Abstract
Description

発明の背景

本明細書で使用される「活性成分」または「活性物」は、本明細書中に記載される乾燥製品によって被験体に送達されることが望まれる任意の化合物または物質であり得る。活性物は、例えば、マトリクス中に分散されて乾燥されるべき香味剤、顔料、酵素、植物抽出物、植物油、微量栄養素(ビタミン、カロテノイド、アルカロイド(ニコチン、メチルキサンチンなど)フラボノイド)、栄養補助食品、化粧品物質、または医薬物質を含む。2種以上の活性物が存在し得、そして活性物は組成物中で2つ以上の役割を果たし得るかまたは2つ以上の機能を有し得る。いくつかの実施態様では、活性物は生物学的に活性な化合物または物質であり、生理学的または精神物理学的応答(感覚および/または知覚の変化)を生じる。活性物は疎水性または親水性であり得る。
(a)乾燥製品が、示差走査熱量測定により、0.23と0.24との間の水分活性で40.5と80.0℃との間のガラス転移温度、および任意に、0.23と0.24との間の水分活性で51.7と68.8℃との間のガラス転移温度を有する;
(b)乾燥製品が、同等の噴霧乾燥組成物と比較した場合、ヒト官能パネルによって決定されるように艶のある、光沢のある、光彩を放つまたはきらめくように見えるフレークであり;そして60度の角度で1.9より大きい平均光沢測定;
(c)活性成分が、担体および/または残りの不揮発性安定化剤中に埋め込まれた小球中に分散しており、乾燥製品が、乾燥製品の重量に基づいて、0.1-71%、任意に1-42wt.%または20-40wt.%の活性物を含有する;
(d)活性成分が、担体中に埋め込まれた小球中に分散しており、小球が100nmと500ミクロンとの間、大部分は500、100、50、20、15または10ミクロン未満(例えば、0.01-20ミクロン)の直径である(すなわち、電子顕微鏡法によって測定されるように、小球の大部分(数)が500、100、50、20、15または10ミクロン未満(例えば、0.01-20ミクロン)の直径である);
(e)乾燥製品が、気密容器中で過剰の塩化マグネシウム溶液の蒸気(32%相対湿度)に1週間曝露された場合に25質量%未満、あるいは20質量%未満、10質量%未満の水;および気密容器中で飽和硫酸ナトリウム溶液の蒸気(81%相対湿度)に1週間曝露された場合に50質量%未満、あるいは40質量%未満、30質量%未満の水を吸収する;
(f)乾燥製品が、0.07-1000ミクロンの間の厚さ、あるいは10-1000ミクロンの厚さ(例えば、約10と1000ミクロンとの間の厚さ)のフレークである;および
(g)乾燥製品が、平均フレークサイズが0.1-200mmであり、そしてしばしば約0.6mmと20mmとの間の円に外接し得るフレークである。

(f)乾燥製品が、0.07-1000ミクロンの間の厚さ、約1および2500ミクロンの厚さ(例えば、約10と1000ミクロンとの間の厚さ)のフレークである;および
(g)乾燥製品が、0.1-200mmの平均サイズを有し、しばしば約0.6mmと20mmとの間(例えば、約6mmと20mmとの間)の円に外接するフレークである。

工程1:カンナビスオイル(大麻油)を加熱する
工程2:ゴムを水に溶解する
工程3:熱いカンナビスオイルを、せん断混合を用いてアラビアゴム/水混合物にゆっくりと添加する
工程4:得られるせん断混合物を薄膜としてCWDベルトに付与し、そして乾燥する。
注釈:ひまわりレシチンなどの乳化剤を水和アラビアゴムと混合またはカンナビスオイルと直接混合することによって使用し得る->ひまわりレシチンを安定化剤と呼ぶ?
工程5:フレークをエリスリトールとともにすりつぶし、そして密封小包中に置く。
より具体的には、アラビアゴムを水に25%(w/w)で溶解し、マトリクス混合物を形成する。(80% CBD)および0% THCを含むカンナビスオイルをひまわりレシチンおよび植物テルペンとともに加熱し、そしてゴム混合物にそれぞれ6.5%、0.2%、および0.05%(w/w)で添加して100%w/wにした。この混合物を次いで乾燥機に付与し、そして180-185Fで28のベルト速度(約3.8メートル/分)で乾燥した。完成した乾燥フレークは19.7%(w/w)の油および16.3%(w/w)の活性物を含んだ。フレークをエリスリトールとともに3%〜6%(w/w)ですりつぶし、そして小包中に置く。5mg〜40mgの活性物を含む小包を用量/応答閾値化研究のために作製した。
工程1:カンナビスオイルを加熱する
工程2:アラビアゴムを水に溶解する
工程3:熱いカンナビスオイルを、せん断混合を用いてアラビアゴム/水混合物にゆっくりと添加する
工程4:得られるせん断混合物を薄膜としてCWDベルトに付与し、そして乾燥する。
注釈:ひまわりレシチンなどの安定化剤を、水和アラビアゴムと混合またはカンナビスオイルと直接混合することによって使用し得る。
工程5 フレークをすりつぶす+一般的オートミールクッキー処方において使用する。フレークを乾燥小麦粉/塩/ベーキングセッド(baking said)に組み込む。油をコントロールのクッキーにおいて使用される生地に組み込む。
Claims (46)
- (a)活性成分を担体および任意の安定化剤と混合して湿潤混合物を作製すること、ここで該活性成分および担体は該湿潤混合物中、約1:1〜約1:250の比であり;
(b)約60℃〜92℃の範囲のベルト温度を有する薄膜ベルト乾燥機のベルト上に該湿潤混合物を広げること;および
(c)該混合物を乾燥して約7%未満の水分、任意に約2-5%の水分、および約0.15および約0.65の水分活性を有する乾燥製品を製造すること
を含む、任意にフレークの形態の乾燥製品を製造するプロセス。 - 活性成分および担体が湿潤混合物中約1:1と約1:20との間の比である、請求項1のプロセス。
- 乾燥製品が以下の特徴の1つ、2つ以上、または全てを有する、請求項1または2のプロセス:
(a)乾燥製品が示差走査熱量測定によって決定される約0.1〜0.6、任意に約0.23-0.239の水分活性で約58.7-62.6℃のガラス転移温度を有する;
(b)乾燥製品が約0.23-0.2の水分活性で測定した場合に約40.5-80.0℃、任意に約51.7-68.8℃のガラス転移温度を有する;
(c)乾燥製品が、類似の組成物の噴霧乾燥製品と比較した場合、ヒト官能パネルによって決定されるように艶のある、光沢のある、光彩を放つまたはきらめくように見え;および/または60度の角度で約1.9より大きい平均光沢測定を有する;
(d)活性成分が親水性担体中に分散されそして埋め込まれ、そして乾燥製品が少なくとも約0.01wt.%の活性成分、または任意にASTM法E-679を用いて決定される検出閾値の少なくとも約100倍の香味料化合物を含有する;
(e)活性成分が担体中に埋め込まれた小球中に分散されており、そして小球が大部分が約500ミクロン未満の直径である;
(f)乾燥製品が気密容器中で過剰の塩化マグネシウム溶液の蒸気(32%相対湿度)に1週間曝された場合に約10質量%未満の水分;および気密容器中で飽和硫酸ナトリウム溶液の蒸気(81%相対湿度)に1週間曝された場合に約30質量%未満の水分を吸収する;
(g)乾燥製品が約0.07と1000ミクロンとの間の厚さ、任意に約10および約1000ミクロンの厚さであるフレークである;および
(h)乾燥製品が約6mmと約20mmとの間の円に外接しているフレークである。 - 活性成分が親水性担体中に分散されそして埋め込まれ、そして乾燥製品が約1-50wt.%の活性成分を含有する、請求項1〜3のいずれかのプロセス。
- 活性成分がコロイド状分散物または乳化物として担体中に埋め込まれた小球中に分散されており、任意に小球が約100nm〜約500ミクロンの直径である、請求項1〜4のいずれかのプロセス。
- 乾燥製品が約0.07-1000ミクロンの平均厚さを有するフレークであり、および/またはフレークの最大寸法において約0.1-20mmの平均サイズを有する、請求項1〜5のいずれかのプロセス。
- 活性成分が疎水性であり、約3〜9のオクタノール/水分配係数(log(Kow)を有し、担体が任意に親水性ポリマー物質を含有し、そして活性成分が任意に疎水性官能基を有する安定化剤を用いて可溶化されている、先行する請求項のいずれかのプロセス。
- 活性成分が約50℃〜約200℃、約75℃〜約175℃または約100℃〜約160℃の沸点を有し、または活性成分が約20-200℃の沸点または融点を有する、先行する請求項のいずれかのプロセス。
- 担体が、アラビアゴム、デンプン、マルトデキストリン、コーンスターチ、ジャガイモデンプン、糖、ラクトース、デキストロース、たんぱく質、オリゴ糖、イヌリン、乳清たんぱく質、微細藻類たんぱく質、酵母またはそれらの組み合わせを含む、先行する請求項のいずれかのプロセス。
- 担体が、麻たんぱく質、発酵酵母または穀物、ワイン醸造からの使用済み酵母、ビール製造からの使用済み穀物、種子デンプン、全種子、脱脂種子粉末またはそれらの組み合わせを含む、先行する請求項のいずれかのプロセス。
- 活性成分または安定化剤が、ステアリン酸、エタノールまたはテルペンを含む、先行する請求項のいずれかのプロセス。
- テルペンが、リモネン、シトラール、メントール、メントン、ショウノウ、シトロネロール、シトロネラル、ゲラニオール、ゲロール(gerol)、α-ピネン、β-ピネン、シトラール、リナロール、α-テルピネオール、α-フェランドレン、サビネン、チモール、シメン、またはミルセンなどのモノテルペン;β-カリオフィレン、α-フムレン、またはノートカトンなどのセスキテルペン;フィトール、ステビオールグリコシド、レチノール、およびレチナールなどのジテルペン;またはα-カロテン、β-カロテン、γ-カロテン、リコピン、ルテイン、ゼアキサンチン、ネオキサンチン、ビオラキサンチン、フラボキサンチン、α-クリプトキサンチン、およびβ-クリプトキサンチンなどのテトラテルペンである、請求項11のプロセス。
- テルペンが、ジテルペン、トリテルペン、ジテルペングリコシド、またはトリテルペングリコシド、任意にスクワレン、ククルビタシン(かぼちゃ抽出物)、またはステビアまたは羅漢果抽出物からのジ-またはトリテルペングリコシドである、請求項11のプロセス。
- 湿潤混合物が約5-50wt.%のアラビアゴムおよび約0.01-30wt.%のテルペンを含有し、該重量パーセントが湿潤混合物の湿潤重量に基づく、先行する請求項のいずれかのプロセス。
- 湿潤混合物が約10-50wt.%のアラビアゴムおよび約1-30wt.%のテルペンを含有し、該重量パーセントが湿潤混合物の湿潤重量に基づく、先行する請求項のいずれかのプロセス。
- 湿潤混合物が約0.1-30wt.%のステアリン酸および約5-50wt.%のアラビアゴムを含有し、該重量パーセントが湿潤混合物の湿潤重量に基づく、先行する請求項のいずれかのプロセス。
- 活性成分がカンナビノイドを含む、先行する請求項のいずれかのプロセス。
- 活性成分が、生大麻、精製または未精製大麻抽出物、カンナビジオール、および/または大麻の葉または他の大麻植物物質を含む、先行する請求項のいずれかのプロセス。
- 活性成分が、テトラヒドロカンナビノール(THC)、カンナビジオール(CBD)、カンナビノール(CBN)、カンナビゲロール(CBG)、テトラヒドロカンナビバリン(THCV)、ヒドロカンナビジバリン(CBDV)、またはカンナビクロメン(CBC)を含む、先行する請求項のいずれかのプロセス。
- 活性成分が、大麻抽出物、任意に大麻の超臨界二酸化炭素、ブタン、エタノール、酢酸エチル、またはイソプロピルアルコール抽出物を含む、先行する請求項のいずれかのプロセス。
- 湿潤混合物が:
活性成分として約0.1%〜30%(w/w)の大麻抽出物;
担体として約5%〜30%(w/w)のアラビアゴム;および
安定化剤として約0%〜30%(w/w)の添加テルペンおよび/またはレシチン、任意に約1%〜30%(w/w)の添加テルペンおよび/またはレシチン
を含有する、上記請求項のいずれかのプロセス。 - 湿潤混合物が:
活性成分として約1%〜30%(w/w)の大麻抽出物;
担体として約10%〜30%(w/w)のアラビアゴム;および
安定化剤として約0%〜30%(w/w)の添加テルペン
を含有する、上記請求項のいずれかのプロセス。 - 上記請求項のいずれかによって製造される乾燥製品。
- 活性成分、担体および任意の安定化剤を含む任意にフレークの形態の乾燥製品であって、該フレークが以下の特徴の1つ、2つ以上、または全てを有する、乾燥製品:
(a)フレークが、示差走査熱量測定によって、約0.1〜0.6、任意に約0.23-0.239の水分活性で約58.7-62.6のガラス転移を有する;
(b)乾燥製品が、約0.23〜約0.2の水分活性で測定した場合、約40.5-80.0℃、任意に約51.7-68.8℃のガラス転移温度を有する;
(c)乾燥製品が、同等の噴霧乾燥組成物と比較した場合、ヒト官能パネルによって決定されるように艶のある、光沢のある、光彩を放つまたはきらめくように見え;および60度の角度で約1.9より大きい平均光沢測定を有する;
(d)活性成分が担体中に埋め込まれた小球中に分散されており、そして乾燥製品が少なくとも約0.01wt.%の活性成分を含有する;
(e)活性成分が担体中に埋め込まれた小球中に分散されており、小球が大部分が約500ミクロン未満の直径である;
(f)乾燥製品が気密容器中で過剰の塩化マグネシウム溶液の蒸気(32%相対湿度)に1週間曝された場合に約10質量%未満の水分;および/または気密容器中で飽和硫酸ナトリウム溶液の蒸気(81%相対湿度)に1週間曝された場合に約30質量%未満の水分を吸収する;
(g)乾燥製品が約10-1000ミクロンの厚さを有するフレークである;および
(h)乾燥製品が約6mmと約20mmとの間の円に外接しているフレークである。 - 活性成分が親水性担体中に分散されそして埋め込まれ、そして乾燥製品が約1-50wt.%の活性成分を含有する、請求項24の乾燥製品。
- 活性成分がコロイド状分散物または乳化物として担体中に埋め込まれた小球中に分散されており、任意に小球が約100nmと約500ミクロンとの間の直径である、請求項24または25の乾燥製品。
- 乾燥製品が約0.07-1000ミクロンの平均厚さを有するフレークであり、および/またはフレークの最大寸法において約0.1-20mmの平均サイズを有する、請求項24〜26のいずれかの乾燥製品。
- 活性成分が大麻抽出物である、請求項24〜27のいずれかの乾燥製品。
- 湿潤混合物が:
活性成分として約0.1%〜30%(w/w)の大麻抽出物;
担体として約5%〜30%(w/w)のアラビアゴム;および
安定化剤として約0%〜30%(w/w)の添加テルペンおよび/またはレシチン、任意に約1%〜30%(w/w)の添加テルペンおよび/またはレシチン
を含有する、請求項24〜28のいずれかの乾燥製品。 - フレークが:
約1-46%(w/w)の大麻抽出物;活性成分;
約54-96%(w/w)のアラビアゴム;および
約0%〜46%(w/w)のテルペン
を含有する、請求項24〜29のいずれかの乾燥製品。 - フレークが、同じまたは類似の組成物の噴霧乾燥サンプルと比較して、官能パネルによって決定されるように艶のあるように見える、請求項24〜30のいずれかの乾燥製品。
- 請求項23〜31のいずれかの乾燥製品を含む、医薬または化粧品組成物、任意に錠剤、カプセルまたはローション。
- 活性成分が安定に乳化している、請求項23〜31のいずれかの乾燥製品を含む飲料。
- 請求項23〜31のいずれかの乾燥製品を含むティーバッグ。
- 活性成分が疎水性であり、そして水を添加するとフレークが溶解し、そして活性成分が水中に分散する、請求項34のティーバッグ。
- パウチが任意に紙または他の繊維状または多孔性物質である、請求項23〜31のいずれかの乾燥製品を含むパウチ。
- 出発物質をエタノール-水混合物、任意に約6%〜35%エタノール中に漬け込むこと、および漬け込んだ物質をコンダクタンス窓乾燥することを含む、着色物質を乾燥するプロセス。
- エタノール エタノール-水混合物が約20-40%のエタノールを含む、請求項37のプロセス。
- エタノール-水混合物が水中約35%のエタノールを含む、請求項38のプロセス。
- 出発物質が植物物質である、請求項37〜39のいずれかのプロセス。
- 植物物質が緑色植物物質である、請求項40のプロセス。
- 請求項32の医薬組成物を被験体に投与することを含む、活性成分を被験体に送達する方法。
- 請求項23〜31の乾燥製品を被験体の皮膚または粘膜に投与することを含む、カンナビノイドを局所送達する方法。
- フレークを局所投与に適した少なくとも1つの賦形剤と混合してクリームまたはローションを形成する、請求項43の方法。
- 活性成分の経皮送達が達成される、請求項43または44の方法。
- 経皮送達が1時間以内に被験体における血漿濃度の検出可能な増加を生じる、請求項45の方法。
