Abstract
Description
本出願は、2018年2月28日に出願された米国特許仮出願第62/636,825号に対する優先権を主張するものであり、なお、当該特許仮出願は、参照によりその全体が本明細書に組み入れられる。
本発明は、抗体に関する。より詳細には、本発明は、癌療法のための抗体に関する。
ヒトにおけるリンパ球の2つの主要なタイプは、T(胸腺由来)およびB(骨髄由来)である。これらの細胞は、リンパ球発達経路のために関与した骨髄および胎児肝臓の造血幹細胞に由来する。これらの幹細胞の後代は、Bリンパ球またはTリンパ球に成熟するための分岐経路に従う。ヒトBリンパ球発達は、完全に骨髄内において生じる。その一方で、T細胞は、骨髄を去って血流によって胸腺まで移動する未成熟前駆体から発達し、そこで、それらは、増殖し、成熟Tリンパ球へと分化する。
T細胞は、最も豊富に存在し(血液リンパ球の約75%)、強力な免疫キラー細胞である。抗腫瘍性免疫応答におけるエフェクターT細胞の役割は、インビトロ研究および、腫瘍のいくつかのタイプにおけるCD8+T細胞の高浸潤が好ましい臨床予後と相関関係を有するという観察(Fridman et al.,2012)によって強く支持される。エフェクターナイーブT細胞の活性化は、少なくとも3つの相補シグナル:(i)共受容体(CD4またはCD8)の支援によるTCR−CD3/Ag−MHC相互作用;(ii)CD80またはCD86などの共刺激性分子のCD28、CD40/CD40Lへの結合;(iii)サイトカインなどのアクセサリ分子を必要とする。
免疫チェックポイントは、通常、付随的な組織損傷を最小限に抑えるため、自己寛容性を維持し、末梢組織における生理学的応答の持続期間および振幅を調節するために、免疫系、具体的にはT細胞媒介性免疫を張り巡らせるリガンド−受容体相互作用によって開始される、阻害性および刺激性経路のグループを意味する(Pardoll,2012)。腫瘍細胞は、免疫抵抗性の主要メカニズムとして、ある特定のチェックポイント経路を選出する。例えば、プログラム細胞死タンパク質1リガンドのPD−L1は、一般的に、ヒト癌の腫瘍細胞表面において上方制御される。PD−L1と、腫瘍浸透リンパ球(TIL)、特にT細胞、において発現される、その受容体であるPD−1との相互作用は、局所的T細胞媒介性応答を阻害することにより、免疫監視から逃れる(Liang et al.,2006;Sznol and Chen,2013)。したがって、癌細胞での免疫抑制性シグナルの阻害、またはT細胞の直接的アゴニスト刺激は、強い持続性の抗腫瘍免疫応答を生じるおよび/または誘導する。最近の臨床研究は、抗体を介した、または可溶性リガンドまたは受容体によって調節される、免疫チェックポイントタンパク質の遮断が、治療的抗腫瘍免疫の活性化に対する最も有望なアプローチであることを強く示唆した(Topalian et al.,2014)。現在、抗PD−1および抗CTLA−4(細胞傷害性Tリンパ球関連抗原−4)抗体が、黒色腫などの疾患を治療するために、FDAによって承認されている。
血管新生、すなわち、既存の血管からの新しい血管の形成は、胎児発達および組織修復にかかわる正常で不可欠なプロセスである。当該プロセスは、血管新生因子および抗血管新生因子の両方によって高度に制御され、ならびに、それは、内皮細胞の遊走および増殖、血管の成熟化および再構成、ならびに細胞外マトリクスの分解を伴う。それは、正常な増殖および発達において重要なプロセスであるが、血管新生は、腫瘍増殖においても重要な役割を果たす。腫瘍は、増殖するために血管供給を必要とし、リガンドの血管内皮増殖因子(VEGF)ファミリーのメンバーを含む血管新生促進増殖因子の発現を介してこれを達成することができる(Hicklin and Ellis,2005)。VEGFおよび他の内皮増殖因子が、内皮細胞上のそれらの受容体に結合する場合、これらの細胞内のシグナルが開始され、それは、新しい血管の成長および生存を促進する。VEGF特異抗体(アバスチン)、可溶性VEGF受容体(アフリベルセプト)、またはVEGFチロシンキナーゼ活性の阻害剤(スニチニブ)によるVEGF活性の阻害は、腫瘍または血管新生タイプの障害(例えば、AMDなど)を治療するために使用されてきた戦略である。
エフェクター免疫細胞に腫瘍細胞を効果的に再標的化させるために二重特異性抗体を使用するアイデアは、1980年代に現れた(Karpovsky et al.,1984;Perez et al.,1985;Staerz et al.,1985)。二重特異性の足場は、一般的に、Fc断片、IgG様分子、および小さい組換え二重特異性形式(それらの大部分は、単鎖可変断片(scFv)に由来する)の有無に基づいて、異なる薬物動力学特性を有する2つの主要な群に分類される。抗体断片は、それらのコンパクトなサイズにより、通常、IgG様分子より効率的に腫瘍に浸透するが、この利点は、それらの全体的腫瘍取り込みおよび滞留時間を制限する短い血清半減期(数時間)によって緩和される(Goldenberg et al.,2007)。対照的に、新生児Fc受容体に結合するFc断片の存在は、IgG様形式に対して長い血清半減期(>10日)を提供し、これは、腫瘍取り込みおよび滞留に有利に働くが、腫瘍浸透を制限する。
実施例が添付の図面に示されている本発明の本実施形態について、以下において詳細に言及されるであろう。同じまたは同様の部分を参照するために、図面および説明において、可能な限り、同じ参照番号が使用される。
PD−L1に対する治療抗体の生成のために、OmniMabファージミドライブラリによる選択を実施した。当該ファージミドライブラリは、百を超える健康なドナーのB細胞のコレクションからAP Biosciences Inc.(APBio Inc.)によって生成される。パンニングの第一ラウンドのためのファージを、Hyperphage(Μ13Κ07ΔρΙΙΙ、Progen、ハイデルベルク、ドイツ)によって調製した。PD−L1特異的バインダー選択およびOmniMabライブラリからの単離に対して、PD−L1に対する固相パンニングおよび細胞パンニングを適用した。固相パンニングは、第一ラウンド選択において組換えヒトPD−L1−Fc(APBio Inc.)を使用して実施し、次いで、追加の第二ラウンドの濃縮に対して、PD−L1を発現したHEK293細胞を使用した。3つのラウンド選択の後、当該特異的PD−L1バインダーをスクリーニングし、対応する組換えタンパク質を用いた直接的ELISAによって単離した(図2Aおよび2B)。予めコーティングしたPD−L1−Fc組換えタンパク質を、レスキューされたファージを含有する上清を用いて1時間ブロットし、0.1%のTween−20を含有するPBSで3回洗浄した。結合したファージを、HRP標識抗M13抗体(Roche)によって検出し、シグナル発生のために、TMB基質を使用した。OD450読み取り値を記録した。ポジティブバインダーを単離し、重鎖および軽鎖の配列および多様性を確認するために配列決定を行った。PD−L1に対して特異的な重鎖および軽鎖の可変領域は、配列番号3から配列番号6で記述され、この場合、配列番号3は、PD−L1クローン6の軽鎖であり、配列番号4は、PD−L1クローン6の重鎖の可変領域であり、配列番号5は、PD−L1クローン32の軽鎖であり、配列番号6は、PD−L1クローン32の重鎖の可変領域である。図2Aおよび2Bに示されるように、いくつかのクローンを単離し、それらは、ネガティブコントロールと比較して、対応する抗原に対して特異的に認識されることが知られている。
T細胞活性化における機能性を有する特異的バインダーの迅速なスクリーニングを容易にするために、ELISAによるPD−L1に対するポジティブバインダーの重鎖および軽鎖を増幅し、消化し、IgG4定常領域(配列番号7)を保持するAPBio専用IgG発現ベクター中へサブクローニングした。配列検証後、プラスミドを調製し、抗体発現のために、293フェクチントランスフェクション試薬(Invitrogen)によってHEK293細胞中へとトランスフェクトした。4日間の培養後、無血清培地中に分泌された抗体を、タンパク質Gクロマトグラフィーによって、培養上清からアフィニティー精製した。次いで、精製された抗体を濃縮し、その後、PBS緩衝液において透析した。透析したタンパク質の最終濃度を、NanoDrop2000分光光度計によって特定し、図3に示されるように、還元試薬を用いてまたは用いずに、SDS−PAGEによって純度および完全性を特定した。精製された様々な抗体リードの完全性は、基準抗体MPDL3280Aと同様に、HEK293細胞において正常である。
直接的コーティングされた設定でのヒトPD−L1−Fcに対するELISA結合キャラクタリゼーションのために、PD−L1に対する精製された抗体リード(抗PD−L1抗体リード)を適用した。図4は、抗PD−L1抗体に対するELISA結合結果を示した。PD−L1特異的抗体の場合、ほとんどのリードは、基準抗体(Ref Ab、MPDL3280A、Roche)に対して、同様のまたはより良い結合活性を示した。
結合活性を特定して、PD−L1を発現したHEK293細胞を用いて結合活性を特定、比較するために、フローサイトメトリに対しても、精製された抗体リード(抗PD−L1抗体リード)を適用した。図5は、安定発現されたPD−L1のHEK293細胞を用いたFACSによって示される、対応する抗体リードの結合活性を示している。
抗体リードは、結合選択性を示し、PD−1へのPD−L1の結合を遮断する能力について本発明の抗PD−L1抗体リードを評価するために親和性アッセイを使用した。
当該PD−1シグナル伝達経路は、中程度のTCR/CD28共刺激シグナルを阻害し、その場合、T細胞増殖を減少することなく、サイトカイン産生がまず減少する。当該TCR/CD28共刺激シグナルが弱まると、当該PD−1経路が支配的となり、増殖の減少に伴い、サイトカイン産生が著しく減少する。したがって、本発明のヒト抗体のPD−L1との相互作用の阻害を介した当該PD−1の阻害がT細胞活性化を高めることを検証するために、混合リンパ球反応(MLR)を実施する。
二重特異性は、VEGF阻害ドメインと融合させたIgGベースとして設計されるため、当該抗PD−L1抗体クローン6は、IgG形式であるように割り当てられ、その一方で、VEGF受容体におけるVEGF結合性ドメインD1およびD2は、抗PD−L1クローン6抗体におけるFc領域のC末端において融合される。Fcアイソタイプまたは遺伝子改変Fcは、哺乳動物細胞における当該二重特異性抗体の有効性または産生を向上させるために重要であるため、結果として、2つの異なるFcアイソタイプであるIgG4(配列番号7)および遺伝子改変IgG1(eIgG1、抗体依存性細胞媒介型細胞傷害(ADCC)の低下型(reduction)、配列番号8)を、二重特異性構築に使用した。VID(配列番号9)と融合した二重特異性抗PD−L1抗体Fcの構築を図8に表した。正確な折り畳みを確実にし、立体障害を最小にするために、短いフレキシブルなペプチドリンカー((GGGGS)3(配列番号10)を、例えば、Fc領域(配列番号4)の抗PD−L1抗体重鎖C末端と、VIDのN末端モジュールとの間に位置した。IgG4およびeIGg1に対する抗PD−L1−VID重鎖のコード配列を、配列番号12および13に示した。構築された抗体Fc融合タンパク質を、シグナルペプチド(配列番号11)によってリードし、哺乳動物細胞によって発現させ、ならびにトランスフェクトされた細胞培養上清から一段階タンパク質Gクロマトグラフィーによって精製した。図9に示されるように、一段階精製プロセスにおいて95%を超える純度を得ることができ、それは、精製された融合タンパク質が正しい分子量(Mw=220kDa)を有することを示す。低い純度または回収率は、化学、製造、および品質管理(CMC)生産における当該二重特異性抗体または融合タンパク質の製造にとって主要な問題であるため、したがって、二重特異性抗体の純度を評価するために、両方の精製された二重特異性抗体を、高速液体クロマトグラフィー(HPLC)によるサイズ溶離カラム(SEC)にも適用した(図10Aおよび10B)。両方の二重特異性抗体抗、PD−L1−VID/IgG4(図10A)または抗PD−L1−VID/eIgG1(図10B)のどちらかは、SEC−HPLC分析において、99%純度より高い純度であることが明らかとなった。それは、当該形式が、CMC開発において容易に処理することができ、ならびに、さらなる開発において上出来の率を提供することを意味する。
PD−L1、またはVEGF−AのスプライシングバリアントであるVEGF165に対する当該二重特異性抗体の結合親和性を測定するために、直接結合酵素結合免疫吸着測定法(ELISA)を使用した。組換えVEGF捕捉タンパク質(VEGFR−Fc)および親抗PD−L1抗体クローン6を、それぞれ、PD−L1およびVEGFに対するポジティブコントロール1として使用した。VEGFR−Fcであるアフリベルセプトは、加齢黄斑変性症(AMD)を治療するための治療用途のために遺伝子改変された可溶性VEGF受容体であり、現在、食品医薬品局(FDA)によって承認されている。VEGFR−Fcは、ヒトIgG1のFc領域に融合されたVEGFR2の第3のIg様ドメイン(D3)に融合されたVEGFR1の第2のIg様ドメイン(D2)を含む(Holash et al.,2002)。
本発明における二重特異性抗体の機能性を試験するために、ヒト臍帯静脈内皮細胞(HUVEC)増殖アッセイを実施した。ポジティブコントロールとして、上記において説明したVEGFR−Fcを使用した。100μLのコーティング溶液(再蒸留水中における1%のゼラチン)を、96ウェルELISAプレートの各ウェルに加え、当該プレートを、37℃で2時間または一晩インキュベートした。当該ウェルを、PBS緩衝液で2回洗浄した。内皮細胞増殖培地における3500のHUVEC細胞を各ウェルに加え、当該プレートを37℃で一晩インキュベートした。示された試料を、30nMの最高タンパク質濃度となるように、アッセイ緩衝液(培地−199 1×アール塩、10%のウシ胎仔血清、10mMのHEPES、1×抗生物質/抗真菌薬)で希釈した。当該試料を、VEGF165(8ng/mL)と混合し、当該混合物を、室温で一晩インキュベートした。次いで、当該ウェルを200μLのPBSで洗浄した。100μLのVEGF165/試料混合物を各ウェルに加え、当該プレートを、5%のCO2において37℃で72時間インキュベートした。インキュベーション後、10μLのMTS検出試薬(蒸留したPBS中における3−(4,5−ジメチルチアゾール−2−イル)−5−(3−カルボキシメトキシフェニル)−2−(4−スルホフェニル)−2H−テトラゾリウム)+フェナジンメトサルフェート)を各ウェルに加え、当該プレートを37℃で2.5時間インキュベートした。各ウェルのODを、ELISAプレートリーダ(Bio−Tek)を使用して、490nmの吸収波長において測定した。吸光度を、試験試料のタンパク質濃度に対してプロットし、当該細胞増殖が50%阻害される濃度(IC50)を特定した。細胞増殖の阻害(IC50)が、試験された本発明の融合タンパク質に対して0.1070nMから0.1233nMの間であることを特定した。二重特異性抗体の1つである抗PD−L1−VID/eIgG1は、別の二重特異性抗体である抗PD−L1−VID/IgG4よりも良好に阻害することが明らかとなった(図12、0.1070nM対0.1233nM)。抗PD−L1−VID/eIgG1のIC50は、ポジティブコントロールであるVEGFR−Fcと同様に良好である(0.1072nM)。
PD−1とPD−L1の間の相互作用の阻害による、T細胞活性化の増強における二重特異性抗体のアンタゴニスト機能性を特定するため。二重特異性抗体リードである抗PD−L1−VID/eIgG1抗体を、上記において説明したMLRに適用した。次いで、抗体処理の3日後または5日後に、IL−2およびIFN−γ産生を記録した。T細胞活性化増進におけるアンタゴニスト機能性を比較するために、単一または二重特異性抗体を等しいモルにおいて適用し、アイソタイプIgGをネガティブコントロールとして使用した。図13Aおよび13Bに示されたデータのように、当該抗PD−L1−VID/eIgG1抗体は、処理の3日後に著しいIL2誘導を示し、処理の5日後に低下した。サイトカイン産生のプロファイルは、基準抗体であるMPDL3280Aと非常に類似性が高い。その一方で、IFN−γ産生も、処理の3日後または5日後に、当該二重特異性抗体リード処理において上方調節され、蓄積される。この示された当該抗PD−L1−VID/eIgG1二重特異性抗体は、本発明におけるいかなる活性の損失もなく、基準抗体と同様に、T細胞活性化におけるアンタゴニスト機能性も有する。
精製した抗PD−L1−VID/eIgG1を、示された異なる種に由来する血清(15μg/mL)と共に、水浴において37でインキュベートした。当該精製した二重特異性抗体を含有する血清試料を、14日目まで、異なる時点において採取した。当該血清試料中の当該二重特異性抗体の濃度は、下記のようなサンドイッチELISAアッセイを使用して特定されるであろう。VEGF165(1μg/mL)で予めコーティングしたウェルを、滴定した濃度の精製した抗PD−L1−VID/eIgG1二重特異性抗体と共にインキュベートして、当該血清(新鮮な調製物、0日)中のAb濃度を計算するための標準曲線とした。検出のため、異なる時点から採取した試料を、予めコーティングしたVEGF165ウェルにも適用した。PBS中における0.1%のTween−20で洗浄した後、インタクトであり、かつ結合したAbを、ビオチン化PD−L1−FcおよびHRP標識ストレプトアビジンで検出し、その後、顕色処理した。外挿法によって当該血清中のAbの濃度を計算し、次いで、Abの半減期を、図14A、14B、および14Cに示されるようにプロットした。抗PD−L1−VID/eIgG1の半減期は、カニクイザル(8.6日)、マウス(9.2日)、またはヒト血清(11.9日)のいずれも、8日よりも長い。当該長い半減期は、将来、動物研究または臨床試験での少ない投与頻度における二重特異性抗体の使用の融通性を提供することが可能であろう。
二重特異性抗体における当該PD−L1の齧歯動物交差反応性の欠如は、当該抗体の抗ヒト腫瘍効力の評価に対する標準的なマウス同系またはヒト異種移植腫瘍モデルの使用を妨げた。したがって、ベージュ(Bg)変異欠如マウスのTおよびBリンパ球および機能性NK細胞を有するSCID−Bgマウス(CB.17/Icr.Cg PkrdcscidLystbg/CrI)を使用して、新規のhuPBL−SCID−Bg異種腫瘍マウスモデルを作製した。当該二重特異性抗体の抗ヒト腫瘍効力を、下記において説明するようにこのモデルを使用して評価した。
下記の実施例は、ヒトPD−L1およびVEGFを標的とする治療目的にとって好適なモノクローナル抗体の作製について説明するものである。複合体、ヒト抗ヒトPD−L1およびVEGF中和ドメインは、それぞれ、抗PD−L1抗体クローン6および、ヒトVEGF受容体由来のVEGF捕捉ドメインから作製した。ヒトV領域配列のセグメントは、無関係のヒト抗体(生殖系列および非生殖系列)配列データベースを供給源とした。
本発明によって提供されるある特定の抗体は、ヒトPD−L1に結合するFabから独自に作製した。当該Fabは、対応するFc融合タンパク質(PD−L1−Fc)および対応するヒトタンパク質(PD−L1)を発現する細胞における交互パンニング(alternating panning)に従って、ファージディスプレイライブラリであるOmniMabファージミドライブラリから選択した。直接的ELISAスクリーニング後、当該ポジティブクローンを、重鎖および軽鎖に対して配列決定した。これらのFabは、PD−L1に対して、「OM−PD−L1−6」および「OM−PD−L1−32」などと命名されるものを含んでいた。本出願において開示されるPD−L1抗体PD−L1−クローン6、およびPD−L1−クローン32は、HEK293細胞またはCHO−S細胞における「OM−PD−L1−6」および「OM−PD−L1−32」から作製した。ならびに二重特異性抗体標的化PD−L1およびVEGFを、同時に、抗PD−L1−VID(VEGF阻害ドメイン)抗体として設計した。所定のFabの軽鎖可変領域および重鎖可変領域のアミノ酸配列は、それぞれ、当該軽鎖可変領域および重鎖可変領域の当該アミノ酸配列と同一である。
抗PD−L1−VID二重特異性抗体を、図8に示されるように構築し、HEK293細胞またはCHO−S細胞において発現させた。二重特異性抗体を含む培地を、プロテインGクロマトグラフィーによって、培養上清からアフィニティー精製した。精製された抗体を濃縮し、その後、PBS緩衝液において透析し、図9に示されるようにSDS−PAGEによって分析した。PD−L1またはVEGF165への精製された融合タンパク質の直接結合をELISAにおいて試験するために、100ng/ウェルの組換えPD−L1を、96ウェルELISAプレートにおいてコーティングした。様々な濃度の精製された抗PD−L1−VID Abを各ウェルに加え、1時間インキュベートした。洗浄後、抗Fabまたは抗FcHRPコンジュゲート(Jackson Immunochemicals)の1:5000希釈を各ウェルに加え、さらに1時間インキュベートした。最終洗浄後、TMB基質(Invitrogen Inc.)を加え、450nmにおけるOD吸光度を測定した。当該データを、GraphPad Prism 5を使用してS字形曲線フィッティングによって解析し、EC50を計算した。
抗PD−L1−VID Ab抗体結合特異性を試験するために、安定PD−L1発現293細胞、IFN−γ刺激A549、またはWiDr(結腸直腸腺癌)を、氷上で1時間かけて1μg/mLの抗PD−L1−VID Ab抗体で染色し、その後、1×PBSで3回洗浄した。結合した抗体融合タンパク質を、Alexa−488標識ヤギ抗ヒトIgG(H+L)で検出し、その後、FACS分析を行った。当該試験のネガティブコントロールとして、アイソタイプ抗体を使用した。結果は、抗PD−L1−VID Abが、抗PD−L1単独と比較して、その抗原結合特異性を維持していることを示した。
T細胞応答性を調節する当該抗PD−L1−VID Abの能力を測定するために、精製されたT細胞が、数日間かけて、GM−CSFおよびIL−4において単球を培養することによって調製した同種異系樹状細胞と共に培養されるであろう。市販のELISAキットを使用して、それぞれ、IL−2およびIFN−γを測定するために、3日目および5日目での上清の収集を可能にするように、平行プレートを準備した。Genentech/Rocheのヒト化抗PD−L1であるMPDL3280Aが、自前で作製され、ポジティブコントロールとして使用されるであろう。図13Aおよび13Bに示されたデータのように、当該IL−2およびIFN−γ産生は、抗体処理の3日後または5日後に、基準抗体と同様に、二重特異性抗体処理において高度に上方調節される。それは、当該二重特異性抗体が、依然として、T細胞活性化を促進するために、T細胞と樹状細胞との間のPD−1/PD−L1相互作用を阻害する能力を有することを明らかにした。
マウスPD−L1との当該PD−L1抗体における検出可能な交差反応の欠如、およびヒト免疫細胞の存在に対する要件は、当該二重特異性抗体のインビボ機能性評価のためのモデルの開発を必要とした。重症複合免疫不全(SCID)変異およびIL−2受容体共通ガンマ鎖(一般的に、NSGと呼ばれる)における欠損を有するNOD遺伝的背景のマウスは、多くのヒト末梢血白血球(huPBL)の移植を支持することができ、少なくとも30日間、移植を維持することができる(King et al.,2008)。huPBL−NSGモデルとしても知られるこのマウスモデルを、ヒト免疫細胞に対する当該抗体のインビボ全身性投与の機能効果を評価するために使用した。
huPBL−NSGマウスにおいて異種移植モデルを確立するために、PD−L1ポジティブヒト前立腺癌細胞株PC−3(ATCC#CRL−1435)または腎癌細胞株A498(ATCC(登録商標)HTB−44(商標))を使用することができる。腫瘍形成のために、3×106のPC−3細胞(またはA498細胞)/マウスが、上記において説明したhuPBL−NSGマウスに、皮下において注入されるであろう。腫瘍増殖に対する阻害効果を評価するために、腫瘍細胞移植の14日後に、0.1〜3mg/kgの異なる濃度の抗PD−L1−VID/eIgG1抗体、基準抗体、またはアイソタイプ抗体が、毎週2回、4週間にわたってマウスに静脈内投与されるであろう。腫瘍増殖は、Fox Chase SCID(登録商標)Beigeマウスモデルにおいて説明したように、最長5週間にわたって毎週2回測定されるであろう。
10mg/kgから40mg/kgの二官能性タンパク質である抗PD−L1−VID/eIgG1が、皮下注入または静脈内注入によってマウスまたはサルに投与されるであろう。注入後、最長15日間まで、異なる時点において、血清試料が採取されるであろう。当該血清試料中のFc融合タンパク質の濃度が、サンドイッチELISAアッセイを使用して特定されるであろう。
本発明は、抗体に関する。より詳細には、本発明は、癌療法のための抗体に関する。
ヒトにおけるリンパ球の2つの主要なタイプは、T(胸腺由来)およびB(骨髄由来)である。これらの細胞は、リンパ球発達経路のために関与した骨髄および胎児肝臓の造血幹細胞に由来する。これらの幹細胞の後代は、Bリンパ球またはTリンパ球に成熟するための分岐経路に従う。ヒトBリンパ球発達は、完全に骨髄内において生じる。その一方で、T細胞は、骨髄を去って血流によって胸腺まで移動する未成熟前駆体から発達し、そこで、それらは、増殖し、成熟Tリンパ球へと分化する。
T細胞は、最も豊富に存在し(血液リンパ球の約75%)、強力な免疫キラー細胞である。抗腫瘍性免疫応答におけるエフェクターT細胞の役割は、インビトロ研究および、腫瘍のいくつかのタイプにおけるCD8+T細胞の高浸潤が好ましい臨床予後と相関関係を有するという観察(Fridman et al.,2012)によって強く支持される。エフェクターナイーブT細胞の活性化は、少なくとも3つの相補シグナル:(i)共受容体(CD4またはCD8)の支援によるTCR−CD3/Ag−MHC相互作用;(ii)CD80またはCD86などの共刺激性分子のCD28、CD40/CD40Lへの結合;(iii)サイトカインなどのアクセサリ分子を必要とする。
免疫チェックポイントは、通常、付随的な組織損傷を最小限に抑えるため、自己寛容性を維持し、末梢組織における生理学的応答の持続期間および振幅を調節するために、免疫系、具体的にはT細胞媒介性免疫を張り巡らせるリガンド−受容体相互作用によって開始される、阻害性および刺激性経路のグループを意味する(Pardoll,2012)。腫瘍細胞は、免疫抵抗性の主要メカニズムとして、ある特定のチェックポイント経路を選出する。例えば、プログラム細胞死タンパク質1リガンドのPD−L1は、一般的に、ヒト癌の腫瘍細胞表面において上方制御される。PD−L1と、腫瘍浸透リンパ球(TIL)、特にT細胞、において発現される、その受容体であるPD−1との相互作用は、局所的T細胞媒介性応答を阻害することにより、免疫監視から逃れる(Liang et al.,2006;Sznol and Chen,2013)。したがって、癌細胞での免疫抑制性シグナルの阻害、またはT細胞の直接的アゴニスト刺激は、強い持続性の抗腫瘍免疫応答を生じるおよび/または誘導する。最近の臨床研究は、抗体を介した、または可溶性リガンドまたは受容体によって調節される、免疫チェックポイントタンパク質の遮断が、治療的抗腫瘍免疫の活性化に対する最も有望なアプローチであることを強く示唆した(Topalian et al.,2014)。現在、抗PD−1および抗CTLA−4(細胞傷害性Tリンパ球関連抗原−4)抗体が、黒色腫などの疾患を治療するために、FDAによって承認されている。
血管新生、すなわち、既存の血管からの新しい血管の形成は、胎児発達および組織修復にかかわる正常で不可欠なプロセスである。当該プロセスは、血管新生因子および抗血管新生因子の両方によって高度に制御され、ならびに、それは、内皮細胞の遊走および増殖、血管の成熟化および再構成、ならびに細胞外マトリクスの分解を伴う。それは、正常な増殖および発達において重要なプロセスであるが、血管新生は、腫瘍増殖においても重要な役割を果たす。腫瘍は、増殖するために血管供給を必要とし、リガンドの血管内皮増殖因子(VEGF)ファミリーのメンバーを含む血管新生促進増殖因子の発現を介してこれを達成することができる(Hicklin and Ellis,2005)。VEGFおよび他の内皮増殖因子が、内皮細胞上のそれらの受容体に結合する場合、これらの細胞内のシグナルが開始され、それは、新しい血管の成長および生存を促進する。VEGF特異抗体(アバスチン)、可溶性VEGF受容体(アフリベルセプト)、またはVEGFチロシンキナーゼ活性の阻害剤(スニチニブ)によるVEGF活性の阻害は、腫瘍または血管新生タイプの障害(例えば、AMDなど)を治療するために使用されてきた戦略である。
エフェクター免疫細胞に腫瘍細胞を効果的に再標的化させるために二重特異性抗体を使用するアイデアは、1980年代に現れた(Karpovsky et al.,1984;Perez et al.,1985;Staerz et al.,1985)。二重特異性の足場は、一般的に、Fc断片、IgG様分子、および小さい組換え二重特異性形式(それらの大部分は、単鎖可変断片(scFv)に由来する)の有無に基づいて、異なる薬物動力学特性を有する2つの主要な群に分類される。抗体断片は、それらのコンパクトなサイズにより、通常、IgG様分子より効率的に腫瘍に浸透するが、この利点は、それらの全体的腫瘍取り込みおよび滞留時間を制限する短い血清半減期(数時間)によって緩和される(Goldenberg et al.,2007)。対照的に、新生児Fc受容体に結合するFc断片の存在は、IgG様形式に対して長い血清半減期(>10日)を提供し、これは、腫瘍取り込みおよび滞留に有利に働くが、腫瘍浸透を制限する。
実施例が添付の図面に示されている本発明の本実施形態について、以下において詳細に言及されるであろう。同じまたは同様の部分を参照するために、図面および説明において、可能な限り、同じ参照番号が使用される。
PD−L1に対する治療抗体の生成のために、OmniMabファージミドライブラリによる選択を実施した。当該ファージミドライブラリは、百を超える健康なドナーのB細胞のコレクションからAP Biosciences Inc.(APBio Inc.)によって生成される。パンニングの第一ラウンドのためのファージを、Hyperphage(Μ13Κ07ΔρΙΙΙ、Progen、ハイデルベルク、ドイツ)によって調製した。PD−L1特異的バインダー選択およびOmniMabライブラリからの単離に対して、PD−L1に対する固相パンニングおよび細胞パンニングを適用した。固相パンニングは、第一ラウンド選択において組換えヒトPD−L1−Fc(APBio Inc.)を使用して実施し、次いで、追加の第二ラウンドの濃縮に対して、PD−L1を発現したHEK293細胞を使用した。3つのラウンド選択の後、当該特異的PD−L1バインダーをスクリーニングし、対応する組換えタンパク質を用いた直接的ELISAによって単離した(図2Aおよび2B)。予めコーティングしたPD−L1−Fc組換えタンパク質を、レスキューされたファージを含有する上清を用いて1時間ブロットし、0.1%のTween−20を含有するPBSで3回洗浄した。結合したファージを、HRP標識抗M13抗体(Roche)によって検出し、シグナル発生のために、TMB基質を使用した。OD450読み取り値を記録した。ポジティブバインダーを単離し、重鎖および軽鎖の配列および多様性を確認するために配列決定を行った。PD−L1に対して特異的な重鎖および軽鎖の可変領域は、配列番号3から配列番号6で記述され、この場合、配列番号3は、PD−L1クローン6の軽鎖であり、配列番号4は、PD−L1クローン6の重鎖の可変領域であり、配列番号5は、PD−L1クローン32の軽鎖であり、配列番号6は、PD−L1クローン32の重鎖の可変領域である。図2Aおよび2Bに示されるように、いくつかのクローンを単離し、それらは、ネガティブコントロールと比較して、対応する抗原に対して特異的に認識されることが知られている。
T細胞活性化における機能性を有する特異的バインダーの迅速なスクリーニングを容易にするために、ELISAによるPD−L1に対するポジティブバインダーの重鎖および軽鎖を増幅し、消化し、IgG4定常領域(配列番号7)を保持するAPBio専用IgG発現ベクター中へサブクローニングした。配列検証後、プラスミドを調製し、抗体発現のために、293フェクチントランスフェクション試薬(Invitrogen)によってHEK293細胞中へとトランスフェクトした。4日間の培養後、無血清培地中に分泌された抗体を、タンパク質Gクロマトグラフィーによって、培養上清からアフィニティー精製した。次いで、精製された抗体を濃縮し、その後、PBS緩衝液において透析した。透析したタンパク質の最終濃度を、NanoDrop2000分光光度計によって特定し、図3に示されるように、還元試薬を用いずに、SDS−PAGEによって純度および完全性を特定した。精製された様々な抗体リードの完全性は、基準抗体MPDL3280Aと同様に、HEK293細胞において正常である。
直接的コーティングされた設定でのヒトPD−L1−Fcに対するELISA結合キャラクタリゼーションのために、PD−L1に対する精製された抗体リード(抗PD−L1抗体リード)を適用した。図4は、抗PD−L1抗体に対するELISA結合結果を示した。PD−L1特異的抗体の場合、ほとんどのリードは、基準抗体(Ref Ab、MPDL3280A、Roche)に対して、同様のまたはより良い結合活性を示した。
結合活性を特定して、PD−L1を発現したHEK293細胞を用いて結合活性を特定、比較するために、フローサイトメトリに対しても、精製された抗体リード(抗PD−L1抗体リード)を適用した。図5は、安定発現されたPD−L1のHEK293細胞を用いたFACSによって示される、対応する抗体リードの結合活性を示している。
抗体リードは、結合選択性を示し、PD−1へのPD−L1の結合を遮断する能力について本発明の抗PD−L1抗体リードを評価するために親和性アッセイを使用した。
当該PD−1シグナル伝達経路は、中程度のTCR/CD28共刺激シグナルを阻害し、その場合、T細胞増殖を減少することなく、サイトカイン産生がまず減少する。当該TCR/CD28共刺激シグナルが弱まると、当該PD−1経路が支配的となり、増殖の減少に伴い、サイトカイン産生が著しく減少する。したがって、本発明のヒト抗体のPD−L1との相互作用の阻害を介した当該PD−1の阻害がT細胞活性化を高めることを検証するために、混合リンパ球反応(MLR)を実施する。
二重特異性は、VEGF阻害ドメインと融合させたIgGベースとして設計されるため、当該抗PD−L1抗体クローン6は、IgG形式であるように割り当てられ、その一方で、VEGF受容体におけるVEGF結合性ドメインD1およびD2は、抗PD−L1クローン6抗体におけるFc領域のC末端において融合される。Fcアイソタイプまたは遺伝子改変Fcは、哺乳動物細胞における当該二重特異性抗体の有効性または産生を向上させるために重要であるため、結果として、2つの異なるFcアイソタイプであるIgG4(配列番号7)および遺伝子改変IgG1(eIgG1、抗体依存性細胞媒介型細胞傷害(ADCC)の低下型(reduction)、配列番号8)を、二重特異性構築に使用した。VID(配列番号9)と融合した二重特異性抗PD−L1抗体Fcの構築を図8に表した。正確な折り畳みを確実にし、立体障害を最小にするために、短いフレキシブルなペプチドリンカー((GGGGS)3(配列番号10)を、例えば、Fc領域(配列番号4)の抗PD−L1抗体重鎖C末端と、VIDのN末端モジュールとの間に位置した。IgG4およびeIGg1に対する抗PD−L1−VID重鎖のコード配列を、配列番号12および13に示した。構築された抗体Fc融合タンパク質を、シグナルペプチド(配列番号11)によってリードし、哺乳動物細胞によって発現させ、ならびにトランスフェクトされた細胞培養上清から一段階タンパク質Gクロマトグラフィーによって精製した。図9に示されるように、一段階精製プロセスにおいて95%を超える純度を得ることができ、それは、精製された融合タンパク質が正しい分子量(Mw=220kDa)を有することを示す。低い純度または回収率は、化学、製造、および品質管理(CMC)生産における当該二重特異性抗体または融合タンパク質の製造にとって主要な問題であるため、したがって、二重特異性抗体の純度を評価するために、両方の精製された二重特異性抗体を、高速液体クロマトグラフィー(HPLC)によるサイズ溶離カラム(SEC)にも適用した(図10Aおよび10B)。両方の二重特異性抗体抗、PD−L1−VID/IgG4(図10A)または抗PD−L1−VID/eIgG1(図10B)のどちらかは、SEC−HPLC分析において、99%純度より高い純度であることが明らかとなった。それは、当該形式が、CMC開発において容易に処理することができ、ならびに、さらなる開発において上出来の率を提供することを意味する。
PD−L1、またはVEGF−AのスプライシングバリアントであるVEGF165に対する当該二重特異性抗体の結合親和性を測定するために、直接結合酵素結合免疫吸着測定法(ELISA)を使用した。組換えVEGF捕捉タンパク質(VEGFR−Fc)および親抗PD−L1抗体クローン6を、それぞれ、PD−L1およびVEGFに対するポジティブコントロール1として使用した。VEGFR−Fcであるアフリベルセプトは、加齢黄斑変性症(AMD)を治療するための治療用途のために遺伝子改変された可溶性VEGF受容体であり、現在、食品医薬品局(FDA)によって承認されている。VEGFR−Fcは、ヒトIgG1のFc領域に融合されたVEGFR2の第3のIg様ドメイン(D3)に融合されたVEGFR1の第2のIg様ドメイン(D2)を含む(Holash et al.,2002)。
本発明における二重特異性抗体の機能性を試験するために、ヒト臍帯静脈内皮細胞(HUVEC)増殖アッセイを実施した。ポジティブコントロールとして、上記において説明したVEGFR−Fcを使用した。100μLのコーティング溶液(再蒸留水中における1%のゼラチン)を、96ウェルELISAプレートの各ウェルに加え、当該プレートを、37℃で2時間または一晩インキュベートした。当該ウェルを、PBS緩衝液で2回洗浄した。内皮細胞増殖培地における3500のHUVEC細胞を各ウェルに加え、当該プレートを37℃で一晩インキュベートした。示された試料を、30nMの最高タンパク質濃度となるように、アッセイ緩衝液(培地−199 1×アール塩、10%のウシ胎仔血清、10mMのHEPES、1×抗生物質/抗真菌薬)で希釈した。当該試料を、VEGF165(8ng/mL)と混合し、当該混合物を、室温で一晩インキュベートした。次いで、当該ウェルを200μLのPBSで洗浄した。100μLのVEGF165/試料混合物を各ウェルに加え、当該プレートを、5%のCO2において37℃で72時間インキュベートした。インキュベーション後、10μLのMTS検出試薬(蒸留したPBS中における3−(4,5−ジメチルチアゾール−2−イル)−5−(3−カルボキシメトキシフェニル)−2−(4−スルホフェニル)−2H−テトラゾリウム)+フェナジンメトサルフェート)を各ウェルに加え、当該プレートを37℃で2.5時間インキュベートした。各ウェルのODを、ELISAプレートリーダ(Bio−Tek)を使用して、490nmの吸収波長において測定した。吸光度を、試験試料のタンパク質濃度に対してプロットし、当該細胞増殖が50%阻害される濃度(IC50)を特定した。細胞増殖の阻害(IC50)が、試験された本発明の融合タンパク質に対して0.1070nMから0.1233nMの間であることを特定した。二重特異性抗体の1つである抗PD−L1−VID/eIgG1は、別の二重特異性抗体である抗PD−L1−VID/IgG4よりも良好に阻害することが明らかとなった(図12、0.1070nM対0.1233nM)。抗PD−L1−VID/eIgG1のIC50は、ポジティブコントロールであるVEGFR−Fcと同様に良好である(0.1072nM)。
PD−1とPD−L1の間の相互作用の阻害による、T細胞活性化の増強における二重特異性抗体のアンタゴニスト機能性を特定するため。二重特異性抗体リードである抗PD−L1−VID/eIgG1抗体を、上記において説明したMLRに適用した。次いで、抗体処理の3日後または5日後に、IL−2およびIFN−γ産生を記録した。T細胞活性化増進におけるアンタゴニスト機能性を比較するために、単一または二重特異性抗体を等しいモルにおいて適用し、アイソタイプIgGをネガティブコントロールとして使用した。図13Aおよび13Bに示されたデータのように、当該抗PD−L1−VID/eIgG1抗体は、処理の3日後に著しいIL2誘導を示し、処理の5日後に低下した。サイトカイン産生のプロファイルは、基準抗体であるMPDL3280Aと非常に類似性が高い。その一方で、IFN−γ産生も、処理の3日後または5日後に、当該二重特異性抗体リード処理において上方調節され、蓄積される。この示された当該抗PD−L1−VID/eIgG1二重特異性抗体は、本発明におけるいかなる活性の損失もなく、基準抗体と同様に、T細胞活性化におけるアンタゴニスト機能性も有する。
精製した抗PD−L1−VID/eIgG1を、示された異なる種に由来する血清(15μg/mL)と共に、水浴において37でインキュベートした。当該精製した二重特異性抗体を含有する血清試料を、14日目まで、異なる時点において採取した。当該血清試料中の当該二重特異性抗体の濃度は、下記のようなサンドイッチELISAアッセイを使用して特定されるであろう。VEGF165(1μg/mL)で予めコーティングしたウェルを、滴定した濃度の精製した抗PD−L1−VID/eIgG1二重特異性抗体と共にインキュベートして、当該血清(新鮮な調製物、0日)中のAb濃度を計算するための標準曲線とした。検出のため、異なる時点から採取した試料を、予めコーティングしたVEGF165ウェルにも適用した。PBS中における0.1%のTween−20で洗浄した後、インタクトであり、かつ結合したAbを、ビオチン化PD−L1−FcおよびHRP標識ストレプトアビジンで検出し、その後、顕色処理した。外挿法によって当該血清中のAbの濃度を計算し、次いで、Abの半減期を、図14A、14B、および14Cに示されるようにプロットした。抗PD−L1−VID/eIgG1の半減期は、カニクイザル(8.6日)、マウス(9.2日)、またはヒト血清(11.9日)のいずれも、8日よりも長い。当該長い半減期は、将来、動物研究または臨床試験での少ない投与頻度における二重特異性抗体の使用の融通性を提供することが可能であろう。
二重特異性抗体における当該PD−L1の齧歯動物交差反応性の欠如は、当該抗体の抗ヒト腫瘍効力の評価に対する標準的なマウス同系またはヒト異種移植腫瘍モデルの使用を妨げた。したがって、ベージュ(Bg)変異欠如マウスのTおよびBリンパ球および機能性NK細胞を有するSCID−Bgマウス(CB.17/Icr.Cg PkrdcscidLystbg/CrI)を使用して、新規のhuPBL−SCID−Bg異種腫瘍マウスモデルを作製した。当該二重特異性抗体の抗ヒト腫瘍効力を、下記において説明するようにこのモデルを使用して評価した。
下記の実施例は、ヒトPD−L1およびVEGFを標的とする治療目的にとって好適なモノクローナル抗体の作製について説明するものである。複合体、ヒト抗ヒトPD−L1およびVEGF中和ドメインは、それぞれ、抗PD−L1抗体クローン6および、ヒトVEGF受容体由来のVEGF捕捉ドメインから作製した。ヒトV領域配列のセグメントは、無関係のヒト抗体(生殖系列および非生殖系列)配列データベースを供給源とした。
本発明によって提供されるある特定の抗体は、ヒトPD−L1に結合するFabから独自に作製した。当該Fabは、対応するFc融合タンパク質(PD−L1−Fc)および対応するヒトタンパク質(PD−L1)を発現する細胞における交互パンニング(alternating panning)に従って、ファージディスプレイライブラリであるOmniMabファージミドライブラリから選択した。直接的ELISAスクリーニング後、当該ポジティブクローンを、重鎖および軽鎖に対して配列決定した。これらのFabは、PD−L1に対して、「OM−PD−L1−6」および「OM−PD−L1−32」などと命名されるものを含んでいた。本出願において開示されるPD−L1抗体PD−L1−クローン6、およびPD−L1−クローン32は、HEK293細胞またはCHO−S細胞における「OM−PD−L1−6」および「OM−PD−L1−32」から作製した。ならびに二重特異性抗体標的化PD−L1およびVEGFを、同時に、抗PD−L1−VID(VEGF阻害ドメイン)抗体として設計した。所定のFabの軽鎖可変領域および重鎖可変領域のアミノ酸配列は、それぞれ、当該軽鎖可変領域および重鎖可変領域の当該アミノ酸配列と同一である。
抗PD−L1−VID二重特異性抗体を、図8に示されるように構築し、HEK293細胞またはCHO−S細胞において発現させた。二重特異性抗体を含む培地を、プロテインGクロマトグラフィーによって、培養上清からアフィニティー精製した。精製された抗体を濃縮し、その後、PBS緩衝液において透析し、図9に示されるようにSDS−PAGEによって分析した。PD−L1またはVEGF165への精製された融合タンパク質の直接結合をELISAにおいて試験するために、100ng/ウェルの組換えPD−L1を、96ウェルELISAプレートにおいてコーティングした。様々な濃度の精製された抗PD−L1−VID Abを各ウェルに加え、1時間インキュベートした。洗浄後、抗Fabまたは抗FcHRPコンジュゲート(Jackson Immunochemicals)の1:5000希釈を各ウェルに加え、さらに1時間インキュベートした。最終洗浄後、TMB基質(Invitrogen Inc.)を加え、450nmにおけるOD吸光度を測定した。当該データを、GraphPad Prism 5を使用してS字形曲線フィッティングによって解析し、EC50を計算した。
T細胞応答性を調節する当該抗PD−L1−VID Abの能力を測定するために、精製されたT細胞が、数日間かけて、GM−CSFおよびIL−4において単球を培養することによって調製した同種異系樹状細胞と共に培養されるであろう。市販のELISAキットを使用して、それぞれ、IL−2およびIFN−γを測定するために、3日目および5日目での上清の収集を可能にするように、平行プレートを準備した。Genentech/Rocheのヒト化抗PD−L1であるMPDL3280Aが、自前で作製され、ポジティブコントロールとして使用されるであろう。図13Aおよび13Bに示されたデータのように、当該IL−2およびIFN−γ産生は、抗体処理の3日後または5日後に、基準抗体と同様に、二重特異性抗体処理において高度に上方調節される。それは、当該二重特異性抗体が、依然として、T細胞活性化を促進するために、T細胞と樹状細胞との間のPD−1/PD−L1相互作用を阻害する能力を有することを明らかにした。
マウスPD−L1との当該PD−L1抗体における検出可能な交差反応の欠如、およびヒト免疫細胞の存在に対する要件は、当該二重特異性抗体のインビボ機能性評価のためのモデルの開発を必要とした。重症複合免疫不全(SCID)変異およびIL−2受容体共通ガンマ鎖(一般的に、NSGと呼ばれる)における欠損を有するNOD遺伝的背景のマウスは、多くのヒト末梢血白血球(huPBL)の移植を支持することができ、少なくとも30日間、移植を維持することができる(King et al.,2008)。huPBL−NSGモデルとしても知られるこのマウスモデルを、ヒト免疫細胞に対する当該抗体のインビボ全身性投与の機能効果を評価するために使用した。
huPBL−NSGマウスにおいて異種移植モデルを確立するために、PD−L1ポジティブヒト前立腺癌細胞株PC−3(ATCC#CRL−1435)または腎癌細胞株A498(ATCC(登録商標)HTB−44(商標))を使用することができる。腫瘍形成のために、3×106のPC−3細胞(またはA498細胞)/マウスが、上記において説明したhuPBL−NSGマウスに、皮下において注入されるであろう。腫瘍増殖に対する阻害効果を評価するために、腫瘍細胞移植の14日後に、0.1〜3mg/kgの異なる濃度の抗PD−L1−VID/eIgG1抗体、基準抗体、またはアイソタイプ抗体が、毎週2回、4週間にわたってマウスに静脈内投与されるであろう。腫瘍増殖は、Fox Chase SCID(登録商標)Beigeマウスモデルにおいて説明したように、最長5週間にわたって毎週2回測定されるであろう。
10mg/kgから40mg/kgの二官能性タンパク質である抗PD−L1−VID/eIgG1が、皮下注入または静脈内注入によってマウスまたはサルに投与されるであろう。注入後、最長15日間まで、異なる時点において、血清試料が採取されるであろう。当該血清試料中のFc融合タンパク質の濃度が、サンドイッチELISAアッセイを使用して特定されるであろう。
Claims (22)
- 少なくとも1つのポリペプチド鎖を含む二重特異性抗体またはその抗原結合性部分であって、該ポリペプチド鎖は、
細胞表面タンパク質に結合する結合性ドメインと、
血管内皮増殖因子(VEGF)阻害性ドメインと、
を含む、二重特異性抗体またはその抗原結合性部分。 - 前記細胞表面タンパク質は、プログラム細胞死タンパク質1リガンド(PD−L1)、プログラム細胞死タンパク質1(PD−1)、上皮増殖因子受容体(EGFR)、ヒト上皮増殖因子受容体2(HER2)、細胞傷害性Tリンパ細胞関連抗原4(CTLA−4)、リンパ球活性化遺伝子3(LAG3)、BおよびTリンパ球アテニュエーター(BTLA)、OX40(分化クラスター134、CD134)、CD27、CD28、腫瘍壊死因子受容体スーパーファミリーメンバー9(TNFRSF9またはCD137)、誘導性T細胞共刺激因子(ICOSまたはCD278)、CD40、またはこれらの組み合わせを含む、請求項1に記載の二重特異性抗体またはその抗原結合性部分。
- 前記結合性ドメインは、前記PD−L1に結合し、ならびに、
配列番号4および6からなる群より選択されるアミノ酸配列に対して少なくとも約80%の配列相同性のアミノ酸配列を含む重鎖可変ドメインと、
配列番号3のアミノ酸1〜111および配列番号5のアミノ酸1〜110からなる群より選択されるアミノ酸配列に対して少なくとも約80%の配列相同性のアミノ酸配列を含む軽鎖可変ドメインと、
を含む、請求項2に記載の二重特異性抗体またはその抗原結合性部分。 - 前記VEGF阻害性ドメインは、ヒトVEGF受容体1(VEGFR−1)またはヒトVEGF受容体2(VEGFR−2)由来である、請求項1に記載の二重特異性抗体またはその抗原結合性部分。
- 前記VEGF阻害性ドメインは、配列番号1、2、9、14、15、16、17、18、19、20、21、22、23、24、25、26、27、およびその組み合わせからなる群より選択されるアミノ酸配列に対して少なくとも約80%の配列相同性のアミノ酸配列を含む、請求項1に記載の二重特異性抗体またはその抗原結合性部分。
- 前記ポリペプチド鎖がさらに、
Fcドメインと、
該FcドメインのN末端に接続されているFab断片であって、前記結合性ドメインを含むFab断片と
を含み、
前記VEGF阻害性ドメインは、該FcドメインのC末端に接続されている、請求項1に記載の二重特異性抗体またはその抗原結合性部分。 - さらに、前記Fcドメインと前記VEGF阻害性ドメインとの間にリンカーを含む、請求項6に記載の二重特異性抗体またはその抗原結合性部分。
- 前記二重特異性抗体は、配列番号12または13に記述されるアミノ酸配列を含む、請求項7に記載の二重特異性抗体またはその抗原結合性部分。
- 前記二重特異性抗体またはその抗原結合性タンパク質は、1対のポリペプチド鎖を含む、請求項1に記載の二重特異性抗体またはその抗原結合性部分。
- 前記二重特異性抗体は、IgG、IgE、IgM、IgD、IgA、またはIgY抗体である、請求項9に記載の二重特異性抗体またはその抗原結合性部分。
- 前記二重特異性抗体は、IgG抗体である、請求項10に記載の二重特異性抗体またはその抗原結合性部分。
- 前記IgG抗体は、IgG1、IgG2、IgG3、またはIgG4抗体である、請求項11に記載の二重特異性抗体またはその抗原結合性部分。
- 前記IgG1抗体は、IgG1抗体の抗体依存性細胞媒介型細胞傷害の低下型である、請求項12に記載の二重特異性抗体またはその抗原結合性部分。
- 前記二重特異性抗体は、ヒト抗体である、請求項1に記載の二重特異性抗体またはその抗原結合性部分。
- 請求項1に記載の二重特異性抗体またはその抗原結合性断片と、少なくとも1種の薬学的に許容され得る担体とを含む医薬組成物。
- 治療薬と、
PD−L1および/またはVEGF阻害性ドメインに結合する二重特異性抗体またはその抗原結合性断片と、
を含む抗体−薬物コンジュゲートであって、
該治療薬は、リンカーによって、該抗体または該抗原結合性断片に共有結合的にコンジュゲートされている、抗体−薬物コンジュゲート。 - 前記二重特異性抗体または前記抗原結合性断片は、請求項3から選択される、請求項16に記載の抗体−薬物コンジュゲート。
- 癌を治療する方法であって、請求項1に記載の二重特異性抗体またはその抗原結合性部分の有効量を、それを必要とする対象に投与することを含む、方法。
- 前記癌が、前立腺癌、肺癌、非小細胞肺癌(NSCLC)、黒色腫、リンパ腫、乳癌、頭頚部癌、腎細胞腫(RCC)、および卵巣癌からなる群より選択される、請求項18に記載の方法。
- 前記有効量が、0.001μg/kgから250mg/kgである、請求項18に記載の方法。
- 請求項3に記載の抗体または抗原結合性部分をコードする核酸分子。
- 対象における癌診断の方法であって、
(a)対象から体液試料または細胞試料を得ることと、
(b)PD−L1およびVEGFからなる群より選択される癌マーカのパネルの発現を検出することができる1種または複数種の抗体に該試料を接触させることと、
(c)該体液試料または該細胞試料に対する該1種または複数種の抗体の結合を検査することと、
(d)該対象における該癌の存在を特定するために、抗体結合のレベルを測定して正常なコントロールと比較することによって、アッセイにおいて該対象の該癌の状態を評価することと、
を含む、方法。
