Abstract
Description
[1] 構造タンパク質を含む組成物であって2%以上7%未満の水分率を有する上記組成物を加熱および加圧する成形工程を含む、モールド成形体の製造方法。
[2] 上記成形工程に先立って、上記構造タンパク質を含む組成物の水分率を2%以上7%未満に調整する水分率調整工程を含む、[1]に記載のモールド成形体の製造方法。
[3] 上記構造タンパク質がクモ糸フィブロインである、[1]または[2]に記載のモールド成形体の製造方法。
構造タンパク質とは、生体構造を構築する役割を有するタンパク質であり、酵素、ホルモン、抗体等の機能タンパク質とは異なる。構造タンパク質としては、例えば、天然に存在するフィブロイン、コラ−ゲン、レシリン、エラスチン及びケラチン等の天然型構造タンパク質を挙げることができる。天然に存在するフィブロインとして、昆虫及びクモ類が産生するフィブロインが知られている。本実施形態において、構造タンパク質はスパイダーシルクタンパク質を含むことが好ましい。
組換えポリペプチドの製造方法について、以下に詳述する。目的とする組換えポリペプチドは、例えば、構造タンパク質をコードする遺伝子配列と、当該遺伝子配列に作動可能に連結された1又は複数の調節配列とを有する発現ベクターで形質転換された宿主により、当該遺伝子を発現させることにより生産することができる。
本実施形態に係るモールド成形体の製造方法は、たとえば上述のようにして得られる構造タンパク質を含む組成物を加熱および加圧する成形工程を含む。この成形工程において、組成物は、2%以上7%未満の水分率を有する。本実施形態に係るモールド成形体の製造方法は、成形工程に先立って、組成物の水分率を2%以上7%未満に調整する水分率調整工程を含んでもよい。水分率調整工程が最初に実施されることにより、構造タンパク質が得られた時点における組成物の水分率に関わらず、成形温度を低く抑えつつ曲げ強度の高いモールド成形体を確実に得ることができる。
後述する実施例で用いたのと同じタンパク質粉末を用いて、タンパク質粉末が樹脂化するタイミングと、加圧成形機の内圧の変化との関係を確認した。より詳細には、上記特許文献2(国際公開第2018/043698号)に記載された加圧成形機と同じ構成を有する加圧成形機を用い、実験用に、金型にガラス窓を設けて金型内部を可視化する改造を施した。さらに、金型の内面に圧力センサーを設置した。
a)タンパク質粉末(Spiber(株)製改変フィブロイン:PRT410)を金型内に投入
b)型締めを行い、内圧が40MPaになるように型締力を調整し、金型の内圧値をリセット(ゼロにする)
c)昇温を開始し(設定40℃/min)、金型内の寸法が変わらないよう型締力を調整
このとき、内圧の上昇が見られた。
d)樹脂化の完了を目視にて確認し、その時点から、金型の温度を5分間保持
樹脂化の確認は、金型内部の色に基づいて行った。粉末の状態では、金型内部の色は不透明の白色であり、樹脂化が始まると金型内部の色が変化し始め、樹脂化が完了すると、金型内部の色は半透明の薄茶色になった。
e)上記樹脂化完了後、金型の温度が80℃になるまで、スポットクーラーにて冷却
f)金型を開放し、モールド成形体を取出し
g)20℃/60%RHの環境下で24時間保管後、後述の曲げ試験を実施
続いて、後述する実施例で用いたのと同じタンパク質粉末を用いて水分率の調整を行い、水分率の違いによる樹脂化温度を確認した。具体的な実験手順としては、まず、加熱乾燥式水分計((株)エー・アンド・デイ製、MS−70)を用いて、1分間の重量変化率が0.01wt%以下になるまで130℃で粉末を乾燥し、この状態をもって水分率0%(すなわち水分が抜けきった状態)とした。続いて、23℃、60%RHの雰囲気下で3分間放置した粉末の水分率を測定したところ2.6%であった。同様に、5分間放置した粉末の水分率を測定したところ5.2%であり、7分間放置した粉末の水分率を測定したところ8.0%であり、20分間放置した粉末を測定したところ10.7%であった。
ネフィラ・クラビペス(Nephila clavipes)由来のフィブロイン(GenBankアクセッション番号:P46804.1、GI:1174415)の塩基配列及びアミノ酸配列をGenBankのウェブデータベースより取得した後、生産性の向上を目的としてアミノ酸残基の置換、挿入及び欠失を施し、さらにN末端に配列番号5で示されるアミノ酸配列(タグ配列及びヒンジ配列)を付加して、配列番号12で示されるアミノ酸配列を有する組換えフィブロイン(「PRT410」ともいう。)を設計した。
上記で得られたPRT410をコードする遺伝子を含むpET22b(+)発現ベクターで、大腸菌BLR(DE3)を形質転換した。形質転換された大腸菌を、アンピシリンを含む2mLのLB培地で15時間培養後、同培養液を、表1に示すシード培養用培地100mLに、OD600が0.005となるように添加した。培養液の温度を30℃に保ち、OD600が5になるまでフラスコにて、さらに約15時間培養を行い、シード培養液を得た。
IPTGを添加してから2時間後に回収した菌体を20mM Tris−HCl緩衝液(pH7.4)で洗浄した。洗浄後の菌体を約1mMのフェニルメチルスルホニルフルオリド(PMSF)を含む20mM Tris−HCl緩衝液(pH7.4)に懸濁させ、高圧ホモジナイザー(GEA Niro Soavi社)で細胞を破砕した。破砕した細胞を遠心分離し、沈殿物を得た。得られた沈殿物を、高純度になるまで20mM Tris−HCl緩衝液(pH7.4)で洗浄した。洗浄後の沈殿物を100mg/mLの濃度になるように8M グアニジン緩衝液(8Mグアニジン塩酸塩、10mMリン酸二水素ナトリウム、20mM NaCl、1mM Tris−HCl、pH7.0)で懸濁し、60℃で30分間、スターラーで撹拌し、溶解させた。溶解後、透析チューブ(三光純薬株式会社製のセルロースチューブ36/32)を用いて水で透析を行った。透析後に得られた白色の凝集タンパク質を遠心分離により回収し、凍結乾燥機で水分を除き、凍結乾燥粉末を回収した。
次に、得られた凍結乾燥粉末の水分を調整した。水分率の調整は、上述の予備実験と同様に行い、水分率0%、2.6%、5.2%、8.0%、10.7%のクモ糸フィブロイン粉末(構造タンパク質粉末)をそれぞれ得た。
上記のように作製した各成形体に対し、卓上形精密万能試験機((株)島津製作所製、オートグラフAG−20)を用いて三点曲げ試験を行った。支点間距離を27mm、測定速度を1mm/分とした。
Claims (3)
- 構造タンパク質を含む組成物であって2%以上7%未満の水分率を有する前記組成物を加熱および加圧する成形工程を含む、モールド成形体の製造方法。
- 前記成形工程に先立って、前記構造タンパク質を含む組成物の水分率を2%以上7%未満に調整する水分率調整工程を含む、請求項1に記載のモールド成形体の製造方法。
- 前記構造タンパク質がクモ糸フィブロインである、請求項1または2に記載のモールド成形体の製造方法。
