTP−マンノース−1−ホスフェートグアニリルトランスフェラーゼ(GTP−mannose−1−phosphate guanylyltransferase)を保有していることを特徴とする、組換えコリネバクテリウム属(Corynebacterium SP.)微生物。
- 前記組換えコリネバクテリウム属(Corynebacterium SP.)微生物は、コリネバクテリウムグルタミクム(Corynebacterium glutamicum)、コリネバクテリウムアンモニアゲネス(Corynebacterium ammoniagenes)及びコリネバクテリウムサーモアミノゲネス(Corynebacterium thermoaminogenes)の中から選ばれるいずれか一つであることを特徴とする、請求項1に記載の組換えコリネバクテリウム属(Corynebacterium SP.)微生物。
- 前記配列番号5に該当するアミノ酸配列を有するα−1,2−フコース転移酵素(α−1,2−fucosyltransferase)は、配列番号4に記載の核酸配列に暗号化されたことを特徴とする、請求項1に記載の組換えコリネバクテリウム属(Corynebacterium SP.)微生物。
- 前記組換えコリネバクテリウム属(Corynebacterium SP.)微生物は、ホスホマンノムターゼ(phosphomannomutase)が過剰発現(overexpression)するように形質転換され、GTP−マンノース−1−ホスフェートグアニリルトランスフェラーゼ(GTP−mannose−1−phosphate guanylytransferase)が過剰発現するように形質転換されたことを特徴とする、請求項1に記載の組換えコリネバクテリウム属(Corynebacterium SP.)微生物。
- ラクトース添加の培地にて、請求項1の組換えコリネバクテリウム属(Corynebacterium SP.)微生物を培養することを特徴とする、2’−フコシルラクトースの生産方法。
- 前記培地は、グルコースをさらに含むことを特徴とする、請求項4に記載の2’−フコシルラクトースの生産方法。
