本発明者らは、驚くべきことに、以下のことを見出した:本発明に従って構築された二機能性融合タンパク質(特異的に結合された標的分子CD20タンパク質の結合構造ドメインと特異的に結合された標的分子CD47タンパク質の結合構造ドメインを含む。)は、CD20陽性腫瘍に対して有意な抑制効果を有し、リツキシマブによるCD20陽性腫瘍への治療効果を強化し、リツキシマブに対して抵抗性を発生するCD20陽性腫瘍を治療し、リツキシマブの増感剤として使用することができる。
本発明に係る組換え型二機能性融合タンパク質は、少なくとも2つの独立した結合構造ドメイン(D1およびD2)を含む。本明細書中で使用される場合、表現「結合構造ドメイン」は、特定目的のタンパク質に特異的に結合できる任意のペプチド、ポリペプチド、核酸分子、足場分子、ペプチドディスプレイ分子またはポリペプチド含有構築物を意味する。

本発明の別の方面において、組換え型二機能性融合タンパク質は腫瘍細胞をターゲティングするために使用されることができる。
本発明はさらに組成物を提供する。好ましい実施形態では、この組成物は、上記の抗体もしくはその活性フラグメントもしくはその融合タンパク質、または上記の組換え型二官能性融合タンパク質、および薬学的に許容されるベクタを含む医薬組成物である。一般に、これらの物質は、毒性のない、不活性かつ薬学的に許容される水性ベクタ媒体中に配合することができる。pHは、調製される物質の性質および治療されるべき病状によって変化することがあるが、通常約5〜8、好ましくは約6〜8である。調製された医薬組成物は、腫瘍内投与、腹腔内投与、静脈内投与、または局所投与を含むがこれらに限定されない従来の経路によって投与することができる。
リツキシマブ重鎖コード配列のカルボキシ末端またはアミノ末端は、ヒトSIRPαの外側における第1の機能領域(SIRPαドメイン1、SD1)の遺伝子コード配列と連結し、配列の両端で2つのクローニング部位点(アミノ末端にあるHindIIIおよびカルボキシ末端にあるSalI)を付加することによって、CD20mAb−H−SD1またはSD1−CD20mAb−H遺伝子コード配列を形成した。次に、CD20mAb−H−SD1、SD1−CD20mAb−H遺伝子配列およびリツキシマブ軽鎖遺伝子コード配列(CD20mAb−L)を合成するために蘇州省心生物技術有限公司に委託し、合成した配列をpMac−HおよびpMac−L発現ベクタ(Macroimmune Inc.から購入)にそれぞれクローニングし、pMac−CD20mAb−H−SD1、pMac−SD1−CD20mAb−HおよびpMac−CD20mAb−L発現ベクタを形成した。
1mgのCD20mAb−SD1またはSD1−CD20mAbタンパク質に対してSEC−HPLC分析を行った。
2つの腫瘍細胞を用いて、CD20およびCD47に対するCD20mAb−SD1およびSD1−CD20mAbタンパク質の結合活性をそれぞれ鑑定し、それらを単一標的対照タンパク質と比較した。Jurkat細胞(CD20−CD47+)を用いてCD47の標的結合活性を分析し、Raji−CD47KO細胞(CD20+CD47−)を用いてCD20の標的結合活性を分析した。
異なる濃度のCD20mAb−SD1、SD1−CD20mAbタンパク質、陽性対照タンパク質SIRPα−Fcおよびリツキシマブを、それぞれ25nMビオチンにより標記されたSIRPα−Fc(Biotin−SIRPα−Fc)と混合し、室温で30分間インキュベートした。その後、混合物をJurkat細胞と室温で1時間インキュベートした。洗浄後、蛍光により標記されたストレプトアビジン(FITC−Strep)と室温で1時間インキュベートした。洗浄後、細胞を100μlの冷リン酸緩衝液(PBS)に懸濁した。最後に、フローサイトメトリー(FACS)を用いて、SIRPα−Fcと細胞との結合状況を分析した。
106/mlのRaji細胞またはCD47ノックアウトRaji細胞(Raji−CD47KO)(標的細胞)をそれぞれ1mMのCFSE(1:500)と混合し、37℃で30分間インキュベートし、10分ごとに1回均等に混合した。NK92MI−CD16a細胞(エフェクター細胞)の密度を6×105/mlに調節した。エフェクター細胞を標的細胞と2:1の比率で混合して、異なる濃度のCD20mAb−SD1、SD1−CD20mAb、陽性対照タンパク質であるリツキシマブ、および陰性対照タンパク質であるハーセプチン/トラスツズマブ(Herceptin/Trastuzumab)を添加し、37℃、5%CO2インキュベーター内で4時間インキュベートした。インキュベ−ションが完了後に、細胞培養プレ−トを室温で10分間静置し、室温に平衡化した後、20μlのPI(ヨウ化プロピオン酸、Propodium Iodide)色素(最終濃度5μg/ml)を各ウェルに添加し、ロ−ガンで吹いたり打ったりすることによって均等に混合した後、FACS分析技術によって異なるタンパク質濃度における細胞PI染色の陽性率を分析し、以下の式に従ってADCC活性を計算した:

第1の実験では、Raji細胞(標的細胞)を、それぞれ異なる濃度のCD20mAb−SD1、陽性対照タンパク質であるリツキシマブ(Rituximab)、および陰性対照タンパク質であるハーセプチン(Herceptin)と混合しました。第2の実験では、Raji細胞(標的細胞)を、それぞれ異なる濃度のCD20mAb−SD1およびSD1−CD20mAbと混合した。次いで一定濃度のウサギ補体を上記混合系に添加し、37℃で、細胞を5%CO2インキュベーター中で4時間インキュベートした。インキュベ−ションが完了後、細胞培養プレ−トを室温で10分間静置し、室温に平衡化した後、20μlのPI(ヨウ化プロピオン酸)色素(最終濃度5μg/ml)を各ウェルに添加し、ロ−ガンで吹いたり打ったりすることによって均等に混合した後、FACS分析技術によって異なるタンパク質濃度における細胞PI染色の陽性率を分析し、以下の式に従ってCDC活性を計算した:

マウスマクロファージ(Ana−1)を96ウェル細胞培養プレ−トに1ウェルあたり1×105細胞で添加し、細胞を細胞培養インキュベーター(37℃、5%CO2)に置いて16〜18時間培養した。CFSEにより標記された標的細胞(HL−60)を、それぞれ異なる濃度のCD20mAb−SD1、SD1−CD20mAb、陽性対照タンパク質であるSIRPaD1−Fc、および陰性対照タンパク質であるリツキシマブと混合し、37℃、5%CO2の条件下で45分間インキュベートした後、マクロファージ(Ana−1)を含む培養プレ−トに細胞を移し、同じ条件下で2時間培養を続け、培養プレ−トを取り出し、懸濁した標的細胞を洗い流し、マクロファージを集め、FACSを用いてマクロファージのCFSE蛍光強度を分析した。
第1の実験では、Raji細胞インサイチュ腫瘍モデルを用いて、CD20mAb−SD1のインビボ抗腫瘍活性を研究した。30匹の免疫不全マウスの尾静脈にLaji細胞を注射し、各マウスに5×106細胞を注射し、注射後の翌日にマウスをランダムに5つのグループに分け、1番目のグループでPBSを腹腔内注射し、2番目のグループでCD20mAb−SD1(5mg/kg)を腹腔内注射し、3番目のグループでSD1−Fc(2.5mg/kg)を腹腔内注射し、4番目のグループでリツキシマブ(2.5mg/kg)を腹腔内注射し、5番目のグループでSD1−Fcおよびリツキシマブ(2.5+2.5mg/kg)を腹腔内注射した。週に一回、5回投与した。マウスの状態を毎日観察し、体重を測定した。
CD20mAb−SD1およびSD1−CD20mAbのタンパク質構造は、図1および図2に示すように、それぞれ1本の重鎖遺伝子および1本の軽鎖遺伝子によってコードされる4本のポリペプチド鎖(2本の重鎖および2本の軽鎖)が対応するセカンドフローキーの接続によって構成される。そのうち、CD20mAb−SD1の重鎖遺伝子コード配列は1749個のヌクレオチドからなる(図3A)。ここで、CD20mAb重鎖は、1350個のヌクレオチドを有し、450個のアミノ酸(1〜450)をコーディングする。SD1配列は、399個のヌクレオシドを有し、133個のアミノ酸(451〜583)をコーディングする。対応するアミノ酸配列を図4Aに示す。SD1−CD20mAbの重鎖遺伝子コード配列は1782個のヌクレオチドからなり(図3C)。そのうち、SD1配列は、399個のヌクレオチドを有し、133個のアミノ酸(1〜133)をコーディングする。連結ペプチド鎖は、30個のヌクレオチドを有し、10個のアミノ酸(134〜143)をコーディングする。CD20mAb重鎖は、1353個のヌクレオチドを有し、451個のアミノ酸(144〜594)をコーディングする。対応するアミノ酸配列を図4Cに示す。CD20mAb−SD1およびSD1−CD20mAbの軽鎖はすべて、639個のヌクレオチド(図3B、3D)を有し、213個のアミノ酸(1〜213)をコーディングする(図4B、4D)。
SEC−HPLC分析により確認されたように、CD20mAb−SD1およびSD1−CD20mAbタンパク質凝集体含量はすべて3%未満であり、低分子量成分(フラグメント化)は1〜3%であり、タンパク質の完全性が非常に良好であることを示した(図5A、5B)。
フローサイトメトリーを用いてCD20とCD47に対するCD20mAb−SD1とSD1−CD20mAbタンパク質の結合活性をそれぞれ分析した結果、両方の構造のタンパク質の二つの標的点に対する結合活性はいずれも高く、EC50がいずれもナノモルレベルにあった。CD20との結合活性については、CD20mAb−SD1とリツキシマブが類似し、EC50がそれぞれ0.08nM(CD20mAb−SD1)および0.06nM(リツキシマブ)であり、一方SD1−CD20mAbの活性はやや低く、EC50が0.4nMであった。CD47との親和性活性については、SD1−CD20mAbがCD20mAb−SD1よりも有意に良好であり、前者のEC50が0.09nMであり、後者のEC50が2.72nMであった。
我々は、Jurkat細胞を使用してCD47に対するCD20mAb−SD1およびSD1−CD20mAbの競合的結合活性を分析した。図7に示すように、両方の構造のタンパク質は、SIRPαとJurkat細胞との結合を有意に阻害したが、SD1−CD20mAb(EC50=19.74nM)の阻害活性は、CD20mAb−SD1(EC50=476.2nM)より有意に優れている。
CD20mAb−SD1およびSD1−CD20mAbタンパク質はADCC(図8のA、B)およびCDC(図8のC、D)活性の両方を有し、両方の構造のタンパク質のADCCに有意な差別はないが、リツキシマブより有意に優れた。Raji細胞に対しては、EC50がそれぞれ、CD20mAb−SD1=0.14ng/ml;SD1−CD20mAb=0.15ng/ml;リツキシマブ=3.82ng/mlであった。Raji−CD47KO細胞に対しては、EC50がそれぞれ、CD20mAb−SD1=0.14ng/ml;SD1−CD20mAb=0.10ng/ml;リツキシマブ=1.13ng/mlであった。両方の構造のタンパク質はいずれも強いCDC活性を有するが、SD1−CD20mAb(EC50=3.36nM)の活性がCD20mAb−SD1(EC50=10.97nM)よりも有意に優れている。
マウスマクロファージ(Ana−1)を用いて、CD20mAb−SD1およびSD1−CD20mAbタンパク質が腫瘍細胞に対するマクロファージの食作用活性を促進することを研究した。図9に示すように、SD1−CD20mAbは強い食作用活性(EC50=0.3μg/ml)を有し、陽性対照タンパク質SIRPaD1−Fc(EC50=0.14μg/ml)に相当する。一方、CD20mAb−SD1は全く食作用活性を示さなかった。
我々は、Raji細胞インサイチュ腫瘍モデルを用いて、CD20mAb−SD1タンパク質のインビボ抗腫瘍活性を調べた。図10Aに示すように、未治療群のマウスは治療開始後38日以内に全部死亡し、リツキシマブおよびSIRPaD1−Fcのような単一の標的で治療された群ではマウスの生存率が大幅に向上し、実験の終わりまでに、生存率がそれぞれ67%および83%であった。しかし、二重標的組換えタンパク質CD20mAb−SD1で治療された群では、生存率が100%であり、これは単一標的のみに作用した薬物よりも著しく良好であった。
Claims (15)
- 組換え型二機能性融合タンパク質であって、
CD20に特異的に結合する抗体または抗体フラグメント、および
CD47に特異的に結合するペプチド、
を含み、
前記ペプチドは前記抗体または前記抗体フラグメントに結合している、
組換え型二機能性融合タンパク質。 - 前記抗体または前記抗体フラグメントは、配列番号5の重鎖可変領域および配列番号7の軽鎖可変領域を含む、請求項1に記載の組換え型二機能性融合タンパク質。
- 前記抗体は、リツキシマブ、オビヌツズマブ、およびオファツムマブからなる群から選択される、請求項1に記載の組換え型二機能性融合タンパク質。
- 前記ペプチドがSIRPαに由来する、請求項1に記載の組換え型二機能性融合タンパク質。
- 前記ペプチドはヒトSIRPαの第一細胞外ドメインである、請求項4に記載の組換え型二機能性融合タンパク質。
- 前記抗体は重鎖および軽鎖を含み、前記ペプチドは前記抗体の前記重鎖のN末端に結合している、請求項5に記載の組換え型二機能性融合タンパク質。
- 前記抗体は重鎖および軽鎖を含み、前記ペプチドは前記抗体の前記重鎖のC末端に結合している、請求項5に記載の組換え型二機能性融合タンパク質。
- 前記重鎖は配列番号5の重鎖可変領域を含み、前記軽鎖は配列番号7の軽鎖可変領域を含む、請求項6または7に記載の組換え型二機能性融合タンパク質。
- 請求項1に記載の組換え型二機能性融合タンパク質をコードするポリヌクレオチド。
- 請求項9に記載のポリヌクレオチドを含むベクタ。
- 請求項10に記載のベクタを含有し、またはゲノムに請求項9に記載のポリヌクレオチドが組込まれた、遺伝子操作された宿主細胞。
- 医薬組成物であって、
(i)請求項1に記載の組換え型二機能性融合タンパク質、および
(ii)薬学的に許容されるベクタ、
を含有する医薬組成物。 - CD20タンパク質を発現する腫瘍を処置するための、請求項1に記載の組換え型二機能性融合タンパク質の使用。
- 前記CD20タンパク質を発現する前記腫瘍は、胃がん、肝臓がん、白血病、腎臓がん、肺がん、小腸がん、骨腫瘍、前立腺がん、直腸結腸がん、乳がん、大腸がん、前立腺がん、子宮頸がん、副腎がん、および膀胱がんからなる群から選択される、請求項13に記載の組換え型二機能性融合タンパク質の使用。
- 組換え型二機能性融合タンパク質を製造する方法であって、
(a)請求項11に記載の宿主細胞を発現に適した条件下で培養すること、
(b)請求項1に記載の組換え型二機能性融合タンパク質を培養物から単離すること、
を含む、請求項1に記載の組換え型二機能性融合タンパク質の製造方法。
