ホスフェート誘導体を使用する場合、治療活性化合物は、脳および血液中において、増大した量で見られる。理論に縛られることなく、活性治療用化合物は、膜結合腸アルカリホスファターゼ(IAP)によってホスフェート誘導体から放出され、故に、消化管上皮に近接し、ここで、血流中に直接移されると想定される。おそらく、化合物は、主として単分散形態で放出され、次いでこれが、消化管から血液へ吸い上げられ得る。ホスフェート誘導体の薬物動態学的特徴は、マウスおよびラットにおいて、脳および血液中における活性化合物の治療的に有用なレベルにつながる。

すべての出発材料および溶媒は、ABCR、Acros、Alfa Aesar、Fluorochem、Sigma−AldrichまたはMerckから購入され、別段の注記がない限り、そのまま使用された。
化合物sery433は、公開されたプロトコール(C.E.Muller、「Prodrug approaches for enhancing the bioavailability of drugs with low solubility」、Chem.Biodiversity 6、2071〜2083(2009);EP2133355A1 2009)に従って調製した。リン酸ジ−tert−ブチルカリウム(35g、141mmol)、K2HPO4・3H2O(127g、557mmol)、n−Bu4NHSO4(4g、11mmol)、H2O(125ml)およびtBuOMe(170ml)の混合物に、tBuOMe(35ml)中のクロロ硫酸クロロメチル(35g、212mmol)の溶液を、継続的に激しく撹拌しながら、0℃にて25分間で滴下添加した。添加が完了した後、撹拌を室温で2時間にわたって続けた(内部温度が30℃を超えた場合には、反応混合物を冷却した)。反応混合物をH2O(350ml)およびtBuOMe(200ml)でクエンチし、有機相を分離し、1M K2HPO4水溶液(200ml)、水(200ml)、ブライン(50ml)で洗浄し、Na2SO4で乾燥させた。硫酸ナトリウムを濾過した後、n−Bu3N(3ml)を溶液に添加し、溶液を減圧下で濃縮して、生成物を油(34.4g、94%)として提供した。冷凍庫(−21℃)内での貯蔵中に安定性を増大させるために、追加分のn−Bu3N(3mL)を生成物に添加した。
DMSO(200ml)中の、anle138b(1)(25g、72.8mmol)、Cs2CO3(35.6g、109mmol)の混合物に、ジ−tert−ブチルクロロメチルホスフェート(28.1g、109mmol)を一度に添加した。室温で5時間にわたって撹拌した後、反応混合物を水(1200ml)で希釈し、ジエチルエーテル(500+200+100ml)で抽出した。合わせた有機画分を、水(500ml)、ブライン(100ml)で洗浄し、Na2SO4で乾燥させた。硫酸ナトリウムを濾過した後、溶液を減圧下で濃縮して、生成物を油(合計49.5g、40.8gのsery432、82%、比2:3を持つ2つの異性体の混合物)として提供した。得られた生成物は、1H NMRスペクトルに基づき、10.5%のsery430および7.1%のBu3Nも含有する。生成物をさらに精製することなく次のステップにおいて使用した。Sery432は、比2:3の異性体の混合物である(1H NMR)。
DCM(400ml)中のsery432(34.4g、60.9mmol)の冷却溶液に、TFA(23.5ml)を、継続的に激しく撹拌しながら0℃にて5分間で添加した。2時間後、追加分のTFA(23ml)を添加し、反応混合物を0℃で8時間にわたって撹拌した。冷トルエン(300ml)による希釈後、反応混合物を0℃にて減圧下で濃縮し、残留物を冷トルエン(300ml)と混合し、0℃で再度濃縮した(DCMを50〜100mbarで蒸発させ、TFAおよびトルエンを高真空ローター(rotor)エバポレーターで蒸発させた)。得られた混合物を冷アセトニトリル(300ml)で希釈し、0℃で1時間にわたって撹拌した。白色沈殿物を濾過し、減圧下で乾燥させて、酸形態のsery433(合計31.0g、29.0gの酸形態のsery433および2gのアセトニトリル)を提供した。酸形態の粗製sery433に、1M NaOH水溶液(129ml、129mmol、2当量)および水(80ml)を添加し、得られた溶液を濾過し(Milliporeエクスプレスプラスフィルター)、濾液を凍結乾燥して、二ナトリウム塩sery433(30.0g、99%、比2:3を持つ2つの異性体の混合物)を白色粉末として提供した。
DMSO(5ml)中の、anle253b(500mg、1.46mmol)、Cs2CO3(620mg、1.9mmol)の混合物に、ジ−tert−ブチルクロロメチルホスフェート(525mg、1.9mmol)を一度に添加した。室温で15時間の撹拌後、反応の完全性が薄層クロマトグラフィー(TLC)(SiO2、ヘキサン:EtOAc=2:1、Rf遊離体0.33、Rf生成物0.18)によって示された。反応混合物を水(30ml)で希釈し、酢酸エチル(2×15ml)で抽出した。合わせた抽出物を、水(10ml)、ブライン(10ml)で洗浄し、Na2SO4で乾燥させ、減圧下で濃縮して、生成物を油(1.08g)として提供した。生成物をさらに精製することなく次のステップにおいて使用した。
DCM(10ml)中のanle423a(1.08g)の冷却懸濁液に、TFA(2ml)を、継続的に激しく撹拌しながら0℃にて1分以内に添加し、反応混合物を0℃で8時間にわたって撹拌した。混合物を濾過し(GHP 0.45μm)、トルエン(10ml)で希釈し、20℃にて減圧下で濃縮し、残留物をトルエン(10ml)と混合し、20℃で再度濃縮した(DCMを50〜100mbarで蒸発させ、TFAおよびトルエンを高真空ローターエバポレーターで蒸発させた)。得られたガラス状の粘性残留物を冷アセトン(30ml)で細砕し、0℃で1時間にわたって撹拌した。白色沈殿物を濾過し、アセトン(10mL)で洗浄し、減圧下で乾燥させて、酸形態のanle423b(448mg、0.99mmol、68%、2D−NOESY NMR実験によれば、比anle423ba:anle423bb=3:2を持つ2つの異性体の混合物)を帯黄色粉末として提供した。anle423b(二酸、156mg、0.345mmol)に、水(3mL)および25%NH4OH(12.6M、150μL、1.89mmol)を添加した。混合物を溶解が完了するまで撹拌し、得られた溶液を凍結させ、凍結乾燥して、二アンモニウム塩anle423b(161mg、331μmol、96%、比anle423ba:anle423bb=3:2を持つ2つの異性体の混合物)を帯黄色粉末として提供した。
α−シヌクレインが黒色腫において発現されること(Matsuo YおよびKamitani T、2010、Parkinson’s disease−related protein,alpha−synuclein,in malignant melanoma、PLoS One、5(5):e10481)およびその過剰発現がB16マウス黒色腫の増殖を強化すること(Israeli Eら、2011、α−Synuclein expression selectively affects tumorigenesis in mice modeling Parkinson’s disease、PLoS One、6(5):e19622)が報告されている。しかしながら、母斑および黒色腫において互いに並び合う3つの主要なPD関連遺伝子/タンパク質−α−シヌクレイン/SNCA、LRRK2/PARK8およびパーキン/PARK2−の発現の状態およびレベルに関する科学データは、入手不可であった。したがって、黒色腫発生の異なる段階において、これらの3つの遺伝子/タンパク質のいずれが、どの程度まで発現されるかを決定するために、本発明者らは、遺伝子SNCA、PARK8またはPARK2によってコードされるそれぞれのタンパク質に対する抗体で、母斑>黒色腫進行組織マイクロアレイ(TMA)の同じ組織をプロービングした(Watson−Hurst KおよびBecker D、2006、The role of N−cadherin,MCAM and beta3 integrin in melanoma progression,proliferation,migration and invasion、Cancer Biol Ther、5(10):1375〜1382)。
VGPおよびMGP黒色腫細胞におけるα−シヌクレインタンパク質の発現レベル、細胞内局在および状態
進行性黒色腫を表す腫瘍に由来する細胞株における、α−シヌクレインタンパク質の発現レベルおよび細胞内局在を決定するために、本発明者らは、VGP黒色腫細胞株WM983−A、3つのMGP黒色腫細胞株WM983−B、SK−MEL−5、WM852、および放射状成長期(RGP)/VGPにおける表在拡大型黒色腫から確立されたWM1158黒色腫細胞株のイムノブロット分析を実施した。図2Aに示される通り、同じ患者のVGPおよびMGP黒色腫に由来した細胞株WM983−AおよびWM983−BならびにMGP黒色腫細胞株SK−MEL−5は、高レベルのα−シヌクレインを発現する。比較すると、WM852(MGP)およびWM1158(RGP/VGP)黒色腫細胞株は、より低レベルのα−シヌクレインタンパク質を含有する(図2A)。
高レベルのα−シヌクレインを発現している黒色腫細胞の、α−シヌクレインに影響を及ぼすオリゴマーモジュレーターを用いる処理は、黒色腫細胞形態学において明白な変化を引き起こし、黒色腫細胞増殖を阻害する
進行性黒色腫におけるα−シヌクレインタンパク質の考えられる機能への洞察を得るために、本発明者らは、黒色腫細胞株WM983−A、WM983−B、SK−MEL−5、WM852およびWM1158を、ジフェニル−ピラゾール(DPP)化合物のパネルで処理した。プリオンタンパク質凝集の阻害、続いて、DPP化合物の医薬品化学最適化によって、小分子DIVERSetライブラリー(Chem−Bridge Corp.、San Diego、CA)から同定されたDPP足場は、anle138bおよびanle138cの同定につながった(Wagnerら、Acta Neuropathol.、2013年6月;125(6):795〜813)。これらの2つの化合物のいずれもα−シヌクレインモノマーとは結合しない(Deegら、Biochim Biophys Acta.、2015、1850(9):1884〜90)が、これらはいずれも毒性オリゴマーの、したがって、間接的に原線維の形成も低減させる。
黒色腫細胞のAnle138b処理は、それらの原形質膜を損傷し、それらのミトコンドリア膜電位を破壊し、オートファジーを異常調節する
黒色腫細胞生存率に対する漸増用量のanle138bの効果ならびにそれが細胞に細胞毒性であるか否か、故にそれらの原形質膜完全性を損ねるか否かを決定するために、本発明者らは、48時間で等価用量の化合物を補充して、96時間にわたって漸増用量のanle138bで処理されたWM983−BおよびWM852黒色腫細胞から収集した培養上清の乳酸デヒドロゲナーゼ(LDH)細胞毒性アッセイを実施した。図4Aに示される通り、高レベルα−シヌクレイン発現WM983−B細胞は、おそらく無血清培地中におけるこの用量を上回るその低溶解度により10μM用量のanle138bで最大に到達する強い殺滅を示した。比較すると、anle138b処理に応答して殺滅された細胞の数は、低レベルα−シヌクレイン発現WM852細胞の場合には有意に低かった(図4A)。
高レベルα−シヌクレイン発現ヒト黒色腫異種移植片を担持するヌードマウスに全身的に投与されたAnle138bは、腫瘍に到達し、それらの形態学およびオートファジーに影響を及ぼす
DPP化合物anle138bが、ヌードマウスにおいて皮下腫瘍として成長させたヒトMGP黒色腫細胞に全身的に投与された場合、腫瘍に到達するか否かを決定するために、本発明者らは、下記の研究を実施した。その右下およびその左下背側に高レベルα−シヌクレイン発現(MGP)WM983−Bヒト黒色腫異種移植片を担持するヌードマウスに、anle138bと混合された飼料ペレットを、7日間にわたって受けさせた。その右下およびその左下背側に同じくWM983−B腫瘍を担持する第2のヌードマウスに、anle138bを含有しない同じ種類の飼料ペレットを7日間にわたって受けさせた。マウスを屠殺した後、本発明者らは、死後切除されおよびその後凍結保存された腫瘍について、下記の分析を実施した。
世界で推定700から1000万人に影響を及ぼすPDと、すべての皮膚がん関連死の75%を占める最も侵攻型の皮膚がんである進行性黒色腫との間の疫学的関連の理由である根本的な機序に関する利用可能な情報は未だほとんどなく、あったとしてもわずかである。遺伝的要因がこれら2つの疾患を関連付けることは認められないため、本発明者らは、関連性が、PDの主要調節因子である、α−シヌクレイン、LRRK2またはパーキンであり得る可能性を探究した。
黒色腫細胞株、TMA、GEOデータセット
ヒト黒色腫細胞株WM983−A、WM983−B、WM852およびWM1158を、記述された通りにインビトロで繁殖させた(Becker D、Meier CBおよびHerlyn M、1989、Proliferation of human malignant melanomas is inhibited by antisense oligodeoxynucleotides targeted against basic fibroblast growth factor、EMBO J、8(12):3685〜3691)。CLS Cell Lines Service GmbH(Eppelheim、Germany)から購入したヒト黒色腫細胞株SK−MEL−5を、非必須アミノ酸、ピルビン酸ナトリウムおよび10%ウシ胎児血清(FBS)を補充したイーグル最小必須培地(MEM)中で繁殖させた。
α−シヌクレインタンパク質発現の分析のために、全細胞溶解液を、12%ドデシル硫酸ナトリウム−ポリアクリルアミドゲル(SDS−PAGE)上で分離し、ポリフッ化ビニリデン(PVDF)膜上に移し、リン酸緩衝生理食塩水(PBS)中0.4%パラホルムアルデヒド(PFA)で室温にて30分間の処理によって架橋させ、5%粉乳でブロックし、α−シヌクレインに対する抗体[MJFR1](Abcam)でプロービングし、続いて、西洋ワサビペルオキシダーゼコンジュゲート(HPR)二次抗体およびスーパーシグナルウエストピコ化学発光基質(Thermo Fisher Scientific Inc.、Schwerte、Germany)とともにインキュベーションした。タンパク質ローディングは、抗アクチン[ab1801]抗体(Abcam)での二次プロービングによって決定した。
WM983−B黒色腫細胞を、0.5%トリトンX−100およびプロテアーゼ阻害剤を含有するPBS中で溶解させた。タンパク質溶解物(2.2mg/0.5ml)を0.45μmの遠心分離管フィルターに通して濾過した後、AKTA精製装置10(GE Healthcare Life Sciences、Uppsala、Sweden)と接続されたスーパーロース6 10/300GLカラム(GE Healthcare Bio−Sciences、Pittsburgh、USA)にロードした。収集画分を10分間にわたって95℃の水に入れ、その後、すべての収集画分の全体積を、ドットブロット真空システムによってニトロセルロース膜上にロードした。5%脱脂粉乳を含有するトリス緩衝生理食塩水−ツイーン20(TBST)中でブロックした後、マウスモノクローナル抗α−シヌクレイン[Syn−1]抗体(BD Biosciences、San Jose、CA、USA)で膜をプロービングし、続いて、対応するHRPコンジュゲート二次抗体および化学発光基質(EMD Millipore)とともにインキュベーションした。SEC−フィルター・トラップ・アッセイを2回繰り返した。
anle138bまたはanle138cの10または30mMストック溶液を、DMSO中で作製した。anle138bおよびanle138cはFBSに強く結合するため、10%FBSを含有する組織培養培地中で24時間にわたって平板培養した黒色腫細胞を、無血清培地で3回すすいだ後、無血清培地中の、anle138b、anle138c、または等価体積のDMSOのみを添加した。48時間後、細胞培養培地を除去し、続いて、無血清培養培地中の、等価用量のanle138bもしくはanle138cまたはDMSOのみを添加した。WM1158(RGP/VGP)黒色腫細胞が無血清培養培地中で十分に増殖しないことから、20ng/mlの組換えヒト塩基性線維芽細胞成長因子(bFGF)を、anle138bまたはanle138cを用いるこれらの細胞の処理中に、無血清培養培地に添加した。DMSOのみを受けた、またはanle138bで処理した黒色腫細胞の増殖は、血球計を用いて細胞をカウントすることによって決定した。
WM983−BおよびWM852黒色腫細胞の複製を、35mmの組織培養プレート中、ウェル当たり3×105細胞で平板培養した。24時間後、細胞を無血清培地で3回すすぎ、続いて、漸増用量のanle138bまたはDMSOのみを添加した。48時間後、細胞培養培地を除去し、続いて、無血清培地中の、等価用量のanle138bまたはDMSOのみを添加した。48時間後に収集された培養培地を、遠心分離してペレット化し、細胞残屑を除去した。PHERAstar FSマイクロプレートリーダー(BMG LABTECH GmbH、Ortenberg、Germany)および分光光度吸収測定を使用して、組織培養培地中へのLDH酵素活性の放出を、LDH細胞毒性アッセイキット(Roche Diagnostics Deutschland GmbH、Mannheim、Germany)を用いて決定した。DMSOのみを受けたWM983−BおよびWM852黒色腫細胞からの無血清培養培地上清は、バックグラウンド対照として役立ち、それらの値を引いた。DMSO対照からの細胞を、製造業者の説明書に従って、トリプシン処理し、カウントし、溶解させた。殺滅された黒色腫細胞の数は、各用量のanle138bについて記録されたLDH放出を、公知の数の黒色腫細胞の溶解時に放出されたLDHに対して正規化することによって取得した(較正曲線)。両方の細胞株についての標準曲線は、重ね合わせることができなかった。
細胞形態学。anle138bもしくはanle138cで種々の時間にわたって処理された、またはDMSOのみを受けた黒色腫細胞の位相差またはDIC画像を、浜松ホトニクス株式会社オルカ電荷結合素子(CCD)カメラ付きZeissアキソバート100顕微鏡上の10×NA0.6対物レンズで捕捉した。培養皿当たり無作為な領域で、各時点について少なくとも4つの画像を記録した。
4週齢の雌ヌードマウス(CAnN.Cg−Foxn1nu/Crl)(Charles River Laboratories、Research Models and Services、Germany GmbH、Sulzfeld、Germany)の右下および左下背側に、WM983−B(MGP)ヒト黒色腫細胞(片側1×107細胞)を皮下注射した。腫瘍が任意の方向に2.3から3.0mmのサイズに到達したら、動物のうち1匹の通常の飼料ペレット食を、anle138bと混合された飼料ペレット[1kgの飼料ペレット当たり2gのanle138b](ssniff Spezialdiaten GmbH、Soest、Germany)に置きかえたのに対し、他の動物の通常の飼料ペレット食は、anle138bを含有しない飼料ペレット(ssniff Spezialdiaten GmbH)に置きかえた。anle138を加えたまたは加えない飼料ペレットを受けさせた7日後に、両方の腫瘍担持動物を屠殺し、切除された腫瘍を直ちに液体窒素中で凍結した。これらの研究は、承認LAVESプロトコール(33.19−42502−04−14/1724)下で行った。

高レベルα−シヌクレイン発現ヒト黒色腫細胞の形態学および細胞死に対するDPP化合物の効果
高レベルα−シヌクレイン発現ヒト黒色腫細胞に対するDPP化合物の影響を決定するために、WM983−B黒色腫細胞を、12ウェル組織培養プレート内、10%ウシ胎児血清を含有する細胞培養培地中、二連で平板培養した。24時間後、細胞を無血清培地で3回すすぎ、続いて、各DPP化合物を10μMの用量で含有する無血清培地を添加した。37℃で48時間にわたるDPP化合物処理細胞のインキュベーション後、各DPP化合物を細胞に2回目の10μMの用量で添加した。光学顕微鏡法を使用して、細胞の形態学における変化>細胞培養皿からの細胞の脱離/細胞培養培地中における浮遊>細胞死を、DPP化合物の添加後24時間、48時間、72時間および96時間の時点において、0、+、++、+++、++++の尺度で記録した。DMSOのみを受けたWM983−B黒色腫細胞およびDPP化合物もDMSOも受けなかったWM983−B黒色腫細胞は、対照として役立った。結果が表1に示される。
DPP化合物を、α−シヌクレイン発現WM983−Bヒト転移性黒色腫細胞に添加した。評価されたDPP化合物の活性は、
++++ DPP化合物の添加48時間後における100%の細胞死
+++ DPP化合物の添加96時間後における100%の細胞死
++ DPP化合物の添加96時間後における50〜90%の細胞死
+ DPP化合物の添加96時間後における20〜50%の細胞死
− DPP化合物の添加または培地中の化合物の沈殿96時間後における20%未満の細胞死
であった。
Claims (12)
- 黒色腫の治療または予防において使用するための、
式(E)によって表される化合物
X、YおよびLは、−C(R1)(R2)−、−C(R3)=、−N(R4)−、−N=,−O−および−S−から独立して、方向性を伴わずに選択され、
MおよびZは、
−−−−は、任意選択の二重結合を表し、
R1、R2、R3、R4およびR5は、水素、C1〜4アルキル、−CH2O−P(=O)(OR)(OR)、−C1〜4アルキレン−ハロゲン、−C1〜4アルキレン−OH、−C1〜4アルキレン−C1〜4アルコキシ、−C(O)−C1〜4アルキルおよびC6〜10アリールから独立して選択され、ここで、前記アリール環は、C1〜4アルキルまたはハロゲンによって置換されていてよく、
Halは、F、Cl、BrおよびIから選択され、
RE1は、ヒドロキシ、C1〜6アルコキシおよび−NRE5RE6から選択され、
RE2は、水素、ハロゲン、ヒドロキシ、C1〜6アルコキシおよび−NRE5RE6から選択されるか、または
RE1およびRE2が、隣接する炭素原子と結合している場合、RE1およびRE2は、一緒になって、構造−T−(CRE7RE8)n−V−{ここで、Tは、CRE9RE10、NR*およびOから選択され、Vは、CRE9RE10、NR*およびOから選択される}および二重結合が存在する対応する構造を、代替として、方向性を伴わずに形成することができ、
RE5およびRE6は、水素およびC1〜6アルキルから独立して選択され、
RE7およびRE8は、独立して、HまたはFであり、
RE9およびRE10は、独立して、HまたはFであり、

RA1およびRA2は、水素、ハロゲン、ヒドロキシ、C1〜6アルコキシおよび−NRA5RA6からそれぞれ独立して選択されるか、または
RA1およびRA2の少なくとも一方は、ヒドロキシ、C1〜6アルコキシもしくは−NRA5RA6であることを条件とし、
代替として、RA1およびRA2は、一緒になって、構造−T−(CRE7RE8)n−V−を、方向性を伴わずに形成することができ、
