実施形態1は、異種導入遺伝子に作動可能に連結されたヨザルヘルペスウイルス(Aotine Herpesvirus)主要最初期プロモーター(AoHV−1プロモーター)を含む組換え核酸分子である。
a)導入遺伝子に作動可能に連結されたAoHV−1プロモーターを含むコンストラクトを調製する工程;および
b)前記コンストラクトを組換えアデノウイルスのゲノムに組み込む工程
を含む方法である。
細胞培養:
HEK293細胞を、10%ウシ胎児血清(FBS)を含むダルベッコ改変イーグル培地(DMEM)中で維持した。ベロ細胞を、5%ウシ胎児血清(FBS)を補充した最小必須培地(MEM、Life Technologies)中で培養した。PER.C6細胞(Fallaux et al.,1998)を、10mM MgCl2を補充した10%ウシ胎児血清(FBS)を含むDMEM中で培養した。
異なるプロモーターコンストラクトを、AvrIIおよびHindIII制限酵素認識部位を用いて、pAdApt35(Vogels et al.2007)にクローニングした。ホタルルシフェラーゼをコードする遺伝子を、HindIIIおよびXbaI制限酵素認識部位を用いて挿入し、異なる試験プロモーター(pAdapt35.P.Luc)下でLuc遺伝子が発現するpAdaptプラスミドを得た。これらのプラスミドを実施例1で使用した。
HEK293細胞をマルチウェル6ポリ−L−リシン(PLL)コーティングプレートに播種し、Lipofectamineを製造業者(Life Technologies)によって提供された使用説明書に従って使用して、1μgのpAdapt.P.Lucプラスミドでトランスフェクトした。その後、プレートを37℃、10%CO2中で24時間インキュベートした。トランスフェクトした24時間後、ルシフェラーゼ活性を、Luminoskan(商標)Ascentマイクロプレート照度計において、0.1%DTT(1M)の存在下、細胞溶解物中で測定した。トランスフェクション効率の対照として、分泌胚性アルカリホスファターゼをコードするプラスミドを同時トランスフェクトした(80ng/ウェル)。SEAPレベルを、Clontech Great EscAPe(商標)SEAP化学発光キット2.0およびTrilux Microbetaワークステーションを用いて測定した。一過性にトランスフェクトしたHEK293細胞における発現分析を、実施例1において使用する。
ベロ細胞を、1.25×104細胞/ウェルの播種密度でマルチウェル96プレートに播種した。播種の1日後に、7xtetO−AoHV−1の制御下で、細胞をガウシアルシフェラーゼ遺伝子を含むpDualLuc由来プラスミドでトランスフェクトした。ルシフェラーゼプラスミドの他に、テトラサイクリン制御トランスアクチベーター(tTA)タンパク質をコードするプラスミド、または陰性対照プラスミド(pBluescript)で、細胞を同時トランスフェクトした。細胞トランスフェクションは、Lipofectamine 3000を用いて、製造業者のプロトコル(Life Technologies)に従って行った。トランスフェクションの24時間後に、ルシフェラーゼ活性を、Luminoskan(商標)Ascentマイクロプレート照度計において、Pierceガイウシアホタルルシフェラーゼデュアルアッセイキット(Thermo Scientific)を用いて測定した。トランスフェクション効率の照合のために、測定されたガウシアルシフェラーゼ単位を、赤色ホタルルシフェラーゼ(AoHV−1ショートによって駆動される発現カセットを、同じpDualLucプラスミドに位置させた)のレベルに正規化した。一過性にトランスフェクトしたベロ細胞における発現分析を、実施例2において行った。
PER.C6細胞を、5×104細胞/ウェルの播種密度でマルチウェル96プレートに播種した。播種の1日後に、実施例6に記載のように、異なるAoHV−1プロモーター変異体の制御下で、ガウシアルシフェラーゼ遺伝子を含むpDualLuc由来プラスミドを使用して細胞をトランスフェクトした。細胞トランスフェクションは、Lipofectamine 2000 CDを用いて、製造業者のプロトコル(Life Technologies)に従って行った。トランスフェクションの24時間後に、ルシフェラーゼ活性を、Pierceガイウシアホタルルシフェラーゼデュアルアッセイキット(Thermo Scientific)およびLuminoskan(商標)Ascentマイクロプレート照度計を用いて測定した。トランスフェクション効率の照合のために、測定されたガウシアルシフェラーゼ単位を、赤色ホタルルシフェラーゼ(AoHV−1ショートによって駆動される発現カセットを、同じpDualLucプラスミドに位置させた)のレベルに正規化した。この方法の項は、実施例6に適用可能である。
TetRを安定に発現する2種類のPER.C6細胞を作製し、ここで使用した。まず、hCMVプロモーターの制御下でTetRを発現するPER.C6−hCMV.TetR細胞を、プラスミドpCDNA6/TR(Thermo Fisher Scientific カタログ番号:V102520)を用い、製造業者のプロトコルに記載されている標準手順に従って、PER.C6細胞を安定にトランスフェクトすることによって生成した。次に、AoHV−1ショートプロモーターの制御下でTetRを発現するPER.C6−AoHV.TetR細胞を、実施例5(および、そこで言及されている方法の項)に記載の手順に従って、PER.C6細胞をプラスミドpC_AoHV_TetRで安定にトランスフェクションすることによって生成した。
PER.C6−hCMV.TetR細胞を、トランスフェクションの1日前に、T80培養フラスコに播種した。トランスフェクションに使用したプラスミドは、AoHV.2×tetOプロモーター(配列番号7)の制御下で目的の遺伝子(GOI)を担持した。さらに、それは、SV40由来のプロモーターによって制御されるネオマイシンホスホトランスフェラーゼII遺伝子発現カセットを含有する。プラスミドは、いくつかのクローニング工程により、pcDNA2004Neo(−)(GenBankアクセッション番号 FB674876)のhCMVプロモーター含有MfeI−XbaI断片を、AoHV.2×tetOを含む断片で置き換え、続いてGOIのコード配列で置き換えることによって構築した。トランスフェクションを、Lipofectamin 2000を用いて製造業者のプロトコル(Invitrogen)に従って行った。トランスフェクションの翌日、細胞を10cm培養皿に1:2、1:4、1:8および1:16の比率で継代し播種した。播種の1日後、細胞培養培地を、0.5mg/mlのジェネティシン(Gibco 10131−019)を補充したPER.C6−hCMV.TetR培地と交換した。培地を週2回交換した。トランスフェクションの2〜3週間後にクローンを採取した。この方法の項は、実施例3に適用可能である。
播種した細胞を、ドキシサイクリン(1μg/ml)と共に、またはドキシサイクリンなしで2日間インキュベートし、RIPA緩衝液(150mM NaCl、1%triton X100、0.5%デオキシコール酸塩、0.1%SDS、50mM トリス−HCL)に回収した。
pDualLuc(配列番号21)は、2つの発現カセット、すなわちヒトサイトメガロウイルス(hCMV)プロモーターの制御下でガウシアルシフェラーゼ(Gluc)を発現する第1の発現カセット、およびAoHV−1ショートプロモーターの制御下で赤色発光ホタルルシフェラーゼ(RFL)を発現する第2のカセットを担持するプラスミドである。pDualLuc内では、GlucおよびRFL発現カセットが互いに対してテールトゥテール配向に位置付けられている。
以前に生成されたAd26ベースのベクターについて記載されているように、pAd26.dE1.dE3.5orf6は、E1欠失、E3欠失、およびAd26 E4 orf6のAd5のそれによる置換を担持する完全長のAd26ベクターゲノムを含むプラスミドである(Abbink et al.,2007)。pAd26.dE1.dE3.5orf6のプラスミド骨格は、pMB1複製起点およびアンピシリン耐性遺伝子を含み、pBR322(GenBankアクセッション番号 J01749.1)に由来する。pAd26.dE1.dE3.5orf6はさらに、ベクターゲノムのE1欠失の代わりに特有のAsiSI制限部位を含み、ベクターゲノムのE4領域と右逆位末端配列(RITR)との間に特有のPacI制限部位を含む。これらの部位を用いて、それぞれの位置に導入遺伝子カセットを挿入することができる。最後に、pAd26.dE1.dE3.5orf6内のアデノウイルスベクターゲノムはその末端の各々にSwaI制限部位が隣接され、SwaI消化によるプラスミド骨格からのその放出を可能にする。
pAd26.CMV_GLuc.AoHV1_RFLは、pAd26.dE1.dE3.5orf6のAd26ベクターゲノムを含むプラスミドであり(上記を参照)、以下の2つの導入遺伝子発現カセットを有するように改変されている:(1)(欠失)E1領域に挿入されたヒトサイトメガロウイルス(hCMV)プロモーター駆動ガウシアルシフェラーゼ(Gluc)発現カセット、および(2)E4とRITRとの間に挿入されたAoHV−1プロモーター駆動赤色蛍光ホタルルシフェラーゼ(RFL)発現カセット。

pC_AoHV_TetRを構築するために、AoHV−1ショート(配列番号30)の後にコドン最適化TetRコード配列を含む1.3Kbp DNA(配列番号20)断片を合成し(GeneArt/LifeTechnolgiesにおいて)、続いてMfeIおよびXbaI制限部位を用いてpcDNA2004Neo(−)(GenBankアクセッション番号 FB674876)にサブクローニングした。図6Aは、1.3Kbp挿入の位置が示されているpC_AoHV_TetRのマップを示す。
安定なPER.C6−AoHV.TetR細胞株を、標準的な細胞株生成手順によって生成した。簡潔に記載すると、4mM L−グルタミン(Lonza)を補充したCDM4 PerMAb培地(HyClone)中で増殖させた無血清増殖懸濁適合PER.C6細胞(Fallaux et al.,1998)を、BioRadエレクトロポレーターを使用するエレクトロポレーションによってプラスミドpC_AoHV_TetRでトランスフェクトした。「PerMAb選択培地」、すなわち、4mM L−グルタミンおよび125μg/mLジェネティシン(Gibco)を補充したCDM4 PerMAb培地中での回収および抗生物質選択の後、トランスフェクトされた細胞を、単一細胞クローニングのために、4mM L−グルタミンおよび125μg/mLジェネティシンを補充したMAb培地(SAFC)に入ったMW96プレートに1ウェルあたり1生細胞で播種した。細胞コロニーが形成されたところで、PerMAb選択培地中、静的培養で、単離されたクローンを数継代培養して増殖させた。続いて、100個のクローンを、抗TetR抗体(Tet03、MoBiTec)を用いるフローサイトメトリー細胞内染色手順によって、TetRを発現する能力について分析した。この最初のスクリーニングにおいて、試験した100個のクローンのうち94個がTetR陽性であることがわかった。これらのTetR陽性クローンのうち47個を選択し、4mMのL−グルタミンおよび125μg/mLのジェネティシンを補充したPermexcis培地(Lonza)中での増殖に適応させた後、フローサイトメトリー細胞内染色手順によるTetR発現、ならびにTetR機能アッセイによるTetR活性について再び試験した(本明細書の方法の項に記載)。次いで、30時間以下の集団倍加時間を示し、広範囲のTetR活性レベルを示す24個のTetR陽性クローンを選択した。PER.C6−AoHV.TetRクローン#1〜#24と示されるこれらのクローンを、振盪フラスコ中で増殖するように(再)適合され、その後、最終的に小規模研究細胞バンク中に凍結保存された。この方法の項は、実施例5に適用可能である。
長期継代を行った際のPER.C6−AoHV.TetR細胞クローンによるTetR発現の安定性の分析。振盪フラスコ中、抗生物質選択が行われた場合と行われなかった場合で、PER.C6−AoHV.TetR細胞クローンを、約60回の集団倍加(または世代)に達するまで、複数継代、増殖させた。TetR発現を、第20代、第40代、および第60代で、フローサイトメトリー細胞内染色によって分析した。これらの時点の各々において、各クローンの細胞をフローサイトメトリー細胞内二重染色プロトコルに供して、TetRおよびチューブリンの両方を検出した。染色は、標準的なフローサイトメトリー細胞内手順に従い、そして2つの抗体インキュベーション工程、すなわち抗TetRマウスモノクローナル抗体(Tet03、MoBiTec)を用いる第1の工程、および抗マウスIgG−FITC複合体(554001、BD Biosciences)を抗チューブリン抗体−Alexa Fluor 647複合体(ab195884、Abcam)と組み合わせて用いる第2の工程を含んだ。クローン毎に、TetR陽性細胞およびTetR陰性細胞の画分を、ストリンジェントなチューブリン陽性ゲート内で決定した。この方法の項は、実施例5に適用可能である。
TetR機能アッセイを、PER.C6−AoHV.TetR細胞クローンの長期継代後に行った。振盪フラスコ中、抗生物質選択が行われた場合と行われなかった場合で、PER.C6−AoHV.TetR細胞クローンを、約60回の集団倍加(または世代)に達するまで、複数継代、増殖させた。その後、PER.C6−AoHV.TetR細胞クローンおよび対照PER.C6細胞を、4mMのL−グルタミンを補充したPermexcis培地の入ったポリ−L−リシン被覆マルチウェル96プレートに播種し、2時間のインキュベーション後、ベクターAd26.CMV_GLuc.AoHV1_RFLおよびAd26.CMVtetO_GLuc.AoHV1_RFL(実施例4に記載)によって4連で感染させた。感染後1日目に、感染細胞をルシフェラーゼ細胞溶解緩衝液(Pierce/Thermo Scientific)中に回収した。続いて、各細胞溶解物の別々のアリコートを使用して、GLucおよびRFL活性を、それぞれ、BioLuxガウシアルシフェラーゼアッセイキット(NEB)およびルシフェラーゼアッセイキット(Pierce/Thermo Scientific)試薬、ならびにLuminoskan Ascentマイクロプレート照度計(Thermo Scientific)を、製造業者の使用説明書に従って使用して測定した。感染複製毎に、Gluc活性をRFL活性によって正規化した。この方法の項は、実施例5に適用可能である。
ウイルスまたは動物/ヒトゲノムに由来するプロモーターは、異種タンパク質発現のために、例えば、細胞または宿主生物において目的の遺伝子を発現するためのプラスミドベクターまたはウイルスベクターにおける発現カセットにおいて頻繁に使用される。この目的のために一般的に使用されるプロモーターは、ヒトサイトメガロウイルス(hCMV)の最初期領域に由来するプロモーターである(Powell et al.,2015)。他の宿主に感染するいくつかのサイトメガロウイルス(CMV)もまた知られている。それらは、例えば、アカゲザル(rhCMV)(Barry,Alcendor,Power,Kerr,& Luciw,1996;Chan,Chiou,Huang,& Hayward,1996;Chang et al.,1993;Hansen,Strelow,Franchi,Anders,& Wong,2003)およびチンパンジー(chCMV)(Chan et al.1996)に感染する。
遺伝子の誘導発現は、バイオテクノロジーの分野、特に毒性遺伝子産物の一過性発現に高い適用性を有し得る。哺乳動物細胞における異種遺伝子調節のための特定のよく確立された系は、hCMVコアプロモーターに連結されたTetオペレーター(TetO)配列の多数のコピーからなる人工プロモーターを利用する(Gossen & Bujard,1992;Gossen et al.,1995)。これらの系において、基礎転写活性を全く示さないか、または非常に低い基礎転写活性を示す前記人工プロモーターは、テトラサイクリン制御性トランスアクチベータータンパク質(tTA)またはリバーステトラサイクリン制御性トランスアクチベータータンパク質(rtTA)として知られる特定の組換え転写因子のTetO配列に特異的に結合することによって活性化され得る。ここでは、hCMVコアプロモーターの代わりに、AoHV−1プロモーターの配列エレメントを用いて、同様のtTA/rtTA応答性人工プロモーターを生成することができるか否かを試験した。
実施例2で述べたとおり、言及したように、細胞株における、例えば、目的の毒性遺伝子の調節発現は、タンパク質またはウイルスベクターの組換え産生に必須であり得る。hCMVプロモーターを有する発現カセットを既に含むウイルスベクターを使用する場合、産生細胞系における目的の遺伝子(GOI)の発現のためには異なる配列を有する強力なプロモーターを使用することが望ましい。hCMVプロモーターの代替物を同定するために、AoHV−1ショートプロモーターおよびヒトホスホグリセリン酸キナーゼ1(hPGK)プロモーター(バイオテクノロジー発現系で使用するために記載された別のプロモーター)を、PER.C6細胞におけるeGFPの一過性発現について試験した。実施例1の結果と一致して、AoHV−1ショートプロモーターは再び、異種遺伝子を発現するための強力なプロモーターであり、eGFP発現レベルの駆動が、(配列番号4の)hCMVと同程度で、hPGKプロモーター(配列番号6)よりもはるかに高いことを示した(図4A)。
AoHV−1ショートプロモーター(AoHV−1ショート)は、ワクチン抗原または他の目的のタンパク質の発現を駆動するウイルスベクターに適用することができる。これは、アデノウイルスベクターAd26.CMV_GLuc.AoHV1_RFLおよびAd26.CMVtetO_GLuc.AoHV1_RFL.との関連におけるAoHV−1ショートの適用によって本明細書中で説明される。これらはいずれも、2つの発現カセット、すなわち(欠失)E1領域の代わりに挿入されたガウシアルシフェラーゼ(GLuc)をコードする第1のカセット、およびE4領域と右の逆位末端配列(RITR)との間に位置する赤色蛍光ホタルルシフェラーゼ(RFL)をコードする第2のカセットを担持する、2つのAd26ベースの組換えアデノウイルスベクターである。2つのウイルスのGluc発現カセットは、それぞれ、hCMVプロモーター(配列番号4)およびCMVtetO(配列番号15)によって駆動される。CMVtetOは、2つのテトラサイクリンオペレーター(TetO)配列モチーフを含む配列の挿入を担持し、このプロモーターがテトラサイクリンリプレッサー(TetR)タンパク質によって抑制可能であるという点で、hCMVプロモーターとは異なる。両ウイルスにおいて、RFL発現カセットは、AoHV−1ショート(配列番号30)の制御下にある。2つのベクターのゲノムの概略図については、図5Aを参照されたい。
AoHV−1プロモーターは、安定な細胞株における異種遺伝子の発現を駆動するために適用することができる。本明細書では、これを、TetR遺伝子発現がAoHV−1プロモーターによって駆動されるプラスミドであるpC_AoHV_TetRを用いて、TetR発現細胞株を生成することによって説明する。
異種遺伝子を発現するためのAoHV−1主要最初期調節領域の必須部分をマッピングするために、AoHV−1プロモーターの異なるバージョンのパネルを作製した。異なる設計を図7Aに示す。この短いプロモーター配列の必須部分をマッピングするために、既に非常に短いAoHV−1ショート配列をトランケートして、2つの設計を行った。他の設計は、AoHV−1ショートの効力が増大され得るか否かを試験するために、さらなる推定ドメインおよび細胞因子結合部位、ならびに予測されるイントロン配列を含む。

上記のように、短くかつ強力なプロモーターが、ヨザルヘルペスウイルス1型(Aotine herpesvirus−1)から特定された。AoHV−1プロモーターは短い配列を有し、かつhCMVプロモーターに匹敵する効力を有する。これらの特徴は、AoHV−1プロモーターを、hCMVプロモーターが使用される状況下で代替として使用する理想的なプロモーターにする。さらに、AoHV−1プロモーターはhCMVプロモーターに対する配列同一性が比較的低く、これは、AoHV−1プロモーターを、同じウイルスベクターから2つの異なる導入遺伝子を発現する場合、または同様の配列を含み得るプロデューサー細胞株を用いてウイルスを生成する場合、hCMVプロモーターと組み合わせて使用するのに適したものとする。AoHV−1プロモーターはまた、AoHV−1プロモーターからの転写の調節に使用され得る調節配列と共に使用するのに好適である。

Claims (25)

- 前記AoHV−1プロモーターは、1つ以上のテトラサイクリンオペレーター配列(tetO部位)を含む調節配列に作動可能に連結されている、請求項1〜3のいずれか一項に記載の核酸分子。
- 請求項1〜4のいずれか一項に記載の核酸分子を含むベクターまたはウイルス。
- 前記ベクターはプラスミドベクターである、請求項5に記載のベクター。
- 前記ウイルスはアデノウイルスである、請求項5に記載のウイルス。
- 前記アデノウイルスは、そのゲノムに、好ましくはそのゲノムの少なくともE1領域に、少なくとも1つの欠失を有する、請求項7に記載のアデノウイルス。
- 請求項1〜4のいずれか一項に記載の核酸分子、または請求項5〜8のいずれか一項に記載のベクターもしくはウイルスを含む細胞。
- そのゲノム中にAoHV−1プロモーターを含む核酸分子を含み、好ましくは、前記プロモーターは目的のタンパク質をコードする核酸に作動可能に連結されている細胞。
- 前記AoHV−1プロモーターは、配列番号25のnt237〜286に対して少なくとも80%の同一性を有する配列を含み、好ましくは、前記AoHV−1プロモーターは、配列番号25のヌクレオチド201〜286と少なくとも少なくとも95%同一である配列を含む、請求項10に記載の細胞。
- 前記ゲノム中の前記AoHV−1プロモーターは、テトラサイクリンリプレッサー(TetR)タンパク質をコードする核酸に作動可能に連結されており、好ましくは前記細胞はPER.C6細胞である、請求項10または11に記載の細胞。
- (i)第1の導入遺伝子に作動可能に連結されたAoHV−1プロモーターを含む核酸分子、および(ii)第2の導入遺伝子に作動可能に連結されたhCMVプロモーターを含む核酸分子を含む細胞。
- 前記AoHV−1プロモーターは、配列番号25のnt237〜286に対して少なくとも80%の同一性を有する配列を含み、好ましくは、前記AoHV−1プロモーターは、配列番号25のヌクレオチド201〜286と少なくとも少なくとも95%同一である配列を含み、かつ前記hCMVプロモーターは、配列番号4のnt578〜736に対して、好ましくはnt564〜736に対して少なくとも95%の同一性を有する配列を含む、請求項13に記載の細胞。
- 前記第1の導入遺伝子はTetRをコードし、好ましくは、前記細胞はPER.C6細胞である、請求項13または14に記載の細胞。
- 第2の導入遺伝子に作動可能に連結されたhCMVプロモーターを含む前記核酸分子は、アデノウイルスの一部である、請求項15に記載の細胞。
- (i)第1の導入遺伝子に作動可能に連結されたAoHV−1プロモーターを含む核酸分子、および(ii)第2の導入遺伝子に作動可能に連結されたhCMVプロモーターを含む核酸分子を含むベクター。
- 前記AoHV−1プロモーターは、配列番号25のnt237〜286に対して少なくとも80%の同一性を有する配列を含み、好ましくは、前記AoHV−1プロモーターは、配列番号25のヌクレオチド201〜286と少なくとも少なくとも95%同一である配列を含む、請求項17に記載のベクター。
- 目的の発現産物を生産する方法であって、請求項1〜8のいずれか一項に記載の核酸分子からの導入遺伝子を宿主細胞で発現させる工程であって、前記導入遺伝子は前記目的の発現産物をコードする工程を含む方法。
- 前記発現産物はタンパク質である、請求項19に記載の方法。
- 前記宿主細胞から、または前記宿主細胞が培養されている培養培地から、または前記宿主細胞および前記培養培地の両方から、前記目的の発現産物を回収する工程をさらに含む、請求項19または20に記載の方法。
- ウイルスを作製する方法であって、前記ウイルスを増殖させる機能を有する異種遺伝子を発現させる細胞中で前記ウイルスを増殖させる工程であって、前記異種遺伝子は、AoHV−1プロモーターの制御下にある工程、および前記細胞から、または前記細胞が培養されている培養培地から、または前記細胞および前記培養培地の両方から、前記ウイルスを回収する工程を含む方法。
- 前記AoHV−1プロモーターは、配列番号25のnt237〜286に対して少なくとも80%の同一性を有する配列を含み、好ましくは、前記AoHV−1プロモーターは、配列番号25のヌクレオチド201〜286と少なくとも少なくとも95%同一である配列を含む、請求項22に記載の方法。
- 請求項5〜8のいずれか一項に記載のベクターまたはウイルス、および薬学的に許容される担体または賦形剤を含む医薬組成物。
- アデノウイルスを調製する方法であって、請求項12に記載の細胞中で、1つ以上のtetO部位に作動可能に連結されたhCMVプロモーターの制御下で、導入遺伝子をコードするアデノウイルスを増殖させる工程、および、好ましくは、前記アデノウイルスを単離する工程を含む方法。
