Abstract
Description

を含む融合分子を調製した。融合タンパク質コンストラクトの概略的を図1に示す。融合タンパク質は、二量体としてBBB通過単一ドメイン抗体、Fc断片、及びアミロイド結合ペプチドを含む。
実施例1に記載されたコンストラクトFC5−mFc−ABP及びヒト化FC5(H3)−hFc−ABPを二量体としてCHO細胞内で発現させ、発現された化合物をMabSelect Sureアフィニティーカラム上で精製した。
実施例2で生成したコンストラクトを免疫組織蛍光アッセイに供し、FC5−Fc−ABP融合分子が、記載されるようなマウス脳の天然に産生されたアミロイド沈着に結合する能力を維持しているかどうかを評価した(Chakravarthy et al., 2014)。野生型(Wt)及びAD遺伝子組換え(AD−Tg)マウス由来の凍結半頭脳をOCTに埋め込み、Jung CM 3000クリオスタットを使用して10μm切片を調製し、−80℃で保存した。組織切片を解凍し、かみそりの刃でOCTを切片から剥離させ、Dakoタンパク質ブロッキング試薬と共に室温で30分間インキュベートした。ブロッキング剤を除去し、切片をTBS中で穏やかに洗浄した。抗体希釈液中のIR800標識FC5−mFc−ABP(5.0μg/μl溶液の1:250希釈)を添加し、室温で1時間インキュベートした。次に、切片をTBSで2回洗浄し、ミリQ水ですすぎ、過剰なすすぎ溶液を除去し、切片にDako Fluorescent Mounting Mediaと共にカバーグラスを載せた。蛍光顕微鏡下で視覚化された切片(図3)。選択的結合(明るい点)は、Aβ沈着物を産生するAD−Tgマウスの脳切片で見られ、アミロイド沈着物を産生しないWtマウスの脳切片では見られず、FC5−Fc−ABPコンストラクトのABPが、AD−Tgマウス脳内で天然に産生されたAβ凝集体に結合する能力を維持することが示唆された。提供されるBBB−Fc−ABPコンストラクトにおいて、BBBはFC5又は抗IGF1R抗体であってもよい。
生体外ラット及びヒト由来のBBBモデルで、FC5−Fc−ABP融合分子のBBB通過を評価した(図4)。Papp測定のための単一時点を使用して、BBBを通過する融合分子を生体外BBB透過性アッセイでスクリーニングした。変異型の定量化をMRM−ILISによって実施した(図4A、4B、及び4C)。
実施例2のコンストラクトが、血液脳関門から脳、特に脳脊髄液(CSF)に移動する能力を生体内で評価し、並びにCSF及び血清中のコンストラクトの存在を定量化した。FC5−mFc−ABPを示された用量(2.5、6.25、12.5、及び25mg/kg)で尾静脈を通じてラットに静脈内投与した。血清及びCSFを連続的に採取した。FC5−Fc−ABPレベルをnanoLC−MRM法を用いて定量化した。
アルブミン比=分析された血漿のnLあたりの強度/分析されたCSFのnLあたりの強度
1500以下の比率は、血液汚染と見なされた。
FC5−mFc−ABPの血清及びCSF PKプロファイルをビーグル犬において評価した。FC5−mFc−ABPを10〜12歳齢のビーグル犬に静脈内注射によって投与し、血清及びCSFを連続的に採取して、上記の実施例5に記載されるように、nanoLC−MRM(左パネル)によって、及びFc特異的抗体を使用したウエスタンブロットによって分析した(図6B)。星印は、血液に汚染されたサンプルを示す(MRM分析には示されない)。見て分かるように、FC5−mFc−ABPはCSF中に時間依存様式で出現し、生体内でイヌ血液脳関門を越えるFC5によるABPの輸送を示唆し、BBB担体の翻訳特性を裏付けた。PKパラメータ及びCSF暴露をWinNonlinソフトウエアによって分析し、以下の表3に示す。
生体内(ラットモデル)においてヒトFc(hFc)と融合しABP(FC5−hFc−ABP)に化学的に連結されたFC5のBBB透過性とCSFの出現。FC5−hFcは、製造会社の使用説明書(ThermoFisher Scientific)に従ってヘテロ二機能性架橋剤スルホ−SMCC(スルホスクシンイミジル4−[N−マレイミドメチル]シクロヘキサン−1−カルボキシレート)を使用して、ABP−シスタミドと連結させた。化学的にコンジュゲートされた分子をラットに6.25mg/kgで尾静脈を介して静脈内投与し、血清及びCSFサンプルを4及び24時間目に採取して、図5に記載されるように分析した。図7に示されるように、FC5−hFc−ABPはCSF中に時間依存様式で出現するが、BBB担体のないFc−ABPは出現せず、血液脳関門を越えるABPのFC5媒介輸送を裏付けた。
実施例2のFC5−Fc−ABPコンストラクトが、生体内で血液脳関門を移動し脳実質に浸透する能力をマウスで評価した。
Tgマウスのアミロイド負荷に対するABPの有効性を評価するために、ABP単独又はFC5−ABPコンストラクトによる処置の結果を比較した。
FC5−ABP(FC5配列番号17;及びABP配列番号36)及びFC5−hFc−ABP(FC5配列番号17;hFc1x7配列番号49及びABP配列番号36]コンストラクトを実施例2に記載されるようにCHO細胞で生成した。血清PKは、実施例5に記載されるように判定した。FC5−ABP及びFC5−Fc−ABPコンストラクトを15mg/kgでラットの尾静脈に静脈内投与した。血清を連続的に採取し、FC5−ABP及びFC5−Fc−ABPレベルをFC5−及びABP−特異的抗体を使用した直接ELISAによって定量化した。血清サンプルをリン酸緩衝生理食塩水(PBS)で希釈(1:5,000)し、Maxisorbプレートに適用して、4℃で一晩インキュベートした。ELISAプレートを3X×100μlのPBSで洗浄し、室温で(RT)30分間、TBST中の1%BSAでブロックした。ブロッキング溶液を除去し、プレートをHRPコンジュゲートFC5モノクローナル抗体と共にインキュベートした(90分間)。インキュベーションと洗浄に続いて、100μlのSureBlue試薬を添加し、室温で10〜15分間暗所でインキュベートした。反応の終了時に、100μlの1M HClを添加し、プレートリーダーで450nmにおける発色を読み取った。図10に示されるように、Fcを含まないFC5−ABPは、Fc構成要素を含むFC5−ABPコンストラクト(FC5−Fc−ABP)と比較して、血清中で非常に急速に(1時間以内)除去された。このアッセイはまた、ABP特異的抗体でも同様の結果で繰り返された。サンプル適用後、ELISAプレートを最初にABPウサギポリクローナル抗体と共にインキュベートし(90分間)、HRPコンジュゲートウサギ二次抗体がそれに続き(30分間)、結合抗体を上記のように検出した(データ未掲載)。
AD−Tgラットに、生理食塩水又はFC5−mFc−ABPのどちらかを4週間にわたり毎週尾静脈から投与した(30mg/kgの初回負荷量及び引き続く15mg/kgの4回の週用量)。FC5−mFc−ABP及びAβのCSFレベルをnanoLC MRMによって分析した。4週間の処置の前後に、特定のAβ結合剤[18F]NAV4694を使用して、PETスキャンによって脳Aβレベルを判定した。FC5−mFc−ABPは、24時間以内にラットにおけるCSFのAβレベルを減少させた。Tgマウスにおけるように、FC5−mFc−ABPのCSFレベルとAβとの逆相関が観察され、FC5によって脳及びCSFに送達されたABPによる、Aβの標的結合と急速なクリアランスが示唆された(図11A及び図11B)。これは、FC5−mFc−ABPによる4週間の処置に続くラット脳Aβレベルの有意な減少(30〜50%)を明確に示したPETスキャンによって、さらに裏付けられた(図11C)。
実施例11に記載される実験では、生理食塩水処置及びFC5−mFc−ABP処置Tgマウスを、処置前に体積測定及び機能的磁気共鳴画像化(MRI)に供した。体積測定MRI(図12A)は、生理食塩水処置対照と比較して、ABP処置Tgマウスにおける海馬の体積の増加を示し、ABPで処置すると海馬の萎縮が停止したことが示唆された。機能的MRI(図12B)は、生理食塩水処置対照と比較して、ABP処置Tgマウスの前帯状回(cingulated)皮質における改善された結合性を示し、神経細胞の結合性の回復が示唆された。このデータは、現在提供されている意義、有効性、及び優れた治療上の利点を裏付ける。
実施例6に記載されるように、FC5−mFc−ABPの血清及びCSFのPKプロファイルを2つの用量(15mg/kg及び30mg/kgで、ビーグル犬において評価した。nanoLC−MRMによるFC5−mFc2a−ABPの血清及びCSFレベルの測定に加えて、AβのCSFレベルもまた、実施例9に記載されるようにnanoLC−MRMによって測定した。見て分かるように、FC5−mFc−ABPは、用量及び時間依存様式でCSF中に出現した(図13B)。最も重要なことは、Tgマウス及びTgラットで見られるように、CSF FC5−mFc2a−ABPレベルに反比例する、CSF Aβレベルの有意な減少があった。
ABP融合分子の汎用性を評価するために、ABPを別のヒト化BBB担体IGF1R5(H2)と成功裏に融合させた。図12に示されるように、分子の二機能性が維持され、ABPがAβオリゴマーに結合する能力(ELISA及びオーバーレイアッセイ)、及びIGF1R5が生体外でBBBモデルを越えてABPを送達する能力(データ未掲載)もまた維持された。これは、ABPが、異なるBBB通過単一ドメイン抗体と融合され、脳に送達され得ることを明確に示唆する。
修飾されたABP(表1及び図15に示される配列番号によって示されるような分子の部位特異的変異又はC末端部分の除去)を有する、様々なFC5−Fc−ABPコンストラクトが提供される。図15に示されるように、全てのコンストラクトは、ELISA法によって、Aβオリゴマーの結合における同様の効力を維持した。
特定の変異を有するFC5−hFc1X7−L−ABP(配列番号35のABP;配列番号36のABP)をCHO細胞で生成し、還元(R)及び非還元(NR)条件下においてSDS−PGE上で分離して、図2に記載されるようにクマシーブルーで染色した。分離されたタンパク質をニトロセルロース膜に転写し、FC5特異的、hFc特異的、及びABP特異的の抗体を用いて免疫ブロット法を実施した。別のセットでは、融合分子中のABPのAβ結合もまたオーバーレイアッセイにより試験した。結合したAβをAβ特異的抗体6E10によって検出した。見て分かるように、特定の変異(例えば、ここに示されているように、ABP配列番号35及びABP配列番号36)によるABPの系統的修飾は、例えば、還元及び非還元条件下での単一タンパク質バンドによって本明細書で明確に示されているように、生成される分子の安定性を実質的に高めた(二重タンパク質バンドが見られる、図2Aのその他のABPコンストラクトと比較して)。融合分子の安定性におけるこの実質的な強化は、有利には、均質分子の生物製造可能性を促進する。
図4に記載されているように、BBB通過を生体外ラットBBBモデルで評価し、血液脳関門を通過する分子をnanoLC−MRM法で検出した。ヒト化FC5及びIGF1R担体に融合された全てのABP変異型は、BBBを効果的に通過した。予測されたように、非BBB透過性sdAbであるA20.1はBBBを通過せず、同様に、A20.1に融合されたABPはBBBを透過しなかった(図17A)。図17Bでは、融合分子の3つの構成要素、FC5、Fc、及びABP全ての「フィンガープリント」ペプチドが、nanoLC−MRMによって検出されたことが示される。
ヒト化FC5−ABP融合分子の血液脳関門通過を実施例4に記載されるように評価した。血液脳関門を通過する融合分子をウエスタンブロット法及びELISA法によって分析した。免疫ブロットは、hFc特異的及びABP特異的抗体で探索した。どちらの抗体もBBB(ボトムチャンバー)を通過する分子を認識し、分子サイズは生体外BBB(上部チャンバー)に適用されたFC5−ABP融合コンストラクトの分子サイズと同一であり、BBBを通過した分子が無傷のままであることを示した。これは、サンドイッチELISAアッセイによって立証され、BBB通過分子がFC5特異的抗体で捕捉され、捕捉された分子がABP抗体を用いて検出された。
マウスにおけるヒト化FC5−ABP融合分子の血液脳関門通過及び脳送達を、実施例8に記載されるように評価した。分子の静脈内投与及び心臓内灌流に続いて4時間後、脳を摘出し、皮質をRIPA緩衝液で抽出し、注入されたFC5−ABP融合分子の存在を、ABP特異的抗体でプローブするウエスタンブロットによって、及び分子をFC5特異的抗体で捕捉してABP特異的抗体で検出するサンドイッチELISAによって、検出した。ウエスタンブロットは、皮質に完全長のFC5−ABPコンストラクトが存在することを明らかにした。これはサンドイッチELISAによって立証され、FC5特異的抗体によって分子が捕捉され、ABP特異的抗体によって捕捉された分子が検出される能力によって示唆されるように、それは分子の無傷の特性を明らかにした。
免疫組織化学検査を実施例3に記載されるように実施した。AD遺伝子組換えマウス由来の脳切片をFC5(H3)−hFc1X7−ABP(ABP、配列番号36)と共にインキュベートし、結合した融合分子はHRPコンジュゲートFC5特異的抗体を用いて視覚化した。陰性対照として、切片をコンストラクトなしで、又は融合ABPなしのFC5−hFcコンストラクトと共に、インキュベートした。簡潔に述べると、APP/PS1遺伝子組換えマウスの皮質及び海馬を含むホルマリン固定40μmのフリー浮遊切片を、10mMクエン酸ナトリウム緩衝液(pH9)中で80°Cで30分間抗原賦活化に供し、次に室温まで冷却した。切片をPBSですすぎ、PBS中の3%H2O2で30分間処理して内因性ペルオキシダーゼをブロックし、再度すすいだ。0.3%トリトンX−100を含むDako無血清タンパク質ブロック中での1時間のインキュベートに続いて、切片を0.3%トリトンX−100を含むDako希釈液中のFC5(H3)−hFc1x7−ABPと、又はFC5−hFc1x7のモル当量と共に、室温で90分間インキュベートした。PBSで完全にすすいだ後、切片をDako希釈液中の抗FC5−HRPと共に室温で60分間インキュベートし、再度すすぎ、次にキットの指示に従ってVector Immpact DABを使用して展開した。切片をSuperfrostプラススライドに載せて一晩風乾させ、次に再水和させてメチルグリーンで対比染色し、アセトン/0.05%酢酸(v/v)に浸して脱水し、透明化してPermountで覆った。FC5(H3)−hFc1X7−ABPコンストラクトでは選択的結合(黒点)が見られたが、ABPのないFC5(H3)−hFc1X7では見られず、脳内のAβ沈着のABP依存性結合(標的結合)が示唆された。アミロイド沈着を生じない野生型マウス由来の脳切片には、結合は見られなかった(データ未掲載)。
フルオロフォア標識(図18A)又は未感作FC5−ABP(図18B)融合分子を、野生型(Wt)及びAD−Tg(Tg)マウスの海馬領域に微量注入した。フルオロフォア標識FC5−ABP融合分子のマイクロインジェクションの30分後、脳を摘出し、切片化して蛍光顕微鏡下で観察した。21Aに示されるように、注入された分子は、Aβ特異的抗体との共局在によって確認されるように、Aβ沈着物に結合した。並行研究では、未感作FC5−ABP融合分子の海馬内注射の4時間後に、注入領域(ips)及び非注入領域(con)からの海馬体を採取して、トリス緩衝化生理食塩水で均質化した。捕捉抗体としてFC5抗体を用い、検出抗体としてABP又はAβ特異的抗体のどちらかを用いる、サンドイッチELISAに海馬抽出物を供した。21Bに示されるように、微量注入されたFC5−ABP分子は無傷のままであり、Aβ特異的抗体によって検出されたプルダウン複合体中のAβの存在によって示されるように、ABPは生体内で標的Aβと会合し結合できた。
FC5−mFc2a−ABP又はFC5(H3)−hFc1x7−ABPを、図5に記載されるように15mg/kgで尾静脈注射を通じてラットに静脈内投与した。血清及びCSFを連続的に採取した。FC5−Fc−ABPレベルをnanoLC−MRM法を用いて定量化した。図22Aに示されるように、血清及びCSFのPKプロファイルは、非ヒト化及びヒト化コンストラクトで非常に良く似ていた。FC5−mFc2a−ABP又はFC5(H3)−hFc1x7−ABPを図9Bに記載されるように15mg/kgで尾静脈注射を通じてTgマウスに静脈内投与した。CSF中のFC5−Fc−ABP及びAβレベルを図9Bに記載されるようにnanoLC−MRMによって測定した。図22Bに示されるように、CSF中の非ヒト化及びヒト化FC5−Fc−ABPのレベルは類似しており、最も重要なことには、CSFのAβレベルの変化(減少)も非常に良く似ており、FC5−Fc−ABPコンストラクトのヒト化は、融合コンストラクのPK及びPDプロファイルに影響を及ぼさなかった。
融合タンパク質を、実施例2及び16に記載のように安定なCHO細胞株中で産生させた。プロテインAアフィニティーカラムを使用した精製の後(実施例2)、融合タンパク質を、陽イオン交換(CEX)クロマトグラフィーを使用してさらに精製した。CEXクロマトグラフィープロファイルは、FC5−(H3)−hFc1x7−ABP(6G)の変異型の存在を示した。それぞれのCEX画分の、抗hFc抗体を用いたウエスタンブロット分析(23A)及び質量分析(23B)の際に、融合タンパク質の変異型の生成が、産生中のABPペプチドのC末端切断に起因することが決定された。C末端切断は、配列番号36のABP配列のC末端切断ペプチドである配列番号37〜配列番号43に示されるように、FC5−(H3)−hFc1x7−ABP(6G)タンパク質中のABP配列の特定の部位で起こる。配列番号36のC末端切断産物又は配列番号47を有するABPは、二量体形式で融合タンパク質中に含有された場合、予想外の安定性及びβアミロイド結合活性を示した。さらに、このようなC末端切断産物は、有利には、ELISA及びウエスタンブロットAβオーバーレイアッセイ(実施例2に記載されるように)によって評価された場合、機能的なホモ二量体形態と比較して、βアミロイド結合活性の同等の生物学的機能(EC50、図23A)を維持した。これらのC末端切断産物は、有利には、機能的且つ安定なβアミロイド結合産物の製造可能性及び単離可能な収率を増加させる。これらの安定な切断産物は、自明ではなく、βアミロイド結合性機能的ペプチド並びに対応する融合ペプチド及び二量体収率に関して有利である。したがって、CHO細胞中での産生中のABPのC末端切断は、図1Ai、1Aii及び1Aiiiに描写されるように、融合タンパク質の二機能性ホモ二量体、二機能性ヘテロ二量体(ABPアームが両方とも機能的であり、任意のABP及び/又はC末端が切断されているが、機能的である任意のABPを含有してもよい)及び一機能性ヘテロ二量体(二量体中の1つの機能的ABP)の生成をもたらした。融合タンパク質(配列番号56)の切断産物の正確な性質は、詳細な分析が行われるまで自明ではなく、生物製造及びタンパク質産生において予測することができなかった。ここで定義されるC末端切断産物が、βアミロイドに有利に結合し、安定なペプチドであることは、知られておらず自明でもなかった。二機能性ホモ二量体、並びに一機能性及び二機能性ヘテロ二量体の混合物を含有するCEX画分のβアミロイド結合活性(ピーク2、図23A、EC50 19nM)は、二機能性ホモ二量体のそれ(ピーク3、図23A、EC50 13nM)と非常によく似ていたが、これは、ヘテロ二量体及びホモ二量体画分(CEXのピーク2及び3)を、βアミロイド結合活性を有意に損なうことなく組み合わせることができることを示している。これは、大規模生物製造中に、完全に機能的な、安定な融合タンパク質としてのFC5(H3)−hFc1x7−ABPの収率及び単離可能性を実質的且つ有利に増加させる。これは、修飾CEX条件が高収率の画分を生成し(ピーク2、図23C)、カラムに印加された総タンパク質の86.4%を占めることを示す、図23Cにおいて示された。ピーク2の典型的な質量分析は、図23Bに示されるように、配列番号56のホモ二量体(MW87,695)と、配列番号56及び配列番号57(MW=87,470);配列番号56及び配列番号60(MW=87,085)並びに配列番号56及び完全に切断された非機能的ABP(MW=84,040)を含むヘテロ二量体との混合物からなる。この組み合わせたプール(画分B3〜B6)のβアミロイド結合活性(EC50)は、図23C、B及びCに示されるように、主にホモ二量体型(C10〜D2、E12)又はヘテロ二量体型(C6〜C8)のいずれかを含有する他のCEX画分と非常によく似ていた。
血液脳関門を通過するFC5(H3)−hFc1x7−ABPの輸送及び脳の標的領域への送達を、実施例8及び19に記載のように評価した。FC5(H3)−hFc1x7−ABP融合タンパク質(実施例23に由来するホモ及びヘテロ二量体の混合物として)を、15mg/kgで、野生型(wt)及びTgマウスの尾静脈を介して投与した。注入の4時間(データ未掲載)又は24時間後、脳の皮質及び海馬領域を切開し、実施例8及び19に記載のように評価した。図24に示されるように、FC5(H3)−hFc1x7−ABP融合タンパク質は、サンドイッチELISA(24A)及びウエスタンブロット分析(24B)によって評価されるように、標的領域中で検出された。これらの結果は、主にホモ二量体型のFC5(H3)−hFc1x7−ABP(配列番号56、図19を参照されたい)の脳への送達と非常によく似ており、混合物中のヘテロ二量体型のFC5(H3)−hFc1x7−ABPが、血液脳関門の通過及び融合タンパク質の脳への送達に影響しないことをさらに裏付けている。
TgマウスへのFC5(H3)−hFc1x7−ABPの注射の静脈内投与の24時間後、CSFを回収し、FC5(H3)−hFc1x7−ABP及びβアミロイドのレベルを、実施例5及び22に記載されたMRMによって測定した。図25に示されるように、FC5(H3)−hFc1x7−ABPのCSF曝露及びCSFのAβレベルの変化(減少)は、主にホモ二量体型のFC5(H3)−hFc1x7−ABP(配列番号56、図22を参照されたい)について見られるものと非常によく似ており、混合物中のFC5(H3)−hFc1x7−ABPが、FC5(H3)−hFc1x7−ABP融合タンパク質の機能的有効性に影響しないことをさらに裏付けている。
