Abstract

米国では毎年22,000件超の卵巣がん新規症例および14,000件超の死亡例があり(Siegel RL, et al. (2016) CA Cancer J Clin 66(1):7-30(非特許文献1))、推定年間6億ドル超の医療負担が伴う(Dizon D MJ (2010) Gynecol Oncol 116(3)(非特許文献2))。化学療法(例えば、カルボプラチン/シスプラチンおよび/またはパクリタキセル)のような従来の手法では、多くの場合、卵巣がんを治癒することができない。患者のおよそ70%は第一選択の化学療法で寛解に到達せず、寛解を得た患者の40〜50%は3年以内に再発する。

本開示の操作されたMSC (または他の操作された免疫細胞)を培養する段階を含む方法も、本明細書において提供される。MSCを培養する方法は公知である。いくつかの態様において、MSCは増殖培地(例えば、MSCGMヒト間葉幹細胞増殖BULLETKIT(商標)培地(血清含有)、THERAPEAK(商標) MSCGM-CD(商標)間葉幹細胞既知組成培地(無血清)、またはRoosterBio xeno-free MSC培地)中で培養される。

4. エフェクタ分子をコードするヌクレオチド配列に機能的に連結されたプロモーターを含む操作された核酸を含む、パラグラフ1〜3のいずれかの間葉幹細胞または組成物。

本実施例では、個別および組み合わせの免疫療法ペイロードを備えたMSCのインビトロでの特性評価について記述する。がん細胞に対する免疫療法発現性MSCの直接的な抗がん効果を最初に測定する。次に、免疫療法発現性MSCが初代免疫細胞(T細胞に焦点を合わせている)とがん細胞との共培養に及ぼす効果を測定する。免疫療法発現性MSCの免疫刺激特性を、マウスとヒトの両方の細胞系における炎症性バイオマーカーパネルに基づいてランク付けする。がん細胞の死滅化を単独でまたはT細胞と一緒に著しく増強する免疫療法発現性MSCを特定し、インビボ試験に進むための上位候補を選択する。
実施例1からの免疫療法発現性MSCを、がん療法に関連するインビトロモデルを用いその機能的効果について評価する。ヒト卵巣がん細胞(例えば、OVCAR8およびSKOV3)ならびに循環血中PBMCから単離されたヒト免疫細胞を用いて、hITを発現するhMSCを試験する。マウス卵巣がん細胞(例えば、ID8)およびマウス免疫細胞を用いて、mITを発現するmMSCを試験する。
細胞結合アッセイ法を用いて、発現された抗体の活性を検証する。抗体の標的であるCTLA4およびPD1は両方とも、T細胞を負に調節するが、T細胞活性化のさまざまな段階で上方制御される(Boutros C, et al. (2016) Nat Rev Clin Oncol 13(8):473-486; Valsecchi ME (2015) New Engl J Med 373(13):1270-1270)。CTLA4はプライミング段階において短時間上方制御されるが、PD1はT細胞活性化のエフェクタ段階において一貫して発現される(Pardoll DM (2012) Nat Rev Cancer 12(4):252-264; Legat A, et al. (2013) Front Immunol 4:455)。抗CTLA4抗体はT細胞表面のCTLA4に結合し、CTLA4が初期段階でT細胞活性化を遮断するのをブロックし、ヒト抗PD1抗体はPD1に結合し、腫瘍細胞がT細胞活性を阻害するのを防ぐ。
細胞遊走アッセイ法およびCCL21走化性に応答するケモカイン受容体CCR7を発現する単離されたナイーブT細胞を用いて、CCL21ケモカインの機能性を確認する。具体的には、CCL21発現MSCまたは対照MSCをトランスウェルの1区画に添加し、その後、他の区画からの単離ナイーブT細胞によって細胞遊走を評価し、引き続いて遊走したT細胞の数を列挙する(Justus CR, et al. (2014) J Vis Exp (88))。
IL2、IL12、およびIL15の活性を測定する。IL2、IL12、およびIL15に特異的なELISAアッセイ法を用いて、MSC上清中のこれらのサイトカインのレベルを検出する。機能的生物活性アッセイ法では、CTLL-2細胞株を利用してIL2またはIL15を介した増殖を評価し、NKG細胞株を利用してMSC上清によるIL12を介したIFN-ガンマ産生を評価する。LUMINEX(登録商標)技術を用いた多重サイトカインプロファイリングアッセイ法を使って、サイトカイン発現および免疫細胞に及ぼす効果を評価してもよい。
STING経路の活性化を、構成的STING変異体ペイロードで測定する。LUMINEX(登録商標)ビーズを用いて、STING変異体の発現を伴うI型インターフェロン(例えばIFN-アルファ2およびIFN-ベータ)の分泌をMSCにおいてプロファイルする。
卵巣がん細胞の成長および生存性に及ぼすMSCのいずれかの直接的な効果を、インビトロにおいて試験する。例えば、mMSCまたはhMSC候補を、マウス卵巣がん細胞株(ID8)またはヒト卵巣がん細胞株(OVCAR8およびSKOV3)と共培養し、がん細胞傷害性を、十分に特徴付けられた乳酸脱水素酵素(LDH)アッセイ法によって測定する。24時間の共培養後、上清を回収し、プレートリーダーでの特定の吸光度によって後に定量化される酵素反応を介して細胞死に関連するLDHレベルを測定する。さらに、トリパンブルー排除によって生細胞 vs 死細胞を、ならびにアネキシン-Vおよびヨウ化プロピジウム染色を用いたフローサイトメトリー測定により生細胞 vs アポトーシス/死細胞をカウントすることにより、がん細胞数を評価する。
試験により、免疫療法発現性MSCがT細胞のような免疫細胞を刺激して、インビトロで卵巣がん細胞に対する改善された抗がん活性を有しうるかどうかを判定する。具体的には、mMSC-mIT候補をマウス脾細胞およびID8がん細胞株と共培養し、またはhMSC-hIT候補をヒトPBMCおよびOVCAR8またはSKOV3細胞株と共培養する。共培養アッセイ法では、MSC有りでまたは無しで、卵巣がん細胞とともにPBMC/脾細胞を使用し、抗CD3/28ビーズで刺激することを伴う。がん細胞死を評価するため、LDH細胞傷害性測定のような技法の使用、色素標識卵巣がん細胞と非標識エフェクタPBMC/脾細胞との一定の比率での組み合わせ、フローサイトメトリーによる死滅化のアッセイ(Jedema I, et al. (2004) Blood 103(7):2677-2682)、およびアネキシン-Vとヨウ化プロピジウムとを用いたフローサイトメトリーによるアポトーシスの読み出しにより16時間の死滅化アッセイ法を実施する。T細胞の活性化/増殖は、3〜5日でのCFSE細胞分裂および1〜3日でのIFN-ガンマのサイトカイン産生による特異的なアッセイである。
本実施例では、同系卵巣がんモデルにおいて免疫療法ペイロードを発現するMSCのインビボでの特性評価について記述する。免疫療法発現性MSCの抗腫瘍効力は、卵巣がんの同系マウスモデル(マウス免疫系を有するmMSC-mIT)を用いて特徴付けられる。同系卵巣マウスモデルにおいて操作されたMSCの腫瘍ホーミングならびに個々のおよび組み合わせでの免疫療法の発現を測定する。操作されたMSC処置での卵巣腫瘍負荷およびマウス生存性も測定する。本実施例では、卵巣腫瘍 vs 他の身体部位での操作されたMSCのTMEへの選択的ホーミングおよび免疫療法因子の局所的産生が実証されるはずである。本実施例では、免疫療法発現性の操作されたMSCによる腫瘍負荷の有意な低減およびマウス生存性の延長も実証されるはずである。
マウス卵巣上皮表面細胞(MOSE)の自発的形質転換に由来するマウスID8細胞株を用いて同系卵巣腫瘍モデルを作製する(Roby KF, et al. (2000) Carcinogenesis 21(4):585-591)。ID8細胞株にウミシイタケ(Renilla)ルシフェラーゼ(rLuc)を発現するレンチウイルスを感染させ、ホタル(Firefly)ルシフェラーゼ(ffLuc)を発現するMSCに無関係なインビボ生物発光画像化を可能にする。成功裏のrLuc発現は、マウスでの同系卵巣がんモデルを樹立する前にインビトロにおいてID8で確認される。同系モデルの場合、6〜8週齢の間に5×105個のID8細胞をC57BL/6マウスの腹腔に注入する(36, 54)。MSCを、ffLuc発現プラスミドとともに、実施例1のプラスミドを発現するペイロードでヌクレオフェクションする。
本実施例では、ヒト免疫細胞を有するマウスでのヒト卵巣がんの異種移植モデルにおいて免疫療法ペイロードを発現するMSCの効力のインビボでの特性評価について記述する。CD34+細胞移植を介してヒト免疫細胞が移植された免疫不全マウスのヒト卵巣がんモデルにおける操作されたMSC (ヒト化免疫系を備えたhMSC-hIT)の活性を試験する。操作されたMSCのホーミングならびにヒト免疫細胞を有するマウスのヒト異種移植卵巣腫瘍における個々のおよび組み合わせでの免疫療法の発現を測定する。操作されたMSC処置での卵巣腫瘍負荷およびマウス生存性も測定する。本実施例では、マウスでのヒト異種移植卵巣腫瘍 vs 他の身体部位への操作されたMSCのホーミングの上昇および免疫療法因子の局所的産生が実証されるはずである。本実施例では、免疫系組成の変化と相関して免疫療法発現性の操作されたMSCによる腫瘍負荷の有意な低減およびマウス生存性の延長も実証されるはずである。
操作されたMSCのヒト臨床試験への移行を可能にするため、ヒトがんのヒト化マウスモデルにおいてhMSC-hIT構築体を試験する。マウスにおける免疫療法発現性hMSCの効果は、CD34+造血幹細胞(HSC)が移植された免疫不全マウス(NSG)におけるヒト卵巣がん細胞株の異種移植片を用いることによりモデル化される。
以下の実験では、MSCを操作して以下のエフェクタ分子の1つを発現させ、その後、単独でまたは組み合わせで、同所性乳がんマウスモデルに投与した: IFNβ、IFNγ、IL12、IL15、IL36γ、IL7、TRAIL、cGAS、CCL21a、OX40L、CD40L、またはHACv-PD1。いくつかの例では、チェックポイント阻害剤(抗CD40抗体、抗PD1抗体、または抗CTLA-4抗体)を、操作されたMSCでの投与と組み合わせて注射した。
以下の実験では、ネズミMSCが乳がんの同所性マウスモデルにおいて腫瘍にホーミングすることを実証する。ルシフェラーゼ発現4T1乳房腫瘍細胞(5×105)を、雌性BALB/cJマウスの背部脂肪体に同所移植した。5日後、マウスに100万個の蛍光標識(XenoLight DiR (Caliper Life Sciences)による)ネズミBM由来MSC (BM-MSC, 治療用細胞)を腹腔内注射した。MSC注入後1日および7日の時点で、Ami HTライブ動物イメージャ(Spectral Instruments)を用いてMSC局在化を判定するために蛍光分析を使用した。7日目に、Ami HTイメージャを用いて4T1細胞のルシフェラーゼ生物発光レポーターにより腫瘍の局在およびサイズを判定した。図3に示されるように、注射されたMSCは腫瘍部位に共局在化し、これらの細胞が実際に4T1乳房腫瘍の部位にインビボで特異的にホーミングすることを示している。注射されたMSCは1日以内に腫瘍にホーミングし、7日を超えて存続する。対照的に、注射されたMSCは、正常マウスにおいて腫瘍の非存在下では背部にホーミングしない。これらの結果は、抗がん分子、タンパク質、または化合物の送達媒体としてMSCを使用できることを示唆している。
以下の実験は、乳がんの同所性モデルにおいて免疫療法エフェクタ(ペイロード)を発現するMSCのインビボでの効力を実証する。4T1-Neo-Flucマウス乳房腫瘍細胞(Imanis Life Sciences, 細胞5×105個)を、雌性BALB/cJマウス(The Jackson Laboratory)の背部脂肪体に同所移植した。次に、腫瘍移植の5日後にマウスを処置群に無作為化した。マウスは、対照MSC成長培地または異なる免疫療法エフェクタ(ペイロード)を発現する操作されたMSC (細胞2×106個)の腹腔内注射を週1回、2週間受けた。各免疫療法は異なるMSCにより表され、組み合わせ処置のためにMSCを組み合わせた(1:1の比率)。腫瘍の成長をキャリパ測定により1日おきにモニターし、マウスの体重を週2回記録した。最初のMSC処置の14日後にマウスを安楽死させ、さらなる分析のために組織を収集した。
GFP群で上記の実験のいくつかにおいて毒性が観察されたため、用量漸増試験を行って根底にある毒性原因を判定した。本実験により、操作されたMSC細胞発現GFPは毒性を誘発せず(図10A〜10B)、MSC懸濁培地が毒性の主な原因でありうることが分かった。
本実験では、IL12およびCCL21aを発現する操作されたマウスMSCが、腫瘍負荷を、より大きな腫瘍(800 mm3超)から低減できるかどうかを試験した。より大きな腫瘍は小さな腫瘍よりも処置することがいっそう困難であり、本実験はこのエフェクタの組み合わせが腫瘍拡大を低減できることを実証している(図12A〜12B)。
図13Aは、IL-12およびCCL21を発現する操作されたMSCが、腫瘍成長を阻害するのに十分であり、注射によるチェックポイント阻害剤(抗PD-1抗体または抗CTLA-4抗体)の添加が効力を増加させなかったことを示す。
以下の実験では、MSCを操作して以下のエフェクタ分子の1つを発現させ、その後、単独でまたは組み合わせで結腸直腸がんマウスモデルに投与した: IFNβ、IL12、IL15、IL36γ、IL7、CCL21a、HACv-PD1、または41BB。いくつかの例では、チェックポイント阻害剤(抗CD40抗体または抗CTLA-4抗体)を、操作されたMSCとの投与と組み合わせて注射した。
以下の実験は、結腸(結腸直腸)がんの皮下マウスモデルで免疫療法エフェクタ(ペイロード)を発現するMSCのインビボでの効力を実証する。CT26-Neo-Flucマウス結腸がん細胞(Imanis Life Sciences, 5×105個)を雌性BALB/cJマウス(The Jackson Laboratory)の脇腹に皮下注射した。腫瘍移植の7日後に、マウスを次いで以下の処置群に無作為化した: 対照MSC成長培地、操作されたMSC (MSC-12+CCL21a)、抗CD40抗体、抗CTLA4抗体(Bio X細胞)、抗CD40抗体と組み合わせたMSC-12+CCL21a、または抗CTLA4抗体と組み合わせたMSC-12+CCL21a。操作されたMSC (細胞2×106個)を2週間、週1回(0日目および7日目)腹腔内(ip)注射した。抗CD40抗体を0日目および3日目にip (100μg)注射した。抗CTLA4抗体を0日目、3日目、および7日目にip (100μg)注射した。腫瘍の成長をキャリパ測定により1日おきにモニターし、マウスの体重を週2回記録した。最初のMSC処置の11日後にマウスを安楽死させ、腫瘍を収集および秤量した。各処置群における個々のマウスの腫瘍重量を測定し、結果を図16B (左グラフ)の左下に示す。各処置群の平均腫瘍体積を経時的にモニターした(図16B, 右グラフ)。処置群2 (IL-12+CCL21a+抗CTLA4抗体)、4 (IL-12+CCL21a)、および7 (IL-12+CCL21a+抗CD40抗体)は、GFP処置マウスと比較してCT26結腸腫瘍の平均成長を阻害した(図16B, 右グラフ)。各処置群における個々のマウスの腫瘍体積を経時的に測定した場合、同様の結果が観察された(図16A)。したがって、免疫療法を発現するMSCによる組み合わせ処置は、インビボでの結腸がん細胞の成長を阻害した。
注射された抗CD40抗体と組み合わせて、異なる濃度の操作されたMSC-IL12およびCCL21a療法をマウスに2回施与した。2回目の施与後、腫瘍負荷が1500 mm3を超えるまで週2回腫瘍体積をモニターし、マウスを殺処理した。図17Aは、個々の群の腫瘍体積を示す。図17Bの左グラフは、個々の群からのマウス体重および腫瘍体積を経時的に追跡している。図17Bの右グラフは、異なる群の生存プロットを示す。
以下の本実験は、ネズミMSCが結腸がんのマウスモデルにおいて腫瘍にホーミングすることを実証する。簡単な実験プロトコルが図19の左上セクションに提供されている。ルシフェラーゼ発現CT26結腸がん腫瘍細胞(5×105個)を、雌性BALB/cJマウスの右大腿に皮下移植した。4日後、Ami HTライブ動物イメージャ(Spectral Instruments)を用いてCT26細胞のルシフェラーゼレポーターにより腫瘍の局在およびサイズを判定した(図19, 左下パネル, ルシフェラーゼシグナル(腫瘍特異的))。腫瘍移植後5日の時点で、XenoLight DiR (Caliper Life Sciences)で蛍光標識された200万個のマウスBM-MSCを、腹腔内注射により腫瘍担持マウス(腫瘍+)に移植した。MSC注射後1日目および3日目の時点で、XenoLight DiR蛍光標識MSCの局在をAmi HTイメージャを用いて判定した(図19, 右パネル, DiRシグナル(MSC特異的))。注射されたMSCはCT26結腸腫瘍の部位に共局在化した(図19, 左下パネルにおけるMSC注射前のマウスでの腫瘍特異的ルシフェラーゼシグナルの局在と、右側の注射後1日目および3日目の腫瘍+マウスでのMSC特異的DiRシグナルを比較している)。それゆえ、MSCはインビボでCT26結腸腫瘍の部位に特異的にホーミングし、これらの結果は、MSCを抗がん分子、タンパク質、または化合物の送達媒体として使用できることを示す。
図21A〜21Bは、腹腔内空間におけるCT26-LUC (ルシフェラーゼ)腫瘍成長の動態を示す。CT26細胞株を0日目に注射し、3匹のマウスを7日目、10日目、14日目、および18日目に収集して、腫瘍成長の動態を判定した。図21Aの第一列では、腫瘍負荷をモニターするために、IVISイメージャでマウスの体重およびROIを測定している。第二列では、各群における個々のマウスの腫瘍重量および腫瘍ROIをモニターしている。第三列では、腫瘍重量を全身ROIまたは腫瘍ROIと相関させている。図21Bは、腫瘍微小環境をさらによく理解するための18日目の群における3匹のマウスの免疫プロファイルを示す。
皮下マウスモデル
図22Aは、IL-12およびCCL21aを発現する操作されたMSCが結腸直腸がんの皮下マウスモデルにおける腫瘍成長を阻害するが、CCL21aおよびIL-36ガンマまたはIL-7を発現するMSCの組み合わせは腫瘍成長を低減しないことを示すデータを含む。図23A〜23Bは、腫瘍免疫浸潤の統計を含む。PBS、ナイーブMSC、およびMSC-IL12+MSC-CCL21a (コンボ)群から3匹のマウスを選択し、フローサイトメトリーを実行して腫瘍微小環境の免疫プロファイルを調べた。図23Aは、ナイーブMSCを投与した群と比較して、コンボ群における浸潤性CD3およびCD8細胞傷害性T集団の有意な増加を示す。図23Bは、ナイーブMSCで処置した群と比較してコンボ群での顆粒球性骨髄由来サプレッサ細胞(gMDSC)およびマクロファージ集団の有意な低減を示す。
図26Aは、IL-12およびCCL21aまたはCCL21aおよびIFN-βを発現する操作されたMSCが結腸直腸がんの同所性マウスモデルにおいて腫瘍成長を阻害するが、CCL21aおよびs41BBLを発現するMSCの組み合わせでは腫瘍成長が低減されないことを示す。各エフェクタは異なるMSCにより発現され、組み合わせ処置のためにMSCを組み合わせた(1:1の比率)。各チャートは、マウス(n = 6〜8)での4T1乳房腫瘍の成長に及ぼす表示された免疫療法を単独でまたは組み合わせで発現する操作されたMSCの効果を示す。図26Aの各線は個々のマウスを表す。図26Bは、各処置における個々のマウスの腫瘍重量を示す。MSC-IL12 + MSC-CCL21aは、ナイーブMSCを注射したマウスと比較して最良の効力を示す。処置効力がまた、MSC-IFNb + MSC-CCL21aで処置された群において観察された。
図25A〜25Bは、腹腔内および皮下結腸直腸がんマウスモデルにおけるMSC-IL-12+CCL21a療法のデータを含む。レンチウイルス形質導入系統の3つの異なるロットをMSC-IL12およびCCL21aについて試験した(TLOO8-3/4、TL019-01/02、およびTL022-01/02; 各TL番号は1ロットを表す)。図25Aは、MSC-IL12 + MSC-CCL21aの全3ロットが、皮下および腹腔内モデルの両方で腫瘍負荷を低減できることを示す(最初の5つのグラフはSCモデルからのものであり、最後の3つはIPモデルからのものである)。全てのマウスからの腫瘍を11日目に収集した。図25Bは、各群からの平均腫瘍重量を示す。
Claims (39)
- 腫瘍媒介免疫抑制機構を調節する複数のエフェクタ分子を産生するように操作された間葉幹細胞を含む組成物を、腫瘍の体積を低減させるのに有効な量で、腫瘍を有する対象に送達する段階を含む、対象における腫瘍体積を低減させる方法。
- 複数のエフェクタ分子が、サイトカイン、受容体/リガンド、抗体、ヌクレオチド、ペプチド、および酵素から選択される、請求項1に記載の方法。
- 腫瘍が、膀胱腫瘍、脳腫瘍、乳房腫瘍、子宮頸部腫瘍、結腸直腸腫瘍、食道腫瘍、神経膠腫、腎臓腫瘍、肝臓腫瘍、肺腫瘍、黒色腫、卵巣腫瘍、膵臓腫瘍、前立腺腫瘍、皮膚腫瘍、甲状腺腫瘍、および子宮腫瘍から選択される、請求項1または2に記載の方法。
- 前記組成物が、(a) 第1のエフェクタ分子を産生するように操作された第1の間葉幹細胞および(b) 第2のエフェクタ分子を産生するように操作された第2の間葉幹細胞を含む、請求項1〜3のいずれか一項に記載の方法。
- 前記組成物が、第1のエフェクタ分子および第2のエフェクタ分子を産生するように操作された間葉幹細胞を含む、請求項1〜3のいずれか一項に記載の方法。
- 第1のエフェクタ分子がIL-12であり、かつ第2のエフェクタ分子がCCL21である、請求項4または5に記載の方法。
- 第1のエフェクタ分子がIFN-βであり、かつ第2のエフェクタ分子がIFN-γである、請求項4または5に記載の方法。
- 間葉幹細胞が、IL-12、CCL21、IFN-β、IFN-γ、またはこれらの2つもしくはそれ以上の任意の組み合わせを産生するように操作されている、請求項1〜7のいずれか一項に記載の方法。
- 前記組成物がチェックポイント阻害剤をさらに含む、請求項1〜8のいずれか一項に記載の方法。
- チェックポイント阻害剤が、抗PD-1抗体、抗PD-1L抗体、または抗CTLA-4抗体である、請求項9に記載の方法。
- チェックポイント阻害剤が抗CTLA-4抗体である、請求項10に記載の方法。

- 腫瘍の体積が対照と比べて少なくとも50%低減され、任意で対照が非改変間葉幹細胞である、請求項15に記載の方法。
- 腫瘍の体積が対照と比べて少なくとも75%低減され、任意で対照が非改変間葉幹細胞である、請求項16に記載の方法。
- 腫瘍が乳房腫瘍であり、第1のエフェクタ分子がIL-12であり、第2のエフェクタ分子がCCL21であり、かつ任意で前記組成物が抗CTLA-4抗体をさらに含む、請求項4または5に記載の方法。
- 腫瘍が結腸直腸腫瘍であり、第1のエフェクタ分子がIL-12であり、第2のエフェクタ分子がCCL21であり、かつ任意で前記組成物が抗CD40抗体をさらに含む、請求項4または5に記載の方法。
- 腫瘍媒介免疫抑制機構を調節する複数のエフェクタ分子を産生するように操作された間葉幹細胞であって、任意で、エフェクタ分子の1つがIL-12であり、別のエフェクタ分子がCCL21である、前記間葉幹細胞。
- 対象における腫瘍の体積を低減させるのに有効な量で製剤化された、腫瘍媒介免疫抑制機構を調節する複数のエフェクタ分子を産生するように操作された間葉幹細胞を含む、組成物。
- 複数のエフェクタ分子が、サイトカイン、受容体/リガンド、抗体、ヌクレオチド、ペプチド、および酵素から選択される、請求項21に記載の組成物。
- 腫瘍が、膀胱腫瘍、脳腫瘍、乳房腫瘍、子宮頸部腫瘍、結腸直腸腫瘍、食道腫瘍、神経膠腫、腎臓腫瘍、肝臓腫瘍、肺腫瘍、黒色腫、卵巣腫瘍、膵臓腫瘍、前立腺腫瘍、皮膚腫瘍、甲状腺腫瘍、および子宮腫瘍から選択される、請求項21または22に記載の組成物。
- (a) 第1のエフェクタ分子を産生するように操作された第1の間葉幹細胞および(b) 第2のエフェクタ分子を産生するように操作された第2の間葉幹細胞を含む、請求項21〜23のいずれか一項に記載の組成物。
- 第1のエフェクタ分子および第2のエフェクタ分子を産生するように操作された間葉幹細胞を含む、請求項21〜23のいずれか一項に記載の組成物。
- 第1のエフェクタ分子がIL-12であり、かつ第2のエフェクタ分子がCCL21である、請求項24または25に記載の組成物。
- 第1のエフェクタ分子がIFN-βであり、かつ第2のエフェクタ分子がIFN-γである、請求項24または25に記載の組成物。
- 間葉幹細胞が、IL-12、CCL21、IFN-β、IFN-γ、またはこれらの2つもしくはそれ以上の任意の組み合わせを産生するように操作されている、請求項21〜27のいずれか一項に記載の組成物。
- チェックポイント阻害剤をさらに含む、請求項21〜28のいずれか一項に記載の組成物。
- チェックポイント阻害剤が、抗PD-1抗体、抗PD-1L抗体、または抗CTLA-4抗体である、請求項29に記載の組成物。

- 腫瘍が乳房腫瘍であり、第1のエフェクタ分子がIL-12であり、第2のエフェクタ分子がCCL21であり、かつ任意で組成物の間葉幹細胞が、抗CTLA-4抗体を産生するように操作されている、請求項24または25に記載の組成物。
- 腫瘍が結腸直腸腫瘍であり、第1のエフェクタ分子がIL-12であり、第2のエフェクタ分子がCCL21であり、かつ任意で組成物の間葉幹細胞が、抗CD40抗体を産生するように操作されている、請求項24または25に記載の組成物。
