特定の実施形態において、バイオマーカーmRNAのレベルは、当技術分野において公知の方法を使用して、生物学的試料においてin situ及びin vitro形式の両方で決定することができる。「生物学的試料」という用語は、被験体から単離された組織、細胞、生物学的流体及びそれらの単離物、並びに被験体内に存在する組織、細胞及び体液を含むことを意図する。多くの発現検出法は、単離RNAを使用する。in vitro法については、mRNAの単離に対して選択しない任意のRNA単離技術を腫瘍細胞からのRNAの精製に利用することができる(例えば、 Ausubel et al., ed., Current Protocols in Molecular Biology, JohnWiley & Sons, New York 1987-1999を参照されたい)。さらに、多数の組織試料を、例えばChomczynski(1989年、米国特許第4,843,155号)の1段階RNA単離プロセスのような当業者に周知の技術を使用して容易に処理することができる。
抗体ベースキットのために、該キットは、例えば、以下のものを含むことができる:(1)バイオマーカータンパク質に結合する(例えば、固体支持体に結合する)第1の抗体;及び、任意に、(2)該タンパク質又は第1の抗体のどちらかと結合し、検出可能な標識とコンジュゲートされた第2の異なる抗体。
被験体は、齧歯類、ヒト、家畜動物、コンパニオンアニマル、又は動物園の動物(zoological animal)であってもよい。一実施形態において、該被験体は、齧歯類、例えばマウス、ラット、モルモットなどであってもよい。別の実施形態において、該被験体は家畜動物であってもよい。適切な家畜動物の非限定的な例は、ブタ、ウシ、ウマ、ヤギ、ヒツジ、ラマ及びアルパカを含んでもよい。さらに別の実施形態において、該被験体はコンパニオンアニマルであってもよい。コンパニオンアニマルの非限定的な例は、イヌ、ネコ、ウサギ、及び鳥類などのペットを含んでもよい。さらに別の実施形態において、該被験体は動物園の動物であってもよい。本明細書において使用される「動物園の動物」とは、動物園で見られる動物を意味する。そのような動物には、非ヒト霊長類、大型ネコ科動物、オオカミ、及びクマを含んでもよい。好ましい実施形態において、該被験体はヒトである。
材料と方法
細胞株及び培養
ヒト包皮線維芽細胞HCA2は、O.Pereira-Smith氏(テキサス大学健康科学センター、サンアントニオ)の研究室から入手した。ヒト包皮線維芽細胞BJは、ATCC(Cat: CRL2522)から購入した。MEFは、公知の方法で13.5日胚から作製された(Demaria 2010)。マウス一次皮膚微小血管内皮細胞は、Cellbiologics(Cat: C57-6064)から購入した。
細胞をほとんどの実験及び検証のために照射後10日目に回収した。時系列について、細胞を照射後4日目、10日目及び20日目に回収した。
細胞を24ウェルプレートに播種し、グルタルアルデヒド/ホルムアルデヒド(2%/2%)で10〜15分間固定し、市販のキット(Biovision)を使用してX-Gal溶液で一晩染色した。
細胞をEdUの存在下で24時間培養し、市販のキット(Click-iTEdu Alexa Fluor 488 Imaging kit;Thermo Fisher Scientific)を使用して固定及び染色した。
画像を400倍の倍率で取得し、ImageJ(www.rsbweb.nih.gov/ij/)を使用して定量化した。試料は、各複製について少なくとも100個の細胞をカウントするtriplicateで行われた。
全RNAをIsolate II RnaMini Kit(Bioline)を用いて調製した。255〜500 ngのRNAをキット(Applied Biosystems)を用いて逆転写した。
qRT-PCR反応は、Universal Probe Libraryシステム(Roche)及びSENSIFast Probe kit(Bioline)を用いて[43]に記載の通りに実施した。チューブリンをCT値の正規化に使用した。全ての試料を、2〜3回の生物学的反復を用いて、技術的反復(technical replicate)で実行した。デルタ-CT値に基づいた統計的有意性を決定するために、unpaired two-tailed Student’s t-testを用いた。0.05以下のP値を有意であるとみなした。
細胞は、RNAeasy mini kit(Invitrogen)を用いてRNA抽出のために調製された。試料をQiasollysis緩衝液で処理し、全RNAを製造元の指示書(Invitrogen)に従ってQiacubeロボットを用いて単離した。NanoDropを用いてRNAを定量し、バイオアナライザチップ(Agilent)を用いてRNAの品質を測定した。精製したRNA試料を、ライブラリー調製(ポリAエンリッチメント(polyA-enrichment))及び製造元のプロトコル(Illumina)に従ったIllumina HiSeq RNA sequencingのためにミネソタ・バイオメディカル・ゲノミクス・センターに送付した。50 bpのpaired endシークエンシングのためのサイズ選抜を〜200bpの挿入サイズに対して行い、HiSeq2500フローセルのレーン当たり220 Mリード以上でシークエンシングを行った。全ての試料にわたる完了したランの平均品質スコアは>30であり、各プールされた試料のリード数の平均は>10,000,000リードであった。生データは、ArrayExpressデータベースに格納されている(https://www.ebi.ac.uk/arrayexpress/; access number: E-MTAB-5403)。
公開データセット及び使用された試料の概要は、表S1及び表S4の通りである。
公開データセットからの生データは、「GEOリポジトリ」から取得した。老化線維芽細胞のトランスクリプトームの以下の6個の公開データセットを含めた。1)Alspach et al, 2014 [16] (「GSE56293」)は、SASP誘導を研究するためにBJ細胞のRSを使用した。2)Dikovskayaet al, 2015 [17](「GSE70668」)は、(Rasによって誘導された)OISにおける多核形成を研究するためにIMR90細胞を使用し、そして有糸分裂において同期した細胞を使用した。3)Herranz et al, 2015 [18](「GSE61130」)は、IMR90細胞における(Rasにより誘導された)OISにおけるSASPを研究した。4)Marthandanet al, 2015 [20] (「GSE63577」)は、異なる集団倍加レベルでのロテノンの効果を研究するためにMRC-5細胞及びHFF細胞を使用し、及び本発明者らは、HFF細胞について最初(増殖)及び最後の時点のみを使用した。5)Marthandan et al, 2016 [19] (「GSE64553」)は、RSを研究するために5つの線維芽細胞株(BJ、WI-38、IMR90、HFF及びMRC-5)を使用した。6)Rai etal, 2014 [21] (“GSE53356”)は、RSのクロマチンランドスケープを研究するためにIMR90細胞を使用した。星状細胞におけるOSISを研究するCrowe et al, 2016 [30] (「GSE58910」)によって作成された1つの公開データセットは、異なる細胞型により共有される老化のコアシグネチャのために使用した。
生データは、SRAToolkit 2.6.2を使用してfastqファイルとしてダウンロードした。本発明者らのものを含む全ての試料の品質管理をFastQCソフトウェアv0.11.5を使用して実行し、低品質のリード(平均品質:<20)を除去した。Trimmommatic 0.36を用いて必要に応じてエンドトリミングを行った。試料をSTAR-2.5.1b alignerを用いてGRCh38ゲノムにアライメントし、生リードカウントテーブルをSTAR出力から直接取得した。protein-codingとしてアノテートされた遺伝子のみを分析に含めた。
データの不均質性は、タンパク質コード遺伝子について対数変換され正規化されたカウントのPCAプロットで評価した。本発明者らは、特異的刺激及び線維芽細胞老化シグネチャのメタ解析のために3つの方法を用いた:負の二項分布の一般化線形モデル(negative-binomial generalized linear model)(GLM)、フィッシャーのp値の組み合わせ( Fisherp-value combination)及び逆正規のp値の組み合わせ(Inverse Normal p-value combination)。最初のアプローチは、主な変数として老化対増殖を用いた発現差異解析のためにR-package DESeq2を使用した。複数の細胞型を用いた場合、細胞型は共変数として含めた。
発現差異解析は、各データセットについて別々にDESeq2を用いて実施し、示差的に発現した遺伝子のリストは、p値を組み合わせることなく、線維芽細胞の老化シグネチャと比較した。全てのデータセット及び線維芽細胞シグネチャ(負の二項GLM法)における多重試験調整された0.01以下のp値(multiple-testing adjusted p-value<=0.01)を有する遺伝子のみがコア老化シグネチャに含まれた。
全てのプロットは、以下のR-packagesを用いて作成された:「pheatmap」、「ggplot2」、「ggfortify」、「RColorBrewer」及び「VennDiagram」。
本実施例は、老化細胞を非老化細胞から識別するために必要かつ十分であるバイオマーカーのパネルからなる「最小のコア老化シグネチャ」を得るための老化シグネチャ内の異なるバイオマーカー遺伝子の遺伝子発現の分析を説明する。
BJ線維芽細胞:対照vs照射(Control vs Irradiated)
メラノサイト:(triplicatesにおける)対照vs照射された及び複製老化(Controlvs Irradiated and replicative senescence (in triplicates))
ケラチノサイト:(duplicatesにおける)対照vsドキソルビシン処理(Controlvs Doxorubicin-treated (in duplicates))
細胞株と培養。
ヒト包皮線維芽細胞BJ、ヒト新生児メラノサイト及びヒト新生児ケラチノサイトは、ATCCから購入した(それぞれ、Cat: CRL-2522、PCS-200-012及びPCS-200-010)。BJ繊維芽細胞を10%ウシ胎児血清(FBS、GE Healthcare Life Sciences)及び1%ペニシリン/ストレプトマイシン(Lonza)を強化したDMEM培地(Thermo FisherScientific)中で培養した。ケラチノサイトは、抗生物質を添加せずに、CnT-Prime, Epithelial Culture Medium(CellnTec, CnT-PR)中で培養した。メラノサイトは、10%ウシ胎児血清及び1%ペニシリン/ストレプトマイシンを強化し、200nM 12-O-テトラカンノイルホルボール 13-アセテート(TPA、Sigma-Aldrich)、200 nMコレラトキシン(Sigma-Aldrich)、10 nMエンドセリン1(Sigma-Aldrich)及び10 ng/mlヒト幹細胞因子(Peprotech)を添加したRPMI培地中で培養した。全ての細胞を5%酸素、5%CO2及び37℃において培養し、マイコプラズマ感染のために定期的に検査した。
電離放射線誘発性老化(IRIS)のために、細胞を137セシウム線源を用いて10Gy線量のガンマ放射線に曝露し、培地を2日ごとに新しく替えた。細胞を照射後4日目及び/又は10日目に回収した。細胞は、複製老化(RS)のために、増殖を停止するまで(〜PD 65)(70-80%コンフルエンスに到達するたびに30-40%密度で再培養して)〜4ヶ月間培養液中で増殖させた。
ドキソルビシン(Tebu-bio)は、250 nMの濃度中で24時間使用した。細胞は、対応する培地で一度洗浄した後、新しい培地を添加し、2日ごとに新しく替えた。細胞を処理後7日目に回収した。ドキソルビシンの場合において処理した試料又は溶媒(PBS)で処理したものと同じPDを有する細胞を刺激し、各条件に対する増殖対照(Proliferating controls)作製した。
細胞は、24ウェルプレート中に播種し、グルタルアルデヒド及びホルムアルデヒドの混合物(2%/2%)中に3〜5分固定して、市販キット(Biovision)を用いたX-Gal溶液で一晩染色した。細胞は、1 μg/mlの4',6-ジアミジノ-2-フェニルインドール(DAPI、Sigma-Aldrich、D9542)溶液で20分間対比染色した。画像を100倍の倍率で取得し、細胞数をソフトウェアImageJ(www.rsbweb.nih.gov/ij/)によりカウントした。陽性細胞の数は、手動でカウントした。SA-bgal染色のデータは示されていない。
全RNAは、Isolate II Rna Mini Kit(Bioline)を用いて調製した。100〜500 ngのRNAは、キット(AppliedBiosystems)を用いてcDNAに逆転写した。qRT-PCR反応は、Universal Probe Libraryシステム(Roche)及びSENSIFast Probe kit(Bioline)を用いて製造元の指示書に従って行った。チューブリン又はアクチンの発現は、CT値の発現を正規化するために用いた。試料は、技術的反復で2〜3回の生物学的反復において実行した。デルタ-CT値に基づいた統計的有意性を決定するために、unpaired two-tailed Student’s t-testを用いた。
主成分分析に用いられる遺伝子は、qPCRの結果における増殖性細胞及び老化細胞の間の発現における変化の再現性に基づいて予め選択した。ほとんどの試料において(RNAseqの結果に基づいた)元の分析により予測されたものと同じ傾向に従った遺伝子をPCAプロットを構築するために用いた。これらの遺伝子は次のものを含む。BCL2L2、C2CD5(プライマー対増幅変異体1、2及び6)、DYNLT3、GDNF(プライマー対増殖変異体1)、MTCYB、PLK3、PLXNA3、SUSD6及びTSPAN13。主成分1(X軸)上の試料分離に対する各遺伝子の寄与を各試料のセットごとに計算した。より寄与の高い遺伝子を「1」として採点し、最も寄与の低い遺伝子を「9」として採点した。分析において使用した全ての試料のリストを構築し、全ての試料上の各遺伝子の全体のスコアを算出し、「1」は全ての試料においてより寄与の高い遺伝子であり、「9」は全ての試料において最も寄与の低い遺伝子であった。最後に、下位に採点された(scored last)3つの遺伝子(7、 8及び9)を除去した。次の6個の最終遺伝子を用いて新しいPCAプロットを構築した:GDNF(プライマー対増幅変異体1)、TSPAN13、BCL2L2、PLK3、SUSD6及びC2CD5(プライマー対増幅変異体1、2及び6)。
図6では、少なくとも確立された老化のマーカーであるチューブリンに対して正規化した LMNB1の下方調節又はp21の上方調節の解析により調査対象の各細胞試料の老化を確認するものである。
TSPAN13は、機能が十分に解明されていない細胞表面タンパク質である。
本実施例では、TSPAN13と細胞の老化との関連性を説明する。
図16は、TSPAN13のmRNAレベルが異なる種類の老化細胞で増加することを示す。図17では、免疫蛍光を使用することによりTSPAN13タンパク質がより発現していることを示す。図18及び図19は、フローサイトメトリー及び追加の細胞/刺激分析を用いて、類似のTSPAN13上方調節を示す。図20において、TSPAN13の高発現又は低発現に基づいて老化細胞集団をソーティングするためにフローサイトメトリーを使用した。興味深いことに、TSPAN13レベルの増加は、他の老化マーカーp16及びp21のレベルの増加と相関することを見出した。
TSPAN13は、その細胞表面局在化に基づき、薬物標的化に理想的である。例えば、老化細胞を殺傷するための薬物複合体は、(i)使用時に、TSPAN13に特異的に標的化及び結合する、TSPAN13標的化剤、及び(ii)結合した老化細胞を殺傷する細胞毒性剤を含むように構成され得る。
遺伝子発現TSPAN13(図16)
ヒト包皮線維芽細胞BJは、ATCCから購入した(Cat: CRL2522)。細胞は、5%酸素、5%CO2及び37℃において、10%ウシ胎児血清(FBS、GE Healthcare LifeSciences)及び1%ペニシリン/ストレプトマイシン(Lonza)を強化したDMEM培地(Thermo Fisher Scientific)を用いて培養した。細胞は、マイコプラズマ感染のために定期的に検査した。
静止状態は、0.2%FBSを添加したDMEM中で48時間細胞を培養することにより誘導した。細胞は、電離放射線誘発性老化(IRIS)のために、137セシウム線源を用いて10Gy線量のガンマ放射線に曝露し、そして培地は2日ごとに新しく替えた。細胞は、照射後10日目に回収した。細胞は、複製老化(RS)のために、増殖を停止するまで(〜PD 65)(70-80%コンフルエンスに到達するたびに30-40%密度で再培養して)〜4ヶ月間培養液中で増殖させた。細胞は、酸化ストレス誘発性老化(OSIS)のために、200 μMの過酸化水素(Sigma Aldrich)で2時間処理した後、10%FBSを添加した新鮮なDMEM中で薬物除去及び培養した。処理は、2日ごとに新しく替えた培地を用いて、0日目、3日目及び6日目に繰り返し、細胞は、最初の処理後10日目に回収した。
ドキソルビシン(Tebu-bio)は、250 nMの濃度で24時間使用した。その後、培地を10%FBSを添加した通常のDMEMで置換し、2日ごとに新しく替えた。細胞は、処理後7日目に回収した。
対応する溶媒を用いて細胞を刺激し、及び/又は処理された試料の同じPDを検討して、各条件に対する増殖対照を作製した。複数の条件に対する対照が1つしか示されていない場合は、各条件に対する対照の平均が示される。
9d IR及びctrl BJ細胞を、両方ともカバースリップ(Sarsteddt、カタログ番号83.1840.002)につき15,000細胞の密度で播種し、細胞培養インキュベーターで一晩インキュベートした(5%酸素)。翌日、細胞をPBSで洗浄し、4%PFA/PBS中で固定した。細胞は、IFまで4℃で保存した。
PBS中5wt%BSA (Sigma)をブロッキングバッファー及び抗体希釈液として使用した。1時間のブロッキングの後に、ウサギ抗ヒトTSPAN13抗体(Genetex、カタログ番号52155)を一次抗体(抗体希釈液中1:50希釈)として用いて4℃で一晩インキュベートした。使用した二次抗体は、ヤギ抗ウサギAlexaFluorl488(Thermo Fisher Scientific、カタログ番号R37116)であった。穏やかに撹拌しながら暗所で90分間インキュベートした。PBSで3回洗浄し、MilliQ水で1回洗浄した後、ProLong Gold Antifade Mountant with DAPI(Thermo Fisher Scientific、カタログ番号P36941)を用いてガラススライド上にカバースリップをマウントした。画像は、Leica DMI6000を用いて作成した。
Ctrl BJ細胞及び8d IR BJ細胞を70%エタノール中で固定し、FACS分析まで4℃で保存した。以降の全てインキュベーションは、全て4℃で行った。使用したFACS緩衝液は、PBS中1%BSAである。全ての洗浄工程は、400 μlFACS緩衝液中、5分間、800x g、4℃である。FACS分析のために、1,000,000個のctrl BJ細胞及び600,000個の8d IR BJ細胞を用いた。使用した一次抗体は、ウサギ抗ヒトTSPAN13抗体(Genetex、カタログ番号52155、FACS緩衝液中1:20希釈)である。一次抗体で1時間インキュベートした。3回の洗浄工程を一次抗体及び二次抗体インキュベーションの間に行った。使用した二次抗体は、ヤギ抗ウサギAlexaFluor488(Thermo Fisher Scientific、カタログ番号R37116)であった。暗所で30分間インキュベートした。細胞をセルストレーナーキャップを通してFACSチューブ(Corning、カタログ番号352235)に移す前に、3回の洗浄工程を行った。蛍光シグナルは、BD FACS CANTO IIを用いて測定した。FACSデータ分析は、Kaluzaソフトウェア (Beckman Coulter)を用いて行った。
FACSを介して細胞膜上のTSPAN13発現を分析するために、ctrl WI-38細胞、(1 uMで毎日処理した後)7d パルボシクリブ及び7dドキソルビシン処理WI-38細胞を用いた。回収後、細胞をPBSで洗浄し、続いてFACS緩衝液(PBS中1%BSA及び0.1%NaN3)中で洗浄した。全ての洗浄工程は、400 μl FACS緩衝液中、3分間、800x g、4℃で行った。特に明記しない限り、以降の全てのインキュベーションは、4℃で行った。一次抗体は、ウサギ抗ヒトTSPAN13抗体(Genetex、カタログ番号52155、FACS緩衝液中1:50希釈)を使用して、30分間インキュベーションした。細胞をFACS緩衝液で洗浄し、PBS中4%パラホルムアルデヒド(ThermoFisher Scientific、カタログ番号28908)中で室温で15分間固定した。FACS緩衝液で3回洗浄した後、細胞をPBS中に4℃で保存した。二次抗体インキュベーションは、ヤギ抗ウサギAF633(Thermo Fisher Scientific、カタログ番号A21070)を用いて30分間行った。細胞をセルストレーナーキャップを通してFACSチューブ(Corning、カタログ番号352235)に移す前に、3回の洗浄工程を行った。蛍光シグナルは、BDFACS CANTO IIを用いて測定した。FACSデータ分析は、Kaluzaソフトウェア (Beckman Coulter)を用いて行った。
TSPAN13発現細胞をソーティングするために、ctrl BJ細胞及び9d IR BJ細胞を使用して、陽性集団(TSPAN13の細胞膜染色)対TSPAN13陰性集団を示した。細胞は固定せず、細胞の回収と同じ日にTSPAN13発現についてソーティングした。FACS緩衝液は、PBS中1%BSAであり、洗浄工程は、FACS緩衝液中、3分間、800x g、4℃で行った 。全てのインキュベーションは、4℃で行った。一次抗体は、ウサギ抗ヒトTSPAN13抗体(Genetex、カタログ番号52155、FACS緩衝液中1:50希釈)を使用して、1時間インキュベーションした。ヤギ抗ウサギAlexaFluor488(Thermo Fisher Scientific、カタログ番号R37116)を用いた二次抗体インキュベーションの前にFACS緩衝液で3回洗浄した。暗所で30分間インキュベートした。細胞をセルストレーナーキャップを通してFACSチューブ(Corning、カタログ番号352235)に移す前に、3回の洗浄工程を行った。BD FACS JAZZを使用して、蛍光シグナルを測定し、細胞をソーティングしてISOLATE II RNA Mini Kit(Bioline、カタログ番号52073)のRNA溶解緩衝液中に分取した。FACSデータ分析は、Kaluzaソフトウェア (BeckmanCoulter)を用いて行った。RNA単離は、マニュアルに従って行った。逆転写酵素PCRは、High-Capacity cDNA Reverse Transcription Kit (Applied Biosystems、カタログ番号4368813)を用いて行った。2つの細胞周期停止遺伝子(p16及びp21)及びTSPAN13についてのqPCRは、LC480 (Roche)及びSensiFASTProbe Lo-ROX Kit (Bioline、カタログ番号84020)を用いて行った。チューブリンをリファレンス遺伝子として使用した。プライマー: p16、F-GAGCAGCATGGAGCCTTC、R-CGTAACTATTCGGTGCGTTG、UPLプローブ#67; p21、F-TCACTGTCTTGTACCCTTGTGC、R-GGCGTTTGGAGTGGTAGAAA、UPLプローブ#32; TSPAN13、F-CCCTCAACCTGCTTTACACC、R-AATCAGCCCGAAGCCAAT、UPLプローブ#84; チューブリン、F-CTTCGTCTCCGCCATCAG、R-CGTGTTCCAGGCAGTAGAGC、UPLプローブ#40;

Claims (20)
- 6個以上のポリペプチド、又はそれらをコードするmRNAを含むバイオマーカーパネルの使用であって、ここで該パネルが少なくともバイオマーカーTSPAN13、GDNF、C2CD5、SUSD6、BCL2L2、PLK3、又はそれらの変異体若しくは断片を細胞老化のバイオマーカーセットとして含む、使用。
- 前記セットが、更にCTLN、FAM214B、PATZ 1、PLXNA3、STAG1、TOLLIP、TRDMT1、ZBTB7A、ARID2、B4GALT7、CHMP5、CREBBP、DDA1, DYNLT3、EFNB3、ICE1、MEIS1、NOL3、PCIF1、PDLIM4, PDS5B、RAI14、RHNO1、SCOC、SLC16A3、SMO、SPIN4、TAF13、TMEM87B、UFM1及びZNHIT1、又はそれらの変異体若しくは断片からなる群から選択される、1以上のバイオマーカーを含む、請求項1に記載の使用。
- 前記セットが、バイオマーカーTSPAN13、GDNF、C2CD5、SUSD6、BCL2L2、PLK3、並びに、DYNLT3及びPLXNA3の1つ若しくは両方、又はそれらの変異体若しくは断片を含む、請求項1又は2に記載の使用。
- 前記セットが、バイオマーカーTSPAN13、GDNF、C2CD5、SUSD6、BCL2L2、PLK3、DYNLT3及びPLXNA3、又はそれらの変異体若しくは断片を含む又はからなる、請求項3に記載の使用。
- TSPAN13ポリペプチド又はそれをコードするmRNA、又はそれらの変異体若しくは断片の細胞老化バイオマーカーとしての使用。
- 試験試料中の老化細胞を検出する方法であって、
該方法は、該試料中で、少なくとも請求項1〜5のいずれか一項に記載の老化細胞バイオマーカーのセット又はバイオマーカーの発現を検出することを含み、ここで参照試料中で検出された発現レベルと比較した少なくとも1つの該バイオマーカーの発現レベルの変化が、該試料中に存在する老化細胞の指標である、方法。
- 参照試料中で検出された発現レベルと比較した、TSPAN13、GDNF、C2CD5、PLXNA3、SUSD6、BCL2L2及び/又はPLK3、又はそれらの変異体若しくは断片の発現レベルの上昇が、該試料中に存在する老化細胞の指標である、請求項5に記載の方法。
- 前記試験試料が被験体から採取された体試料であり、好ましくは、ここで該試料が、血液、血漿、血清、髄液、尿、汗、唾液、涙、乳房吸引液、前立腺液、精液、膣液、糞便、子宮頸管掻爬、細胞、羊水、眼内液、粘液、息の水分、動物組織、細胞溶解物、腫瘍組織、髪、皮膚、腔内粘膜、爪、骨髄、軟骨、プリオン、骨粉末、耳垢、又はそれらの組み合わせを含む、請求項6又は7に記載の方法。
- 前記被験体が、実験動物又はヒトである、請求項6〜8のいずれか一項に記載の方法。
- 試料中の老化細胞を検出するための老化細胞検出キットであって、
該キットは、被験体由来の試料中の少なくとも請求項1〜4のいずれか一項に記載のバイオマーカーポリペプチド又はmRNAのセットの存在を検出するための手段を含む、キット。
- 少なくとも1つの対照試料又は参照試料を含み、好ましくは陰性対照及び/又は陽性対照を含む、請求項10に記載のキット。
- 老化細胞を殺傷するための薬物複合体であって、
該複合体は、(i)使用中に、TSPAN13、GDNF、FAM214B、PLXNA3、SUSD6、TOLLIP、ZBTB7A、B4GALT7、BCL2L2、CHMP5、DDA1、DYNLT3、NOL3、PDLIM4、PLK3、RAI14、SCOC、SLC16A3、TAF13、TMEM87B、UFM1及びZNHIT1からなる群から選択される少なくとも1つの老化細胞バイオマーカーに特異的に標的化及び結合するように構成された老化細胞標的化剤と、好ましくはここで該標的化剤がTSPAN13、PLXNA3、SUSD6、GDNF、FAM214B、TOLLIP and ZBTB7Aからなる群から選択されるバイオマーカーに結合する、並びに (ii) 結合した老化細胞を殺傷する細胞毒性剤とを含む、薬物複合体。
- 使用時に、TSPAN13に特異的に標的化及び結合するように構成された標的化剤を含む、請求項12に記載の薬物複合体。
- 前記標的化剤が、抗体若しくはその抗原結合断片、アプタマー、プラスチック抗体又は小分子である、請求項12又は13に記載の薬物複合体。
- 前記細胞毒性剤が、老化細胞抑制剤、放射性同位元素、毒素又は毒性ペプチドであり、好ましくは老化細胞抑制剤である、請求項12〜14のいずれか一項に記載の薬物複合体。
- 前記細胞毒性 (senolytic)が、(a)Bcl-2抗アポトーシスタンパク質ファミリーメンバーの阻害剤、(b)MMD2阻害剤、又は(c)Akt特異的阻害剤、好ましくは1以上のBCL-2抗アポトーシスタンパク質ファミリーメンバーの阻害剤であり、ここで該阻害剤が少なくともBcl-xLを阻害し、より好ましくはABT-263、ABT-737、WEHI-539、及びA-1 155463から選択される、請求項15に記載の薬物複合体。
- 医薬品として使用するための、請求項12〜16のいずれか一項に記載の薬物複合体。
- 加齢性疾患の治療、遅延、予防又は改善における使用のための、請求項12〜17のいずれか一項に記載の薬物複合体であって、好ましくはここで該加齢性疾患がアテローム性動脈硬化症、循環器疾患、癌、関節炎、緑内障、白内障、骨粗鬆症、2型糖尿病、高血圧症、アルツハイマー病又は他のタイプの認知症である、薬物複合体。
- 請求項12〜16のいずれか一項に記載の薬物複合体と、薬学的に許容される溶媒とを含む、医薬組成物。
- 治療上有効な量の、非老化細胞よりも老化細胞を選択的に殺傷する、請求項12〜16のいずれか一項に記載の薬物複合体を含む医薬組成物をそれを必要とする被験体に投与することを含む、老化関連疾患又は障害を治療する方法。
