Abstract
Description
本発明は、生物医学の分野に関し、より具体的には、抗B7-H3抗体ならびにその調製および適用に関する。
腫瘍の成長および転移は、宿主の免疫監視を回避し、かつ宿主の防御を克服する能力に大きく依存する。ほとんどの腫瘍は、宿主の免疫系によって様々な程度で識別される抗原を発現するが、多くの場合、エフェクターT細胞の非効率的な活性化により、不十分な免疫応答を誘発する。
[発明が解決しようとする課題]
本発明の目的は、抗B7-H3抗体ならびにその調製および適用を提供することである。

細胞免疫応答において、T細胞の増殖および活性化は、T細胞受容体TCRがAPCまたは腫瘍細胞の表面のMHCによって提示された第1のシグナルを識別する必要があるだけでなく、共刺激分子によって提供された第2のシグナルをさらに必要とする。B7-CD28スーパーファミリーは、これまでに発見された共刺激分子ファミリーの一つであり、免疫グロブリンスーパーファミリーに属する。B7ファミリー分子は、T細胞免疫応答を増強および維持するための刺激シグナルを提供することができ、T細胞免疫応答を制限および弱体化させるための阻害シグナルを生成することもできる。従って、当該ファミリーは、腫瘍疾患、臓器移植および自己免疫疾患において重要な役割を果たす。

以下、本発明は、具体的実施例と併せてされに説明される。これらの実施例は、本発明を説明するためにのみ使用され、本発明の範囲を限定するものではないことを理解されたい。以下の実施例において、具体的条件を示さない実験方法は、通常、例えば、Sambrookら、分子クローニング:実験マニュアル(New York:Cold Spring Harbor Laboratory Press、1989)に記載される条件等の従来の条件、またはメーカーによって提案された条件に従う。特に明記されない限り、パーセンテージおよび部数は、重量パーセンテージおよび重量部数で計算される。
健康なBalb/cマウスを選択し、皮下、腹腔内に複数回注射して免疫し、2週間に1回免疫する。マウスの一つのグループをB7-H3hisタンパク質で免疫し、B7-H3を過剰発現するCHOK細胞でマウスの別のグループを免疫する。最初の免疫にフロイントの完全アジュバントを使用し、続いてフロイントの不完全アジュバントを使用して免疫する。3~4回免疫した後、ELISAでマウスの血清力価を検出し、力価が最も高いマウスを犠牲にした後に脾臓を取ってファージディスプレイライブラリー構築する。
免疫後の合格的力価を有するマウス脾臓を採取し、マウス脾臓の総RNAをクロロホルムで抽出し、次いで逆転写PCRによってcDNAライブラリーを調製する。Sanyou Bio独自のコンビネーションプライマーでマウスの軽鎖および重鎖の可変領域遺伝子を増幅し、組み合わされたFabフラグメントは、Fab抗体ライブラリーを構築するためにファージミドベクターに挿入される。ライブラリーをファージディスプレイライブラリーとして調製し、液相スクリーニング方法を使用して、B7-H3hisbiotinを磁性ビーズにコーティングして、3回の液相スクリーニングを行う。3回の液相スクリーニング後、B7-H3Hisでマイクロタイタープレートをコーティングし、次いで抗B7-H3のFab上清を加えて、ELISAスクリーニングによって、B7-H3に特異的に結合する陽性クローンを取得する。
Sanyou Bio数千億レベルの抗体ライブラリーの構築:4000を超えるヒトPBMC試料をRNA抽出に使用し、IMGTデータベースに従って様々な生殖細胞株(germline)のプライマーとして設計し、文献で報告されたヒト由来遺伝子の比率に従って、ライブラリー構築計画を立て、生殖細胞株の比率が異なる合計四つの数百億のライブラリーを確立し、数千億の抗体ライブラリーに統合する。
抗体の軽鎖および重鎖を有するプラスミドをExpiCHOS細胞に同時トランスフェクトし、7日間発現した後に細胞発現上清を収集し、プロテインA親和性クロマトグラフィーカラムを使用して精製し、具体的な実験段階については、AKTAのマニュアルを参照する。精製抗体を透析によりPH7.2のPBS緩衝液に交換する。合計23のマウス免疫ライブラリーおよび39のヒトライブラリーの抗体に対して発現および精製し、正常に発現および精製した合計57の抗体を最終的に取得し、ここで、19は、マウス免疫ライブラリーに由来し、38は、ヒトライブラリーに由来する。
CDR移植により、マウス由来抗体(IL-P7-C05、IL-P5-C06)のCDRは、対応するヒト抗体骨格に移植され、次いで3D構造シミュレーションによって軽鎖および重鎖の突然変異体のいくつかの対を設計し、一過性発現後に抗体の活性を検証する。
実施例5.タンパク質レベル結合および種交差検出
ヒトB7-H3his、マウスB7-H3hisおよびサルB7-H3hisをPBSで2μg/mLに希釈し、マイクロタイタープレートの各ウェルに30μLを加え、次いでマイクロタイタープレートを4℃の冷蔵庫で16~20時間インキュベートする。翌日、洗浄液(0.05%Tween20を含むPBS)でマイクロタイタープレートを1回洗浄し、次いで5%脱脂粉乳を加えて1時間ブロッキングする。ブロッキング完了後、マイクロタイタープレートを1回洗浄し、次いで勾配希釈した抗B7-H3抗体を加え、室温下で1時間インキュベートする。インキュベートした後、マイクロタイタープレートを3回洗浄し、次いでヤギ抗ヒト抗を標識二次抗体を加え、室温下で1時間インキュベートする。インキュベートした後、マイクロタイタープレートを3回洗浄し、次いでTMBを加えて5~15分間発色させ、最後に2M硫酸で停止させ、マイクロプレートリーダーでデータを記録する。
B7-H3を過剰発現するCHOK細胞またはB7-H3を内因的に発現するNCIH322細胞を丸底96ウェルプレートにウェルあたり100000細胞で加え、遠心分離して上清を除去し、次いで勾配希釈した抗B7-H3抗体を加え、4℃下で1時間インキュベートする。インキュベートした後、FACS緩衝液(2%FBSを含むPBS)で細胞を2回洗浄し、次いで蛍光標識したヤギ抗ヒト二次抗体(Abcam、ab98596)を加え、30分間インキュベートする。インキュベートした後にFACS緩衝液で細胞を2回洗浄し、FACS緩衝液で細胞を再懸濁した後にフローサイトメーター(Beckman、cytoFlex)で検出する。
B7-H3を過剰発現するCHOK細胞またはB7-H3を内因的に発現するNCIH322細胞を96ウェルプレートに播種し、37℃下でインキュベーター一晩培養する。まず毒素を含む二次抗体(ATS、IT51)で抗B7-H3抗体を標識し、次いで抗体を細胞に加え、37℃下でインキュベーターで3日間培養する。抗体がエンドサイトーシスされる場合、抗体に標識された毒素は、抗体のエンドサイトーシスとともに細胞に入り、細胞に毒性を引き起こし、細胞の生存率を低下させ、最後に細胞毒性検出キット(Promega、G3581)で細胞の活性を検出する。
カップリングエフェクター分子としてMMAE(Monomethyl auristatin E)を使用し、前述で調製した抗体IL-P7-C05-H4L3を使用して抗体薬物コンジュゲートIL-P7-C05-H4L3-MMAEを構築し、インビトロ薬力学評価を実施する。
ヒト非小細胞肺がんNCI-H1975細胞BALB/cヌードマウス皮下移植腫瘍モデルにおいて、IL-P7-C05-H4L3-MMAEおよびアイソタイプ対照Isotype-MMAEの単回投与による腫瘍増殖の阻害効果を検出する。
結果は、1mg/kgおよび3mg/kgの用量で尾静脈から1回投与されたIL-P7-C05-H4L3-MMAEは、NCI-H1975皮下移植腫瘍の増殖に対して有意な阻害効果を示し(P<0.01)、実験終了後(投与後21日)の腫瘍成長阻害率(TGI)は、それぞれ78.4%および101.8%であり、二つの投与群の相対腫瘍増殖率(%T/C)は、それぞれ26.2%および4.2%であり、3mg/kg投与群の腫瘍は、縮小し、消失する。3mg/kgの投与量下でアイソタイプ対照Isotype-MMAEを尾静脈から単回投与した後、溶媒対照群と比較して腫瘍成長に統計学的差はない(P>0.05)。1mg/kgおよび3mg/kgの用量でのIL-P7-C05-H4L3-MMAEの単回投与、および3mg/kgの用量でのアイソタイプ対照Isotype-MMAEの単回投与は、動物の体重変化に有意な影響を及ぼさず、明らかな異常な臨床徴候は見られず、動物は十分に耐える。

SEQ ID NO:3n+1に示されるVH-CDR2、および
SEQ ID NO:3n+2に示されるVH-CDR3の三つの相補性決定領域CDRを含み、
ここで、各nは、独立して、11、12、13、14、15、16、または17であり、
または、前記重鎖可変領域は、
SEQ ID NO:97に示されるVH-CDR1、
SEQ ID NO:54に示されるVH-CDR2、および
SEQ ID NO:35に示されるVH-CDR3、
もしくは
SEQ ID NO:33に示されるVH-CDR1、
SEQ ID NO:54に示されるVH-CDR2、および
SEQ ID NO:35に示されるVH-CDR3、
もしくは
SEQ ID NO:36に示されるVH-CDR1、
SEQ ID NO:55に示されるVH-CDR2、および
SEQ ID NO:56に示されるVH-CDR3、
もしくは
SEQ ID NO:57に示されるVH-CDR1、
SEQ ID NO:40に示されるVH-CDR2、および
SEQ ID NO:41に示されるVH-CDR3、

SEQ ID NO:51に示されるVH-CDR1、

ここで、前記アミノ酸配列におけるいずれか一つのアミノ酸配列は、任意選択で少なくとも一つのアミノ酸が付加、欠失、修飾および/または置換され、かつB7-H3結合親和性を保持できる誘導配列をさらに含むことを特徴とする、前記抗体の重鎖可変領域。 - 抗体の重鎖であって、
前記重鎖は、請求項1に記載の重鎖可変領域を有することを特徴とする、前記抗体の重鎖。 - 抗体の軽鎖可変領域であって、
前記軽鎖可変領域は、
SEQ ID NO:3m+1に示されるVL-CDR1、
SEQ ID NO:3m+2に示されるVL-CDR2、および
SEQ ID NO:3m+3に示されるVL-CDR3の三つの相補性決定領域CDRを含み、
ここで、各mは、独立して、0、1、2、3、4、5、6、または7であり、
または、前記軽鎖可変領域は、
SEQ ID NO:1に示されるVL-CDR1、
SEQ ID NO:25に示されるVL-CDR2、および
SEQ ID NO:3に示されるVL-CDR3、
もしくは
SEQ ID NO:1に示されるVL-CDR1、
SEQ ID NO:26に示されるVL-CDR2、および

ここで、前記アミノ酸配列におけるいずれか一つのアミノ酸配列は、任意選択で少なくとも一つのアミノ酸が付加、欠失、修飾および/または置換され、かつB7-H3結合親和性を保持できる誘導配列をさらに含むことを特徴とする、前記抗体。 - 前記抗体は、請求項1に記載の重鎖可変領域および請求項3に記載の軽鎖可変領域を有し、ここで、

前記組換えタンパク質は、
(i)請求項1に記載の重鎖可変領域、請求項2に記載の重鎖、請求項3に記載の軽鎖可変領域、請求項4に記載の軽鎖、または請求項5~7のいずれか1項に記載の抗体、および
(ii)発現および/または精製を補助する任意選択のタグ配列を含むことを特徴とする、前記組換えタンパク質。 - 抗体コンジュゲートであって、
当該抗体コンジュゲートは、
(a)請求項1に記載の重鎖可変領域、請求項2に記載の重鎖、請求項3に記載の軽鎖可変領域、請求項4に記載の軽鎖、または請求項5~7のいずれか1項に記載の抗体、またはその組み合わせからなる群から選択される抗体部分、および
(b)検出可能な標識、薬物、毒素、サイトカイン、放射性核種、酵素、またはその組み合わせからなる群から選択される前記抗体部分とカップリングされるカップリング部分を含むことを特徴とする、前記抗体コンジュゲート。 - 有効成分の用途であって、
前記有効成分は、請求項1に記載の重鎖可変領域、請求項2に記載の重鎖、請求項3に記載の軽鎖可変領域、請求項4に記載の軽鎖、請求項5~7のいずれか1項に記載の抗体、請求項8に記載の組換えタンパク質、または請求項9に記載の抗体コンジュゲート、またはその組み合わせからなる群から選択され、ここで、前記有効成分は、(a)診断試薬またはキットの調製、および/または(b)B7-H3の発現または機能異常に関連する疾患を予防および/または治療するための薬物の調製に使用されることを特徴とする、前記有効成分の用途。
