Abstract
Description
本明細書で使われる用語「膜融合性膜タンパク質(fusogenic membrane protein)」は、ウイルス由来の膜タンパク質であって、ウイルスが宿主細胞に浸透するために、宿主細胞への付着及び膜融合を誘発するに当たって、核心的な役割を行うタンパク質を意味する。代表的な例としては、水疱性口内炎ウイルス由来の外被糖タンパク質(VSV−G)が存在する。
本発明の一態様によれば、162番目のアミノ酸であるヒスチジンがアルギニンに置換されたVSV−G変異体タンパク質が膜に導入された組換え原形質膜をベースとする小胞体が提供される。
本発明者らは、VSV−G H162R変異体タンパク質を含む組換えエクソソームを製造するために、まず、VSV−Gの162番目(GenBank No.CAC47944に開示されたタンパク質基準178番目)のアミノ酸残基であるヒスチジンがアルギニンに置換されたH162R変異体タンパク質をコードする遺伝子コンストラクトを製造した。具体的に、このために、野生型VSV−Gタンパク質(配列番号1)をコードするポリヌクレオチド(配列番号2)が含まれたプラスミドDNA(pCMV−VSV−G Envelope Vector、RV−110、Cell Biolabs、以下、‘VSV−Gコンストラクト’と略称する)を鋳型として配列番号3に記載されるフォワードプライマー
を用いて位置指定突然変異を行った(太字のコードは、変異アミノ酸であるアルギニンに対応する)。
本発明者らは、前記実施例1から製造されたVSV−G H162Rタンパク質(配列番号5)をコードする遺伝子(配列番号6)が挿入されたpCMV−VSV−G H162Rプラスミドベクター(以下、‘VSV−G H162Rコンストラクト’と略称する、図2A及び図2B)をHEK293T細胞に形質移入させた後、48時間培養した。次に、細胞培養液を回収して、300gで10分間、2,000gで10分、10,000gで30分の順次遠心分離を行った後、0.2μmフィルターを用いて濾過した後、再び150,000gで3時間限外濾過を行って、ペレットを回収した(図3)。
本発明者らは、比較例として162番のアミノ酸が変異されていない野生型VSV−Gタンパク質が発現されるように遺伝子コンストラクトを製造した後、実施例2の方法を用いて野生型VSV−Gタンパク質が膜に存在する組換えエクソソーム(wtVSVG−Exo)を製造した。
前記実施例2から製造された組換えエクソソームに免疫原性細胞死誘導剤であるドキソルビシン(DOX)をロードするために、精製されたエクソソーム(〜1011エクソソーム)を、先に1mL PBS中のDOXと混合する。引き続き、DOX−組換えエクソソーム混合物は、次のような設定で超音波処理する:0.25インチチップがあるModel 505 Sonic Dismembratorを用いて20%振幅、30秒オン/オフ及びサイクル間2分の冷却器を有する3分1回転を合計6回転行い、超音波処理後、Exo−DOX溶液を37℃で60分間培養して、エクソソーム膜を回収する。過量の遊離薬物は、NAP−10 Sephadex G25カラム(GE Healthcare,Buckinghamshire,UK)を使用してサイズ排除クロマトグラフィーで除去する。
本発明者らは、前記実施例2から製造された組換えエクソソームが実際試験管内条件で癌細胞と融合するか否かを確認しようとした。
引き続き、本発明者らは、mVSVG−Exoが癌細胞表面に融合された後、癌細胞同士の融合を促進するか否かを確認しようとした。このために、具体的に、乳癌細胞株である4T1−Luc、リンパ腫細胞株であるEL4−Ova、大腸癌細胞株であるCT26.CL25のそれぞれ1×105個をディープレッド(Deep red)1μMまたはGreen CMFDA 1μMで染色し、前記実験例1に記載された方法通りにmVSVG−Exoを処理して、pH7.4またはpH6.8の条件で融合を行った。次に、35パイ培養皿に播種し、24時間経過後、癌細胞融合の程度(Green CMFDA Signal、Deep red signal double positive cell)をフローサイトメトリーにより調査した。その結果、図7A及び図7Bで確認されるように、4T1−Luc、CT26.CL25の場合、pH7.4よりもpH6.8でmVSVG−Exoによって細胞間融合が有意に促進されることを確認し、EL4−Ovaの場合にも、たとえ有意していないとしても、融合が増加する傾向を示した。
本発明者らは、本発明の一実施形態による融合性エクソソームが大食細胞及び樹状細胞による癌細胞の食細胞作用(phagocytosis)を促進させるかを確認するために、食細胞作用分析を行った。
VSV−Gタンパク質は、TLR4作用剤として知られている。これにより、本発明者らは、本発明の一実施形態による組換えエクソソームが、TLR4経路を通じて樹状細胞の成熟を増進させるか否かを調査した。このために、具体的に、6ウェル培養皿に骨髄由来樹状細胞1×106個を播種し、mVSVG−Exo 500ngまたはCon−Exo 500ngを24時間処理した。次に、細胞を分離して遠心分離し、樹状細胞の成熟を反映する抗CD40抗体及び抗CD86抗体を用いて染色した後、フローサイトメトーターにより樹状細胞の成熟を評価した。実験の結果、図9で確認されるように、mVSVG−Exoによって樹状細胞でのCD40及びCD86発現量が増加することを確認し、これは、mVSVG−Exoが樹状細胞の成熟を促進することができることを意味する。
本発明者らは、前記実験例1〜実験例4の結果から本発明の一実施形態による組換えエクソソームが生体内条件で癌細胞の成長を抑制することができるか否かを調査した。
本発明者らは、前記実験例5で示された本発明の一実施形態による組換えエクソソームの生体内抗癌効果が、エクソソームに導入されたmVSV−Gタンパク質による作用であるかを確認するために、腫瘍細胞表面のVSVGタンパク質の発現レベルを調査した。このために、具体的に、C57BL/6野生型マウスの左側の背中にEL4−Ova癌細胞1×106個を皮下注入して癌を誘発させ、癌細胞注入後、腫瘍サイズが100mm3に到達した時、wtVSVG−Exo 100μg、mVSVG−Exo 100μg、Con−Exo 100μgまたはPBSを腫瘍内に注射を用いて投与した。投与2時間後、癌組織を摘出し、単一細胞化し、抗VSVG抗体を利用した染色を伴ったフローサイトメトリーを行った(図12)。実験の結果、図12で示すように、mVSVG−Exoグループのみで癌細胞膜表面でVSVGが発現されることを確認した。
7−1:樹状細胞に及ぼす影響調査
本発明者らは、本発明の一実施形態による組換えエクソソームが樹状細胞の機能を活性化させることにより、抗癌活性を示すか否かを調査した。このために、具体的に、C57BL/6野生型マウスの左側の背中にEL4−Ova癌細胞1×106個を皮下注入して癌を誘発させ、癌細胞注入後、6日目(癌細胞注入した日を0日目とする)、7日目にwtVSVG−Exo 100μg、mVSVG−Exo 100μg、Con−Exo 100μg、またはPBSを腫瘍内投与した。癌細胞注入後、18日目にマウスを屠殺して癌組織及び腫瘍排液リンパ節を摘出した。次に、樹状細胞の癌抗原表出の程度を分析するために、単一細胞化した腫瘍排液リンパ節を樹状細胞のマーカーである抗CD11c抗体と癌特異的な抗原である卵アルブミン(Ovalbumin)がMHC−1にロードされたものを測定することができる抗H−2kb Ova抗体で染色した後、フローサイトメーターで分析した(図13A)。その結果、図13Aで確認されるように、mVSVG−Exo投与群で最も優れた樹状細胞の癌特異的な抗原表出の程度を示した。また、樹状細胞の成熟程度を分析するために、単一細胞化した腫瘍排液リンパ節を樹状細胞のマーカーである抗CD11c抗体と樹状細胞の成熟の程度を評価することができる抗CD40抗体及び抗CD86抗体でそれぞれ染色した後、フローサイトメーターで分析した(図13B及び図13C)。実験の結果、wtVSVG−ExoとmVSVG−Exoグループいずれも樹状細胞の成熟を促進することができることを確認した。したがって、本発明の一実施形態による組換えエクソソームの樹状細胞機能促進効果は、癌細胞への組換えエクソソームの融合と関係のないVSVG自体の機能によるものであることが分かった。
本発明者らは、本発明の一実施形態による組換えエクソソームがCD8 T細胞の癌組織内への浸潤に及ぼす影響を調査した。このために、具体的に、前記実験例7−1の実験動物から摘出された癌組織をOCT compoundに包埋して冷凍させた後、凍結薄片を製造して、抗CD8抗体を用いて染色した後、蛍光顕微鏡により癌組織内CD8 T細胞の浸潤程度を比較分析した(図13D及び図13E)。その結果、mVSVG−Exo投与群で最も優れたCD8 T細胞の浸潤程度を示した。特に、wtVSVG−Exo投与群の場合、対照群や対照エクソソーム投与群と差が存在せず、本発明の一実施形態による組換えエクソソームのCD8 T細胞の浸潤促進活性は、VSVG自体の機能だけではなく、腫瘍微小環境での癌細胞との融合による効果であることが分かった。
本発明者らは、本発明の一実施形態による組換えエクソソームがCD8 T細胞に対する交差能力があるかを調査した。このために、具体的に、C57BL/6野生型マウスの左側の背中にEL4−Ova癌細胞1×106個を皮下注入して癌を誘発し、癌細胞注入後、6日目(癌細胞注入した日を0日目とする)、7日目にwtVSVG−Exo 100μg、mVSVG−Exo 100μg、Con−Exo 100μg、PBSを腫瘍内に注射を用いて投与した。癌細胞注入後、18日目にマウスを屠殺して癌組織と脾臓組織とを摘出した。大食細胞と樹状細胞との交差減作能力(cross−prime ability)を評価するために、単一細胞化した癌組織でF4/80またはCD11c磁性粒子を用いて樹状細胞(CD11c−陽性細胞)と大食細胞(F4/80−陽性細胞)とを分離した。次に、MHC1にロードされた卵アルブミンを認識することができるOT−1 CD8 T細胞を有しているOT−1形質転換マウスの脾臓からCD8 T細胞カラムを用いてOT−1 CD8 T細胞を分離し、癌組織から分離した樹状細胞または大食細胞及び前記OT−1 CD8 T細胞をそれぞれ1:5の比率で3日間培養培地で共培養した。3日後に培地を回収して、グループ別に抗IFN−γ抗体を利用したELISA分析を行って、INF−γの発現レベルを分析した(図14A)。その結果、他のグループに比べて、mVSVG−Exoを処理したラットの癌組織で摘出した樹状細胞の交差減作能力が有意に向上していることを確認した。
本発明者らは、本発明の一実施形態による組換えエクソソームが癌治療が終わった後、同じ癌が再発する場合、癌抗原を記憶して癌特異的免疫反応を触発させるか否かを調査した。このために、前記実験例7−3の実験動物から摘出された脾臓組織を単一細胞化した後、5×106個を12ウェル培養皿に1ml培養培地と共に播種し、PBSまたは癌特異的な抗原である卵アルブミン10μg/mlで24時間処理した。次に、培地を回収して、グループ別に抗IFN−γ抗体を用いてELISA分析を行うことにより、培地内のIFN−γの発現レベルを分析した(図14B)。その結果、図14Bで確認されるように、wtVSVG−Exo投与群とmVSVG−Exo投与群とで有意な癌抗原特異的免疫反応が増加し、特に、mVSVG−Exo投与群で最も優れた癌抗原特異的免疫反応が観察された。
本発明者らは、本発明の一実施形態による組換えエクソソームがどのような免疫細胞に依存的であるかを調査しようとした。まず、T細胞免疫が欠けているヌードマウスでも、食細胞の貪食作用の亢進による抗癌効果が示されるかを確認するために、EL4−Ova癌細胞1×106個をヌードマウスの左側の背中に皮下注入して癌を誘発させ、癌細胞注入後、6日目(癌細胞注入した日を0日目とする)、7日目にmVSVG−Exo 100μg、Con−Exo 100μgまたはPBSを腫瘍内に注射を用いて投与した。3日間隔で癌のサイズを測定し、癌細胞注入後、18日目にマウスを屠殺して癌組織を摘出して重量を測定した(図15A)。その結果、図15Aで確認されるように、T細胞が存在したC57BL/6マウスで示されたmVSVG−Exoの抗癌効果がヌードマウスでは消えることを確認し、それを通じて本発明の一実施形態による組換えエクソソームの抗癌効果は、T細胞免疫に依存的であることが分かる。T細胞免疫が欠けているヌードマウスでも、食細胞の貪食作用の亢進による抗癌効果が示されるかを確認するために、EL4−Ova癌細胞1×106個をヌードマウスの左側の背中に皮下注入して癌を誘発させ、癌細胞注入後、5日目(癌細胞注入した日を0日目とする)、6日目、7日目、8日目にmVSVG−Exo 100μg、Con−Exo 100μgまたはPBSを腫瘍内に注射を用いて投与した(他の実験に比べて、2倍多い量である)。3日間隔で癌のサイズを測定し、癌細胞注入後、17日目にマウスを屠殺して癌組織を摘出して重量を測定した(図15B)。その結果、図15Bで確認されるように、既存の薬物投与量を2倍に上げ、さらに癌が小さな時点で実験を進行した時、mVSVG−Exoの抗癌効果がヌードマウスでも示されることを確認した。これは、本発明の組換えエクソソームがCD8 T細胞の浸潤能の増加以外にも、食細胞の貪食作用を促進することにより、先天性免疫反応を誘導するためであると推定される。
Claims (30)
- 162番目のアミノ酸であるヒスチジンがアルギニンに置換されたVSV−G変異体タンパク質が膜に導入された組換え原形質膜をベースとする小胞体。
- エクソソーム、細胞外小胞または細胞由来ナノベジクルである請求項1に記載の組換え原形質膜をベースとする小胞体。
- 前記VSV−G変異体タンパク質をコードするポリヌクレオチドを含む遺伝子コンストラクトに形質転換されて、前記VSV−G変異体タンパク質を過発現する哺乳動物細胞から分離及び精製されたものである請求項1に記載の組換え原形質膜をベースとする小胞体。
- 162番目のアミノ酸であるヒスチジンがアルギニンに置換されたVSV−G変異体タンパク質が膜に導入され、内部に1つまたは2つ以上の免疫原性細胞死誘導剤が取り込まれた組換え原形質膜をベースとする小胞体。
- 前記免疫原性細胞死誘導剤は、アントラサイクリン系抗癌剤、タキサン系抗癌剤、抗EGFR抗体、BKチャネル作用剤、ボルテゾミブ、強心性配糖体、シクロホスファミド系抗癌剤、GADD34/PP1阻害剤、LV−tSMAC、麻疹ウイルス、ブレオマイシン、ミトキサントロンまたはオキサリプラチンである請求項4に記載の組換え原形質膜をベースとする小胞体。
- 請求項1〜請求項5のいずれか一項に記載の組換え原形質膜をベースとする小胞体を有効成分として含む癌治療用薬学的組成物。
- 1つまたは2つ以上の抗癌化合物をさらに含む請求項6に記載の癌治療用薬学的組成物。
- 前記抗癌化合物は、免疫原性細胞死誘導剤または免疫チェックポイント阻害剤である請求項7に記載の癌治療用薬学的組成物。
- 前記免疫原性細胞死誘導剤は、アントラサイクリン系抗癌剤、タキサン系抗癌剤、抗EGFR抗体、BKチャネル作用剤、ボルテゾミブ、強心性配糖体、シクロホスファミド系抗癌剤、GADD34/PP1阻害剤、LV−tSMAC、麻疹ウイルス、ブレオマイシン、ミトキサントロンまたはオキサリプラチンである請求項8に記載の癌治療用薬学的組成物。
- 前記アントラサイクリン系抗癌剤は、ダウノルビシン、ドキソルビシン、エピルビシン、イダルビシン、ピクサントロン、サバルビシン、またはバルビシンである請求項9に記載の癌治療用薬学的組成物。
- 前記タキサン系抗癌剤は、パクリタキセルまたはドセタキセルである請求項9に記載の癌治療用薬学的組成物。
- 前記抗EGFR抗体は、セツキシマブである請求項9に記載の癌治療用薬学的組成物。
- ウイルス由来の膜融合性膜タンパク質が膜に導入された組換え原形質膜をベースとする小胞体を有効成分として含む癌治療用薬学的組成物。
- 前記原形質膜をベースとする小胞体は、エクソソーム、細胞外小胞または細胞由来ナノベジクルである請求項13に記載の癌治療用薬学的組成物。
- 前記ウイルス由来の膜融合性膜タンパク質は、水疱性口内炎ウイルスVSV−Gタンパク質、テナガザル白血病ウイルスGALV.fus、インフルエンザウイルスヘマグルチニン、呼吸器細胞融合ウイルスFタンパク質、ヒト免疫不全ウイルスgp120またはgp41、フラビウイルスEタンパク質、アルファウイルスE1タンパク質、バキュロウイルスgp64、C型肝炎ウイルスgp31またはgp70、麻疹ウイルスHタンパク質またはFタンパク質、またはエボラウイルスgp1またはgp2である請求項13に記載の癌治療用組成物。
- 1つまたは2つ以上の抗癌化合物をさらに含む請求項12〜請求項15のいずれか一項に記載の癌治療用薬学的組成物。
- 前記抗癌化合物は、免疫原性細胞死誘導剤または免疫チェックポイント阻害剤である請求項16に記載の癌治療用薬学的組成物。
- 前記免疫原性細胞死誘導剤は、アントラサイクリン系抗癌剤、タキサン系抗癌剤、抗EGFR抗体、BKチャネル作用剤、ボルテゾミブ、強心性配糖体、シクロホスファミド系抗癌剤、GADD34/PP1阻害剤、LV−tSMAC、麻疹ウイルス、ブレオマイシン、ミトキサントロンまたはオキサリプラチンである請求項17に記載の癌治療用薬学的組成物。
- 前記アントラサイクリン系抗癌剤は、ダウノルビシン、ドキソルビシン、エピルビシン、イダルビシン、ピクサントロン、サバルビシン、またはバルビシンである請求項18に記載の癌治療用薬学的組成物。
- 前記タキサン系抗癌剤は、パクリタキセルまたはドセタキセルである請求項18に記載の癌治療用薬学的組成物。
- 前記抗EGFR抗体は、セツキシマブである請求項18に記載の癌治療用薬学的組成物。
- 前記免疫チェックポイント阻害剤は、PD−1/PD−L1相互作用阻害剤またはCTLA−4/B7−1/B7−2相互作用阻害剤である請求項30に記載の癌治療用薬学的組成物。
- 前記PD−1/PD−L1相互作用阻害剤は、PD−1またはPDL1を標的とする抗体または前記抗体の機能性断片または一本鎖をベースとする抗体類似体である請求項17に記載の癌治療用薬学的組成物。
- 前記PD−1またはPDL1を標的とする抗体は、ペムブロリズマブ、ニボルマブ、アテゾリズマブまたはアベルマブである請求項23に記載の癌治療用薬学的組成物。
- 前記CTLA−4/B7−1/B7−2相互作用阻害剤は、CTLA−4、B7−1またはB7−2を標的とする抗体または前記抗体の機能性断片または一本鎖をベースとする抗体類似体である請求項22に記載の癌治療用薬学的組成物。
- 前記CTLA−4/B7−1/B7−2相互作用阻害剤は、イピリムマブである請求項25に記載の癌治療用薬学的組成物。
- 前記一本鎖をベースとする抗体類似体は、scFv、sdAb、ダイアボディ(diabody)、モノボディ(monobody)、可変性リンパ球受容体(VLR)、ナノボディ(nanobody)またはラクダ科重鎖断片(VHH)である請求項23または請求項25に記載の癌治療用薬学的組成物。
- 前記抗癌化合物は、前記組換えエクソソームの内部に封入された請求項16に記載の癌治療用薬学的組成物。
- 治療的に有効な量の請求項6に記載の癌治療用薬学的組成物を癌にかかった個体に投与する段階を含む前記個体の癌治療方法。
- 治療的に有効な量の請求項13に記載の癌治療用薬学的組成物を癌にかかった個体に投与する段階を含む前記個体の癌治療方法。
